Opinnäytetyön tarkoituksena on tutkia mitä subjektiivinen elokuvakerronta on ja millä keinoin sitä luodaan. Työn ydin keskittyy tutkimaan POV-otosta sekä yleisesti subjektiivisen ja objektiivisen kuvan suhdetta. Työssä käydään läpi seikkoja, joita elokuvan pääkuvaajan täytyy ottaa huomioon niin kuvasuunnittelussa kuin teknisissä valinnoissaan, pyrkimyksenään luoda subjektiivista elokuvakerrontaa. Tämän pohjalta pohditaan millaista subjektiivinen ja objektiivinen kuvakerronta on. Työssä on keskitytty tekijöihin, jotka vaikuttavat näkökulman valintaan sekä siihen kuinka elokuvantekijä luo mahdollisimman vahvan yhteyden, samaistumisen, katsojan ja kuvattavan henkilöhahmon välille. Opinnäytetyön aineistona on käytetty sekä teoreettisia lähteitä, että käytännön elokuvaesimerkkejä. Opinnäytetyössä havainnollistetaan aihetta muun muassa tekijän opinnäytetyöelokuvan Metsästysmaan sekä taide-elokuvan Fall Downin kautta. Työn viimeisessä osiossa on tehty kaksi kohtausanalyysiä POV-otosta esimerkillisesti käyttävistä elokuvista. Tutkimusta tehdään sekä yleisesti, että kauhuelokuvagenren kontekstissa. Subjektiivinen otos antaa katsojalle näkymän henkilöhahmon ajatuksiin ja tuntetilaan. Subjektiivisen kuvan eri muodot jaetaan yleisesti viiteen kategoriaan, joita ovat POV-otos, takauma, puhdas mielikuva, kuvitteellinen- tai fantasiakerronta ja objektiivisen subjektivisointi. Subjektiivinen elokuvakerronta puolestaan muodostuu useammasta elementistä ja pelkkä subjektiivinen kuva ei vielä takaa subjektiivista kokemusta. Subjektiivinen suhde henkilöhahmoon voidaan saada aikaan myös objektiivisen kuvan keinoin, objektiivis-subjektiivisella otoksella. Vaikka elokuvan kuvakerronta koostuu sekä subjektiivisista, että objektiivisista otoksista, se on kuitenkin tavalla tai toisella aina subjektiivista. Elokuvantekijän tavoitteena on saada katsoja samaistumaan elokuvan henkilöhahmoihin. Samaistumiseen vaaditaan, että katsoja näkee henkilöhahmossa piirteitä, jotka ovat hänelle itselleen tuttuja tai sellaisia ominaisuuksia, joita hän toivoisi itsellään olevan. POV-otos ei yksinään välttämättä riitä samaistumisen saavuttamiseen, vaan katsojan on nähtävä myös ulkoisesti miltä henkilöhahmo näyttää ja kuinka hän käyttäytyy. POV-otoksessa katsoja jakaa henkilöhahmon kanssa elokuvallisen ajan ja paikan ja katsoja ei tiedä enempää eikä vähempää kuin itse henkilöhahmo. Sen ohella, että POV-otos näyttää mitä nähdään, se voi myös näyttää kuinka henkilöhahmo näkee. POV-otos on olellinen osa subjektiivista elokuvakerrontaa ja myös objektiivisen kuvan avulla voidaan luoda subjektiivinen näkymä henkilöhahmon sisimpään. Lopulta nämä kaksi yhdessä luovat kokonaisvaltaisimman subjektiivisen kokemuksen.

Opinnäytteeni tavoitteena oli luoda Tampereen Ammattikorkeakoulun uuden pelilaboratorion, eli Pelistudion, tulevaisuuden suunnittelu peli-sv:n opettajien ja opiskelijoiden toiveiden mukaan. Pelilaboratorion siirryttyä vuoden 2014 helmikuussa neljännen kerroksen neuvottelutilasta kolmannen kerroksen suurempaan luokkaan, saatiin enemmän tilaa tietokoneille, pelikoneille ja neuvotteluun. Pelistudion on tarkoitus avata ovensa vuoden 2014 syyslukukauden alkaessa. Opinnäytteeni oli mielipidetutkimuksen lisäksi tilasuunnittelua. Oli pohdittava tarkkaan, mitkä opettajien ja opiskelijoiden toiveista voitiin toteuttaa, sekä se, mitä pitää toteuttaa. Huomioitavana oli Pelistudion kyky toteuttaa sen tarkoitus peli-sv:n opiskelijoiden työpisteenä, sekä taata paikan mukavuus niin ergonomisesti kuin sisustukseltaan. Tilojen laatu oli vaihtelevaa. Pelikoneita oli tarpeeksi, mutta tietokoneita olisi voinut olla enemmän, ja tarpeetonta roskaa oli siellä täällä. Tietokoneissa ei myöskään ollut kaikkia tarvittavia ohjelmia, jättäen niiden käyttökyvyn kyseenalaiseksi. Motion Capture -tila, jossa oli tarkoitus tehdä liikkeitä peleihin, oli vasta alkutekijöissä Pelistudion mielipidetutkimukset järjestettiin aluksi vain opettajille ja opiskelijoille, mutta myöhemmin lisättiin mukaan pari muiden koulujen pelilaboratoriota. Peli-sv:n opiskelijoille lähetettiin mielipidekysymys Pelistudiosta. Seitsemän opiskelijaa vastasi kysymykseen, mutta he olivat kaikki käyneet Pelistudiossa aikaisemmin. Opettajien kanssa järjestettiin haastattelu, josta saatiin yksityiskohtaisemmin tietoa Pelistudion kehityksestä ja jo suunnitellusta tulevaisuudesta. Muiden pelilaboratorioiden ylläpitäjille lähetettiin kysely tiloista, työvälineistöstä ja toiminnasta. Tuloksista ja havainnoinneista tultiin siihen tulokseen, että vaikka Pelistudiossa oli jo paljon työvälineistöä, niin lisää sekä tarvittiin että toivottiin. Tietokoneita ja huonekaluja hankittaisiin lisää, roskat siivottaisiin pois ja luokka asennettaisiin WPK-verkkoon, jotta ohjelmia voitaisiin asentaa enemmän.

The purpose of my thesis was to create a plan for the future of the gaming laboratory of Tampere University of Applied Sciences, also known as the Gaming studio, according to the wishes of the Game Production programme students. After the gaming lab transferred from the meeting room in the fourth floor to the larger classroom in the third floor in February 2014, more space was available for computers, game consoles and conferencing. Gaming studio is supposed to open its doors at the start of the 2014 autumn term. In addition to an opinion poll, my thesis was about space planning. Careful consideration was needed on the matters of what wishes of the teachers and students could be realized and what else needed to be actualized. Gaming studio’s ability to fulfill its purpose as the workstation for game production programme students and to ensure its convenience both ergonomically and interiorwise, had to be considered. Introdução - O carcinoma da bexiga é considerado o quarto tumor maligno mais comum e a oitava causa de morte por cancro. O tipo mais comum é o carcinoma urotelial da bexiga (CUB) correspondendo, aproximadamente, a 90% dos casos. O sistema de estadiamento TNM fornece estimativas úteis sobre o risco de progressão/recorrência da doença e sobrevida dos pacientes, mas a heterogeneidade da biologia tumoral conduz a variações significativas. A O-glicosilação aberrante é uma das mais comuns modificações pós-tradução associadas ao cancro. A utilização de biomarcadores (como antigénios carbohidratados associados a tumor - TACAs) poderá contribui para um melhor entendimento dos mecanismos biológicos subjacentes à progressão tumoral, especialmente no que se refere ao papel da glicosilação na evolução do carcinoma da bexiga. Materiais e Métodos - Neste trabalho, composto por amostras de 96 doentes com CUB de vários graus e estádios, foi avaliada a imunoexpressão dos antigénios glicosídicos sialil Tn (STn) e sialil T (S6T) e também do índice de proliferação Ki67, tendo sido correlacionada com os parâmetros clínico-patológicos em estudo, e com a sobrevida global. Resultados - Verificou-se que 39 casos tinham expressão negativa para o STn e 57 tinham expressão positiva. A expressão do antigénio glicosídico STn associou-se ao alto grau de diferenciação celular (p=0,012), estádio tumoral avançado (p=0,001), morte por cancro (p=0,030), metastização ganglionar (p=0,043) e a tumores com proliferação celular descontrolada (p=0,022). Foi, também, verificado que a sobrevida global nos doentes com tumores que expressavam o STn era significativamente menor (p=0,034). Neste trabalho, o antigénio S6T não se mostrou informativo. Relativamente ao Ki67, os resultados mostraram que este marcador se correlaciona com o estádio de invasão e com grau de diferenciação. Conclusões - Neste estudo, o antigénio STn revelou-se um bom marcador de diagnóstico e subsequentemente de prognóstico do cancro, podendo representar um alvo terapêutico útil na imunização de doentes com CUB. ▼ A terapia anti-cancro baseada em anticorpos monoclonais tem vindo a ser atractiva na medida em que os anticorpos têm a capacidade de reconhecer especificamente antigénios alvo associados a cancro e de serem reconhecidos por células do sistema imunitário. Um dos antigénios que tem vindo a ser estudado é o glicano sialil-Tn (STn), presente na maioria dos carcinomas humanos e aproximadamente em 30% dos casos de cancro da mama. O objectivo geral deste trabalho foi o desenvolvimento de novos anticorpos terapêuticos contra STn. Especificamente, foi estudada a imunogenicidade de diferentes antigénios baseados em STn na produção desses anticorpos. Através da análise dos soros, verificou-se uma resposta imune com produção de anticorpos pelos murganhos imunizados, em separado, com mucina1 decorada com STn, mucinas de origem animal STn+ e lisados celulares STn+. Contrariamente, a imunização com STn associado a treonina ou poliacrilamida não desencadeou nenhuma resposta imunológica. Assim, percebeu-se que o STn é muito pouco imunogénico, e que a imunogenicidade deste depende do seu acoplamento a proteínas (mucinas) e da densidade do glicano a decorar as mesmas. Os sobrenadantes da cultura dos hibridomas obtidos pela fusão celular foram analisados por citometria de fluxo. Embora, se esperasse obter hibridomas produtores de anticorpos anti-STn, isto não se verificou. Uma das hipóteses levantadas está relacionada com a tolerância imunológica induzida por este antigénio, tal como verificado anteriormente pelo nosso e outros grupos de investigação. Assim, novas imunizações estão a ser realizadas com alterações relevantes no protocolo. A segunda parte deste trabalho consistiu na criação (transdução lentiviral) e análise (citometria de fluxo e RT-PCR) da linha celular fluorescente MCF-7/GFP STn+, que permitirá analisar o mecanismo de acção dos anticorpos anti-STn futuramente produzidos. A realização deste trabalho permitiu optimizar diversas técnicas e passos-chave importantes para o desenvolvimento de anticorpos terapêuticos anti-STn para cancro da mama ou outros cancros que expressem STn. This dissertation analyzes the SV/VS order variations in inaccusative verb constructions in 18 (eighteen interviews, taken from The Basic Corpus of the Rural Afro-Brazilian Dialect, in the State of Bahia, trying to reconcile a variational socio-linguistic approach (cf. LABOV, 1972) with a theoretical generative apparatus (cf. CHOMSKY, 1981; 1986). The analyzed corpus deals with three communities in different parts of the State of Bahia, in which slave workers were settled: ?Helvécia,? in the far south of the State; ?Cinzento,? in the semiarid; and ?Sapé,? on the environs of the All Saints Bay. As object of this study, the perspective of one of the orders ? the VS order ? is taken, aiming to characterize the linguistic and extra-linguistic factors that condition it, the latter ones in function of the external motivations (social-historical), linked to these rather isolated rural Afro-Brazilian communities. The statistical analysis shows that there are syntactic and semantic-discourse factors, relevant to the VS order, predominating over the inaccusative verb nature, in relation to the argument it selects. This analysis concludes the SV/VS orders are not exactly a case of variation, but a phenomenon of heterogeneous configuration, in complementary distribution.

Esta dissertação analisa a variação da ordem SV/VS em construções com verbos inacusativos de 18 (dezoito) entrevistas, extraídas do Corpus Base do Dialeto Rural Afro-Brasileiro, da região da Bahia, procurando conciliar uma abordagem sociolingüística variacionista com um aparato teórico gerativista. O corpus analisado reúne três comunidades localizadas em regiões diferentes do estado da Bahia, em que se fixou mão-de-obra escrava: Helvécia, no extremo-sul do Estado; Cinzento, no semi-árido; e Sapé, no Recôncavo Baiano. Assume-se, como objeto de estudo, a perspectiva de uma das ordens, a ordem VS, procurando caracterizar os fatores lingüísticos e extralingüísticos que a condicionam, estes últimos em função de motivações externas (sócio-históricas), ligadas a essas comunidades rurais afro-brasileiras, vivendo em relativo isolamento. A análise estatística demonstra que há fatores sintáticos e semântico-discursivos relevantes no condicionamento da ordem VS, sobrelevando a natureza do verbo inacusativo em relação com o argumento interno que seleciona. Esta análise conclui que as ordens SV e VS não constituem propriamente um caso de variação, mas sim, um fenômeno de configuração heterogênea, em distribuição complementar.

O interesse por substâncias antioxidantes aumentou consideravelmente nos últimos anos, pois moléculas que diminuem a peroxidação de lipídeos podem minimizar ou inibir os danos oxidativos causados ao DNA, proteínas e carboidratos. Os flavonóides, um grupo de metabólitos secundários, amplamente distribuídos no reino vegetal, apresentam propriedades antiinflamatórias, antibacterianas, antivirais, antialérgicas e antitumorais, além de possuírem propriedades antioxidantes sobre radicais livres de forma a inativá-los e a prevenir os danos oxidativos em nível celular. Acredita-se que a atividade biológica dos flavonóides seja devido às suas propriedades antioxidantes. Devido à grande importância dada atualmente aos radicais livres e antioxidantes, existe uma forte demanda por técnicas analíticas capazes de identificar e quantificar esses dois grupos de compostos. Este trabalho descreve o estudo do poder anti-radicalar dos flavonóides canferol, fisetina, quercetina, quercitrina, miricetina e rutina frente à radiação UV e ao radical HO. O método empregado baseou-se na geração fotoquímica artificial do radical HO pela fotodecomposição do H2O2. Os flavonóides estudados foram fotoestáveis frente à radiação UV e eficientes no combate ao radical HO. A quercitrina foi o flavonóide com maior ação anti-radicalar, seguida da rutina, canferol, fisetina, quercetina e miricetina. A metodologia proposta possibilita a comparação quantitativa da ação anti-radicalar de antioxidantes com base na cinética de reação das espécies com o radical HO. Da mesma forma, o estudo in vitro da interação entre radicais HO e extratos de plantas com caráter antioxidante pode ser realizado pelo método proposto.

The interest in antioxidants has increased considerably in recent years as molecules that reduce lipid peroxidation may minimize or inhibit the oxidative damage caused to DNA, proteins and carbohydrates. Flavonoids, a group of secondary metabolites widely distributed in the plant kingdom, have properties of anti-inflammatory, antibacterial, antiviral, anti-allergic and anti-tumor, in addition to possessing antioxidant and free radicals to inactivate them and prevent oxidative damage at cell level. It is believed that the biological activity of flavonoids is due to its antioxidant properties. Due to the great importance given today to free radicals and antioxidants, there is a strong demand for analytical techniques able to identify and quantify these two groups of compounds. This work describes the study of anti-radical power of the flavonoids kaempferol, fisetin, quercetin, quercitrin, myricetin and rutin against UV radiation and HO radical. The method used was based on artificial photochemical generation of HO radicals by photolysis of H2O2. The flavonoids studied were photpstable towards UV radiation and efficient in the fight against HO radical. La mise au point de médicaments ciblant la neuroinflammation, une composante importante de la physiopathologie des maladies neurodégénératives, fait l’objet de nombreuses recherches. Dans ce travail de thèse, nous avons étudié les effets de deux molécules potentiellement anti-inflammatoires et neuroprotectrices : l’hémine, un inducteur de l’hème oxygénase 1(HO-1) et ; le C16, un inhibiteur de la protéine kinase activée par l’ARN (PKR) dans un modèle de neuroinflammation in vivo par injection intrastriatale d’acide quinolinique (AQ) chez le rat. Nos résultats ont montré que l’induction de la HO-1 produit des effets délétères tandis que l’inhibition de la PKR induit des effets neuroprotecteurs et antiapoptotiques. Ce travail a par ailleurs permis de décrire l’évolution cinétique de la neuroinflammation sur 90 jours dans le modèle AQ, la capacité du tissu cérébral à se régénérer après la lésion et l’intérêt de ce modèle dans l’étude des effets d’agents neuroprotecteurs administrés au long cours.

Neuroinflammation is a key part of the physiopathology of neurodegenerative diseases and is an interesting target in their treatment. In this PhD work, we studied the effects of two potentially anti-inflammatory and neuroprotective molecules, hemin and C16, in an in vivo rat model of neuroinflammation by intrastriatal injection of quinolinic acid (QA). We showed that heme oxygenase 1 (HO-1) induction by hemin has deleterious effects whereas inhibition of the protein kinase RNA activated (PKR) by C16 treatment induced neuroprotective and anti-inflammatory effects. Concurrently, we evaluated longitudinal evolution of neuroinflammation in our model. Results showed the kinetic of the inflammatory phenomena; the ability of cerebral tissue to recover integrity and the capability of this model to evaluate potential neuroprotective and anti-inflammatory drugs in a long-time study.

Bakgrund: Deep Brain Stimulation (DBS) i nucleus subthalamicus (STN) och i kaudala zona incerta (cZi) har visat positiva effekter på motoriska symtom (rörelseförmåga och balans) vid Parkinsons sjukdom. Efter DBS har dock negativa taleffekter noterats, inklusive nedsatt artikulatorisk precision och stabilitet. Nedsatt artikulatorisk precisionoch stabilitet påverkar klusilproduktionen mest vilket kan resultera i att klusilexplosionen uteblir eller att multipla explosioner uppstår. Mål: Att undersöka artikulatorisk precision och stabilitet vid klusilexplosionen efter DBS i STN respektive cZi. Metod: I studien deltog 19 patienter varav 9 deltagare stimulerats i STN och 10 deltagare i cZi. Talinspelningar gjordes innan operation och ett år efter operation med DBSstimulering på och av. Klusilerna i talmaterialet delades in i tre kategorier, de med en klusilexplosion, de med två eller flera klusilexplosioner samt de utan klusilexplosion. Hypotesprövning gjordes gällande fördelningen av klusiler med multipla explosioner samt klusiler utan explosion mellan och inom patientgrupperna samt inom respektive klusil. Resultat: Multipla explosioner ökade hos cZi-patienterna och minskade hos STNpatienterna. Klusiler som saknade explosion ökade hos STN-patienterna medan de minskade hos cZi-patienterna. För båda patientgrupperna ökade multipla explosioner och klusiler utan explosion då stimuleringen var på jämfört med av. Skillnaderna mellan och inom grupperna var inte signifikanta. Slutsats: Artikulatorisk precision och stabilitet påverkades efter DBS i form av bristande stabilitet, för kort slutningsrörelse och felaktig koordination av slutningsrörelsen. Detta resulterade i multipla explosioner och uteblivna explosioner med olika effekter för elektrodlokalisationerna.

Background: Deep Brain Stimulation (DBS) in Subthalamicus Nucleus (STN) and Caudala zona incerta (cZi) have shown positive effects on motor symptoms in Parkinson‘s disease. Negative effects on speech after DBS has been noted including reduced articulatory precision. Reduced articulatory precision and stability affects the production of stop consonants and as a result, loss of burst or multiple burst can occur. Aim: To investigate articulatory precision and stability regarding the burst in stop consonants after DBS of STN or cZi. Method: The study included 19 patients with 9 patients stimulated in STN and 10 patientsin cZi. Speech recordings were made before surgery and one year after with stimulation ON and OFF. The stop consonants were divided into three categories, those with one burst, those with two or more bursts (multiple burst) and those with loss of burst. Hypothesis testing was done on the categorization of the stop consonants in and between the groups and between the stop consonants types.
