▼ In this study, theoretical and computational approaches have been utilized to investigate the mechanisms of natural and artificial
metalloproteases. The active sites of most natural
metalloproteases contain a tetrahedral zinc center, coordinated by three amino acid residues combinated from His(N), Cys(S), Glu(O), and Asp(O) with a water molecule as the fourth ligand. However, the roles played by the ligands environment in the catalytic functions of enzyme are not clear. In this study, the effects of different ligand combinations (NS2, N2S, N2O, N3, S3, NO2 and NSO) in the mechanism were investigated energy barriers were compared. The machanism and energetics of the substrate bound artificial
metalloproteases Ni(II)cyclen (cyclen: 1,4,7,10-tetraazacyclododecane) and Cd(II)cyclen have been investigated. In addition, the mechanism of hydrolysis of Phe-Phe peptide bond catalyzed by another artificial metalloprotease [Pd(H2O)4]2+ has also been studied.

▼ Tuberculosis is considered one of the most prevalent diseases worldwide, claiming the lives of
approximately 1.2 to 1.5 million people each year. It is caused by a member of a class of bacteria
called Mycobacterium (M. tuberculosis), which includes saprophytic and pathogenic species. M.
bovis causes similar disease in cattle. This latter species is used for the production of all currently
available tuberculosis Bacillus Calmette-Guérin (BCG) vaccines. There has been progress made
in the past 20 years in understanding the pathogenesis of Tuberculosis but much more research is
needed to fully understand the mechanisms that mediate the survival of the pathogen in
phagocytic cells. Metalloproteases are a subfamily of proteases that utilize metals, mostly zinc,
for their catalytic activities. They are involved in virulence, cell wall processes and intermediary
metabolism in bacteria. Metalloprotease activity in the culture supernatant of M. tuberculosis has
been detected previously in other laboratories and we are able to detect this activity in the culture
supernatant of M. bovis BCG. Bioinformatics analysis of the proteome of M. tuberculosis reveals
a large number of putative proteases (66 in total) with 23 having the zinc-binding motif
(PROSITE PDOC00129). The goal of this study was the direct identification of the protein responsible for the protease activity in the culture supernatant of M. bovis BCG. The protease in
the supernatant was identified by zymography, with partial purification of the protease activity
by ion exchange chromatography. A band on electrophoresis gels associated with the protease
activity was then subjected to N-terminal sequencing of the purified protein, which identified
hexapeptide sequence VTGGGA. Using a bioinformatics approach, the best matching protein
with this sequence was identified as PstS, a phosphate binding protein normally considered a
component of the phosphate transport system.

▼ Abstract: Trypanosoma cruzi, a protozoan from the family Trypanosomatidae, is responsible for Chagas disease which affects about 6 to 8 million people in Latin America. The origin of this family can be studied through molecular techniques, for instance the investigation of the region V7V8 ¿ SSurRNA. The protozoan is subdivided in six independents groups TcI-TcVI known as Discrete Typing of Units (DTUs). The biological and molecular characterization of eleven strains of T. cruzi from the group TcI (Bolivia; Tlenti; Tmelanocephala; SC90), TcII (Famema; SC96; SI8; Y) and TcIII (QMM3; QMM5; SI5) isolated from five different species of triatomine are able to elucidate biological factors through the kinetic growth, parasitemic curve, cell infection rate, molecular characterization, metalloproteinase action, proteic and serological profile. This investigation was conducted to provide a biological and molecular characterization of T. cruzi isolated from specimens of triatomines. The group TcII of T. cruzi demonstrated higher replicative capacity in epimastigotes forms during the kinetic growth curve, followed by TcI and TcIII. The parasitemic curve demonstrated variability between Balb/c mice; however, the groups TcI, TcII and TcIII showed equivalent parasitemic profile. Furthermore, the cell infection rate in J774 cellular lineages and peritoneal macrophages was used to corroborate the biological data. The group TcI of T. cruzi demonstrated higher infection rate and less time to amastigotes forms multiplication and the peritoneal macrophages showed more attractive to T. cruzi. The molecular characterization through the V7V8 region indicated that these strains belong to DTUs TcI, TcII and TcIII. The groups segregation is important to compare the proteic profile of epimastigotes and tripomastigotes forms of T. cruzi. The groups TcI presented more proteins in acrylamide gels, which can be associated with intra-specif of TcI. The metalloproteinase action was observed in epimastigotes and tripomastigotes forms, demonstrating active and stable presence at the parasite life cycle. The serological reactivity was proven in the groups TcI, TcII and TcIII through ELISA dilutions of 1/100 to 1/12.800. Moreover, the Western Blotting technique was added to SDS-PAGE experiments in infected animals¿ serum after 50 dias and the proteic profile of Y strain was observed in nitrocellulose membrane. The results demonstrated that these eleven strains of T. cruzi have differences between the groups of the parasite. The group TcI showed higher infection rate in cells; TcII, higher value for kinetic growth and TcIII is phylogenetically closer to TcV, which is a hybrid group. The association between parasite and host is able to explain the biological and molecular differences in T. cruzi strains; for this reason, the study of eleven strains isolated from different hosts can add information to the literature and clarify some of the biological aspects of the parasite

▼ Objective: The aim of this study was to investigate the protein expression of KISS-1 gene products in adenomyosis lesions compared with matched orthotopic endometrium, as well as with endometrium from patients without adenomyosis. KPs are multifunctional peptides which have been shown to be inhibitors of tumour metastasis across a range of cancers and are also involved in cell invasion through regulation of matrix metalloproteinases (MMPs). Design: Laboratory, comparative study. Setting: Tertiary academic center. Patient(s): Women with histologically proven adenomyosis after hysterectomy, and women without adenomyosis. Intervention(s): None. Main outcome measure(s): Expression on the protein level of KISS-1 was analyzed immunohistochemically in formalin-fixed, paraffin-embedded adenomyotic tissue (n=29) and normal endometrium specimens (n=29). Result(s): By applying a semiquantitative immunohistochemical evaluation approach, we found that KISS-1 protein expression was higher in adenomyosis lesions compared with matched orthotopic endometrium. Furthermore, KISS-1 protein expression in the orthotopic endometrium of adenomyosis patients was increased compared with that from patients without adenomyosis. Conclusion(s): KISS-1 protein expression appears to be up-regulated in adenomyosis lesions as compared with orthotopic endometrium of adenomyotic patients as well as women without adenomyosis. These findings are suggestive of the involvement of KISS-1 in the pathogenesis and maintenance of adenomyosis.
Σκοπός: Σκοπός της μελέτης αυτής ήταν η διερεύνηση της πρωτεϊνικής έκφρασης του γονιδίου KISS-1 σε αδενομυωσικούς ιστούς συγκριτικά με το αντίστοιχο ορθότοπο ενδομήτριο και με ενδομήτριο ασθενών δίχως αδενομύωση. Οι ΚΡ είναι πεπτίδια με πολλαπλή λειτουργικότητα, τα οποία λειτουργούν αποδεδειγμένα ως αναστολείς της μετάστασης των κακοήθων νεοπλασιών και συμμετέχουν επίσης στην κυτταρική διείσδυση μέσω της ρύθμισης των μεταλλοπρωτεασών της εξωκυττάριας θεμέλιας ουσίας (matrix metalloproteinases - MMPs).Σχεδιασμός: Συγκριτική εργαστηριακή μελέτη.Τόπος διεξαγωγής: Τριτοβάθμιο ακαδημαϊκό κέντρο. Ασθενείς: Γυναίκες με ιστολογικά αποδεδειγμένη αδενομύωση μετά από υστερεκτομή, και γυναίκες χωρίς αδενομύωση. Παρεμβάσεις: Καμία. Κύριες παράμετροι που μετρήθηκαν: Η πρωτεϊνική έκφραση των KiSS-1 αναλύθηκε ανοσοϊστοχημικά σε ιστολογικά παρασκευάσματα τοποθετημένα σε παραφίνη μετά από μονιμοποίηση σε φορμόλη, συγκεκριμένα 29 παρασκευάσματα αδενομυωσικών ιστών (n=29) και 29 παρασκευάσματα φυσιολογικού ενδομητρίου (n=29). Αποτελέσματα: Χρησιμοποιώντας μια ημιποσοτική προσέγγιση ανοσοϊστοχημικής αξιολόγησης, καταλήξαμε στο συμπέρασμα πως η έκφραση της πρωτεΐνης KISS-1 ήταν υψηλότερη σε αδενομυωσικές εστίες συγκριτικά με το αντίστοιχο ορθότοπο ενδομήτριο. ▼ This thesis concerns the investigation of the pathophysiology of idiopathic pulmonary hypertension (IPAH), a rare disease with very severe morbidity and a high mortality rate. During the last 20 years great progress has been achieved in understanding the pathophysiology of the disease, which includes the improvement of both the early diagnosis of patients and pharmacotherapy. The prevalent specific medication is based upon enhancing prostacyclin’s and nitrogen oxide’s action (e.g. sildenafil) as well as the inhibition of endothelin-1 (e.g. bosentani). One of the fields that remain to be explained is the contribution of the extracellular matrix molecules in tissue remodeling observed in IAPH, given that extracellular matrix (ECM) molecules such as glycosaminoglycans (GAG), proteoglycans (PG) and the connective tissue metalloproteases (MMP) involved in tissue remodeling and have been reported to be associated with other lung diseases that exhibit tissue remodeling. In the present study we investigated the role of GAG, PG and MMP in IPAH. The study was performed on homogenized lung tissue and on primary cultures of pulmonary artery smooth muscle cells (PASMC) obtained from 12 patients with IPAH and 10 healthy organ donors. The PASMCs were also investigated for the effect of drugs sildenafil and bosentani in the expression of GAG, PG and MMP, as well as the effect of the transforming growth factor-b1 (TGF-b1) and the bone morphogenetic protein-2 (BMP-2). These two growth factors are key regulators of the synthesis of ECM molecules and they are directly involved in the pathophysiology of IPAH. This system is the closest simulation of an experimental model in tissue remodeling associated with IPAH. It was discovered that the samples of lung tissue and the PASMCs express the following GAGs: HA, heparan sulfate, dermatan sulfate and chondroitin sulfate. A statistically significant increase of HA and reduction of heparan sulfate, dermatan sulfate and chondroitin sulfate was observed in the lungs of patients with IPAH. Also, the molecular mass of HA in the healthy lung tissue was found over 1000 kDa, whereas HA from patients with IPAH was estimated at approximately 200 kDa. These differences in HA levels are owed to the modulation of expression of the enzymes that metabolize it. The samples of lung tissue and PASMCs express the enzymes that synthesize HA: Has1, Has2 and Has3 and the enzymes that degrade HA: Hyal1, Hyal2, Hyal3 and Hyal4, as well as the HA receptors: CD44 and Rhamm. The expression and protein levels of HAS1 are statistically significantly increased, whereas the expression of HYAL1 is reduced in patients with IPAH. The results support the theory that high molecular weight HA is associated with physiological processes, while the low molecular weight HA is associated with inflammatory conditions. ▼ Endometriosis is defined as the presence of functional endometrial tissue, including endometrial glands and stroma cells, outside of the uterus. The most widely accepted theory for the etiopathogenesis of endometriosis is the theory of ectopic implantation of the endometrium through reflux menstruation. In general, the correlation between endometriosis and cancer remains unclear. Numeral studies have shown that various enzymes of the extracellular matrix are active in endometriotic lesions or even in the eutopic endometrium of women with endometriosis leading to self-destruction of the extracellular matrix. Among these various enzymes of the extracellular matrix those better studied include the metalloproteinases’ and the plasminogen system. The MMPs’ system (matrix metalloproteases or metalloproteinases) consists of an enzymatic element, the MMPs and a tissue-inhibitor of metalloproteinases or TIMP. The MMP’s system plays a crucial role in the normal development and growth of the endometrium as well as in various other normal processes in different tissues. Given the necessity for balance between MMPs and TIMPs, it seems reasonable that differential expression between MMPs and TIMPs is correlated with the pathophysiology of endometriosis. Both the ectopic and the eutopic endometrium of women with endometriosis present differential expression of MMPs and TIMPs and this favors tissue infiltration and remodeling of endometriotic tissue. On this basis, it has been hypothesized that a therapeutic intervention at the MMP’s system could inhibit the necessary steps for the induction and progression of endometriosis, thus opening new perspectives for the medical treatment of endometriosis. The aim of the present doctoral thesis was the analysis of protein expression of Tissue Inhibitor of Metaloproteinase 3 (TIMP-3) in the endometrium of patients with and without endometriosis. Tissue samples were collected prospectively from 35 patients with endometriosis and 29 without endometriosis. All women were premenopausal and underwent operative laparoscopy during the proliferative phase of the menstrual cycle; diagnosis or absence of endometriosis was confirmed histologically. Protein expression of TIMP-3 was analyzed immunohistochemically in endometriotic lesions and the eutopic endometrium of women with endometriosis and women without endometriosis. TIMP3 expression was significantly decreased in endometriotic cysts from endometriosis patients as compared with the eutopic endometrium from patients without endometriosis (p = 0.006). On the other hand, in endometriosis patients, TIMP3 expression was decreased in the ectopic as compared with the eutopic endometrium, but this difference was not statistically significantly (p = 0.17). ▼ As metaloproteases AAA/FtsH são componentes chave do controle da qualidade das proteínas inseridas nas membranas de mitocôndrias e cloroplastos. Em Arabidopsis thaliana, as proteases FtsH presentes nas membranas dos tilacóides formam um complexo heterohexamérico composto pelas subunidades FtsH1/FtsH5 (tipo A) e FtsH2/FtsH8 (tipo B). Este complexo está envolvido na reciclagem de proteínas foto-danificadas, especialmente da proteína D1, centro de reação do PSII. Algumas linhas de evidências indicam ainda que existe um limiar de concentração das proteases FtsH, necessário para a correta formação e desenvolvimento dos cloroplastos. Apesar da extensiva caracterização genética e molecular das proteases FtsH, o mecanismo regulatório do complexo FtsH dos cloroplastos não foi totalmente elucidado até o momento, contudo existem evidências de que a sua ativação pode estar relacionada a alta incidência luminosa e a outras condições de estresse. A presença de fatores proteicos auxiliares, foi testada como hipótese alternativa por nosso grupo, através do uso da protease FtsH5 como isca em um ensaio de duplo híbrido de leveduras. Este ensaio identificou uma proteína interagente putativa, nomeada FIP (FtsH5 Interacting Protein), a qual comprovadamente interage com FtsH5 e está localizada nas membranas dos tilacóides. De modo a investigar o papel regulatório putativo de FIP sobre a atividade do complexo FtsH, nós analisamos os padrões de expressão em uma ampla gama de condições de estresse a partir de dados públicos de microarranjos de DNA. Os perfis de expressão indicam que FIP pode ser um regulador negativo da atividade do complexo. Os resultados também sugerem que o complexo pode estar envolvido na resposta do cloroplasto a diferentes tipos de condições de estresse. O estudo da história evolutiva das proteínas interagentes FtsH5 e FIP evidenciou que as sequências homólogas a FIP são encontradas exclusivamente em musgos e plantas superiores, sugerindo assim que a origem de FIP pode estar relacionada a colonização terrestre. Todos os genes codificantes das proteases FtsH do complexo foram usados como \"query ̈na busca por sequências homólogas, permitindo a classificação das proteases FtsH nos tipos A e B por inferência filogenética Bayesiana. Análises filogenéticas Bayesianas também foram feitas para FIP e as proteases FtsH tipos A e B, independentemente. A análise Mirrortree suportou a existência de coevolução entre FIP e as proteases FtsH tipo A. Por outro lado, nenhuma correlação foi encontrada entre FIP e as proteases FtsH tipo B, o que corrobora nossas observações experimentais anteriores. Além disso, o agrupamento portador de homólogos FIP pôde ser recuperado em uma filogenia mais completa das proteases FtsH do tipo A. ▼ A doença periodontal (DP) e o diabetes mellitus (DM) são doenças inflamatórias crônicas, e quando associadas aumentam a destruição da região periodontal. Por esta razão, o objetivo do nosso trabalho foi avaliar o papel do diabetes sobre o processo inflamatório, reabsorção óssea alveolar, produção de metaloproteinase de matriz (MMP), colágeno e TGF-β1 em camundongos com DP experimental. Foram utilizados 40 camundongos Balb/c machos de 18 – 22 g, distribuídos igualmente em 4 grupos, sendo estes, normal (N), normal com doença periodontal (N+DP), diabético (DM) e diabético com doença periodontal (DM+DP). O diabetes foi induzido por tratamento com estreptozotocina 200 mg/kg por via intraperitoneal. Passados 7 dias os índices glicêmicos foram verificados, e os animais foram considerados diabéticos quando a glicemia apresentava-se igual ou superior a 250 mg/dL. Posteriormente, os camundongos do grupo com DP foram submetidos a uma ligadura em torno dos primeiros molares inferiores, e no 15º dia, estes animais foram eutanasiados e as amostras foram coletadas para as análises seguintes. O tecido gengival marginal foi coletado para as análises de marcadores inflamatórios, tais como, níveis de CXCL2, CCL3 e TGF-β1 por ELISA, expressão gênica de MMP-2, MMP-9, TGF-β1 e col1a2 por RT-PCR em tempo real, e atividade enzimática de MMP-2 e MMP-9 por zimografia. Os parâmetros de reabsorção de osso alveolar, recrutamento de neutrófilos e quantificação de colágeno também foram avaliados, pelo processamento histológico das hemimandíbulas, os quais apresentaram diferença significativa no grupo DM+DP. A DP foi capaz de aumentar a produção de CXCL2, CCL3 e TGF-β1, contudo somente a produção de CXCL2 e CCL3 foi aumentada no grupo DM+DP. A expressão gênica de MMP-2 não apresentou diferença estatística nos grupos analisados, em contrapartida a expressão de MMP-9 foi diminuída com a DP, apresentado diferença significativa no grupo DM+DP, assim como a expressão de TGF-β1 e col1a2. Nos grupos com DP, houve aumento significativo na atividade de MMP-2 e MMP-9, com potencializada atividade de MMP- 2 no grupo DM+DP. Em resumo, nós observamos que o DM mais a DP agravam a resposta inflamatória e processo destrutivo na região periodontal, caracterizada por aumento da migração de neutrófilos, reabsorção óssea, aumento na produção de CXCL2 e CCL3, aumento na atividade de MMP-2 e diminuição da porcentagem de colágeno, sugerindo assim um desequilíbrio no reparo tecidual da região periodontal.

▼ Abstract: The effect of Periodontal Disease (PD) in the pulp is not well known. Bactéria and their products play an important role in the development and perpetuation of pulp and periodontal diseases. Endotoxin (LPS) is a lipopolysaccharide found in the cell wall of gram-negative bactéria and it is a potent stimulator of innate response of the host defense and can lead to irreversible tissue damage and bone destruction. The metalloproteinases (MMPs) are enzymes capable of degrading extracellular matrix proteins including collagen, especially during the destruction of the periodontal tissues. The aim of this study was investigate in periodontal pockets (PP) and root canal (RC) the microbiological, endotoxic content and metalloproteinases levels of teeth with positive pulpal response and periodontal disease, before and after mechanical-chemical preparation, and after intracanal medication. It was investigated: 1) the profile and microbial diversity, by simple PCR and sequencing of the gene 16S rRNA; 2) quantify the LPS levels, determining endodontic and periodontal content; 3) determine the MMP-2, MMP-3, MMP-8, MMP-9 and MMP-13 levels in periodontal and endodontic tissues; and 5) evaluate the effect of chemical-mechanical preparation (CMP) with chlorhexidine gel 2% (CLX), EDTA 17% and intracanal medication (ICM) in the reduction of microorganisms and LPS. 32 teeth with PD and positive pulp response that were under periodontal treatment for at least 6 months were selected. Samples were collected at three different times of endodontic therapy: before endodontic treatment (C1), after CMP (C2), and after ICM (C3) for 30 days. Apyrogenic/sterile paper points were used. After the CMP teeth were divided into groups according to the presence or absence of ICM: single visit G1 (n = 10); GII - Ca(OH) 2 + CLX (n = 11); GIII - Ca(OH) 2 + saline solution (SS) (n = 11). PCR and pirosequencing of gene16S rRNA were used for bacterial investigation. The LAL assay was used to quantify LPS levels and the amounts of MMPs were measured by ELISA assay. The Friedman test were used to assess the micro-organisms by PCR and in each endodontic stage. The Wilcoxon test was used to endotoxins levels. The linear regression for the correlation between bacterias and endotoxins. Pearson for bacteria correlation. For assess the MMPS levels, ANOVA and Tuckey test were used. The level of significance was 5% (p <0.05). The results showed that: 1) through the PCR targets bactérias were identified in PP and RC in all stages of endodontic treatment, but more often in PP. After ICM, Porphyromonas gingivalis and Parvimonas micra were prevalent in RC and PP; On the other hand, the ICM diversified the microbiome in both sites; Through sequencing it was observed that the initial microbiome of PP are different from initial RC, with predominance of gram-negative species in PP. 2)LPS were recovered in 100% of the initial PP, after CMP and ICM with median of 132 EU/mL, 90,4 EU/mL e 46,85 EU/mL respectively. ▼ Abstract: In this work we described the purification, structural characterization and the local and systemic effects of a new low molecular weight snake venom metalloproteinase, named BtaHF. This protein was purified from the Bothriopsis taeniata crude venom combining conventional molecular exclusion (Sephadex G-75 column) followed by an ion exchange (DEAE 8HR APMinicolumn) chromatography on a HPLC system. The new metalloproteinase BtaHF showed a high degree of purity and molecular homogeneity with a molecular mass of 25968.16 Da. BtaHF showed caseinolytic activity with an optimum temperature and pH of 37-40 °C and 8, respectively. The caseinolytic activity was inhibited by EGTA, EDTA and DTT, but PMSF and SBT-I did not show inhibitory effect. The Ca+2 ion shown to be important for protein stability enhancing its caseinolytic activity, on the contrary, Z+2 and Mn+2 showed inhibitory effects upon the enzymatic activity of this protein. The metalloproteinase BtaHF is an ?-fibrigenolytic enzyme, rapidly degraded fibrinogen A? chain within 15 minutes, while fibrinogen B? chain is degraded after 6 hours. This enzyme did not show fibrinolytic or arginine amidase activities BtaHF is an acidic protein due the mayor proportion of acidic amino acid in its amino acid composition with 6 cysteinyl residues. By the homology study BtaHF share a high sequence identity other weakly hemorrhagic P-I class SVMP such BaP1 isolated from Bothrops asper snake venom This new metalloproteinase, BtaHF, have a weak hemorrhagic with a minimum hemorrhagic dose of 20.14 ?g. Also, this metalloproteinase showed dose-dependent paw edemaforming activity, this effect remain six hours after inoculation. Both hemorrhage and edemaforming activities were related to the enzymatic activity and correct fold of BtaHF, since chelating (EDTA and EGTA) and reducing (DTT) agents inhibited these activities. On the other hand, this enzyme did not show miotoxic activity, although, showed citotoxic activity on C2C12 fibroblast. Histological analysis of mice gastrocnemius muscle confirmed that the metalloproteinase produce local hemorrhage and pro-inflammatory effects, as well as, the absence of miotoxic activity. This metalloproteinase, BtaHF, produce specific systemic effects when injected endovenously in mice. On lung tissue level these toxin produce alteration of alveolus structure with significant hemorrhage, thickness of alveolus septum and inflammation. On renal tissue BtaHF did not produce hemorrhage, but, was observe alteration of glomerulus structure, along with cytoplasmatic alterations of distal and proximal tubulus cells. On the other hand, were not observed any alterations on hepatic tissue. BtaHF deplete completely plasma fibrinogen levels in a time and dose-depended manner, reaching blood incoagulability after one hour. ▼ Abstract: Lipopolysaccharides (LPSs, known as endotoxins) present on the outer layers of Gram-negative bacterial envelope, and released during bacteria multiplication and death, can egress into periradicular tissues, acting as one of the most potent stimuli for immunocompetent cells in the release of inflammatory mediators and matrix metalloproteinases (MMPs). Thus, the objectives of this study were: 1) To evaluate the influence of primarily infected root canal contents (bacterial and endotoxin contents) on host-immune response by interleukin (IL)-1?, IL-1?, tumor necrosis factor ? (TNF-?), prostaglandin E2 (PGE2), matrix metalloproteinases (MMP) MMP-2, MMP-3, MMP-8, MMP-9 and MMP-13 production and to correlate their levels with clinical features (chapter 1); 2) To investigate the influence of 17% ethylenediaminetetraacetic acid (EDTA) ultrasonic activation after chemo-mechanical preparation (CMP) on eliminating/reducing oral bacterial lipopolysaccharide (LPS, known as endotoxins) and cultivable bacteria in teeth with pulp necrosis and apical periodontitis (chapter 2); 3) To investigate the endotoxin levels from primary endodontic infection before and after CMP, and to determine their antigenicity against 3T3 fibroblasts through gelatinolytic activity of MMPs (chapter 3). Methods: Root canal content samples were taken from 24 primarily infected root canals with apical periodontitis by using sterile/apyrogenic paper points. Samples were taken at different clinical times: S1- before CMP; S2- after CMP; S3- after EDTA: G1- with ultrasonic activation (n=12) and G2- without ultrasonic activation (n=12). PCR technique (16S rRNA) was used for the detection of the target bacteria. Culture techniques were used to determine the number of colony-forming units (CFU). Limulus Amebocyte Lysate (LAL) was used to measure endotoxin. Cytokines and MMPs levels were measured by enzyme-linked immunosorbent assay (ELISA) from samples that were taken passing 2 mm through the root apex after CMP. The levels of MMP-2 and MMP-9 gelatinolytic activity were measured using the zymography technique. The Pearson coefficient was used to correlate the amount of endotoxins with cytokine and MMP levels. Clinical data were set as dependent variables and correlated by individual correlation with each cytokine and MMP level. Friedman¿s and Wilcoxon tests was used to compare the amount of bacteria and endotoxin contents at each clinical time. Data obtained from gelatinolytic activity was analysed using Anova and Tukey¿s tests. The significance levels always were set at 5% (p<0,05). Results: IL-1?, IL-1?, TNF-? and PGE2, as well as, MMP-2, MMP-3, MMP-8, MMP-9 and MMP-13 were detected in all samples. A correlation between endotoxin levels with cytokines and MMPs production was found (p<0.05). Root canal exudation was positively correlated with high levels of TNF-?, and symptomatic teeth were correlated with IL-1?, PGE2 and MMP-8 (p<0.05). ▼ Introdução: A Organização Mundial da Saúde estimou para o ano de 2008 aproximadamente 1.38 milhões de casos novos de câncer de mama no mundo e 458 mil mortes. A maioria dos casos (56%) e das mortes (64%), ocorrendo em países economicamente desenvolvidos. Apesar dos avanços e do diagnostico precoce, um número significativo de mulheres com tumores da mama operáveis apresentando evolução desfavorável vêm à sucumbir devido ao surgimento de doença metastática. Uma melhor compreensão da heterogeneidade do tumor e das características microambientais subjacentes, bem como dos mecanismos e as consequências das suas interações é essencial para melhorar o direcionamento das terapias existentes e desenvolver novos agentes terapêuticos para o câncer. Objetivos: Descrever as características clínicas, anatomopatológicas e imuno-histoquímicas de um grupo de pacientes com câncer de mama operável, e estudar o impacto destas características no estadiamento da doença, sobrevivência livre de recorrência e sobrevivência global. Além disto, analisar as potenciais correlações existentes entre estas características. Métodos: Estudo de coorte retrospectiva de base hospitalar envolvendo 86 mulheres com câncer primário de mama, submetidas a tratamento entre julho de 1999 e dezembro de 2004, no Serviço de Mastologia do Hospital de Clinicas de Porto Alegre. Dados clinicopatológicos e imuno-histoquimicos (RE, RP, HER2, Ki67 e p53) foram coletados dos registros hospitalares. Expressão do VEGF, MMP-2, MMP-9, TIMP-1 e TIMP-2 foram analisadas através de imuno-histoquimica. Variáveis contínuas foram analisadas pelo coeficiente de correlação de Spearman, ou pelo teste não paramétrico U de Mann-Whitney e H de Kruskal-Wallis, quando comparadas com variáveis categóricas. Variáveis categóricas foram analisadas pelo teste 2 de Pearson. Estimativas da probabilidade de sobrevivência foram obtidas pelo estimador não paramétrico de Kaplan-Meier e pelo semiparamétrico modelo de regressão de Cox. Comparação entre as curvas de sobrevivência foi realizada pelo teste estatístico de log-rank. O IC foi calculado em 95% e valores p< 0,05 foram considerados estatisticamente significativos. Resultados: A sobrevivência livre de recorrência em 5 e 10 anos foi de 82,2% e 68%, e a global foi de 90,2% e 82,9%, respectivamente. Número de linfonodos positivos (p= 0,00; p= 0,03), diâmetro tumoral (p= 0,01; p= 0,01) e estádio (p= 0,00; p= 0,02) são fatores de risco isolado para recorrência e óbito, respectivamente. Superexpressão de HER2 é um fator de risco isolado para recorrência (p= 0,04). Existe uma correlação positiva significativa entre: VEGF e MMP-9 (rs: 0,246; p= 0,023); TIMP-2 e MMP-2 (rs: 0,358; p= 0,001). Também foram encontradas associações significativas entre as variáveis: a) VEGF e receptor de progesterona positivo (p= 0,045); b) TIMP-2 e idade ≥ 50 anos (p= 0,002), e diâmetro ≤ 2,0 cm (p= 0,016); c) TIMP-1 e menarca ≤ 12 anos (p= 0,038); d) Maior diâmetro e alto grau histológico (2: 19,3; p= 0,004), invasão vascular (2: 12,6; p= 0,006), status do linfonodo…

▼ A resistência que apresentam as serpentes peçonhentas às suas próprias peçonhas, assim como a resistência observada em alguns animais está bem documentada, e é atribuída a fatores solúveis presentes no seu plasma, soro ou músculo. No caso das serpentes peçonhentas estes fatores as amparam de sofrer danos decorrentes da própria peçonha. No presente trabalho foi isolado do soro da serpente Bothrops alternatus um inibidor de metaloproteases, denominado BaltMPI, mediante cromatografia em DEAE Sepharose(TM), Superdex(TM) 200, MonoQ(TM) 5/50 GL e cromatografia de fase reversa em C18, com uma recuperação proteica de 0,3%. A massa molecular determinada para o BaltMPI por SDS - PAGE foi de 60,5 kDa, e por espectrometria de massas MALDI/TOF de 42,4 kDa. Seu ponto isoelétrico, determinado por focalização isoelétrica, é 5,27. Portanto, o BaltMPI, é um SVMPIs com caráter ácido, pertencente aos SVMPIs de baixa massa molecular. Os primeiros 60 aminoácidos da região N-terminal do BaltMPI foram determinados mediante degradação de Edman, e apresentou alto grau de homologia com a sequência para a mesma região de outros SVMPIs isolados a partir do soro de serpentes peçonhentas. Outros segmentos da sequência também foram determinados após clivagem com tripsina e posterior análise por espectrometria de massas. Posteriormente realizou-se o alinhamento da sequência parcial determinada para o BaltMPI, com a sequência do BJ46a, um SVMPI proveniente da Bothrops jararaca, encontrando alta homologia entre elas. Tanto o soro da serpente, quanto o inibidor, BaltMPI, inibiram a atividade hemorrágica da Batroxase, uma SVMP da classe P-I, e a da BjussuMP-I, uma SVMP da classe P-III. Já, em relação à DHM da Batroxase, foi determinado que o BaltMPI tem uma DIHM de 5ug, e uma CE50% de 0,857ug. O BaltMPI também apresentou um efeito inibitório sobre a ação proteolítica da Batroxase, sobre os substratos: fibrinogênio, fibrina e azocaseína. Sobre a atividade fibrinogenolítica da serinoprotease BjSP, o BaltMPI não apresentou um efeito inibitório, corroborando assim com a especificidade descrita para inibir as SVMPs que possuem os SVMPIs. O BaltMPI inibiu a ação hemorrágica e proteolítica da Batroxase, ao formar um complexo mediante ligações não covalentes com esta SVMP. O inibidor BaltMPI, ao igualmente aos demais SVMPIs descritos, é estável em uma ampla faixa de pH (1 - 9), e a temperaturas elevadas, sendo que em temperaturas acima de 60°C foi observada uma diminuição na sua capacidade de inibir a atividade hemorrágica da Batroxase. A inibição da atividade hemorrágica da Batroxase, quando avaliado o potencial do BaltMPI como complemento à soroterapia, foi menor à inibição observada ao realizar os testes com incubação prévia entre o inibidor e a metaloprotease. ▼ Lesão hepática é uma das complicações decorrentes de pancreatite aguda (PA) e mostra uma correlação positiva com a gravidade da doença. Recentemente foi demonstrado que tratamento com solução hipertônica (SH) reduz inflamação e mortalidade na PA. O presente trabalho investigou os efeitos de SH na modulação do estresse oxidativo, apoptose e remodelamento tecidual no fígado durante a PA. Ratos machos foram divididos em quatro grupos: C animais que não foram submetidos à PA ou tratamento; past animais submetidos à indução de PA e que não receberam tratamento; pas animais submetidos à PA e tratados com solução fisiológica (NaCl 0.9%); pah animais submetidos à indução de PA e que receberam SH (NaCl 7.5%). PA foi induzida pela injeção retrógrada de ácido taurocólico (2.5%) no ducto bilo-pancreático. Após 4, 12 e 24 horas os animais foram sacrificados para coleta das amostras de interesse (fígado e plasma) submetidas a ensaios para análise de expressão (western blot e PCR) de Óxido Nítrico Sintase induzível (iNOS), metaloproteinases (MMP) -2 e -9, Heat Schock Protein (HSP) 47, 60, 70 e 90, colágenos tipos I e III, caspases 2 e 9, APAF-1 e AIF, formação de nitrotirosina (imunohistoquímica), peroxidação lipídica (TBARs), atividade de alaninoaminotransferase (ALT), produção de nitrito/nitrato (Reação de Griess) e das citocinas TNF-, IL-1, IL-6 e IL-10 (ELISA). O tratamento com SH reduziu significantemente o estresse oxidativo hepático com a redução da expressão gênica de iNOS (p<0,01 vs. pas), dos níveis de nitrito/nitrato (P<0,01 vs. pas), liberação de ALT (p<0,01 vs. pas) e inibição da formação de peroxinitrito após 12 horas da indução de PA. Conseqüentemente, a expressão de HSP70 não foi ativada devido à proteção hepática causada pela administração de SH. A expressão e atividade de MMP-9 aumentaram significativamente nos grupos pas e past, entretanto o tratamento com SH manteve os níveis basais (p<0,05 vs. past, pas). Aumento da expressão de HSP47 e alterações na expressão de colágeno indicaram intenso remodelamento de matriz extracelular nos grupos não tratados ou tratados com solução fisiológica, permanecendo semelhante ao controle no grupo pah. Não ocorreram mudanças significativas na expressão das proteínas envolvidas no processo apoptótico. A SH diminui o estresse oxidativo no período crítico da PA, resultando na diminuição da lesão hepática e do remodelamento tecidual, mantendo dessa forma a integridade de matriz extracelular

▼ A matriz extracelular e as células musculares lisas das vias aéreas estão intimamente interrelacionadas. Poucos estudos porém, avaliaram a composição dos diferentes componentes da matriz extracelular e seus reguladores na camada do músculo liso brônquico na asma. Utilizando um programa de análise de imagens, a área fracionada do colágeno total e das fibras elásticas foi quantificada no interior do músculo liso brônquico de 35 indivíduos que faleceram devido a um ataque de asma (Asma Fatal), e comparada com 10 casos de indivíduos com asma e que faleceram de outras causas (Asma Não Fatal), e com 22 indivíduos controles sem patologia pulmonar. Expressão dos colágenos I e III, fibronectina, versicam, metaloprotease (MMP)-1, 2, 9 e 12, e inibidores de metaloprotease 1 e 2 foram quantificados no interior do músculo liso brônquico de 22 casos de asma fatal e 10 controles. Nas grandes vias aéreas dos casos de asma fatal, a área fracionada das fibras elásticas foi significativamente maior na camada do músculo liso brônquico quando comparada com os grupos de Asma Não Fatal e Controle. Semelhantemente, fibronectina, MMP-9 e MMP-12 estavam aumentadas no músculo liso das grandes vias aéreas nos casos de asma fatal quando comparadas aos controles. Apenas aumento das fibras elásticas foi observado nas pequenas vias aéreas na Asma Fatal, e somente quando comparadas aos casos de Asma Não Fatal. O conjunto dos resultados mostra que há uma composição alterada dos elementos da matriz extracelular e um ambiente de degradação protéica no músculo liso brônquico de indivíduos que morreram por asma, o qual pode acarretar importantes conseqüências nas funções sintéticas e mecânicas do músculo liso das vias áreas.
There is an intimate relationship between the extracellular matrix (ECM) and smooth muscle cells within the airways. Few studies have comprehensively assessed the composition of different ECM components and its regulators within the airway smooth muscle (ASM) in asthma. With the aid of image analysis, the fractional area of total collagen and elastic fibres was quantified within the ASM of 35 subjects with Fatal Asthma (FA) and compared with 10 Nonfatal Asthma (NFA) patients and 22 nonasthmatic control cases. Expression of collagen I and III, fibronectin, versican, matrix metalloprotease (MMP)-1, 2, 9 and 12 and tissue inhibitor of metalloprotease-1 and 2 was quantified within the ASM in 22 FA and 10 control cases. In the large airways of FA cases, the fractional area of elastic fibres within the ASM was increased compared with NFA and controls. Similarly, fibronectin, MMP-9 and MMP-12 were increased within the ASM in large airways of FA cases compared with controls. Elastic fibres were increased in small airways in FA only in comparison with NFA cases. The results shower that, there is altered extracellular matrix composition and a degradative environment within the airway smooth muscle in fatal asthma patients, which may have important consequences for the mechanical and synthetic functions of airway smooth muscle.

▼ Cellular senescence is considered to be a potent anticancer mechanism. However, it has been proposed that senescent stromal cells may enhance the growth of adjacent malignant epithelial cells. Exposure of tumours to repeated low doses of chemotherapeutic drugs and ionizing radiation is a common treatment regime in several tissues. However, the effect of this stress to the neighboring stromal cells and the interaction of the latter with cancer cells has not been adequately investigated. In this study we have exposed confluent cultures of human lung fibroblasts and human bone marrow-derived mesenchymal stem cells (hBM-MSC) to repeated subcytotoxic doses of Taxol, Cisplatin, Doxorubicin, STI571 and ionizing radiation. We have found that a single dose of Cisplatin and Doxorubicin, but not Taxol and STI571, immediately activates a DNA damage response, as shown by the activation of ATM/Chk2/p53/p21CIP1/WAF1 axis. After a series of doses with Cisplatin, Doxorubicin and Taxol but not STI571, cellular senescence was accelerated, as shown by morphological alterations, growth arrest, p21CIP1/WAF1 and p16INK4A upregulation and senescence-associated β galactosidase staining. Interestingly, we found that in the case of γ-radiation-induced premature senescence this process is p53-dependent. Next, we studied the effect of these prematurely senescent cells on the growth of human malignant lung cell lines (A549). Medium conditioned by young and prematurely senescent cells has a slightly inhibitory effect on the proliferation of A549 cells. Furthermore, in co-culture studies we have found that the growth of cancer cells was merely enhanced when cultured on drug-induced prematurely senescent fibroblasts. In addition, in immunocompromised mice taxol-induced, as well as cisplatin-induced senescent cells intensely promoted A549 cells to form tumours. In addition, prematurely senescent fibroblasts secrete MMP that are in part responsible for the synergy between senescent and cancer cells in tumor growth.Furtermore we investigated the effect of chemotherapeutic drugs and in some cases ionizing radiation on the ability of hBM-MSC towards adipogenic, osteogenic and chondrogenic differentiation and we found that both chemotherapeutic drugs and ionizing radiation significantly diminished MSCs’ ability towards differentiation. These findings support the idea that replicative- or stress-induced senescence of stromal cells represent a major challenge for tissue homeostasis.

▼ Abstract: The Achilles tendon has a high incidence of rupture, particularly in sports activities. The healing process leads to a disorganized extracellular matrix (ECM) which reduces its tensile strength and lead to a high rate of injury recurrence. Low level laser therapy (LLLT) has been effective, but there are conflicting results regarding their protocols. Our purpose was to evaluate the effects of different therapies with LLLT in partially tenotomized Achilles tendon. Adult male Wistar rats were divided into 7 groups: G1 ? intact; G2 ? injured; G3 ? injured + LLL (4 J/cm2 continuous); G4 ? injured + LLL (4 J/cm2 ? 20 Hz); G5 ? injured; G6 ? injured + LLL (4 J/cm2 continuous); and G7 ? injured + LLL (4 J/cm2 ? 20 Hz until the 7th day and 2 kHz from 8?14 days). G2, G3 and G4 were euthanized 8 days after injury, and G5, G6 and G7 were euthanized on the 15th day. Quantification of non-collagenous protein (NCP), glycosaminoglycans (GAG) and hydroxyproline (HOPro) was performed, in addition to electrophoresis on agarose gel, zymography for MMP-2 and -9, western blotting (WB) for collagen types I and III. The structural analysis was done by hematoxylin-eosin (HE) and toluidine blue (TB) staines with in addition to birefringence measurements and analysis by polarization microscopy. Functional analysis was performed using the gait assessment of the animals in the Catwalk. Regarding HOPro there was a significant decrease in all groups when compared to the G1, except G7. The dosage of GAG showed an increase in all groups except G5 and agarose gel showed an increase in the content of dermatan sulfat in all transected groups, being less expressive in G4 and G7. In sections stained with TB the tenotomized groups showed metachromasia, particularly the treated groups. WB for collagen I showed an increase in G4 and G7. In G4 collagen III showed higher values when compared to G2. The zymography for MMPs-2 showed higher values for G4 and G7. MMP-9 increased in G3 and G4. Analysis of birefringence showed marked disorganization in all groups, with a significant improvement in G7. The catwalk results showed that after surgery, G4 had better results when compared to other groups. Our results show that the continuous and pulsed LLLT has different effects on the process of tendon repair. Pulsed laser acted on the inflammatory process improving the gait and promoting collagen synthesis and organization. Thus we believe that this treatment protocol can be adapted for use in rehabilitation clinics in order to accelerate the repair and enhance the morphological, biochemical and functional characteristics of the tendon

metalloproteases, composed of alpha and/or beta subunits, consist of membrane-bound and secreted forms that are abundantly expressed in kidney and intestinal epithelial cells. They are also expressed in the skin and in certain populations of leukocytes. Meprins have been implicated in several inflammatory diseases, such as renal ischemia, diabetic nephropathy, and inflammatory bowel disease, indicating a role for these enzymes in modulation of the immune response. Prior to the initiation of this work, extensive information about the structure and in vitro behavior of the meprins was known, but little was known about their in vivo roles in immune modulation. These studies are the first to demonstrate a relationship between the inflammatory response and the meprins in both systemic and bladder challenge models.
The aim of this work was to determine the role of meprins in the host response to gram-negative uropathic Escherichia coli (E. coli). Initial studies demonstrated marked increases in meprin alpha expression in the urine of women with active urinary tract infections (UTIs), implicating meprin involvement in the host response to bacterial infections. To examine further the role of meprins in the host response to UTI, meprin alpha knockout (alphaKO) and wild-type (WT) mice were challenged with a transurethral inoculation of uropathic E. coli. In this localized model of inflammation, bladder myeloperoxidase (MPO) activity, bladder weight, and bladder permeability were significantly less in alphaKO compared to WT mice after induction of UTI. These data indicate that meprin A is pro-inflammatory, contributing to leukocyte infiltration, edema, and epithelial damage. To determine whether the action of meprin A was the result of a direct interaction with E. coli, extensive in vitro studies were carried out. Meprin A did not decrease the binding of E. coli to bladder cells in culture, nor did it degrade the pili that are responsible for the attachment of E. coli to the bladder epithelium. No evidence of direct interactions between meprin A and E. coli has been found, indicating that the differential response observed in meprin alphaKO mice in comparison to WT is indirect and likely involves the immune system.
A large component of the immune response to E. coli is directed toward lipopolysaccharide (LPS) in the bacterial cell wall. Therefore, the responses of meprin alphaKO, meprin beta knockout (betaKO), meprin alphabeta double knockout (alphabetaKO), and WT mice after intraperitoneal (i.p.) LPS challenge were examined. Genotype-specific differences in response to LPS were observed as early as 1 h after challenge with 2.5 mg/kg i.p. E. coli LPS. Meprin alphaKO mice displayed a decreased systemic response to LPS compared to WT mice and meprin betaKO mice, as indicated by lower blood urea nitrogen (BUN) levels, lower serum TNFalpha levels, and less severe hypothermia, implicating meprin in modulation of the host immune response to endotoxin. ▼ En 2017, il a été estimé que 425 millions de personnes souffraient de diabète. Les raisons de ce boom, liées au contexte socio-économique actuel, représentent une dépense globale de 1.3 milliard de dollars par an avec des estimations atteignant jusqu’à 2.1 milliards de dollars en 2030 (Bommer et al., Diabetes Care, 2018). Au-delà de dérèglements physiologiques, cette maladie peut engendrer de nombreux effets secondaires si elle n’est pas traitée correctement, notamment la formation de plaies chroniques, plaies ne suivant pas le schéma de cicatrisation habituel. Parmi les différentes plaies chroniques se trouvent les plaies du pied diabétique avec une prévalence globale de 6,3% (Sen, Advances in Wound Care, 2019) et une probabilité de développement chez les personnes diabétiques de 25% (Boulton et al., The Lancet, 2005). Ces plaies possèdent de nombreux facteurs participant à ce phénomène de pérennisation, dont les métalloprotéases matricielles ou MMP, endopeptidases à zinc aux rôles variés.Ces travaux de thèse s’intéressent à l’élaboration d’un dispositif de pansement connecté pour le suivi de l’évolution des plaies du pied diabétique. Ce suivi se fait par mesure de la concentration d’une enzyme clé dans les phénomènes d’inflammation de ce type de plaie, la métalloprotéase matricielle 9 (MMP9), également appelée Gelatinase B. Ce système se base sur l’utilisation d’alumine poreuse fonctionnalisée avec un Élément de Reconnaissance Biologique (ERB) capable d’interagir avec la cible, MMP9.Dans un premier temps, une recherche d’ERB affine pour la cible est étudiée. Cette recherche se base sur un screening de banques d’anticorps de camélidé (VHH) et de banques peptidiques par la méthode de phage display.La suite du projet se focalise sur l’élaboration de deux prototypes de biocapteurs capacitifs, l’un avec l’étude de la fonctionnalisation d’un anticorps conventionnel anti-MMP9, l’autre avec l’utilisation d’un VHH tiers, affine pour la GFP. Ces deux preuves de concept seront utiles pour la continuité du projet.
In 2017, it was estimated that 425 million people worldwide had diabetes. The main reason for this skyrocketing number is related to the current socio-economic context of the world with global expenses reaching 1.3 billion dollars a year with estimates reaching up to 2.1 billion dollars by 2030 (Bommer et al., Diabetes Care, 2018). Above all physiological unbalances, this disease can cause many side effects if left untreated, one of them being chronic wounds, wounds that don’t follow the usual healing process. Among the different types of chronic wounds is diabetic foot ulcers (DFUs), with a prevalence of 6.3% (Sen, Advances in Wound Care, 2019) and a lifetime risk for the diabetic population as high as 25% (Boulton et al., The Lancet, 2005). ▼ La membrane basale (MB) est une feuille dense de matrice extracellulaire spécialisée qui sépare l'épithélium du tissu sous-adjacent. La pénétration des cellules à travers les barrières des MB, c’est appelée «invasion», est un processus important pour le développement normal des tissus et dans la métastase du cancer. Beaucoup a été compris sur la génétique et la signalisation sur comment les trous sont formés dans le MB pendant de l'invasion ont été compris. Cependant, les forces physiques impliquées sont moins comprises: comment la contractilité de la myosine participe à l'élimination de la MB et comment les différents facteurs de polymérisation de l’actine et les protéines de réticulation contribuent au processus invasif. Pour répondre à ces questions, nous avons étudié un événement d'invasion dans un processus de développement, l’invasion de la cellule anchre (AC) chez Caenorhabditis elegans. La rupture de la MB par l’AC est connue pour dépendre d'une protrusion riche en actine et de l'activité des métalloprotéases (MMP), similaire à l'invasion de cellules cancéreuses. Inactivation génique par RNAi de différents activateurs et nucléateurs de la polymérisation d'actine, et l'expression spécifiquement dans l'AC d'une forme négative dominante d'un activateur du complexe Arp2/3 a montré que l'invasion de l’AC dépendes fortement des filaments ramifiés formés via l'activation WASP / WSP-1 du complexe Arp2/3. La microscopie à haute résolution a indiqué que la protrusion invasive de l’AC était densément assemblée, en accord avec l'idée que la protrusion invasive était fortement ramifiée. Nous avons également montré qu'un autre activateur du complexe Arp2/3, WAVE / WVE-1, pouvait permettre une invasion lorsque WASP / WSP-1 était absent. Les formines semblaient ne pas de jouer un rôle majeur et les protéines de réticulation d'actine étaient également dispensables pour l’invasion de l’AC. Dans les vers de type normaux, nous avons observé que l'activité de la myosine n'était pas nécessaire pour l'invasion. Cependant, il a été rapporté que les cellules cancéreuses augmentent la contractilité de la myosine pour envahir en l'absence de protéases, nous avons donc utilisé un ver sans les cinq principales MMPs du génome du ver pour tester le rôle de la myosine dans ce contexte. L'invasion de l’AC a eu lieu dans en l’absence des MMPs, mais avec un retard. L’inactivation génique par RNAi de différents composants lies à l’activité de la myosine n'a pas amélioré le défaut d'invasion. En plus, la visualisation du cytosquelette d'actine dans les vers sans MMPs a révélé que l'actine était concentrée dans la protrusion de l’AC et à peine détectable dans le cortex, ce qui rendait improbable que la contraction de la myosine du cortex aiderait la compression cellulaire à travers de la MB comme il a été reporté dans les cellules cancéreuses en l'absence de protéases. Tous ces résultats ensemble, ont montré que la cellule invasive a adapté sa polymérisation de filaments d'actine ramifiée pour maintenir l'invasion dans différents contextes…

▼ The thesis is focused on an attempt to delay
photoreceptor cell death in the rd1 mouse using Lens Epithelium
Derived Growth Factor (LEDGF), Glutathione S-Transferase mu (GST-??
and Glutathione S-Transferase alpha (GST-?? in an organ culture
paradigm as well as the role of metalloproteases (MMPs) and their
tissue inhibitors in photoreceptor degeneration (TIMPs). The rd1
mouse model displays a retina degeneration which starts around day
10 with rod photoreceptor cell death. By 21 days after birth
virtually all rod photoreceptors in the retina have died, and
subsequent cone degeneration starts. The latter process is somewhat
slower and by the age of 3-4 months, the animal has practically no
photoreceptors left. In order to study the effect of factors which
delay or halt the photoreceptor degeneration, a long term retina
organ culture has been used. The culture medium of this culture
paradigm was devoid of fetal calf serum (FCS) as FCS by itself
contains an unknown number of growth factors which can
synergistically interact on the rescue mechanisms together with the
tested substance. In order to provide a chemically defined
environment for the test paradigm, the culture medium was further
developed to function without the addition of any FCS. Using this
improved culture paradigm, a moderate but clear photoreceptor
rescue was demonstrated using LEDGF. Also GSTs were tested and
found to exhibit a rescue effect on photoreceptor degeneration. The
mechanism(s) of photoreceptor cell death (apoptosis) in the rd1
mouse is not completely understood and is probably not involving
the established caspase dependent apoptotic pathways. MMPs and
TIMPs are involved in apoptotic processes. To enlighten their role
in photoreceptor cell death, a study on the normal presence of
these substances in the wild type retina and their changes in the
rd1 mouse was performed.

▼ The primary contributor to Vibrio fischeri aminopeptidase activity is aminopeptidase N, PepN. Colonization assays revealed the pepN mutant strain to be deficient at forming dense aggregates and populating the host’s light organ compared to wildtype within the first 12 hours of colonization; however the mutant competed normally at 24 hours. To address the role of PepN in colonization initiation and establish additional phenotypes for the pepN mutant strain, stress response and other physiological assays were employed. Marked differences were found between pepN mutant and wildtype strain in response to salinity, acidity, and antibiotic tolerance. This study has provided a foundation for future work on identifying a putative role for V. fischeri PepN in regulating stress response.

▼ Introduction: Since its discovery in 1926, relaxin has been regarded as a peptide hormone that plays a role in the enlargement of pubic symphysis during the perinatal period. Since then, it has been found to exhibit a variety of biological functions in various organs and systems, especially the cardiovascular system. The specific mechanisms through which it exerts its effects on the cardiovascular system have not yet been elucidated. What is known is that of the three types of relaxin found in humans (RL1, RL2, RL3), the major isoform is relaxin 2, commonly referred to as "relaxin". Relaxin family peptides target specific G protein-coupled receptors (G-coupled receptors), defined as relaxin family peptide receptors (RXFP), of which RXFP1 is the specific receptor for RL2. RL2 has been found to exert some of its biological activity through intracellular nitric oxide (NO), which is a potent vasodilator and its lack or production is a cause of atheromatosis according to modern atherogenesis theories. In other studies it has been found to affect the synthesis of metalloproteinases (MMPs) 2 and 9 in various tissues including the arteries. The role of metalloproteinases is well known in modern theories for aneurysm replacement because of their action on the destruction of collagen fibers and elastin in the aneurysm wall. Therefore, there is a theoretical background linking relaxin to vascular disease, but to date it has only been studied in experimental animals and not in humans. Aim: The purpose of the present study was to investigate the amount of relaxin released in the serum and the expression of the relaxin gene in arterial preparations of patients with atherosclerosis and arterial aneurysm, in relation to the type and severity of vascular disease, as well as in control subjects without evidence of vascular disease. Meanwhile, the potential mechanisms of action of relaxin will be elucidated by simultaneously measuring the expression of MMP-2, MMP-9 and eNOS in arterial preparations. Our aim is to demonstrate any predictive value of relaxin in the presence and severity of vascular disease, as well as the possibility of using relaxin as a biological indicator of early diagnosis of aneurysm and its potential therapeutic applications in atherosclerosis. ▼ Les phosphates de calcium sont des biomatériaux couramment utilisés dans de nombreuses spécialités médicales. L'amélioration de ces biomatériaux vise à augmenter leur ostéointégration et leur bioactivité. Le strontium possédant d'intéressantes capacités de modification de la physiologie osseuse, l'incorporation de ce dernier au sein de phosphates de calcium par substitution ionique pourrait permettre un déplacement de la balance osseuse vers la formation osseuse.Notre travail a permis de démontrer la capacité des particules de phosphates de calcium substitués en strontium à augmenter la prolifération des ostéoblastes en culture et à modifier l'expression et la synthèse des principales protéines impliquées dans la physiologie osseuse (Collagène de type I, Serpine H1, métalloprotéinases matricielles 1 et 2, inhibiteurs tissulaires des MMPs). Par ailleurs, la poudre de phosphates de calcium ne contenant pas de strontium a entrainé une sécrétion accrue de chimiokines pro-inflammatoires (MCP-1 et GRO-?) qui n'a pas été observée pour la poudre substituée. Enfin, des études in-vivo réalisées dans un modèle de défaut osseux murin a permis de démontrer une plus grande résorbabilité de la poudre contenant du strontium et sa plus grande capacité à stimuler la réparation osseuse.
Calcium phosphate are widely used in medicine. Their upgrade tend to enhance their biocompatibility and their bioactivity. Strontium has interesting capability to modify the bone physiologie. Its incorporation in calcium phosphates could lead to modify the bone balance toward osteogenesis.The present work reveal the capacities of such biomaterials to enhance the replication of osteoblasts ant to modify the expression and the synthesis of proteins implicated in the bone balance (type I collagen, serpinH1, Matrix metalloproteinases 1 and 2, tissular inhibitors of MMPs). Moreover, non substituted calcium phosphate powders enhance the expression and synthesis of inflammatory cytokines (MCP-1 and Gro-a). This fact was not observed with the non substituted powder. In-vivo studies on a mouse model permit us to demonstrate the higher resorbability and the higher bone healing capability of the substituted powder.

▼ Disintegrins and disintegrin-like peptides interact with various cell surface integrins and block cell-cell and cell-matrix interactions and induce apoptosis in various cell types. A novel disintegrin-like snake venom gene, Acocostatin, was isolated from the venom gland mRNA of Agkistrodon contortrix contortrix. The cDNA was cloned and expressed, and the role of the recombinant protein was tested on human umbilical vein cells (HUVECs). Translation of the cloned cDNA sequence revealed that acocostatin belongs to the PIII-SVMP subfamily of disintegrin proteins. It is composed of 139 amino acids with a Glu-Cys-Asp (ECD) disintegrin-like domain and conserved cysteine residues. Acocostatin showed 93% amino acid identity with other PIII-SVMPs such as VAP-1 (vascular apoptosis-inducing protein-1) and crotastatin-1. The recombinant protein had a molecular weight of 16 kDa when analyzed by SDS-PAGE. Incubation of actively proliferating HUVECs with 5 ìM GST-acocostatin for 24 h induced 19.68% of early apoptotic cells. Incubation of HUVECs with 5 ìM captothecin revealed 35% early apoptotic cells. Untreated and 5 ìM GST-treated cells showed 1.2% and 2.4% early apoptotic cells, respectively, as detected by annexin V-FITC staining and flow cytometric analysis. Typical apoptotic changes such as nuclear condensation and DNA fragmentation were also observed when GST-cocostatin treated cells were stained with Hoechst 33342 and observed under fluorescence microscopy.

▼ As metaloproteinases de matriz (MMPs) formam uma família de endopeptidades cálcio-dependentes capazes de degradar componentes da matriz extracelular, influenciando em processos como migração e interações célula-célula. Além disso, atuam em condições patológicas e em danos teciduais participando de doenças do sistema nervoso e doenças infecciosas, principalmente quando em desequilíbrio com seu inibidor tecidual específico, os TIMPs. A hanseníase é caracterizada por lesões de pele e nervo, esta última, conseqüência da interação do M.leprae (ML) com a Célula de Schwann (CS). A expressão de MMP-2 e MMP-9 encontra-se aumentada em lesões de nervo periférico de pacientes com hanseníase sugerindo uma participação dessas MMPs nesta doença. O objetivo deste trabalho é elucidar se as CS são capazes de produzir MMP-2 e MMP-9 quando infectadas com M.leprae e se o TNF, uma importante citocina proinflamatória na hanseníase, está contribuindo com essa produção. Para isso, CS humanas da linhagem ST88-14 e CS primárias foram cultivadas e estimuladas com ML e/ou TNF. Em seguida, o RNA total dessas células foi extraído para avaliar a expressão de MMPs e TIMP-1 por RT-PCR em tempo real. Para verificar o perfil de secreção das MMPs e TIMP-1 as células foram cultivadas por 24 horas nas mesmas condições descritas anteriormente e os sobrenadantes foram então analisados por ELISA. Além disso, os mesmos sobrenadantes foram submetidos à técnica de zimografia para avaliar a atividade proteolítica de MMP- 2 e MMP-9. A expressão de mRNA de MMPs foi detectada constitutivamente nessas células. No entanto, quando as culturas foram estimuladas com ML, os níveis de mRNA e de proteína de MMP-2 e MMP-9 aumentaram em relação ao controle enquanto que os níveis de TIMP-1 não foram alterados. Quando a razão entre MMP-9 e TIMP-1 foi avaliada um desequilíbrio favorecendo MMP-9 foi observado. O TNF associado ao ML também aumentou a expressão e secreção de MMP-9, porém não aumentou a secreção MMP-2. O mesmo perfil observado para MMP-9 foi visto para TIMP-1, porém com valores em menor escala sugerindo novamente um desequilíbrio entre MMP-9 e seu inibidor. Não foi observada enzima ativa em nenhum dos sobrenadantes analisados pela zimografia. Para os ensaios in vivo, Em paralelo, biopsias de nervo de pacientes com hanseníase foram divididas em 4 grupos: G1 (sem alterações histopatológicas), G2 (com fibrose), G3 (com infiltrado inflamatório e sem fibrose) e G4 (com infiltrado inflamatório e fibrose). A expressão e produção de MMPs foram avaliadas por RTPCR em tempo real e imunohistoquímica respectivamente. Nos ensaios in vivo níveis de RNAm de MMP-2 estavam aumentados em biópsias de nervo do grupo G3 em contraste com os níveis de MMP- 9 que tiveram maior aumento nas biópsias do grupo G4. TIMP-1 também sofreu aumento em biopsias do grupo G4, porém, novamente em menor escala que TIMP-1. ▼ Nosso grupo já vem há muitos anos investigando a resistência natural de animais a venenos de serpentes. Do soro do gambá (Didelphis marsupialis) foi isolada e caracterizada uma glicoproteína ácida de 43kDa (DM43), capaz de inibir as atividades de metaloproteases encontradas nos venenos por formação de complexos não-covalentes (Neves-Ferreira et al., 2000). No atual trabalho, procuramos avaliar se DM43 seria capaz de inibir outras metaloproteases, como as MMPs, que estão envolvidas em diversos processos patológicos, como a osteoartrite e câncer. Utilizamos linhagens celulares normais (3T3-fibroblastos) e tumorais (MDA-adenocarcinoma de próstata e MCF7-adenocarcinoma de mama) e líquido sinovial de pacientes com osteoartrite, como fontes de MMPs. As MMPs interagiram com DM43 acoplada a uma coluna de afinidade. Por imunorevelação foram identificadas como MMPs 2, 3 e 9. Em ensaios utilizando a fibronectina e a caseína como substratos, DM43 mostrou-se capaz de inibir a atividade proteolítica destas metaloproteases. O número de células, o ciclo e a morte celular foram analisadas utilizando como tratamento DM43 em 3 diferentes doses (10, 250 e 1000 ng/mL). Nossos resultados demonstraram uma diminuição no número de fibroblastos e de células de adenocarcinoma de mama, utilizando as três doses de tratamento. A análise do ciclo celular não demonstrou alteração, em relação ao controle. Portanto, a diminuição de celularidade ocorreu em conseqüência à indução de morte celular nestas linhagens, por via ainda não elucidada. O aumento da morte, em relação ao controle não tratado, foi de pelo menos 2x em fibroblastos e de quase 7x em adenocarcinoma de mama (utilizando a maior dose de DM43). Além disto, verificou-se a capacidade de DM43 inibir a degradação da matriz extracelular produzida por estas enzimas, levando a um intenso depósitofibronectina, e por conseguinte, um grande aumento de adesão celular. Estes resultados indicam o uso potencial de DM43 na terapia anti-tumoral, bem como, para pacientes com osteoartrite.
Natural resistance of marsupials against the toxic effects of venoms is due to the presence of soluble neutralizing proteins in their sera. We have isolated from opossum serum (Didelphis marsupialis) an acidic glycoprotein with 43 kDa (DM43), with an effective inhibition action to metalloproteinases snake venoms, by non-covalent complex formation (Neves-Ferreira et al., 2000). In this study, we have been investigating the effect of DM43 against MMPs involved in some pathologic processes, like osteoarthritis and cancer. In order to test if DM43 is a potential inhibitor of MMPs, we submitted supernatants of stromal cells (3T3), adenocarcinoma cell line (prostate and breast) and synovial fluid from osteoarthritis to an affinity column coupled with DM43. ▼ A degradação da interface adesiva em dentina pode acometer tanto a porção resinosa como a porção orgânica da camada híbrida. Evidências indicam que a degradação da porção orgânica pode ocorrer em parte devido a atividade enzimática de proteases da própria dentina. As metaloproteinases da matriz (MMPs) constituem a primeira classe de enzimas relacionada com a degradação da matriz orgânica. É possível que as MMPs, possam ser reativadas durante o procedimento adesivo em conseqüência da queda do pH do meio. Objetivo: Esta investigação teve como objetivos avaliar a influência de soluções desmineralizadoras com diferentes concentrações de ácido fosfórico (AF1%, AF10% e AF37%) na atividade proteolítica da dentina humana sadia, na degradação do colágeno exposto pela desmineralização, além de identificar imunologicamente as metaloproteinases. Material e Métodos: Vinte dentes molares permanentes foram triturados para obtenção de um pó de dentina. Alíquotas de 1g do pó foram acondicionadas em tubos plásticos para que a desmineralização do substrato pudesse ser realizada de acordo com as três diferentes concentrações de ácido fosfórico testadas. Após a neutralização do ácido com NaOH, as amostras foram centrifugadas. O sobrenadante foi utilizado para mensuração espectrofotométrica do conteúdo de Cálcio (Ca) liberado pela desmineralização. O precipitado obtido foi incubado com tampão de extração de proteínas por 24h a 4°C sob agitação. Nova centrifugação foi realizada e o sobrenadante utilizado para avaliação da atividade da MMP-2 e MMP-9 por zimografia e para a identificação imunológica das metaloproteinases por western blot. O precipitado obtido pela centrifugação foi liofilizado e separado em alíquotas de 40mg (n=10) de pó de dentina para cada condição experimental. As alíquotas foram incubadas em saliva artificial por 24h a 37°C sob agitação e hidrolisadas em autoclave. Após incubação a 65°C por 40 min, as amostras foram avaliadas por espectrofotometria para que a mensuração da concentração do aminoácido hidroxiprolina (HYP) liberado pela degradação do colágeno exposto pudesse ser obtida. Fatias de 0.3 mm da dentina coronária de outro dente foram obtidas e desmineralizadas, de acordo com as diferentes concentrações de ácido fosfórico para avaliação da atividade enzimática através do método da zimografia in situ. Os resultados foram submetidos à análise de variância de um fator sem vinculação ou teste de Kruskal-Wallis para identificação das diferenças existentes entre os grupos experimentais. Para contraste das médias foi realizado o teste de Student-Newman-Keuls com nível de significância de 5% (=0,05). Resultados: A atividade enzimática de MMP-2 foi estatisticamente maior quando a dentina foi desmineralizada com AF 1% ou 10%. O uso do ácido fosfórico 37% diminuiu consideravelmente a atividade desta enzima, em relação ao AF10% (<0,05). ▼ A lesão pulmonar é determinante da morbi-mortalidade na pancreatite aguda (PA). Neutrófilos (PMN) e mediadores inflamatórios são responsáveis pelo desenvolvimento da lesão. A solução hipertônica modula a reposta inflamatória tendo como resultante um efeito imunomodulatório capaz de prevenir a lesão tecidual. Neste trabalho, investigamos os efeitos da solução hipertônica sobre os níveis de HSPs, MMPs e citocinas no tecido pulmonar, bem como, o mecanismo envolvido em sua regulação. Ratos machos Wistar foram submetidos a pancreatite pela injeção retrógrada de 1 ml/Kg de taurocolato de sódio 2,5%. Os animais foram randomizados em 4 grupos: 1) controle: não foi submetido a qualquer procedimento; 2) pancreatite sem tratamento (ST); 3) pancreatite e tratamento com solução fisiológica (SF); 4) pancreatite e tratamento com solução hipertônica (SH). Os animais dos grupos 3 e 4 tiveram a veia jugular cateterizada e receberam infusão de solução hipertônica ou fisiológica 1 hora após a indução de pancreatite. Os animais com PA foram sacrificados após 4, 12 e 24 horas. Os pulmões foram processados e submetidos a histologia com HE e dosagem de mieloperoxidase (MPO) para quantificação do infiltrado de neutrófilos. A produção e atividade das MMPs 2 e 9 foi analisada por zimografia. Western Blotting foi utilizado na detecção das HSPs 60, 70 e 90. As alterações na expressão gênica foram analisadas por RTPCR. Os níveis de peroxidação lipidica dosados por TBARs. A concentração de citocinas foi medida por ELISA. A reposição volêmica com SHT diminui o infiltrado inflamatório (PMN) 12 horas após a indução da PA. Concomitante a redução dos PMNs vimos a queda na atividade e expressão da MMP-9 e aumento da expressão protéica de HSP70 no tecido pulmonar. Tardiamente, 24 horas, o grupo tratado com SHT mostrou aumento na produção de IL-10, uma citocina anti-inflamatória. A SHT modula a resposta inflamatória, promovendo proteção ao tecido pulmonar contra os efeitos deletérios decorrentes da PA. Seus efeitos benéficos envolvem alterações celulares e moleculares que levam à redução da lesão tecidual.
Acute Pancreatitis (AP) is an inflammatory process of the pancreas with variable involvement of other organs and systems. The lungs are the most common distant organs affected by severe acute pancreatitis. The immunomodulatory effects of hypertonic solution (HS) provide potential strategies to attenuate inappropriate inflammatory reactions. This study tested the hypothesis that administration of HS modulates the development of lung injury in pancreatitis model. HS resuscitation results in a significant attenuation of lung injury following AP by modulate MMP 2 and 9 activity and protected tissue by increased HSPs 70 and 90 expression.

▼ INTRODUÇÃO: A degeneração do disco intervertebral (DIV) é um processo crônico e apontado como o maior causador de cervicalgia e lombalgia. Esse processo geralmente conta com a degradação da matriz extracelular, expressão de citocinas inflamatórias e fatores angiogênicos e axonogênicos. Entretanto, muito pouco se sabe sobre esse processo em DIVs assintomáticos durante o envelhecimento, principalmente no segmento cervical. O objetivo desse estudo foi de delinear o perfil de moléculas relacionadas à degeneração discal em DIVs cervicais e lombares. MÉTODOS: Discos intervertebrais humanos cervicais e lombares (C4-C6 e L4-S1) foram coletados em autópsia de 30 indivíduos presumivelmente assintomáticos e divididos em grupos jovem (GJ < 35 anos, n=60) e idoso (GI > 65 anos, n=60). O nível de degeneração foi constatado pela escala de Thompson, e foi correlacionado com a detecção imuno-histoquímica das moléculas de MMP-1, -2, -3, TIMP-1, IL-1beta, TNF-alfa, VEGF, NGF-beta e BDNF. RESULTADOS: Todos os DIVs mostraram algum grau de degeneração, embora mais acentuadas no GI. As moléculas empenhadas no estudo foram identificadas em ambos grupos. A detecção imuno-histoquímica foi prevalente no citoplasma das células nativas do DIV e na região de interseção entre a placa vertebral e o arranjo fibro-colágeno. O envelhecimento propiciou, no disco cervical, maior expressão de MMP-2, -3, VEGF, NGF-beta e BDNF, enquanto que no disco lombar, a maior expressão foi de MMP-1, -2 -3, TIMP-1, TNF-alfa, VEGF e NGF-beta. DISCUSSÃO: O envelhecimento de DIVs cervicais e lombares caracterizou-se por exibir um processo catabólico e extensivo remodelamento da matriz extracelular, os quais podem ser interpretados como eventos que antecipam a doença degenerativa discal. Esse processo é capaz de levar a angiogênese e axonogênese de modo a ampliar o metabolismo aeróbio do DIV e captar informação nociceptiva como forma de defesa, uma vez que até nos discos lombares de indivíduos jovens essa última característica pôde ser observada. Discos assintomáticos também exibem moléculas relacionadas à doença degenerativa discal e talvez a inibição de parte dessas possa resultar em terapia preventiva
