Požadavek činoherního divadla a přítomnost dalších klasických divadel ve městě vedlo k inspiraci divadlem řeckým; japonským; středověkým; alžbětinským; totálním; osvobozeným; arénovým. Moderní budova činohry národního divadla se nemůže spoléhat pouze na klasická schémata; i když i ta mohou být požadována. Návrh reflektuje divadlo především jako místo střetnutí dvou protichůdných světů; světa reálného; v němž žijeme; a světa zidealizovaného; přetvořeného; který vídáme na jevišti; světa skutečnosti a jejího obrazu; světa tragédie a komedie; světa skutečných tváří herců a jejich masek; jejich skutečných osobností a jejich rolí; světa skutečných věcí a kulis; světa prostorového a plošně zjednodušeného. Také dva základní protichůdné pohledy na divadelní umění jako na svět ideální; pompézní; výchovný; poučný a zábavný a na divadlo jako nemilosrdné zrcadlo našeho života; subjektivní; věrné či sarkastické; i rozpor těchto dvou chápání divadla můžeme zahrnout mezi divadelní dvojakosti. BUDOVA Nová budova divadla chce být zhmotněním dialogu; základního stavebního prvku divadelní tvorby; kterým Aischylos povýšil do té doby všeplatné zpěvné a taneční rituály na skutečné drama. Dialog spolu vedou dvě protichůdné formy – budova divadla a její prázdný otisk – veřejné prostranství. Klasifikace dialog a monolog se stává subjektivní; uvažujeme-li za druhý subjekt prázdno. Tato představa dodává úvaze existenciální náboj. Samotné dvě formy – plná a prázdná – mají kromě parafráze divadelní protichůdnosti; pověstného rozhraní dvou světů; přesah do mnoha dalších oblastí života – podobně jako samotné divadlo. Tvar budovy - zužující a zvyšující se hmota - vznikl spolu s prostranstvím spontánně jako asociace doplňkovosti. Tento tvar však reaguje kromě urbanistických a provozních návazností také na ideu moderního divadla; které by mělo být proměnlivé a dynamické. Vzletný tvar budovy zhmotňuje nejen tuto abstraktní dynamiku; ale je také intuitivním přepisem dynamiky hereckého projevu; přetlaku energie sršící z člověka; nepostradatelné nejen k hereckému výkonu; ale také ke všem lidským tvůrčím činnostem.; THEATRE Dramatic art is as old as human society. During its existence it has passed through long and colourful evolution which we charted probably only really partially. Requierement of a drama theatre and presence of another classical thetre buildings in the town caused an inspiration by Greek; Japonese; medieval; Elizabethan; Total; Exempt or arena theatre. A modern bulding of national drama theatre can´t depend of only calssical schemes; though even those could be reguired. The project reflect theatre first of all as a place of a battle of two contradictory worlds; a real world which we live in it; and an ideal; transformated world which we use to see on a stage; a world of reality and a world of its reflex; a world of tragedy and a of comedy; a world of existing faces of actors and a world of their masks; their real personalities and their parts; a world of true things and a world od scenes; a three-dimensional world and a 2D-simplified world. Also the two contradictory views of dramatic art; the first one as an ideal world; pompous; educational; instructive and funy and the second as a merciless mirror of our own lifes; subjective; authentic or caustic; so even this discrepancy of those two different opinions of theatre we could add in the theatre duplicity. BUILDING The new theatre building wants to be an objectification of a dialog; which is the basic structural element of dramatic production; whith which Aischylos sublimated singing and dancig rituals which were of binding force in that times to a real drama. This dialog is runned by two antagonistic forms – the theatre building and its empty impression – a public space. Established classification of dialog and monolog becomes subjective; if we suppose an emptiness as the second object. This idea gives our meditation an existencial character a bit. The two forms by themselves – the full one and the empty one –except of an imitation of theatre dialectics; a proverbial boundery-line of two worlds; also transcend into many other spheres of human life – analogous to a theatre. The shape of the building – a diminished and raising mass – has arised within the open space spontaneously as an association of complementing. But this shape – except other urbanistic and functional sequences - also reacts to an ide of modern theatre; which should be variable and dynamic. The poetic shape of the building not only materializes thist abstract dynamism but it is also an intuitive transscription of dynamism of an actor´s manifestation; an overpressure of human energy which is indispensable not only for a performance but also for all human creative activities.

▼ Emil Petrovics (1930-2011) was an award-winning, prominent music figure in the history of Hungarian music. Known primarily for his one-act opera C'est la guerre, Petrovics also wrote film music, an oratorio, a string quartet, and numerous instrumental and vocal works. Among his instrumental music, Concertino is the only solo work Petrovics ever composed for solo trumpet with orchestra. Significant professional posts for Petrovics included professor of composition and conducting at the Franz Liszt Academy of Music, musical director of the Petofi Theatre in Budapest, president of the Hungarian Association for Copyright Protection, and director of the Hungarian State Opera. In addition, Petrovics was a member of the Hungarian Parliament. The purpose of this study is to promote the work of Emil Petrovics through a performance edition of Concertino transcribed for brass ensemble. A secondary purpose of this study is to provide insight into the compositional process of Petrovics with the support of personal interviews with the composer prior to his death. The document presents a biographical sketch of Petrovics, a brief description of the three-movement work, and includes the arrangement of the Concertino for brass ensemble. Trumpeter Gyorgy Geiger and the Budapest Radio Symphony Orchestra commissioned the orchestral version of Concertino by Petrovics in 1990. Editio Musica Budapest published a piano reduction of Concertino, but the orchestral score was not published. Special permission to arrange the work was required and subsequently granted by the composer and EMB in December 2010. Although the trumpet solo line to Concertino has remained the same as the original, the accompaniment was rescored from woodwind and string instruments to brass and percussion instruments. Occasionally, parts that are not possible to play on brass instruments because of range, speed, or other technical difficulties are accomplished in the percussion lines. Prior to his death in June 2011, Petrovics reviewed the transcription of the first movement of the Concertino. He suggested the addition of string bass to the orchestration of the accompaniment, making it unique from other brass ensemble arrangements, and he hoped that this arrangement would inspire a performance in Budapest someday.

▼ Research Doctorate - Doctor of Philosophy (PhD) p>In spite of the highly fluctuating oil prices seen over the past number of years, the availability of relatively inexpensive sources of liquid hydrocarbons is diminishing, and the development of economically and environmentally viable alternative technologies to supply these hydrocarbons is vital. Projecting into the short and mid-term futures, the world economy is expected to continue to depend on hydrocarbon resources for stability, and thus technologies to produce these resources are important. Synthesis gas (CO and H₂), can be produced from a number of sources and feedstocks, such as biomass, waste and natural gas, and synthesis gas can be converted into liquid fuels such as hydrocarbons and alcohols, which are currently produced in large quantities in the conventional petrochemical industry. The selective oxidation of hydrocarbons, and in particular natural gas of which CH₄ the primary constituent, is notoriously difficult to control, mainly due to propensity of the reaction to generate to products of complete combustion. N₂O is a potent greenhouse gas (CO₂(e) = 300) and its concentration in the atmosphere is continuously increasing [Lee, Ryu et al, International Journal of Greenhouse Gas Control, January 2011, Pages 167–176]. Despite its greenhouse gas notoriety, N₂O is also recognised as a more selective oxidant than O₂ and has been used to selectively oxidise CH₄ to produce valuable products including synthesis gas. The production of synthesis gas, via its reaction with N₂O could be beneficial environmentally, as it could be used to provide valuable products when used to reaction with natural gas. The main aim of this research work is to investigate the scope of selective oxidation of natural gas (mainly CH₄) with N₂O over cobalt catalysts. The catalytic work was carried out in a continuous flow fixed bed micro reactor. Catalysts were prepared according to literature methods, as well as a new novel deposition method. The techniques used for catalyst characterization were powder X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), In situ Fourier transform infrared spectroscopy (FTIR), nitrogen adsorption/desorption isotherms, temperature program desorption (TPD), chemisorption, transmission electron microscopy (TEM). The elemental composition of catalyst samples was determined by using inductively coupled plasma optical emission spectrometer (ICP-OES). Based upon the experimental results obtained, the oxidation of CH₄ with N₂O over cobalt exchanged ZSM-5 (Co-ZSM-5) catalyst produced synthesis gas at a relatively high yield (19 %). When O₂ (instead of N₂O) was used as an oxidant over Co-ZSM-5 catalyst, synthesis gas was not formed. Similarly, when CH₄ and N₂O reacted over a cobalt on alumina catalyst, synthesis gas was not produced. Interestingly, H₂ was not detected as a reaction product when CH₄ with N₂O reacted over a Fe-ZSM-5 catalyst and only CO_x and H₂O were formed. Halliday’s metafunctional functionalism, which forms the basis for the semantics of his grammar, is based upon a revision of Bühler’s triadic functions of language. However, the more philosophical, classical American branch of the pragmatic tradition developed a likewise triadic understanding of experience, which also differs from Bühler’s. Whitehead’s pragmatic metaphysics provides the most comprehensive example of such a non-reductive philosophical system, crowned by an aesthetically-based general theory of value. p>These different functional trajectories suggest the possibility of reconstructing Halliday’s central functional notions, including his three metafunctions and subject notions (Theme, Subject and Actor), thereby radicalizing existing critiques advanced by McGregor, Martin, Bateman and others. On this view, the organic unity of language is based upon its expressive compositionality, grounded in the compositionality of value rather than independent metafunctions. An emphasis on action, motivated by the expression and realization of value, means that the underlying conceptions of language also differ. Although Halliday’s grammar is amongst the more comprehensive, functional linguistic theories, his functionalism is based upon the system network formalism. On this pragmatic view, the contents conveyed by a grammar are not abstract, general categories as Halliday holds, but rather valuations intrinsic to the organization of language itself. These function to organize the now grammatically central activity of reference, understood not representationally, as language transcendent, but immanently. The content of the grammar, now understood as composed of indexicals or shifters, generalized beyond words to all forms of grammatical expression, is, then, essentially reflexive, relative and variable. ▼ Abstract The present study is concerned with the mechanism of growth of porous anodic films formed on high purity aluminium and sputtering deposited aluminium over a wide range of current density between 5 to 50 mA/cm2 for times up to 5400 s in 24.5 wt % sulphuric acid and at temperatures of either 0 or 20 0C. The resultant films were examined using scanning electron microscopy (SEM), transmission electron microscopy (TEM), Rutherford backscattering spectroscopy (RBS), nuclear reaction analysis (NRA), optical interferometery, microhardness and nanoindentation.The RBS analysis enabled determination of the composition of the porous films, which was expressed as Al2O3.xAl2(SO4)3, with the sulphur species content increasing with increase in current density and decrease in temperature. The average expansion factor (expressing, the ratio of the film thickness to the oxidized aluminium thickness) increased between 5 mA/cm2 and 50 mA/cm2 for films formed at 0 0C, extending from 1.58 to 1.88 and from 1.57 to 1.78 according to SEM and TEM respectively. For films fabricated at 20 0C, the average expansion factor increased from 1.45 to 1.66 and from 1.42 to 1.67 derived from SEM and TEM respectively. The expansion factor increases as the current density increases for both temperatures, and decreases as electrolyte temperature for a given current density increases. The increase of expansion factor is also associated with a rise in the steady voltage during film growth. However, the film expansion does not depend on the anodizing time. The increase in expansion factor correlates with a small increase in the amount of sulphur in the film, which increases with rise of current density. The surface of the porous alumina revealed a network of furrows and ridges, reflecting the pattern of the cellular textures on the topography of the elecropolished aluminium. The retention of topography indicates that the thinning of the film due to chemical dissolution by the electrolyte is negligible, although softening of the film toward the film surface increases with rise of electrolyte temperature and anodizing time as determined by microhardness measurements on film cross-sections. For films fabricated at 0 0C, nodules appeared with a low population density on the film surfaces formed at 20 mA/cm2 for 5400 s, and a locally high population density, but non-uniform distribution, for films formed at 30 to 50 mA/cm2 for a wide range of anodizing times. NRA determined the oxygen concentrations in the films, from which the efficiency of the film grown was derived. The efficiency showed a correlation with the expansion factor, with values increasing with rise of current density and with decrease in the anodizing temperature, ranging from 72 % to 87 % between 5 mA/cm2 and 50 mA/cm2, for an electrolyte temperature of 0 0C, and between 66 % to 75 %, for the same range of current density, for an electrolyte temperature of 20 0C. A utilização destas redes revela-se de particular importância nas situações em que a instalação antecipada de infra-estruturas de suporte não é possível ou desejável. Os algoritmos de difusão realizam a entrega de mensagens ao maior número possível de participantes na rede. A difusão é uma peça fundamental de muitos dos protocolos que se antecipa virem a ser usados nas MANETs. Dada a escassez de recursos que caracteriza estas redes, é da maior importância que a disseminação seja concretizada com o menor custo possível, tendo sido propostas alternativas ao dispendioso mas popular algoritmo de inundação. Este trabalho avalia experimentalmente, através de simulações, o impacto de quatro algoritmos de difusão no desempenho do Ad hoc On Demand Distance Vector (AODV), um dos protocolos de encaminhamento mais citados para MANETs. Adicionalmente, é apresentada uma biblioteca para o sistema operativo Linux, que concretiza um dos algoritmos de difusão estudados. p>Mobile ad hoc networks (MANETs) are composed exclusively of the participants’ devices. These networks are particularly relevant in scenarios where the deployment in advance of an infra-structure is not possible or desirable. In MANETs, broadcast algorithms aim at delivering messages to the biggest number of participants. Broadcast operations are a fundamental building block for many protocols used in MANETs. Given that devices have limited resources, it is of utmost importance to reduce the resource consumption of each broadcast. Therefore, many alternatives to the costly, but popular, flooding algorithm, have been presented. This document reports the experiments and evaluation, through simulations, of the performance of the Ad hoc On-demand Distance Vector (AODV) protocol with four distinct broadcast algorithms. ▼ Deterministic Lateral Displacement (DLD) is a method capable of sorting cells based on size where mechanicalinteractions between a sufficiently large particle and obstacles in a microfludic pillar array force the particle tofollow a different trajectory than their smaller counterparts, resulting in continuous lateral separation. To extendthe capability of DLD, electrical interaction between particles and pillars can be employed to complement themechanical interaction, making electrical/dielectric properties additional parameters for sorting. Another idea isto exploit the morphologies of cells and as a concequence, their dynamical properties, to sort them in DLD. Thedevelopment of DLD cell sorting methods based on those two ideas has brought forth five papers appended to thisthesis: paper I, III, and V (combination of electrokinetics and DLD), and paper II and IV (exploiting morphologyin sorting by DLD).In the first topic, differences in electric properties or dielectric properties of particles and cells are employed toextend the capability of DLD. In Paper I, an AC electric field was applied across DLD devices having insulatingpillars to sort similar-sized polystyrene particles having different surface charge, viable from non-viable yeast cells,and viable from non-viable E. coli bacteria. In Paper III, the same method was utilised on open channel DLDdevices, showing unaltered effectiveness but offering the ability to flexibly change the distance between the electrodes.Also in the topic of combining electrokinetics and DLD, Paper V introduced a new type of DLD devicewhere the electrodes were defined locally on every pillar, making it easier to generate a high electric field strength.Besides electrical properties, morphology is another useful accompaniment to DLD. In Paper II, pathogenicStreptococcus pneumoniae bacteria were fractionated in DLD devices according to the difference in their morphology,viz. their chain length. It was also demonstrated, in paper IV, that an AC field can be used to rotatenon-spherical red blood cells and in turn, change their trajectory in a DLD device. This implies an opportunity tosort red blood cells from cells having different morphology, either spherical cells or parasites like trypanosomes.

▼ Middle managers are tasked with supporting new initiatives and change, while at the same time experiencing challenges in making sense of the strategic initiative and giving sense to other members of the organisation. This dissonance experienced by middle managers tasked with change informed the topic of the research. The current study explores the strategising of middle managers through sensemaking and sensegiving during a strategic initiative in a single case study within a financial services provider in South Africa. The study seeks to examine the phenomenon through the theoretical lenses of the strategy-as-practice perspective, middle manager perspective and the theory of sensemaking and sensegiving. The study adopted a qualitative-exploratory design which involved collecting data through semi-structured in-depth individual faceto-face interviews. Data was analysed through coding, using thematic analysis and the categorisation and interpretation of common themes. Three main themes emerged from the study which were categorised as middle manager dissonance, middle manager sensemaking and sensegiving actions and middle manager practices in sensemaking and sensegiving. These themes offer insight into middle manager strategising, sensemaking and sensegiving during a new strategic initiative. The study concludes that when the organisation introduced a new strategic initiative middle managers’ knowledge was disturbed. These middle managers experienced various emotions as they were trying to make sense of the changes while giving sense to other members of the organisation. Findings confirmed delays in implementation due to lack of understanding of the change by middle managers. The study also concludes that although middle managers experienced challenges initially, they moved into a task of “selling” the new initiative through sensemaking and sensegiving. Middle managers got involved in various practices such as team discussions and information sharing sessions as they make sense and give sense to other team members. Through this interaction, middle managers aligned others and created common understanding while they influence them through sensegiving. Findings of the current study may offer valuable knowledge to organisations in the financial sector and practitioners tasked with v new strategic initiatives. OBJETIVOS: 1. Profundizar en el conocimiento morfológico de las especies ibéricas perennes y así contrastar los tratamientos previos: • Grupo de Chaenorhinum origanifolium; • Grupo de Chaenorhinum macropodum; • Grupo de Chaenorhinum villosum; • Especies gipsófilas de Chaenorhinum. 2. Sentar las bases morfológicas para la diferenciación de los táxones conflictivos o dudosos del género Chaenorhinum, para poder abordar sobre ellas futuros estudios sistemáticos o filogenéticos basados en los caracteres moleculares. 3. Comprobar la utilidad de los caracteres micromorfológicos para la descripción de nuevas especies. METODOLOGIA: Material. Las especies seleccionadas del género Chaenorhinum son todas las perennes, originarias del Mediterráneo occidental y la mayoría endémicas de la Península Ibérica. El material estudiado procede tanto de muestras procedentes del material seco cedido por varios herbarios, como de material fresco procedente de recolecciones propias efectuadas directamente en el campo. Los estudios morfológicos de semillas y tricomas se han realizado sobre 3-6 poblaciones por taxón, teniéndose en cuenta que las poblaciones estuvieran repartidas por toda el área de distribución de cada taxon. Técnicas de observación. Se recurre al análisis morfológico de los caracteres diferenciales para poder establecer los grupos de estudio en función de las ordenaciones previas realizadas sobre el género. La dificultad de encontrar caracteres macromorfológicos que nos permitan diferenciar los distintos táxones nos obliga al estudio de los caracteres micromorfológicos: tricomas e indumento, y la ornamentación de las semillas, que han demostrado ser de gran utilidad en estudios previos realizados en el género. Las técnicas de observación empleadas son: Microscopía esteroscópica: nos ha permitido tomar las medidas de los ejes mayores de las semillas, así como observar su forma y la ornamentación de sus crestas. Nos ha proporcionado también, la visión de la superficie de las plantas a escaso aumento y, al ser un medio de observación directa que no precisaba de la preparación previa del material, hemos podido estudiar muchos individuos en poco tiempo. Microscopía óptica: se ha utilizado para medir los tricomas, así como para ver su forma, y el número, forma y disposición de las células que los componen. Microscopía electrónica de barrido: nos ha permitido observar la forma y la ornamentación superficial de las semillas y tomar las medidas de sus elementos, así como apreciar el indumento y la forma y ornamentación de los tricomas. ▼ The present study is concerned with the mechanism of growth of porous anodic films formed on high purity aluminium and sputtering deposited aluminium over a wide range of current density between 5 to 50 mA/cm2 for times up to 5400 s in 24.5 wt % sulphuric acid and at temperatures of either 0 or 20 0C. The resultant films were examined using scanning electron microscopy (SEM), transmission electron microscopy (TEM), Rutherford backscattering spectroscopy (RBS), nuclear reaction analysis (NRA), optical interferometery, microhardness and nanoindentation.The RBS analysis enabled determination of the composition of the porous films, which was expressed as Al2O3.xAl2(SO4)3, with the sulphur species content increasing with increase in current density and decrease in temperature. The average expansion factor (expressing, the ratio of the film thickness to the oxidized aluminium thickness) increased between 5 mA/cm2 and 50 mA/cm2 for films formed at 0 0C, extending from 1.58 to 1.88 and from 1.57 to 1.78 according to SEM and TEM respectively. For films fabricated at 20 0C, the average expansion factor increased from 1.45 to 1.66 and from 1.42 to 1.67 derived from SEM and TEM respectively. The expansion factor increases as the current density increases for both temperatures, and decreases as electrolyte temperature for a given current density increases. The increase of expansion factor is also associated with a rise in the steady voltage during film growth. However, the film expansion does not depend on the anodizing time. The increase in expansion factor correlates with a small increase in the amount of sulphur in the film, which increases with rise of current density. The surface of the porous alumina revealed a network of furrows and ridges, reflecting the pattern of the cellular textures on the topography of the elecropolished aluminium. The retention of topography indicates that the thinning of the film due to chemical dissolution by the electrolyte is negligible, although softening of the film toward the film surface increases with rise of electrolyte temperature and anodizing time as determined by microhardness measurements on film cross-sections. For films fabricated at 0 0C, nodules appeared with a low population density on the film surfaces formed at 20 mA/cm2 for 5400 s, and a locally high population density, but non-uniform distribution, for films formed at 30 to 50 mA/cm2 for a wide range of anodizing times. NRA determined the oxygen concentrations in the films, from which the efficiency of the film grown was derived. The efficiency showed a correlation with the expansion factor, with values increasing with rise of current density and with decrease in the anodizing temperature, ranging from 72 % to 87 % between 5 mA/cm2 and 50 mA/cm2, for an electrolyte temperature of 0 0C, and between 66 % to 75 %, for the same range of current density, for an electrolyte temperature of 20 0C. ▼ Metallosupramolecular chemistry involves the construction of nanoscale molecular assemblies by reacting metal atoms with bridging organic ligands. The metal atoms act as a type of molecular ‘glue’ binding together the organic ligands in specific orientations. Thus, appropriate combinations of metal ions and ligands lead to the controlled self-assembly of interesting one-, two- and three-dimensional molecular aggregates. This thesis details the preparation of a range of novel flexible bridging heterocyclic ligands using conventional organic synthesis, and then explores their reactions with a variety of transition metal precursors. By varying the nature of the organic ligand and the transition metal precursor, new and exciting supramolecular topologies and architectures can be formed. A total of forty-eight ligands were synthesised in this work, forty-seven of which are new compounds. The majority of the ligands synthesised were based around commercially available bisphenol cores. All forty-eight of the ligands had nitrogen heterocyclic groups as coordinating units. The ligands discussed in this thesis can be divided into three main sections. The first involves the synthesis and coordination chemistry of two-armed ligands based around the Bisphenol A, Bisphenol Z and Bisphenol AP cores. The second section describes the synthesis and coordination chemistry of the larger Bisphenol P and Bisphenol M based two-armed bridging ligands. The third section describes the synthesis and coordination chemistry of various multi-substituted ligands, including tripodal ligands based around a trisphenol core, four-armed ligands and six-armed ligands. The two-armed bisphenol based ligands proved very successful as synthons in metallosupramolecular chemistry and produced many products with a variety of different metal atoms. The complexes characterised included discrete dimeric products, coordination polymers and a number of helicates, including a dinuclear quadruply-stranded helicate. Multi-armed ligands are topical, because they have multiple coordination sites that are capable of binding and bridging multiple metal atoms. Such coordination can lead to the construction of cage-like species and complicated networks. A series of three-armed ligands based around a trisphenol core were synthesised with the intention to use these to form such species on coordination with appropriate metal salts. Indeed, one of the products of self-assembly was an interesting M₃L₂ cage. Various other multi-armed ligands were also investigated. The ligands and complexes in this thesis were characterised by a variety of structural techniques, such as ¹H NMR, ¹³C NMR, mass spectrometry, elemental analysis and X-ray crystallography when crystals were obtained. The crystal structures of twenty-seven ligands and forty-three complexes are described.

▼ A leucodistrofia metacromática (MLD) é um erro inato do metabolismo de herança autossômica recessiva, dividido em três subtipos clínicos de acordo com a idade de início dos sintomas. Na MLD ocorre deposição intralisossômica de glicolipídeos sulfatados principalmente no sistema nervoso central. A causa dessa doença é a deficiência de arilsulfatase A (ARSA), uma glicoproteína lisossômica que catalisa a hidrólise de glicolipídeos sulfatados constituintes da bainha de mielina. Até o momento cerca de 140 mutações relacionadas à manifestação de MLD foram descritas no gene da ARSA, das quais duas delas ocorrem com alta frequência na maioria das populações estudadas. Ademais, entre 7% e 12% das pessoas saudáveis têm deficiência in vitro de ARSA, condição denominada pseudodeficiência de ARSA (PD-ARSA). Não existem dados de caracterização genotípica de pacientes brasileiros com MLD. Da mesma forma, dados sobre a dinâmica molecular (DM) da ARSA também são escassos, em parte por dificuldades metodológicas implicadas no estudo desses aspectos em glicoproteínas. Nesse contexto, os objetivos desse trabalho foram identificar os alelos mutantes no gene da ARSA em pacientes brasileiros com MLD e avaliar os efeitos da glicosilação e do pH na DM da ARSA. A amostra foi composta por 27 pacientes com MLD. A mutação mais frequente (c.459+1G>A) e polimorfismos constituintes do alelo PD-ARSA foram detectados por PCR em tempo real e as demais mutações por sequenciamento direto do gene da ARSA. As DM realizadas foram de (i) ARSA não glicosilada em pH~7 e (ii) em pH~5, (iii) ARSA triplamente glicosilada e (iv) deficientemente glicosilada p.N350S, usando GROMACS. Nesse estudo a mutação c.459+1G>A foi a mais frequente (0,50), conforme esperado pela alta prevalência de pacientes com MLD infantil em nossa coorte. As mutações p.P426L (0,08) e c.103_110del8 (0,08) também apresentaram frequências relevantes quando comparadas às demais mutações. No conjunto, 11 mutações raras foram identificadas, incluindo 2 mutações novas: p.S44P e p.P284S. Além das mutações potencialmente deletérias, foram identificados 2 polimorfismos neutros frequentemente associados à mutação c.459+1G>A (p.W193C [0,54] e p.T391S [0,65]) e 2 polimorfismos constituintes do alelo PD-ARSA (p.N350S [0,15] e c.1524+95A>G [0,04]).
