This thesis explores the Rapid Recommendations process, a new responsive way of creating clinical practice guidelines.

Healthcare workers rely on clinical practice guidelines to inform their practice. However, most guidelines are not trustworthy when judged by accepted standards and they typically take several years to produce. Guideline trustworthiness is undermined by panel members who often have conflicts of interest, by including representation from only a subset of stakeholders, by failing to examine the entirety of the evidence systematically, and by rapid obsolescence. Further, they are often difficult for users to understand in limited time. Rather than updating guidance on a fixed schedule, the Rapid Recommendations approach involves continuous monitoring of the literature and produces guidelines in response to new potentially practice-changing evidence. A collaborative network of clinicians, methodologists, and patients respond by rapidly producing trustworthy evidence syntheses and guidance. We have identified efficiencies at every step of the guideline development process. The guideline panel does not include anyone with a financial conflict of interest and there are strict limits professional and intellectual conflicts. Systematic reviews are produced on the relative effects of each option, on prognosis, and on patient values and preferences with the explicit intent to inform the question at hand. The panel also considers practical issues. Rapid Recommendations are published in a concise multilayered user-friendly format headed by an interactive infographic that contains all of the necessary information for users need to make informed decisions at the point of care. The guideline is published simultaneously in print and electronically, including decision aids that can be used at the point of care and integrated into electronic medical records. In this thesis, you will find a selection of exemplary publications relevant to the Rapid Recommendations process. We show that a responsive approach to rapid and trustworthy guideline creation is possible. It represents a way forward from the current limitations that plague most current clinical practice guidelines.

Candidate in Philosophy

Healthcare workers often decide what to do in practice based on the advice of experts through clinical practice guidelines. However, most clinical practice guidelines are not completely trustworthy. Guideline authors often have conflicts of interest, do not include patients or patient views, and are created so slowly that they rapidly fall into obsolescence. This thesis explores a new way of developing clinical practice guidelines that we call Rapid Recommendations. Instead of creating them on a fixed schedule (i.e., every few years), they are created in response to new studies that might change practice. Objetivou-se no presente trabalho a avaliação de metodologias existentes e novas para monitoramento da qualidade dos grãos de café durante seu processamento, bem como a adaptação das metodologias que determinam a qualidade de sementes, para uso na avaliação da qualidade de grãos de café, visando uma maior eficiência na avaliação destes . Os cafés utilizados no experimento foram separados de acordo com seu estádio de maturação, formando dois lotes: frutos maduros e frutos imaturos, que posteriormente foram descascados e levados ao terreiro para uma pré-secagem.A secagem mecânica foi conduzida em três secadores de camada fixa nas seguintes temperaturas: 35C, 40C e 45C, ocorrendo amostragem dos grãos de duas em duas horas, para análises pelo Lercafé e Biospeckle.Após secos os dois lotes foram subdivididos, sendo uma parcela beneficiada mecanicamente e a outra parcela teve seu pergaminho retirado manualmente. Após a aplicação dos tratamentos, os cafés foram degustados segundo o sistema de avaliação proposto pela Associação Americana de Cafés Especiais. Além da análise sensorial foram feitas as análises da Condutividade elétrica, Tetrazólio, Lercafé e Biospeckle Laser. Pode-se observar que os testes Lercafé e Biospeckle não conseguiram identificar nos grãos de cafés danos ocorridos durante o processo de secagem. No entanto o Lercafé apresentou-se eficaz, como também uma metodologia rápida na avaliação final dos grãos, sendo capaz de diferenciar danos térmicos, diferentes estádios de maturação e injúrias ocorridas pelo beneficiamento. O método do Biospeckle Laser não se mostrou assertivo em relação as análises conduzidas, todavia apresentou resultados que apontam para sua capacidade de perceber o grau de desestruturação molecular sofrido após a secagem.

The aim of this work is to evaluate the existing methodologies for monitoring of the coffee quality of the grains of coffee during processing, adapting the methodology that determine the seed quality for use in evaluate of the coffee quality, in order increase efficiency in the evaluation of the same and exploitation biospeckle as a tool non destructive for evaluation of the biological activity of the grain and its quality. The coffees used in the experiment were separated according to their maturation stage, making two groups: ripe fruit and immature fruit, where, after were stripped and taken to the yard for a pre-drying. The mechanical drying was conducted in three dryers machines with layer fine at the following temperatures: 35 C, 40 C and 45 C, grain sampling occurring each two hours for analysis by Lercafé and biospeckle. After dry the two set were subdivided, which a part were mechanically benefit and the other part had his parchment removed manually. After treatment application, the coffees were tasted by a system of evaluation proposed by Specialty Coffee American Association. In addition the sensorial analysis, were made by electrical conductivity analysis, tetrazolium, Lercafé and biospeckle. It can be observed that the Lercafé tests and biospeckle technique…

▼ Quantitative analysis of casein content in cheese milk can give a better control over cheese yield and understand cheese quality. Traditional analytical methods for casein measurement rely on nitrogen-based quantification and involve time-consuming sample preparation steps. The current study applied front-face fluorescence spectroscopy (FFFS) combined with chemometrics to quantify casein and casein-to-crude-protein ratio (CN/CP) in milk intended for cheese manufacturing. FFFS spectra of acid-precipitated casein milk dispersion (pH = 4.6) were collected from 20 ultrafiltered (UF) model milk samples with different casein contents. A preliminary calibration model was developed with principal component regression (PCR) using reference casein contents and the FFFS spectra. The model was externally validated with 20 raw milk samples and a root mean square error (RMSE) of 0.15% was found between the predicted and reference casein contents. A relative prediction error (RPE) of 6.7% indicating usefulness for quality control purposes. To further refine the FFFS-based casein quantification method, 30 model milk samples prepared from UF and microfiltration (MF) permeates and retentates to generate different casein contents and CN/CP. The FFFS spectra were collected following the same procedure and used as predictors for casein and CN/CP quantifications. Calibration models were developed using partial least squares regression (PLSR) and elastic net regression (ENR) and the models were further optimized using 20 samples including raw, skim, and UF milk. The optimized PLSR and ENR models were again tested using 20 test samples including raw, skim, and UF milk and evaluated in terms of RMSE, residual prediction deviation (RPD), and RPE. The PLSR and ENR models reduced the RMSE for casein quantification to 0.13% with RPD ranged from 3.2 to 3.4, indicating practical model performances. For CN/CP quantification, PLSR models resulted in useful predictions with an RMSE of 0.024, an RPD of 1.5, and an RPE of 3.0%. The FFFS-based casein quantification method provides a rapid casein measurement in fluid milk and can be implemented in the cheese industry for routine measurements. In a different study, FFFS was utilized to predict the protein leaks in permeate during membrane processing of skim milk and whey. Protein leak occurs when proteinous matters pass through the UF membrane into the permeate stream leading to financial losses and product quality defects. FFFS as a sensitive and specific instrument was applied to characterize protein leak occurrences in UF permeate, develop chemometrics models to quantify true protein (TP) content in permeate streams, and classify sources of protein leak in the feed material. Measurements of crude protein (CP), non-protein nitrogen, TP, tryptone-equivalent peptide, α-lactalbumin (α-LA), and β-lactoglobulin (β-LG) were performed on 33 lots of commercial whey permeate and 29 lots of commercial milk permeate. Este estudo foi realizado com o objetivo de desenvolvimento e validação do método microbiológico rápido empregando a técnica de bioluminescência de Adenosina Trifosfato (ATP) como método alternativo para o teste de esterilidade. O ATP reage com o sistema enzimático luciferina/luciferase e gera um fóton de luz em presença de íons magnésio, reação que pode ser utilizada na detecção de microrganismos. A luz gerada na reação é medida por um dispositivo chamado luminômetro, que traduz o sinal em unidades relativas de luz (URL), metodologia altamente sensível que pode ser utilizada na análise de produtos estéreis com o objetivo de diminuição no tempo do ensaio. Enquanto a turbidez do meio de cultura só pode ser visualizada quando o contaminante chega à concentração de 106 UFC/ml, a tecnologia de bioluminescência de ATP pode detectar amostras com concentração em torno de 104 UFC. Foram empregados na validação dados obtidos a partir do método tradicional de esterilidade (técnica de filtração) realizado paralelamente ao método alternativo. As soluções parenterais utilizadas nos ensaios foram: solução fisiológica 0,9%; solução de dextrose 5%; ringer lactato; e solução de metronidazol 0,5%. As soluções-teste foram inoculadas intencionalmente com suspensões microbianas preparadas através da diluição de Bioballs®, com concentrações de 10 UFC/100 ml, 2 UFC/100 ml e 0,4 UFC/100 ml. Após a realização do teste convencional, as membranas resultantes do ensaio foram incubadas nos meios de cultura caldo caseína de soja e tioglicolato. Alíquotas destes meios foram retiradas após 96 horas de incubação para análise pelo método alternativo. Os seguintes microrganismos foram selecionados para o estudo: Staphylococcus aureus, Bacilus subtilis, Pseudomonas aeruginosa, Candida albicans, Clostridium sporogenes, Aspergillus brasiliensis, Kocuria rosea e Micrococcus luteus. A análise dos resultados obtidos mostrou que o método alternativo é capaz de detectar os microrganismos testados. Quanto à sensibilidade, o método alternativo apresentou vantagem na concentração 2 UFC/100 ml, e equivalência nas outras concentrações. A não interferência dos diferentes produtos e meios nos resultados encontrados permite vislumbrar evidência de robustez do método. Adicionalmente, em relação ao tempo de resposta, o método alternativo demonstrou ser equivalente ao convencional (p-valor=0,43).

This study is being conducted with a goal of validating and developing the fast microbiological method of ATP bioluminescence, as an alternative method to the sterility test. Salmonella spp é um dos microrganismos patogênicos de relevância em alimentos. Sua detecção em alimentos por metodologia de cultura é trabalhosa e demorada, o que explica a grande variedade de sistemas automatizados e kits para detecção rápida desse patógeno existentes no mercado. Muitos desses métodos alternativos são validados por organizações internacionalmente reconhecidas, mas tais validações têm sido efetuadas em condições laboratoriais artificiais e com matrizes alimentares experimentalmente contaminadas, dificilmente refletindo a realidade de um laboratório de rotina de análises microbiológicas de alimentos. Nesse trabalho, avaliou-se o desempenho de sete métodos rápidos alternativos genotípicos e imunológicos, usados simultaneamente para detecção de Salmonella spp em 244 amostras de alimentos sem contaminação experimental, empregando o método de cultura ISO 6579:2002 como método de referência. Os métodos avaliados foram BAX Salmonella,VIDAS Salmonella (SLM),Transia Plate Salmonella Gold,VIP Salmonella,TECRA UNQUIQUE SALMONELLA,Singlepath Salmonella e 1-2 Test. O nível de detecção para cada método também foi determinado, trabalhando-se com três matrizes alimentares diferentes, experimentalmente contaminadas com cinco níveis de inóculo. O método de referência detectou 50 amostras (20,5%) positivas para Salmonella spp. Nas condições em que o trabalho foi realizado, a porcentagem de concordância dos resultados positivos obtidos pelos métodos alternativos avaliados em relação aos resultados positivos obtidos pelo método ISO 6579:2002 variou de 42,3% a 100% nas amostras sem contaminação experimental e de 75% a 100% nas amostras artificialmente contaminadas, dependendo do método avaliado e da matriz alimentar analisada. Todos os métodos rápidos avaliados apresentaram resultados falso-positivos e falso-negativos, em freqüências dependentes também do método avaliado e da matriz alimentar analisada. Os resultados obtidos evidenciaram que os métodos rápidos avaliados devem ser utilizados unicamente para fins de triagem, podendo haver um risco intrínseco na escolha de um método rápido para detectar Salmonella spp em determinada matriz alimentar. A seleção de um método rápido, em particular, só deve ser feita após a determinação do grau de risco que se queira assumir.

Salmonella spp is an important foodborne pathogen. The detection in foods by the cultural methodology takes at least five days to be completed and several automated systems and kits are available in the market for rapid detection of this pathogen. Most rapid methods are validated by internationally recognized organizations, but most validations have been conducted in artificial laboratory conditions, working with inoculated food samples, hardly reflecting the real conditions of a routine testing laboratory. This test makes it possible to detect antibodies against tetanus in equine serum within one hour. Moreover the prototype allows making a statement about the antibody titer. The standard values of the OIE (WORLD ORGANISATION FOR ANIMAL HEALTH, 2009) to validate in vitro-diagnostical tests for veterinary medicine were followed. The rapid test achieves results comparable to the ELISA. The cut off of the rapid test averaged 0,0921 IU/ml. The sensitivity reached 95,65%. There could not be detected any cross reactions, which equals a specificity of 100%. There are limitations to analyzing serum samples with an antibody titer below the cut off. The prototype of a “rapid test” also should only be used to analyze serum samples of horses vaccinated more than two weeks ago. This is important, because it is not possible to differentiate between IgM and IgG. Thus, the prototype could give a false positive result. IgM can attach to the tetanustoxin, but there does not proceed any neutralization reaction of the toxin (OURTH et MAC DONALD, 1977). The handling of the prototype of a “rapid test” would be easier, if whole blood could be used. A display panel in front of the analyzer showing the antibody titer would also expedite the process. The results of the in field tests are the following: -All horses of a group were vaccinated according to the same pattern. They showed however different antibody titers. For this reason, the directions of the producer of the vaccine should be followed strictly in order to ensure the immunization protection. -Foals whose mothers were not vaccinated against tetanus during the pregnancy should be tested for their antibody titer against tetanus at the age of three months, if there could not be assured an adequate ingestion of colostrum. -Foals should be vaccinated at the age of five or six months, if their mothers were not vaccinated additionally during the pregnancy. It is possible to variate the beginning of the vaccination, if the antibody titer is beyond the cut off. Therefore, the tetanus antibody titer of foals has to be detected. -The maximum antibody titer can be detected 14 days after the first vaccination. After the second vaccination, the maximum antibody titer is already reached after five days. -After the periodical booster vaccination the maximum antibody titer can be detected after 14 days. -There could be detected an increase of the antibody titer in horses which came down with tetanus and were medicated with a serotherapy. ▼ Ɣ-ray bursts (GRBs) are the Universe's most luminous transient events. Since the discovery of GRBs was announced in 1973, efforts have been ongoing to obtain data over a broader range of the electromagnetic spectrum at the earliest possible times following the initial detection. The discovery of the theorized ``afterglow'' emission in radio through X-ray bands in the late 1990s confirmed the cosmological nature of these events. At present, GRB afterglows are among the best probes of the early Universe (z ≳ 9). In addition to informing theories about GRBs themselves, observations of afterglows probe the circum-burst medium (CBM), properties of the host galaxies and the progress of cosmic reionization. To explore the early-time variability of afterglows, I have developed a generalized analysis framework which models near-infrared (NIR), optical, ultra-violet (UV) and X-ray light curves without assuming an underlying model. These fits are then used to construct the spectral energy distribution (SED) of afterglows at arbitrary times within the observed window. Physical models are then used to explore the evolution of the SED parameter space with time. I demonstrate that this framework produces evidence of the photodestruction of dust in the CBM of GRB 120119A, similar to the findings from a previous study of this afterglow. The framework is additionally applied to the afterglows of GRB 140419A and GRB 080607. In these cases the evolution of the SEDs appears consistent with the standard fireball model. Having introduced the scientific motivations for early-time observations, I introduce the Rapid Infrared Imager-Spectrometer (RIMAS). Once commissioned on the 4.3 meter Discovery Channel Telescope (DCT), RIMAS will be used to study the afterglows of GRBs through photometric and spectroscopic observations beginning within minutes of the initial burst. The instrument will operate in the NIR, from 0.97 μm to 2.37 μm, permitting the detection of very high redshift (z ≳ 7) afterglows which are attenuated at shorter wavelengths by Lyman-ɑ absorption in the intergalactic medium (IGM). A majority of my graduate work has been spent designing and aligning RIMAS's cryogenic (~80 K) optical systems. Design efforts have included an original camera used to image the field surrounding spectroscopic slits, tolerancing and optimizing all of the instrument's optics, thermal modeling of optomechanical systems, and modeling the diffraction efficiencies for some of the dispersive elements. To align the cryogenic optics, I developed a procedure that was successfully used for a majority of the instrument's sub-assemblies. My work on this cryogenic instrument has necessitated experimental and computational projects to design and validate designs of several subsystems. ▼ Polycyclic aromatic hydrocarbons (PAHs) are a class of potent environmental pollutants which exhibit carcinogenic, mutagenic, and teratogenic properties. High levels of PAHs are found in food products cooked at high temperatures, such as fried foods and repeatedly used or illicitly recycled oils. Screening and detection of PAH compounds are an urgent demand to ensure cooking oil safety, but current techniques require labor-intensive and time-consuming sample preparation procedures. To shorten the detection time and avoid complicated sample pretreatment, a new technique combining ultra-thin layer chromatography (UTLC) and surface enhanced Raman spectroscopy (SERS) has been proposed and evaluated for rapid screening of PAHs in oil samples. The UTLC-SERS method utilizes the nanoporous and Raman enhancing properties of the silver nanorod (AgNR) substrates for chromatographic separation of mixture samples directly on the sensing surface within minutes. In this dissertation, the UTLC-SERS principles were demonstrated using model Raman reporter molecules, and subsequently applied in the detection of PAHs. The AgNR substrate was modified with mercaptoethanol (ME), which served as the stationary phase in UTLC. The mobile phase was optimized for the separation of three representative PAHs, benz(a)anthracene (BaA), benzo(a)pyrene (BaP), and pyrene (P). PAHs were extracted from artificially contaminated vegetable oil samples through a 1-min acetonitrile extraction procedure, and the organic phase was directly used for UTLC-SERS without further treatment. The UTLC process served to elute target PAHs from the oil matrix to near the solvent front, where the SERS signal of BaA, BaP, and P could be identified at concentrations as low as 50 µg PAH/mL oil. The UTLC-SERS approach was demonstrated to rapidly identify BaA, BaP, and P from vegetable oil at relatively high contamination levels without complicated sample preparation. To achieve lower detection limits and more efficient separation among the PAHs, more in-depth work is needed on improving the UTLC solvent migration as well as refining the sample preparation protocols.

▼ Uncorrected refractive errors (URE) and presbyopia are public health challenges in community eye health today. Effective planning and management of the services to tackle these twin problems requires information for needs assessment and monitoring. Conventional cross sectional studies (population-based) which can provide this information are resource intensive and hence not repeatable. The current research was undertaken to develop a low cost rapid assessment methodology that can fill this gap. A two stage approach was adopted. In the first stage, the RARE (Rapid Assessment of Refractive Errors) methodology was piloted and the definition for URE was validated against the standard definition for refractive errors. In the second stage, the validated methodology was used in Mahbubnagar district in Andhra Pradesh, India. The results from this study were compared with those obtained from a conventional cross sectional study conducted a decade ago. Cluster random sampling with ‘population proportionate to size (PPS)’ was employed as done in other rapid assessments. Distance visual acuity (VA) (aided, unaided and pinhole) was measured using a LogMAR Chart (English alphabet optotypes & tumbling E), near vision (unaided, aided and with age appropriate near addition) was measured with near vision chart with N notation held at 35 cm distance. Visual impairment (VI) was defined as binocular presenting VA<6/12, URE was defined as presenting VA<6/12 but improving to at least 6/12 or better on using a pinhole. Uncorrected Presbyopia was defined as binocular unaided near vision <N8 in subjects aged >35 years and who had no distance VI. In the pilot study, the definition for URE based on pinhole showed good sensitivity and specificity. The main study revealed a large ‘unmet need’ for correction of uncorrected refractive errors and presbyopia. The spectacle coverage was low for both refractive errors and presbyopia. However in comparison with a cross sectional study, prevalence of URE has decreased and there was an increase in the prevalence of spectacle use. The current research demonstrated that RARE can provide vital information for planning and management of refraction services and can assist in achieving the aim of the global initiative, VISION 2020: The Right to Sight. ▼ The research in this manuscript highlights new and improved methods to concentrate pathogens from a complex food matrix and detect them via PCR. These contributions to the science of rapid pathogen detection are unique in that they are designed to measure the efficacy of the concentration or capture technique without using a pre-enrichment step. To address the potential of pathogen concentration to facilitate PCR detection, the following objectives were performed: (i) evaluating the performance of differential centrifugation as a means to concentrate and clarify the food sample for rapid PCR detection of Listeria monocytogenes, without pre-enrichment and (ii) investigating the efficacy of a novel immuno-capture device to capture E. coli O157:H7 and Salmonella typhimurium from foods. For the first objective, we investigated filtration followed by a two-step, differential centrifugation as a means to concentrate bacteria and remove a large portion of the food sample prior to DNA extraction, PCR amplification, and Southern hybridization of L. monocytogenes targeting a unique region of 16S rDNA. Simple high speed centrifugation (11,950 x g) was also investigated to test the efficacy of our two-step method. Our method incorporated use of a 11g sample of ready-to-eat deli salad diluted 1:10 with 99ml 0.9% sterile normal saline. The two-step method was able to reduce the sample volume by approximately 10-fold rather than only 5-fold for simple high speed centrifugation. The two-step method was 1,000 fold (10⁶ to 10³ CFU/g) more sensitive than when using high speed centrifugation alone, and bacterial recoveries indicated that both methods produced similar recoveries. Following DNA extraction, PCR amplification, and Southern hybridization, detection was achieved at input levels of 10⁵ CFU/g for chicken salad, 10⁴ CFU/g for macaroni salad, and 10³ CFU/g for potato and seafood salads, with no pre-enrichment. In our second objective, we evaluated the efficacy of a novel immuno-capture system (Pathatrix™) to capture S. typhimurium in buffered peptone water (BPW), ground turkey and nonfat dry milk and E. coli O157:H7 from BPW, ground beef and romaine lettuce. The Pathatrix system is unique in that it is designed to sample an entire 25g sample by circulating the homogenized 250ml volume across a surface of immunomagnetic beads. The samples were seeded with bacteria at levels ranging from 10⁶ to 10⁰ CFU/25g. The E. coli O157 format was able to capture 100 % of input E. coli O157:H7 and PCR amplification was able to detect the pathogen at 10⁰ CFU/25g. The Salmonella format was not as robust, only capturing approximately < 1 % of cell input, but was able to produce a significant PCR detection limit of 10² CFU/25g when Pathatrix was preceded with the two-step centrifugation method. The two-step centrifugation further clarified the sample and improved detection by 1000-fold (10⁵ to 10² CFU/25g). ▼ Submarine groundwater discharge (SGD) can be an important pathway for nutrients entering coastal systems. However SGD flow paths can be difficult to identify and flow volumes difficult to quantify. This study assesses whether geophysical techniques are potentially cost effective methods for detecting the presence or lack of SGD within an estuary environment found in Sarasota Bay, Florida. In this area, a rapid increase in urbanization has led to increased nitrogen loading into the bay, with some 10% of this loading attributed to SGD. Discharging groundwater is expected to be fresher and hence higher resistivity, than "background" surface waters. Thus resistivity surveys sensitive to seafloor conductivities may be useful for identifying zones of SGD. However, terrain resistivities are influenced by matrix geology as well as pore water resistivity. In this study we compare the results of marine resistivity surveys against both geochemical measures of SGD (radon tracers) and seismic profiles indicative of subsurface structure to better determine the relative impacts of geology and SGD on marine resistivity measurements in Sarasota Bay. On both regional (kilometers to tens of kilometers) and local scales (hundreds of meters) the relationship between marine resistivity and tracer-based SGD estimates does not follow the expected pattern of higher resistivities associated with higher SGD flux. Seafloor resistivities instead appear primarily influenced by stratigraphy, particularly the presence of a clay layer at ~10-15 m depth in the southern part of the bay. In the southern bay, resistivities decrease at the depths associated with the clay layer. On the local (hundreds of meters) scale, lateral variations in resistivities derived from inversions of resistivity data were not found to be reproducible; nearly-coincident lines collected 30 minutes apart in time show different local signatures. This apparent local lateral variability in the resistivity profiles is inferred to be a result of inversion of noisy streaming resistivity data.

Abstract Management of product development activities has become increasingly important, as cycle times of product development have shortened. Smaller product development projects are often conducted rapidly at companies based on customer or sales requests to answer the need for faster cycle times. However, this is often done without fully realizing the impact of the new projects on the larger project portfolio or organizational effectiveness. The main objective of this dissertation is to increase knowledge on the use of agile development methods in small, rapid product development projects, and on the implementation of a rapid product development model. The dissertation is formulated as a qualitative, inductive study based on the research results of four original publications and a summary combining the results. The results of the dissertation show that it is beneficial to separate a rapid product development process for certain types of customer- or sales-initiated projects. A new rapid development model with principles and guidelines is introduced to help organizations facilitate this separation. The implementation of the model can be supported with agile development practices, of which self-managing teams are studied in more detail. There is significant overlap between case companies’ perceived success factors for rapid development and self-management. The results imply that a functional rapid development model can be utilized as a strategic asset at companies. The results also provide empirical evidence that agile development practices can be utilized in product development. In addition to providing empirical evidence in scientific discussion about combining product development and agile software development practices, the results can be used to create better definitions of product development processes in general.

Tiivistelmä Tuotekehityksen johtamisesta ja hallinnasta on tullut entistä haastavampaa ja tärkeämpää, kun tuotekehitysprojektien läpimenoajat ovat lyhentyneet. Yritykset tekevät kasvamassa määrin lyhyitä tuotekehitysprojekteja asiakaspyyntöjen tai myynnin aloitteesta vastatakseen markkinoiden vaatimuksiin nopeasta kehityksestä. Tällaisten nopeiden tuotekehitysprojektien käynnistäminen ja toteutus tehdään usein ymmärtämättä yksittäisen projektin vaikutusta koko projektiportfolioon tai organisaation tehokkuuteen. Tämän väitöskirjan päätavoitteena on tutkia ohjelmistokehityksestä tunnettujen ketterien kehitysmenetelmien käyttöä nopeissa tuotekehitysprojekteissa ja uudenlaisen nopean tuotekehityksen mallin käyttöönotossa. ▼ This work develops optimal assembly sequences for modular building blocks. The underlying concept is that an automated device could take a virtual shape such as a CAD file, and automatically decide how to physically build the shape using simple, identical building blocks. This entails deciding where to place blocks inside the shape and generating an efficient assembly sequence that a robot could use to build the shape. The blocks are defined in a general, parameterized manner such that the model can be easily modified in the future. The primary focus of this work is the development of methods for generating assembly sequences in a time-feasible manner that ensure static stability at each step of the assembly. Most existing research focuses on complete enumeration of every possible assembly sequence and evaluation of many possible sequences. This, however, is not practical for systems with a large number of parts for two reasons: (1) the number of possible assembly sequences is exponential in the number of parts, and (2) each static stability test is very time-consuming. The approach proposed here is to develop a multi-hierarchical rule-based approach to assembly sequences. This is accomplished by formalizing and justifying both high-level and mid-level assembly rules based on static considerations. Application of these rules helps develop assembly sequences rapidly. The assembly sequence is developed in a time-feasible manner according to the geometry of the structure, rather than evaluating statics along the way. This work only evaluates the static stability of each step of the assembly once. The behavior of the various rules is observed both numerically and through theory, and guidelines are developed to suggest which rules to apply. A secondary focus of this work is to introduce methods by which the inside of the structure can be optimized. This structure optimization research is implemented by genetic algorithms that solve the multi-objective optimization problem in two dimensions, and can be extended to three dimensions. ▼ Professors need alternative programs to support their online teaching. This dissertation reports an initial study in a long-term research agenda for developing a faculty online teaching solution. The primary purpose of the study is to explore faculty perceptions of a case library to help decision makers and researchers determine whether they would pursue the use of such a tool to support faculty online teaching. The secondary purpose of the study is to generate design knowledge to inform future development of and research on this or similar case libraries. The methodology of this study includes three components: development research, rapid prototyping, and qualitative methods. Development research and rapid prototyping provided a three-stage framework for this study: conceptualization, development, and research. I synthesized the literature to create conceptual models of an Online Teaching Case Library (OTCL) at the conceptualization stage, built a prototype to implement the models at the development stage, and conducted research to evaluate the prototype at the research stage. Qualitative methods guided data gathering and analysis. I recruited seven faculty participants based on a purposeful sampling technique. To gather the data, I followed a three-step data collection process: initial interviews, contextual interviews, and final interviews. This process allowed me to observe and interview faculty participants while they were exploring the prototype. I analyzed the data by following an 11-step procedure synthesized from the works of Miles and Huberman (1994) as well as LeCompte and Schensul (1999a). This study found that on one hand, faculty members might use an OTCL, because they perceived that this tool could support their apprenticeship approach to learning to teach. On the other hand, however, their perceived decision to use an OTCL would also be influenced by the perceptions of the usefulness and usability of the tool. The study identified the initial evidence supporting an OTCL as an online teaching resource and the challenges involved in developing and implementing such a solution. It provides a base for decision makers to determine whether they would adopt this tool. It also offers some design guidance for those who do want to pursue this solution to faculty development. ▼ ECA Check® Easygel® Plus (ECA) is a pectin-based gelling system that reacts with calcium ions bound to a pre-treated Petri dish, eliminating autoclaving prior to use. It can chromogenically and/or fluorogenically distinguish three organisms: Escherichia coli, Salmonella spp., and coliforms. This study compared the recovery of these organisms to conventional media using stock culture, inoculated, and non-inoculated ground beef and ground turkey. ECA was compared to Violet Red Bile Agar (VRB), Violet Red Bile Agar with 4-methylumbelliferyl-β-D-glucuronide (VRB-MUG), Xylose Lysine Desoxycholate Agar (XLD), Escherichia coli/Coliform (ECC) Count Plate Petrifilm™, and Tryptic Soy Agar (TSA). The stock culture recovery of Salmonella Typhimurium for ECA, TSA, and XLD were 8.62, 8.69, and 6.82 log CFU/ml, respectively. There was very little difference between the media in the recovery of Escherichia coli and Cronobacter spp., formerly referred to as Enterobacter sakazakii. Mean counts of presumptive E. coli in ground beef were 7.24 and 7.41 logs for ECA and VRB-MUG. Total coliform mean counts were 7.43, 7.63, and 7.37 logs for ECA, Petrifilm™, and VRB. Presumptive Salmonella means were 6.68 and 6.21 logs on ECA and XLD, while total aerobic counts were 7.84 and 6.51 logs on ECA and TSA. At 6.72 logs, ECA recovered considerably more Salmonella than XLD (5.71 logs) from the inoculated ground turkey; ECA recovered 7.62 logs total aerobic count which was significantly more than TSA at 6.89 logs. Total counts for both non-inoculated ground meats resulted in significant differences between TSA recovery and all other media. ECA also recovered significantly more than Petrifilm™ from both non-inoculated foods. The randomly selected organisms recovered from ECA were identified using BBL™ Crystal™ Enteric/Nonfermenter ID or Gram-Positive kits, and correlated precisely to the chromogenic reaction of the colonies. ECA Check® Easygel® was efficient, less labor-intensive, comparable to, and, in some instances, better than conventional media at recovering target organisms. Advisors/Committee Members: Daniel Y.C. ▼ Traditionally, the design of complex aerospace systems requires iteration among segregated disciplines such as aerodynamic modeling and trajectory optimization. Multidisciplinary design optimization algorithms have been developed to efficiently orchestrate the interaction among these disciplines during the design process. For example, vehicle capability is generally obtained through sequential iteration among vehicle shape, aerodynamic performance, and trajectory optimization routines in which aerodynamic performance is obtained from large pre-computed tables that are a function of angle of attack, sideslip, and flight conditions. This numerical approach segregates advancements in vehicle shape design from advancements in trajectory optimization. This investigation advances the state-of-the-art in conceptual hypersonic aerodynamic analysis and trajectory optimization by removing the source of iteration between aerodynamic and trajectory analyses and capitalizing on fundamental linkages across hypersonic solutions. Analytic aerodynamic relations, like those derived in this investigation, are possible in any flow regime in which the flowfield can be accurately described analytically. These relations eliminate the large aerodynamic tables that contribute to the segregation of disciplinary advancements. Within the limits of Newtonian flow theory, many of the analytic expressions derived in this investigation provide exact solutions that eliminate the computational error of approximate methods widely used today while simultaneously improving computational performance. To address the mathematical limit of analytic solutions, additional relations are developed that fundamentally alter the manner in which Newtonian aerodynamics are calculated. The resulting aerodynamic expressions provide an analytic mapping of vehicle shape to trajectory performance. This analytic mapping collapses the traditional, segregated design environment into a single, unified, mathematical framework which enables fast, specialized trajectory optimization methods to be extended to also include vehicle shape. A rapid trajectory optimization methodology suitable for this new, mathematically integrated design environment is also developed by relying on the continuation of solutions found via indirect methods. Examples demonstrate that families of optimal hypersonic trajectories can be quickly constructed for varying trajectory parameters, vehicle shapes, atmospheric properties, and gravity models to support design space exploration, trade studies, and vehicle requirements definition. These results validate the hypothesis that many hypersonic trajectory solutions are connected through fast indirect optimization methods. The extension of this trajectory optimization methodology to include vehicle shape through the development of analytic hypersonic aerodynamic relations enables the construction of a unified mathematical framework to perform rapid, simultaneous hypersonic aerodynamic and trajectory optimization.
