▼ Drug resistant infections with Gram negative bacteria have become increasingly common. Some of these Gram negative bacteria are pan resistant. To better understand the spread of drug resistant infections we used E. coli as a model system. By using this system we investigated both the spread of drug resistance genes and bacteria that are drug resistant.Drug resistance genes and their mobile genetic elements are frequently identified from environmental saprophytic organisms. It is widely accepted that the use of antibiotics in agriculture selects for drug resistant microorganisms, which are then spread from the farm environment to humans through the consumption of contaminated food products. We wished to identify novel drug resistance genes from microbial communities on retail food products. To do this, we created metagenomic plasmid libraries from microbiota isolated from retail spinach samples. From these libraries we identified five unique plasmids that increased resistance to antimicrobial drugs. These plasmids were identified in E. coli that grew on plates that contained ampicillin (pAMP), aztreonam (pAZT), ciprofloxacin (pCIP), trimethoprim (pTRM), and trimethoprim-sulfamethoxazole (pSXT). We identified open reading frames with similarity to known classes of drug resistance genes in the DNA inserts of all five plasmids. These drug resistance genes conferred resistance to fluoroquinolones, cephalosporins, and trimethoprim, which are classes of antimicrobial drugs frequently used to treat human Gram negative bacterial infections. These results show that novel drug resistance genes are found in microbiota on retail produce items. Food saprophytes may serve as an important reservoir for new drug-resistance determinants in human pathogens.The clinical management of infections caused by E. coli, including meningitis, is greatly complicated when the organism becomes resistant to broad-spectrum antibiotics. We sought to characterize the antimicrobial susceptibility, multilocus sequence type (MLST), and presence of known drug resistance genes of E. coli that caused meningitis between 1996 and 2011 in Salvador, Brazil. We then compared these findings to E. coli isolates from community acquired urinary tract infections (UTI) that occurred during the same time period and in the same city. We found 19% of E. coli from cases of meningitis and less than 1% of isolates from UTI to be resistant to third-generation cephalosporins. The sequence types of E. coli from cases of meningitis included ST 131, ST 69, ST 405, and ST 62, which were also found among isolates from UTI. These sequence types of E. coli have previously been isolated from produce items and food animals. Additionally, among the E. coli isolates that were resistant to third-generation cephalosporins, we found genes that encode the extended-spectrum beta-lactamases CTX-M-2, CTX-M-14, and CTX-M-15. ▼ This dissertation investigates the correlation between prosody and the speaker's bias in Japanese negative polar questions (henceforth, NPQs) without a sentence final particle. I argue that the prosodic contrast of the negative morpheme -na'i between accented and deaccented variants is correlated with the presence or polarity of the speaker's epistemic bias: while NPQs with deaccented -nai (P-type NPQs) often convey positive epistemic bias, NPQs with accented -na'i (NN-type NPQs) convey negative epistemic bias or represent that the speaker has no epistemic bias. In this respect, Japanese NPQs show a bifold difference with English NPQs, in the way they express the polarity of bias, and in the way they divide NPQs into subgroups. Some evidence is given to show that the prosodic patterns in Japanese NPQs are neither accidental nor in free variation, but rather intimately and robustly related to distinctions in the speaker's bias. Also, a case where the prosodic contrast disappears is discussed. I reject the analysis which claims that the prosodic contrast is due to post-focus pitch reduction, and argue that the contrast is due to deaacentuation, a phenomenon which should not be identified with is demonstrably distinct from post-focus reduction. I also discuss the non-truth-conditional aspect of the meanings of Japanese NPQs (e.g. bias). I argue that NN-type NPQs (those with negative bias) and P-type NPQs also demonstrate epistemic bias (concerning belief or expectation) and desiderative bias (concerning desire). I clarify several conditions which are required for each type of Japanese NPQ to be asked felicitously to convey a specific kind of epistemic bias ("epistemic" here is the general term independent of specific modal flavors). These findings point to directions for further work on other types of Japanese NPQs. I also introduce the results of two types of experiment, a naturalness rating test and a comprehension test, which were conducted to support my claim concerning the correlation between prosody and bias in Japanese NPQs. ▼ The majority of empirical research on responses to negative feedback has focused on affective responses to negative feedback, which have largely been adverse. The purpose of this study was to examine how negative feedback enhances motivation. A key feature of this study is the conceptualization of motivation using Edward Deci and Richard Ryan’s self-determination theory. Self-determination theory proposes a continuum of motivation, based on one’s regulation, or contingency for performance. Goal orientation and social dominance orientation are proposed as two moderators of the negative feedback-regulation relationship. Two studies were conducted to examine the relationship between negative feedback and regulation. Study 1 used a survey-based instrument with a work sample after a performance appraisal was conducted (N = 221), and Study 2 took place in a psychology statistics undergraduate course (N = 156). Negative feedback yielded a decrease in obligated motivation in Study 1. Mastery prove goal orientation and performance prove goal orientation were consistent significant moderators of the negative feedback-regulation relationship, such that individuals with high levels of Mastery prove goal orientation increased their autonomous regulation at higher levels of negative feedback, while individuals with high levels of performance prove goal orientation decreased their autonomous regulation at higher levels of negative feedback. Implications for feedback delivery are discussed. This study contributes to the literature by being the first to examine the effects of negative feedback on all forms of regulation, and is the first to use goal orientation and social dominance orientation as moderators of the negative feedback – regulation relationship. Recent studies indicate a correlation between high expression of the POZ-ZF transcription factor Kaiso, and the aggressiveness of the triple negative breast cancer (TNBC) subtype. However, little is known about the biological roles of Kaiso in TNBC tumorigenesis and metastasis, which laid the foundation for this thesis. To elucidate Kaiso’s role in TNBC, we generated stable Kaiso depletion in two well-established TNBC cell lines – MDA-MB-231 and Hs578T – using RNA interference technology. Intriguingly, we observed that Kaiso depletion delayed the tumor onset of MDA-MB-231 but not Hs578T cells, and led to the reduced expression of the c-Myc oncoprotein in MDA-MB-231 but not Hs578T cells. We postulate that this reduction in c-Myc expression is partly responsible for the delayed tumor onset observed in MDA-MB-231 cells. Additionally, loss of Kaiso expression resulted in increased apoptosis of both MDA-MB-231 and Hs578T cells in vitro and in vivo, which was accompanied by reduced expression of the DNA repair protein BRCA1. Remarkably, bioinformatic analysis revealed that high Kaiso and BRCA1 mRNA expression correlates with the reduced survival rates of TNBC patients. Further characterization of the Kaiso-depleted cells revealed that loss of Kaiso expression strongly inhibited the metastatic abilities of MDA-MB-231 and Hs578T cells. Importantly, Kaiso depletion led to decreased TGFβ-receptor I and II (TGFβRI and II) expression that is essential for the activation of the TGFβ signaling cascade. Concomitantly, suppressing Kaiso led to reduced TGFβ signaling. As increased TGFβRI expression is independently associated with the poor prognostic outcome of breast tumors, and the TGFβ signaling pathway is highly involved in breast tumor metastasis, we hypothesize that Kaiso functions together with TGFβRI and the TGFβ signaling cascade to promote TNBC metastasis. An additional goal of this thesis was to investigate the role of Kaiso in the prevalence of TNBC in women of African ancestry (WAA) compared to Caucasian women – since increased Kaiso expression is implicated in the poor survival outcomes of breast cancer patients of African ancestry relative to their Caucasian counterparts. Using tissue microarray and immunohistochemical analyses, we revealed for the first time a high nuclear expression of Kaiso in TNBC tissues of WAA (Nigerian, Barbadian, African American) compared to TNBC tissues of Caucasian women. Collectively, these findings unveiled functional oncogenic roles for Kaiso in the tumorigenesis and metastasis of TNBC, and revealed a plausible link between high Kaiso expression, high African ancestry and the predisposition of young WAA to TNBC.

▼ There are a number of reasons why informal settlements exist and why people decide to establish and develop them. The main concept is that informal settlements have become a perpetual challenge and adversely imposing negative impacts to the environment. Living in informal settlements leads to the exposure and vulnerability to environmental hazards to people and the land which they occupy. Informal settlements are characterized by a lack of basic services, pollution, overcrowding and poor waste management. These characteristics impact negatively on the environment posing risk and susceptibility to health problems associated with informal settlements. Apart from the living conditions which are found in informal settlements, the notion of informal settlements being self-built by families and using temporary materials has its own negative impact on the environment. People residing in informal settlements face challenges of service delivery. This is mainly due to the fact that land was occupied illegally, thus impacting negatively on the environment. A lack of basic needs and services leads to informal settlements having impacts on the environment. This includes inadequate sanitation which leads occupants to pollution, the use of coal and biomass fuel for lighting, causing air pollution and in turn contributing to the emission of greenhouse gases. Waste removal is an issue with which informal settlers’ struggle and which causes land pollution and emerging and re-emerging infectious disease hazards. Thus, waste removal is a problem which the study is to discuss and to examine possible solutions thereof. The main aim of the study was to investigate the impacts which informal settlements have on the environment and raise environmental awareness by illustrating how extreme the impacts of informal settlements are in impacting the environment. The main aim was to be achieved by using the following objectives: identifying environmental challenges which are caused by informal settlements, examining the sources of environmental challenges experienced in informal settlements, investigating solutions to reduce the negative impacts which informal settlements have on the environment, identifying health risks associated with the environmental impacts present in informal settlements and examining how waste is disposed of in informal settlements. The research method used for the study was qualitative research. This type of scientific research method seeks answers to a question, systematically using a predefined set of procedures to answer the question, collects evidence and provides findings that were not determined in advance. This method was used in conjunction with interviews with municipal officials and questionnaires handed to 40 of Jika Joe informal settlement residents who were selected using random sampling. ▼ Plant cells contain two or even three types of vacuoles: the lytic, the (seed) protein storage and vegetative storage vacuoles. Soluble vacuolar proteins are sorted through the secretory pathway to these vacuoles by three different routes, depending on different types of Vacuolar Sorting Determinants (VSD) and involving several types of receptors and vesicles. The AtRMR1 protein has been identified in cellular structures associated with the seed storage vacuole pathway (Jiang et al. 2000). Based on its localisation and homology to a known vacuolar receptor, it has been hypothesised to be a receptor protein for the C-terminal type of VSD (CtVSD) involved in sorting to the storage vacuole. The genome of Arabidopsis thaliana contains 5 genes homologous to AtRMR1. The main goal of this study was to test the involvement of AtRMR1 in vacuolar sorting and to test the specificity of the different AtRMR proteins for different known CtVSDs. To test the involvement of AtRMR proteins in vacuolar sorting we studied the effects of manipulations of these genes on targeting of vacuolar reporter proteins. I used two different models: A. thaliana leaf protoplasts and whole plants of insertional mutants from the SALK Institute collection. The protoplast model allowed me to study in vivo the effects on vacuolar sorting of versions of AtRMR1 with loss of function deletions or modified functions. I obtained interesting results with two of these constructs: one lacking the luminal VSD-binding domain (RMRΔlum) and one consisting of this soluble luminal domain retained in the ER by the addition of a HDEL peptide signal (RMRlumER). When overexpressed with GFP fused to the ssVSD of barley aleurain, the RMRΔlum construct showed a different pattern of fluorescence compared with the control: in RMRΔlum, a significant proportion of protoplasts showed a dot-like fluorescent pattern, whereas the fluorescence was more often found in the central vacuole or ER in the control. When I used either the CtVSD of tobacco chitinase or a short version of the barley aleurain ssVSD, I didn’t see a different fluorescent pattern with or without the dominant negative construct. To better estimate the effects of the dominant negative constructs, the level of secretion of enzymatic reporters was investigated. The reporters were α-amylase fused either to the CtVSD of tobacco chitinase, the CtVSD of barley lectin or the ssVSD of sweet potato sporamin. For these three different reporters, overexpression of RMRΔlum induced an increased secretion of the reporter, whereas a simple fractionation showed that the RMRlumER construct provoked their accumulation in microsomal compartments, presumably ER. Two other constructs, where either the luminal domain of AtRMR1 was redirected to the PM (RMRlumPM) or the soluble cytosolic domain was overexpressed (RMRcyt) didn’t have any effect on the sorting of enzymatic reporters, suggesting that the transmembrane and cytosolic domains are both important for the VSD binding and that the cytosolic domain alone is not able to interfere with… Advisors/Committee Members: Jean-Marc (Dir.).

▼ Infections caused by extended-spectrum β-lactamase (ESBL)-producing Gram-negative bacteria are associated with increased mortality and cost of care. These pathogens are particularly challenging to treat due to their resistance to many antibiotics. Although β-lactam/β-lactamase inhibitors have proven in vitro activity against ESBL-producing pathogens, their use for ESBL infections remains controversial due to reports of treatment failure and a limited understanding of the pharmacokinetic/pharmacodynamics (PK/PD) governing these combinations. As a result, carbapenems have remained the preferred agents for treating infections due to ESBL-producing organisms. However, the heavy reliance on these carbapenems may have contributed to the rapid dissemination of carbapenemases that further limit treatment options. This has heightened the need to optimize the use of available antibiotics against these clinically-challenging pathogens. Thus, the goal of this project was to improve the rational basis for the dosing and pairing of β-lactams and β-lactamase inhibitors that would extend their usefulness against resistant organisms. We used a commercially available and widely used combination (piperacillin/tazobactam) to illustrate our approach. In our third chapter, we explored the prevalence and mechanism of resistance for extended-spectrum β-lactamases produced by Klebsiella pneumoniae isolated at a local tertiary care hospital. In so doing, we were able to focus our efforts on the most clinically-relevant β-lactamases to our geographic region. Using a collection of β-lactamase-producing clinical isolates, in our fourth chapter, we demonstrated a novel approach to susceptibility profiling for β-lactam/β-lactamase inhibitor combinations that could better inform dosing. Using a theoretical concept called the instantaneous MIC (MICi), we illustrated how susceptibility may be dependent on inhibitor concentrations and differed for each clinical isolate. In chapter 4, we also illustrated that the efficacy of combinations such as piperacillin/tazobactam was dependent on the concentration-response relationship between the β-lactamase producing isolate and the inhibitor (tazobactam). We showed that the current fixed dose ratio for piperacillin/tazobactam was inadequate against our test isolates, even when they tested susceptible by current susceptibility standards. Instead, escalated exposures of tazobactam were required to optimize dosing for certain clinical isolates. Finally, in chapter 5, we showed that for isolates that failed to respond to optimal piperacillin/tazobactam dosing, pairing piperacillin to an alternative inhibitor improved efficacy. The results from our studies have contributed to a better understanding of the PK/PD governing the joint effect of β-lactam/β-lactamase inhibitor combinations like piperacillin/tazobactam. Moreover, our results also highlighted additional considerations for the optimal pairing of β-lactams and β-lactamase inhibitors. ▼ This thesis investigates bullying and negative behaviour among chefs working in commercial kitchens. The idea for this study arose due to evidence from the hospitality industry and amongst chefs in particular which suggested that negative behaviour and bullying were widely accepted practices. However, much of this evidence has been either anecdotal from media reporting or based on small scale studies.The industry has also complained about high labour turnover and the need for a trained workforce. Therefore, this study examines the behaviours to which chefs are exposed and if negative behaviours cause them to leave the industry.This thesis has used a questionnaire to measure responses from chefs who were either in training in catering colleges or working in the industry. Questionnaires were distributed to first year student chefs (n = 202), final year student chefs (n = 153) and working chefs (n = 304). Working chefs and final year student chefs were given questionnaires that included a behavioural method of measuring bullying, known as the NAQ-R, a self labelling method of measuring bullying and items about kitchen specific behaviours. Working chefs and final year student chefs were also asked about positive aspects of work and job satisfaction. First year student chefs were also given a personality instrument as well as being asked their opinions of kitchen specific behaviours.The findings suggest chefs’ exposure to regular bullying was in line with another major UK study (Hoel, 2002). However, occasional bullying was much higher. An examination of industry specific behaviours revealed that chefs tolerate a range of behaviours from verbal abuse on the one hand to physical and sexual abuse on the other. There were also positive aspects about the freedom of work and job satisfaction but this study was unable to find any evidence as to what made chefs stay in the industry.The study found that as student chefs become socialised into their role they were more likely to tolerate negative work behaviours and could identify reasons for their use although, this did not mean that they necessarily agreed with them. In fact, as the working chef sample was very different in ethnicity, gender and nationality from the student sample it could mean that due to negative behaviours on graduation students may not work as chefs.

▼ With identity negative priming (INP) designs, subjects respond on the basis of a target stimulus’ identity and trials are presented in pairs; first the ‘prime’ and then the ‘probe’; with the target and/or to-be-ignored distractor objects appearing on either trial. The latency-based INP phenomenon reflects the fact that probe trial target reactions are significantly slower when this target has just served as the prime distractor object, relative to when it was new. One explanation for the INP phenomenon is that the prime distractor event is inhibited and representations of this are stored so that they are retrieved when the probe trial is delivered and then participates in related processing on this trial. Its participation produces inhibitory after-effects such as the INP effect. The specific question here is what aspect of the prime distractor event is inhibited that causes the INP effect (i.e., locus question); its object (i.e., Object NP), its response (i.e., Response NP) or both? The task employed here to examine the locus of the INP effect used the numbers 1-8 as inputs that were mapped in pairs onto a single button-press finger response (many:1 mapping), allowing us to (a) repeat both the prime distractor object and its response as the probe target (ignored-repetition), (b) repeat the prime distractor response but not the prime distractor object (distractor-response repeat) and, (c) repeat nothing from the prime trial (control trial) [RT(a) > RT(b) = Object NP; RT(b) > RT(c) = Response NP]. In addition to this manipulation, we used three Response Conflict trial types on the probe trials (conflict distractor, neutral distractor, no probe distractor). This allowed us to test the assertion that an INP would only materialize if the probe contained a conflict distractor (which had an assigned experimental response). Finally, two groups were distinguished on the basis that one had pre-experimental trials to enhance the number-to-finger association strength (Practice Group), the other did not (No Practice Group). The main findings of note and their implications were as follows: (i) as expected, prime trial distractor events were processed, seemingly to the point of the activation and subsequent inhibition of their related response (i.e., ‘conflict’ [assigned response] but not ‘neutral’ [no assigned response] probe distractors produced significantly elevated reaction times beyond that of the control condition, indicating that this latency elevation was due to the distractor response, likely because it caused a response conflict which took time to resolve), (ii) stored prime distractor response processing was retrieved and participated in ‘related’ probe trial processing (i.e., evidenced in the inhibitory after-effects they produced), such as on ignored-repetition and distractor-response repeat trials. The nature of the probe trial inhibitory after-effects produced by the retrieved prime distractor response was somewhat unexpected, however. L’oeuvre de Foucault, des années 1970 jusqu’à sa disparition en 1984, fut marquée par une critique du «complexe d’Oedipe», concept essentiel à Freud. La radicalité des critiques foucaldiennes aurait impliqué, d’une part, un certain éloignement des psychanalystes de ses travaux et, d’autre part, que ses lecteurs prennent une certaine distance de la lettre freudienne. Un détail fondamental à la compréhension de cette difficile interlocution: les références du philosophe au «complexe d’Oedipe» furent toujours formulées sans qu’il ne présente à ses lecteurs aucune citation de Freud. De l’intérieur de son propre texte, qui se rapporte systématiquement à l’oeuvre du psychanalyste, Foucault convoqua, à chaque fois, son interlocuteur sans jamais lui donner la parole. Quelle lecture de Freud aurait-il engagé? Comment put-il autant écrire sur le «complexe d’Oedipe» sans jamais citer Freud? Afin de dégager les modalités, les aspects, les formes de cette opération de lecture effacée, on abordera les principales références foucaldiennes au «complexe d’OEdipe» et leur logique négative.

Foucault's work, from the 1970s until his death in 1984, was marked by a critique of the "Oedipus complex", a concept essential to Freud. The radicalism of Foucault's critics would have implied, on the one hand, a certain distance from the psychoanalysts of his works and, on the other hand, that his readers take a certain distance from the Freudian letter. A fundamental detail to the understanding of this difficult interlocution: the references of the philosopher to the "Oedipus complex" were always formulated without presenting to his readers any quote from Freud. From within his own text, which systematically refers to the work of the psychoanalyst, Foucault summoned each time his interlocutor without ever giving him the floor. What reading of Freud would he have done? How could he write so much on the "Oedipus complex" without ever mentioning Freud? In order to identify the modalities, the aspects, the forms of this erased reading operation, we will approach the main Foucaldian references to the "Oedipus complex" and their negative logic.
