▼ Greyback canegrubs cost the Australian sugarcane industry around $13 million per annum in damage and control. A novel and cost effective biocontrol bacterium could play an important role in the integrated pest management program currently in place to reduce damage and control associated costs. During the course of this project, terminal restriction fragment length polymorphism (TRFLP), 16-S rDNA cloning, suppressive subtractive hybridisation (SSH) and entomopathogen-specific PCR screening were used to investigate the little studied canegrub-associated microflora in an attempt to discover novel pathogens from putatively-diseased specimens. Microflora associated with these soil-dwelling insects was found to be both highly diverse and divergent between individual specimens. Dominant members detected in live specimens were predominantly from taxa of known insect symbionts while dominant sequences amplified from dead grubs were homologous to putativelysaprophytic bacteria and bacteria able to grow during refrigeration. A number of entomopathogenic bacteria were identified such as Photorhabdus luminescens and Pseudomonas fluorescens. Dead canegrubs prior to decomposition need to be analysed if these bacteria are to be isolated. Novel strategies to enrich putative pathogen-associated sequences (SSH and PCR screening) were shown to be promising approaches for pathogen discovery and the investigation of canegrubsassociated microflora. However, due to inter- and intra-grub-associated community diversity, dead grub decomposition and PCR-specific methodological limitations (PCR bias, primer specificity, BLAST database restrictions, 16-S gene copy number and heterogeneity), recommendations have been made to improve the efficiency of such techniques. Improved specimen collection procedures and utilisation of emerging high-throughput sequencing technologies may be required to examine these complex communities in more detail. This is the first study to perform a whole-grub analysis and comparison of greyback canegrub-associated microbial communities. This work also describes the development of a novel V3-PCR based SSH technique. This was the first SSH technique to use V3-PCR products as a starting material and specifically compare bacterial species present in a complex community.

▼ ENGLISH SUMMARY : Crops expressing genes from the bacterium Bacillus Thuringiensis (Bt) produce a protein toxic to members of the order Lepidoptera and are a popular alternative to sprayed insecticides. Although these Bt crops are considered to be an effective pest control method, careless usage may add selective pressure on the pest population to develop resistance to the protein over time. One method of limiting the rate of resistance development is to keep small portions of the cultivated land planted with the non-GMO crop which then acts as a refuge area for susceptible pests, limiting its exposure to the protein. Varieties of Bt sugarcane for the South African market that should limit the damage caused by the stalk borer moth, Eldana Saccharina Walker, are being developed, and a prerequisite to releasing such a product is to manage resistance development. Refuge areas are the primary way of managing resistance, but a recommendation is required on the size and layout of the refuge areas to be planted, as an area too small may not curb the rate of resistance development enough, but an area too large may not be economically viable for the industry. An agent-based simulation model is presented where individual moths are modelled as agents on an underlying sugarcane field that can either be Bt or refuge. The spatial aspect of the simulation model allows for testing the effectiveness of different shapes of refuge and not just the size. To reduce the complexity of the model, the field is divided into a series of smaller cells that can interact with each other, which allows us to model the impact of a severe infestation on an area the size of an average sugarcane farm. The simulation divides the simulated space into two grids representing the layers of the sugarcane that the different life stages of the insect move in. The first layer is the ground layer, representing the lower portions of the sugarcane where the eggs, larvae, pupae and egg-laying female moths are found. These cells are smaller as the immature life stages are nearly stationary and the female moth is assumed to have a much lower dispersal rate when searching for sights to lay eggs. The second layer is the sky layer with larger cells, where adult males and unmated females are able to move over larger distances to find mates. To further reduce complexity, every cluster of eggs laid by a female moth is modelled as a single agent that progresses as a unit through the immature life stages before being converted to many individual moth agents when they mature. The model is then applied to three hypothetical case studies, each focussing on a specific aspect of refuge planning. The first case study focusses on the size and distribution of refuge, the second on the shape of the refuge, and the third assumes that the Bt sugarcane is introduced gradually and how this may affect the refuge strategy. ▼ ENGLISH ABSTRACT: An agent-based simulation model is formulated in this dissertation in order to simulate the population dynamics of Eldana saccharina Walker infestation in sugarcane. The simulation model comprises four distinct building blocks, namely a graphical user interface, the implementation of the life cycle and associated influence of temperature on E. saccharina, the mating process of E. saccharina and the oviposition and dispersal of E. saccharina. These building blocks are based on existing literature pertaining to the biology and behaviour of the pest and, in cases where the relevant literature is insu cient or non-existent, expert opinion and careful assumption. In order to select areas from existing sugarcane farms on which to execute location-speci c experiments, functionality which allows Geographic Information Systems (GIS) data importation is included as a platform on which to run the simulation model. These data inform the model in respect of the shape and characteristics of the underlying sugarcane elds in which the simulated E. saccharina population interacts. The model interacts and operates within an AnyLogic simulation software environment and, in so doing, aims to emulate the behaviour of a population of E. saccharina moths in sugarcane. It is anticipated that the model implementation may serve as a basis for facilitating future design and testing of control measures in order to suppress the pest and its consequent detrimental e ect to sugarcane through infestation and feeding on interior stalk nutrients. Numerous working mathematical models of the pest exist in the literature, but, in all previous cases, intricate aspects of the stalk borer's biology have been aggregated on a population level and average population changes have been a ected at discrete time steps. The resulting analyses therefore yield conclusions that do not necessarily re ect the continuous, changing nature of E. saccharina on a localised level. Using agent-based modelling, however, the pest's behaviour may be modelled in more detail so as to facilitate more thorough investigation of potential control strategies and their expected e cacy on the pest at di erent points in its life cycle. The agent-based simulation model designed in this dissertation is subjected to a number of veri cation and validation techniques. Furthermore, a pilot sensitivity analysis is conducted to identify the most in uential parameters in the simulation model. These parameters are then considered further in a comprehensive parameter variation analysis in order to illustrate the exibility and diversity of the model in terms of the variety of scenarios pertaining to E. saccha- rina population behaviour that it can accommodate. ▼ Rattus sordidus (canefield rat) is a major pest in north Queensland sugarcane growing areas, causing losses of between 2 and 4 million each year. A further 0.2 to 0.4 million is expended on annual baiting campaigns. This procedure provides only short-term results, is seldom cost-effective and is hazardous to non-target species and the environment. There was a close relationship between the dynamics of R. sordidus populations and the crop cycle. During the 1987/88 crop cycle, breeding occurred over the January- July period and coincided with the period when summergrasses were abundant in crops and the predominant food source of R. sordidus. The relationship between the onset of breeding, the first appearance of summergrasses and diet of R. sordidus indicates that breeding is dependent on the availability of a non-cane food source. The level of in-crop weed biomass may therefore play an important role in the regulation of population and damage levels throughout a district. Consequently, a number of agricultural practices which affect in-crop weed levels may have a significant effect on R. sordidus populations. The practice of green cane harvest and trash-blanketing has increased in most northern sugarcane growing districts since the early 1980's. This practice differs to the conventional technique in that crops are not burnt prior to harvest and following harvest the trash, comprising the green "tops" and dry leaf material, is left as a "trash-blanket" on the ground and minimum or zero tillage techniques employed. Suppression of weed germination and growth is a major agronomic advantage associated with this technique. In-crop weed control may also be achieved through the application of herbicides. The monetary benefit associated with yield increases as a result of reduced weed competition in young crops has lead to an increase in the number of crops treated with herbicide. This study investigated the effect of green cane harvesting and trashblanketing, the conventional agricultural practice and herbicide applications on the dynamics of R. sordidus populations during the 1987/88 crop cycle.

ENGLISH ABSTRACT: Eldana saccharina Walker (Lepidoptera: Pyralidae) is a stem borer and food crop pest of economic importance. Temperature and moisture availability possibly influence E. saccharina distribution and abundance, however, the thermal biology and desiccation physiology of E. saccharina are not fully understood. Furthermore, physiological adaptation probably facilitates the invasion success of E. saccharina into novel environments and this too remains unstudied. Here, the thermal- and desiccation-trait variation of E. saccharina were studied and population responses were modelled. The results of this work provided insights into novel physiological outcomes of E. saccharina that is coupled with its environmental climatic stress resistance, overwintering ability and population fitness in general. In determining thermal limits to activity and survival of E. saccharina results showed that chill coma onset temperature (CTmin) and critical maximum temperature (CTmax) of E. saccharina moths collected from sugarcane (Saccharum spp. hybrids) were significantly lower than those from Cyperus papyrus L. (CTmin = 2.8 ± 0.4 vs. 3.9 ± 0.4 °C; CTmax = 44.6 ± 0.1 vs. 44.9 ± 0.2 °C, P < 0.0001 in both cases). These results holds important implications for habitat management (or ‘push-pull’) strategies in the sense that host plant may strongly mediate lower critical thermal limits. Results for pronounced variation in adult CTmin (± 4 °C) across the geographic range of E. saccharina in South Africa was found and it was significantly positively correlated with the climatic mean minimum temperature. Slower developmental time in the most low-temperature tolerant population suggests lower CTmin adaptation has come at a cost to fitness, but allows greater survival and activity in that environment. There are a significant reduction of phenotypic plasticity in the laboratory population and a strong genetic component to CTmin trait variation. Physiological acclimation within a single generation, during immature life stages, resulted in altered adult water balance physiology to enhance fitness. Results from a biophysical population model showed that over-wintering life stage and climate significantly affected the number of E. saccharina generations, predicted stress, relative moth fitness and relative adult abundance. Larval overwintering led to less generations and more frequent cold- and heat stress at a cold field site compared to a warm one. This in turn reflected on the relative adult fitness and –abundance. Larval presence predictions overlapped well with positive scout records averaged across a matrix of sugarcane ages and cultivars. The results from this work are important on which to base integrated pest management strategies and are applicable to a large audience across agricultural landscapes and in the sugarcane industry of South Africa.

▼ South Africa is the leading producer of sugarcane in Africa and one of the largest sugarcane producers in the world. Sugarcane is grown under a wide range of climatic, agronomic, and socio-economic conditions in the country. Stress factors such as water and nutrient deficiencies, and insect pests and diseases are among the most important factors affecting sugarcane production in the country. Monitoring of stress in sugarcane is therefore essential for assessing the consequences on yield and for taking action of their mitigation. The prediction of sugarcane yield, on the other hand is also a significant practice for making informed decisions for effective and sound crop planning and management efforts regarding e.g., milling schedules, marketing, pricing, and cash flows. In South Africa, the detection of stress factors such as nitrogen (N) deficiency and sugarcane thrips (Fulmekiola serrata Kobus) damage and infestation are made using traditional direct methods whereby leaf samples are collected from sugarcane fields and the appropriate laboratory analysis is then performed. These methods are regarded as being time-consuming, labour-intensive, costly, and can be biased as often they are not uniformly applied across sugarcane growing areas in the country. In this regard, the development of systematically organised geo-and time-referenced accurate methods that can detect sugarcane stress factors and predict yields are required. Remote sensing offers near-real-time, potentially inexpensive, quick and repetitive data that could be used for sugarcane monitoring. Processing techniques of such data have recently witnessed more development leading to more effective extraction of information. In this study the aim was to explore the potential use of remote sensing to quantify stress in and predict yield of sugarcane in South Africa. In the first part of this study, the potential use of hyperspectral remote sensing (i.e. with information on many, very fine, contiguous spectral bands) in estimating sugarcane leaf N concentration was examined. The results showed that sugarcane leaf N can be predicted at high accuracy using spectral data collected using a handheld spectroradiometer (ASD) under controlled laboratory and natural field conditions. These positive results prompted the need to test the use of canopy level hyperspectral data in predicting sugarcane leaf N concentration. Using narrow NDVI-based vegetation indices calculated from Hyperion data, sugarcane leaf N concentration could reliably be estimated. In the second part of this study, the focus was on whether leaf level hyperspectral data could detect sugarcane thrips damage and predict the incidence of the insect. The results indicated that specific wavelengths located in the visible region of the electromagnetic spectrum have the highest possibility of detecting sugarcane thrips damage. Thrips counts could also adequately be predicted for younger sugarcane crops (4–5 months). ENGLISH ABSTRACT: Contrary to expectations, Leuconostoc mesenteroides was not the major gum-producing bacterium isolated from sugarcane. Instead, high cell numbers of W. confusa and W. cibaria were recorded. The dominance of Weissella spp. on the prepared cane is significant because these bacteria are not usually associated with deteriorated sugarcane and have not previously been isolated from a sugarcane processing factory. This study also report, for the first time, on the isolation of B. amyloliquefaciens from a sugarcane processing factory. The second aim of the study was to evaluate the efficacy of biocides to prevent/limit microbial growth in the factory. Two dithiocarbamate biocides, Busan®1021 and Preventol®Z, each at a 20 ppm dose, were tested against seven gum-producing bacteria. Preventol®Z demonstrated a bactericidal (killing) effect against Le. mesenteroides A16-9, Le. lactis B9-3, B. subtilis B7-19 and B. amyloliquefaciens B7-51 after 6 h of contact, but had only a bacteriostatic (growth inhibiting) effect on W. cibaria A1-17, W. confusa B1-24 and Lb. fermentum B19-18 when tested under the same conditions. Busan®1021 had a bactericidal effect on all seven species. Bacillus subtilis B7-19 and B. amyloliquefaciens B7-51 were susceptible to both biocides, but only for the first 2 h of exposure, after which the killing effect remained constant. Based on results obtained in this study, the concentrations of Preventol®Z and Busan®1021 may need to be increased, or dosage intervals altered, to kill all gum-producing bacteria. Although sugarcane processing factories have little to no control over the quality of cane entering the factory, poor sanitation in the factory and incorrect process control can also contribute to sucrose loss due to microbial activities and subsequent gum formation. Factories should therefore be mindful of correctly controlling high-temperature processes and reduce the recirculation of sump contents which are not treated with biocides. This study provided valuable knowledge on the identities of gum-producing bacteria in sugarcane processing factories, and their susceptibility to two commercial biocides.

No Brasil, o bicudo da cana-de-açúcar, Sphenophorus levis Vaurie (Coleoptera: Curculionidae) tem se tornado nos últimos anos praga importante da cultura, principalmente no estado de São Paulo onde as perdas alcançam 30 ton/ha/ano e no Sul do estado de Minas Gerais. Isto devido à ausência de medidas preventivas para a disseminação e ao crescimento da área plantada, decorrente da alta demanda de álcool. Existem diversos métodos para o seu controle e um dos mais empregados são as iscas tóxicas; apenas impregnadas com o inseticida carbaril não existindo alternativas, o que pode favorecer a aparecimento de populações resistentes do inseto, dificultando o controle. Sendo assim, é evidente a necessidade de investigar outros inseticidas tão ou mais eficientes visando aumentar inclusive a eficiência das iscas utilizadas atualmente através da incorporação de substâncias mais atraentes. O presente trabalho foi realizado no Departamento de Entomologia, Fitopatologia e Zoologia Agrícola da Escola Superior de Agricultura \"Luiz de Queiroz\", Universidade de São Paulo, e teve como objetivos avaliar a atração de adultos de S. levis por meio de olfatômetro \"Yä toletes de cana-de-açúcar com e sem melaço a 10% e casca de abacaxi, em diferentes períodos de fermentação (24, 48 e 72hs), horários (diurno e noturno) e sexos (machos e fêmeas) e estabelecer a relação entre os compostos emitidos pelos substratos, acetato de etila e etanol, por meio de cromatografia gasosa, com a atração dos insetos a cada substrato testado; assim como avaliar o efeito letal dos inseticidas cartap e tiametoxam em diferentes dosagens fornecidas através de iscas de cana-de-açúcar no laboratório é validá-los em campo, comparando-os com as capturas das iscas contendo carbaril. Os resultados do laboratório demonstraram que a mistura cana-de-açúcar com melaço fermentado por 24 horas, atraiu os insetos em até 90% e que a resposta dos insetos foi maior no horário diurno, sem diferenças estatísticas entre os sexos. Os inseticidas cartap e tiametoxam nas dosagens 2,0 g p.c/L de água e 0,6 gp.c/L de água respectivamente, ocasionaram níveis de mortalidade superiores a 80% no laboratório. No campo a mortalidade em iscas contendo cartap (2,0 g p.c/L) foi menor que as iscas contendo carbaril e tiametoxam (0,6 g p.c/L) as quais tiveram resultados semelhantes. O maior número de insetos capturados nas iscas tóxicas foi registrado aos 7 dias, nos dias subseqüentes as capturas diminuíram.

In Brazil, the sugarcane weevil, Sphenophorus levis Vaurie (Coleoptera: Curculionidae), has become a serious pest of sugarcane in recent years mainly in the states of São Paulo and south of Minas Gerais where losses caused by this pest surpasses 30 ton/ha/year. ▼ The interactions between insect herbivores and their host plants have fascinated scientists for generations. There is a vast diversity of these insects, which have a variety of different feeding strategies and diet breadth. Many have become significant pests of managed ecosystems, such as forest and crop production systems. The majority of research has focussed on aboveground insect defoliators rather than root feeding insects. This is surprising as root feeding insects are some of the most economically damaging and difficult to control. Several scarab species (Coleoptera: Scarabaeidae) are among the most significant insect pests to agriculture, particularly during the root feeding larval stages. The soil environment in which scarab larvae feed and develop involves several abiotic and biotic factors which can be highly influential in shaping the relationship between these root feeders and their host plants. Soil nutrients including nitrogen (N), phosphorus (P) and potassium (K), along with soil moisture, are important to plant growth and quality, and thereby are important to the belowground insects which feed on their roots. Silicon (Si) is known to impact plant growth, but can also act as a plant defence mechanism against insect herbivores, although this remains untested in root feeding insects. Biotic soil influences include arbuscular mycorrhizal (AM) fungi, which associate with the majority of land plants, and can alter plant quality and defences, while negatively impacting root herbivore performance, although the mechanisms remain unclear. This work addresses how these different factors within the soil environment affect root feeding scarab larvae. This is initially investigated at the community level, then subsequently using the grass crop plant sugarcane, Saccharum species hybrids, and the canegrub, Dermolepida albohirtum. Recent literature on the ecology of scarab larval pests within Australasia and the effects of different soil factors on scarab larvae are synthesised in chapters one and two, respectively. These chapters highlight the paucity of knowledge surrounding the ecology of root feeding scarab larvae and the importance of factors such as soil nutrients (N,P,K and Si) and microbial communities (AM fungi) on plant-herbivore interactions belowground. Chapter three investigates how fertilisation and irrigation practices, which alter soil nutrients (N,P,K) and moisture, impact host plant communities, scarab larval communities and their natural enemies (entomopathogenic nematodes). Scarab larval communities were positively affected by fertilisation, increasing their abundance by 52%. While irrigation did not impact scarab communities, there was an increase in entomopathogenic nematode presence by 78%, suggesting scarab populations were suppressed by their natural enemies. ▼ In the KwaZulu-Natal (KZN) Midlands North region of South Africa, the importance and increased prevalence of endemic scarabaeids, particularly Hypopholis sommeri Burmeister and Schizonycha affinis Boheman (Coleoptera: Melolonthinae), as soil pests of sugarcane, and a need for their control was established. The development of a mycoinsecticide offers an environmentally friendly alternative to chemical insecticides. The identification of a diversity of white grub species, in two Scarabaeidae subfamilies, representing seven genera were collected in sugarcane as a pest complex. Hypopholis sommeri and S. affinis were the most prevalent species. The increased seasonal abundances, diversity and highly aggregated nature of these scarabaeid species in summer months, suggested that targeting and control strategies for these pests should be considered in this season. Increased rainfall, relative humidity and soil temperatures were linked to the increased occurrence of scarab adults and neonate grubs. Beauveria brongniartii (Saccardo) Petch epizootics were recorded at two sites in the KZN Midlands North on H. sommeri. Seventeen different fluorescently-labelled microsatellite PCR primers were used to target 78 isolates of Beauveria sp. DNA. Microsatellite data resolved two distinct clusters of Beauveria isolates which represented the Beauveria bassiana senso stricto (Balsamo) Vuillemin and B. brongniartii species groups. These groupings were supported by two gene regions, the nuclear ribosomal Internal Transcribed Spacer (ITS) and the nuclear B locus (Bloc) gene of which 23 exemplar Beauveria isolates were represented and sequenced. When microsatellite data were analysed, 26 haplotypes among 58 isolates of B. brongniartii were distinguished. Relatively low levels of genetic diversity were detected in B. brongniartii and isolates were shown to be closely related. There was no genetic differentiation between the two sites, Harden Heights and Canema in the KZN Midlands North. High gene flow from swarming H. sommeri beetles is the proposed mechanism for this lack of genetic differentiation between populations. Microsatellite analyses also showed that B. brongniartii conidia were being cycled from arboreal to subterranean habitats in the environment by H. sommeri beetles. This was the first record of this species of fungus causing epizootics on the larvae and adults of H. sommeri in South Africa. The virulence of 21 isolates of Beauveria brongniartii and two isolates of B. bassiana were evaluated against the adults and larvae of S. affinis and the adults of H. sommeri and Tenebrio molitor Linnaeus (Coleoptera: Tenebrionidae). Despite being closely-related, B. brongniartii isolates varied significantly in their virulence towards different hosts and highlighted the host specific nature of B. brongniartii towards S. affinis when compared to B. bassiana. A broca da cana-de-açúcar, Diatraea saccharalis (Fabricius, 1794) (Lepidoptera: Crambidae) é naturalmente infectada por diversos patógenos, sendo os mais importantes Metarhizium anisopliae (Metsch.) (Ascomycota: Clavicipitaceae), Beauveria bassiana (Bals.) (Ascomycota: Cordycipitaceae) e vírus da granulose. Esses microrganismos podem co-infectar um mesmo indivíduo em condições de campo, mas os resultados das interações desses patógenos não são conhecidos. O presente trabalho objetivou determinar a virulência desses patógenos aplicados de forma isolada e em infecções mistas na fase larval de D. saccharalis em condições de laboratório sobre dieta artificial e em plantas de milho. A maioria das combinações resultou em efeito aditivo na mortalidade dos insetos (M. anisopliae + B. bassiana, M. anisopliae + DsGV e M. anisopliae + B. bassiana + DsGV), entretanto ficou evidenciado antagonismo entre B. bassiana e o granulovírus. Todos os cadáveres oriundos das aplicações associadas apresentaram sintomas de apenas um dos patógenos envolvidos na infecção. A produção de conídios de uma espécie de fungo nos cadáveres submetidos à co-infecção foi semelhante à produção de conídios da mesma espécie na infecção isolada. Na infecção mista, os dois fungos se desenvolveram na hemolinfa do hospedeiro, entretanto em um estágio tardio do processo infectivo ocorreu a exclusão de um dos patógenos. A infecção por B. bassiana diminuiu drasticamente a densidade de hemócitos circulantes na hemolinfa das lagartas, efeito não observado para M. anisopliae. Duas populações de laboratório de D. saccharalis, uma oriunda de Piracicaba-SP e outra de Araras-SP, foram testadas quanto à suscetibilidade aos fungos M. anisopliae e B. bassiana. A população de insetos de Piracicaba foi menos suscetível em 2009 do que a mesma população em 2008 e também em relação à população de Araras. Dessa forma, foi realizado um bioensaio comparando a suscetibilidade de insetos das duas populações e da progênie (F1) dos cruzamentos diretos entre elas, criadas nas mesmas condições bióticas e abióticas, para determinar se a suscetibilidade diferencial estaria relacionada a fatores genéticos. Não foram observadas diferenças significativas na suscetibilidade das duas populações e dos cruzamentos aos fungos. A suscetibilidade diferencial observada nos dois primeiros bioensaios provavelmente esteja relacionada às variações no sistema de criação, em componentes nutricionais e antimicrobianos da dieta e não a variações entre as populações dos hospedeiros.

Esta nova tendência de colheita irá resultar em mudanças nas relações tróficas presentes no canavial. A broca da cana (Diatraea saccharalis) é considerada uma pragachave da cultura e é responsável por uma perda significante de produtividade. O controle biológico da broca é feito através da liberação do parasitóide Cotesia flavipes. Este método de controle aplicado está sujeito a interrupções que comprometem seu sucesso. A broca tem nos dermápteros um agente predador, de ocorrência natural nos canaviais. Muito pouco se sabe sobre estes insetos. Informações sobre os aspectos bioecológicos destes predadores são fundamentais para a sua avaliação como potenciais agentes alternativos de controle da broca da cana, fortalecendo os programas de controle biológico adotados nos canaviais atualmente. Esta pesquisa tem como objetivo avaliar alguns aspectos biológicos e analisar aspectos comportamentais de dermápteros presentes em canaviais da região de Piracicaba. Foram identificadas quatro espécies de tesourinhas: Euborellia janeirensis, Euborellia peregrina, Cosmiella brasiliensis e Doru luteipes. Devido ao baixo número de fêmeas de C. brasiliensis as variáveis: número de posturas, ovos por postura, período embrionário, período de cuidado maternal e porcentagem de eclosão desta espécie, não integraram a análise estatística. Todas as espécies apresentaram dimorfismo sexual nos cercos e cuidado maternal com ovos e ninfas. Em relação a duração do período de cuidado maternal, E. peregrina e E. janeirensis obtiveram os maiores valores, 24,83 e 24,81 respectivamente. D. luteipes diferiu estatisticamente exibindo cuidado maternal por 14,6 dias. Não houve diferença estatística entre as espécies E. janeirensis e E. peregrina quanto o período embrionário (12,06 e 11,83 dias), porém D. luteipes diferiu estatisticamente apresentando duração de apenas 7 dias. Não houve diferença estatística entre as três espécies no que diz respeito a porcentagem de eclosão, sendo que o maior valor foi registrado para E. janeirensis (77,4%), seguida de D. luteipes (76,48%) e E. peregrina (69,65%). Quando comparadas as médias do número de ovos por postura, as três espécies diferem estatisticamente sendo o maior valor registrado em E. janeirensis (46,33) seguido por E. peregrina (34,16) e D. luteipes (27,60). A duração das fases ninfal e adulta foram analisadas para as quatro espécies. A maior duração de fase ninfal foi registrada em E. janeirensis (51,95 dias) e em E. peregrina (50,70 dias). C. brasiliensis diferiu estatisticamente das duas espécies do gênero Euborellia, apresentando duração de 33,65 dias. A menor duração foi registrada em D. luteipes (24,10 dias), diferindo estatisticamente de todas as espécies. A duração da fase adulta foi maior em D. luteipes (219,65 dias), E. peregrina (212,5 dias) e E. janeirensis (210,30 dias). Apenas C. brasiliensis diferiu estatisticamente (183,50 dias). ▼ A necessidade de economias sustentáveis tem aumentado o interesse no desenvolvimento de plataformas microbianas para novos processos, tanto para a produção de biocombustíveis quanto para a síntese de compostos que demandam alta capacidade energética e processamento químico em sua produção. O isolamento de microrganismos, capazes de degradação materiais lignocelulósicos, resistentes a diferentes inibidores e com rendimento elevado na biossíntese de moléculas específicas faz-se necessário para atender tais propósitos. Neste trabalho, microrganismos cultiváveis isolados do trato digestivo de larvas dos insetos pragas de cana-de-açúcar, besouro da raiz, Migdolus fryanus, bicudo da cana, Sphenophorus levis, broca-gigante da cana, Telchin licus licus e de broca-da-cana, Diatraea saccharalis, foram caracterizados quanto a atividade enzimática em fontes de carbono e identificados por técnicas moleculares. Larvas no terceiro ínstar de M. fryanus e S. levis e quinto ínstar de T. licus licus e D. saccharalis foram coletadas em plantios de cana-de-açúcar no interior de São Paulo. Um total de 341 microrganismos cultiváveis foram avaliados quanto a capacidade de degradação de substratos lignocelulósicos, como única fonte de carbono em meio sólido usando o índice de atividade enzimática (I.E.). Os isolados foram identificados por sequênciamento das regiões do 16S rRNA para bactérias, ITS rDNA para fungos e 26S rDNA para leveduras. Bactérias com atividade enzimática foram relacionadas aos gêneros do filo Firmicutes Bacillus, Enterobacter Serratia e Citrobacter. Os isolados relacionados à B. amyloliquefaciens destacaram-se na degradação da celulose. Entre fungos filamentosos, leveduras e leveduriformes, a atividade enzimática foi destacada para a degradação da hemicelulose. Fungos filamentosos e leveduras pertencem ao filo Ascomycota e os leveduriformes ao filo Chlorophyta. Fungos que apresentaram hidrólise de xilano foram relacionados aos gêneros Pyrenophora, Aspergillus e Penicillium. As leveduras relacionam-se a nove gêneros, majoitariamente aos gêneros Meryerozyma e Candida. Leveduras com capacidade hidrolítica destacada foram relacionadas à Meyerozyma guilliermondii, Cryptococcus laurentti, Candida pseudointermedia, C. parapsilosis, C. solani e Aureobasidium pullulans. Entre as leveduras, 40 isolados apresentam sequências que diferem em mais de 1% das sequências referência, podendo-se inferir a respeito de possíveis novas espécies. No ensaio cinético, realizado para a triagem de leveduras com capacidade de bioconversão da D-xilose em xilitol e etanol, o maior rendimento em xilitol foi demonstrado pelo microrganismo relacionado à Prototheca zopfi var. hydrocarbonea. A capacidade desse organismo de produzir xilitol não havia sido descrita. ▼ The fungal disease smut (causal agent: Ustilago scitaminea H. & P. Sydow) and stalk borer Eldana saccharina Walker place major constraints on sugarcane agriculture in South Africa. The best approach for management is the introduction of resistant cultivars; however, conventional field-based screening for pest and disease resistance is a lengthy process. This study evaluated in vitro techniques combined with artificial inoculation of 12 week old in vitro plantlets and 8-10 week old embryogenic calli as rapid screening methods. Preliminary investigations were conducted on cultivars with known field ratings to smut and E. saccharina: NCo376, N26 and N39; and 5 „test‟ cultivars, whose identities were undisclosed until completion of experiments, were used to assess the accuracy of protocols. Infective U. scitaminea sporidia generated from teliospores, were used as inocula. Development of a callus protocol was unsuccessful due to sporidial and mycelial overgrowth, despite addition of a contact fungicide, Dithane M-45® (0.025 g/l) and a biocide/fungicide, PPMTM (5 ml/l), to media. Plantlet inoculation by injection, 1 cm above the apical meristem, resulted in 12% and 20% of smut susceptible NCo376 plantlets producing smut whips after 5 weeks when inoculated with 1 x 106 and 1 x 109 sporidia/ml, respectively. Smut whip production in 5 of the 8 (63%) cultivars inoculated with the lower sporidial concentration correlated with their field resistance ratings. In addition, whips harvested from in vitro plantlets were a valuable source of aseptic teliospores for future research. Ongoing work involves inoculation of NCo376 calli with such teliospores and maintenance on medium with PPMTM - emergence of whips from plantlets remains to be assessed. The E. saccharina screening protocol involved surface decontamination of eggs with 1% sodium hypochlorite (NaOCl) for 15 min. Feeding bioassays were conducted by placement of first instar larvae on in vitro plantlets and calli for 3 and 2 weeks, respectively. Larval mass, length and percentage infestation were recorded. Although greater larval size was expected in susceptible compared with resistant cultivars, the results did not support this. Significant differences in plantlet infestation were observed between susceptible (94-98%) and resistant (72-86%) lines. No significant differences were found in the callus feeding bioassay. However, a 24 h callus choice bioassay which investigated larval preference between callus genotypes compared with NCo376, showed significant differences and correctly discerned cultivar susceptibility according to field ratings. A cana-de-açúcar é uma das mais importantes culturas no Brasil. No entanto, muitas pragas atacam esta cultura causando prejuízos econômicos. O gorgulho da cana-deaçúcar, Sphenophorus levis, é uma importante praga na cultura canavieira no Estado de São Paulo. Para completar o seu ciclo de vida, S. levis parece depender de microorganismos que habitam o seu trato intestinal e desempenham um importante função na fisiologia e nutrição do inseto. Neste estudo, nós realizamos a caracterização da microbiota intestinal de população de larvas de inseto campo e de laboratório. As análises das seqüências de 16S rDNA revelaram um total de catorze gêneros, um grupo da categoria Candidatus e dois grupos nãocultiváveis representados pelos filos Alfa-Proteobacteria, Beta-Proteobacteria, Gamma- Proteobacteria, Firmicutes e Bacteroidetes tanto em larvas do campo quanto nas do laboratório. Os microrganismos isolados através dos métodos dependentes de cultura foram agrupados de acordo com parâmetros morfológicos e através do marcador moléculas 16S rDNA. Além das bactérias também foram isolados quatro fungos filamentosos. Observou-se uma diversidade bacteriana levemente superior no campo do que no laboratório de acordo com o índice de Shannon (Campo H = 3,36; Laboratório H = 3,26). É também nosso objetivo neste trabalho encontrar microorganismos capazes de degradar celulose, um importante evento no ataque do inseto. Dos microorganismos cultiváveis, cinco gêneros de bactérias e dois fungos filamentosos apresentaram atividade celulolítica. Este é o primeiro estudo sobre a microbiota do S. levis que pode contribuir para a compreensão da interação planta-patógeno e também ser útil para o futuro desenvolvimento de novas estratégias de controle de S. levis no cultivo da cana-de-açúcar.

The sugarcane weevil, Sphenophorus levis, is an important pest in sugarcane culture in São Paulo state, Brazil. To complete its life cycle, S. levis may depends on microorganisms that inhabit its intestinal tract and play an important key in the insect physiology and nutrition. In this study we report the characterization of the intestinal microbiota from population of insect larvae from field and laboratory. Analysis of 16S rDNA sequences revealed a total of fourteen genera, one group from Candidatus category and two uncultivable groups represented by Alfa-Proteobacteria, Beta-Proteobacteria, Gamma-Proteobacteria, Firmicutes and Bacteroidetes phylum. Microorganisms isolated through culture-dependent methods were classified according morphological parameters and using 16S rDNA molecular marker. In addition to bacteria, four filamentous fungi were isolated. It was observed a slightly higher bacterial diversity in field than in laboratory according to Shannon index (Field H = 3,36; Laboratory H = 3,26). ▼ O Brasil é o maior produtor mundial de cana-de-açúcar. Embora essa cultura seja um exemplo onde se emprega o controle biológico de pragas, especificamente de Diatraea saccharalis, o uso de inseticidas químicos tem sido necessário no controle de outros insetos. Mato Grosso do Sul é um dos Estados que apresentam os maiores aumentos de área cultivada de cana-de-açúcar no país, com plantio previsto, inclusive, em extensas áreas adjacentes ao Bioma Pantanal. O estudo com substâncias derivadas de plantas com potencial no controle de pragas justifica-se pela necessidade de compostos menos agressivos ao meio ambiente. Extratos do cajuzinho-do-cerrado, Anacardium humile, têm mostrado potencial como inseticida sobre alguns insetos. A cigarrinha-das-raízes, Mahanarva fimbriolata (Hemiptera: Cercopidae) é considerada praga importante nessa cultura, ocasionando danos severos e sensível redução na produção. Portanto, o objetivo deste trabalho foi avaliar o efeito do óleo e dos extratos aquoso, etanólico e hexânico de A. humile, nas concentrações 0,05, 0,4 e 1,0%, no controle de ninfas de M. fimbriolata, em cana-de-açúcar. O experimento foi constituído por dez repetições de cada tratamento, sendo cada uma composta por um pote de plástico, com tampa vazada para encaixe do copo com a muda, cujo fundo foi recortado de modo a expor as raízes. Após a aplicação dos produtos nessas raízes, cada muda foi infestada com seis ninfas recém eclodidas. As avaliações foram feitas aos 5, 10, 15 e 20 dias após a aplicação dos tratamentos e os parâmetros avaliados foram: mortalidade ninfal, período ninfal, longevidade dos machos e fêmeas, peso seco dos adultos, fecundidade e viabilidade dos ovos. A ação do óleo e extratos de A. humile resultou em baixos percentuais de mortalidade de M. fimbriolata. A maior mortalidade ninfal registrada foi de 53,1% na concentração 1,0% do extrato aquoso, valor superior aquele verificado na testemunha negativa. O prolongamento do período ninfal, com exceção do extrato aquoso, foi constatado nos demais compostos e/ou concentrações testadas. A longevidade das fêmeas foi alterada no tratamento com extrato hexânico nas três concentrações. A longevidade e o peso dos machos de M. fimbriolata, nos diferentes tratamentos, não diferiram da testemunha. O óleo e demais extratos de A. humile determinaram malformações em M. fimbriolata.
