▼ This dissertation describes the application and development of molecular tools that have great potential for use in studying variation in strawberry germplasm. The first study evaluated 91 microsatellite (simple sequence repeat, SSR) markers for transferability in 22 Fragaria species and their utility in fingerprinting octoploid strawberries. Out of the transferable markers, a reduced set of four SSRs was developed based on polymorphism, multiplexing ability, reproducibility and ease of scoring. Unique SSR fingerprints were obtained for over 175 Fragaria samples representing 22 Fragaria species used in the study. Testing of two molecular markers linked to the red stele and anthracnose resistances identified potential sources of resistance in previously untested genotypes. Further characterization of these accessions is warranted to validate resistance and usefulness in breeding. In the second study, 20 SSRs polymorphic in wild Asian diploids, F. iinumae and F. nipponica, from Hokkaido, Japan were selected for genetic analysis of 137 accessions from 22 locations. Principal coordinate analysis followed by non-parametric modal clustering grouped accessions into two groups representing the two species. Further clustering within the groups resulted in 10 groups (7-F. iinumae, 3-F. nipponica) that suggest lineages representative of the diversity in Hokkaido, Japan. The third study tested plant DNA barcodes, the nuclear ribosomal Internal Transcribed and the chloroplast PsbA-trnH spacers, for Fragaria species identification. The 'barcoding gap', between within species and between species variation, was absent and prevented identification of Fragaria species. The fourth study evaluated the genetic diversity of 94 accessions representing 22 Fragaria species using four universal chloroplast SSR loci. Genetic diversity was moderate (0.54) despite the homoplasy observed. Species-specific haplotypes for F. nipponica, F. orientalis, F. iinumae and F. nilgerrensis were identified. Sequencing whole chloroplast genomes using Illumina in a final study revealed a close maternal genome relationship between F. vesca ssp. bracteata and the octoploid species supporting a North American origin of the octoploids and the polyphyly of F. vesca. Calculation of divergence time of Fragaria revealed young evolutionary age of the genus at 2.7 million years and of the octoploids at 450,000 years. Advisors/Committee Members: Bassil, Nahla V. ▼ Two studies were initiated in June of 1981 in order to gain greater understanding of the growth and fruiting habits of field grown strawberries in Oregon's Willamette Valley. Yield component analysis of 'Benton', Oregon's leading cultivar and an advanced breeding selection, 'OR-US 4356' was conducted during the first (1981) and second (1982) fruiting years. In each season, the two genotypes produced essentially the same yields/plant, averaging 0.82 kg in 1981 and 0.94 kg in 1982. 'Benton' produced 25-30% more crowns/plant than 'OR-US 4356'. 'Benton' had fewed trusses/crown in 1981 (0.93 vs. 1.47) but in 1982 their numbers were the same (1.5). 'OR-US 4356' had about 60% higher numbers of fruit/truss in each season. However, 'Benton' averaged 75% greater mean berry weight than 'OR-US 4356'. There was no significant difference in total achenes/berry between genotypes (202 in 1981 and 239 in 1982). Larger fruit had a greater number of achenes and linear regressions of berry weight versus achenes/berry were significant different between genotypes in each season. 'Benton' had less achenes/gram fresh weight of berry (24 vs. 34) and achenes/cm2 (10.6 vs. 14.0) than 'OR-US 4356'. This indicates that 'OR-US 4356' fell short of its yield potential due to its inability to reach maximum berry expansion. Frequent sampling and functional growth analysis were used to quantify growth of field grown strawberry plants ('OR-US 4681') during the summer of establishment and through flowering and fruiting the next spring. Plant dry weight and leaf area increased rapidly during mid-summer, but thereafter gains slowed and finally ceased in October. This was reflected in the absolute growth rate (AGR) which peaked at 1 g dry matter/day near September 1, then fell to zero by early October. Over this period, weekly mean temperature, solar radiation, and daylength decreased by 37%, 47% and 35% respectively. Relative growth rate (RGR) declined steadily from a high of 44 mg/g/day at the end of June. Unit leaf rate (ULR) showed a similar pattern, starting at 9 g/m2/day. During the following April through June, both plant dry weight and leaf area increased at exponential rates. The growth environment improved during this period and the AGR increased continuously but RGR remained constant at 19 mg/g/day. The rate of dry matter accumulation in fruit was exponential, whereas that in leaf lamina and stems (crowns plus petioles) showed linear increases. Because of fruiting, a much smaller proportion of dry matter was partitioned to leaves than during plant establishment. However, ULR rose from 5.5 to 6.5 g/m2/day, indicating that dry matter production was relatively constant during flower and fruit production.

▼ The purpose of this thesis was to evaluate two methods of estimating 1953 costs of producing strawberries. To accomplish this the following steps were taken: 1. To estimate production costs in 1953 by using 1947 input data and 1953 prices for those inputs, (Method 1); 2. To compare the results of the preceding method with the index number method of revising the cost data, (Method 2); To evaluate the two methods of revising the cost data. By the use of Method 1 the production cost per pound was 16.4 based on an average yield of 39609 pounds per acre. This amounted to an average total cost of $590.30 per acre. Considerable variation existed among the 53 producers whose records were used, with the higher yielding producers tending to have lower costs. When the index number method was used, the cost was 16.5 cents per pound. This is only 0.1 cents more than costs revised with the actual prices paid for the inputs. Because these costs are nearly identical, no significant difference appears to exist between the results of the two methods. However, it must be noted that this comparison is for only one crop and one crop year. Different results may be obtained for different crops and for different years. 3. The following conclusions were drawn with respect to the two methods: 1. Method 1 is based on prices paid to produce the crop. This is in contrast to costs revised with indices which include items not actually used in the production of strawberries. 2. With Method 1 only prices prevailing in the production area are used. On the other hand, costs revised with index numbers are affected by state and/or national prices for commodities which may or may not be used to produce strawberries. 3. More computations are necessary to revise the costs in Method 1 than Method 2. Costs revised by Method 1 require about 25 computations to revise each producer's data plus about 20 more to determine the sample average. By contrast, only six computations are necessary to revise cost data with index numbers. 4. By using the principles of both methods it may be possible to retain some of the advantages of both methods. A cost index based on local prices would be more realistic than more general indices and would retain the simplicity of the index number method now used.

▼ Size and color are important visual quantity attributes of the fruit of strawberry (Fmgaria x ananassa Duch.). Strawberry cultivars produce fruit of a characteristic size and color, the later being the result of accumulation of particular anthocyanins. This research sought to determine the basis for genotypic differences in fruit size and to monitoring changes in the amount of anthocyanins and related phenols, and the activity of associated enzymes during strawberry fruit development. Fruit size (weight and volume), number cells per fruit, and mean cell size were determined throughout the development of secondary fruit of the day-neutral strawberry cultivars Tillikum, Tristar, and Selva. Fruit size among the cultivars showed a four fold range and resulted from differences in the number of cells per fruit. The number of cells per mature fruit was 0.72, 1.96, and 2.94 x 106 for `Tillikum', `Tristar', and `Selva', respectively. The relative difference in cell number among cultivars was established by anthesis. In all cultivars, mean size of fruit cells was similar (z 6 x 106 μm3 at maturity) and cell division continued for about 12 days after anthesis. Young green fruit of `Tillikum' contained high levels of soluble phenols, composed mostly of tannins (;u 44%) and flavonoids (=51%). The concentration of phenols rapidly decreased as fruit developed, but the amount per fruit slowly increased. The concentration of anthocyanins was low throughout most of fruit development, but at ripening showed an 18-fold increase in 3 days. The activity of phenylalanine ammonia-lyase (PAL), an initial and key regulatory enzyme in the biosynthesis of phenolics, peaked in green fruit and again during ripening. The second rise in activity paralleled the rapid accumulation of anthocyanins. The activity of PAL was low during the white berry stage, when fruit growth was rapid. A UDP-glucose:flavonoid glucosyltransferase was isolated and purified about 100 fold from ripening Tillikum' fruit. The soluble enzyme showed a pH optimum of 7.5 and a molecular weight of about 55,000 Da. The Km for UDP-glucose was 143 μM and for the flavonol, quercetin, 4.5 μM. The enzyme had a broad substrate specificity and the isoflavone, biochanin A, was the best substrate among the flavonoid aglycones examined. ▼ Mechanisms by which plant organs accumulate photoassimilate are important in determining final sink size and may involve invertase activity. Because strawberry fruit are strong sinks for sugars, changes in invertase levels during development and mechanisms of sugar uptake within the fruit were examined. Fruit from greenhouse-grown, day-neutral strawberries, (Fragaria X ananassa Duch. cv Brighton), were used for all studies. Fruit invertase levels were analyzed from 0 to 2 5 days post anthesis (DPA). Soluble and insoluble acid invertases were found, both with optimal activity at pH 4.6. Soluble invertase activity was highest at anthesis and decreased during development. Insoluble invertase activity was low at anthesis, but increased by 5 DPA where it remained for the following 20 days. Insoluble invertase was solubilized in 1 M NaCl and is presumably ionically bound to the cell wall. At 15 DPA, histochemical staining showed invertase activity (mostly insoluble) throughout the tissue, but denser around vascular bundles; cortex tissue stained predominantly in the cell walls. Location of invertase near vascular bundles suggests a function in phloem unloading. Sugar uptake kinetics of cortex protoplasts from 16-18 DPA fruit were similar to those found previously in tissue disks. Glucose uptake was higher than sucrose or fructose and exhibited simple saturation kinetics. Sucrose and fructose had similar uptake rates with biphasic curves; saturable kinetics was observed at low sugar levels (below 10 mM) and first-order (linear) kinetics at higher levels. Apoplastic sucrose, glucose and fructose concentrations were estimated to be 80, 80, and 120 mM, respectively. Higher fructose levels may result from sucrose hydrolysis by cell wall invertase followed by more rapid uptake of glucose. The high apoplastic sugar concentrations indicate that the linear component of sucrose and fructose uptake is important in sugar uptake in vivo. No competition was seen among the three sugars, suggesting separate uptake pathways. 1'-Fluorosucrose, a sucrose analog, was not hydrolyzed by cell wall invertase and showed uptake kinetics similar to sucrose in cortex tissue. This, coupled with a lack of competition in sucrose uptake and demonstration of sucrose uptake by protoplasts, suggests that hydrolysis of sucrose by cell wall invertase is not a prerequisite for its uptake in strawberry fruit. ▼ Recent research shows that flower parts of strawberry (Fragaria X ananassa Duch.) differ in their capacity to support the growth of Botrytis cinerea, the fungus which causes gray mold on fruit. Growth in styles is slow and hyphae rarely enter the carpel (achene), suggesting that this pathway is not the predominant mode for fruit infection. However, the fungus is observed to grow to the distal end of stamens and into subtending calyx and receptacle tissue. Since gray mold is frequently first observed at the calyx end of the fruit, fungal entry via stamens may be a significant source of fruit infection. Therefore, genotypic variation of susceptibility of strawberry stamens to B. cinerea was surveyed. Newly open flowers of 42 strawberry cultivars were inoculated with dry conidia of B. cinema in the greenhouse and/or field, and stamens collected after 14, 21 and 28 days. Length of filament, colonization and sporulation on anthers, filament length colonized, and the proportion of filaments with hyphae present in their base were evaluated by UV fluorescence microscopy after staining stamens in aniline blue. Conidia germinated on anthers and hyphae grew into the anthers and filaments, colonization of which was more rapid and extensive in the field than greenhouse. Sporulation occurred on anthers in a majority of genotypes in the field, but was not observed in the greenhouse. The fungus grew to the base of stamens in some genotypes within 14 days in the field, whereas in the greenhouse this was not observed until 21 days. Filaments of over half the genotypes in the field were colonized to greater than 80% of their length in 28 days. Infection of filaments of non-inoculated controls by R cinema did not occur in the greenhouse but was common in the field, and here hyphae reached the base of at least one filament in control flowers of 11 of 30 genotypes. Genotypes varied in both the area of anther and filament length colonized by the fungus during the 4 week period following inoculation. Less than 2% of the anther surface of greenhouse-grown 'Capron' {Fragaria moschata Duch.) was colonized and no hyphae were observed to enter the filaments. Conversely, in the cultivars Podnyaya Zagorya', 'Elsanta', 'Tangi', 'Primella', and 'Senga Sengana', colonization of the length of filaments in the field varied from 80% to 100% and anther colonization ranged from 51% to 70%. Of the 33 genotypes evaluated in the field and greenhouse, these five genotypes ranked as the most susceptible in both environments, whereas 'Marlate' and 'Massey' showed relatively low susceptibility. Some genotypes exhibited low susceptibility in the greenhouse but not in the field, and the correlation between percent filament length colonized by genotypes in the two environments was low. Genotypic differences in susceptibility of stamens to attack by B. cinema suggest that stamen tissues of some genotypes are able to inhibit the advanced of the fungus. ▼ Two experiments were conducted from 1987 to 1990 to study the effect of date of renovation on strawberry yield and yield components. In the first experiment, 'Benton' plants were renovated from July 6, 1988 (1 week after harvest - WAH) to August 24, 1988 (8 WAH). In the second experiment 'Benton', 'Totem', and 'Redcrest' were studied. Treatments were date of renovation from July 12, 1989 (2 WAH) to August 16, 1989 (7 WAH), and a control (un-renovated). Date of renovation affected the number of runners per plant in fall, 1988 and 1989. The number of runners showed either a negative linear relationship ('Totem', 1989) or a quadratic relationship ('Benton', 1988 and 1989; 'Redcrest' 1989) with date of renovation — early-renovated plants (3 WAH or early) had the greatest number of runners. Thus, early-renovation may be more important in a matted row system than in a hill system. In the fall, 1988, early renovated 'Benton' plants (3 WAH or earlier) had a greater number of trusses and flowers per plant. The following summer the pattern was reversed; late-renovated plants (4 to 8 WAH) produced a higher yield. There was a positive linear relationship between date of renovation and the number of trusses and flowers per plant in 'Benton' in the summer of 1989. In the second experiment, 'Benton', 'Totem' and 'Redcrest' responded differently to date of renovation in the fall of 1989 as well as in the summer of 1990. There was very little flowering of 'Totem' in fall, 1989. In 'Benton', early-renovated plants (up to 6 WAH) did not differ significantly in number of trusses and flowers per plant compared to the control. However, early-renovated plants had a greater number of trusses and flowers per plant than late-renovated plants in the fall of 1989. In 'Redcrest', a negative linear relationship was present between date of renovation and the number of trusses and flowers per plant the fall of 1989. In summer, 1990, there was no significant difference between treatments and the control in number of trusses, number of flowers, leaf area (LA) per plant, plant fresh weight (PFW), and yield per plant in 'Totem'. Response of 'Redcrest' plants in summer, 1990 to date of renovation in 1989 was similar to 'Totem' expect that PFW differed between the treatments and control groups. Early-renovation (2 to 5 WAH) led to higher LA per plant. In 'Benton', plants renovated from 3 to 5 WAH had more trusses, flowers and LA per plant, and more PFW than late renovated plants (6 and 7 WAH). Un-renovated control plants tended to have the highest LA and PFW but did not differ significantly from early-renovated plants (2 to 5 WAH). The number of trusses and flowers on control plants was significantly lower than on early-renovated plants (2 to 5 WAH). ▼ The effects of manganese nutrition on growth, mineral composition, carbohydrate and nitrogen metabolism, IAA oxidase and phenols of 'Northwest' strawberry plants are reported. Growth and mineral composition response were investigated in sand culture and Hoagland's solution 2 modified to contain 0.0, 0.5, 2.5, 12.5, 25.0, 62.5 and 125.0 ppm substrate manganese. The remaining aspects of the study were conducted using the Hoaglands solution in a hydroponics system with manganese levels of 0.0, 0.5, 2.5, 12.5 and 25.0 ppm in the nutrient solution. In sand culture, symptoms of manganese toxicity occurred first in old leaves after three weeks of 125.0 ppm Mn treatment; after five weeks at 62.5 ppm; and after 13 weeks at 25.0 ppm substrate Mn. Toxicity symptoms appeared as necrotic spots along the margins of leaves; these spots enlarged greatly within a few days and ultimately killed all the leaves. All the plants treated with 62.5 and 125.0 ppm Mn died within 7 and 11 weeks, respectively. Young leaves of runners from the plants treated with 0.0 ppm Mn developed interveinal chlorosis and cupping after 12 weeks. Later, these leaves developed chlorotic spots which expanded to chlorotic bands. Plant weight and total leaf area of the plants harvested after 20 weeks was greatest with 2.5 ppm Mn treatment, and decreased both at lower or higher substrate manganese. The most leaves and runners were produced at 25.0 ppm treatment; whereas area per leaf was highest at 0.5 ppm Mn. Manganese present in tissue was directly correlated to substrate manganese levels and was also related to the plant part. Accumulation of manganese occurred in all tissues at high substrate Mn levels, indicating that the tolerance of strawberry plants to high Mn is not because of low uptake but due to their ability to endure large amounts of accumulated Mn. In Mn deficient plants, the differences in the concentration of Mn in the various tissues were small, suggesting the possibility of redistribution of the element under conditions of deficiency. Highest levels of tissue iron were found in 0.0 ppm Mn treatment followed in order by 2.5, 0.5, 12.5 and 25.0 ppm substrate Mn. A strong inverse relationship was noted between the tissue iron/ manganese ratio and the substrate Mn. The deficiency and excess of Mn resulted in a decrease in the concentration of Mg and P in the leaves. Highest levels of these elements were observed with 2.5 ppm Mn treatment. The most vigorous plants were produced at 2.5 ppm Mn treatment when old and young leaves contained 955.7 and 617.4 ppm Mn, respectively. Typical toxicity symptoms were shown by plants when Mn in young leaves reached about 4,000 and in old leaves 9,000 ppm. In the hydroponic system, manganese deficiency and toxicity resulted in a decrease in sugar as well as starch content in the leaves, except in old leaves at 25.0 ppm Mn treatment which contained the highest levels of starch. ▼ Three rates of urea, urea-DCD, and NH₄N0₃ were soil applied in April and August 1986 to strawberry selection OR-US 4930 and cultivar 'Sumas' to determine effects of the nitrification inhibitor dicyandiamide (DCD) and nitrogen source and rate on soil NH₄ and N0₃ levels, fruit yield and quality, and mineral composition of foliage and fruit. There were four replications of zero nitrogen plots. For both OR-US 4930 and 'Sumas', soil NH₄ levels reflected the seasonal mineralization pattern rather than a fertilizer source or rate pattern. After the August nitrogen treatments soil N0₃ was lower with urea-DCD than with urea. Urea-DCD altered mineral uptake by the plant, but did not affect total marketable yield or quality of OR-US 4930 or 'Sumas'. Petiole NO3-N was significantly (p < .05) increased by increasing N rate in "Sumas' in March 1987. Petiole N0₃-N of 'Sumas' and OR-US 4930 in June 1986 was significantly (p < .05) affected by an interaction of N rate and source. OR-US 4930 petiole N0₃-N at the middle N rate of urea was greater than at the same rate of urea-DCD. At the high N rate there were significant differences among all three sources. 'Sumas' petiole N0₃-N at the high N rate of urea was greater than the same rate of NH₄N0₃. OR-US 4930 leaf N content in August 1986 and fruit N content in 1987 were significantly (p < .05) increased by increasing nitrogen. 'Sumas' leaf N content in June and November 1986 and in March 1987, fruit N content and petiole N0₃-N in March 1987 were significantly (p < .05) increased by increasing N rate. Leaf N content of OR-US 4930 in June 1986 was significantly (p < .05) greater with urea than with urea- DCD. Fruit N content was significantly (p < .05) lower with urea-DCD than with NH₄N0₃. Leaf N content of OR-US 4930 in November 1986 was significantly (p < .05) increased by increasing N applied as NH₄N0₃ or urea-DCD. Fruit S content of OR-US 4930 significantly (p < .05) increased with increasing N. ▼ Polyphenol oxidase (PPO) from strawberries was purified, partially characterized, and used to study its role in the degradation of anthocyanin pigments. The extraction method included the use of a buffered solution containing the phenolic binders Polyclar AT and Amberlite XAD-4. Addition of Triton X-100 resulted in an increase of the extracted activity. Higher levels of PPO activity were obtained using citrate rather than acetate in the buffer. This was possibly due to the capacity of citrate to chelate calcium ions, and, therefore, to inhibit crosslinking of the pectic polymers which would trap the enzyme. Using a Phenyl Sepharose CL-4B (PS) hydrophobic column two fractions of the enzyme, Fl and F2, were separated from the pectic material and the bulk of the 280 nm absorbing material. PP0-F1 obtained from the PS column was less hydrophobic than F2, and showed a molecular weight of 111,000 by gel filtration on Sephacryl S-300. Form Fl eluted in the void volume of an anion exchange column and did not penetrate into the running gel during electrophoresis. The binding of PP0-F1 to a Concanavalin-Agarose (ConA) column demonstrated the presence of carbohydrate associated with the enzyme. PP0-F2 showed a molecular weight of 34,500 and could be resolved by anion exchange chromatography into one large peak and a smaller one at its shoulder. Fraction F2 of the enzyme resolved into two bands during electrophoresis, and did not bind to the ConA column, Both fractions of the enzyme showed multiple banding on lithium dodecyl sulfate – polyacrylamide gradient gels. Both fractions of the enzyme obtained from the PS column showed very high activity in the presence of D-catechin. The Michaelis constants for D-catechin for PPO fractions Fl and F2 were 0.50mM and O.4lmM respectively, and the maximum velocity 82,700 and 18,800 nmoles 0₂ per min per ml respectively. D-catechin in the presence of PP0-F1 was rapidly oxidized and the resulting solution had a maximum absorption at 390 nm. When D-catechin and PP0-F1 were combined in model systems with either pure cyanin or pelargonin, an absorbance peak at 390 nm was noticed with increased absorbance in the region of the anthocyanin pigments. After 24 hr, 50% of the pelargonidin and 60% of the cyanin was destroyed and a brown precipitate was formed. PPO-Fl seemed to oxidize cyanin at a very slow rate but did not oxidize pelargonin. It was suggested that the anthocyanin pigments were destroyed by either the direct oxidation by the quinones formed from D-catechin by PPO, or that the anthocyanin pigments were co-polymerized into the brown polymeric pigment, tannin, formed from D-catechin-quinone polymerization. ▼ Preservation of food by freezing is a common method of processing. Of relatively recent origin is the freeze-drying method. This method has several advantages over other methods of preservation, but is not free of problems, one of which is the maintenance of appearance and texture. In the present study, the effects of freezing, thawing, freeze-drying, and reconstitution on the physico-chemical properties of Northwest variety strawberries with particular reference to textural characteristics were investigated. Very little prior work is reported with regards to the effect of these factors on fruits. Strawberries were frozen at two different rates of freezing - slow frozen at 0°F and quick frozen in a blast freezer at -20°F. Frozen berries were examined by physical and chemical methods to evaluate the changes. In a separate study, berries were thawed under standard conditions of relative humidity and temperature and the drip collected for analysis. Thawed berries were examined by both physical and chemical methods. To study the reconstitution behaviour of the berries, they were freeze-dried and reconstituted under standard conditions of berry to water ratio, temperature of water and time of reconstitution. Reconstituted berries and the remaining solutions were then examined. Measurements of the texture, color, total solids, alcohol insoluble solids, pectins, cellulose, ash, and constituents of ash were made on raw, frozen, thawed, and reconstituted berries. The results indicated the following conclusions: 1. An increase in percent soluble solids, pH, and titratable acids and a decrease in total solids and AIS of the berries were observed upon freezing. Slow frozen berries showed more pronounced effects. Smaller berries were observed to change the least. No significant differences were found in the other constituents. 2. Strawberries lose weight upon thawing depending on the chemical composition of the berries. An inverse relationship was observed between weight loss and total solids, AIS, pectic substances, cellulose, ash and mineral content of the berries. With gradual increase in the weight loss upon thawing, there was a progressive softening of the berries. A direct relationship was found between weight loss and the amount of pectic substances, sugars, titratable acids, ash, and minerals found in the drip. Slow freezing resulted in higher weight loss and smaller berries lost the least. 3. A linear relationship exists between weight loss of the berries upon thawing and the area of the drip. This method offers a convenient and quick procedure for evaluation of the quality of frozen strawberries, with the added advantage of collecting the drip and storing it for further analysis at a convenient time. 4. Ability to reconstitute was found to be related to rate of freezing and size of berry. Quick freezing and smaller berries gave the best results. 5.
