▼ The oestrous cycle of the domestic bitch is unique with regard to its considerable length compared with that of most other domestic animals. A non-seasonal anoestrus, with a duration that may last from 2 to 10 months, follows each oestrous cycle. The anoestrus can be prolonged by oestrus-preventing drugs such as progestins. The general aim of the studies in this thesis was to get further insight into the endocrinology of the physiological and the progestin-induced canine anoestrus. Studies performed throughout recent years have made clear that during the course of canine anoestrus many endocrinological changes take place at the hypothalamic, pituitary, and ovarian level. Apart from these changes, there are indications of involvement of dopaminergic influences. The results of the study described in Chapter 3 provide forceful evidence that the bromocriptine-induced shortening of the interoestrous interval is not triggered by a decline in plasma prolactin concentration. The results of the study described in Chapter 4 provides additional evidence that the premature onset of oestrus brought about by a dopamine agonist must be due to some other dopamine-agonistic effect, probably increased FSH secretion. The study in Chapter 5 is a report on the effects of GnRH administration on the plasma concentrations of reproductive hormones in intact and OVX bitches. The data presented in this study demonstrate that provocative testing of the pituitary-ovarian axis using GnRH may be helpful in differentiating between bitches in anoestrus and neutered bitches. The study reported in Chapter 6 describes the basal and GnRH-induced plasma concentrations of FSH and LH in four anoestrous and four OVX bitches. In this report indications were found that measurement of the plasma FSH concentration in a single plasma sample may prove reliable for determination of neuter status. Taken together, the results of the studies described in Chapters 5 and 6 provide a basis for the diagnosis of remnant ovarian tissue and verification of neuter status in the bitch. Chapters 7 and 8 are reports on the effect of MPA on canine adenohypophyseal function. The results of both studies demonstrate that treatment with MPA affects the hypothalamic-pituitary-ovarian axis. Oestrus, ovulation, and a subsequent luteal phase did not occur in any of the bitches during treatment with MPA. The prevention of oestrus by MPA cannot be ascribed to a significant reduction in circulating levels of either FSH or LH. On the contrary, during the first months of MPA treatment there was an increase in basal plasma FSH and LH, which may be due to a direct oestrus-preventing effect of MPA at the ovarian level. With continuing MPA treatment, basal plasma gonadotrophin concentrations returned to pretreatment levels and the pituitary FSH response to GnRH stimulation decreased, while several LH pulses were not accompanied by a significant FSH pulse, suggesting that MPA treatment attenuated pituitary FSH sensitivity to endogenous GnRH. ▼ Pharmaceuticals have routinely been detected in the environment resulting in a growing concern about whether these drugs could elicit effects on aquatic organisms. The concerns are centered on the highly conserved nature of mammalian therapeutic targets in fish. These pharmaceuticals are found at very low levels in the environment, which can result in sub-lethal effects in aquatic organisms. Therefore, 28 d early-life stage studies were conducted on six pharmaceuticals to assess their impacts on survival and growth fathead minnow larvae. Two pharmaceuticals tested, carbamazepine and fenofibrate, resulted in no alterations to survival and growth. However, amiodarone, clozapine, dexamethasone, and levonorgestrel (LNG) reduced survival at concentrations tested with LNG being the most potent at 462 ng/L. Survival was increased with amiodarone and clozapine; however LNG significantly decreased growth at 86 ng/L. Therefore, the most potent pharmaceutical tested was the synthetic progestin LNG with survival and growth impacts at concentrations less than 1 μg/L. Further analysis was conducted by measuring specific endocrine related mRNA transcript profiles in FHM larvae following the 28 d ELS exposure to LNG. Transcripts of 3β-HSD, 20β-HSD, and FSH were significantly down-regulated following 28 d exposure to both 16.3 and 86.9 ng/L LNG. Also, CYP19a expression was significantly down-regulated at 86.9 and 2392 ng/L LNG. Subsequently, a second study examined time periods that may be most sensitive (e.g., windows of sensitivity) for FHM larvae exposed to LNG. Larvae were exposed to a single concentration of LNG (i.e. LOECgrowth of 86.2 ng/L as determined in the 28 d ELS study) for different time periods starting with fertilized egg through 28 dph. Growth and mRNA expression of the four differentially expressed transcripts from the first study were measured. Regardless of the duration of exposure, LNG significantly decreased growth in fathead minnow larvae at day 28. For both 20β-HSD and CYP19a, mRNA expression was decreased following exposure to LNG; however, these transcripts returned to baseline levels after removal of LNG. 3β-HSD and FSH showed similar trends after exposure to LNG with 7-14 d and 14-28 d exposures exhibiting a decrease in expression; however, FSH expression returned to baseline once removed for LNG exposure. Based on these data, 3β-HSD was the only transcript to remain down regulated after LNG exposure. Together these data suggest LNG can negatively impact FHM larval survival and growth, with significant alterations in endocrine related responses. However, these changes in endocrine related responses may not directly correlate to the changes in growth demonstrated with LNG exposure to fathead minnows. Therefore, additional research is warranted to ascertain additional mechanisms, either endocrine related or non-endocrine functions, related to changes in growth of larval fathead minnows.

▼ The overall goal of this research was to verify the involvement of membrane progestin receptor alpha (mPRα) in mediating progestin-stimulated sperm hypermotility in the Atlantic croaker and southern flounder. Sperm motility in Atlantic croaker and southern flounder were tested with both the endogenous progestin, 17,20β,21-trihydroxy-4-pregnen-3-one (20β-S) or the selective mPRα agonist, 10-ethenyl-19-norprogesterone (Org OD 02-0). In croaker, the Pi3k/Akt/Pde and ErbB2/Mapk intracellular signaling pathways were examined. The role of mPRα in mediating sperm hypermotility and fertility in southern flounder was also studied. The effects of seasonal hypoxia on sperm motility in croaker were investigated in a field study in the northern Gulf of Mexico in the fall of 2010. Finally, the effects of acidified activator solution (simulating ocean acidification) were studied in the laboratory. In vitro, Org OD 02-0 mimicked the stimulatory actions of 20β-S in inducing sperm hypermotility and intracellular signaling cascades in croaker and flounder sperm, indicating that mPRα is the mediator of progestin signaling in the sperm of these species. In croaker sperm, both the Pi3k/Akt/Pde and ErbB2/Mapk intracellular signaling pathways were shown to be important mediators of progestin-induced sperm hypermotility, suggesting novel functions of G [subscript olf] βγ-subunits in teleost sperm. In flounder sperm, mPRα was shown to be important in mediating sperm hypermotility as only high motility sperm with high expression of mPRα were responsive to progestin stimulation, resulting in higher fertilization success compared to low motility sperm. A single LHRHa injection resulted in increased sperm motility and fertility, associated with an increase in mPRα expression in the sperm plasma membrane. The results also suggest that the mPRα/Acy/cAMP pathway first described in croaker sperm is present in flounder sperm. Field studies of male Atlantic croaker exposed to chronic seasonal hypoxia showed that hypoxia exposure resulted in smaller gonads, lower spermatogenesis, reduced testicular mPRα expression, and in some sites, reduced sperm motility. Studies with croaker sperm using acidified activator solution to simulate ocean acidification indicated that croaker sperm were sensitive to environmental insult. Furthermore, the results suggested that the progestin signaling mechanism is more sensitive to changes in ocean pH levels than the mechanism that controls sperm motility.

▼ O experimento 1 verificou o efeito de um protocolo de indução e sincronização de estros em novilhas sobre a cronologia das parições e a repetição de crias como primíparas. 194 novilhas Hereford e Braford foram divididas em 2 grupos: Tratamento, submetido a um protocolo misto de observação e sincronização de estros e inseminação artificial a tempo fixo (IATF); e Controle, submetido ao manejo convencional de inseminação artificial (IA). Ambos grupos foram submetidos a repasse por touros. O Experimento 2 avaliou o efeito de um pré-tratamento com norgestomet (NOR), prévio a um protocolo de sincronização para IATF em novilhas, sobre as taxas de prenhez, assim como os efeitos de características relacionadas com a reprodução. 74 novilhas Angus e cruzas, 18 e 24 meses de idade, com peso médio de 276 kg, condição corporal (CC) mínima de 3 (1-5) e avaliadas por ultra-som para o status ovariano, onde utilizaram-se somente novilhas 2 e 3 (1-3). Pesagens a cada 30 dias, desde os 60 dias anteriores aos serviços. Coletado sangue para dosagem de progesterona. Os animais foram divididos em 2 grupos: Tratamento, que recebeu um implante com 6 mg de NOR, na orelha direita, permanecendo por 14 dias; e Controle, que não recebeu implante. Dez dias após a retirada dos implantes, todas as novilhas foram a um protocolo para IATF, com uma injeção de GnRH, 7 dias depois uma aplicação de prostaglandina (PGF) e 48 horas mais tarde, as IATF e outra aplicação de GnRH. Quatorze dias após iniciou-se repasse com 2 touros, por mais 45 dias. O Experimento 3 avaliou o efeito da suplementação com progestágeno (acetato de medroxi-progesterona; MAP) ou progesterona, anteriormente a um protocolo para IATF, e verificou o efeito de características associadas à reprodução de novilhas, e de distintos manejos alimentares sobre as taxas de prenhez. Utilizaram-se 102 novilhas Angus, cruzas Angus e Braford, 22-24 meses, peso médio 241 kg e CC mínima de 2,5. Pesagens a cada 30 dias, a partir dos 60 dias anteriores aos serviços e ultra-som para o status ovariano (1-3). Quarenta e cinco dias antes da estação foram divididas em 2 grupos, Sem Suplementação, com 27 novilhas exclusivamente sobre pastagem natural, e Com Suplementação, com 75 novilhas consumindo 1% do peso em concentrado. Coletas de sangue para dosagem de progesterona. Grupos novamente divididos em 3, para receber dispositivo com progesterona, ou uma esponja intravaginal com MAP, ambos por 10 dias. O grupo Controle não recebeu nenhum tratamento. Na retirada dos implantes, todas as novilhas receberam PGF, e 4 dias depois, foram a um protocolo para IATF, com 2 mg de benzoato de estradiol na colocação de uma esponja com MAP. Sete dias depois, a esponja foi retirada, sendo aplicada PGF, seguindo-se nova aplicação de BE 24 horas depois, e IATF 52-56 horas após a PGF. Observação de estros 2 vezes por dia, desde a PGF até a IATF. Passados 14 dias, iniciou repasse por 4 touros, durante 45 dias. ▼ Abstract: The decrease in bone mineral density (BMD), weight gain and changes in uterine bleeding patterns are often associated with the use of progestin-only methods. Aspects not still elucidated and the moment of occurrence could harm the continuation and provoke premature discontinuation or induce morbidities. Objectives: To evaluate BMD, weight gain and discontinuation rates for bleeding disturbances of progestin-only methods. Subjects and Methods: Four studies were conducted with depot medroxyprogesterone acetate (DMPA) users: A) regarding BMD and body composition (BC) from 12 months to 23 years of use; B) on weight gain among DMPA users, the levonorgestrel-releasing intrauterine system (LNG-IUS) and cooper-intrauterine device (IUD) up to 10 years of use; C) on the influence of the etonogestrel-releasing implant (ENG) upon BMD and BC up to 24 months of follow-up; and D) to evaluate the influence of two counseling strategies regarding to bleeding disorders in continuation rates of the users of the LNG-IUS, ENG-implant and IUD. Results: BMD after 12 months of DMPA use was lower at the lumbar spine compared to IUD users and 29.8% has osteoporosis among those women who had used DMPA for 10 years or more compared to 2.4% of IUD users. Regarding BC, at 12 months of use it was observed, an increase of 2 kg of fat mass and 2% in the percentage of fat mass in DMPA users; however, in the long-term use, there was no difference in the amount of fat mass compared to IUD users. The weight increased at the end of the first year was 1.3kg, 0.7kg and 0.2kg and, at 10 years, was 6.6kg, 4.0kg and 4.9kg among DMPA-, LNG-IUS- and IUD-users, respectively. Users of the ENG-implant showed a decrease in BMD at lumbar spine after 12 months of use and an increase of 2% to 2.7% of fat mass at 12 and 24 months when compared to IUD-users. Women who received routine or "intensive" counseling about the expected bleeding patterns showed no significant differences regarding the rates of discontinuation of LNG-IUS, ENG-implant and IUD. Conclusions: Users of DMPA showed a decrease in BMD at the end of the first year of use, the decrease was progressive and an increased prevalence of osteoporosis in the long-term use was observed. DMPA, LNG-IUS and IUD users showed weight gain after 10 years of use, higher in DMPA users. Fat mass increased in the first year of DMPA use; however, was not significant in the long-term when compared to IUD-users. In ENG-implant users it was found a weight gain and increase of fat mass at 24 months and a reduction in BMD after 12 months of use. The routine and "intensive" counseling showed no differences in rates of continuation in ENG-implant, LNG-IUS- and IUD-users

▼ Eight postpartum bitches diagnosed with subinvolution of placental sites (SIPS) were followed in this retrospective study to investigate the efficacy of treatment with low oral doses of
progestins in stopping bloody vulvar discharge. Two other bitches diagnosed with SIPS were included in this study not receiving treatment.
A progestin derivate was administered orally once a day in low dose for two weeks, with half the daily dose reduced the second week. Cessation of vulvar discharge was seen in all eight treated dogs within this two week treatment period. The untreated dogs persisted with the problem respectively until a few weeks before and till onset of the next proestrus. No adverse effects on future heat, fertility, gestation and parturition was found in both the treated and untreated dogs. The results of this study indicate that oral low dose progestin therapy is effective and probably safe in treatment of SIPS.

Faculdade de Ciências Farmacêuticas de Ribeirão Preto Universidade de São Paulo, Ribeirão Preto, 2011. Introdução: Mais de 100 milhões de mulheres no mundo fazem uso de métodos contraceptivos hormonais. Apesar de seu desejado efeito na contracepção, sua metabolização ocorre no fígado estimulando a síntese de proteínas plasmáticas, dentre elas, as que controlam os sistemas de coagulação e fibrinólise, conferindo um estado de hipercoagulabilidade em suas usuárias. A literatura aponta também alterações no metabolismo dos lipídios e carboidratos, além de modular a expressão de moléculas de adesão presentes no endotélio. Para combater os efeitos indesejáveis destes medicamentos, baixas concentrações de estrógenos e diferentes progestágenos foram introduzidos, pois, a estes últimos é conferida a atividade anti estrogênica destas formulações. Estudos independentes sugerem que progesteronas de terceira e quarta geração possuem atividade anti-estrogênica menor em relação as de segunda geração. Objetivos: avaliar a ocorrência de alterações hemostáticas, analisar o perfil lipídico e quantificar moléculas de adesão no soro ou plasma da população feminina brasileira usuária de contraceptivos orais de segunda e quarta gerações. Materiais e Métodos: 70 participantes distribuídas em quatro grupos a saber : grupo I (controle- 20 pacientes); grupo II (DRSP/30EE- 20 pacientes); grupo III (DRSP/20EE- 16 pacientes) e grupo IV (LNG/30EE-14 pacientes). Foram avaliados os seguintes parâmetros: TP, Fator VII, TTPA, Fator XII, Fibrinogênio, Fator 1+2, Proteína C, Proteína S, Antitrombina, D dímeros, PAI 1, VCAM, ICAM, E- selectina, HDL, LDL,VLDL, Colesterol total e Triglicérides. Resultados e discussões: grupo II promoveu diminuição em TP e Prot. S; e aumento em HDL,VLDL,Col. total e TG; grupo III diminuiu TP, TTPA, Prot.S, ICAM e VCAM ; e aumentou Fibrinogênio, D-dímeros, HDL,VLDL,Col. total e TG e grupo IV diminuiu TP, Prot.C e aumentou ICAM e VCAM. Conclusões: Dos medicamentos estudados apontamos que: o grupo II promoveu alterações significativas no perfil lipídico caracterizando um estado pró-trombótico, apesar de apresentar o maior aumento nos valores de HDL e poucas alterações associadas à hipercoagulabilidade; o grupo III promoveu o maior número de alterações hipercoagulantes, alterou também perfil lipídico contribuindo para um estado pró- trombótico, embora tenha apresentado proteção endotelial e o grupo IV foi o medicamento que promoveu melhor proteção endotelial, não alterou perfil lipídico e ocasionou poucas alterações associadas à hipercoagulabilidade

▼ Le rôle des progestatifs et du récepteur de la progestérone (PR) dans la carcinogénèse mammaire est maintenant établi. Le PR est exprimé sous deux isoformes PRA et PRB dont l'expression est équimolaire mais qui différent par leur activité transcriptionnelle des gènes cibles. La surexpression de PRA est associée à un pronostic défavorable du cancer mammaire, et est observée chez les femmes à haut risque génétique de cancer du sein. L'utilisation d'antagonistes pour inhiber l'activité de PR pourrait constituer une stratégie thérapeutique potentielle. L'objectif de cette thèse a été d'évaluer les effets de l'ulipristal acétate (UPA), un anti-progestatif, sur la tumorigenèse mammaire dans deux modèles d'études complémentaires. In vitro, dans la lignée cancéreuse mammaire bi-inductible (MDA-iPRAB) développée au laboratoire. Nous avons démontré qu'UPA inhibe la prolifération cellulaire induite par la progestérone en présence de PRA. L'expression de certains gènes clés de la tumorigenèse mammaire, cibles de PRA a été étudiée. Une corrélation entre avec la prolifération cellulaire et l'expression du facteur anti-apoptotique BCL2-L1 (messager et protéine) a été démontrée. L'activation transcriptionnelle, dépendante de la progestérone et inhibée par l'UPA, s'accompagne d'un recrutement spécifique et sélectif de PRA sur des séquences régulatrices du gène BCL2-L1, comme le démontrent les expériences de ChIP. In vivo, nous avons évalué l'efficacité anti-tumorale de l'UPA ainsi qu'une nouvelle classe d'antagoniste sélective et passive de PR, les « APRns » dans un modèle murin de xénogreffe de tumeur mammaire humaine HBCx34. UPA ralentit la croissance tumorale, diminue la prolifération cellulaire (Ki67, PCNA) et inhibe l'expression de BCL2-L1. L'ensemble de nos résultats démontrent une action antiproliférative et apoptotique de l'UPA ce qui suggère une utilisation potentielle d'antagonistes de PR dans la prise en charge du cancer du sein.
The role of progestins and the progesterone receptor (PR) in breast tumorigenesis has now been elucidated. PR is expressed as two isoforms PRA and PRB, differing by their structure and their transcriptional activity. They are often co-expressed in normal breast tissue but the predominance of PRA expression is associated with mammary carcinogenesis, a bad prognosis and an endocrine-resistance. Antiprogestins inhibit mammary tumor growth in several experimental models. The aim of this thesis was to investigate the effect of a well-known selective PR modulator (SPRM), Ulipristal acetate (UPA) in two complementary breast cancer models. In vitro, we used the newly established bi-inducible breast cancer cell line “MDA-iPRAB”, and demonstrated an anti-proliferative activity of UPA in progesterone-dependent, PRA-specific MDA-iPRAB cell proliferation. ▼ The oestrous cycle of the domestic bitch is unique with regard to its considerable length compared with that of most other domestic animals. A non-seasonal anoestrus, with a duration that may last from 2 to 10 months, follows each oestrous cycle. The anoestrus can be prolonged by oestrus-preventing drugs such as progestins. The general aim of the studies in this thesis was to get further insight into the endocrinology of the physiological and the progestin-induced canine anoestrus. Studies performed throughout recent years have made clear that during the course of canine anoestrus many endocrinological changes take place at the hypothalamic, pituitary, and ovarian level. Apart from these changes, there are indications of involvement of dopaminergic influences. The results of the study described in Chapter 3 provide forceful evidence that the bromocriptine-induced shortening of the interoestrous interval is not triggered by a decline in plasma prolactin concentration. The results of the study described in Chapter 4 provides additional evidence that the premature onset of oestrus brought about by a dopamine agonist must be due to some other dopamine-agonistic effect, probably increased FSH secretion. The study in Chapter 5 is a report on the effects of GnRH administration on the plasma concentrations of reproductive hormones in intact and OVX bitches. The data presented in this study demonstrate that provocative testing of the pituitary-ovarian axis using GnRH may be helpful in differentiating between bitches in anoestrus and neutered bitches. The study reported in Chapter 6 describes the basal and GnRH-induced plasma concentrations of FSH and LH in four anoestrous and four OVX bitches. In this report indications were found that measurement of the plasma FSH concentration in a single plasma sample may prove reliable for determination of neuter status. Taken together, the results of the studies described in Chapters 5 and 6 provide a basis for the diagnosis of remnant ovarian tissue and verification of neuter status in the bitch. Chapters 7 and 8 are reports on the effect of MPA on canine adenohypophyseal function. The results of both studies demonstrate that treatment with MPA affects the hypothalamic-pituitary-ovarian axis. Oestrus, ovulation, and a subsequent luteal phase did not occur in any of the bitches during treatment with MPA. The prevention of oestrus by MPA cannot be ascribed to a significant reduction in circulating levels of either FSH or LH. On the contrary, during the first months of MPA treatment there was an increase in basal plasma FSH and LH, which may be due to a direct oestrus-preventing effect of MPA at the ovarian level. With continuing MPA treatment, basal plasma gonadotrophin concentrations returned to pretreatment levels and the pituitary FSH response to GnRH stimulation decreased, while several LH pulses were not accompanied by a significant FSH pulse, suggesting that MPA treatment attenuated pituitary FSH sensitivity to endogenous GnRH. ▼ Dois conjuntos comerciais, o \'ImmuChem Doubly Antibody Corticosterona 125I RIEd́a ICN \'Biomedicalsé o \'Coat-a-count Cortisol 125I RIEd́a DPC foram utilizados na mensuração dos metabólitos de glicocorticóides de fezes de jaguatiricas (Leopardus pardalis) desafiadas com ACTH com o objetivo de comparar os resultados obtidos por cada um dos conjuntos. Paralelamente foram mensuradas as progestinas dos animais desafiados visando detectar fontes extra-gonadais de progesterona. Dois tipos de extração de metabólitos fecais também foram testados. O conjunto comercial da ICN provou ser mais confiável na detecção de metabólitos fecais de glicocorticóides. As duas técnicas de extração obtiveram resultados semelhantes, demonstrando ser indiferente a escolha da técnica para a extração de metabólitos de glicocorticóides em fezes de jaguatirica. Existem fortes indícios da produção de progesterona pela adrenal de jaguatiricas submetidas ao desafio com ACTH.
Two commercial kits, the ImmuChem Doubly Antibody Corticosterona 125I RIE from ICN Biomedicals and Coat-a-count Cortisol 125I RIE from DPC were used on fecal glucocorticoids measurement of ocelots (Leopardus pardalis) submitted to ACTH challenge aiming to compare the performance of both kits. In another way progestins from the same animals submitted to ACTH challenge were measured aiming to find progesterone from extra gonadal sources. Two different fecal metabolites extraction protocols were tested as well. The ICN commercial kit had better results on glucocorticoids metabolites measurement. Both extraction protocols had similar performance and this data indicates that any both can be used for this purpose. There are strong evidences of adrenal secretion of progesterone after ACTH challenge.
