▼ O glutamato (Glu) é o neurotransmissor excitatório mais abundante do sistema nervoso central e está envolvido em funções cerebrais como aprendizado/memória, desenvolvimento e envelhecimento cerebral. Mudanças na vulnerabilidade neuronal durante o desenvolvimento pós-natal podem ser parcialmente explicadas por mudanças na expressão das subunidades de receptores de Glu (GluRs). O objetivo desse trabalho foi determinar o padrão de expressão das subunidades de GluRs do tipo N-metil-D-aspartato (NMDA) em quatro estruturas cerebrais (cerebelo, córtex, estriato e hipocampo) em diferentes estágios do desenvolvimento pós-natal de ratos por quantificação relativa por PCR em tempo real. Mudanças significativas na expressão de Grin1 foram observadas em quase todas as estruturas em relação aos níveis pós-natais de expressão. Níveis de expressão de Grin2A foram menores no cerebelo com um padrão de aumento gradual e níveis elevados na vida adulta no córtex e no hipocampo. A expressão do Grin2B demonstrou um padrão decrescente no cerebelo assim como no hipocampo. Por outro lado, o padrão de expressão de Grin2C mostrou uma tendência a aumentar nas quatro estruturas, a qual foi mais evidente no cerebelo com um aumento significativo nos níveis de expressão. O padrão de expressão do Grin2D foi também caracterizado por uma tendência para uma diminuição nos níveis de expressão pela vida adulta. Essa tendência foi mais evidente no cerebelo e no hipocampo. Expressões de Grin3A e Grin3B não demonstraram uma tendência específica. No caso de Grin3B, níveis semelhantes de expressão foram observados com a exceção de um pico de expressão no cerebelo de ratos de 10 dias. Os dados obtidos nesse estudo permitiram-nos introduzir novos aspectos na área de expressão de GluRs e poderão contribuir para desvendar potenciais alvos para o desenvolvimento de terapias mais efetivas para muitas doenças.
Glutamate (Glu) is the most abundant excitatory neurotransmitter of the central nervous system and is involved in cerebral functions such as learning/memory, development, and cerebral aging. The changes in vulnerability of neurons during postnatal development can be partially explained by changes in expression of glutamate receptors subunits (GluRs). In this work, we aimed to determine expression profile of ionotropic Nmethil- D-aspartate (NMDA) GluR subunits in four brain structures (cortex, cerebellum, striatum and hippocampus) in different stages of postnatal rat development by relative quantification real-time PCR using glyceraldehyde 3-phosphate dehydrogenase (Gapdh) as endogenous control. Significant changes in Grin1 expression were demonstrated in almost each age from all structures in relation to the initial postnatal expression level. Levels of expression of Grin2A were lower in cerebellum with a gradual raising pattern and high levels in adulthood in cortex and hippocampus. ▼ Omega-3 polyunsaturated fatty acids (PUFA) and vitamin
D both have anti-cancer effects through common and unique pathways. The hypothesis of this thesis is that the combination of
n-3 PUFA with 1,25(OH)2D3 will inhibit breast cancer cell growth in an additive or synergistic manner. A 3X3 factorial design was used to test the combinations of five PUFA treatments (α-linoleic acid (ALA, 18:3n3), eicosapentaenoic acid (EPA, 20:5n3) and docosahexaenoic acid (DHA, 22:6n3), γ-linolenic acid (GLA, 18:3n6) and arachidonic acid (AA, 20:4n6)) with 1,25(OH)2D3 on MCF-7, MDA-MB-231, and MCF-10A cell growth, and determine any potential synergism in combination treatments. MCF-7 and MCF-10A cells responded to PUFA and 1,25(OH)2D3 treatments, but combinations provided no potential synergism. MDA-MB-231 growth was not affected by 1,25(OH)2D3, while combinations treatments involving ALA, EPA, GLA, and AA caused potentially synergistic growth inhibition. This thesis presents the novel observation that PUFA are sensitizing MDA-MB-231 cells to 1,25(OH)2D3 treatment.

▼ N-acylethanolamines (NAEs) including anandamide are lipid derivative molecules, which play vital roles in physiological and developmental processes in plants and animals and mediate stress responses. In mammals, NAEs are synthesized from hydrolysis of their precursor molecule N-acylphosphatidylethanolamine (NAPE) by NAPE-specific phospholipaseD (NAPE-PLD). All NAEs including anandamide (NAE20:4) are hydrolyzed by fatty acid amide hydrolase (FAAH) into free fatty acid and ethanolamine. To date, different NAEs including anandamide have been identified in Physcomitrella patens but its metabolic pathway remains undiscovered. It is hypothesized that NAE metabolic pathway in P. patens is conserved and is similar to that of other eukaryotic systems. To this extent, putative PpNAPE-PLD and PpFAAH were identified and cloned for heterologous expression and characterization. Expression of PpFAAH was further verified by Western blot analysis. Future studies will involve biochemical characterization of putative PpNAPE-PLD and PpFAAH, to establish the evolutionarily conserved nature of NAE functions in early land plants.

▼ This research focused on the study of the amino sugar recycling and sialic acid degradation pathway as a possible entry point for N-acyl glucosamines for the production of sialic acid analogs. Meeting this objective would allow the development of a bacterial strain capable of producing non-natural nonulosonic acids that could be used in the development of medicines, vaccines or useful compounds for the study of interactions between pathogenic organisms and their host.
The first step was to understand how N-acetyl-D-glucosamine-6-phosphate deacetylase reacts to different types of substrates in order to determine its tolerance to the size of acyl groups in acyl amino sugars. This was achieved by studying the enzymatic activity in an in vitro system. We determine that the enzyme has a preference for small and slightly bulky acyl groups. Then, an in silico docking modeling and an in vivo system experiment were carried out. These experiments allowed to confirm the previous results.
The second project was carried out due to the uncertainty of whether the kinase involved in the catabolic pathway would be able to phosphorylate the substrates. By quantifying residual ATP, the high specificity of N-acetyl-D-glucosamine kinase could be verified. This result led us to think about the design of an organic synthesis strategy that would allow the phosphorylation of glucosamine in carbon 6. A simple synthetic route was designed based on the protection of the two most reactive moieties of the amino sugars and the reactivity of the hydroxy group on carbon 6. However, we had problems with the purification step of the final product due to its high polarity.
The next stage of this investigation was to confirm the transformation of GlcNAc into ManNAc. For this, an NMR analysis was designed that would detect the presence of both sugars in the reaction system. The epimerization of ManNAc to GlcNAc was detected successfully. Notwithstanding, the reverse reaction could not be detected. Based on the results obtained in the previous stage, we realized that an error was made in the epimerization reaction since we placed the wrong kinase because we did not take into account its substrate specificity.
Finally, we tried to produce sialic acid analogs in a fermentative system using different genetic variants of Escherichia coli. Two of the expected analogs, Neu5Pr and Neu5nBu, were obtained. In addition, NagA activity towards substrates with small acyl groups was confirmed.

▼ Neste trabalho estudamos a existência de ordem de longo alcance em modelos ferromagnéticos na presença de um campo externo cuja configuração apresenta um padrão tipicamente aleatório. Provamos por meio do argumento de Peierls modificado por Griffiths para o estudo de um antiferromagneto, que o modelo de Ising ferromagnético bidimensional exibe, para um campo alternado de intensidade fraca, ordem de longo alcance `a temperatura finita. Propomos dar um passo além considerando campos auto-similares esparsos, cuja soma é nula em todas as escalas. Estudamos também o modelo hierárquico em duas dimensões, para o qual provamos a existência de ordem de longo alcance a temperatura finita, na ausência de campo externo e para um campo com regiões irregulares esparsas. Provamos que os resultados do modelo de contornos hierárquicos são equivalentes aos resultados do modelo hierárquico em duas dimensões. Por fim, provamos através do método do limite infravermelho existência de ordem de longo alcance no modelo N-vetorial com campo alternado, de intensidade fraca, para d >= 3, sob a hipótese de que a variância do estado associado `a interação com o campo apresenta cardinalidade inferior a do volume do sistema. Mostramos, sob hipóteses similares, que o modelo N-vetorial hierárquico com campo externo, esparso e de intensidade pequena, apresenta ordem de longo alcance a baixas temperaturas.
In this work we study the existence of long range order for ferromagnetic models in the presence of an external field whose configuration has a pattern typically random. We prove, via the Peierlsárgument modified by Griffiths in his study of an antiferromagnet, that the two dimensional ferromagnetic Ising model for a staggered field exhibits long-range order at finite temperature and small field intensity. We propose to give a further step considering sparse self similar fields, whose sum is zero in all scales. We study as well the hierarchical model in two dimensions, where we prove existence of long-range order at finite temperature in the absence of external field and for a field configuration with sparse irregular regions. We prove that the results for the two-dimensional hierarchical contours model are equivalent to the results of the hierarchical model in two dimensions. Lastly, we prove via infrared bound method, existence of long range order in the N-vector model with a staggered and weak external field for d >= 3, under the hypothesis that the variance of the state connected with the field interaction has cardinality lower than volume. We show, under similar hypotheses, that the N-vector hierarchical model with a sparse field of low intensity has long range ordem at low temperatures.

▼ A β-D-N-acetilglucosaminidase extracted and partially isolated from crustacean Artemia franciscana by ammonium sulfate precipitation and filtration gel chromatography Bio Gel A 1.5m. the enzyme was immobilized on ferromagnetic Dacron yielding a insoluble active derivative with 5.0 units/mg protein and 10.35% of the soluble enzyme activity. β-D-N-acetilglucosaminidase-ferromagnetic Dacron was easily removed from the reaction mixture by a magnetic field, it was reused for ten times without loss in its activity. The ferromagnetic Dacron was better activated at pH 5.0. The particles visualized at scanning electron microscope (SEM) had presented different sizes, varying between 721nm and 100m. Infra red confirmed immobilization on support, as showed by primary amino peaks at 1640 and 1560 cm-1 . The immobilize enzyme presented Km of 2.32 0.48 mM and optimum temperature of 50C. Bought presented the same thermal stable of the soluble enzyme and larger enzymatic activity at pH 5.5. β-D-N-acetilglucosaminidase-Dacron ferromagnético showed sensible for some íons as the silver (AgNO3), with loss of activity. The β-D-N acetilglucosaminidase activity for mercury chloride (HgCl2), whom is one of the most toxic substance joined in nature, it was presented activity already diminished at 0,01mM and lost total activity at 4mM, indicating sensitivity for this type of metal. β-D-N-acetilglucosaminidase-ferromagnetic Dacron showed degradative capacity on heparan sulfate, the enzyme still demonstrated degradative capacity on heparan sulphate, suggesting a possible application to produce fractions of this glycosaminoglycan
A β-D-N-acetilglucosaminidase, extraída e parcialmente isolada do crustáceo Artemia franciscana através de precipitação com sulfato de amônio e cromatografia em gel filtração Bio Gel A 1.5m foi imobilizada em Dacron ferromagnético rendendo um derivado insolúvel ativo contendo 5,0 unid/mg de proteína e retendo 10,35% da atividade da enzima solúvel. A β-D-N-acetilglucosaminidase-Dacron ferromagnético foi facilmente removida do meio reacional com o auxílio de um campo magnético e pôde ser reutilizada por dez vezes seguidas sem perda de atividade. O Dacron ferromagnético foi melhor ativado a pH 5,0 As partículas visualizadas no microscópio eletrônico de varredura (MEV) apresentaram diferentes tamanhos, variando entre 721nm e 100m. O infra vermelho confirmou a imobilização ao suporte, quando exibiu os picos de aminas primárias a 1640 e 1560 cm-1 . A enzima imobilizada apresentou Km aparente de 2,32 0,48 mM e atividade ótima a temperatura de 50C. Ambos apresentaram praticamente a mesma estabilidade térmica da enzima solúvel e maior atividade enzimática no pH 5,5. gel chromatography Bio Gel A 1.5m. the enzyme was immobilized on ferromagnetic Dacron yielding a insoluble active derivative with 5.0 units/mg
protein and 10.35% of the soluble enzyme activity. β-
D-
N-acetilglucosaminidase-ferromagnetic Dacron was easily removed from the reaction mixture by a magnetic field, it was reused for ten times without loss in its activity. The ferromagnetic Dacron was better activated at pH 5.0. The particles visualized at scanning electron microscope (SEM) had presented different sizes, varying between 721nm and 100µm. Infra red confirmed immobilization on support, as showed by primary amino peaks at 1640 and 1560 cm-1
. The immobilize enzyme presented Km of 2.32 ± 0.48 mM and optimum temperature of 50°C. Bought
presented the same thermal stable of the soluble enzyme and larger enzymatic activity at pH 5.5. β-
D-
N-acetilglucosaminidase-Dacron ferromagnético showed
sensible for some íons as the silver (AgNO3), with loss of activity. The β-
D-
N acetilglucosaminidase activity for mercury chloride (HgCl2), whom is one of the
most toxic substance joined in nature, it was presented activity already diminished at 0,01mM and lost total activity at 4mM, indicating sensitivity for this
type of metal. β-
D-
N-acetilglucosaminidase-ferromagnetic Dacron showed degradative capacity on heparan sulfate, the enzyme still demonstrated
degradative capacity on heparan sulphate, suggesting a possible application to produce fractions of this glycosaminoglycan

sistema bei surastos reakcijos sąlygos, leidžiančios
regioselektyviai atlikti Suzuki kryžminio jungimo reakciją
2,4-dichlorpirolo[2,3-d]pirimidine. Ištirta variu(I) katalizuojama
N-arilinimo reakcija tarp 2,4-diarilpirolo[2,3-d]pirimidinų ir
įvairių arilhalogenidų, dihalokarbazolų ir dihalofluorenų bei
susintetinti 2,4,7-triarilpirolo[2,3-d]pirimidinai,
bis(pirolo[2,3-d]pirimidin-7-il)karbazolai ir atitinkami fluorenai.
Sukurtas būdas, leidžiantis sintetinti pirolo[2,3-d]pirimidinus su
įvairiais pakaitais 2-, 4- ir 7-oje heterociklo padėtyse. Pirmą
kartą nustatyta, kad pirolo[2,3-d]pirimidino oligoarilenai pasižymi
mėlyna-UV fluorescencija tirpale ir kietame būvyje. Ištirta
arilgrupių struktūros, padėties pirolo[2,3-d]pirimidino žiede bei
pi-konjuguotos sistemos dydžio įtaka junginių fotoliuminescencinėms
savybėms. Nustatyta, kad kai kurie pirolo[2,3-d]pirimidino
oligoarilenai pasižymi teigiamu solvatofluorochrominiu efektu,
sudaro nanoagregatus tetrahidrofurano/vandens mišinyje. Nustatyta,
kad molekulių agregacija indukuoja emisijos padidėjimą. Parodyta,
kad pirolo[2,3-d]pirimidino oligomerai su išplėsta pi-konjuguota
sistema efektyviau fluorescuoja kietame būvyje nei
tirpale.
The main goal of the present work was to
synthesize pyrrolo[2,3-d]pyrimidine core based oligoarylenes and to
evaluate their photophysical properties. As a result, the Suzuki
reaction of 2,4-dichloropyrrolo[2,3-d]pyrimidine with arylboronic
acids was studied and novel 4-aryl- and
2,4-diarylpyrrolo[2,3-d]pyrimidines were synthesized. The reaction
conditions including an effective catalyst system for the Suzuki
cross-coupling reaction were proposed to achieve site-selectivity
of the reaction studied. A general approach for the preparation of
2,4,7-triarylpyrrolo[2,3-d]pyrimidines,
bis(pyrrolo[2,3-d]pyrimidin-7-yl)carbazoles and -fluorenes were
developed by copper-catalyzed N-arylation reaction of
2,4-diarylpyrrolo[2,3-d]pyrimidines with aryl halogenides,
dihalocarbazoles or dihalofluorenes. Synthetic strategies leading
to pyrrolo[2,3-d]pyrimidines with different aryl moieties in
positions 2, 4 and 7 of the heterocycle were studied and an
efficient method for their synthesis was developed. The synthesized
pyrrolo[2,3-d]pyrimidine-core based oligoarylenes were found to
exhibit blue-UV fluorescence in solution and in solid state.
Influence of structure of aryl groups, their position in the
pyrrolo[2,3-d]pyrimidine nucleous and extent of pi-conjugated
aromatic system on the photoluminescent characteristics were
evaluated. Some pyrrolo[2,3-d]pyrimidine derivatives were found to
exhibit positive solvatofluorochromism and to form nanoaggregates

D-hidantoinases (dihidropirimidina amidrohidrolase EC 3.5.2.2) são enzimas que catalisam a hidrólise de hidantoínas, com abertura do anel, para o correspondente N-carbamoil-D-aminoácido e são usadas na produção de Daminoácidos, incluindo D-fenilglicina e D-p-hidroxifenilglicina. Este trabalho relata os estudos desenvolvidos para a imobilização e estabilização de D-hidantoinase de Vigna angularis (3.5.2.2.). Foram abordadas diferentes estratégias de imobilização multipontual em suportes orgânicos como quitosana e agarose. Diferentes protocolos de imobilização foram empregados, sendo adição de íons durante a redução com NaBH4 importante para proteção do centro catalítico da enzima. Os derivados ativos e estabilizados foram empregados na reação de hidrólise da fenilhidantoína. O estudo de temperatura e pH de máxima atividade da enzima foi 60C e pH 10,0. As subunidades da enzima apresentam peso molecular, com valor próximo a 50kDa. A enzima imobilizada em glioxil-agarose na presença dos íons Zn2+ durante a etapa de redução, com tempo de imobilização de 24 h foi o derivado mais estável sendo 89 vezes mais estável que a enzima solúvel. A análise de aminoácidos mostrou que aproximadamente 50% dos resíduos de lisina presentes na enzima foram covalentemente ligados no derivado de glioxil-agarose. A enzima imobilizada em quitosana-alginato-epoxilado foi 20 vezes mais estável que a enzima solúvel. Todos os procedimentos de imobilização testados levaram a 100% de rendimento de imobilização. Enzima solúvel e os melhores derivados obtidos por imobilização em glioxil e quitosana foram usados na catálise da hidrólise de fenilhidantoína, produzindo 99% de N-Carbamoil-D-fenilglicina nos tempos de 3, 9 e 15 h, respectivamente.
Immobilization and stabilization of enzymes increases their potential for use in industrial scale. D-hydantoinases (dihidropirimidina amidrohidrolase EC 3.5.2.2) catalyze the hydrolysis of D-hydantoins, generating the corresponding Ncarbamoil- D-amino acid and are used in the production of D-amino acids, including Dphenylglycine and D-p-hydroxyphenylglycine.This work reports studies for immobilization and stabilization of D-hydantoinase from Vigna angularis (E.C. 3.5.2.2.). Different strategies of multipoint covalent attachment in organic supports as chitosan and agarose were used. Different protocols of immobilization were employed, being the adittion of ions during the reduction step with the NaBH4 important to protect enzyme catalytic site. The active and stabilized derivatives were used to catalyze the hydrolysis of D-phenylhydantoin. The temperature and pH enzyme profiles showed maximum enzyme activity at 60C and pH 10,0. ▼ The work presented in this thesis is focused on the experimental study of the two deuterium burning channels 2H(d,p)3H and 2H(d,n)3He in the Big Bang Nucleosynthesis (BBN) astrophysical scenario.
In the beginning is presented this phase of the early universe and why the BBN model is still fine tuned with new results coming from observations and laboratory measurements. This model (if BBN is considered standard) can be adjusted such to have only a free parameter, the baryon to photon ratio of the universe eta at a certain time. Also the WMAP satellite results have been of great help to fix eta (and then all the others BBN numbers), but still persist some discrepancies, as for the lithium primordial abundances, between what is theoretically predicted and what is observed. The deuterium (and the reactions in which is involved) plays a key role in the evaluation of eta, thus for all BBN, so that is called the best baryometer.
Then is explained that the need of new cross sections (and reaction rates) measurements for astrophysics can not be satisfied. This is because of the problems given by their measurements in the laboratory, as the presence of Coulomb barrier (that makes cross sections exponentially decrease in the energy range of interest) and the electron screening effect.
For all these reasons the present measurements have been performed through the Trojan Horse Method (THM), an indirect method that allow to have a bare-nucleus cross section of the two-body reaction of astrophysical interest that is free of the Coulomb suppression. This is accomplished via the selection of the quasi-free mechanism in an appropriate three-body reaction, whose center-of-mass energy is greater than the Coulomb barrier.
Two experimental runs have been carried out at the Nuclear Physics Institute of the Academy of Science of Czech Republic, in Rez (Prague). In the first one, with a 17 MeV 3He beam (in which only 2H(d,p)3H has been measured) the presence of quasi-free mechanism events has been ascertained. The result obtained is fair but not good enough for the error reduction needed for astrophysics. Instead, to optimize the result in the region relecant for astrophysics, a the second run (where the 3He beam energy was 18 MeV) has been performed. In particular for the first time the technique of measuring one of the two-body reactions participant ejectile and the spectator particle, in this case a proton, instead of both the ejectiles. This has also allowed the measure of the 2H(d,n)3He reaction without the complexity of the neutron detection, so with a very good precision. All the off-line analysis done until the S-factor extraction is detailed explained, including the MPWBA analysis by Dr. S. Typel. Also the screening potential has been evaluated, obtaining a value of 13.2±1.8 eV for 2H(d,p)3H and 11.7±1.6 eV for 2H(d,n)3He, very close to the adiabatic limit, as expected. ▼ O Status epilepticus induzido por LiCl-pilocarpina causa uma massiva neurodegeneração em diversas regiões cerebrais, e a maioria dos fármacos antiepilépticos clássicos não são suficientemente eficazes para proteger os neurônios da morte. Antagonistas do receptor N-metil-D-aspartato apresentam ação anticonvulsiva e são atualmente capazes de proporcionar neuroprotecção contra a morte neuronal induzida pelo SE. No entanto, estes compostos demonstraram inúmeros efeitos colaterais. Apesar disso, a memantina, um antagonista NMDA de baixa afinidade, é clinicamente bem tolerada e não induz tantos efeitos indesejados. Portanto, o objetivo do presente estudo foi investigar o efeito neuroprotetor da memantina contra a degeneração neuronal induzida pelo status epilepticus induzido por LiCl-pilocarpina nos períodos iniciais da vida. Filhotes de ratos Wistar receberam memantina (20 mg / kg ip) em diferentes regimes de tratamento. A neurodegeneração foi quantificada por fluoro jade C. Em CA1 a neurodegeneração diminuiu significativamente quando os animais receberam memantina nos regimes de pós-tratamento realizados às 0 e 0,25 horas. Além disso, o pós-tratamento foi capaz de proteger contra a neurodegeneração em uma região da amígdala. O pré-tratamento com memantina mostrou uma redução da morte celular no tálamo. Nossos resultados demonstram o efeito neuroprotetor da memantina na morte neuronal induzida pelo SE no início da vida. Este efeito é dependente do tempo de tratamento e da região cerebral.
Status epilepticus induced by LiCl-pilocarpine is accompanied by neurodegeneration and the classical antiepileptic drugs are not effective in protect neurons from death. Typical N-methyl-D-aspartate receptor antagonists exhibit anticonvulsant action and are currently able to provide neuroprotection against SE-induced neuronal death. However, these compounds have demonstrated undesirable side effects. Despite this, memantine, a low-affinity NMDA antagonist, is clinically well tolerate and does not induce side effects. Therefore, the aim of the present study was to investigate the neuroprotective effect of the memantine against neuronal degeneration induced by early life LiCl-Pilocarpine-induced status epilepticus. Wistar pup rats received memantine (20 mg/kg i.p.) at different treatment regimens before and after SE induction. Neurodegeneration was quantified by fluoro jade C staining. In CA1 subfield neurodegeneration was significantly decreased when animals received memantine at 0h and 0.25h post-treatment regimens. Additionally, post-treatment was able to protect against neurodegeneration in amygdala region. Memantine pre-treatment showed a reduction of cell death in thalamus. Our results demonstrated the neuroprotective effect of memantine on neuronal loss induced by SE early in life. This effect was time- and region-dependent.

N-methyl-
D-aspartate receptors (NMDARs) is known to play an important role in synaptic plasticity, learning and memory. Moreover, NMDAR dysfunction may contribute to a variety of neuropsychiatric and neurological disorders including schizophrenia, epilepsy, stroke and trauma. Pregnenolone sulfate (PS) is one of the most commonly occurring neurosteroids in the central nervous system and influences function of several receptors. PS modulates NMDARs and has been shown to have both positive and negative modulatory effects on NMDAR currents generally in a subtype-selective manner. We assessed the gating mechanism of PS modulation of GluN1/GluN2A receptors transiently expressed in HEK 293 cells using whole-cell and single-channel electrophysiology. Only a modest effect on the whole-cell responses was observed by PS in dialyzed (non-perforated) whole-cell recordings. Interestingly, in perforated conditions, PS was found to increase the whole-cell currents in the absence of nominal extracellular Ca2+ whereas PS produced an inhibition of the current responses in the presence of 0.5 mM extracellular Ca2+. The Ca2+-binding DRPEER motif and GluN1 exon-5 were found to be critical for the Ca2+-dependent bidirectional effect of PS. Single-channel cell-attached analysis demonstrated that PS primarily affected the mean open time to produce its effects, with positive modulation mediated by an increase in duration of open time constants while negative modulation mediated by a reduction in the time spent in a long-lived open state of the receptor. Further kinetic modeling of the single-channel data suggested that the positive and negative modulatory effects are mediated by different gating steps which may represent GluN2 and GluN1 subunit-selective conformational changes respectively. Our studies provide a unique mechanism of modulation of NMDARs by an endogenous neurosteroid which has implications for identifying state-dependent molecules. UBP (University of Bristol Pharmaceuticals) series compounds act at allosteric binding sites on the NMDARs and have been able to achieve subtype-selectivity. We found that UBP684 significantly increased the whole-cell current and mean open time and reduced the mean shut time for GluN1/GluN2A receptors. Mutagenesis studies suggest that UBP684 can stabilize the closed cleft of the LBD and drive the receptor into the more stable open states.

▼ The NMDA receptor (NMDAR) hypofunction hypothesis in schizophrenia posits that reduced NMDAR function leads to behavioral abnormalities observed in schizophrenia. Lower expression of GluN2C subunit of NMDAR has been reported in the cortex and thalamus in postmortem brains from schizophrenic patients, however its functional implications remain unknown. In the present study, we used two GluN2C KO mouse model to further characterize the deficits associated with a reduction in GluN2C, as well as to elucidate the expression pattern of the GluN2C subunit. We found cellular and synaptic deficits in GluN2C knockout mice including lower parvalbumin labeling and lower dendritic spine density, features similar to those observed in human condition. GluN2C heterozygous and knockout mice also exhibit behavioral and cognitive deficits relevant to schizophrenia. Moreover, a GluN2C/GluN2D potentiator, CIQ, attenuated NMDAR channel blocker-induced hyperlocomotion, working memory deficit as well as prepulse inhibition deficit in wildtype and heterozygous mice but not in GluN2C knockout mice. These results demonstrate that deficit in GluN2C-containing NMDARs leads to schizophrenia-like phenotypes, and establish that pharmacologic enhancement of GluN2C-containing NMDARs may lead to beneficial effects for behavioral and cognitive deficits in schizophrenia.

▼ Spontaneous neurotransmission is a stochastic mode of neurotransmitter release that is not coupled to action potential arrival at the presynapse. Recent evidence has shed light upon the presynaptic and vesicular fusion machinery that makes this mode of release unique to action potential driven, or evoked release, yet the postsynaptic reception of these differing signals has yet to be probed in detail. Here, I illustrate that evoked and spontaneous release modes coexist within the same synapses, yet the neurotransmitter released falls upon separate subsets of postsynaptic receptors. As a consequence of this phenomenon, in glutamatergic hippocampal synapses the NMDA receptors activated by spontaneous release are able to drive large calcium transients that maintain synaptic homeostasis and suppress local protein synthesis.

▼ The N-methyl-D-aspartate (NMDA) subtype of excitatory, ionotropic L-glutamate neurotransmitter receptor is a cation-selective, ligand-gated ion channel composed of NRl and NR2 subunits. NMDA receptors are localised on the dendritic spines of neurons where they are tightly associated with proteins of the post-synaptic density such as the scaffolding protein, post-synaptic density 95 (PSD-95). Many studies have characterised the pharmacological properties of NMDA receptors expressed in non-neuronal cell lines which lack the proteins of the post-synaptic density. The aim of this thesis was to investigate the effect of PSD-95 on the biochemical and pharmacological properties of NMDA receptors following their co-expression in mammalian cells. Various homomeric and heteromeric combinations of NR1-1 a, NR1-2a, NR1-4b, NR2A and NR2B subunits were expressed ± PSD-95 in human embryonic kidney 293 cells and analysed by quantitative immunoblotting, radioligand binding and immunoprecipitation. It was found that PSD-95 both enhanced the level of NR2A and NR2B subunit expression by approximately three-fold and induced a three-fold increase in the [3H]MK801 radioligand binding sites for NR1-1a/NR2A receptors. PSD-95 also increased the EC50 values for the L-glutamate and glycine enhancement of [3H]MK801 binding to NR1-1a/NR2A receptors by four-fold and six-fold respectively. PSD-95 however, had no effect on the affinity of NRl-la/NR2A receptors for L-glutamate and glycine as determined by [3H]CGP39653 and [3H]MDL105-519 displacement assays respectively suggesting that the reduced EC50 may be due to decreased channel gating. Deletion studies showed that both the biochemical and pharmacological effects of PDS-95 on expressed NMDA receptors were dependent upon the NR2 C-terminal ESDV motif, the PSD-95-binding domain. Further, mutagenesis of the two N-terminal cysteine residues of PSD-95 demonstrated that PSD-95/NMDA receptor clustering is necessary for the changes in NMDA receptor properties induced by PSD-95. These results suggest that the interaction between PSD-95 and native NMDA receptors may be important for the regulation of both receptor function and receptor number at post- synaptic sites.

▼ Podocytes play a dynamic role in regulating glomerular filtration. The focus here is on the regulatory mechanisms of podocyte expressed transient receptor potential 6 (TRPC6) channels, an ion channel implicated in certain forms of proteinuric kidney disease. TRPC6 channels are polymodal and can be activated by either chemical or mechanical stimuli. Chemical stimulation is mediated by surface-expressed receptors, and the roles of angiotensin II type 1 receptors (AT1R), insulin receptors, and
N-methyl-
D-aspartate receptors (NMDAR) on TRPC6 activity are studied here. In acutely isolated rat glomeruli, angiotensin causes an upregulation of TRPC6 activity, and this is mediated by the Gαq /PLC pathway. This angiotensin-evoked upregulation is partially dependent on the formation of reactive oxygen species (ROS). In mouse podocyte cell lines, insulin causes upregulation of TRPC6 through increasing ROS formation via NADPH oxidase 4 (NOX4). Similarly, NMDAR activation upregulates TRPC6, albeit through NOX2. The previously uncharacterized podocyte NMDAR has unusual properties with strong physiological implications. Specifically, podocyte NMDA receptors are essentially unresponsive to L-glutamate and L-aspartate and do not show glycine-mediated potentiation. These receptors respond to the agonists L-homocysteate and
D-aspartate with large ionic currents that are potentiated by
D-serine. Given their resistance to L-glutamate-induced activation, podocyte NMDA receptors likely do not act in a localized glomerular signaling system. However, their response to ligands that circulate in both the normal and pathological state suggests a role for podocyte NMDA receptors in normal glomerular function. Receptor-driven upregulation of TRPC6 comprises a class of potential targets for prevention and treatment of multiple acquired kidney diseases. Independent of receptor-mediated response, TRPC6 channels are mechanosentive and can be activated by membrane deformation in both podocyte cell lines and isolated glomeruli. This mechanosensitivity is repressed by podocin, a cholesterol-binding, membrane-associated partner of the TRPC6 channel. In addition, podocin mediates diacylglycerol activation of TRPC6, suggesting that podocin determines the favored mode of TRPC6 activation in podocytes. It is possible that disruption of the podocin-TRPC6 complex at the slit diaphragm contributes to Ca2+ overload and eventual foot process effacement. Drugs that selectively target and suppress TRPC6 mechanosensitivity could potentially serve as treatments for glomerular diseases.

Célunk három Penicillium faj morfológiai változásának és öregedésének vizsgálata volt.
Felületi tenyészetekben a sejtfal-integritást megzavaró kongóvörös festék alkalmazása eltérő eredményre vezetett. A P. pinophilum volt a festékre legérzékenyebb, ennél a sejtfal integritás megzavarása növekedés-gátlást okozott. A P. frequentans és P. chrysogenum esetében a sporuláció gátlását tapasztaltuk már igen kis festék-koncentrációnál (5 mg ml-1).
Süllyesztett tenyészetekből vett minták mikroszkópi vizsgálata eseté
n láthatók voltak az exponenciálisan növekedő és autolizáló gombák morfológiai különbségei. Az öregedő tenyészetekben megfigyelhető fragmentálódás mértéke szorosan összefüggött a kitináz és Nacetil-ß-

▼ My thesis works as an experiment, or rather a series of experiments, in methods of thinking about historical material. These methods come from anthropology and engage with myths and ritual, with the concept of “complementary others”, and the concept of “schismogenesis" as it has been developed by Gregory Bateson and advanced further by Marshall Sahlins. My overall goal is not to re-describe a well-researched historical event, but to explore how different ways of analysis, using different analytical frameworks, could lead to valuable explanations of the same political-cum-cultural event. The phenomenon I engage with is the last Oecumenical Council, a major religious event in the history of Councils within already schismatic societies. For this reason, I treat this Council in particular, as a ritual, unprecedented in scale and ambiguous in its inception. I am examining the structure and the return of this Event in History, and the controversies and tensions in the diachrony of East and West. I do this not only through the notion of schismo-genesis and ritual, but specifically the notion of sacrifice as developed by Maurice Bloch, in which the journey from Constantinople to Italy becomes a historical metaphor of mythical realities, regarding the Emperor John VIII Palaiologos. And finally, I explore the significance of Bessarion and complementary others within the notion of transformation and alterity. What I establish through discussions of the historical material, which span eleven centuries of history, is first of all, that there is no event without a system; that means the journey can acquire the form of the ritual. I argue that the relation between the myth and the idea of unity is dialectical in nature; the Event of a Union, which could bring peace in the one Church of Christ, from this moment of realisation becomes a fabrication, a mystery to the witnesses, and all the other myths that will be developed on the way become even more imperative and melancholic, because they seek to express a negative and unavoidable truth. The Event doesn’t portray reality any more, it exists despite it and becomes an extreme position, almost like a dream, and it justifies the vision one wished to be possible, only to show that it is untenable: the “what if it could be”; the possibility of all parts being aspirations to the whole, oecumenicity as a goal rather than unity. Overall, this thesis is about the presence of the past in the present, in relevance to the future.

▼ Conantokins, which are neuroactive peptides
found in the venom of marine snails, contain post-translationally
modified &gamma-carboxyglutamate residues (Gla; γ). The Gla
residues are integral for their structure, function, and
subunit-selective antagonism of ion channels of the
N-Methyl-
D-Aspartate Receptors (NMDAR). The functional
heterotetramer of the receptor is composed of GluN1 and GluN2
subunits which impart specific biochemical and physiological
properties to the ion channels. The optimum spacing of Gla residues
in conantokins stabilize the structure when complexed to divalent
ions and mediate NMDAR-subunit selective properties. In this study,
the structural and antagonistic properties of conantokins
ConPr-1,-2, and -3 from Conus parius and ConRl-B from Conus rolani
species were studied. The Conus parius family of conantokin
peptides are GluN2B selective and adopt a low properties of α-helix
conformation in the presence of Mg
2+. These
peptides also display the ability to diminish downstream
signaling events. ConRl-B, ConPr-2, and ConPr-3, additionally
contain the post-translationally modified 4-transhydroxyproline
(HyP; O) residue. The Hyp10 residue in ConRl-B, not only changes
the Gla positioning in the C-terminal part of ConRl-B, but also
affects the structural properties of the peptide. A combination of
biophysical techniques, e.g., CD,
1H NMR and
ITC, was employed to study the structure and metal binding
properties of Mg
2+/ConRl-B. The structure
determined by high-field 1H NMR, shows disruption in the α-helix
due to HyP10 further interfering with
Mg
2+-binding, thus resulting in solvent
accessible backbone amide protons. To assess the role of HyP and
the molecular requisites for the unique structural and functional
properties of ConRl-B, mutant peptides, ConG[10 ▼O] and
ConRl-B[ΔKAO▼NQ] were synthesized. Although, the 3D structure and
Mg
2+ binding abilities of ConRl-B[ΔKAO▼NQ]
were similar to Con-G, this peptide displays complete
non-selectivity towards NMDAR. Local disruption of the
α-helix backbone in Con-G on the introduction of HyP,
differs from that of ConRl-B. Additionally, Con-G[10 ▼O] loses
selectivity towards ion channels containing the GluN2B subunit.
These results suggest that the natural occurrence of HyP in the
ConRl-B backbone is important for high GluN2B selectivity. In
conclusion, the presence of HyP residue changes the structural and
functional properties of conantokins.

▼ Studien undersöker invånare och verksammas upplevelser av ett, enligt en rapport frånunderrättelseenheten, utsatt bostadsområde i Västerås. För att besvara frågeställningen har vigenomfört 14 intervjuer varav 5 var semistrukturerade och resterande var ostrukturerade. Vihar även genomfört 6 observationer, där samtliga var icke deltagande observationer. Dåmålsättningen var att undersöka så förutsättningslöst som möjligt har vi använt oss av BarneyGlasers variant av grundad teori som metodologiskt angreppssätt. Tidigare forskning som vibelyser är delade i tre teman: Segregation, familjestrukturer och sysselsättning. En teorisom är grundad i data presenteras i resultatdelen och beskriver det som sker i Bäckby.Studiens teoretiska referensramar är Erving Goffmans teori om stigma, Howard. S. Beckersteori om avvikande samt Giorgio Agambens teori om undantag och belägring som fungerarsom en komplettering till förståelsen av hur det är i ett utsatt bostadsområde. Slutsatsernavisar att det är en handfull ungdomar som står för oron som sker i Bäckby och dess handlingarmöjliggörs på grund av misslyckade strukturella åtgärder. De förhållanden vi har funnitavgörande för trygghets upplevelse i Bäckby är inte endast att ungdomar behöversysselsättning, utan det behövs även en spridning av människor med utländsk bakgrund blandetniska svenskar och förebyggande åtgärder från externa aktörer.

▼ In this thesis, a model for argument revision is presented, in terms of the expansion and contraction of a system of structured argumentation. At its core, the model uses the belief revision concept of minimal change, but without requiring a pre-determined entrenchment ordering to establish minimality. In the first part of the thesis, a model for argument revision is defined and described. Specified in terms of the ASPIC\+ framework for argumentation, the model is divided into two main concepts: argument expansion, whose goal is to make certain arguments acceptable in the system, possibly by adding them; and argument contraction, whose goal is to make certain arguments unacceptable in the system, possibly by removing them. The goal of a revision process can be achieved in multiple different ways, thus a method of choosing which, based on measures of minimal change, is also specified. The second part of the thesis demonstrates two applications of the model in the context of multi-agent dialogue. The first is used to assist a participant when faced with a need to update its commitment store during persuasion dialogue, while the second shows how a participant can use argument revision techniques to both assess and maintain a lie.The main contributions of the thesis are twofold. First, the characterisation of a model for argument revision, based on established belief revision principles but with a key difference. The model for argument revision demonstrates how it is possible to use measurable effects on the system when determining minimal change instead of relying on a pre-determined, qualitative entrenchment ordering.Second, the thesis demonstrates two applications of argument revision in dialogue. The first is in assisting an agent in retracting a commitment that has been defeated, and for which it can offer no defence. When retracting a claim, the participant may also be required to retract other claims from which the defeated one is a consequence. Applying argument revision techniques allow the participant to reason about what constitutes a minimal set of retractions, in terms of current commitments and potential future communications in the dialogue.The second dialogical application relates to the opposite of retraction; instead of choosing to retract an undefended claim, the participant could instead choose to lie in order to defend it. Argument revision allows the participant to not only assess whether or not lying is ``minimal'' (compared to retracting), but to also to maintain the lie, by using the measures of minimal change.Overall, the thesis shows that not only is justifiable argument revision possible without relying on a pre-determined entrenchment ordering, it is also a powerful tool for participants in a dialogue, assisting with dialogue move selection.

▼ Visualisation is a graphical representation that is used to aid understanding and gain insight into information. Similarly, visualising arguments can help people to explore argument structure and comprehend knowledge associated with tough problems. In this thesis, we focus on representing arguments, especially when data size increase, which causes difficulty in exploring and understanding arguments. A fundamental problem from an argument perspective is that understanding argument requires users to understand the overview, read the content of arguments, and recognise how the arguments support, compete, and conflict with each other. Applying information visualisation techniques or combination between them in an argumentation domain can improve the navigation and exploration in argument data. The main contribution of this thesis is proposing various visualisation approaches for gaining insights on argument data and helping experts to understand arguments. Based on collected user requirements, the work presents several prototypes that provide an overview of arguments while giving users the ability to read the argument text. To compare and evaluate our proposed techniques and tools, a controlled user study and interviews with argument experts are conducted. The collected qualitative and quantitative results are thoroughly analysed using relevant statistical tests.

▼ The full-length cDNA Pbnag enconding a N-acetyl-β-Dglucosaminidase (NAG) GeneBank accession number AF 419158 of Paracoccidioides brasiliensis was characterized and classified as a NAG belonging to the glycosil hydrolase family 20. The PbNAG was expressed in Escherichia coli and purified to homogeneity by affinity chromatography on glutathione-Sepharose 4B. SDS-PAGE analysis revealed the presence of one protein species with molecular mass estimated of 64.8 kDa. Immunoelectron microscopy demonstrated the presence of the native protein in the cell wall of P. brasiliensis. The recombinant protein was evaluated regarding to its kinetics properties The Km and Vmax values for the enzyme using ρ- nitrophenyl-β-D-N-acetylglucosamine (ρNP-GlcNAc) as substrate, were 0.112 mM and 0.0017 mM, respectively. The pH optimum for the enzyme was 4.4 and maximum activity was obtained at 37 C. The enzyme activity was poorly affected by glucose and was inhibited by glucosamine, Nacetylglucosaminide. The studies show that PbNAG is present in the cell wall of yeast cells of P. brasiliensis suggesting its potential role in the fungal cell wall additionally.
Um cDNA (Pbnag, n de acesso GenBank AF 419158) codificante para N-acetil-β-D-glicosaminidase (NAG) de Paracoccidioides brasiliensis, isolado Pb 01 (ATCC MYA826) foi caracterizado e classificado como uma enzima pertencente à família 20 de glicosil hidrolase. PbNAG foi clonado em pGEX-4T3 e expresso em Escherichia coli. A análise em gel apresentou a presença de uma proteína de massa molecular estimada em 64,8 kDA. A proteína expressa foi purificada em resina glutationa-sepharose 4B e utilizada para produção de anticorpo policlonal. O anticorpo produzido anti-PbNAG apresentou reação com a proteína nativa, em extrato total, de micélio e levedura de P. brasiliensis. Na imunocitolocalização o anticorpo anti-PbNAG reagiu com a proteína NAG nativa presente na parede de células leveduriformes P. brasiliensis. A enzima recombinante PbNAG foi caracterizada apresentando afinidade pelo substrato ρ-nitrofenil-β-D-N-acetilglucosamina (ρNP-GlcNAc), com Km de 0.112 mM, Vmax de 0.0017 mM min-1 mg proteina-1, temperatura ótima de 37C e pH ótimo de 4.4. A enzima foi pouco inibida por glicose e apresentou inibição por glicosamina, N-acetilglicosamina. Os estudos mostram que PbNAG está presente na parede celular de células leveduriformes de P. brasiliensis, sugerindo seu papel no metabolismo da parede fúngica.

▼ Efficient and effective delivery of fertilisers, herbicides, pesticides and growth regulating compounds to plants is the
subject of much ongoing research. The objective of this research was to develop nano-formulations for delivery of compounds to plants. Two formulations were developed: the first was solution-based focused on encapsulation of the active ingredient in a nanoemulsion. Nanoemulsions should be ideal for facilitating transfer of compounds to plant leaves as their size correlates well with the nanoscale surface features of leaves, achieving significantly greater total contact area between the oil droplets and the leaves. The second nano-formulation was solid-state based, focused on locating the active ingredient within the tubules of a nanotube clay. For proof-of-concept two synthetic plant hormones,
N-phenyl-N‘-(2-chloro-4-pyridyl)urea (CPPU) or forchlorfenuron, a synthetic cytokinin, and 2,4-dichlorophenoxyacetic acid (2,4-
D), a synthetic auxin, were chosen for encapsulation. CPPU is a phenylurea derivative that shows strikingly strong cytokinin-like activity in plants, including delaying senescence. It is highly water insoluble, but soluble in organic solvents. It is widely used in a variety of crops, particularly kiwifruit and table grapes. Delivery of CPPU safely, efficiently and at the appropriate dosage is a priority as overdosing or incorrect timing of application causes detrimental effects on fruit firmness and other quality attributes. Auxins are also a group of plant hormone. 2,4-
D is a synthetic auxin which has been widely used at high concentrations as a herbicide, at medium concentrations for fruit thinning, and at low concentrations promotes root initiation, but at even lower concentrations promote root elongation. Consequently, careful control of dosage is required to obtain the desired effect. The nanoemulsion system developed was water/polysorbate 80/glycerol/soybean oil. The active ingredient, CPPU, was incorporated into the nanoemulsion via the oil phase in a pre-concentrate which was then crash diluted to yield the final nanoemulsion. Nanoemulsions are created only when the concentrate is located in the bicontinuous or oil-in-water microemulsion regions of the phase diagram. The droplet size of the nanoemulsions was measured using dynamic light scattering with droplets ranging in size from 30 – 100 nm. The CPPU-loaded nanoemulsions were stable for more than three days. To determine if the nanoemulsion was an effective delivery system, a leaf senescence bioassay was conducted to test the senescence-delaying effect of the CPPU-loaded nanoemulsions when applied to explants. The nanoemulsions were applied directly to the leaves of dwarf bean explants. Chlorophyll was extracted from the leaves and measured spectrophotometrically before and several days following treatment. The CPPU-loaded nanoemulsions enhanced the effectiveness of CPPU in delaying leaf senescence compared with the control experiments, including direct application of CPPU. [Mn(II), Co(II), Ni(II), Cu(II) i Zn(II)] sa anjonima aromatičnih
polikarboksilnih kiselina (ftalne, izoftalne, tereftalne i
piromelitne) kao O,O-donorskim ligandima i aromatičnim

▼ Efficient and effective delivery of fertilisers, herbicides, pesticides and growth regulating compounds to plants is the
subject of much ongoing research. The objective of this research was to develop nano-formulations for delivery of compounds to plants. Two formulations were developed: the first was solution-based focused on encapsulation of the active ingredient in a nanoemulsion. Nanoemulsions should be ideal for facilitating transfer of compounds to plant leaves as their size correlates well with the nanoscale surface features of leaves, achieving significantly greater total contact area between the oil droplets and the leaves. The second nano-formulation was solid-state based, focused on locating the active ingredient within the tubules of a nanotube clay.
For proof-of-concept two synthetic plant hormones,
N-phenyl-N‘-(2-chloro-4-pyridyl)urea (CPPU) or forchlorfenuron, a synthetic cytokinin, and 2,4-dichlorophenoxyacetic acid (2,4-
D), a synthetic auxin, were chosen for encapsulation. CPPU is a phenylurea derivative that shows strikingly strong cytokinin-like activity in plants, including delaying senescence. It is highly water insoluble, but soluble in organic solvents. It is widely used in a variety of crops, particularly kiwifruit and table grapes. Delivery of CPPU safely, efficiently and at the appropriate dosage is a priority as overdosing or incorrect timing of application causes detrimental effects on fruit firmness and other quality attributes. Auxins are also a group of plant hormone. 2,4-
D is a synthetic auxin which has been widely used at high concentrations as a herbicide, at medium concentrations for fruit thinning, and at low concentrations promotes root initiation, but at even lower concentrations promote root elongation. Consequently, careful control of dosage is required to obtain the desired effect.
The nanoemulsion system developed was water/polysorbate 80/glycerol/soybean oil. The active ingredient, CPPU, was incorporated into the nanoemulsion via the oil phase in a pre-concentrate which was then crash diluted to yield the final nanoemulsion. Nanoemulsions are created only when the concentrate is located in the bicontinuous or oil-in-water microemulsion regions of the phase diagram. The droplet size of the nanoemulsions was measured using dynamic light scattering with droplets ranging in size from 30 – 100 nm. The CPPU-loaded nanoemulsions were stable for more than three days.
To determine if the nanoemulsion was an effective delivery system, a leaf senescence bioassay was conducted to test the senescence-delaying effect of the CPPU-loaded nanoemulsions when applied to explants. The nanoemulsions were applied directly to the leaves of dwarf bean explants. Chlorophyll was extracted from the leaves and measured spectrophotometrically before and several days following treatment. The CPPU-loaded nanoemulsions enhanced the effectiveness of CPPU in delaying leaf senescence compared with the control experiments, including direct application of CPPU. ▼ Zoltán Kodály is primarily known outside of Hungary for his pedagogical work. However, his compositions should also be recognized for their excellence. His art songs for voice and piano, though mostly composed earlier in his career, represent an important contribution to the art song genre. Kodály was able to synthesize elements of foreign art song composers and his own folk culture to create an effective and important contribution to art song. He, along with Bartók, was one of the first and most significant Hungarian contributors to this genre. These works are rarely performed outside of Hungary due to language issues and availability. Through historical, textual, musical analysis, translations, and IPA, this dissertation will provide a performers’ guide for Kodály’s Op. 9 Öt dal. In addition, reasons for choosing Op. 9 include texts written by the contemporary poets Endre Ady and Béla Balázs, a lack of current scholarship, no complete English translations, and more compositional maturity while using Hungarian elements. This dissertation will allow more non-Hungarian musicians to teach and perform Kodály’s art songs.

▼ This study presents an analytical discussion of blurring techniques found in the first movement of Fauré's Piano Trio Op. 120. After a brief overview of Fauré's compositional style and the piano trio, the analysis proceeds in two parts. The first part examines small-scale blurring within the elements of cadence, phrase structure, and sequence. The second part observes large-scale blurring within the elements of key area and formal function. The paper concludes by suggesting that Fauré's systematic blurring of structural elements results in the decreased perception of musical architecture.

▼ The Wanderer Fantasy integrates qualities derived from three different genres: fantasy, tone poem, and sonata, and thus demonstrates a fusion of compositional styles that culminates in a complex formal structure. This document examines the Wanderer Fantasy through the individual lens of each genre and suggests a broader view and approach to the sonata form, one that considers Schubert’s distinctive musical language, as well as necessary structural adjustments influenced by the tropes of the fantasy genre. Such adaptations allow the projection of the two-dimensional sonata concept onto Schubert’s Wanderer Fantasy and thus provide an additional avenue to comprehend the structure of the piece.
