1. Lu, Chiao-Hui. The pregnancy outcomes for human embryos cultured in single step medium and sequential medium.

Since Louise Joy Brown, the first baby of In Vitro Fertilization-Embryo Transfer (IVF-ET), was born in the United Kingdom in 1978, there had been over five million successful cases over the course of 35 years. There are two commercially available culture media in the market: Single Step Medium (Global Medium) and Sequential Medium (Sage Medium). The development of these two media had enhanced the quality of embryo and increased the pregnancy rate in IVF-ET. In this collection of 210 cases of IVF-ET, oocytes were fertilized by intracytoplasmic sperm injectionï¼ICSIï¼and randomly assigned to the two commercially-available culture media. The quantity and quality of embryos were observed and recorded for three consecutive days in order to compare the development of embryos in the two commercial culture medium. A fertilized rate of 75.5% in Global Medium and 77.5% in Sage Medium were observed on the first day. There was a higher number of cleaved cells in Global Medium and embryos of Grade 1 cells than those in Sage Medium both on the 2nd and the 3rd day, though no significant differences were noted in the pregnancy rates between the two samples. In the group of those over 38 years old, there was a higher number of cells in Global Medium, but exhibited no difference in either embryonic pattern or in the pregnancy rate. Though the development of commercial culture medium had enabled a longer cultivation, some cases still failed be cultured to the blastocytic stage due to other limitations. It had been attempted with morphologic selection in embryos by the 3rd day of culture, with a criteria of over six cells per embryo and minimal cell fragmentation, to reach compatible pregnancy rate of embryo implantation of the 5th day by conventional method. Our study indicated that the there was higher number of Grade 1 embryos with more than 6 cells and lower embryo fragmentation in Single Step Medium, thus yielded a better overall quality of embryo than Sequential Medium. It can be speculated, therefore, that the Single Step Medium would offer a valuable choice in patients with difficulty in reaching blastocytic stage by conventional method due to various factors. O estudo teve como objetivo investigar os efeitos da adição de gás carbônico (CO2) na fase inicial de incubação sobre a eclodibilidade e a qualidade do neonato de frangos de corte. Duas diferentes condições atmosféricas de nível de CO2 foram estabelecidas em incubadoras de estágio único com capacidade para 20640 ovos cada, injetando-se em uma delas 4000ppm de CO2 durante os 4 primeiros dias de incubação. Ambas incubadoras (sem e com adição de CO2) foram controladas para obtenção da temperatura média de 37,5ºC, umidade relativa de 60%, uma viragem a cada hora por 19 dias e ventilação, garantindo-se 96 trocas gasosas durante 24 h. Cada tratamento foi repetido em quatro incubações sequenciais de ovos originados de mesmo lote de matrizes pesadas entre 34 e 41 semanas de idade. As características avaliadas foram: perda de peso do ovo (1 a 19 dias), eclodibilidade, mortalidade embrionária, observação dos ovos bicados com embriões vivos ou mortos, fertilidade aparente, percentagem de nascimento sobre ovos férteis, comprimento e peso dos neonatos. Os resultados foram analisados segundo delineamento de blocos casualizados com dois tratamentos (com e sem adição de CO2) e quatro repetições (sequência de incubação), totalizando 82560 ovos incubados por tratamento. Não houve diferença significativa entre os tratamentos para todas as características avaliadas. Apesar de não letal para os embriões, a adição de 4000ppm de CO2 nos quatro primeiros dias de incubação de ovos de matrizes pesadas não influenciou positivamente os resultados da incubação ou a qualidade dos pintainhos

The aim of is experiment was to study effects of high levels of carbonic gas (CO2) at initial phase of incubation upon hatchability and chick qualities. Two different CO2 atmosphere conditions in single stage incubators of 20640 eggs each were tested: injecting in one of them 4000ppm of CO2 on the four first days of incubation. Our management of both machines (with and without CO2 injection) included control of temperature (37,5ºC), relative humidity (60%), rotation each hour up to 19 days of incubation, and ventilation at 96 gas exchanges rate per 24 h. Treatments were replicated four times on four sequential incubations of eggs obtained from the same flock breeders aging 34 to 41 weeks. The evaluated parameters were: weight egg loss (1 to 19 days), hatchability, embryo mortality, alive piped eggs, death piped eggs, fertility, % hatching per fertile egg, length and weight of neonates. Results were analyzed according to a randomized block design with two treatments (with and without CO2 injection) and four replicates, totaling 82560 incubated eggs per treatment.
