▼ Mosaic is a viral disease of sugarcane caused primarily by Sorghum mosaic virus (SrMV) in Louisiana. Low mosaic incidence has resulted from successful breeding for resistance. However, mosaic was detected in breeding program experimental clones and a new cultivar, HoCP 09-804.Therefore, multiple research approaches were undertaken to assess the current status of mosaic in Louisiana sugarcane and prevent it from re-emerging as an important problem. Field surveys conducted during 2016-2018 of breeding program yield trials and experimental clone seed-cane increases determined disease incidence and distribution. Mosaic was detected in three of five sugarcane production areas and incidence ranged from 0 to 10% in HoCP 09-804. Symptomatic and asymptomatic leaves were tested for SrMV with reverse transcription polymerase chain reaction (RT-PCR). All symptomatic leaves tested positive for SrMV confirming it is the current causal virus species. A low percentage of asymptomatic leaves (0.3%) tested positive for SrMV. Runs analysis detected aggregation of infected plants in rows of surveyed fields. The geographic and within field distribution suggested the source of disease was infected seed-cane. Subsequent surveys of the same locations detected incidence increases and decreases in first ratoon, but incidence decreased for all in second ratoon. The results suggest high rates of disease increase due to aphid transmission are not occurring under current conditions. Recovery from mosaic was evaluated as the emergence of asymptomatic plants from symptomatic stalks. L 10-147 had a higher frequency of recovery (9.4-18.9%) than HoCP 09-804 (0.9-2.3%) across two experiments. RT-PCR failed to detect SrMV in 83% of HoCP 09-804 and 97% of L 10-147 of recovered plant samples. Comparison of asymptomatic and symptomatic stalk plantings found mosaic reduced yield in HoCP 09-804 but not L 10-147. Sources of susceptibility were evaluated in the basic and commercial parent populations by mechanical inoculations. Mosaic susceptible parents were identified within both populations. The level of susceptibility within the basic parent population was low indicating it will continue to be a resource for continued introgression of mosaic resistance. The results will allow the elimination of sources of susceptibility and informed crossing to continue successful management of mosaic with host plant resistance.

Dans le royaume végétal, les plantes doivent faire face à de nombreux facteurs de stress environnementaux abiotiques et biotiques. La diversité des agents pathogènes conduit encore aujourd’hui à des pertes économiques importantes en impactant le rendement et la qualité des fruits des plantes cultivées. Si pour certains agents pathogènes les traitements chimiques sont adaptés, ils restent inopérants contre les virus menant à l’utilisation de moyen empirique fastidieux à mettre en place et souvent onéreux. La tomate est une plante d’intérêt agricole majeur dont l’un des challenges les plus importants consiste à lutter contre les maladies et ravageurs pouvant provoquer de graves dégâts industriels. Parmi ces fléaux, le PepMV fait partie d’un groupe de virus émergeants dont les connaissances générales sont encore très limitées et méritent d’être approfondies compte tenu de son impact agronomique important. Puisqu’aucune résistance naturelle n’a encore été identifiée chez la tomate vis-à-vis du PepMV, il devient urgent de développer des moyens de lutte efficace afin d’améliorer la production maraichère. Les études développées lors de ce doctorat se sont donc concentrées sur les nouvelles stratégies d’amélioration des plantes en identifiant dans un premier temps des mécanismes d’interaction du pathosystème PepMV/tomate par la mise en place d’une stratégie sans a priori en levure. Cette étape a permis de déterminer des gènes candidats de tomate dont certains potentiellement indispensables au cycle viral du PepMV. Des cribles par la méthode TILLING ont été réalisés avec les facteurs hôtes de sensibilité afin d’acquérir, si possible, des plantes possédant une résistance durable, à large spectre vis-à-vis de nombreux virus ou agents pathogènes et commercialisables en Europe car non OGM. D’autres candidats ne pouvant pas être exploités par une perte de fonction, qui conduirait à une sensibilité accrue des plantes suite à l’infection du PepMV, ont alors fait l’objet d’une étude scientifique plus fondamentale. Ces études ont permis de souligner le mécanisme et le rôle de résistance de plusieurs candidats dans le pathosystème PepMV/tomate.

Within the plant kingdom, crops struggle with numerous abiotic and biotic stresses. The fabulous diversity of pathogens still leads to dramatic economic losses by impairing yield and fruit quality. Chemical treatments are available and effective against some pathogens but unfortunately drive to critic environmental damage and remain useless to face viruses. Nowadays, best ways to eradicate or at least to restrain viruses are empirical and expensive. Tomato is one of the most cultivated plants among vegetable crops in agriculture and the major challenge consists in a intense battle against serious damages on industrial benefits caused by pathogens diseases. The emergent virus PepMV is a worldwide scourge and the limited knowledge deserves to be explored. ▼ An epidemiological and molecular characterization study of cassava mosaic disease was carried out in four cassava growing Provinces of Zambia in 2013. The objectives of the study were to determine the incidence and severity of Cassava mosaic disease (CMD) in the study areas; to determine molecular diversity of Cassava mosaic geminiviruses (CMGs); to evaluate the effect of CMD on yield of selected cassava genotypes. The study involved survey of CMD in four cassava producing provinces, virus isolate characterization using the polymerase chain reaction (PCR) and sequencing of purified PCR products and evaluation of nine landrace and improved cassava genotypes to CMD. The results from the survey showed CMD incidence and severity was 61.5 % and 2.7, respectively. The results also showed that common transmission of the disease was due to cutting infection (57.7 %) compared to whitefly infection (3.8 %). The polymerase chain reaction showed single infections of African cassava mosaic virus (ACMV) in 34.7 % of the positive samples while East African cassava mosaic virus (EACMV) occurred in 4.5 % of the positive samples. The dual infections of ACMV and EACMV were detected in 60.8 % of the positive samples. Restriction fragment length polymorphisms (RFLP‟s), complimented by sequence analysis done by multiple sequence analysis established the existence of different CMG isolates in Zambia. The sequence identity analysis and phylogenetic analysis displayed high nucleotide (nt) sequence identities amongst one another with the isolate sequences showing identities of 89 % to 99 % within the coat protein (CP) gene of ACMV. The Zambian isolates in this study showed substantial homology with sequences of ACMV-UGMild Uganda (AF126800.1), ACMV-UGSvr Uganda (AF126802.1), ACMV-[MG:MG310A1] Madagascar, and ACMV-CM39 Cameroon (AY211462.1) with sequence identities of 97 %, 97 %, 97 % and 98 %, respectively. However, the Zambian isolates showed greater variability within the EACMV species nucleotide sequence divergence ranging between 77 % to 99 %. The isolates showed similarity to the Kenyan (EACMV-KE), Malawian (EACMMV) and Tanzanian (EACMV-TZT) with sequence identities of 96 %, 90 % and 96 %, respectively. Among the genotypes evaluated for CMD response, Manyopola and Bangweulu were found to be susceptible (3.5) whilst Kampolombo was resistant (1.7). Mweru had the highest root yield (1.6 kg plant-1) while Kapumba, the second most susceptible genotype, had the lowest root yield (0.2 kg plant-1). The current study showed slight epidemiological variation from previous results in same areas. This underlines the importance of continuous evaluation of the disease in order to develop effective control measures.

Control relies on cultural practices to control the vector, the wheat curl mite (Aceria tosichella Keifer, WCM), and the use of resistant or tolerant varieties. WSMV and WCM depend on living tissue for survival and reproduction, including common grassy weeds. Little is known about the relative importance of these weeds as alternative hosts of WSMV. The purpose of these studies was to evaluate the risk of infection with WSMV in commonly grown wheat varieties and various grassy weed species, information useful to understanding WSMV epidemiology and control. Winter wheat, spring wheat and barley varieties in Montana were evaluated in the field by measuring the effect of fall vs. spring inoculation and variety on incidence, symptom severity, and yield components. Winter wheat varieties from five states, and spring wheat and barley varieties from Montana were tested for incidence and absorbance in greenhouse. Fall-inoculated winter wheat had less effect of WSMV inoculation compared to spring-inoculated winter wheat. Yields of spring wheat varieties were largely reduced by WSMV inoculation. There was no correlation between yield and incidence or symptom severity. In greenhouse studies, the highest incidence was observed in varieties from Idaho and Nebraska, whereas the highest relative absorbance was observed in varieties from Montana. In 2008 and 2009, surveys of common grassy weeds were conducted. Grass species from croplands in six states were selected and mechanically inoculated to determine the susceptibility to WSMV. Grassy weeds were also evaluated as a source of WSMV by measuring transmission efficiency with virulifeous WCM. Bromus tectorum was the most prevalent grassy weed and the most frequent viral host. Aegilops cylindrica, and Avena fatua had the highest incidence and relative absorbance. There were no differences in the susceptibility of grass species to WSMV by their state of origin. WCM transmission study indicated infected grass species had lower transmission efficiency than from infected wheat. These studies will benefit producers in Montana to assess their risk of WSMV based on variety selection and the presence of grassy weeds. ▼ Field images are becoming more frequently used for sensing the crucial properties of the crops in Precision Agriculture. Some of them contain distortions that need to be removed before further analysis. Data from images, such as the coordinates of the crops, also need specific algorithm to be extracted. For these purposes, two computer vision algorithms were developed for pre-processing the field images from two monocular vision systems. One algorithm was used for a tower remote sensing system data pre-processing for image distortion removal and the mosaic to generate geo-referenced images. The other was for the image data interpretation used for the vision system of a field robot. Satellite and aerial remote sensing systems are the two major platforms for collecting remote sensing images for agriculture. However, due to the critical drawbacks of these systems, such as low spatial and temporal resolution, a tower remote sensing system with a 360-degree rotatable camera on the top has been established in the experiment field to obtain the multi-spectra images for monitoring the status of the plants. In this research, the geo-reference and image mosaic algorithms were developed for data acquisition. While taking remote sensing images of the field, the camera will turn 360o horizontally and 90o vertically. This creates the difficulty of geo-reference because different images have different distortions. Therefore, traditional ways of geo-reference, such as using Ground Control Points (GCP), are no longer appropriate. A three-axis digital compass was used to provide the absolute orientation of the camera, which can be used to geo-reference a single image. The calibration of both camera and compass was introduced, and necessary parameters for geo-reference were estimated. Based on the angles, positions and optical parameters of the camera, a transformation from the image coordinate system to the ground coordinate system was introduced. After the transformation, the performance of the geo-reference method was evaluated with data from a Real-Time Kinematic Global Positioning System (RTK-GPS) to assure the accuracy. Since there are not enough features in the field, the algorithm of the image mosaic for the tower system is based on geographical information rather than features. Moreover, comparing to aerial and satellite systems, the images from a tower remote sensing system have usually 10 to 15 times larger pitch angles that result in large geometric distortion. Thus further processing is needed to remove distortion. The algorithm firstly used coordinate transformation to compute top-view coordinates of all pixels in the image. The new coordinates were used for reorganizing the pixels. Due to the large geo-metric distortion, the Pixel Combination was applied. After computing the top-view image, global alignment was applied to generate an initial image mosaic. ▼ Mosaic Arthroplasty is a well-accepted surgical approach to treating focal cartilage defects of the knee. However, the task of creating the mosaic of osteochondral grafts that optimally restores the cartilage surface is technically demanding. Conventional techniques require the surgeon to reconstruct a complex, three-dimensional surface by eye and experience only. There is evidence that this type of procedure is sensitive to technique: grafts that are transplanted proud of the native cartilage surface tend to show evidence of cartilage fissuring and fibrillation prematurely. Two computer-assisted techniques (navigation by optical tracking and guidance by patient-specific templates) were investigated to determine whether they would have a beneficial effect on surgical execution, and whether any differences in execution had any correlation to surgical outcome. The experimental work can be broken into two parts: an in vitro study that compared the accuracy of execution of an optically navigated group versus a template guided group, and an in vivo animal trial that compared both computer assisted techniques to the conventional, non-assisted approach. The results of the pilot study indicated that, while there were higher errors in the individual measures of position, orientation, and plug depth in the optically navigated group, there was no significant difference in the overall fidelity of the geometric surface produced between the two groups. The results of the animal trial indicated that both computer assisted techniques produced morphological results that were superior to the conventional technique. The two computer-assisted techniques also showed a significantly better treatment effect as seen by their higher histological scoring. Furthermore, a significant linear correlation was found between morphological results and histological score. Overall, the experiments demonstrated that surgeons and patients could potentially benefit from the use of computer-assisted techniques in the short term. Further work is required to prove long-term beneficial effect.

▼ Sugarcane mosaic virus (SCMV) belongs to the Potyvirus genus and Potyviridae family of single-stranded RNA viruses. It is a disease of great economic importance, especially in sugarcane and maize in many parts of the world. SCMV has been understudied at both the genomic level with a greater emphasis put on the coat protein gene (CP) for molecular epidemiology. This was a computational study using publicly available sequences (NCBI GenBank) that sought to understand the evolutionary processes that shape SCMV molecular evolution such as phylogeny, recombination, selection pressure and nucleotide diversity by focusing on the individual genes of the polypeptide. Recombination was found to be a major driver of evolution with breakpoints found in P3, HC-Pro, C1, NIa-Pro, NIb, and CP. No statistically significant recombination was detected in the P1, 6K1, 6K2, and VPg genes most probably because of their relatively shorter length. After the removal of recombinants from the initial 82 full polyprotein-coding sequences, 57 sequences were used in phylogenetic analysis using a Bayesian MCMC framework implemented in BEAST2 with particular substitution models for each gene. All the ten genes gave similar low mean nucleotide substitution rates of between 3.35 x 10-3 (C1) to 4.29 x 10-3 (CP) and time to the most recent common ancestor (TMRCA) of 219 years (CP) to 264 years (P1). This would mean that the community’s over-reliance on the CP may produce results accurate for the whole genome, provided the researchers controlled for the effects of recombination, but that sufficient data exists for other genes as well, so there is no rational justification for excluding them from similar studies. The SCMV strains clustered into two distinct groups with sub-clustering well defined by their geographical isolation points, with sequences from Argentina closely grouping with Chinese strains. This observation may not be conclusive as most of the sequences studied were from China. Analysis of dN/dS shows significant negative selection in all the genes with P1 and CP registering relatively lower levels (SLAC: 0.192 for P1 and 0.153 for CP). A few sites in P1, HC-Pro, P3, NIa-VP, NIb, and CP appear to be under diversifying selection. P1 and CP also had the highest nucleotide diversity while the overlapping P3N-PIPO region had the least diversity relative to other regions of the polypeptide. The overlapping region is understandably highly conserved and presumably under strong purifying selection due to its double role in translation. This knowledge from this study enhances the understanding of SCMV evolution, highlights residues in several genes that may be affecting SCMV-crop interactions and will help in developing strategies for the control of the diseases in plants. The yam (Dioscorea spp.) is affected by several diseases and pests occurring in field and/or during transport and storage. It is a crop with a high social economic importance in Northeast of Brazil, showing a good expansion potential by tuber export, especially to Europe. An evaluation of disease and pest development in important producing areas was undertaken, as well as a study on the quality of tuber seed used by the growers. Visits to seven counties of Pernambuco and three of Paraíba, totaling 30 farms were done. In order to evaluate the occurrence of diseases and the damages caused by pests under field conditions, in each field five lines of 10 plants were marked, while for virus evaluation, 30 plants were randomly selected. The plants were analyzed 30 days before and at the harvest day. The nematodes were extracted by association the Jenkins method and identified by their structures under light microscope. For fungus identification, isolation and inoculation were doneand the structures were observed under light microscope. For the viruses, observation under electron microscopy, triple antibody sandwich enzyme-linked immunosorbent assay (TAS-ELISA), with polyclonal and monoclonal antibodies specific to Yam mosaic virus (YMV), from IITA, Nigeria, and a polymerase chain reaction (PCR) with degenerated primers for detecting badnavirus and duplex reverse transcription (RT-PCR) with specific primers to YMV and Yam mild mosaic virus (YMMV) were used. The PCR and RT-PCR products were purified and sequenced in order to validate the tests used for detecting badnavirus andidentification of the potyviruses by the comparative analyses of the sequences available in the GenBank. It was observed the leaf spot caused by Curvularia eragrostidis in all areas studied, with a higher intensity on cv. Inhame-da-Costa of D. cayennensis-D. rotundata complex, whereas the damages caused by leaf wor (Pseudoplusia oo), present in all fields, were higher on cv. São Tomé of D. alata plants. The seed tubers of both yam species, collected in five farms representative of the growing areas of Pernambuco and Paraíba, after 60 days of storage, presented serious problems of green rot caused by Penicilium sclerotigenum, mealybugs (Planoccocus sp.) and tuber worms (Araecerus fasciculatus). It was identified the potyviruses YMV and YMMV, occurring in single and mixed infections with badnavirus, consisting the first detailed report with molecular information on theseviruses in Brazil.

O inhame (Dioscorea spp.) tem sido afetado por a vários problemas fitossanitários, que se manifestam na lavoura e durante o transporte e/ou armazenamento. É uma cultura de elevada importância sócio-econômica para a região Nordeste do Brasil, inclusive com potencial de expansão via exportação de túberas, especialmente para a Europa. Foi efetuada uma avaliação do desenvolvimento de doenças e pragas em importantes áreas produtoras, bem como um estudo da qualidade do material de propagação utilizado pelos produtores. Foram visitados sete municípios de Pernambuco e três da Paraíba, totalizando 30 propriedades. Para avaliação da ocorrência das doenças da parte aérea e danos causados por lagarta, foram marcadas em cada campo, 5 fileiras de 10 plantas e mais 30 plantas, escolhidas aleatoriamente, para análise de viroses, as quais foram avaliadas um mês antes e no período da colheita. Para extração dos nematóides foi usado método de centrifugação rápida associadoao de Jenkins, os quais foram identificados por suas estruturas observadas ao microscópio ótico. Os fungos detectados foram isolados e identificados e observando-se suas estruturas reprodutivas. Para identificação dos vírus foram efetuadas observações ao microscópio eletrônico de transmissão, teste triple antibody sandwich enzyme-linked immunosorbent assay (TAS-ELISA), com anti-corpos policlonais e monoclonais específicos para Yam mosaic virus (YMV), obtidos do IITA, Nigéria e reaçaão de PCR (polimerase chain reaction) com oligonucleotídeos degenerados paradetecção de badnavírus e duplex reverse transcription (RT)-PCR com oligonucleotídeos específicos para YMV e Yam mild mosaic virus (YMMV). Os produtos de PCR e RT-PCR foram purificados e seqüenciados para validação dos testes usados na detecção de badnavírus e identificação dos potyvírus, por análise comparativa com as seqüências disponíveis no GenBank. Ficou comprovada a ocorrência de pinta preta, causada por Curvularia eragrostidis em todas as áreas estudadas, com maior intensidade na cv. Inhame-da-Costa do complexo D. cayennensis-D. rotundata, enquanto que o ataque de lagarta das folhas (Pseudoplusia oo), presente em todos os campos, teve maior severidade na cv. São Tomé de D. alata. As túberas-semente de ambas as espécies de inhame, coletadas em cinco propriedades representativas das áreas de cultivo em Pernambuco eParaíba, após 60 dias de armazenamento, apresentaram sérios problemas de Podridão Verde, causada por Penicillium sclerotigenum, cochonilhas (Planoccocus sp.) e brocas das túberas (Araecerus fasciculatus). Foram identificados os potyvírus YMV e YMMV, ocorrendo em infecção simples e mista com badnavírus, constituindose o primeiro registro detalhado com informações moleculares destes vírus no Brasil.

▼ As cucurbitáceas podem ser afetadas por várias doenças, dentre elas as viróticas, as quais podem provocar perdas de até 100% da produção. As plantas infectadas por vírus apresentam mosaico, redução do limbo foliar e deformação nas folhas e frutos, podendo a sintomatologia variar tanto com o hospedeiro infectado quanto com a ocorrência de infecções mistas. Devido às suas condições climáticas, favoráveis ao aparecimento das viroses, o estado do Tocantins tem apresentado grande incidência dessas doenças no campo. Contudo, não existem estudos direcionados à sua caracterização e identificação, dificultando, assim, o desenvolvimento de programas de melhoramento visando à resistência a doenças em abóboras. Nesse contexto, este trabalho foi realizado com a finalidade de identificar sorológica e molecularmente isolados de abóbora e melancia, coletados naquele estado, em colaboração com pesquisadores da Universidade Federal do Tocantins (UFT). A identificação desses isolados será de fundamental importância para fornecer subsídios aos programas de melhoramento da UFT, bem como estabelecer medidas de controle para as viroses de cucurbitáceas no campo. Foram coletadas 25 amostras de plantas de cucurbitáceas que apresentavam sintomas característicos de viroses, em diversos municípios de Tocantins. Estes isolados foram inoculados em diferentes espécies, como abobrinha-de-moita (Cucurbita pepo cv. Caserta), abóbora- rasteira (C. moschata Menina Rajada), melancia (Citrullus lanatus Crimson Sweet), melão (Cucumis melo), mamão (Carica papaya), pepino (Cucumis sativus), Chenopodium amaranticolor e Chenopodium quinoa, para a identificação biológica. Além disso, foi realizado teste de diagnose DAS-ELISA com antissoros específicos, diagnose por meio de RT-PCR e sequenciamento genômico de fragmentos da região do gene do capsídeo. Quatorze dos 25 isolados estudados, ou seja, 56%, foram identificados como Squash mosaic vírus (SqMV), tendo sete deles sido provenientes de abóbora Caserta e sete de melancia. Os demais isolados foram identificados como Zucchini yellow mosaic vírus (ZYMV). Ao contrário do que ocorre em outras regiões brasileiras, o SqMV foi o vírus com maior incidência nas amostras. Os isolados de SqMV, ao infectarem plantas de melancia, causaram mosaico e distorção foliar, diferentemente do que tem si do descrito por diversos autores, em outras partes do Brasil. A análise da capa protéica dos 14 isolados de SqMV mostraram uma identidade de nucleotídeos entre eles que variou de 86 a 100% e, entre eles e os seis isolados disponíveis no GenBank de 86 a 88%. A analise de aminoácidos dessa região, mostrou identidade de 95 a 100% entre eles e 91 a 97% entre eles e os isolados estrangeiros. O isolado que apresentou maior variabilidade foi o de Porto Nacional, denominado de PN2. ▼ No name seems to have been associated with more systematic criticism in regard to political and social thought during the magisterial Reformation than that of the Old Testament lawgiver, Moses. Beginning early in the Reformation era, rejections of the need for Mosaic judicial laws are varied, broad, and explicit. In some cases, such as Luther’s and Melanchthon’s attacks on Andreas Karlstadt, alleged proponents of Mosaic civil law are given by name. In other cases they are anonymous. But what is less clear is whether anyone actually held the views attributed. After a review of literature of Melanchthon, Jacob Strauss, Karlstadt, Zwingli, Thomas Müntzer, the peasants of the Peasant War (1524–5), Luther, the Anabaptists of Münster, Calvin, and others, it is confirmed that none of the implicated writers between key dates of 1520 and 1536 actually held the view of exclusive Mosaic Law attributed, particularly by Calvin. Other motivations must have been involved in the accusations. An analysis of literature from Luther and Calvin as well as the historical background of the period makes it clear that social, political, and economic pressures influenced the magisterial reformers in regard to crucial theological expressions in which they strongly rejected the need for Mosaic civil law in society. The reformers in question restrained or altered their expressions according to the pressures of external circumstances - most importantly war and rebellion spurred by so-called “radical” reformers. As alleged theological positions were weaved with reports and denunciations of violence, Mosaic Law emerged as an allegedly dangerous ideological force, the accusation of which could marginalize opponents. In this crucible of history, in which the long shadows of rebellion and war were cast over Mosaic Law during the mid-1520s and mid-1530s, we find both Luther and Calvin (among others) writing their most vehement denunciations of Mosaic Law. Particularly, we find young Calvin, exiled, sitting down to write his denunciation of “some” who rejected the validity of a commonwealth unless it relied exclusively upon Mosaic civil polity. Luther, Calvin, and others thus warned against applying Moses in the civil realm and linked his laws with sedition and rebellion (even though the association was not accurate in any given case) mainly for their own utilitarian causes. Both Calvin and Luther subsequently employ the doctrine of two kingdoms in distancing themselves and their movements from the need for Mosaic laws in the civil realm, as well as to impede opponents who would use civil power to enforce reforms contrary to them, and yet both act inconsistently when enforcement of the first table of the Decalogue would favor their own reforms. As well, both go on to advance and approve of non-biblical civil laws more invasive and extensive than Mosaic polity would have allowed - including the execution of Anabaptists - all the while denouncing alleged proponents of Moses as dangerous, seditious, barbaric, murderous, and bloodthirsty. ▼ No name seems to have been associated with more systematic criticism in regard to political and social thought during the magisterial Reformation than that of the Old Testament lawgiver, Moses. Beginning early in the Reformation era, rejections of the need for Mosaic judicial laws are varied, broad, and explicit. In some cases, such as Luther’s and Melanchthon’s attacks on Andreas Karlstadt, alleged proponents of Mosaic civil law are given by name. In other cases they are anonymous. But what is less clear is whether anyone actually held the views attributed. After a review of literature of Melanchthon, Jacob Strauss, Karlstadt, Zwingli, Thomas Müntzer, the peasants of the Peasant War (1524–5), Luther, the Anabaptists of Münster, Calvin, and others, it is confirmed that none of the implicated writers between key dates of 1520 and 1536 actually held the view of exclusive Mosaic Law attributed, particularly by Calvin. Other motivations must have been involved in the accusations. An analysis of literature from Luther and Calvin as well as the historical background of the period makes it clear that social, political, and economic pressures influenced the magisterial reformers in regard to crucial theological expressions in which they strongly rejected the need for Mosaic civil law in society. The reformers in question restrained or altered their expressions according to the pressures of external circumstances - most importantly war and rebellion spurred by so-called “radical” reformers. As alleged theological positions were weaved with reports and denunciations of violence, Mosaic Law emerged as an allegedly dangerous ideological force, the accusation of which could marginalize opponents. In this crucible of history, in which the long shadows of rebellion and war were cast over Mosaic Law during the mid-1520s and mid-1530s, we find both Luther and Calvin (among others) writing their most vehement denunciations of Mosaic Law. Particularly, we find young Calvin, exiled, sitting down to write his denunciation of “some” who rejected the validity of a commonwealth unless it relied exclusively upon Mosaic civil polity. Luther, Calvin, and others thus warned against applying Moses in the civil realm and linked his laws with sedition and rebellion (even though the association was not accurate in any given case) mainly for their own utilitarian causes. Both Calvin and Luther subsequently employ the doctrine of two kingdoms in distancing themselves and their movements from the need for Mosaic laws in the civil realm, as well as to impede opponents who would use civil power to enforce reforms contrary to them, and yet both act inconsistently when enforcement of the first table of the Decalogue would favor their own reforms. As well, both go on to advance and approve of non-biblical civil laws more invasive and extensive than Mosaic polity would have allowed - including the execution of Anabaptists - all the while denouncing alleged proponents of Moses as dangerous, seditious, barbaric, murderous, and bloodthirsty. ▼ A sensitive assay for the analysis of contaminating segments in pseudorecombinants constructed from a seed-transmissible (CVM-Pg) and a non-seed-transmissible (CMV-Le) strain of cucumber mosaic virus was developed using partial RNase T₁ digestion. Isolated genomic RNA segments produced unique digestion patterns when 3'- end labeled, treated with RNase T₁, and subjected to polyacrylamide gel electrophoresis. Although unique bands were present in these patterns, an overall similarity existed, resulting from putative insertions and deletions in the RNA of each strain. Northern blot hybridization studies of the homology between CMV-Pg and CMV-Le indicated that all 4 heterologous RNA segments contained sequences that were greater than 83% homologous. Homology between the total RNA from each strain was estimated at 70% when assayed by S₁ nuclease digestion following hybridization of RNA to a complementary DNA probe under stringent conditions. A comparison of the electrophoretic mobility of each RNA of both strains in agarose, glyoxal and polyacrylamide gels suggested that each segment was identical in size. RNA segments 1 and 2 when compared to the heterologous strain, however, had discernable differences in the secondary structure. A 5th RNA was detected in both strains. It migrated as two distinct bands when subjected to polyacrylamide gel electrophoresis, and was 3'-end labeled preferentially when present with segments 1-4. Complementary DNA synthesized from RNA 5 of one strain hybridized to segments 1-4 of both strains as determined by northern blot hybridization. RNA 5 of both strains produced identical patterns when subjected to partial RNase T₁ digestion, yet they differed significantly when compared to the digestion patterns from RNA 4 of the homologous strain. These results indicated that RNA 5 of both strains contained virtually identical nucleotide sequences and that they originated from RNA's 1-4, but not from the extreme 3'-terminus. Total CMV RNA, subjected to RNase H treatment after hybridization to a specific deoxyribohexanucleotide, produced two novel bands which were visualized by ethidium bromide staining. Double-stranded cDNA synthesized from RNA of each strain, cloned by blunt-end ligation, produced recombinant clones that contained only inserts of insignificant size. ▼ A survey of major bean (Phaseolus vulgaris) breeding programs and seed producing areas in Idaho, Washington, and Oregon was conducted to determine the occurrence of cucumber mosaic virus (CMV), recently reported to be seedborne in beans. Some 259 plants, sampled from breeding lines of diverse germplasm, were found by enzyme-linked immunosorbent assay (ELISA), bioassay, or gel double-diffusion serology to be free of seedborne CMV. CMV was, however, detected in 1 of 32 Plant Introduction (PI) bean accessions tested by the same methods. Seed-transmission rates of selected isolates (isolates will hereafter refer to those reportedly seedborne and one not reported to be seedborne in beans) were determined under field and greenhouse conditions. An isolate from PI No. 271998 (originating in Spain) was found to be seed-transmitted in up to 49% of the progeny of infected 'Top-crop' bean plants. Seed-transmission depended on early inoculation of plants, i.e., less than four weeks after planting. Rates of transmission under our conditions differed from values reported by others. Most notably, seed-transmission of CMV-F, reported at a rate of 54% in seeds of 'Toperop' (61), was found in our studies to be 3.3% in this cultivar. CMV was distributed uniformly throughout seedlings arising from infected seed but relative virus concentration was most consistently higher near the growing point. Symptoms were mild or masked in plants arising from infected seed. Cucumber (Cucumis sativus 'Lemon') and tobacco (Nicotiana tabacum 'Xanthi') plants infected with CMV were detected by gel serology using standard procedures. To obtain gel serological reactions with infected bean or cowpea tissue, a 0.5 M citrate buffer, pH 6.5 with 0.1% sodium thioglycollate and 0.1% Triton X-100, was employed. CMV was readily detected in infected plant tissues by ELISA. Purified CMV was detected by gel serology at concentrations of 0.05-0.1 mg/ml and by ELISA at concentrations of 6 ng/ml. Isolates were compared for differences in host range, infectivity, serological relatedness, electrophoretic mobility of protein and nucleic acid species, and presence of minor or satellite RNA components. No differences among isolates were found in any of the above properties, except that CMV-Pg seemed to produce lower yields in beans and tobacco and that its major RNA components appeared to fragment into unique electrophoretic species. Otherwise, the greater seed-transmission rate of CMV-Pg was the principal distinction among these isolates. The nucleic acid components of CMV-Pg (seedborne) and CMV-Le (nonseedborne) were separated by sucrose density gradient centrifugation. Pseudorecombinants were prepared among RNA components of these isolates for continuing studies of the genomic determinants of CMV seed-transmissibility in beans.
