Le caribou montagnard des monts Torngat situé dans le nord du Québec et du Labrador connait présentement un déclin rapide. Cependant, la démographie, l’utilisation de l’espace et la sélection de l’habitat en relation avec le déclin de cette population sont méconnues. À l’aide de 35 caribous adultes munis de colliers satellites, nous avons évalué la sélection des habitats à deux échelles spatiales, la survie, et l’effet de la chasse ainsi que du chevauchement avec la population voisine de caribous migrateurs de la Rivière-George. Nous avons démontré que la sélection de l’habitat est similaire entre les deux échelles spatiales étudiées, mais variable selon les saisons. Nos résultats suggèrent que la sélection est déterminée par l’accessibilité et la disponibilité des ressources alimentaires en hiver et durant la pré mise-bas, l’évitement des insectes en été, et par la présence de plantes graminoïdes lors du rut. Bien que la prédation soit connue pour être le principal facteur limitant les populations et la sélection chez le caribou lors de la mise-bas, aucun comportement anti-prédateur n’a été détecté chez les femelles lors de cette saison. Nos résultats démontrent que le déclin est principalement attribuable à une faible survie des femelles adultes due à la chasse et à la prédation. Nous avons observé que l’utilisation de l’aire de distribution du caribou des monts Torngat par le troupeau Rivière-George a grandement diminué durant les 25 dernières années en raison de leur déclin. Ceci pourrait causer une diminution du flux génique et des possibilités d’immigration entre les deux troupeaux. Nos modèles démographiques suggèrent que le caribou des monts Torngat continuerait de décliner rapidement si aucune mesure de gestion n’était prise de façon à augmenter la survie des adultes.

The Torngat Mountains caribou herd in northeastern Québec-Labrador is rapidly declining. Few studies have examined the causes of this decline and little is known about the habitat selection, survival and space use of this herd. Using 35 caribou equipped with satellite collars between 2011 and 2015, we evaluated the Torngat Mountains caribou habitat selection at two different spatial and temporal scales, survival, and effect of harvest on future population trends. We also evaluated the Torngat Mountains caribou space use in relationship with its neighbor, the Rivière-George migratory caribou. We found little variation in habitat selection between the two spatial scales studied. However, there was variation in factors limiting habitat selection among different seasons. Habitat selection was primarily driven by resource availability and abundance during winter and pre-calving, by insect avoidance in summer and by the presence of graminoids during the rut. Although predation is known to be the main limiting factor of caribou populations and habitat selection by female caribou during calving, we found no evidence of predator avoidance behavior during this season. L’objectif de ce projet de recherche est de développer un modèle d’évaporation de l’eau d’une rivière en milieu boréal. Un site expérimental sur la rivière Natashquan dans la région de la Côte-Nord au Québec a été sélectionné pour y installer une station hydrométéorologique faisant un suivi des variables environnementales locales. Cette rivière est marquante dans le paysage de la Minganie par la superficie de 15 930 km2 de son bassin versant et son débit moyen annuel de 349 m3/s. Ce puissant cours d’eau est une des dernières rivières naturelles de la Côte-Nord n’étant pas harnachée par un barrage. Une campagne constituée de 5 sorties de mesures intensives sur le terrain a été effectuée afin d’observer l’évaporation de l’eau de la rivière par la méthode des mini-lysimètres flottants. Ces derniers s’inspirent du processus derrière le bac évaporatoire de classe A pour obtenir une estimation de l’évaporation in-situ en ayant un petit bac évaporatoire portatif à la surface de l’eau de la rivière. Un modèle d’évaporation par transfert de masse a été développé avec les données récoltées et 8 fonctions de vent de la littérature ont été testées. Puis, un modèle d’évaporation par ensemble de réseaux de neurones a été retenu, car il offrait une meilleure performance que les approches classiques. Ce réseau de neurones ayant comme variables d’entrée la tension de vapeur à saturation, le rayonnement incident d’ondes longues, la vitesse du vent et la tension de vapeur de l’air, permet d’explorer une non linéarité entre les variables qui n’est pas accessible à l’approche de transfert de masse. Ce modèle met en évidence le manque de recherche sur les processus gouvernant l’évaporation fluviale dans le cadre de rivière de grande envergure et particulièrement l’évaporation des rivières du milieu boréal.

The objective of this study was to develop a river evaporation model for the boreal biome. A hydrometeorological station was installed on the bank of the Natashquan river to collect environmental data. The Natashquan river is major hydrographic component of the region with its 15 930 km2 watershed and its 349 m3/s mean annual flow. This powerful watercourse is one of the only remaining significant river that has not yet been dammed in the Côte-Nord region. Five intensive field campaigns were carried out targeting different conditions during the summers of 2018 and 2019. River water evaporation was observed using the floating minipan method. This method, inspired by the process behind the class A evaporation pan, allows small portable evaporation pans to be deployed on the water surface and to monitor in-situ evaporation in more holistic conditions. Observations were used to calibrate the wind function of a mass-transfer evaporation model and compare it with different wind functions taken from the literature. A stacked neural network was selected as the river model based on a higher performance compared to the mass transfer approach. ▼ Les perturbations humaines sont en augmentation dans de nombreuses régions du monde et entraînent des conséquences graves sur les écosystèmes. Les régions nordiques et arctiques pourraient être particulièrement touchées par le développement anthropique à cause du fort potentiel en ressources naturelles et la faible résilience de ces écosystèmes. Le caribou migrateur (Rangifer tarandus) est au coeur de ces préoccupations puisqu’il constitue une espèce clé de cet écosystème. La majorité des troupeaux de caribous est toutefois en déclin et la nature généralisée et parfois synchrone de ces déclins suggère que des changements globaux, comme les changements climatiques et le développement anthropique, pourraient être responsables. Dans le nord du Québec et au Labrador, les caribous migrateurs des troupeaux Rivière-aux-Feuilles (TRAF) et Rivière-George (TRG) ne font pas exception à la tendance observée. Ces troupeaux ont subi une importante diminution au cours des dernières décennies. Simultanément à ces déclins, la région a connu une augmentation marquée des activités humaines, particulièrement dans le secteur minier. Toutefois, la contribution des perturbations humaines aux déclins de populations actuellement observés reste inconnue. Dans le cadre de cette thèse, je me suis intéressée à quantifier les effets des perturbations humaines sur l’utilisation de l’espace et la survie des caribous migrateurs du TRAF et TRG. Mes travaux sont divisés en quatre objectifs : 1) Évaluer les effets des perturbations humaines sur le comportement des caribous en évaluant l’impact des infrastructures linéaires sur les déplacements et en estimant les zones d’influence autour des infrastructures; 2) Évaluer la perte cumulée d’habitat associée à l’évitement des perturbations; 3) Évaluer l’effet des perturbations humaines sur la vulnérabilité des caribous à la chasse sportive; 4) Évaluer les effets des perturbations humaines sur le risque de mortalité des caribous et leur importance relative par rapport aux facteurs naturels; Mes travaux ont démontré que les caribous migrateurs répondent aux perturbations humaines à différentes échelles spatiotemporelles. À fine échelle, les caribous accéléraient leurs déplacements lors de la traversée d’une route. Une des routes sur l’aire de répartition du TRAF (route Raglan) semblait jouer un rôle de barrière aux mouvements des caribous. Les caribous évitaient les infrastructures sur des distances pouvant aller jusqu’à 23 km et l’évitement semblait exacerbé durant la période de chasse sportive. À plus large échelle, l’évitement des infrastructures s’est traduit en une perte cumulée d’habitat qui pouvait atteindre jusqu’à 30% des aires saisonnières et jusqu’à 37% des habitats fortement utilisés par les caribous. Mes travaux ont également démontré que la vulnérabilité des caribous à la chasse sportive était principalement affectée par les caractéristiques du paysage qui influencent la visibilité et l’accessibilité des chasseurs aux zones de chasse. ▼ This thesis presents the development of a software-based decoder design tool (DDT) for producing Ambisonic decoders optimised for playback over 5-speaker layouts. The research specifically focuses on developing decoders for irregular layouts with loudspeakers at a constant radial distance from the central listening position. It was motivated by the desire to provide better surround sound over the standard ITU 5-speaker layout for listeners in the sweet spot and off-centre positions. A wide-ranging literature review is presented revealing the need for such work. The DDT employs the Tabu Search algorithm to seek improved decoder parameters according to a multi-objective fitness function. The fitness function encapsulates criteria from psychoacoustic models as a set of objectives. In order to ensure the objectives were treated equally a method known as „range-removal‟ was used for the first time in Ambisonic decoder design. A companion technique termed „importance‟ allows the systematic prioritisation of range-removed objectives giving a designer control over desired decoder criteria. Additional elements exist in the DDT that can be turned on or off in different combinations. They include: a novel component for producing decoders with even performance by angle, a novel component for producing performance that correlates with the pattern of human spatial resolution estimated in previous Minimum Audible Angle experiments, and the ability to produce frequency dependent or independent decoders of different orders. Moreover, the user of the DDT can optimise performance for a single listener or multiple distributed listeners. To make the DDT as interactive as possible searches can optionally run on a High Performance Computer. This thesis also details the extensive testing of Ambisonic decoders for the ITU layout. Decoders have been assessed subjectively in listening tests and objectively using binaural measurements which has verified the methods developed in this research and the DDT‟s concept. Furthermore, decoders derived by the DDT have been compared to existing decoders and the results show they give equal or better performance. The development of a fully-functioning DDT which incorporates techniques for range-removal, importance, even performance by angle, minimum audible angle, off-centre listeners and their use in any combination represent the key outcomes of this work.

▼ This thesis offers a phenomenologically framed exploration of “computer-mediated” (Duignan, 2010) musical "creativities” (Burnard, 2012) and draws upon established creativity models to inform the development of a creativity framework that speaks specifically to the domain of computer-mediated musical practice. It is important to understand that the framework itself is not the focus of this thesis; rather, the aim of this thesis is to disclose transparently a method for developing a personalised understanding of my computer-mediated musical creativities as a template for other practitioners. Drawing from contemporary literature on creativity from a sociocultural perspective (Amabile, 1996), this study implements a phenomenological (Van Mannen, 2007) approach in order to capture a detailed impression of how my creativity is informed by concrete (computers, technology) and conceptual (socio-cultural background, cognitive processes) contextual resources. More specifically, this research focuses on “little c” (Craft, 2000) creativity, the skills possessed by all individuals as evidenced in learning and development literature (Craft, 2000; Arvaja, 2007; Burnard & Younker, 2010). This thesis offers a longitudinal case study of the sociological contexts of computer-mediated music creativities by focusing on myself as a subject for this work. Various types of data collection were collected, including: comprehensive written protocol (note-taking), audio excerpts of deconstructed musical materials; and audiovisual screen-capture of real-time creative practices. The creativity stages outlined by Sawyer in Explaining Creativity (2012) and Zig Zag (2013) were used as data sets for template analysis (King, 1998). Data that met these criteria were analysed and represented through structural mapping and analysis of the musical product, methods drawn from Collins (2005). The findings demonstrate that the chronology and direction of my creative focus between stages is intuitive and context dependent. It was also found that the influence of concrete and conceptual resources can be observed and understood through metacognitive investigation. This guided the development of a framework for the understanding of my computer-mediated musical creativity. This thesis encourages digitally informed musicians to think critically about their own creativity and guides the exploration of sociocultural context as a mediating resource and inspiration.

▼ This thesis presents and discusses research undertaken into the detection and measurement of colouration produced by audio transformers. It is common for transformers to be subjectively described by audio professionals as ‘warm’, ‘fat’, ‘smooth’, etc. however there is little evidence to show if there is an audible difference and if there is a correlation between these levels of perceptual attributes and the performance of the device. Therefore, the research question was defined as: is there an audible difference with a transformer? A review of the objective and subjective elements of the study was conducted. First the history, application and operation of the audio transformer with an aim to understand the objective measures and performance of the device with focus on the nonlinear distortion response. This also includes the design and testing of a suitable test circuit used in the measurement method. The subjective review concerns perception of distortion and testing methodologies for investigating audible differences. The conclusion of the review is that few pieces of research exist showing the relationship between device, distortion and perception. The testing of each transformer involved the use of a specially designed test circuit using a variety of measures including THD+N and frequency domain analysis, to provide the most information about the operation of the device. Using the same setup, the device responses to a variety of samples were recorded and implemented in a double-blind triple-stimulus with hidden reference test using trained listeners in accordance with the ITU-R BS.1116-3 recommendation. The test results were then analysed for a random distribution using a 1-tailed binomial test. The results of the analysis show a high likelihood that the bass samples were audibly different shown by the significant p-values of all 3 samples at less than 0.001. A slight correlation seems to exist with THD+N, 3rd harmonic distortion and level however with no other obvious trends, it was concluded that the distortion and therefore the audibility is programme dependent. It was concluded that transformers are likely to produce a level of distortion deemed audible although the effect is considered to be programme dependent.

Urbanistickým konceptem bylo vytvořit propojení příjemným způsobem s městem na východ od zadaného pozemku. Městské lázně jsem pojala jako opravdové lázně s lázeňskými procedurami – rašelinovými (slatinnými) koupelemi a zábaly; dále vodoléčbou; ale zároveň i jako místo pro rekreaci a relax. Tvar objektu hmotově naznačuje zkosené písmeno U; kde vnitřní prostor je veřejně přístupný. Recepcí je možné vejít do areálu lázní; nebo projít do veřejného prostoru náměstí s jednotlivými obchody v přízemí na jedné straně budovy a na druhé s prosklenou fasádou areálu lázní. Náměstí je klidné a nabízí možný průchod skrz celé území Starého Brna příjemným prostředím. Budova je dispozičně rozdělena na dvě části. Jedna je pronajímatelná pro obchody a služby. Tato funkce sahá přes dvě patra. Ve třetím patře jsou pronajímatelné plochy lékařských ordinací; rehabilitace a zázemí pro cvičení skupin; služby s funkcí zdraví a péče o tělo a mysl. Druhá část objektu je tvořena areálem lázní. Jediným vchodem z recepce; která obě části rozděluje; jsou vstupy do šaten; které tvoří filtr; jak pro kontrolu; tak i pro čistotu. Z šaten se dostaneme do otevřeného půdorysu s jednotlivými funkcemi (vodoléčba; slatinné koupele; sauna; pára a další); který sahá do stejné výšky jako dvě patra obchodů. Jsou zde totiž umístěny solné zdi a lehátka; která dohromady se solnými zdmi vytváří příjemná zákoutí k odpočinku. Prostor tak tvoří obří „solnou jeskyni“ pro kvalitní a příjemný vzduch. Ve druhém; technicky třetím podlaží areálu lázní najdeme whirlpooly; bazény; jak vnitřní; tak i venkovní bazén s terasou; masérny; pronajímatelnou saunu a páru; whirlpooly specifických funkcí; opět odpočívárny tvořené zákoutími lehátek a příčkami ze soli. V tomto podlaží je možné také vnitřní bazén se zázemím šaten plně uzavřít k soukromým akcím. Ve dvou podzemních podlažích se nachází parkování o celkové kapacitě 172 stání. Hlavní povrch tvoří černý a krémově bílý mramor. Na vnějších zdech je mramor leštěný; na vnitřních zdech v areálu lázní je matný. Tento luxusní materiál je doplněn krémovou keramickou velkoformátovou dlažbou v interiéru; velikými prosklenými plochami a příčkami z lámané soli; které dotváří specifickou atmosféru. Technologické rozvody pod stropy jsou zakryty sádrokartonovými podhledy s výjimkou parkovacích podlaží; kde jsou instalace vedeny volně pod stropem. Nosná obvodová stěna je navržena z vápenopískových bloků; systém jinak monolitický železobetonový skelet. Stejně tak vnitřní příčky jsou tvořeny vápenopískovými bloky s mramorovým obkladem. Okenní otvory jsou osazeny do hliníkových rámu Schueco v tmavé antracitové barvě. Hlavní schodiště v lázních je železobetonové.; Territory is located in the District of Pisárky. The only entrance on reception; which separates the two parts are inputs into changing rooms that make up the filter; both the control and purity. From changing rooms is an open floor plan with the individual features (hydrotherapy; peat bath; sauna; steam room and more); which goes back to the same height as the two floors of shops. There are indeed placed salt walls and sunbeds; which together with the salt walls creates a pleasant place to relax. Space is a giant "salty cave" for high quality and pleasant air. In the second; third floor; the baths of sophisticated features whirlpooly; swimming pools; both indoor and outdoor pool with terrace; masseurs; leaseable sauna and steam; whirlpooly-specific features; again rest rooms consisting of frills bunks and bars of salt. In this deck is also an indoor pool with changing rooms fully close the private actions. In two ground floors; parking on the total capacity of 172. The main surface is black and cream-white marble. On the external walls; marble polished; internal walls in a matte. This luxury material is accompanied by a large format perfect ceramics tile in the Interior; large glass surfaces and the bars of the smattering salt which musicians. Technological divorce under the ceilings are covered with pasterboard ceilings except parking deck; where installation is kept loose under the ceiling. A perimeter wall is designed from gange-sand blocks; otherwise a monolithic system consist of skeleton. As well as internal splitter are gange-sand blocks with marble coating. The window openings are fitted with aluminium frame Schueco in a dark metallic colour. The main staircase at the steel cocrete.

▼ L’étude porte sur les retombées économiques de la mine de Voisey’s Bay sur les conditions socioéconomiques des communautés de Rigolet, Makkovik, Hopedale, Nain et de Postville. Elle adopte une approche multidimensionnelle à la fois descriptive et comparative. Les données utilisées proviennent des recensements de la population de 1991, 1996, 2001 et de 2006 conduits par Statistique Canada. Elle utilise aussi les données de l’Enquête nationale auprès des ménages de 2011. Les données internes à la mine proviennent des Rapports de responsabilités sociales de la firme VBNC qui se charge d’exploiter la mine. Nous avons également en recours aux données des rapports du Gouvernement du Nunatsiavut depuis sa mise en place en 2005. Cette étude s’inscrit dans un contexte d’exploitation des ressources naturelles. Elle montre que, contrairement à des considérations voulant que celles-ci constituent une source de richesses, leurs effets sur les conditions socioéconomiques des communautés locales restent mitigés. Les firmes minières s’acharnent à maximiser leurs profits et les conditions de vie des communautés locales n’en bénéficient que partiellement. Pour notre étude, nous faisons l’hypothèse que cette mine ait eu de nombreuses retombées directes ou indirectes (emplois, achats de biens et services, taxes minières, investissements et redevances) et que celles-ci ont modifié les conditions de vie des communautés concernées. Nous avons opté pour une méthodologie basée sur un modèle de retombées économiques de la mine de Voisey’s Bay; un outil conçu avec l’inspiration de la grille d’évaluation des projets miniers, gaziers et pétroliers sur les conditions socioéconomiques de Paul Kishchuk. Ce modèle nous a permis d’analyser les effets de Voisey’s Bay sur la démographie, le marché du travail, la scolarité, le revenu et le logement. Les dimensions identifiées font état d’une légère amélioration sur les conditions de vie des communautés, bien que l’effet ne puisse pas être totalement attribué à la mine, étant donné le rôle important joué par le Gouvernement du Nunatsiavut depuis sa création. Este estudo avaliou o efeito de fontes e níveis de suplementação de selênio (Se) em dietas de aves reprodutoras pesadas sobre produção de ovo e a concentração de Se no conteúdo total do ovo. Foram utilizadas 50 reprodutoras Cobb 500 de 22 semanas, alojadas individualmente e alimentadas com dieta basal sem a suplementação de Se por três semanas. Após as aves receberam cinco tratamentos dietéticos com 10 repetições de uma ave cada, sendo compostos por selenito de sódio (inorgânico; Na2SeO3, 45% Se) e Zn-LSe- metionina (orgânico; ZnSeMet, 0,1% Se), como segue: T1- 0,15% inorgânico; T2- 0,30% inorgânico; T3- 0,15% orgânico; T4- 0,30% orgânico; T5- 0,15% inorgânico e 0,15% orgânico. Avaliações foram realizadas em dois períodos de 4 semanas. A produção de ovo e o peso do ovo foram diariamente registrados e a gravidade específica foi medida duas vezes por semana até o final do estudo. No primeiro período os animais alimentados com 0,30 ppm de Se orgânico apresentaram maior percentual de produção de ovo (P < 0,05), sendo que, no segundo período, nenhuma diferença foi observada entre os tratamentos (P > 0,05). As avaliações entre os períodos apresentaram que o peso do ovo não foi diferente (P > 0,05), sendo que a gravidade específica foi menor no segundo período (P < 0,05) e a concentração de Se no ovo aumentou com o tempo de suplementação (P < 0,05), independentemente da fonte de Se suplementada. A comparação dos tratamentos que continham apenas uma única fonte de Se demonstrou que a concentração de Se no ovo foi maior com a suplementação do maior nível de ZnSeMet (P < 0,05). Entretanto, a suplementação com a combinação das fontes (Na2SeO3 e ZnSeMet) produziu similares concentrações de Se no ovo. O aumentou da dose de selenito de sódio de 0,15 para 0,30 ppm não foi acompanhado por um aumento da deposição de Se no ovo.

This study evaluated effects of sources and levels of Se in broiler breeder diets on egg production and Se concentration in eggs. Fifty Cobb 500 hens 22 wks of age were individually placed in steel cages and fed a basal diet without Se supplementation for 3 wks. Birds were then provided five dietary treatments with 10 replicates of one individual hen, which had dietary selenium supplied from sodium selenite (inorganic; Na2SeO3, 45% Se) and/or from Zn-LSe- methionine (organic; ZnSeMet, 0.1% Se) as follow: T1- 0.15% Se from Na2SeO3; T2- 0.30% Se from Na2SeO3; T3- 0.15% Se from ZnSeMet; T4- 0.3ç0% Se from ZnSeMet; T5- 0.15% Se from Na2SeO3 + 0.15% Se from ZnSeMet. Evaluations were conducted in two periods of 4 wks each. Experimental diets were prepared through the supplementation of corn-soybean meal diets. Egg production and egg weight were recorded daily, whereas specific gravity was measured twice a wk towards the end of the study. A partir de températures de surface mesurées sur une base régulière et répétitive par des capteurs embarqués sur des satellites, il a été décidé de produire des cartes de températures de surface, d’indices de gel et de dégel et de limites de répartition du pergélisol sur les territoires arctiques et subarctiques. Le territoire cartographié est le territoire du Québec-Labrador. Les températures mesurées par satellite ont été comparées avec les températures de différentes stations situées en région de pergélisol continu, en Alaska et au Québec (chapitre 1). Les coefficients de corrélations sur l’ensemble des stations entre les températures de l’air et les « Land Surface Temperatures » (LST) du capteur MODIS des satellites Aqua et Terra sont tous supérieurs à 0.80. Ces mesures ont donc un extraordinaire potentiel pour déterminer les températures de surface sur le vaste territoire nordique où les stations de mesures sont très dispersées dans l’espace. Comme le territoire couvert par les satellites est de 2300 km à chaque passage (ils passent deux fois par jour), la possibilité de couvrir des étendues régionales à peu de frais est immense. Cependant, les régions arctiques connaissent un ennuagement assez important et les données satellitaires ne sont pas continues sur l’année. Leur nombre est tout de même suffisant pour établir une interpolation temporelle sur chaque pixel. Au chapitre 2, il est montré que le modèle mathématique utilisé permet de calculer les températures moyennes annuelles et les indices de gel à partir des LST très bien corrélées, et les indices de dégel corrélés de façon satisfaisante avec ceux calculés aux stations. A partir de ces comparaisons entre les pixels et les stations présentes dans un pixel, les cartes ont été produites. L’analyse de ces cartes, moyenne annuelle, indice de gel et de dégel montre des similitudes dans les régions où les données existent mais surtout établissent une représentation originale sur les parties de territoire où aucune station de mesures n’existe et apportent un élément nouveau à la compréhension du climat du Québec-Labrador. Au chapitre 3, les indices habituels qui servent à délimiter le pergélisol sur des échelles régionales sont appliqués à ce territoire. Là encore, il est démontré que la limite sud du pergélisol continu ainsi dessinée est comparable à celle des cartes existantes. Toutefois, mesurées entre 2000 et 2005, les conditions thermiques propres à cette limite ont récemment migré vers le nord. Le modèle mis au point dans cette thèse est une avancée exceptionnelle pour la cartographie du pergélisol.

Infecções causadas por tripanossomatídeos causam milhares de mortes anualmente, além de levarem a redução da capacidade produtiva, com elevada morbidade na população acometida. A eficácia terapêutica é limitada na maioria dos casos, sendo o benzonidazol o único fármaco aprovado para uso do tratamento do Trypanosoma cruzi, sendo ativo somente na fase aguda da Doença de Chagas, com grande efeitos colaterais. No caso das leishmanioses, as substâncias de tratamento existentes causam toxicidade renal e cardíaca, além de induzirem a resistência e apresentarem eficácia insuficiente. Observando todo o contexto torna-se necessário a busca por novas substâncias que sejam mais eficazes e menos tóxicas. Assim, o trabalho representa uma contribuição para a busca de novas moléculas bioativas para o tratamento da doença de Chagas e leishmaniose a partir da padronização e realização de ensaios celulares usando a forma epimastigota da cepa Y do T. cruzi e promastigota para Leishmania chagasi e L. amazonensis. A padronização do método colorimétrico de MTT foi realizada a partir da comparação com o método tradicional de contagem por microscopia usando câmara de Neubauer, seguido do estudo de viabilidade por citometria de fluxo, como método confirmatório e, finalmente a padronização do ensaio de ciclo celular usando os fármacos de referência benzonidazol (T. cruzi) e anfotericina B (Leishmania spp.). Os compostos testados neste estudo são proveniente de classes distintas de inibidores de alvos macromoleculares (cisteíno proteases, DHODH, GAPDH, quinases) e foram planejados e/ou selecionados pelo Grupo de Química Medicinal (NEQUIMED). Dentre todas as substâncias, inibidores de quinases apresentaram maior potencial para estudos subsequentes, sendo o T. cruzi o parasito mais sensível, onde um grande número de substâncias apresentou atividade nos estudos de triagem. Para uma delas (Neq0474) foi realizado o ensaio de dose-resposta, com EC50 = 53 µmol/L. L. chagasi apresentou a maior resistência dentre todos os parasitos estudados neste trabalho, enquanto que L. amazonensis foi sensível para Neq0438. Algumas substâncias estudadas apresentaram potencial para estudos mais aprofundados visando identificar novas alternativas terapêuticas para estas doenças parasitárias.

Infections caused by trypanosomatides lead to thousands of deaths annually, besides the reduction of the quality of life and working capability, with high morbidity to the patients. The therapy efficacy is limited in most cases, being benznidazole the only approved drug in Brazil, which works only in the acute phase of the Chagas Disease with severe collateral effects. For leishmaniasis, the drugs cause renal and cardiac toxicity and trigger resistance. In this context, novel efficacious and secure substantes are necessary to improve the current therapeutic strategies, which was the goal of this project by means of cell-based assays.
