▼ Myocardial infarction (MI) is a leading cause of congestive heart failure (CHF), and its course of cardiac remodeling is of paramount importance in prevention and treatment of cardiac dysfunction. Activation of the sympathetic nervous system (SNS), and the renin angiotensin system (RAS), is critical in cardiac subcellular remodeling post MI. Although blockade has prevented remodeling, little is known regarding the beneficial effects in reversing subcelllular changes in failing hearts. Upon rat coronary artery occlusion - losartan, metoprolol, and prazosin were initiated 12 weeks post MI for 8 weeks, to evaluate their reversal effects at the molecular and cellular levels. As the sarcoplasmic reticulum (SR) primary regulates intracellular Ca2+ in cardiac contraction/relaxation, we hypothesized that abnormalities in its function and regulation contribute to contractile dysfunction. We examined cardiac performance, SR function, SR molecular expression and plasma catecholamine levels 20 weeks post MI. The fibrinous myocardium, cardiac hypertrophy and pulmonary edema all portrayed CHF, in addition to elevated LVEDP and depressed dP/dt. Ejection fraction, fractional shortening, and cardiac output were all significantly reduced, as were IVSs, LVIDd, LVPWs, and LVPWd. SR preparations showed altered phospholamban (PLB) and sarcoplasmic reticulum Ca2+ ATPase (SERCA2a) mRNA, which appropriately matched their protein expression. These modifications correlated to decreased cardiac SR Ca2+-uptake, providing further disruptions in Ca2+ homeostasis. Therefore, these modifications in the mRNA of PLB and SERCA2a are postulated to play a critical role in SR protein remodeling. Supplementary studies addressed remodeling of mRNA myofibrils, which revealed a decreased α-MHC isozyme with an increased β-MHC isozyme, and reduced myofibrillar Ca2+-stimulated ATPase post MI. Finally, circulating plasma catecholamine levels of norepinephrine, epinephrine and dopamine were significantly elevated. Losartan, metoprolol, and prazosin corrected lung edema, myocardial hypertrophy, cardiac contractile dysfunction, and attenuated PLB and SERCA2a proteins, whereas α- and β- MHC mRNA were only attenuated by losartan; and metoprolol only attenuated β-MHC mRNA. SR Ca2+-uptake activities and plasma catecholamines of norepinephrine were partially reversed, yet dopamine was only affected by losartan. Early work previously focused on prevention, but this study is one of the first to attempt reversal of cardiac subcellular remodeling in CHF due to MI. Advisors/Committee Members: Dhalla, Naranjan S. ▼ For decades, social scientists and practitioners have viewed public opposition to land-use change, including the development of affordable housing, through the lens of NIMBYism (“Not in My Back Yard”). Currently, there are two main limitations with research on the subject. First, much of what is understood about community opposition to affordable housing is investigated through the perspectives of third parties, such as developers and government officials, based on their experiences with the broader public. Second, there is relatively little qualitative research on what drives support from individuals and communities during the affordable housing development process. These constraints mean that knowledge of community reactions to affordable housing is both speculative and highly circumscribed. Building on a “strengths-based” approach inspired by Kurt Lewin’s force-field analysis framework, this research seeks to close these gaps by better understanding how community members describe their own support for affordable housing. To this end, I conducted 19 in-depth interviews with individuals connected to affordable housing issues in Washtenaw County, Michigan e.g. nonprofit leaders, current and former government officials, small business owners, and developers. Although my interview pool was similar to that of previous research, the analytical focus on personal support for affordable housing offers a fresh perspective to the field. I found that community leaders largely couched their support in the community-level benefits of affordable housing and in their own personal values, particularly those of equity and fairness. Looking to public education campaigns, I use these results to elaborate on untapped opportunities to advocate for affordable housing in Ann Arbor, MI, by centering campaign messaging on these themes. Overall, this study has demonstrated that we must look beyond merely responding to perceived “NIMBY” reactions and learn how to amplify the existing sources of support already existing in the community when advocating for affordable housing. This new approach to understanding and leveraging public support could be replicated in other contentious land-use change scenarios of public goods, including the siting of renewable energy generation and other human service facilities. ▼ Los acuerdos comerciales desempeñan un papel importante por las economías de los países en vías de desarrollo, incluyendo los Países Terceros Mediterráneos (PTM). Desde los años ochenta, la apertura comercial de Marruecos se ha ido concretando mediante la firma de acuerdos de libre comercio con varios países y bloques comerciales (UE, USA, Turquía, países de Agadir¿). Con la mayoría de estos socios comerciales la balanza comercial general y agrícola de Marruecos se mantiene en términos deficitarios a pesar de los esfuerzos de competitividad y establecer el equilibrio. La mayoría de las exportaciones agrícolas de los PTM se destinan al mercado europeo, sobre todo en las frutas y hortalizas, productos para los cuales disponen de ventajas comparativas y condiciones de acceso preferente. La literatura ha señalado que Marruecos recibe altos márgenes de preferencia por parte de la UE para sus frutas y hortalizas exportadas al mercado europeo. Con esta situación, el objetivo principal de esta Tesis es analizar cuantitativamente las implicaciones para Marruecos de las preferencias actuales y de hipotéticas liberalizaciones comerciales futuras. Consecuentemente se realizan dos análisis complementarios. En primer lugar, se calcula el Valor del Margen de Preferencia para varias frutas y hortalizas de importancia en las exportaciones marroquíes a la UE (tomate, naranja, pepino y clementina), considerando el nuevo acuerdo comercial entre la UE y Marruecos. Los resultados obtenidos señalan que el tomate es el producto con mayor alcance de estas preferencias, que la reducción del precio de entrada tiene diferentes implicaciones prácticas entre estos productos, y que la revisión del acuerdo ha supuesto mayores transferencias potenciales para Marruecos. En segundo lugar, en esta investigación se exploran diversas posibilidades de liberalización comercial de las importaciones de la UE para estos mismos productos, posibilidades que van desde una liberalización limitada a los productos marroquíes, a otras más generalizadas. Se realizan diversas simulaciones de equilibrio parcial teniendo en cuenta las peculiaridades de la protección comercial en frutas y hortalizas y diferentes orígenes de los productos. Los resultados de las mismas muestran caídas de precios en el mercado europeo y aumentos notables del volumen de las importaciones, y al mismo tiempo aumentos de los precios en frontera para el país exportador. Además, en todos los casos cabe destacar una pérdida en el bienestar de consumidor marroquí, y ganancias para los productores de este país. ▼ Syftet med studien var att undersöka klienternas och socialarbetarnas upplevelse av metoden motiverande samtal (motivational interviewing, MI), vilket besvaras genom frågeställningarna; hur upplever klienten kontakten med socialarbetaren som använder MI och vad är socialarbetarens upplevelse av MI metoden. Studien är genomförd med en triangulerande kvalitativ metod där studiens syfte blir belyst från tre olika perspektiv. Empirin baseras därför på två stycken intervjuer med klienter, två stycken intervjuer med socialarbetare och två stycken inspelade verkliga MI-sessioner mellan klient och handläggare. Som teoretisk referensram har resultatet analyserats utifrån begreppet empowerment. Resultatet visar att relationen mellan socialarbetare och klient är en avgörande faktor för att kunna bryta ett destruktivt beteende. Att formulera delmål på vägen mot ett större mål anser samtliga informanter vara av stor vikt i förändringsarbetet. Klienten har ett egenansvar om att förändra sin livssituation, där den ena klienten ställer sig negativ till eget ansvar. Båda klienterna berättar att, när de befunnit sig i ett aktivt missbruk har kontakten med socialtjänsten endast handlat om att få pengar eller utnyttja systemet på något sätt. I denna fas finns inte någon motivation att förändras, vilket leder till svårigheter. Resultatet visar att klienterna har upplevt sin handläggare som stöttande och en trygghet i processen att bli fri från sitt missbruk. p> The aim of the study was to investigate the clients 'and the social workers' experience of the method of motivational interviewing, which is answered by the questions; how do clients experience the contact with social workers who use MI as a method and what is the social worker's experience of MI as a method. The study was conducted with a triangulation qualitative method where the purpose of the study was illuminated from three different perspectives. The empirical data is therefore based on two interviews with clients, two interviews with social workers and two MI-recorded real-sections between the client and the social worker. As a theoretical framework, the result has been analyzed based on the concept of empowerment. The result shows that the relationship between social worker and client is an essential factor to break destructive behavior. All of the informants believes that formulating partial goals is an important part in the changing process. The client has a personal responsibility to change their situation, one client is negative to that responsibility. Both of the clients report that, when in an active addiction, the contact with the social services only were about gain money or use the system. In this phase there is no motivation to change, which leads to difficulties. The results show that clients have experienced their administrators as supportive and secure in the process of becoming free from their addiction. p>

▼ Bakgrund: MI är en metod för rådgivning och för att hjälpa individen att hitta och bibehålla motivation till förändrat beteende. På senare år har metoden börjat användas även inom tandvården. Syfte: Syftet med studien var att genom en litteraturöversikt sammanställa effekten av MI på parodontala sjukdomar och karies hos barn, ungdomar och vuxna. Metod: Studien består av en litteraturöversikt där vetenskapliga artiklar har tagits fram genom sökningar i databaserna PUBMED, DOSS, Medline, Psychinfo och CINAHL som sedan granskats och sammanställts. Sjutton artiklar har utvalts utifrån de inklusion/exklusion kriterier som fanns. De studierna som blev inkluderade i resultatet har blivit granskade med hjälp av en mall för att kunna identifiera studiernas värde för litteraturstudien. Resultat: De utvalda artiklarna var från Frankrike, Kanada, Kina, Tyskland, Iran, Indien, Serbien, Spanien, Sverige och USA. Ungefär hälften av artiklarna påvisade ett positivt resultat för patienter som genomgått MI-samtal i kombination med vanlig behandling jämfört med kontrollgrupper. Den andra hälften såg inga dylika resultat. Slutsats: Metoden tycks ha effekt i vissa sammanhang men inte i andra. En faktor som inte visade någon påverkan på effektiviteten av MI var patienternas ålder. Ytterligare forskning krävs för att fastställa vilka faktorer som påverkar resultatet vid användning av MI inom tandvården. p> Background: MI is a method for counseling and for helping individuals find and maintain motivation for change of behavior. In later years the method has also seen some use in dental care. Aim: To study the effect of MI on parodontal diseases and caries in children, adolescents and adults. Method: The study design was a literature review where scientific articles were gathered by searching through databases like for example PUBMED, DOSS, Medline, Psychinfo and CINAHL and then scrutinized and summarized. Seventeen articles were selected according to existing inclusion-/exclusion criteria. The included studies were reviewed with a modified review template to present studies of strong/moderate value. Result: The selected articles were from Canada, China, France, Germany, Iran, India, Serbia, Spain, Sweden and USA. About 50% of the articles indicated a positive result for patients who undertook MI-sessions in combination with regular treatment, compared to control groups. The other half could see no such results. Conclusion: The method seems to have effect in some instances but not in others. A factor that showed no alteration of MI effectiveness was the patients’ age. Further research is required to determine what factors might influence the outcome of using MI in dental care. p>

▼ Low levels of physical activity (PA) during pregnancy are associated with detrimental health outcomes, not only for the mother but also for her child. Pregnant WMO are less likely to meet recommended PA (20-30 minutes/day), most days/week. They have higher overweight (76.9%) and obesity (45%) rates compared to the US population, and face increased risk for excessive GWG, GDM and T2DM. Nonetheless, most research available on PA during pregnancy has focused on NHW women. Employing a socioecological approach, this dissertation built on literature describing pathways between PA-related beliefs, attitudes, subjective norms and perceived behavioral control (individual level), social support (interpersonal level), and neighborhood poverty, social cohesion, safety, walkability, presence/condition of parks/playgrounds, and car/truck traffic (neighborhood level) and PA among pregnant and non-pregnant NHW, and racial and ethnically minority women, and the literature on interventions aimed at increasing PA. However, these pathways and interventions have not been examined among pregnant and postpartum WMO. The purpose of this dissertation is to examine pathways that individual, interpersonal and neighborhood factors may influence PA and assess the efficacy of a behavioral intervention to reduce PA declines among pregnant and early postpartum WMO. The Theories of Planned Behavior and Social Support grounded this research. Using a dataset from an RCT of a behavioral intervention, first I examined independent and joint relationships of individual-level factors and PA intention/behavior; and social support moderating effects. Findings suggest significant associations between attitude and PBC with PA intention but not PA behavior. PA-social support did not moderate relationships of PBC-PA intention nor PBC-PA behavior. Independent associations between social support and PA behavior were significant. Joint relationships between TPB constructs and social support, and PA accounted for 6.8% of the variance in regression models. Second, I examined relationships of neighborhood characteristics and PA. I found positive significant independent and joint associations between neighborhood social cohesion, walkability, and presence/condition of parks/playgrounds, and PA. Neighborhood poverty, safety and heavy car/truck traffic were not significantly associated with PA. Neighborhood safety perceptions moderated the neighborhood poverty-PA association. Associations of PA-Neighborhood characteristics accounted for 22% of the PA variance. Third, I examined the efficacy of a lifestyle intervention to reduce PA declines among pregnant and postpartum WMO. Findings suggested a significant PA decline from T0(baseline) to T1(follow up), and an increase from T1 to T2(postpartum), but still lower PA at T2 compared to T0. The PA- HLI group had significantly lower PA declines than the comparison group during pregnancy and early postpartum. ▼ CELL AND DEVELOPMENTAL BIOLOGY The role of the BMP antagonist Gremlin2 during cardiac tissue repair Lehanna Nalani Sanders Dissertation under the direction of Professor Antonis Hatzopoulos Acute myocardial infarction (MI) or heart attack occurs when an artery becomes occluded, preventing blood flow thereby causing severe cardiac cell loss. Massive cell death induces an inflammatory response, which leads to granulation tissue deposition and eventual scar formation. This results in cardiac tissue remodeling and heart failure. Although these sequential events of cardiac tissue repair have been well described, the regulation of each phase is not entirely understood. Molecular pathways necessary for development are often re-activated during injury. The BMP pathway has been known to be important for cardiac development and morphogenesis as well as being induced post-MI. However it’s role in the tissue repair process or how it is regulated is not well known. We recently showed that the Bone Morphogenetic Protein (BMP) antagonist Gremlin 2 (Grem2) is required for cardiac tube looping and cardiomyocyte differentiation during zebrafish development. In this work we show that in adult mice, BMP signaling components including Grem2 is strongly and transiently induced after myocardial infarction (MI) within the peri-infarct area during the inflammatory phase of cardiac tissue repair. By engineering loss- (Grem2-/-) and gain- (TGGrem2) of-Grem2-function mice, we discovered that Grem2 controls the magnitude of the inflammatory response and limits infiltration of inflammatory cells in peri-infarct ventricular tissue, improving cardiac function. Excessive inflammation in Grem2-/- mice after MI was due to overactivation of canonical BMP signaling and suppressed by the BMP inhibitor DMH1. Furthermore, intra-peritoneal administration of Grem2 protein in wild-type mice was sufficient to reduce inflammation after MI. Cellular analyses showed BMP2 acts synergistically with TNFα to induce expression of pro-inflammatory proteins in endothelial cells, whereas Grem2 specifically inhibits the BMP2 effect. The role of Grem2 appears to be specific to the inflammatory phase since changes in Grem2 expression did not directly alter the phenotype of the granulation tissue deposition and fibrotic phases of recovery. Finally, high-throughput sequencing analysis is being implemented in order to determine any previously unknown targets and interactions of Grem2 for future analysis. ▼ As concerns regarding the growing human population, rising seafood demand, and up to present limited success of fisheries management intensify, aquaculture is increasingly posited as a means to more efficiently produce seafood commodities. However, aquaculture expansion raises contentious issues itself. The current study addresses a number of these issues in attempts to improve hatchery production and related activities. Investigation of less-harmful chemotherapeutants for disease reduction at the egg stage could lead to improved hatchery microbial management, increased survival during early larval stages, increased efficacy of bacterial probiotic incorporation, and reductions in disease transfer between hatcheries and locations. Attempts to surface sterilize and disinfect cobia Rachycentron canadum eggs with 3 and 2% hydrogen peroxide solutions significantly reduced survival through the yolk-sac larvae stage. Furthermore, timing of treatment application at differing stages of egg development was found to significantly impact survival, highlighting the importance of this compounding factor. Bacterial probiotics can remediate water quality, reduce target host stress, and improve survival and population growth rates of live feed organisms. Two of the following studies investigated the benefits of incorporation of a Bacillus spp. probiotic blend in aquaculture activities. During closed container mock shipment of yellowfin tuna Thunnus albacares yolk-sac larvae, significant reductions in total ammonia nitrogen resultant from probiotic incorporation were observed. Furthermore, significantly reduced dissolved oxygen utilization might have resulted from stress reduction, as suggested by a non-significant lower degree in the breakdown of osmoregulation of the larvae. Incorporation of probiotics within rotifer Brachionus plicatilis cultures resulted in significantly higher daily mean populations, significantly lower population variability among replicates, and non-coincident logistic population growth regressions which yielded higher population growth rates and system carrying capacities. Broodstock management should not only maximize quantity of reproductive output, but quality as well. Significant increases in cobia egg diameter with increasing broodstock female age were detected, as well as significant increases in egg diameter with decreasing salinity. Confirmation of potentially increased larval growth rates resultant from increased energy stores of larger eggs when produced by older females, as demonstrated in other species, is warranted. Attempts to find production parameters which maximize cobia larval growth and survival would lead to increased sustainability via reducing demand for wild-collected Artemia and economic efficiency. Gaps in knowledge for achieving this goal continue to exist, but conclusions drawn from analysis of multiple production scale trials indicate temperatures of 29-31 °C maximize growth and survival. ▼ Motiverande samtal (MI) är en populär samtalsmetod med fokus på motivations- och förändringsarbete. MI utgår från ett teoretiskt underbyggt förhållningssätt där rådgivaren och klienten tillsammans utforskar möjliga vägar till förändring. För att säkerställa att MI används på bästa sätt kan samtalsledarens skicklighet och förmåga observeras kodas enligt olika variabler. Dessa mätningar bör i sin tur besitta en hög interbedömarreliabilitet (IBR) för att ge tillförlitliga resultat. Absolut överensstämmelse i samtliga variabler ger maximal IBR, medan en låg IBR kan tyda på att mätinstrumentet är otydligt, att det finns brister hos bedömarna, eller på en kombination av detta. I den här uppsatsen undersöks IBR i The Motivational Interviewing Treatment Integrity Code (MITI), ett kodningsverktyg som tagits fram för att vara reliabelt och lätt att använda. Samtalssekvenser om tjugo minuter kodas enligt fjorton variabler. Fyra oberoende kodare har kodat tolv samtal enligt den senaste versionen av mätinstrumentet (MITI 4.2.1). Samtliga kodningar genomfördes inom ramen för en kodarutbildning som hålls av ett professionellt kodningslabb. Intraklasskorrelation (ICC), Krippendorffs α (Kalpha) samt Cronbachs α redovisas parallellt som mått på IBR. Flera variabler visar en otillfredsställande eller, beroende på vilken koefficient som används, oacceptabel IBR. De fyra kodarna visar en lägre samstämmighet i bedömningen av kvalitativa variabler samt variabler avseende beteenden ej förenliga med MI. ICC för fyra kodare ligger generellt på en måttlig men godkänd nivå, medan Kalpha genomgående ligger under gränsen för det godtagbara. Cronbachs α ligger överlag något över ICC vilket tyder på en viss systematik i kodarnas bristande överensstämmelse. En tänkbar orsak till en bitvis sviktande IBR är att de undersökta kodarna vid mättillfället var relativt oerfarna och skulle behövt mer praktisk träning i MITI 4.2.1. Vidare kan en bristande förståelse för instrumentets teoretiska grunder ha inverkat negativt på IBR. För att MITI ska kunna utvärderas korrekt i framtida forskning krävs ett enhetligt grepp för hur IBR ska mätas. En högre förståelse för metoderna bakom mätninga av IBR är önskvärd.

▼ MI (Motivational Interviewing) är en samtalsmetod som avser att förbereda individer för förändringar. Socialstyrelsen (2004) anger i sina riktlinjer att svensk skolhälsovård ska använda evidensbaserade metoder i det hälsopedagogiska arbetet. Fortlöpande utbildning i MI har genomförts de senaste åren. Skolsköterskan har en god inblick i skolelevers hälsoläge avseende ohälsosamma levnadsvanor, och har dessutom en möjlighet att påverka dessa. Syftet med studien är att undersöka skolsköterskors användning av MI i sitt omvårdnadsarbete med gymnasieelever. Som omvårdnadsteoretisk ram användes Dorothea Orems egenvårdsteori samt Aaron Antonovskys salutogena perspektiv. Studien är en kvantitativ stickprovsundersökning. Enkätfrågorna omfattar skolsköterskors användning av MI i dagligt arbete, MI-teknikens grundprinciper samt eventuella behov av stöd och handledning för att vidmakthålla sina kunskaper i MI-tekniken. Resultatet visar att skolsköterskor använder MI i sitt omvårdnadsarbete i stor utsträckning och att metoden anpassas för implementering i det dagliga arbetet. Majoriteten anser att MI-metoden fungerar bättre än traditionell rådgivning vid livsstilsförändringar. Begränsningar utgjordes dock av grundprincipen att uppmärksamma förändringsprat hos eleven, samt att det finns ett stort behov av handledning och stöd för att vidmakthålla MI-kunskaperna. I vidare forskning skulle det vara intressant att undersöka vilket stöd och handledning det finns behov av och hur stöd och handledning bör utformas. p> MI (Motivational Interviewing) is a conversational approach designed to prepare individuals for changes. Social Board specifies in its guidelines to Swedish school health services that they should use evidence-based practices in the health pedagogic work. Continuous education in MI has been implemented in recent years. School nurses have a good insight into the school students´ health mode for unhealthy living habits, and they also have an opportunity to influence these. The purpose of the study is to examine school nurses´ use of MI in their nursing work with secondary school students. Dorothea Orem’s self-care theory and Aaron Antonovsky’s salutogenic perspective were used as a theoretical frame. The study is a quantitative sampling. Survey questions covered the school nurses´ use of MI in daily work, MI-technique fundamentals, along with any need for assistance and guidance in order to maintain their skills in MI-technology. The result shows that school nurses are using their nursing work extensively and that the method adapts for implementation in their daily work. The majority believes that the MI-method is working better than traditional advice at lifestyle changes. O livro foi publicado na Argentina, trata-se de obra de carÃter autobiogrÃfico que ratifica a imagem de uma mulher que se tornou um Ãcone do peronismo de visÃo quase messiÃnica no contexto latino. A representaÃÃo de Eva PerÃn evoca tambÃm consideraÃÃes de gÃnero, ou seja, revela a consciÃncia de uma mulher que transgrediu os padrÃes de uma Ãpoca, conseguindo transformar-se em lÃder de um movimento polÃtico, o movimento do partido peronista, movimento este que surgiu na Argentina na dÃcada de 1940, quando o paÃs esteve sobre o comando dos militares. Desde a sua morte, em 26 de julho de 1952, a histÃria de Eva PerÃn ou âEvitaâ foi escrita e reescrita por historiadores e romancistas. AlÃm disso, muitos escritores se ofereceram como testemunha para contribuir na escrituraÃÃo desta histÃria, colaborando para a continuidade deste mito e na recriaÃÃo de sua imagem, sempre dialogando com sua autobiografia. La razÃn de mi vida permitiu a Eva PerÃn incrustar no Ãmbito discursivo sua autorrepresentaÃÃo como mulher de Estado. Nesta dissertaÃÃo, a obra Ã estudada na condiÃÃo de gÃnero autobiogrÃfico e se investigam as relaÃÃes histÃricas, literÃrias e ideolÃgicas que tal gÃnero pode estabelecer, a partir dos pressupostos teÃricos de Lejeune (1975), May (1979), Pouillon (1974), Bakhtin (2000), Rodrigues (2007) entre outros. Interessa-nos analisar como a escrita do eu se manifesta e se configura na obra atravÃs de uma estrutura composta por relatos de experiÃncia do cotidiano que acabam por revelar presente e passado, individual e coletivo. p>This essayâs main purpose was to make a study about the book La razÃn de mi vida (1951) of Eva PerÃn (The reason of my life), whose authorship or ghostwriter, was attributed to Manuel Penella da Silva, a Spanish writer that according to scholars about the myth of Eva following the example of the sociologist Horacio GonzÃlez (2009). The book that was published in Argentina is an autobiography that ratifies an image of a woman that became an icon of the Peronism of an almost messianic vision within the Latin context. The representation of Eva PerÃn also evokes some consideration of gender, in other words, it reveals the conscience of a woman that broke behavior patterns of an era, even managing to become the leader of a political movement. The political movement in question took place in Argentina in the 40âs when the country was under the command of the military. Since her in in July 26th 1952, the story of Eva PerÃn or âEvitaâ has been written and rewritten by historians and novelists. ▼ A novel fuzzy learning framework that employs fuzzy inference to solve the problem of multiple instance learning (MIL) is presented. The framework introduces a new class of fuzzy inference systems called Multiple Instance Fuzzy Inference Systems (MI-FIS). Fuzzy inference is a powerful modeling framework that can handle computing with knowledge uncertainty and measurement imprecision effectively. Fuzzy Inference performs a non-linear mapping from an input space to an output space by deriving conclusions from a set of fuzzy if-then rules and known facts. Rules can be identified from expert knowledge, or learned from data. In multiple instance problems, the training data is ambiguously labeled. Instances are grouped into bags, labels of bags are known but not those of individual instances. MIL deals with learning a classifier at the bag level. Over the years, many solutions to this problem have been proposed. However, no MIL formulation employing fuzzy inference exists in the literature. In this dissertation, we introduce multiple instance fuzzy logic that enables fuzzy reasoning with bags of instances. Accordingly, different multiple instance fuzzy inference styles are proposed. The Multiple Instance Mamdani style fuzzy inference (MI-Mamdani) extends the standard Mamdani style inference to compute with multiple instances. The Multiple Instance Sugeno style fuzzy inference (MI-Sugeno) is an extension of the standard Sugeno style inference to handle reasoning with multiple instances. In addition to the MI-FIS inference styles, one of the main contributions of this work is an adaptive neuro-fuzzy architecture designed to handle bags of instances as input and capable of learning from ambiguously labeled data. The proposed architecture, called Multiple Instance-ANFIS (MI-ANFIS), extends the standard Adaptive Neuro Fuzzy Inference System (ANFIS). We also propose different methods to identify and learn fuzzy if-then rules in the context of MIL. In particular, a novel learning algorithm for MI-ANFIS is derived. The learning is achieved by using the backpropagation algorithm to identify the premise parameters and consequent parameters of the network. The proposed framework is tested and validated using synthetic and benchmark datasets suitable for MIL problems. Additionally, we apply the proposed Multiple Instance Inference to the problem of region-based image categorization as well as to fuse the output of multiple discrimination algorithms for the purpose of landmine detection using Ground Penetrating Radar. För att inte de ohälsosamma levnadsvanorna ska ge framtida komplikationer är det viktigt att se till att barn och ungdomar får den hjälp de behöver för att motiveras till en beteendeförändring så tidigt som möjligt. Motiverande samtal (MI) är en samtalsmetod för att få individer motiverade till en förändring. Denna studie kommer att fokusera på just hälsosamma levnadsvanor. Olika hälso- och sjukvårds professioners använder motiverande samtal som metod för att stödja en beteendeförändring hos överviktiga ungdomar. Därutifrån har syftet till denna studie skapats vilket är att undersöka professionernas upplevelser av hur metoden fungerar på barn och ungdomar med övervikt. Studien har en kvalitativ insats som inkluderade intervjuer med sex stycken professioner. Intervjuerna spelades in och analyserades. Resultatet av studien visade att professionerna upplevde MI som en mycket positiv och användbar samtalsmetod och att kontinuitet behövdes för att upprätthålla kunskapen. Möjligheterna med att använda MI menar de professionella är att de ofta kan se en positiv förändring hos patientens beteende samt att de ser positivt på användningen av MI i framtiden. Författaren anser att det är viktigt att fortsätta utbilda personal som jobbar med människor inom MI då samhället troligtvis kan “spara” både pengar och tid hos personalen genom att använda MI i exempelvis primärvården. p> The unhealthy lifestyles such as poor diets and lack of physical activity can lead to overweight and obesity for children and adolescents today. To not have the unhealthy lifestyles lead to future complications it’s important to give them the help and support they need to get motivated to change theirs behavior. Motivational interviewing (MI) is a method to help people change their behavior, this study will focus on a healthy lifestyle changes. A profession in healthcare uses the MI as a method to support the lifestyle change for children and adolescents with obesity. The purpose of this study is to examine what the professions experiences is concerning how the method works with children and adolescents with overweight. This study has used a qualitative method with six interviews. The interviews were recorded and analyzed. The result of this study showed that professions experiences MI as a very positive and useful method, also that the professions needed to use the method continuity to maintain the knowledge. The potential of MI as a method was that the professions often could see a positive change in the patient and the respondent had a positive view in using MI in the future. The author of this study believes that it’s incredibly important to keep working with MI as it probably can "save" money both for society and the work hours for professions in healthcare. p>

▼ Antalet ensamkommande barn ökar, både i Sverige och i övriga Europa. Trots det finns det inga tydliga riktlinjer för hur arbetet med barn som fått permanent uppehållstillstånd ska se ut. Tidigare forskning tar upp resilience som en viktig aspekt i samband med ensamkommande barn. Teorin resilience handlar om förmågan att återhämta sig och att man når ett tillfredsställande resultat trots erfarenheter av situationer som innebär en risk att utveckla psykosociala problem. Vårt syfte med uppsatsen var därför att undersöka om personalens arbete leder till att öka barnens resilience. För att studera detta ville vi ta reda på vilka egenskaper hos barnen som personalen ser som viktiga för en sund utveckling och hur personalen stöttar dem att tillvarata sin inre styrka. Vi ville även studera hur personalen arbetar med att främja stödjande faktorer i barnens omgivning vilka kan öka deras resilience. Vi har gjort en kvalitativ studie där vi har intervjuat fyra respondenter som arbetar på boenden för ensamkommande barn. Resultatet visar på att personalen kontinuerligt diskuterar barnens styrkor och generellt ser de barnen som starka individer. På boendena arbetar man framförallt med att skapa en trygg miljö och att skapa ett förtroende mellan personalen och barnen. En god relation skapas genom att personalen finns till för barnet och anpassar sig till deras nivå samt utgår från varje individs personliga förutsättningar. Personalen hjälper barnen att reflektera och de vill att de ska bli självständiga och få kontroll över sina egna liv. p> The number of unaccompanied children is increasing, both in Sweden and elsewhere in Europe. Nevertheless there are no clear guidelines on how to work with children with permanent residence. Previous research is addressing resilience as an important aspect in working with unaccompanied children. The theory of resilience is about the ability to recover and to reach a satisfactory result despite experiences of situations involving a risk of developing psychosocial problems. The purpose of this paper was to investigate whether the personnel activities leads to strengthen children's resilience. To see this, we wanted to find out the capacities of the children which the personnel considers as important for their healthy development and how they are supporting them to attend to their inner strength. We also wanted to study how the personnel work to promote supportive factors in the environment for the children which may increase their resilience. We have conducted a qualitative study where we interviewed four respondents who work at shelters for unaccompanied children. The result indicates that the personnel continuously discuss the children’s strengths and generally sees the children as strong individuals. At the residence the personnel primarily works with creating a safe environment and to create a trust between themselves and the children. ▼ The 250 ka centred on the Oligocene/Miocene boundary (23.03 Ma) were witness to a transient glaciation of Antarctica known as the Mi-1 event. Although a number of deep-sea and Antarctic sedimentary records exist from this period and suggest some relationship between ice volume and obliquity, the rapid rate of change in Antarctic ice volume coupled with the relatively low resolution (~1,000–5,000 years) of the available records means that the details and drivers of this event are still unclear. There is little to no evidence from the Southern Hemisphere mid-latitudes, and thus little knowledge of how the glaciation affected the climate further afield. The Foulden Maar diatomite is a maar-lake deposit from the South Island of New Zealand. The deposit is finely laminated, and pollen evidence indicates a date in the early Miocene. I use the presence of orbital cycles within the physical properties records to show that individual pairs of light/dark laminae represent single years, and that the deposit covers ~100 ka. I then combine this information with 40Ar/39Ar dates from the base of the core and a magnetic reversal at ~106 m to show that the diatomite was being deposited during the period ~23.03–22.93 Ma, the peak and initial deglaciation phase of Mi-1. The density and reflectance of the Foulden Maar deposit covary with Antarctic ice volume as inferred from Ocean Drilling Program (ODP) cores. During periods of greater Antarctic ice volume, conditions were cooler and drier at 45°S. Tropical influence on the site can be inferred from the presence of a half-precession cycle and significant variability in the El Niño–Southern Oscillation (ENSO) band. The ENSO-band variability persists throughout the depositional period, indicating that the increasing warmth during the Mi-1 deglaciation does not result in permanent El Niño-like conditions. While the physical properties measurements imply increasing warmth throughout the depositional period, I also infer four millennial-scale cool reversals. The stomatal index data indicate a rapid increase in atmospheric carbon dioxide (pCO2) from <2x pre-industrial atmosopheric levels (PAL) to >3xPAL around 22.95 Ma, followed by a more gradual decrease back to around 2xPAL over the rest of the depositional period. This carbon dioxide pulse coincides with the initiation of rapid deglaciation in Antarctica, and suggests that pCO2 changes may have been connected to the deglaciation phase of Mi-1. I use a number of techniques and lines of evidence for both the age model and the cyclostratigraphic parts of this study. Where only one technique is possible, evidence to support my conclusions is drawn from other sources. However, some of the stomatal index evidence is not statistically reliable, and more data is required before firm conclusions can be drawn. A transient increase in pCO2 at ~22.95 Ma is correlated with the initiation of the deglaciation phase of Mi-1. ▼ There has been significant interest in the therapeutic potential of the adult stem/progenitor cells from bone marrow called multipotent mesenchymal stromal cells (MSCs). Signals from injured tissues activate MSCs to secrete beneficial factors and contribute to immune/inflammatory modulation and tissue healing. In order to enhance the therapeutic potential of MSCs hanging drop culture method was used to preactivate the cells in vitro and eliminate the lag period required for their activation in vivo. Stanniocalcin-1 (STC-1) is a potent anti-inflammatory and anti-apoptotic protein secreted from activated MSCs and has been considered as a substitute for MSCs in several disease conditions; therefore, the effects of STC-1 were studied on monocyte fate in vitro and in a mouse model of ischemic myocardial injury. Aggregated MSCs in hanging drops were self-activated to produce several therapeutic factors such as anti-inflammatory protein STC-1 and TNFα stimulated gene/protein 6 (TSG-6). MSCs dissociated from spheroids were also smaller than MSCs from standard 2 dimensional cultures, and as a result, larger numbers of them trafficked through the lung of mice after intravenous administration. Notably, spheroid MSCs were more effective than MSCs from standard cultures in suppressing inflammatory responses in a co-culture system with activated macrophages and in a mouse model of peritonitis. The data suggest enhanced therapeutic potential of spheroid MSCs for diseases caused by unresolved inflammation. Treatment with STC-1 reduced expression of CD14, a coreceptor for toll-like receptors, in differentiating monocytes/macrophages and suppressed the inflammatory responses of the cells to endotoxin. Administration of STC-1 also reduced CD14 expression in monocytes stimulated with various danger signals and in hearts of mice after myocardial infarction. These findings may explain the observed decreases in cardiac inflammation following myocardial infarction, and the improvements in ejection fraction and infarct size. The results suggest that STC-1 is a promising therapy to protect the heart and other tissues from ischemic injury.
