▼ During the summer trial lactating dairy cows were randomly assigned to four treatment groups. These groups included a control group that received no hormonal supplement, and three groups that received supplemental gonadotropin releasing hormone (GnRH) on either day 5, day 11, or both days 5 and 11 post insemination. The cows during the winter trial were randomly assigned to three treatment groups, a control, and cows receiving hormonal supplements on day 5, or days 5 and 11 post insemination. Blood samples and rectal temperatures were taken 9 and 2 days prior to insemination and every other day there after beginning on day 5 post insemination and ending around day 30. Pregnancy was checked on day 30 and then again between days 45 and 60 by ultrasonography. Daily maximum and minimum temperatures and humidity values were recorded in order to calculate the temperature-humidity index and determine the level of heat stress the cows were experiencing. Serum progesterone (P4) and cortisol concentrations from all samples were analyzed using RIA. Environmental information for the summer indicated that the cows experienced an overall mild heat stress by an average THI of 76, where as the winter environmental data showed an average THI of 43, within the thermo-neutral range. However, on individual days during the summer the THI reached values between 79 and 85, indicating that cows experienced medium heat stress. Serum P4 concentrations were data was analyzed between days 11 through 17 post insemination, the expected luteal phase, during the summer. Treatment groups were compared and indicated that the serum P4 concentration was greater in GnRH-D11 treated cows than control cows (P < 0.05) and than GnRH-D5 treated cows (P < 0.05). Concentrations of P4 between groups during the winter were also analyzed between days 11 and 22 post insemination, however the results were different. GnRH-D5 and GnRH-D5+11 were both greater than control concentrations, 6.87 ± 0.69 (P < 0.09) and 7.10 ± 0.56 (P < 0.05), respectively. Winter progesterone concentrations were greater than those of summer between treatment groups (P < 0.05), indicating that there was an effect of environment on hormonal levels. Analysis of cortisol concentrations during the summer indicated no significant difference between treatment groups, as was the same for winter groups, and there was no difference between seasons. Administration of GnRH appeared to have a beneficial effect on pregnancy rates during the summer between days 45 and 60. This was determined when all groups receiving the hormone supplement were combined (P < 0.05). In conclusion, the administration of GnRH to heat stressed cows increased P4 concentrations and appeared to have a positive effect on pregnancy rates. Advisors/Committee Members: Dr. C.S. O sucesso de um programa comercial de transferência de embriões depende principalmente da interação dos fatores que afetam a recolha embrionária, a gestação e a morte embrionária após a transferência. O objetivo deste trabalho consistiu na avaliação de alguns destes fatores, de forma a minimizar aqueles que possam ter uma influência negativa e permitindo também informar e aconselhar os proprietários dos animais de uma forma mais clara e correta. O presente estudo foi realizado na Central Equina de Reprodução, no estado de São Paulo, Brasil. Foram analisados os dados relativos a 2086 recolhas e 1090 transferências, efetuadas ao longo de duas épocas reprodutivas consecutivas. Neste trabalho, foi avaliada a influência na taxa de recolha embrionária, da época reprodutiva, do dia da recolha, do tipo de sémen (refrigerado ou congelado) utilizado na inseminação artificial e da raça da dadora. Relativamente às taxas de gestação e de morte embrionária, foi avaliada a influência da época reprodutiva, da idade do embrião, da ciclicidade (recetoras cíclicas e acíclicas) e do dia pós-ovulação em que a recetora se encontrava no dia da transferência. As variáveis foram analisadas pelo teste de Fisher ou Qui-quadrado em tabelas de contingência 2x2, sendo as diferenças consideradas significativas se p <0,05. O tipo de sémen teve uma influência significativa na taxa de recolha embrionária. Foi possível concluir que em éguas inseminadas com sémen congelado a taxa de recolha embrionária é menor (p <0,05), sendo por isso preferível utilizar sémen fresco ou refrigerado. A época reprodutiva afetou significativamente a taxa de gestação, tendo esta sido superior no ano de 2011-2012 (p <0,05). No entanto, em ambas as épocas reprodutivas, as taxas de gestação foram consideradas satisfatórias, sendo de 76,0% em 2010-2011 e de 81,8% em 2011-2012. Tanto a idade do embrião como a ciclicidade e o dia pós-ovulação da recetora não influenciaram as taxas de gestação nem as de morte embrionária. Estes resultados permitem concluir que é possível transferir embriões entre os 6 e os 10 dias de idade, utilizar éguas recetoras entre os 3 e os 9 dias pós-ovulação bem como éguas em anestro suplementadas com progesterona de longa ação, sem que isso afete o sucesso do programa de transferências.

The success of a commercial embryo transfer program depends mainly on the interaction of factors that affect embryo recovery, pregnancy and embryonic loss after the embryo transfer. A transferência de embriões (TE) - tecnologia reprodutiva de 2ª geração possui um sucesso comprovado, tendo actualmente uma expressão mundial significativa. O objectivo deste trabalho consistiu no estudo das taxas de gestação (TG) após transferência de embriões bovinos descongelados, usando gonadotrofina coriónica humana (hCG) ou o anti-inflamatório não esteróide, meloxicam, em novilhas da raça Frísia Holstein pertencentes a produtores de quatro ilhas do grupo central da Região Autónoma dos Açores. Apesar da amostra estudada ser reduzida (n=100), os resultados das TG parecem-nos aceitáveis para todos os grupos: grupo não tratado (n=48) (64,6%); grupo meloxicam dia 6 (n=15) (66,6%); grupo meloxicam dia 14 (n=19) (63,2%); grupo hCG dia 6 (n=18) (55,5%). Contudo, os dados não evidenciaram diferenças significativas entre as TG nos diferentes grupos de tratamento pelo teste exacto de Fisher bilateral, com p=0,907. Também não se encontrou qualquer evidência de associação entre “Grupo tratado” ou “Grupo não tratado” versus “Diagnóstico de gestação” pelo teste exacto de Fisher bilateral, com p=0,837. Os dados também não evidenciaram diferenças entre o tempo mediano de TE (7 min) entre novilhas gestantes e não gestantes, pelo teste de Mann-Whitney U, com p=0,850. Assim, apesar do estudo não revelar resultados estatísticos significativos, parece-nos admissível sugerir o uso de meloxicam como tratamento coadjuvante, sobretudo após a transferência de embriões cuja viabilidade esteja ainda mais comprometida que os embriões usados no presente trabalho. Em conclusão, o presente ensaio poderá ser o precursor de outros a serem realizados com uma amostra maior e eventualmente com um desenho experimental mais completo. Futuros estudos poderão incluir um maior número de grupos para análise e avaliarem a eventual diferença entre tratamentos, na perspectiva da obtenção de uma relação custo-benefício favorável com recurso à transferência de embriões de alto valor genético.

ABSTRACT - The effect of human chorionic gonadotrophin or meloxicam administration on the pregnancy rate after transfer of frozen-thawed Holstein bovine embryos to dairy heifers - Embryo transfer (ET) - 2nd generation of reproductive technologies has an approved success and a significative expression worldwide. The aim of this work was to study the pregnancy rate (PR) after ET using thawed bovine embryos injecting human chorionic gonadotrophin (hCG) or the non steroidal anti-inflammatory meloxicam in a group of Holstein Frisiean heifers belonging to farmers allocated in four islands of the central group of Azores autonomous region. Despite the reduced studied population (n=100), the results in PR seem to us acceptable to all groups: Untreated group (n=48) (64,6%); meloxicam day 6 group (n=15) (66,6%); meloxicam day 14 group (n=19) (63,2%) and hCG day 6 group (n=18) (55,5%). However, the data revealed no significant differences between the PR in the different treated groups with Fisher’s exact bilateral test…

▼ Um dos objetivos deste trabalho foi avaliar e comparar a taxa de prenhez de receptoras bovinas nulíparas, após transferência de embriões fecundados in vitro ou in vivo, associados ou não a um embrião partenogenético. Um total de 239 novilhas foram utilizadas, e as médias das taxas de prenhez aos 30 (TP30d) e 60 dias (TP 60d), foram comparadas em arranjo fatorial 8x2 entre G1 e G2 (fecundados in vitro), e em análise fatorial 3x2, entre G3 e G4 (fecundados in vivo). O delineamento utilizado foi: G1) transferência de 1 embrião fecundado in vitro (n=86); G2) transferência de 1 embrião partenogenético associado a 1 embrião fecundado in vitro (n=81); G3) transferência de 1 embrião fecundado in vivo (n=36) e G4) transferência de 1 embrião partenogenético associado a 1 embrião fecundado in vivo (n=36). A taxa de prenhez aos 30 dias para o G1 foi de 31,3% (27/86) e do G2 de 37,0% (30/81); P= 0,28. Para o G3, de 55,5% (20/36) e G4, de 75,0% (27/36); P=0,09. Não houve diferença (P=0,92) para taxa de prenhez aos 60 dias para o G1 em relação ao G2, com taxas de 24,6% (22/86) e 28,2% (23/81); respectivamente. Também não houve diferença (P=0,08) aos 60 dias para o grupo G3 em relação ao G4, com taxas de 44,4 % (16/36) e 61,1% (22/36), respectivamente. As perdas embrionárias no período foram de 14,81% para o G1, 33,34% para o G2, 25,00% para o G3 e 18,52% para o G4. As médias de peso ao nascimento dos animais pertencentes ao G1 e G2, foram de 26,3 Kg (±1,2; n=6) e 28,8 Kg (±2,1; n=7) respectivamente. As médias dos dias de gestação das receptoras dos grupos G1e G2 foram de 290 dias (±2,1) e 292,8 dias (±1,1) respectivamente. Para avaliar o embrião partenogenético em relação aos embriões fecundados in vitro após 9 dias de cultivo embrionário, foi determinado os resultados para as seguintes variáveis dependentes: taxa de clivagem, taxa de blastocisto, taxa de blastocisto eclodido, número de núcleos e freqüência do RNAm do IFN-τ em 6 sessões de produção embrionária. Os resultados foram analisados em fatorial 6x2. Foi observada diferença (P<0.05) nas taxas de clivagem, blastocisto e eclosão entre os tratamentos, houve diferença entre número de núcleos, 231 para os fecundados e 156 para os partenogenéticos (P=0.001), e para a expressão do gene do IFN-τ, os embriões partenogenéticos expressaram mais o gene do IFN em relação aos fecundados (P=0.001). Finalmente, Western blots foram realizados para identificar a proteína do IFN-τ bovino em embriões fecundados e partenogenéticos após 12 dias de cultivo. O objetivo do estudo foi avaliar a influência do benzoato de estradiol, associado ao CIDR, na sincronização do estro e da ovulação sobre a taxa de sincronização e prenhez em vacas de corte zebuínas lactantes no pós-parto. Foram utilizadas 226 vacas zebuínas, previamente submetidas a avaliação para condição corporal, divididas aleatoriamente em três tratamentos: MN - monta natural (n=77) com estação de monta de 21 dias; T1 (n=70) as fêmeas receberam um dispositivo intra-vaginal de liberação de progesterona (CIDR) e uma injeção de benzoato de estradiol BE (dia zero do tratamento). No dia 7, foi removido o CIDR e aplicado uma dose única de PGF2α. As vacas foram inseminadas 12 horas após a observação do estro; T2 (n=79) as fêmeas foram submetidas ao mesmo protocolo empregado no T1, adicionando outra dose de BE no dia 8 e todas as fêmeas foram inseminadas 56 horas após a retirada do CIDR. As fêmeas dos tratamentos T1 e T2 manifestaram o estro na proporção de 96,7 e 100,0% respectivamente, sendo que a taxa de prenhez foi de: 42,9, 18,3 e 42,7% para os tratamentos MN, T1 e T2 respectivamente. A concentração plasmática média de progesterona no dia 0 foi de 11,71 ng/mL e de 8,87 ng/mL, no dia 7. Não foi observado influência do escore de condição corporal (ECC) sobre as taxas de prenhez das vacas estudadas neste experimento. No presente estudo, a utilização da dose suplementar de BE, 24 horas após a retirada do implante contendo progestágeno foi o protocolo mais indicado para vacas de corte lactantes no período pós-parto.

The objective of this research was to evaluate the effect of estradiol benzoate plus CIDR on estrus and ovulation synchronization and pregnancy rates in zebu lactating cows at postpartum. Two hundred and twenty six zebu cows, were randomly assigned to three different treatments: MN (n=77) - cows were submitted to a breeding season of 21 days, cows of the T1 (n=70) received an intravaginal progesterone device (CIDR) and an injection of estradiol benzoate (EB) (day 0 of treatment). On day 7, the CIDRs were removed and the animals received an injection of a PGF2α. The cows were inseminated 12 hours after estrus detection. Females of the T2 (n=79) received the same protocol as T1, but plus another injection of EB on day 8 and were inseminated at 56 hours after the CIDR implant removal. The estrus induction rates for the treatments T1 and T2 were 96.7 and 100.0% respectively. The pregnancy rates were respectively 42.9, 18.3 and 42.7% for the MN, T1 and T2 groups. The progesterone plasma concentration were 11.71 ng/mL (day 0) and 8.87 ng/mL (day 7). The body condition score (BCS) did not affect pregnancy rates. Estradiol benzoate injected 24 hours after progestágeno removal gave a better response for lactating beef cows at post-partum.

▼ Assessment of trophic interactions between increasing populations of New Zealand fur seals (Arctocephalus forsteri) and fisheries in southern Australia is limited due to a lack of species specific demographic data and an understanding of the factors influencing population growth. To establish species specific demographic parameters a cross-sectional sample of New Zealand fur seal females (330) and males (100) were caught and individually-marked on Kangaroo Island, South Australia between 2000 and 2003. The seals were aged through examination of a postcanine tooth, which was removed from each animal to investigate age-specific life-history parameters. Annual formation of cementum layers was confirmed and accuracy in age estimation was determined by examination of teeth removed from individuals of known-age. Indirect methods of assessing reproductive maturity based on mammary teat characteristics indicated that females first gave birth between 4-8 years of age, with an average age at reproductive maturity of 5 years. Among reproductively mature females, age-specific reproductive rates increased rapidly between 4-7 years of age, reaching maximum rates of 70-81% between 8-13 years, and gradually decreased in older females. No females older than 22 years were recorded to pup. Age of first territory tenure in males ranged from 8-10 years. The oldest female and male were 25 and 19 years old, respectively. Post-weaning growth in females was monophasic, characterised by high growth rates in length and mass during the juvenile growth stage, followed by a gradual decline in growth rates after reproductive maturity. ▼ Assessment of trophic interactions between increasing populations of New Zealand fur seals (Arctocephalus forsteri) and fisheries in southern Australia is limited due to a lack of species specific demographic data and an understanding of the factors influencing population growth. To establish species specific demographic parameters a cross-sectional sample of New Zealand fur seal females (330) and males (100) were caught and individually-marked on Kangaroo Island, South Australia between 2000 and 2003. The seals were aged through examination of a postcanine tooth, which was removed from each animal to investigate age-specific life-history parameters. Annual formation of cementum layers was confirmed and accuracy in age estimation was determined by examination of teeth removed from individuals of known-age.Indirect methods of assessing reproductive maturity based on mammary teat characteristics indicated that females first gave birth between 4-8 years of age, with an average age at reproductive maturity of 5 years. Among reproductively mature females, age-specific reproductive rates increased rapidly between 4-7 years of age, reaching maximum rates of 70-81% between 8-13 years, and gradually decreased in older females. No females older than 22 years were recorded to pup. Age of first territory tenure in males ranged from 8-10 years. The oldest female and male were 25 and 19 years old, respectively.
