▼ Although speech and language therapy practitioners commonly place great weight on standardised, static assessment, the procedures may not be fully representative, and reveal little about the child’s learning potential or the direction that intervention should take. Vygotsky’s theories, particularly his notion of the Zone of Proximal Development, underpin a range of approaches within the complementary assessment paradigm known as Dynamic Assessment (DA) (Vygotsky 1986). The term is used for assessments consisting of ‘active intervention by the examiners and assessment of examinees’ response to intervention” (Haywood and Lidz 2007 P1) The current project investigated the application of Dynamic Assessment to a population of children with previously identified Language Impairments. As in parallel studies of intelligence, both manifest skills of language, and underlying processes used in manipulating and constructing language as a tool, were elucidated. The contribution that such an assessment can make to extending the understanding of language impairment, and in devising intervention programmes was investigated. This thesis describes the development of a Dynamic Assessment task requiring implicit knowledge of syntactic structure. The construction of the procedure was a novel adaptation and combination of established DA methodologies that are described and evaluated in Chapter 1. The task, which is essentially a sentence anagram, comprised 12 items specifically selected to assess particular grammatical structures reported in Chapter 2 to be problematic in children with Specific Language Impairments (SLI). The details of the task construction are reported in Chapter 3. The measure was employed on 24 children aged 8-10, with identified language impairment, and the results are reported in Chapter 4. Inter-rater reliability of the test measure was 88%, and the sensitivity of the test to change over time was demonstrated. Information about participants’ ability to transfer learning between items, their ability to use less directive prompts, their strategy use, and their metalinguistic and metacognitive awareness was extracted, and reported to the speech and language therapists working with the children. Evaluation of the test is discussed in Chapter 5. The thesis also reports on an investigation of the role of the information derived from the DA in informing intervention programmes (Chapter 6). The same cohort of 24 children with SLI was randomly allocated to two groups. Reports from the DA were used to inform the ongoing language intervention of one of the groups of children. In Chapter 7 the outcomes of therapy from that group were compared to the outcomes of the group receiving regular intervention. Differences between groups were nonsignificant although the gains achieved by subgroups of children were predicted, and in particular children making little progress in their ongoing therapy were shown to derive most benefit from the modified intervention. ▼ In last 25 years, the relation between cardiac diseases and periodontal disease has been extensively studied. Among the approaches used with this purpose, there is the detection of periodontopathogens in both, oral cavity and atheroma lesions. The aims of this study were: 1) determine the prevalence of Aggregatibacter actinomycetemcomitans and Porphyromonas gingivalis in the subgingival biofilm of patients with cardiac diseases and with different periodontal conditions; 2) determine the prevalence the prevalence of A. actinomycetemcomitans and P. gingivalis in atheromatous lesions biopsies. Previous to the cardiac surgery, patients were submitted to oral clinical evaluation and the subgingival biofilm were collected with curettes and suspended in TE (pH 7.6), the biopsies of atheromatous lesions were obtained during the surgery and stored in TE (pH 8.0) at -20C until processing. The DNA of both samples (biofilm and biopsies) were extracted and submitted to polymerase chain reaction with 16SrRNA species-specific primers in order to detect the species above. The results showed that, in oral biofilm, A. actinomycetemcomitans and P. gingivalis were detected in 3 (14,28%) and 10 (47,62%) out 21 patients respectively. In atheromatous lesions, P. gingivalis could be detected in 5 (23,8%) and A. actinomycetemcomitans in 3 (14,28%) of 21 biopsies. These data suggest that eventhough the atheromatous lesions can harborthe periodontopathogens (A. actinomycetemcomitans and P. gingivalis) they are not always found together in atheromatous lesions and oral biofilm. p>Nos últimos 25 anos, a relação entre a doença periodontal e doenças cardíacas tem sido amplamente estudada. Entre as abordagens utilizadas com essa finalidade, existe a detecção de periodontopatógenos em ambos, cavidade oral e lesões de ateroma. Os objetivos deste estudo foram: 1) determinar a prevalência de Aggregatibacter actinomycetemcomitans e Porphyromonas gingivalis no biofilme subgengival de pacientes com doenças cardíacas e com diferentes condições periodontais; 2) determinar a prevalência de A. actinomycetemcomitans e P. gingivalis em biópsias de lesões ateromatosas. Anteriormente à cirurgia cardíaca, os pacientes foram submetidos à avaliação clínica oral e foram coletados amostras de biofilme subgengival com curetas e suspensas em TE (pH 7,6), as biópsias de lesões ateromatosas foram obtidas durante a cirurgia e armazenados em TE (pH 8,0) à -20C até o processamento. O DNA de ambas as amostras (biópsias e biofilme) foram extraídos e submetidos à reação de cadeia polimerase com primers espécies-específicos 16SrRNA a fim de detectar as espécies acima. ▼ Le parasite Plasmodium falciparum, responsable des formes graves du paludisme chez l'homme, est transmis par Anopheles gambiae, son principal vecteur en Afrique sub-saharienne. Les nouvelles stratégies de lutte contre la maladie visent à limiter ou à interrompre le développement du parasite chez le moustique vecteur, et il est donc nécessaire d'améliorer notre compréhension des interactions entre le vecteur, son environnement et le parasite. L'objectif de ce projet de thèse a été de caractériser la flore microbienne du vecteur An. gambiae en conditions naturelles de transmission, d'étudier le rôle des principales espèces bactériennes colonisant l'estomac du moustique sur le développement de P. falciparum et de mesurer l'influence des bactéries sur la réponse immunitaire des moustiques femelles et leur capacité à transmettre le parasite. Pour mener à bien ce projet, nous avons collecté des populations de moustiques sauvages au Cameroun pour la caractérisation de la flore microbienne, nous avons ensuite exposé des moustiques de la colonie de laboratoire Ngousso à des cultures bactériennes puis infecté ces moustiques avec des isolats naturels de P. falciparum. Notre étude a montré que les souches bactériennes naturelles de l'intestin du moustique Serratia, Pseudomonas et Escherichia réduisaient la prévalence et l'intensité de l'infection et que le degré d'inhibition variait selon les taxons bactériens et les porteurs de gamétocytes. L'analyse des flores bactériennes des différents épithéliums de l'insecte par pyroséquençage a révélé des similarités entre la flore intestinale et celles retrouvées dans les ovaires et les glandes salivaires pour un même moustique. Les analyses d'expression suggèrent que la régulation de l'expression des gènes l'immunité par les bactéries intestinales pourrait participer à la modulation de la réponse antiplasmodiale. Les mécanismes impliqués dans les interactions bactéries-Plasmodium-vecteur sont complexes et multifactorielle et la modélisation de l'ensemble des interactions qui permettent à P. falciparum d'accomplir son cycle chez le moustique vecteur sera nécessaire pour envisager de nouvelles méthodes de lutte efficaces et durables. p>Plasmodium falciparum, the parasite responsible for the severe form of malaria, is transmitted by Anopheles gambiae, its major vector in sub-Saharan Africa. ▼ O fósforo (P) é um elemento essencial para o desenvolvimento das plantas e precisa ser suprido em quantidades adequadas para obtenção de altas produtividades das culturas agrícolas. O conteúdo de P no solo e consequente perda deste para as águas superficiais são importantes fatores relacionados a eutrofização de ambientes aquáticos. Nesse trabalho foi obtido o Grau de saturação de P (GSP) em 29 solos tropicais altamente intemperizados e seus valores foram correlacionados com atributos químicos, físicos e mineralógicos dos solos. Antes de calcular o GSP, foram determinados parâmetros como Índice de sorção de P (ISP), Capacidade de sorção de P (CSP) e fator de escala ?. Os solos foram divididos em grupos, e os solos de cada grupo receberam doses diferenciadas de P para obtenção do ISP devido a variação na capacidade de adsorção,. As doses utilizadas foram 200, 500, 1000, 1500 e 3000 mg L-1 de P. Os períodos de reação avaliados foram 1, 3, 7, 21, 42, e 84 d. A porcentagem média de P adsorvida no final do período de contato (84 d) variou de 23% a 49% do P adicionado inicialmente. Os Latossolos, os Gleissolos e os Nitossolos apresentaram os maiores valores de ISP, enquanto os Argissolos e os Neossolos tiveram os menores valores. Os maiores valores foram encontrados nos solos com elevados teores de argila, matéria orgânica (MO) e óxidos cristalinos e mal cristalizados de Fe e de Al. O ISP foi maior no final do período de contato (84 d), o que realça a influência do tempo na adsorção do P. O fator de escala ? e o GSP foram calculados em todos os períodos de contato. Os valores de ? aumentaram e os de GSP diminuíram em função do tempo devido à maior interação do P com o solo. Os menores valores de GSP foram obtidos nos solos com alta capacidade de adsorção de P, ao passo que os maiores valores de GSP foram dos solos com menor capacidade de adsorção. O maior GSP (31%) foi obtido no Neossolo Litólico. O valor limite de GSP considerado nesse trabalho foi 23%. As relações entre GSP, CSP, ISP e alguns atributos dos solos foram abordadas por meio da análise de componentes principais. Com o agrupamento de todos os solos, os teores de óxido mal cristalizado de alumínio (Alox), argila, MO e de óxido cristalino de alumínio (AlDCB) foram os que melhor se correlacionaram com o ISP e a CSP. Avaliando somente os Latossolos, os teores de argila e de óxidos cristalinos e mal cristalizados de ferro e de alumínio (Feox, FeDCB, AlDCB e Alox) representaram os componentes principais ligados à adsorção de P. Com a adição dos demais solos, os teores de fósforo extraídos por solução ácida de oxalato de amônio (Pox), Alox, argila e MO formaram os componentes principais para ISP e CSP. O GSP não correlacionou significativamente com nenhum dos atributos. A partir do dendograma, os solos foram separados em dois grupos. O grupo I foi composto por solos pouco suscetíveis a perdas de P em função dos valores de CSP e de GSP. Já o grupo II reuniu os solos mais suscetíveis. Abstract In order to gain better understanding on the impacts of sewage treatment plant (STP) effluent on marine environment, the present study was undertaken to investigate the structure of benthic community and protein expression patterns of 4 benthic species in STP effluent areas in Kaohsiung, Taiwan. Six sites were selected, 3 near Jhong Jhou STP, 2 near NSYSU STP and one reference site. Samples were collected in December 2009 (winter) and July 2010 (summer) by bottom trawling. In winter, temperature was 26.8 °C; salinity and pH were 32. 2‰ and 8.1. In summer, environmental data were 33.7 °C, 31.5‰ and 8.0, respectively. In all, 5 phyla, 31 families and 48 different species were captured in two seasons. Proteomic approach by one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (1D SDS-PAGE) was conducted on 3 fishes, Leiognathus splendens, Apogon fasciatus, Engyprosopon multisquama, and one crab Portunus hastatoides. Environment characteristics and structure of benthic community had no significant difference among the 6 sampling sites. Protein expression patterns based on the four examined species also indicated no significant difference among sites. The complexity of pollutants in Kaohsiung Harbor is probably the main reason affecting all sampling sites. In other words, contaminated area in Kaohsiung Harbor may cover a wide range, at least III over the present sampling distance, i.e. 8km which causes no difference in community structure and protein expression pattern among control, Jhong Jhou STP and NSYSU STP sites. ▼ Thirteen South African medicinal plants which are used traditionally to treat symptoms associated with Listeria monocytogenes infections, were screened for activity against the pathogen. Different plant parts were extracted separately with ethyl acetate or chloroform. All the extracts were first screened against the bacteria using the disc diffusion method. Zones of inhibition observed in the presence of the chloroform extracts of Eucomis autumnalis, ethyl acetate extracts of Acacia karroo and Plectranthus ecklonii (50 mg/ml) were 12 mm, 14 mm and 15 mm respectively. Active extracts were further tested against the bacteria for minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) using the microtitre dilution method. Ethyl acetate extracts of A. karroo exhibited MIC of 3.1 mg/ml and MBC of 6.25 mg/ml. Ethyl acetate extracts of P. ecklonii showed MIC of 0.5 mg/ml and MBC of 1.0 mg/ml. Five samples namely A. karroo (ethyl acetate extract), P ecklonii (ethyl acetate extract), Senecio inonartus (ethyl acetate extract), S. inonartus (chloroform extract) and Aloe arborescens (ethyl acetate extract) showed good MIC against L. monocytogenes and a MBC range from 1.0 to 12.5 mg/ml. The two plants, A. karroo and P. ecklonii were further selected for the isolation of the active compound(s). Column chromatographic purification of ethyl acetate extracts of the leaves of A. karroo led to the isolation of three known pure compounds namely â-sitosterol, epigallocatechin and epicatechin. The MICs of the â-sitosterol and epigallocatechin that were isolated from A. karrroo were found to be 31.25 µg/ml and 62.5 µg/ml respectively against L. monocytogenes. The confocal scanning laser microscopy (CSLM) showed that the biomass of the listerial biofilms were reduced when the isolated compounds were added and slightly reduced when the crude extract was added. The aggregation of cells which were exposed to â-sitosterol and epigallocatechin was reduced from 25 µm as observed in untreated cells to < 10 µm in length. Therefore as one of the local South African plants identified in the present study, the pure compounds isolated from A. karroo, could be used as a potential natural alternative for eliminating L. monocytogenes biofilms from food processing surfaces. This could help in combating the problem of food contamination and food poisoning caused by the pathogen. It could also help in preparing antibiofilm agents that are cost effective and easily accessible to the public. A. karroo should be further be explored in this regard. The present study reports for the first time the isolation of the three compounds, â-sitosterol, and epicatechin and epigallocatechin from A. karroo. Bioassay-guided fractionation of the P. ecklonii ethyl acetate extract led to the isolation of two known compounds, parvifloron D and parvifloron F. Parvifloron D and F exhibited a minimum inhibitory concentration (MIC) of 15.6 and 31.25 µg/ml respectively against L. monocytogenes. ▼ Over the last three decades, the medical community has seen a drastic increase in the number of antibiotic resistant infections, particularly caused by the ESKAPE (Enterococcus faecium, Staphylococcus aureus, Klebsiella penumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa and Enterobacter spp.) pathogens. While all six species are known to cause an array of human disease, I have been primarily concerned with bacteremia and skin and soft tissue infections associated with A. baumannii, S. aureus, and P. aeruginosa. Bacteremia and skin based infections have the highest mortality rates and exhibit physiological differences compared to laboratory culture conditions. Indeed, the antibiotic resistant nature of the ESKAPE pathogens, combined with the dwindling antibiotic pipeline, has resulted in a need for new therapeutic options, thus my research has focused on the development of three new antimicrobial therapies. First, a topical ointment containing mupirocin and neomycin has been shown to have a wide spectrum of activity, with efficacy against S. aureus, A. baumannii, and P. aeruginosa, and has been shown to clear colonization of multi-drug resistant S. aureus. A second topical ointment containing compounds found to be effective against Pseudomonas biofilms also demonstrates efficacy against a murine wound model infected with A. baumannii, P. aeruginosa, and S. aureus. The third therapy was the identification and characterization of two compounds with activity as efflux pump inhibitors for A. baumannii and P. aeruginosa that could be used as adjunctive therapy in combination with conventional antibiotics. Seven genes that are likely to be the targets of these compounds have been identified, allowing for an early model of their cellular mechanism of action for future testing. All together the three newly described therapeutic candidates we have developed may provide new strategies for the therapeutic intervention of extremely antibiotic resistant bacterial species.

▼ Thirteen South African medicinal plants which are used traditionally to treat symptoms associated with Listeria monocytogenes infections, were screened for activity against the pathogen. Different plant parts were extracted separately with ethyl acetate or chloroform. All the extracts were first screened against the bacteria using the disc diffusion method. Zones of inhibition observed in the presence of the chloroform extracts of Eucomis autumnalis, ethyl acetate extracts of Acacia karroo and Plectranthus ecklonii (50 mg/ml) were 12 mm, 14 mm and 15 mm respectively. Active extracts were further tested against the bacteria for minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) using the microtitre dilution method. Ethyl acetate extracts of A. karroo exhibited MIC of 3.1 mg/ml and MBC of 6.25 mg/ml. Ethyl acetate extracts of P. ecklonii showed MIC of 0.5 mg/ml and MBC of 1.0 mg/ml. Five samples namely A. karroo (ethyl acetate extract), P ecklonii (ethyl acetate extract), Senecio inonartus (ethyl acetate extract), S. inonartus (chloroform extract) and Aloe arborescens (ethyl acetate extract) showed good MIC against L. monocytogenes and a MBC range from 1.0 to 12.5 mg/ml. The two plants, A. karroo and P. ecklonii were further selected for the isolation of the active compound(s). Column chromatographic purification of ethyl acetate extracts of the leaves of A. karroo led to the isolation of three known pure compounds namely â-sitosterol, epigallocatechin and epicatechin. The MICs of the â-sitosterol and epigallocatechin that were isolated from A. karrroo were found to be 31.25 µg/ml and 62.5 µg/ml respectively against L. monocytogenes. The confocal scanning laser microscopy (CSLM) showed that the biomass of the listerial biofilms were reduced when the isolated compounds were added and slightly reduced when the crude extract was added. The aggregation of cells which were exposed to â-sitosterol and epigallocatechin was reduced from 25 µm as observed in untreated cells to < 10 µm in length. Therefore as one of the local South African plants identified in the present study, the pure compounds isolated from A. karroo, could be used as a potential natural alternative for eliminating L. monocytogenes biofilms from food processing surfaces. This could help in combating the problem of food contamination and food poisoning caused by the pathogen. It could also help in preparing antibiofilm agents that are cost effective and easily accessible to the public. A. karroo should be further be explored in this regard. The present study reports for the first time the isolation of the three compounds, â-sitosterol, and epicatechin and epigallocatechin from A. karroo. Bioassay-guided fractionation of the P. ecklonii ethyl acetate extract led to the isolation of two known compounds, parvifloron D and parvifloron F. ▼ Abstract The present study is concerned with the mechanism of growth of porous anodic films formed on high purity aluminium and sputtering deposited aluminium over a wide range of current density between 5 to 50 mA/cm2 for times up to 5400 s in 24.5 wt % sulphuric acid and at temperatures of either 0 or 20 0C. The resultant films were examined using scanning electron microscopy (SEM), transmission electron microscopy (TEM), Rutherford backscattering spectroscopy (RBS), nuclear reaction analysis (NRA), optical interferometery, microhardness and nanoindentation.The RBS analysis enabled determination of the composition of the porous films, which was expressed as Al2O3.xAl2(SO4)3, with the sulphur species content increasing with increase in current density and decrease in temperature. The average expansion factor (expressing, the ratio of the film thickness to the oxidized aluminium thickness) increased between 5 mA/cm2 and 50 mA/cm2 for films formed at 0 0C, extending from 1.58 to 1.88 and from 1.57 to 1.78 according to SEM and TEM respectively. For films fabricated at 20 0C, the average expansion factor increased from 1.45 to 1.66 and from 1.42 to 1.67 derived from SEM and TEM respectively. The expansion factor increases as the current density increases for both temperatures, and decreases as electrolyte temperature for a given current density increases. The increase of expansion factor is also associated with a rise in the steady voltage during film growth. However, the film expansion does not depend on the anodizing time. The increase in expansion factor correlates with a small increase in the amount of sulphur in the film, which increases with rise of current density. The surface of the porous alumina revealed a network of furrows and ridges, reflecting the pattern of the cellular textures on the topography of the elecropolished aluminium. The retention of topography indicates that the thinning of the film due to chemical dissolution by the electrolyte is negligible, although softening of the film toward the film surface increases with rise of electrolyte temperature and anodizing time as determined by microhardness measurements on film cross-sections. For films fabricated at 0 0C, nodules appeared with a low population density on the film surfaces formed at 20 mA/cm2 for 5400 s, and a locally high population density, but non-uniform distribution, for films formed at 30 to 50 mA/cm2 for a wide range of anodizing times. NRA determined the oxygen concentrations in the films, from which the efficiency of the film grown was derived. The efficiency showed a correlation with the expansion factor, with values increasing with rise of current density and with decrease in the anodizing temperature, ranging from 72 % to 87 % between 5 mA/cm2 and 50 mA/cm2, for an electrolyte temperature of 0 0C, and between 66 % to 75 %, for the same range of current density, for an electrolyte temperature of 20 0C. ▼ Some filamentous fungi of the genera Aspergillus and Penicillium are of high impact for food safety due to their ability to produce a toxic secondary metabolite, ochratoxin A (OTA). OTA is well known for its carcinogenic, immunosuppressive and teratogenic properties. OTA has also been extensively documented as a contaminant of a wide variety of foods including cereals, green coffee, spices, nuts, dried fruits, beer, wine, grapes, and grape juice. Following, a literature review on OTA and existing knowledge of three main ochratoxigenic fungi (Aspergillus westerdijkiae, A. carbonarius and Penicillium verrucosum) the objectives of the present thesis were: i) to study the effect of different ecophysiological factors such as pH, aw, microstructure (intrinsic factors), temperature (extrinsic factor), and initial inoculum level (implicit factors) on fungal growth and OTA production, ii) to develop kinetic or stochastic models by using the primary data in order to predict fungal growth and OTA production and validate the aforementioned models on different foodstuffs, iii) to correlate different evaluation methods regarding fungal growth (colony diameter, dry fungal biomass, and spores population) and OTA production (ELISA and HPLC analysis), and iv) to study potential biological (e.g., microorganisms- bacterial and yeast composites) or alternative methods (e.g., essential oils- vapor phase and enclosed in edible coatings) for fungal inhibition and OTA reduction. At preliminary stage (Chapter 3), the potential of the ELISA method in the determination of the produced OTA by A. westerdijkiae, A. carbonarius and P. verrucosum in Malt Extract Agar at different pH (3.9, 5.1, 5.9 and 6.8), aw (0.87, 0.93 and 0.99), and temperature (10, 15, 20, 25, 30 and 40°C) levels was evaluated, providing a rapid screening for the optimum and marginal conditions of OTA production. Moreover, selected samples were analyzed for OTA by HPLC in order to verify ELISA method. The knowledge of optimal and marginal levels of ecological factors in order to optimize post-harvest and storage of food products may significantly affect the production of OTA. Following this screening, the study was focused on food microstructure (e.g., liquid, semi-liquid, solid) (expressed as % w/v gelatin) and how it may affect growth and OTA production of A. carbonarius in combination with factors such as aw, pH and temperature (Chapter 4). The primary data were used in order to develop secondary (kinetic) models for growth and OTA production rates by A. carbonarius. Finally, the performance of the developed models was evaluated in food matrices of different viscosity such as jelly, custard and marmalade. The findings of this chapter may provide a basis for reliable assessment of the risk of fungal growth and OTA production in foods of different structural and rheological properties. ▼ As populações de aves antárticas têm sido estudadas e consideradas indicadoras da qualidade do ecossistema marinho, especialmente dos oceanos do sul, ao longo dos últimos 50 anos. Existem cerca de 40 espécies de aves marinhas que se reproduzem em áreas descobertas de gelo. Dentre as aves marinhas antárticas, os pinguins são os que têm a maior representatividade ecológica e são considerados espécies sentinelas para o estudo das mudanças ambientais nesse continente. Esses animais representam 90% da biomassa de aves nos oceanos do sul, e suas colônias estão distribuídas nas ilhas antárticas e subantárticas bem como sobre o Continente Antártico. Há três espécies de pigoscelídeos mais representativos desta biomassa, são eles: Pygoscelis papua (Pinguim gentoo), Pygoscelis adeliae (Pinguim-de-adélia) e Pygoscelis antarcticus (Pinguim-de-barbicha). Os pinguins antárticos estão entre as aves de menor contato com humanos, o que os torna possíveis indicadores da presença natural de genes de resistência a antimicrobianos na microbiota intestinal de aves e no ambiente. O objetivo desta dissertação foi avaliar a presença de resistência a antimicrobianos na microbiota intestinal de P. antarcticus e de P. papua, e compará-las à microbiota de Spheniscus magellanicus (pinguins-de-magalhães). Os S. magellanicus habitam a Argentina, Chile e Ilhas Malvinas, locais em que há variadas atividades humanas. Foram coletadas amostras de fezes aparentemente frescas de 46 pinguins P.antarcticus e de 12 pinguins P. papua, nas suas respectivas colônias na Ilha Elefante, em dezembro de 2014. De S. magellanicus foram coletadas, com auxílio de suabes cloacais, amostras de 19 indivíduos, encontrados na costa norte do Rio Grande do Sul, de Quintão a Torres, durante os meses de inverno de 2015 e de 2016. As amostras de microbiota dos pinguins foram cultivadas em ágar LB e os isolados bacterianos foram triados para os seguintes antimicrobianos: eritromicina (≥ 8μg/mL), estreptomicina (≥ 2.000 μg/mL), tetraciclina (≥ 16 μg/mL) e vancomicina (≥ 32 μg/mL). Em 10 amostras de P. antarcticus e em 15 amostras de S. magellanicus foram identificadas bactérias resistentes a pelo menos um dos antimicrobianos testados. Todas as amostras de P. papua foram sensíveis a esses antimicrobianos. As espécies dos isolados resistentes foram identificadas pelo sequenciamento do rRNA 16S, que revelou sete gêneros, sendo os mais recorrentes Enterococcus sp. e Staphylococcus sp. Esses isolados resistentes também foram triados para a presença de genes de resistência aos antimicrobianos. O tet(M) foi mais abundante em S. magellanicus (5) do que em P. antarcticus (3), ao passo que o int e van(B) foram identificados somente em P. antarcticus (três e um, respectivamente). O gene erm(B) não foi encontrado em nenhum dos isolados. Uma vez que a fração não cultivável das fezes também pode apresentar genes de resistência, foi realizada a extração do DNA das fezes de pinguins antárticos para obtermos DNA de todos os micro-organismos presentes. ▼ Hemija - Organska hemija / Chemistry - Organic Chemistry p>Rod Euphorbia, sa više od 2.000 vrsta jednogodišnjih, dvogodišnjih ili višegodišnjih cvetnica koje su članovi familije Euphorbiaceae, jedan je od najvećih i najraznovrsnijih rodova ne samo u svojoj familiji, već u celom biljnom carstvu. Biljke ovog roda koriste se u tradicionalnoj medicini još od antičkih vremena. Dosadašnja fitohemijska proučavanja pokazala su da vrste roda Euphorbia proizvode veoma raznovrsne sekundarne metabolite, uključujući različite terpene (seskviterpeni, diterpeni i triterpeni), steroide, cerebrozide, glicerole i fenolna jedinjenja (floracetofenoni, flavonoidi, tanini, kumarini). Mnoga od ovih jedinjenja pokazuju biološke aktivnosti, a najvažnije su reverzija višestruke rezistencije na lekove, antimitotsko, antiproliferativno, citotoksično, antivirusno, antimikrobno, antitumorsko, antiinflamatorno, tumor promotorsko i proinflamatorno dejstvo. Vrste iz roda Euphorbia su bogat izvor jatrofana, makrocikličnih diterpena osnovne trans-biciklo[10.3.0]pentadekanske strukture, za koje je karakteristično postojanje veoma fleksibilnog dvanaestočlanog prstena. To su veoma funkcionalizovana, različito oksigenovana jedinjenja; u biljkama se obično nalaze u obliku polihidroksilovanih diterpena esterifikovanih različitim kiselinama (acetil, propanoil, nbutanoil, izobutanoil, izovaleril, angeloil, tigloil, benzoil, nikotinoil derivati, itd.), zbog čega su poznati i pod nazivom jatrofanski poliestri. U okviru ove disertacije ispitivani su jatrofanski diterpeni iz vrste Euphorbia dendroides L. poreklom iz Crne Gore koja do sada nije fitohemijski proučavana. Proučavanje je obuhvatalo dve celine: nadzemni deo i mlečni lateks. Iz nadzemnog dela E. dendroides izolovano je i okarakterisano šest novih jatrofana, koji su nazvani eufodendrofani A – F (1 – 5, 16), a iz mlečnog lateksa E. dendroides izolovano je i okarakterisano trinaest novih jatrofana, eufodendrofani G – S (6 – 15, 17 – 19), kao i tri jatrofana izolovana iz nadzemnih delova (1, 2, 16). Izolovani jatrofani imaju vrlo slične strukture. Jatrofani 1 – 15 se međusobno razlikuju samo po vrsti, broju i položaju supstituenata, dok se jatrofani 16 – 19 od njih razlikuju po tome što sadrže endocikličnu (Δ5) umesto egzociklične (Δ6(17)) dvostruke veze.
