▼ As artificial intelligence development has been rapidly advancing, the goal of creating artificial agents that can mimic human behavior is becoming a reality. Artificial agents are becoming capable of reflecting human behavior and decision making such as drawing creative art pieces and playing video games [10][24]. Therefore, they should be able to mimic one of the greatest human strengths, cooperation. Cooperation is an integral skill that allows humans to achieve feats that they cannot do alone. It is also a highly valuable skill that can be developed for artificial agents as software with intelligent programming becomes integrated into human society. As advanced neural network architectures become widely available, cooperative artificial agents will aid humans in a wide variety of fields. Thus, it becomes vital to discuss the quality of interactions between society and artificial systems and analyze how these systems should interact with the public.
This study attempts to emulate past experimentation with cooperative agents and how well it performs in the context of a cooperative video game, NBA
Jam for the Super Nintendo Entertainment System. NBA
Jam is a great testbed for testing cooperative agents because it is well known in the gaming community for its difficulty [25]. NBA
Jam includes a complex set of inputs and combinations that allow the player to play with a unique style. This experiment seeks to explore the results of training an intelligent artificial agent that attempts to maximize cooperation between itself and its teammate. The inclusion of well programmed cooperative agents should allow players to win more games and have an enjoyable experience. By adjusting the rewards systems of neural networks, this project attempts to explore the nuances of developing cooperative agents for video games.

▼ I denna uppsats undersöker vi Game/Media Jam, Hackathon och liknande koncept och försökerskapa en metod för att ta fram en modell för deras topologi. Vi ämnar att undersöka degemensamma punkterna Jams och Hackathons har via deras regler, samt jämföra detta med JamSessions. Vidare kommer vi försöka identifiera reglernas syften. Dessa syften sätter vi i entopologiskt mätbar intervall som sedan kan överföras på den topologisk modellen. Sedan användsdessa resultat för att jämföra regelrätta Jams/Hackathons, närliggande koncept och arrangemangsom endast har vissa gemensamma punkter med Jams, för att se om modellen kan påvisa om ettarrangemang har en känsla av Jam utan att faktiskt vara det.
In this thesis we will study Game/Media-Jams, Hackathons and similar concepts and try to establisha method to create a model for their topology. We intend to investigate what these concepts have incommon through their rules and compare that to Jam Sessions. Following this we will try to extractthe purpose of the rules. These purposes will then be put in topologically measurable intervals thatcan be transfered onto the topological model. The results will then be used to compareJams/Hackathons, similar concepts and events that only have a few things in common with Jams, tosee if the model can determine if an event is in the spirit of a Jam without actually being one.

▼ This study was part of the development of an ice jam
flood forecasting system for Hay River, NWT. 2-D numerical models
were used to simulate ice processes in an effort to predict ice jam
formation. A summer survey was conducted to finalize the bathymetry
of the Hay River Delta. Observations were undertaken during
freeze-up and the winter to better understand Hay River ice
conditions. Ice jam events were surveyed during breakup in 2008 and
2009 for model testing. The data collected was used to develop
CRISSP2D and River2D models to simulate observed conditions. Simple
tests were conducted with CRISSP2D to better understand the model
inputs. CRISSP2D modeling of Hay River was unsuccessful and it
limitations were discussed. River2D was able to match observed ice
jam profiles. The results were used to create an Ice Jam Profile
Generator to assist the Town of Hay River with evacuation
planning.

▼ Syftet med detta arbete är att jämföra effekterna av game feel och detaljerad grafik på en spelares immersion. Arbetet är riktat mot korta produktioner och har som mål att redovisa data som gör att spelutvecklare kan göra informerade val vid implementation av funktionalitet och grafik. Tre prototyper skapades för att mäta spelarnas immersion: en med abstrakt grafik, en med detaljerad grafik och en med game feel. Försökpersonernas upplevelse av prototyperna samlades in med hjälp av en webbbaserad enkät. Försökpersonernas svar visar inga säkra resultat men pekar på att försökspersonerna som spelade prototypen med game feel upplevde mest immersion.

▼ The Town of Hay River experiences significant threats
to life and property each spring as ice jam release events from
upstream bring waves of ice and water to the town. The development
of a forecasting tool for ice jam release events has been limited
by insufficient data, especially regarding the speed of ice runs
associated with ice jam release events. The purpose of this
research was to document and analyze ice jam release events to
provide the town warning of their potential timing and magnitude,
and to contribute to general knowledge on ice jam release.
Comprehensive field programs were undertaken from 2007 to 2009, and
this new data was used to assess the River1D ice jam release
forecasting model. Although the model showed reasonable
approximations for wave arrival times for flood forecasting
purposes, the predicted speeds and arrival times of ice runs did
not agree very well with field observations.

▼ Ice jams have caused serious hazards such as floods in northern countries. Despite several studies focused on river ice jams, a lack of research has been identified in the existing literature to analyze impacts of an ice
jam on turbulent flow characteristics. Thus, this research attempts to address this knowledge gap. A better comprehension of flow features beneath an ice
jam will help river ice engineers to reduce economic, environmental, and ecological damages.
Technical difficulties and safety concerns limit data collection in ice
jam field studies. Therefore, a physical model of a river ice
jam was constructed in the Hydraulics Research & Testing Facility at the University of Manitoba. Two scenarios were proposed to simulate an ice
jam. The first scenario was to use a basic model with constant upstream and downstream water depth. The second scenario was developed as an extension of the first scenario, with a modified setup to allow the upstream water depth to be greater than the downstream water depth. This was done to increase similarity with the real ice
jam in nature and improve the results. For the second scenario, different roughness ratios of channel bed to ice
jam were defined. Experiments were conducted to quantify the turbulent flow characteristics including streamwise velocity and streamwise turbulence intensity distribution beneath the ice
jam. The flow velocities were measured at different locations beneath the ice
jam using an acoustic Doppler velocimeter. The turbulence intensities and bed shear stresses were calculated using the measured velocities.
The results indicated that the flow velocity and turbulence intensity were a function of the streamwise location beneath the ice
jam as well as the roughness of the bed and ice
jam. Significant changes were observed in the velocity and turbulence intensity profiles where the boundaries were fully rough, which could influence an ice
jam formation, sediment concentration and water quality, as well as river bed deformation. It is believed that the results from this research will improve our fundamental understanding of flow beneath an ice
jam, and can be used for validation of subsequent ice
jam numerical models.

Slutsatsen är att metoden gick att använda för att kategorisera idéflödet men att ett material som sträcker sig över många ämnen tenderar att sänka tillförlitligheten på resultatet. ▼ L’adhésion, la migration cellulaire et l’environnement stromal sont intimement liés pour garantir l’homéostasie du système immuno-hématopoïétique. Néanmoins, nos connaissances des mécanismes responsables du maintien de ce processus fonctionnel restent fragmentaires. Notre étude a permis de mieux caractériser le stroma ganglionnaire et médullaire dans lesquels nous avons démontré de nouveaux rôles immuno-régulateurs des molécules d’adhésion jonctionnelle JAM-B et JAM-C. Dans la zone T des ganglions lymphatiques, les cellules réticulaires fibroblastiques (FRC) sécrètent des composés de la matrice extracellulaire et des chimiokines homéostatiques, nécessaires à la migration intranodale des lymphocytes T naïfs. La génération de nouveaux anticorps monoclonaux a permis d’identifier une diversité phénotypique et fonctionnelle au sein de la population FRC. L’un d’entre eux reconnaît la Thrombomoduline permettant d’identifier une population de FRC exprimant les protéines JAM-C et PDGFRα. Cette population cellulaire, dénommée FRCDP (Double Positive) sécrète des chimiokines homéostatiques, ce qui la distingue de la population FRCDN (Double Negative). Les souris sauvages traitées avec l’anticorps anti-JAM-C présentent une diminution significative du taux intranodal des chimiokines CXCL12, CCL19, CCL21 affectant la recirculation des cellules T naïves. De façon similaire, les cellules stromales des niches hématopoïétiques fournissent un environnement fonctionnel, nécessaire à l’homéostasie du système hématopoïétique. Les molécules d’adhésion sont connues pour contrôler ces mécanismes. JAM-C est exprimée à la surface des cellules souches hématopoïétiques (CSH) mais son rôle dans l’hématopoïèse reste inconnu. Notre étude montre que la molécule JAM-B est exprimée par l’environnement médullaire et interagit spécifiquement avec JAM-C sur les CSH. Les souris déficientes pour le gène jam-b présentent une diminution du nombre de CSH quiescentes et une réponse accrue aux agents mobilisants, démontrant ainsi que le couple JAM-B/JAM-C est nécessaire au maintien et à la rétention des CSH dans la moelle osseuse.
Homeostasis of the immune and hematopoietic system is dependent of cell adhesion, cell migration and stromal environment. Nevertheless, the molecular mechanisms involved in the crosstalk between hematopoietic and stromal cells have remained elusive. Our studies allowed to better characterize lymph node (LN) and bone marrow (BM) stromal compartments through the demonstration that expression of junctional adhesion molecules (JAM) in these compartments is necessary for the maintenance of immune and hematopoietic homeostasis. In the T cell zone (LN), extracellular matrix and homeostatic chemokines are secreted by fibroblastic reticular cells (FRC) which control naive T cell migration. We have identified new FRC subsets using a monoclonal antibody based approach to identify new cell surface markers of stromal cells. ▼ Different chemical components and their locations in strawberry (Fragaria x ananassa) achenes, vascular bundles and cortical cell walls were studied by means of bright-field microscopy using different staining systems and by Fourier transform infrared (FT-IR) microscopy. The structural changes in pretreated strawberry tissues during freezing and thawing were studied by means of physical and chemical analyses as well as by bright-field and FT-IR microscopy, and by confocal laser scanning (CLS) microscopy. In addition, traditional jams made from pretreated frozen strawberries were investigated by means of physical and chemical analyses and by sensory evaluation. Basic microstructural studies showed that all the cell walls were complex and consisted of several components. Lignin was an important component of achenes and vascular tissues, whereas the cortical cell walls contained mainly cellulose, pectin and some protein. Cellulose and pectin were also detectable in the intercellular spaces of cortex, in the vascular tissue and pith. Protein was deposited inside the cortical cells, as well as in the vascular tissue and pith. The microscopical methods used gave comparable results. According to microscopical studies both the pretreatments with calcium chloride (CaCl2) and crystallised sucrose as well with CaCl2 and pectin methyesterase (PME) in a vacuum affected the microstructure of strawberry tissues. These pretreatments especially affected pectin, protein, lignin and structural carbohydrates in the vascular tissue and cortex compared to the untreated reference samples. The use of a vacuum appeared to make the pretreatment solutions absorb more efficiently to the cortex and pith, thus improving the stabilisation particularly of pectin and structural carbohydrates. Firmness of thawed and particularly of jam strawberries pretreated with CaCl2 and PME in a vacuum was higher than that of other pretreated or untreated berries. In all the prefreezing treatments studied, dipping of strawberries into a CaCl2 solution with PME in a vacuum resulted in a significantly different sensory profile than was found in the other jams. The sensory attributes wholeness of the berries (p<0.001), firmness, clarity and evenness of the jam medium (p<0.001), softness of the berries (p<0.001) and faultlessness of odour and flavour (p<0.001) in particular were statistically significantly different among the strawberry jams. Sensory quality was perceived to decrease during 4 months of storage, even though the shapes of the sensory profiles of the studied jams did not change significantly from those evaluated after 2 weeks storage. For achieving high quality jams the pretreatment time should be short (about 5-15 min), the temperature low (below 20 °C), the vacuum level high (pressure less than 10 kPa), the CaCl2 concentration moderate (about 1%) and the dosage of PME comparatively low (about 50-100 μkat/kg strawberries).

The purpose of this intrinsic case study was to understand the function and significance of Java Jam – an annual music performance event at a public high school in which students select and rehearse music to perform. Five questions guided the study: How do different groups of Pikeford High School community members view Java Jam? How does Java Jam influence music-making? What are the successes and failures of Java Jam? What are the connections between Java Jam participants and school music? And, how does Java Jam affect the Pikeford High School community? In this study, I provide a detailed description of Java Jam through the perspective of 24 participants representing varied roles among PHS community members: students, teachers, parents, and alumni. The study was bound by both time and place, and data were collected at Pikeford High School during the 2013-2014 school year. Data used in this study consist of personal interviews, a focus group session, observations, and archival data submitted by participants. The research results are presented as both a personal narrative and case description from participant perspectives. This study provides an example of extracurricular alternative music making in which students engage in self-directed learning and peer learning. This study impacts the field of music education in that it demonstrates positive outcomes from an event such as Java Jam, including increased student autonomy, space for musical creativity, and increased confidence among student participants. I present additional implications for the field of music education, music teacher education, and future research to close the study.

▼ River ice breakup and associated flooding are
realities for many northern communities. This is certainly the case
in Hay River, NWT, which is located at the junction of the Hay
River and Great Slave Lake. Hay River experiences a wide range of
spring river ice scenarios; from docile thermal melt outs, to
severe ice jams resulting in life-threatening, disastrous flooding.
This study involved the analysis of five seasons of aerial and
time-lapse photographs, water level measurements and
hydrometeorologic data. This work also compiled an extended
historical record of breakup in the Hay River delta, which was
compared against the field data gathered for this study; combining
local, experiential knowledge with scientific observation into a
cohesive description of breakup. This will be used to advise the
non-technical flood watch community on the patterns of timing and
sequencing of breakup, which is critical for evacuation
planning.

▼ The vulnerability of GPS to interference has been a major concern for both military and civilian applications, including small UAVs. Various signal processing techniques have been developed to improve the reliability of GPS receivers against different types of interference. Among these techniques, null steering is recognized as an effective method to protect GPS against both narrowband and broadband interference. However, due to the requirement of multiple antenna channels, it has mainly been implemented for large platform applications.
This thesis examines the suitability of null steering techniques for small UAV applications and determines the practically achievable anti-jamming ability by implementing a two-element miniaturized adaptive antenna array.
The adaptive antenna array is tested against a 2MHz broadband jamming signal under both laboratory testing conditions and a real jamming environment. Approximately 40dB anti-jamming range was achieved in the laboratory testing conditions. 38dB and 42dB were obtained in a real jamming environment with different antenna configuration. The likely performance limitations and possible further performance enhancements are also outlined in this thesis.

▼ This essay describes the work to document and evaluate tools for front-end development at the company B3 Consulting Group. The methods that are evaluated is AMP-stack and JAM-stack. AMP-stack uses Apache, MySQL and PHP, Wordpress is used to build with that method. JAM-stack uses JavaScript, API and Markup, for that method GatsbyJS and Contentful is used. The evaluation is done both through user tests and how the methods are percived from a developer perspective. The project also includes work with customers at the company the project is done, Wordpress is used for those web pages.
Denna rapport beskriver arbetet med att dokumentera och utvärdera verktyg för frontend-utveckling vid företaget B3 Consulting Group. Metoderna som utvärderas är AMP-stack och JAM-stack. AMP-stack bygger på Apache, MySQL och PHP, för att bygga med den metoden har Wordpress använts. JAMstack bygger på JavaScript, API samt Markup, för den metoden har GatsbyJS samt Contentful använts. Utvärdering sker både genom användartester och hur metoderna upplevs ur ett utvecklarperspektiv. I arbetet ingår även arbete med kunder till företaget projektet utförts hos, för deras webbplatser har Wordpress använts.

▼ Collective cell migration is a fundamental process throughout development. The migrating border cell cluster (BCC) in the Drosophila ovary is a good in-vivo model system to study this process. My work focuses on the function of the large Maf transcription factor, Traffic Jam (Tj), in regulating BCC migration. Tj has a dual function in both promoting and inhibiting BCC migration, as reduction or increase of tj expression leads to migration delays. I found a genetic interaction between Tj and three factors known to promote BCC migration: an inhibitor of the JAK/Stat pathway, Socs36E, the transcription factor Slow border cells (Slbo), and the adhesion molecule DEcadherin (DEcad). At high expression level, Tj enhances the expression of Socs36E, which subsequently inhibits JAK/Stat and causes reduction of Slbo. High Tj expression also inhibits DEcad upregulation in the BCC. Activation of Slbo, which is induced by JAK/Stat, leads to a natural downregulation of Tj. This low expression level of Tj maintains Slbo expression in the BCC, which allows for DEcad upregulation. My work has shown that Tj is an important component of this regulatory network, which allows for proper BCC migration.
Analysis of BCC morphology reveals that Tj also functions in regulating the formation of actin-based cellular protrusions. These protrusions are normally extended by border cells and are necessary for its movement. Altering Tj expression leads to abnormal organization of the actin cytoskeleton and morphology of the protrusions. This effect does not appear to be mediated through Slbo or DEcad, which are known to regulate actin. A genome-wide microarray screen identified serine/threonine kinase Protein Kinase C δ (PKCδ) as a putative target of Tj. Changing PKCδ expression in the BCC also causes abnormal organization of actin filaments. Genetic interaction studies suggest that Tj regulates actin organization through upregulating PKCδ. Furthermore, altering Tj or PKCδ expression changes the distribution pattern of an actin filament elongation factor, Enabled, in the BCC. My results have identified Tj and its putative downstream effector, PKCδ, as crucial regulators of the actin cytoskeleton and cellular protrusions in the BCC.

▼ Many rivers in cold climate regions can experience ice jams, which can be significant contributors to heavy flooding, especially when coinciding with higher spring runoff flows. Due to the danger and unpredictability inherent with ice jams, laboratory studies are often used to analyse flow conditions underneath ice jams. An experiment was conducted in the small-scale Particle Image Velocimetry (PIV) flume at the University of Manitoba, using polyethylene pellets to simulate the accumulation of ice pieces in a breakup ice
jam. The goals of this experiment were to examine the flow conditions under a simulated ice
jam using PIV, and develop a methodology for calculating void seepage, the portion of total river flow within the
jam. Contours and profiles were presented for average velocities and turbulence statistics for three test conditions at a constant Froude number and varied upstream water depths of 5, 10, and 15 cm. Velocity profiles were integrated to calculate void seepage along the
jam, with the percentage of seepage then related to the relative thickness of the ice
jam. Seepage values were found to be often near 5% of total flume discharge, reaching as high as 14% at the thickest portions of the ice
jam. A cubic polynomial was fitted to the dataset relating the recorded seepage ratios to the relative
jam thickness. The recorded seepage flow and estimated hydraulic gradient was also used to calculate a minimum seepage velocity parameter for the model ice
jam, found to be λ = 0.57 m/s.

▼ Bluegrass and old-time are genres founded on Appalachian music traditions, as mediated through the early recording industry. While initially considered performance genres, the styles have become the foundation for
jam sessions across the United States. These
jam sessions are participatory, inclusive spaces, in which anyone with a basic knowledge of the style and the proper instrumentation can, in theory, join in the musicking (as defined by Christopher Small, 1998). For the musician with a disability, these informal
jam sessions, founded on musical sociability, demonstrate an alternative value structure, as they are mostly unregulated by the Americans with Disabilities Act of 1990 (ADA), an economic-based antidiscrimination law. As this leaves accommodation up to the discretion of the participants, the accommodations found within these settings portray a different perspective of disability—one which allows for possibility and participation. I term this as a participatory model of accommodation and, through ethnography and autoethnography, illustrate how this peer-based model promotes inclusion within the social model of disability. Drawing on Thomas Turino’s (2008) definitions of participatory music and Tom Shakespeare’s (2006, 2010) articulation of the social model of disability, I analyze the musical and social dynamics of the
jam session, drawing on my own experience as a fiddle player with a hearing impairment. Further, using ethnographic research—specifically, interviews and participant-observation—of musicians with disabilities in these sessions in Tennessee, Washington, and Illinois, I illustrate the importance of interpersonal relationships, rather than power dynamics, in the
jam session. Moreover, I demonstrate how the aging population of these
jam sessions impacts ideas of disability, impairment, and mortality. Finally, I argue that this narrative of inclusion and practices of accommodation in
jam sessions can change individual perspectives on disability.

Resumo: O termo "processamento de alimentos" vem sofrendo diversas críticas na mídia oficial entre profissionais de diversas áreas do conhecimento. De forma geral, alguns profissionais vêm considerando o processamento feito pela indústria desvantajoso sob o ponto de vista nutricional, ambiental e social, recomendando, desta forma, o preparo caseiro de alimentos. Neste projeto foi proposto o estudo do processamento da geleia de morango obtida de forma industrial, comparando àquelas elaboradas por receitas e utensílios caseiros. Foram avaliados, em ambos os processos, o consumo de energia e de água, geração de resíduos da
fruta e, sob o ponto de vista sensorial e nutricional, a cor, a retenção de antocianinas e de vitamina C, discutindo as vantagens e desvantagens de ambos os processos. Foram avaliadas geleias caseiras preparadas em tempos e proporções diferentes de fruta e açúcar e utilizando diferentes potências do queimador do fogão. Foram feitas também geleias em equipamentos em escala piloto, assim como adquiridas geleias industriais, todas avaliadas durante o armazenamento. A perda na limpeza do morango foi de 7 a 13% no processo caseiro, e de 15% segundo informações da indústria, contudo, deve-se considerar que, muitas vezes, os morangos que foram para a indústria partem de um reaproveitamento de morangos que não vão para o consumidor. O consumo de água na lavagem caseira dos frutos (litros/kg de morango) variou de 9,5 a 20 quando realizado por água corrente, e de 1,54 a 1,78 por simples imersão, maiores que o obtido pelo processo industrial, de 1,5. A energia consumida (kcal/kg água evaporada)
no processo de concentração variou de 1278 a 2999 na geleia caseira e de 638 a 770 nas geleias produzidas em escala piloto e industrial. A maior retenção de antocianinas (97,65%) e ácido ascórbico (83%) ocorreu na geleia com maior quantidade de açúcar (50%) e menor tempo de exposição ao calor (20 minutos), enquanto a menor retenção de antocianinas ocorreu na geleia caseira com maiores quantidade de açúcar (50%), tempo de exposição ao calor (60 minutos) e consumo de energia (688 Kcal/batelada). As geleias feitas em planta piloto retiveram 58±5,83% de antocianinas, sendo semelhante estatisticamente (p>5%) a geleia caseira de maior tempo de exposição ao calor. As geleias industriais seguiram um modelo de degradação de primeira ordem para antocianinas e ácido ascórbico, com D25 °C = 150 dias e 140 dias, respectivamente. Houve degradação da cor vermelha durante o processo e armazenamento. A qualidade da geleia, do ponto de preservação de cor e antocianinas pode ser obtida tanto de

Resumo: A mobilidade urbana é um dos grandes desafios nos centros das principais cidades pelo mundo a serem compreendidos de como se pode melhorá-lo, pois naturalmente afeta a qualidade de vida das pessoas que vivem e trabalham nestas regiões. Diante do paradoxo de como os veículos po-dem ser produzidos com alta eficiência e eficácia, mas o propósito deles não atende o desloca-mento diário dos centros das grandes cidades em tempos de trajetos razoáveis, utiliza-se as ferra-mentas consagradas na fabricação de veículos por meio da melhoria contínua, manufatura enxuta, fluxo contínuo e teoria das restrições para
balancear as opções de modalidades de transporte. Os cenários são feitos a paritr de dezessete vias, sendo dezesseis arteriais e uma expressa, delimitadas no centro expandido da cidade de São Paulo por intermédio dos dados oficiais de fluxo de veícu-los. Os cenários são analisados na promoção do desenvolvimento sustentável e na contribuição da melhoria da qualidade de vida dos cidadãos. Na tese é trabalhada a hipótese de que se a unidade conceitual de medição dos veículos, dada por volume ou fluxo de veículos, sendo a unidade con-sagrada e utilizada, que é veículos/h, for alterada para pessoa por m²/h, as análises e os resultados são mais efetivos com as propostas de monitoramento. A pesquisa realizada aponta duas conclu-sões principais: que a alteração da métrica por meio da tecnologia no modelamento de uma estru-tura conceitual de um ambiente ciber-físico, se torna balizador nas tomadas de decisão na gestão da engenharia de tráfego na cidade de São Paulo e, que os números de
pessoas que se deslocam nas vias tradicionais do Plano de Avenidas, equivalem a retirar 29,5% dos automóveis por hora nos horários de pico e reduzir a emissão de gases em 64,0% proveniente dos veículos automoto-res, transportando o mesmo número de pessoas nas vias analisadas, sem alterar a modalidade de transporte. Há ganhos na perspectiva social devido ao estímulo de compartilhamento de veículos, sejam privados ou coletivos; na perspectiva ambiental há reduções nas emissões de poluentes de-vido ao estímulo de ônibus fretado e veículos ultracompactos elétricos e compartilhados; e por último, há ganhos econômicos devido à redução dos tempos de viagem e por parte dos modais terem a cobrança variável e compartilhada nos deslocamentos das vias principais da cidade

O objetivo deste trabalho foi desenvolver uma geleia light de maná cubiu adoçada com sucralose, avaliar sua composição e aceitabilidade. No fruto in natura e na polpa branqueada de maná cubiu foram realizadas as seguintes análises físico-químicas (Sólidos Solúveis, pH, Acidez Titulável, açúcar redutor), quantificação da cor instrumental e análises bioquímicas (Compostos Fenólicos Totais e Atividade Antioxidante pelo método DPPH). As geleias foram preparadas com uma concentração de 60:40 (polpa:açúcar cristal) para a convencional e 60:40 (polpa:açúcar cristal + sucralose) para as geleias light. Para que ocorresse a geleificação das mesmas, foi utilizada pectina cítrica. A geleia foi envasada à quente
em embalagens de vidro com capacidade para 320g, previamente esterilizadas fechadas e invertidas. Após o envase as geleias foram resfriadas naturalmente e estocados à temperatura ambiente. Nas geleias foram feitas as seguintes análises: físico-químicas (Sólidos Solúveis, pH, Acidez Titulável, açúcar redutor e açúcar redutor total), quantificação da cor instrumental, análises bioquímicas (Compostos Fenólicos Totais e Atividade Antioxidante pelo método DPPH), análise microbiológica e análise sensorial, durante o período de armazenamento de 120 dias. Os teores de Sólidos Solúveis aumentaram durante o tempo de armazenamento. Os tratamentos que apresentaram adição de edulcorante (formulações light), ocasionou pequena elevação do valor do pH quando comparados com a geleia convencional. Durante o período de armazenamento houve pequena elevação nos teores de Acidez Titulável, principalmente nas formulações light. The objective of this work was to develop a light jelly manna cubiu sweetened with sucralose, evaluate its composition and acceptability. In the fresh fruit and bleached pulp manna cubiu were held the following physico-chemical analysis (soluble solids, pH, Acidity, reducing sugar), quantification of instrumental color and chemistries (Phenolic Compounds and Antioxidant Activity Totals by DPPH method). The jellies were prepared with a concentration of 60:40 (pulp crystallized sugar) for conventional and 60:40 (pulp: Crystal + sucralose sugar) for light jellies. dabartinis vaiko priežiūros atostogų teisinis reguliavimas, dėmesį
sutelkiant ties šių atostogų samprata ir subjektais, turinčiais
teisę į vaiko priežiūros atostogas. Siekiant geriau suprasti
nagrinėjamus teisės aktus, susijusius su vaiko priežiūros
atostogomis, ir teismų praktiką taikant juos, atlikta teisės aktų
istorinės raidos apžvalga. Praktinio taikymo probleminių aspektų
tyrimo metu nagrinėjamas darbo sutarties nutraukimas, darbuotojui
esant vaiko priežiūros atostogose, probleminius aspektus rūšiuojant
pagal darbo sutarties pasibaigimo pagrindą. Be to, įvardintos
darbuotojo teisės ir socialinės garantijos vaiko priežiūros
atostogų metu bei piktnaudžiavimo teise, naudojantis teisės aktų

The thesis provides an analysis of the legal
regulation of child care leave and highlights fundamental problems
as regards practical application as well as suggests possible means
by which a relevant solution could be developed. This paper reviews
the existing child care legal regulation including the general
clause and subjects who are eligible to take child care leave. In
order to further understand the legislation related to child care
leave the historical overview of legal development is covered. The
thesis examines the employment contract termination while employee
is in child care leave. The problem issues are being sorted on the
basis of the employment contract termination. Moreover, rights,
social guarantees of the employee in child care leave and problems,
which arise due to violation of law by using legal loopholes, are
considered. The final chapter of the thesis discusses the issues as
regards employees’ work after they return from child care leave
(taking into account that employee’s child is under three
years).

▼ The amount and type of antioxidants present in raspberries is dependent upon cultivar, ripeness, and growing conditions. Previous research on raspberry jam has reported some color, antioxidant, and flavor loss after processing and storage, though it is unknown to what extent similar changes will occur in raspberries grown in Utah. Sugar concentration and heating temperature as well as storage time were evaluated in an effort to maximize color retention, flavor, antioxidant content, shelf life, and consumer acceptance of Utah-grown raspberry jam. Four types of jams were processed in two batches each: low-sugar (40-42 Brix) and typical sugar (65-68 Brix) at 85 and 95 C, from two separate farms in Utah. Oxygen Radical Absorbance Capacity (ORAC), total anthocyanin content (TAC), color, headspace, Brix, pH, consistency, and water activity were measured in fresh jam, and after one and three months of typical storage (dark, room temperature) to evaluate changes after storage. A sensory analysis compared three-month stored jam to fresh jam made from the same berry crop. ORAC significantly declined in all jams during storage. Fresh low-sugar jam was found to contain higher ORAC values than high-sugar jam after processing and after three months of storage. All jams retained their initial anthocyanins over the first month and significantly lost an average of 28.8% anthocyanins between months 1 and 3 of storage. Color loss was found to be less pronounced than anthocyanin degradation, though nearly all jams maintained initial L*, C*, and h* values over the first month then significantly decreased by the third month of storage. When comparing fresh and three-month jam, significant sensory differences were found in color, overall acceptability, flavor, and texture. All parameters scored higher for freshly-made jam, though three-month stored jam was still found to be acceptable to consumers. In summary, after three months of storage, significant nutrient quantity and sensory quality remains in Utah raspberry jam, despite significant declines in several assays and significant differences between treatments.

▼ New tendencies in the development of innovative food packages could affect product microbial stability during storage. In this sense, antifungal systems such as emulsions, nanoemulsions and mesoporous silica supports, may be promising alternatives to preserve the product quality without having to apply further thermal treatments.
The formulation of stable antifungal systems and the optimisation of the methodology to be employed to prepare these systems play an important role in: i) their antifungal effectiveness; ii) bioactive compounds losses during their preparation; and iii) their impact on the food product's sensory profile.
Nanoemulsions prepared with essential oils generally presented higher antifungal effect than free essential oils applied at the same concentrations. Furthermore, the emulsifier played a key role in the resulting antifungal activity. Whey protein isolated-based nanoemulsions were more effective in inhibiting mycelial mould growth and spore germination than Tween 80-based ones.
The great in vitro effectiveness of nanoemulsions to control mould development allowed to check their application to food products, and strawberry
jam was selected as the food matrix. To this end, clove and cinnamon leaf emulsions were incorporated into strawberry jams to control fungal decay. Although these emulsions were able to reduce
jam spoilage, their incorporation negatively affected the aroma, taste and the overall acceptance of the
jam.
Regarding the methodology employed to prepare oil-in-water emulsions, the magnetic stirring and high pressure homogenisation combination was the most effective to reduce essential oil losses. Moreover, stable cinnamon bark-xanthan gum emulsions with good in vitro antifungal properties against the target fungi were obtained. Despite the promising use of cinnamon bark emulsions to control fungi decay in strawberry
jam, their incorporation negatively affected the product's aroma, taste and overall acceptance.
Given the strong impact of essential oils on the food product's sensory profile, two new approaches were investigated: i) combinations of different antifungal compounds in emulsions; and ii) immobilisation of bioactive compounds on mesoporous silica surfaces. The cinnamon bark essential oil, zinc gluconate and trans-ferulic acid combination allowed the obtention of a product considered microbiologically acceptable and with good organoleptic characteristics. However, total mould inhibition was not achieved. Consequently, the antifungal and sensory properties of the bioactive agents (eugenol and thymol) immobilised on mesoporous silica surfaces were studied. The preparation of jams with eugenol immobilised on MCM-41 microparticles induced better control of the fungal inhibition compared with the samples functionalised with thymol. The sensory analysis of the jams after incorporating the solids revealed that eugenol and thymol immobilisation cushioned the impact of these compounds on the

▼ Home canning has been used as a method of food preservation for hundreds of years. The United States has a regulatory agency known as the National Center for Home Food Preservation (NCHFP), which gives guidelines on canning safely to decrease the risk of foodborne illness. The NCHFP provides tips for fruit jams and jellies; however, savory
jam is rising in popularity. Savory
jam includes jams with a savory main ingredient, such as tomatoes, bacon, onion, or chili peppers. There are currently no guidelines for safe preparation of savory jams. Research is needed to see if recipes for savory
jam are safe, particularly in the YouTube space, as the quality of food safety messaging on this platform has not been investigated. YouTube allows creators with a variety of expertise levels in food and safety to upload recipes for home canners. Analysis of safety messages in these cooking videos will help determine the level of risk associated with this emerging platform. Additionally, a survey was conducted to assess whether home canners are following the same "safe" and "unsafe" practices observed in YouTube videos. Finally, home canners followed a YouTube video to prepare
jam. The
jam then underwent physical, chemical, and microbial analysis to assess its suitability for safe water bath canning. ii In the evaluation of YouTube savory
jam recipe videos we found that the expertise level of the content creator significantly impacted the quality and prevalence of food safety messages. Amateur creators received lower scores than those of expert level creators. Recipes for canned jams (as opposed to those stored under refrigeration) demonstrated a higher percentage of correct guidance, however, these videos still failed to include all the necessary information for safe canning. This analysis demonstrated that there is continued research and education needed for both producers and consumers of such content to keep home canners safe. When savory
jam was made according to the instructions of amateur level videos found on YouTube, pH was the only factor analyzed for which all samples met the current recommendation for fruit-based jams of 4.6 and below. Both Brix and water activity had values below recommended levels which created a product that is potentially unsuitable for water bath canning. Using this method of processing for such a recipe could contribute to increased risk of illness and spoilage. When home canner habits were surveyed it revealed that older participants in the survey were more likely to follow safe standard procedures for canning. However, this is concerning for newer home canners that are of younger age who are less likely to follow these procedures to keep them safe. As newer forms of recipe sourcing rise, and recipes we’ve never seen before become trending, this interaction of food and safety needs to be further explored to ensure safety at home. ▼ Sixty years ago, Francis Crick first stated the central dogma of molecular biology: DNA makes RNA, RNA makes protein. At that time the central dogma could only be imagined, but over the past two decades revolutionary advances in fluorescence microscopy have now made it possible to directly image transcription and translation in living cells and organisms. A key breakthrough was the discovery and development of GFP, which can be genetically fused to other proteins to selectively light them up and track their expression in vivo. While this powerful technology can illuminate mature protein products in live cells, processes like translation remain in the dark. This is because fluorescent fusion tags take too long to mature and light up. By the time the fluorescence becomes visible, translation of a nascent protein is over and has long since separated from its parental RNA strand. This fundamental challenge has made it difficult to visualize, quantify, and study translational gene regulatory mechanisms in living cells and organisms with fluorescence microscopy. To achieve this, nascent chain tracking (NCT) was developed, a technique that uses multi-epitope tags and antibody-based fluorescent probes to visualize and quantify protein synthesis dynamics at the single-RNA level. NCT revealed an elongation rate of ~10 amino acids per second, with initiation occurring stochastically every ~30 seconds. Polysomes contain ~1 ribosome every 200 to 900 nucleotides. By employing a multi-color probe strategy, NCT shows that a small fraction (~5%) form multi-RNA sites in which two distinct RNAs are translated simultaneously while spatially overlapping. NCT was then applied to further understand the dynamics of translating RNAs interactions with ribonucleoprotein (RNP) granules during cellular stress. NCT was used to image real-time single RNAs, their translational output, and RNA-granule interactions during stress. Although translating mRNAs only interact with RNP granules dynamically, non-translating mRNAs can form stable, and sometimes rigid, associations with RNP granules. These stable associations increase with both mRNA length and granule size. Live and fixed-cell imaging demonstrated that mRNAs can extend beyond the protein surface of a stress granule, which may facilitate interactions between RNP granules. Thus, the recruitment of mRNPs to RNP granules involves dynamic, stable and extended interactions affected by translation status, mRNA length and granule size that collectively regulate RNP granule dynamics. Finally, NCT was utilized to quantify the dynamics of multiple open reading frames at the individual RNA level in a living system. An NCT multi-color imaging modality was used to investigate ribosomal frameshifting during translation. Frameshifts occur through a ribosomal +1(-2) or -1 (+2) nucleotide(s) "slip" into another frame and are implicated in both human disease and viral infections. ▼ Consumer acceptability, or “liking,” is a complex
consumer reaction to food which impacts food-derived enjoyment,
purchasing behavior, and satiety, and is thus of interest to the
food industry as well as consumers and researchers. While numerous
factors impact acceptability, food flavor plays an outsize role in
determining if a food is liked or disliked. Traditional methods of
analyzing flavor identify individual compounds in isolation and out
of context, primarily due to instrumental limitations. These
methods have limited capability to understand acceptability, itself
more a consumer reaction to the holistic experience provided by
food than to individual sensory attributes or impact compounds.
Chemical profiling and multivariate modeling of metabolites to find
compounds predictive of effects, termed “metabolomics,” has
recently been applied to the study of flavor. The resulting
technique, termed flavoromics, offers the unprecedented ability to
directly tie the underlying chemistry of a food to acceptability
and other complex sensory percepts through statistical modeling.
Compounds predicted to be important to acceptability can be
recombined with the original food system and tasted to determine if
they drive liking or are merely correlated, providing crucial
confirmation for further application and use of findings. While
showing great potential for compound discovery, flavoromics remains
an emerging technique. Within this dissertation, flavoromics was
used across two studies to understand chemical drivers of liking in
fruit spreads. Sugar-free fruit spreads have been observed to
universally present atypical flavor defects not seen in traditional
products and are less-liked than full-sugar spreads. To understand
chemical drivers of positively perceived flavors in traditional
spreads, flavoromics was used to find universal differences between
sugar-free and traditional jams across eight fruit varieties
through class-based modeling to remove the impact of fruit variety.
Four non-volatile compounds predictive of traditional spreads as
compared to sugar-free spreads, regardless of fruit, were
identified and tasted. Two compounds, including one which had not
previously been described for flavor activity, were revealed to
have sensory impact which made sugar-free spreads taste
more-similar to traditional spreads, although further sensory
validation is needed. This study showed the ability of flavoromics
to relate chemistry to flavor-relevant class-based differences
regardless of orthogonal variations such as fruit type, and to
uncover flavor-active compounds linked to an effect. This study
will provide molecular targets to the
jam industry for use in
producing better-liked sugar-free products, helping dieting
consumers achieve sugar-reduction goals without sacrificing flavor
enjoyment. The second study focused on linking chemistry directly
to consumer ratings of acceptability in strawberry preserves.

▼ This thesis contributes to debates concerning the role of the Muslim Brotherhood and the study of political systems in transition, with a focus on Egypt following the 25 January 2011 revolution. Orthodox studies on political systems within Political Science, including scholarship on large-scale transitions and (liberal) democratisation, often focus on political institution-building, regime elites, civil/political rights, and purely structuralist approaches as explaining political dynamics and change. This study argues that discourse, power relations, and forms of resistance indicate and shape political processes, including change and transition. The originality of this work thus derives from its focus on discourse and power dynamics, analysed within the narrative accounts of informal political actors collected during Egypt's transition. This thesis also argues that the pillars of (liberal) democratisation have made possible the analytical location of the Brotherhood within these debates. For example, political systems have been measured on a (sometimes linear) scale from authoritarian to liberal democracy and this has made possible the framing of Islamic political movements on a scale from 'radical' to 'moderate', or constructed binaries in which movements like the Brothers are categorised. Through this work's analytical and empirical approach, these categories are shown to be simplistic and fragile. Two central themes are developed in relation to discourse, power relations, and the Brotherhood: democratisation as discourse and political violence. Through this study's framework political change and transformation are studied beyond the level of the regime and political elite, as is often the focus within political systems/transitions literature. Conceptions of Egypt's political transition and of the Brothers are grounded in empirical research and the experiences of political actors on the ground.
