disorders are attributed to stress on the disparity
vergence
system. This system is composed of an open-loop burst signal that
initiates changes in the ocular
vergence angle, and a closed-loop
sustained response that completes and maintains this angle.
Disparity
vergence also contains an adaptable component known as
phoria adaptation (PA). The ability of the sustained response to
generate PA has been extensively studied, however little is known
regarding the burst signal's contribution to
PA. The phoria is the ocular angle in the absence
of a disparity
vergence response. However the time course for
depleting PA makes measuring the true phoria challenging and is
typically done by extended periods of monocular occlusion. As these
extended periods of occlusion are not practical in a clinical
setting, two
forced vergence cover test (FVCT) procedures have
previously been proposed for rapidly depleting PA and revealing the
true phoria. In a series of experiments, this study evaluated these
FVCT procedures and the burst
vergence contribution to
PA. The FVCTs were performed before and after
significant PA was generated by fifteen minutes of binocularly
viewing through base-out prisms. Pre- and post-adaptation FVCT
results were significantly different (p<0.01) indicating that
the procedures did not eliminate existing phoria
adaptation. In a second experiment, PA generated
in response to three over-convergence adapting scenarios was
measured. These scenarios consisted of 30-seconds of steady fusion
(steady), 3-seconds of fusion (quick-step), 27-seconds of a flashed
target presentation (flash) that repeatedly presented the stimuli
binocularly when subjects were under-converged, and monocularly
when
subject's
vergence angle was near that required of the
stimuli; the flash period concluded with 3-seconds of fusion. The
flash period was designed to generate repeated burst
vergence
responses with minimal influence from the closed-loop response.
Greater PA was generated by the flash period than the quick-step
(p=0.025). Equivalent PA was found following the flash and the
steady periods. In conclusion, currently
described FVCT procedures do not eliminate PA and thus are unlikely
to reveal the true phoria. Additionally, the burst
vergence
response can generate significant PA.

vergence is the subtype of
vergence that is
stimulated by visual cues other than blur and disparity. There are
two major clinical methods to assess proximal
vergence. In one of
these methods, termed the AC/A differencing method, proximal
vergence is determined by calculating the difference in
vergence
change from the far-near AC/A method and the
vergence change from
the gradient AC/A method (equated for the accommodative demand). In
the other method, termed the +2.50D method, the change in
vergence
posture between distance viewing and near viewing through a +2.50D
lens is calculated. In assessing these values, it is typically
assumed that response accommodation matched the change in
accommodative demand from distance to near, which would be 2.50D
for a 40cm near viewing distance. However, individuals often do not
alter their accommodation by the amount of the accommodative
demand. Therefore, proximal values calculated using accommodative
responses might vary from proximal values that are calculated using
the accommodative demand. The purpose of the present research is to
determine the extent to which response accommodation could
influence these methods of assessing proximal
vergence, in order to
better understand how proximal
vergence is measured and therefore
to better understand the relationship between proximal
vergence and
the other
vergence subtypes in future studies. Thirteen subjects
were recruited, ages 22-37, who underwent a battery of measurements
consisting of interpupillary distance, visual acuity, stereoacuity,
accommodative amplitudes, step
vergence ranges, distance
heterophoria, and near heterophoria using various refractive
lenses. A Grand Seiko WR5100K autorefractor measured accommodation.
Accommodative data revealed high accommodative lags in many
subjects, which influenced response AC/A ratios and response
proximal
vergence results. Proximal
vergence was then calculated in
four distinct ways, two based on stimulus measurements and two
taking response accommodation into consideration. Statistical
analysis using t-tests showed no statistically significant
difference between any of these proximal
vergence calculations
after using the Bonferonni correction (p>0.0083 for all
comparisons). Additionally, no statistically significant
relationship between measures of fusional
vergence and measures for
proximal
vergence were found (p>0.05) using linear regression
analysis. In conclusion, this study found that all four clinical
methods, using stimulus and response measurements, yielded similar
values.

▼ Zinc deficiency is associated with both anxiety and depression in animal models. Zinc
supplementation reduces depression-like behaviors in the
forced swim
test and increases
grooming behavior and line crossing in the open field
test. Zinc is a toxin, however, and
excess zinc has been associated with increased anxiety-like behaviors, including a
reduced ability to extinguish fear and greater thigmotaxicity in the Morris water maze.
The addition of copper was reported to remediate the negative effects of zinc
supplementation in this case. Many of the studies reporting the anti-depressant and anti-anxiety
effects of zinc supplementation employed a single, acute zinc dose, or treatment
over a short period of time. This study employed a chronic, pre- and post-natal zinc
supplementation paradigm to determine if there would be a difference between zinc, zinc
plus copper, or lab water fed rats, on behavioral measures of anxiety and depression in
rats. It was hypothesized that the zinc-supplemented rats would display a higher amount of anxiety- and depression-like behaviors than controls, and that the zinc + plus copper
animals would show fewer anxiety- and depression-like behaviors than the zinc-supplemented
rats, similar to controls. However, the results showed that there was no
difference in depression-like behaviors between the groups on the
forced swim
test.
There was also no difference in anxiety-like behaviors between the groups on the
elevated plus maze or on day one of the open field
test. On day two of the open field
test,
there was a significant interaction between water group and time. The zinc rats spent
more time in the center at the end of the trial than at the beginning, while the zinc +
copper animals and the lab animals spent less time in the center at the end of the trial than
at the beginning. The multiple, conflicting results in this field of research may be due to
the complex mechanisms behind the relationship between zinc homeostasis and affective
disorders, but are more likely due to differences in methodology and high variability.
Although zinc supplementation seems to be a promising treatment, it should seek to
restore zinc homeostasis without leading to a state of excess zinc.

▼ The effects of THC, CBD and a combination of CBD and THC (15:1) were tested in mice models of anxiety (elevated plus maze EPM) and depression (forced swim test FST). Adult, male CD1 mice were injected intraperitoneally (I.P.) with CBD (0-120mg/kg), THC (0-8mg/), CBD + THC (up to 120+8mg/kg), diazepam (0, 2.5mg/kg) or imipramine (0, 30mg/kg). EPM ran for 5mins, FST for 6mins, and open field test for 10mins. CBD was injected 30 or 60mins prior to testing and THC was injected 30mins prior. CBD decreased immobility time in FST. THC increased time spent in the open arms in EPM, and decreased immobility time in FST. The combination of CBD and THC was better than CBD or THC alone in FST at 30mins. CBD did not show anxiolytic effects but showed anti-depressant effects. THC had both anxiolytic and antidepressant effects at some doses. CBD and THC combined might be useful in patients with anxiety and depression.

In a 2017 report, the World Health Organisation estimated that 320 million people suffered from depression globally. Although several antidepressant drugs have been delivered to the market over recent years, many patients do not respond adequately, or at all, to treatment. This has created an urgent need for safer and more effective treatments, which requires a better understanding of the underlying pathophysiology of depression. Disruption in neuronal plasticity; the ability of neurons to physically and functionally change over time, has been implicated in the pathophysiology of depression in both preclinical and clinical investigations. Microtubule dynamics, the rapid assembly and disassembly of microtubules and its constituents is a cellular mechanism allowing for neuronal plasticity. Microtubules contribute to this process primarily through axonal and dendritic remodelling that is required for the maintenance of synaptic connections. ▼ Every year around 2 million people in the United States (US) suffer a traumatic brain injury. Those people are often at a higher risk of post-TBI psychiatric illness, like anxiety and depression. Animal models of TBI are a useful way to measure such psychiatric illnesses in a pre-clinical setting. There were two purposes of this study: the first being to
test the modified Marmarou TBI model, and second to investigate anxiety- and depression-like symptoms in C57Bl6/J mice following a TBI. The modified Marmarou model used different weights (95g, 30g, & sham) to see how severity affected the manifestation of anxiety-like and depression-like symptoms. Anxiety-like behavior was tested using the elevated zero maze (ZM), and the open field
test (OF). Depression-related behavior was tested using the
forced swim
test (FST). Lastly, the effects that the TBI had on motor coordination was also tested using a rotarod. The mice were put through four trials in each
test over seven days post-TBI with a three day inter-trial interval between the first two trials and the last two. Results indicated that TBI did not cause anxiety or depression-like behaviors or any deficits involving motor coordination. In conclusion, future studies are needed in order to understand how effective this weight-drop model is, or if it was the behavioral tests that needed to be altered in order to measure anxiety and depression-like behavior more accurately.

Medical image processing can contribute to the detection and diagnosis of human body anomalies, as an important toll to minimizes the degree of uncertainty in diagnosis, and provides specialists with an additional source of information. Strabismus is one of these anomalies, which affects approximately 4% of the population. Strabismus modify the mechanism of vision, resulting in aesthetical and sensory problems. The first case is reversible at any age. Regarding sensory problems, late treatment may lead to an irreversible condition. The cover test is one
of the types of exams used to diagnose this pathology. The use of low cost computational resources to aid in the diagnosis and treatment within the strabismus subspecialty is not yet a reality. Therefore, this work presents a computational methodology to automatically pre-diagnosis strabismus through digital videos featuring cover test. To do this, the methodology was divided in 8 stages: Acquisition (1), Detection of eyes region (2), Location of pupil (3), Location of limbus (4) , Tracking eyes (5), Detection of occluder (6) , Detection (7) and Diagnosis of strabismus (8). The effectiveness of the method in the indication of the diagnosis was evaluated by comparisons made with the diagnosis provided by the expert. For detection of strabismus, the method proposed achieved 100% specificity, sensitivity of 80% and 93.33% accuracy and was demonstrated to be 87% accurate in diagnosing strabismus, while acknowledging measures lower than 1 prismatic diopter, and an average error of 2.57
prismatic diopters in deviation measure. So, was demonstrated the feasibility of using computational resources based on images processing techniques to achieve diagnosing strabismus through cover test.
O processamento de imagens médicas vem contribuindo para a detecção e o diagnóstico de anomalias no corpo humano, sendo uma importante ferramenta na redução do grau de incerteza do diagnóstico, provendo uma fonte adicional de informação aos especialistas. Uma dessas anomalias é o estrabismo, que afeta aproximadamente 4% da população e modifica o mecanismo de visão, resultando em problemas estéticos e sensoriais. O primeiro caso é reversível a qualquer idade. Já em relação aos problemas sensoriais, o tratamento tardio pode levar á um quadro irreversível. To compare with different rate of Dyneema fiber added at different ratio of AE water-reducing in the condition of concrete slump
test and slump flow
test. We planning in different water-cement ratio 0.48(non AE water-reducing)ã0.4(add AE water-reducing), to
test for its fresh properties and hardened properties, and discussion the effect by AE water-reducing and Dyneema fiber on the marine engineering.
According to this study, adding Dyneema fiber will make the slump and slump flow value dropped, and affecting the workability. Because of Dyneema fiber will tangle when add too much Dyneema fiber in concrete. So in mixing time, the Dyneema fiber and concrete will form clumps, in this study, we add the AE water-reducing to improve.
After we add AE water-reducing, the slump, slump flow, compressive strength, are increase, but we still had to pay attention to the ratio between Dyneema fiber and AE water-reducing, the strength of structure perhaps decrease if added too much Dyneema fiber or AE water-reducing.
This study can get the best positive effect when added 1.5% volume volume of Dyneema fiber.

▼ Numerous efforts have been made to illuminate the mechanisms underlying neurological and psychiatric disorders, in order to develop treatments for these diseases. With the advantage of transgenic mouse models to recapitulate human pathological alterations and to support drug screening, promising discoveries have been made over the past century. In this dissertation, I am focusing on Alzheimer’s disease (AD), which is a neurodegenerative disorder with significant public health concern. Using one of the newest mouse models of AD, the AppNL-F mouse model, I have investigated synaptic, network, and cognitive alterations related to this disease, with techniques from in vitro and in vivo electrophysiology for examining abnormalities in the brain to behavior tests for assessing the cognitive functions of the animals. In the meantime, novel quantitative approaches have been developed and applied to objectively interpret the experimental data. In AppNL-F homozygous mice, we have found synaptic alterations consistent with deficits in parvalbumin-expressing interneurons (PV INs). Moreover, through long-term in vivo local field potential (LFP) recording, we have discovered perturbed network activities associated with disrupted sleep patterns, which happened during a critical period in the disease progression and may lead to further cognitive deficits in these animals. Besides well-known cognitive dysfunctions such as loss of memory and reasoning as well as language difficulties, there is a second group of non-cognitive symptoms including psychiatric disorders and behavioral disturbances, such as depression, hallucination, delusions, and agitations(Burns et al., 1990; Burns and Iliffe, 2009). Therefore, I have also investigated into the mouse forced-swim test (FST), which is widely used in the screening of antidepressants. By quantitative analyses on this test, we have discovered a new confounding factor, and developed novel methods to determine the important parameters in the mouse FST, which could be applied to mouse AD models to study depression and the effect of antidepressants in AD. In conclusion, the application of quantitative approaches in neuroscience studies have led to the discovery of intriguing results and provided ground for exciting future research.

▼ Neurotensin is a neuropeptide that influences monoaminergic neurotransmission in areas of the brain involved in the pathophysiology of depression. The
forced swim
test is a commonly used screening model for putative antidepressant medications. Drugs that have antidepressant effects in humans reliably decrease the time animals spend in an immobile posture in the
forced swim
test without increasing general locomotor activity as measured in an open field
test. The present study sought to examine the effects of the neurotensin NTS
1 receptor agonist PD149163 and the tricyclic antidepressant drug imipramine on immobility in the
forced swim
test and on locomotor activity in an open field in mice. PD149163 decreased the total time spent immobile at doses of 0.1 mg/kg and 1.0 mg/kg, without increasing overall locomotor behavior in the open field
test. Imipramine produced a reduction in immobility at a dose of 10.0 mg/kg, but not at 1.0 mg/kg, without increasing locomotor activity. These results indicate that since drugs that target the neurotensin system display antidepressant properties in rodent models of depression they may represent a novel mechanism for treatment of depressive symptoms in humans.

▼ The interactions between stress and allergic pulmonary inflammation are not well understood. Experiments were conducted utilizing the ovalbulmin (OVA) sensitization and aerosol challenge to induce allergic pulmonary inflammation in mice and stimulate the T helper (Th2) milieu immunity in female Balb/c mice. An Ovalbumin (OVA) sensitization and aerosol challenge model was utilized. The first study utilized an amphetamine withdrawal model as the stressor. There were no significant immune x stress interactions. An additive effect was observed for some Th2 cytokines and lung inflammation. The second study evaluated lipopolysaccahride challenge and
forced swim
test (FST), effects on mouse plasma corticocosterone levels at 20, 50, and 80 min after the start of exposure. FST treated mice, 50 minutes after exposure had the highest levels of corticosterone. Control mice also had elevated levels of corticosterone compared to literature baseline levels. The third study attempted to establish a time when full lung inflammation occurred. Sampling times included 0, 12, 24, 36 and 48 h after aerosol challenge. Mice harvested 0 to 24 h post allergen exposure had the most complete inflammatory response (not significant). The final study examined the effects of the FST on allergic pulmonary inflammation using the OVA induced allergy mouse model. Th2 cytokines in BAL fluid were elevated among allergic mice, the stressor reduced IL-5 and IL-6 concentrations in allergic mice, while the stressor alone (non-allergic) caused an increase in IL-4 and IL-10.

▼ Receptores CB1 e TRPV1 desempenham papéis opostos na modulação da atividade neuronal e, possivelmente, na regulação da resposta ao estresse. A exposição ao estresse reduz a neurotransmissão mediada por receptores CB1, enquanto que a facilitação do sistema endocanabinóide produz efeito tipoantidepressivo. Por outro lado, estudos farmacológicos e de manipulação genética apontam que a diminuição da sinalização mediada por receptores TRPV1 produz efeito tipo-antidepressivo em modelos animais. Evidências científicas apontam que a modulação da neurotransmissão glutamatérgica, dependente de receptores NMDA, esteja envolvida em respostas mediadas por CB1 e TRPV1. Ambos os receptores são amplamente expressos em estruturas cerebrais envolvidas na resposta emocional, incluindo o córtex pré-frontal medial ventral (CPFmv), o que aponta para essa estrutura como importante alvo para os efeitos mediados por receptores CB1, TRPV1 e NMDA. Entretanto, pouco se sabe
sobre a interação entre CB1 e TRPV1 corticais na resposta ao estresse. Dessa forma, avaliamos se receptores CB1 e TRPV1 localizados no CPFmv-pré-límbico (PL) podem contribuir, de maneiras opostas, para as mudanças comportamentais induzidas pelo estresse do teste do nado forçado (TNF), um teste preditivo de efeito tipo-antidepressivo. Em um primeiro grupo experimental observou-se que a AEA induz efeito tipo-antidepressivo com uma curva dose-resposta em U, quando administrada no CPFmv-PL. Além disso, o efeito tipo-antidepressivo da AEA foi prevenido pela pré-administração de AM251, um antagonista para receptores CB1. Por outro lado, a maior dose de AEA, que não induz efeito per se, produziu efeito tipo-antidepressivo quando combinada ao bloqueio de receptores TRPV1 por meio do pré-tratamento com SB366791. Corroborando estes dados, observou-se que a administração intra-CPFmv-PL de um bloqueador dual da enzima FAAH e de receptores TRPV1 (AA-5HT), induziu efeito tipo-antidepressivo no
teste do nado forçado. Além disso, a administração do inibidor da FAAH (URB597) e do antagonista de receptores TRPV1 (SB366791), em grupos independentes de animais, reduziu o tempo de imobilidade no teste do nado forçado. Ademais, a coadministração de doses subefetivas de URB597 e SB366791 reduziu o tempo de imobilidade no mesmo teste comportamental, evidenciando somação de efeito do bloqueio combinado da FAAH com receptores TRPV1. Em conjunto, esses ii resultados indicam que tanto a facilitação da neurotransmissão mediada por CB1R quanto o bloqueio de TRPV1R no CPFmv-PL promove efeito tipo-antidepressivo, sugerindo que ambos os receptores corticais são importantes na modulação de respostas comportamentais ao estresse e, possivelmente, na neurobiologia da depressão, porém de maneiras opostas. ▼ Hibiscus tiliaceus L. (Malvaceae) é usado popularmente em desordens do pós-parto. O extrato metanólico de flores de H. tiliaceus apresentou atividade antidepressiva-like nos testes de nado forçado e de suspensão da cauda, ambos são amplamente usados como modelos animais preditivos da atividade antidepressiva. Além disso, o extrato demonstrou uma tendência de aumento do tempo gasto nos braços abertos no labirinto em cruz elevado. Considerando que o extrato de H. tiliaceus, que contém fitoesteróis como o estigmasterol, o stigmastadienol e o stigmastadienone, pode ser útil no tratamento ou prevenção da depressão pós-parto relacionadas à retirada crônica de altos níveis de hormônios associados à gestação. Nosso objetivo foi avaliar o efeito de extratos de H. tiliaceus em modelo animal de depressão pós-parto e ansiedade. Utilizamos ratas Wistar fêmeas submetidas ao modelo de depressão pós-parto induzido por hormônios (estradiol e progesterona),
administrados por via subcutânea. Após período gestacional induzido, os animais receberam água, veiculo ou extrato de H. tiliaceus nas doses de 100 e 400mg/kg, via oral (gavagem). No período pós-parto, as ratas foram submetidas a testes comportamentais de nado forçado, o labirinto em cruz elevado e de preferência claro-escuro como modelos preditivo de depressão e ansiedade. Foi obsevado um aumento no tempo de imobilidade associado a uma diminuição na tentativa de fuga, no teste do nado forçado, em animais submetidos ao modelo de depressão pós-parto comparado ao grupo controle, sugerindo que este modelo é capaz reproduzir sintomas de depressão pós-parto. Não houve diferença significativa nos testes de ansiedade entre os animais submetidos ao modelo de depressão pós-parto comparado aos controles. A administração de extrato metanólico de H. tiliaceus não foi capaz de alterar o comportamento relacionado à depressão e ansiedade em ratas Wistar.
Postpartum affective disorders are rarely
modeled. The depressive-like behavior of hormone withdrawal following hormone-simulated "pregnancy" was described in Long-Evans and Sprague Dawley rats. Our aim was to evaluate the validity of hormone withdrawal following hormonesimulated "pregnancy" method in Wistar rats as a model of depression and/or anxiety. Recently, it was demonstrated an antidepressant-like profile of methanol extract of Hibiscus tiliaceus L., a plant used in postpartum disorders, in adult male Swiss albino mice, then, we also investigated the antidepressant and anxiolytic-like activities of the methanol extract of H. tiliaceus flowers using this animal model of postpartum disorder. ▼ The United States contains an estimated 100,000 miles (160000 km) of levees in
which erosion related issues are the top priorities. Proper documentation of overtopping
induced erosion is a complicated issue involving the collection and analysis of timesensitive
field data and personal observations. This thesis is a study of the performance
of the Midwest Levee system during the 2008 flooding events.
The goal of the Midwest Levee investigation was to gather and analyze
perishable data in an effort to provide a comprehensive overview at each breach
location. To predict how a site will perform during a particular flood event, there are
three main inputs: the flood conditions, the site conditions, and the soil properties. Site
geometry and imperfections can greatly affect the performance of a levee system. Any
low spots or potential seepage paths can concentrate the flow and be detrimental to the
levee.
The vegetative
cover is the single most important condition at a site. As seen in
the Brevator case, vegetative armor can prevent failure of a levee comprised of less
resistant soils subjected to long periods of overtopping. Recommended grasses include:
Switchgrass, Smooth Brome, Reed Canarygrass, and Tall Fescue. It is also recommended that grasses are kept at least 0.5 m tall during the flood season and to limit
the presence of trees to 10 m beyond the levee toe.
The erosion resistance of the materials comprising the levee is also important.
From the correlations in this study, it was determined that erodibility is influenced by
grain size, relative compaction, clay content, and activity. Devices like the Torvane and
Pocket Erodometer can also be used to get a quick field estimate of erosion. While these
correlations and field devices give insight into an erodibility value, they are no substitute
for site specific analysis with laboratory equipment such as the Erosion Function
Apparatus. Soil behavior is highly nonlinear and the entire erosion function is needed to
get an accurate measure of the erodibility of a soil. By combining these properties in an
erosion matrix, a prediction of whether a site will withstand a given flood event can be
made.

▼ Since 1971, a number of flood frequency models have been developed for river basins in
Malawi for use in the design of hydraulic structures, but the varied nature of their results
have most often given a dilemma to the design engineer due to differences in magnitudes
of calculated floods for given return periods. All the known methods for flood frequency
analysis developed in country so far have not used a homogeneity
test for the river basins
from which the hydrological data has been obtained. This study was thus conducted with a
view to resolving this problem and hence improve the design of hydraulic structures such
as culverts, bridges, water intake points for irrigation schemes, and flood protection dykes.
In light of the above, during the course of this study the applicability of existing methods
in the design of hydraulic structures was assessed. Also, the study investigated how land
use and land
cover change influence the frequency and magnitude of floods in the study
area, and how their deleterious impacts on the socio-economic and natural environment in
the river basins could be mitigated

Land use / land cover (LULC) change detection based on remote sensing (RS) data
is an important source of information for various decision support systems. In East
Timor where forest plays a key role in sustaining communities’ livelihoods the
information derived from LULC change detection is invaluable to the conservation,
sustainable development and management of forest resources.
To assess the patterns of land cover change, as a result of complex socio-economic
factors, satellite imagery and image processing techniques can be useful. This study
is concerned with identifying change in land use and land cover types in East Timor
between 1972 and 2011, using satellite images from Landsat MSS, TM and ETM+
sensors. Seven major cover types were identified in this study including forest,
mixed rangeland, grassland, farmland, built-up areas, bare soil and water. A
combination of NDVI differencing, supervised and unsupervised classification was
used to derive final classification maps. Due to the lack of ground truth data, further
processing were performed to improve the final classification maps by applying
rationality change test.
Post-classification comparison change detection technique was used to assess
categorical changes between 1972 and 2011. The results highlight a significant
level of deforestation due to uncontrolled illegal logging and increase in farmland,
built-up areas, as well as bare soil. This decline has had considerable impact on the
livelihoods of rural communities. As the new nation of Timor-Leste establishes
itself, it must consider its current stock and distribution of natural resources to
ensure that development efforts are geared towards sustainable outcomes. Without
this information historical patterns of resource consumption, development efforts
may, unwittingly, lead to continuing decline in forest resources.

▼ This study was to evaluate clinical efficacy of photography with Spielmann translucent occluder in intermittent exotropia. A total of 271 patients with intermittent exotropia were included. Photographs were obtained in alternate cover test with Spielmann translucent occluder, and analyzed by 3 observers (2 strabismus pediatric ophthalmology specialists and 1 ophthalmology resident). The angle of photo-analysis was compared with that of prism alternate cover test. We also measured inter-observer variability and the effect of the type of exotropia, the direction of suppression, eyelid features and control grade. Photo-analysis and prism alternate cover test showed a strong positive correlation (at distant R=0.605, at near R=0.531; P <0.001), and the standard error was 3.6 prism diopter (PD). The standard error in inter-observer difference was 3.3PD (ICC=0.786; P <0.001). In basic type of intermittent exotropia, there was positive correlation between the angle of photoanalysis and that of prism alternate cover test, while no correlation in convergence insufficiency type and divergence excess type. The angle of deviation in photo analysis was lower than that of prism alternate cover test in patient who have good control grade, but not statistically significant. There was no difference according to the direction of suppression. Photography with Spielmann translucent occluder can be used as an objective tool for measuring the angle of exotropia deviation. However, there is possibility of errors according to the degree and type of exotropia and control grade.
반투명가리개를 이용하여 간헐외사시를 촬영한 사진의 유용성을 평가하고자 하였다. 간헐외사시 환자 271명을 대상으로 Spielmann반투명가리개를 이용하여 한눈씩 교대로 가리면서 정면사진을 촬영하였다. 사진에서 보이는 사시각을 안과의사 3명이 표준사진을 바탕으로 판독하여 프리즘교대가림검사로 측정한 사시각과 비교하였다. 검사방법간, 검사자간 차이를 분석하였고, 간헐외사시의 종류, 억제, 눈꺼풀모양, 조절정도가 사진촬영에 영향을 미치는지 조사하였다. 사진 사시각은 실제 사시각과 뚜렷한 양의 상관관계가 있었고(원거리 R=0.605, 근거리 R=0.531, P <0.001), 3.6프리즘디옵터(PD)의 표준오차를 보였다. 사진 사시각의 검사자간 표준오차는 3.3PD이었다(ICC=0.786, P <0.001). 기본형에서는 사진 사시각이 실제 사시각을 반영하였지만, 눈모음부족형과 눈벌림과다형에서는 상관관계가 없었다. 조절정도가 좋은 환자에서 사진 사시각이 실제 사시각보다 적은 경향이 있었으나, 억제유무와 방향에 따른 차이는 없었다. 간헐외사시에서 반투명가리개를 이용하여 촬영한 사진 사시각은 실제 사시각을 비교적 반영하였지만, 사시 정도와 종류, 조절정도에 따른 영향이 있을 수 있으므로 해석에 주의하여야 하겠다.

▼ There is a lack of rapid test methods for accurately assessing the impact of curing on the differential hydration through the depth of the concrete cover as well as the impact on chloride penetration resistance. In the absence of adequate performance assessment tools, prescriptive curing specifications have been adopted for concretes exposed to chlorides and other exposures, such as in CSA A23.1-19. There is a desire to switch to a performance specifications for curing, particularly by the precast concrete industry, that could be used to assess the impact of using of accelerated heat curing methods widely used to obtain high early-strength gain and maturity.
Two methods are presented and evaluated in this thesis. The first involves proling the initial rate of absorption of a sodium chloride solution. The second makes use of embedded arrays of electrodes to map the formation factor with depth, enabling a multi-mechanistic approach to the problem.

▼ Vergence is the disjunctive movement of the eyes to maintain single binocular vision.
Vergence eye movements are necessary to maintain the object of interest on the fovea of each eye as an individual looks from one object to another. Recent studies show that office based
vergence/accommodative therapy (OBVAT) is an effective treatment for the binocular dysfunction known as convergence insufficiency. This study was performed to investigate the changes in oculomotor parameters parameter data for pre- and post-therapy subjects who are binocularly normal controls. A haploscope was used to collect eye movement data pre- and post-therapy. The analysis of the eye movements was done in MATLAB. Fifty binocularly normal controls participated in 12 hours of office-based therapy where half participated in OBVAT and the remaining half participated in office-based placebo therapy (OBPT) therapy. The latency, time to peak velocity, peak velocity, response amplitude, final amplitude, and main sequence ratio were measured for participant’s responses to 4-degree and 6-degree ramps, 4- and 6-degree disappearing steps, 6- and 10-degree stepramps, and 5- and 10-degree saccades. Peak velocity was significantly greater post OBVAT therapy compared to baseline, most of them having a p≤.001. Clinically meaning differences were not observed post OVPT compared to baseline. Results support that OBVAT significantly changes
vergence dynamics and may be used for sports enhancement.

▼ Treatment of major depression, posttraumatic stress disorder and other psychopathologies with antidepressants can be associated with improvement of the cognitive deficits related to these disorders. Although the mechanisms of these effects are not completely elucidated, alterations in extinction of aversive memories are believed to be present in these psychopathologies. Moreover, researches with laboratory animals usually focus on male subjects, and we have recently verified that extinction of an aversive task is reduced in female rats when compared to males. In the present study, female rats were long-term treated with clinically used antidepressants (fluoxetine, nortriptyline or mirtazapine) and tested in the plus-maze discriminative avoidance and
forced swimming tests in order to evaluate learning, memory, extinction, anxiety and depression-related behaviors. All groups learned the task, but learning was somewhat faster in nortriptyline and mirtazapine-treated animals . Task retrieval was also showed by all experimental groups. Chronic treatment with fluoxetine, but not with the other antidepressants, increased extinction of the discriminative task. In the
forced swimming
test, animals treated with fluoxetine and mirtazapine showed decreased immobility duration. In conclusion, antidepressants interfere with learning and female rats treated with fluoxetine presented increased extinction of the aversive memory task. On the other hand, both fluoxetine and mirtazapine were effective in the
forced swimming
test, suggesting dissociation between the antidepressant effects and the extinction of aversive memories

Previous researches showed that depression is the most frequently diagnosed psychiatric disorder after traumatic brain injury (TBI). Neuroinflammation plays an important role in this issue. Our previous study demonstrated that hyperbaric oxygen (HBO) therapy in after TBI can attenuate neuroinflammation, resulting in a neuroprotective effect. We also found preexisting hyperlipidemia to be an independent predictor of new-onset depression and anxiety in patients with TBI. In this thesis, in the basic research, we will elucidate the therapeutic effect of HBO and Simvastatin in acute stage of TBI-induced anti-depression-like behavior by rat model. In addition to TBI, psychological disorders may also occur after traumatic spinal cord injury (tSCI). Therefore, in the clinical research, we conducted a large nationwide Taiwanese population-based study to retrospectively examine the relationship between tSCI and the risk of new-onset anxiety or depression in comparison with other health condition groups. We also hypothesized that preexisting hyperlipidemia may also a risk factor for new-onset of anxiety and depression in patient with tSCI.
Methods and results
There are basic research and clinical research two parts in this study:
Fluid percussion device was used in anesthetized rats to produce a moderate severity brain trauma. In the basic research I study, animals were divided into 3 groups: sham operation plus normobaric air (NBA), TBI plus NBA, and TBI plus HBO. HBO was applied immediately for 60min/d after TBI for 3 days.
In the basic research II study, animals were divided into 5 groups: sham-operated controls, TBI controls, and TBI treatment with Simvastatin 4, 10, or 20mg/kg. Simvastatin was intraperitoneally injected 0, 24, and 48 hours after TBI. The motor function was measured using an inclined plane, and depression-like behavior was evaluated using
forced swimming tests on the 4th, 8th, and 15th day after TBI in all groups. Neuronal apoptosis, microglia and astrocyte activation, and TNF-α expression in the microglia and astrocytes of the hippocampal CA3 area were investigated using immunofluorescence assay.
TBI-induced depression-like behavior, which increased duration of immobility, was significant attenuated by HBO or 20mg Simvastatin therapy on day 15 after TBI. TBI-induced neuronal apoptosis, microglia and astrocyte activation, and TNF-α expression in the microglia and astrocytes of the CA3 area of the hippocampus were significant reduced by HBO or Simvastatin treatment, particularly when 20mg/kg was administered for 3 days.
In the clinical research for tSCI, the Longitudinal Health Insurance Database (LHID) from Taiwanâs National Heath Insurance Research Database (NHIRD) was used for retrospective cohort study. Individuals with tSCI were identified using the International Classification of Diseases, Ninth Revision, Clinical Modification (ICD-9-CM) diagnostic codes. ▼ Direct sun drying is one of the oldest methods of food preservation. A drying method that utilizes solar energy while minimizing negative aspects of sun drying is a solar dryer. However, research about the quality characteristics of fruits and vegetables dried using a solar dryer compared to a traditional dryer is lacking. To measure this, apple, tomato and carrot slices, and grape halves and whole peas were prepared and dried using either a solar or FA dehydrator (FA), Consumer liking was measured using two sensory panels that asked panelists 9-point hedonic questions between like products. Quality differences were analyzed using a TA.XT2 texture analyzer. Color was measured using a Hunterlab colorimeter. There were no significant differences in consumer liking (p<0.05) between drying methods for any fruits and vegetables with regards to overall acceptability. Grapes, tomatoes and carrots all had significant differences in consumer liking with regards to appearance, which correlates well with observed differences in a* values. Consumers liked the aroma of solar-dried peas significantly more. The texture of FA grapes was liked significantly more than their solar counterparts, which likely contributed to consumers significantly ranking the FA grapes higher than their solar counterparts when asked to rank which one they prefer.

▼ Voluntary wheel running is rewarding and believed to activate the same brain reward system as in alcohol and drug addiction. Brain-derived neurotrophic factor (BDNF), a well-known growth factor widely expressed in the brain, is modulated by both voluntary exercise and alcohol consumption. The aim of this study was to evaluate how voluntary exercise affects the expression levels of BDNF and its receptor TrkB in brain regions involved in positive and negative reinforcement. Additionally we wanted to evaluate the effect it may have on alcohol drinking behaviors in C57BL/6 mice, a mouse model which are naturally prone for engaging in voluntary exercise and voluntary alcohol consumption. We found a small upregulation in DG and CA1 after three weeks of exercise, confirming findings by others, and a significant 3-fold downregulation of BDNF in NAc after both three weeks of exercise and exercise followed by a five week period of either ethanol intake or not. Interestingly, we here show a significant 100-fold increase in BDNF after exercise and a 120-fold increase after both exercise and alcohol consumption in amygdala, a region involved in regulation of anxiety-related behavior and negative reinforcement. Additionally a slightly lower 10-fold increase in BDNF was seen after exercise and a 15-fold increase after exercise followed by ethanol in prefrontal cortex, a structure contributing to reward-related behavior. Behaviorally, we could not either directly following exercise or at five weeks post-exercise detect any significant effect of wheel-running on depression-related behavior. However, we did find that exercise significantly increased the alcohol intake.

▼ Early life events, in both humans and animals, may influence on brain development, leading to long-term consequences on adult behavior. Thus, this study was carried out in order to assess the influence of neonatal tactile stimulation (TS), when associated to sub-therapeutic administration of sertraline (SERT) in rats, on behavioral parameters related to depression-like. We also evaluated the influence of the TS-SERT association on the hypothalamic-pituitary-adrenal axis, as well as on the development of anxiety-like symptoms, assessed in different behavioral. Male rat pups were daily submitted to 10 min TS, from postnatal day (PND) 8 to PND 14. On DPN50, rats were first exposed to forced swimming test (FST). Then, after 24h, it was conducted the SERT administration and, 30 minutes later, animals were submitted to new evaluation on FST. On DPN51, rats were evaluated on the elevated plus maze (EPM). The animals were euthanized 24h after the
behavioral tasks, followed by biochemical assessments. TS per se decreased depression behaviors, since there was an increase on swimming time and a decrease on immobility time in FST. On the EPM task, TS diminished behaviors related to anxiety, as shown in the increase of time on open arms and decrease on anxiety rates. In addition, TS also decreased corticosterone and cortisol plasma levels. All these results were enhanced when TS was associated to a sub-therapeutic dose of SERT. From these results, we suggest that neonatal handling TS may accomplish beneficial influence on parameters of depression, promoting the ability to cope with stressful situations in adulthood.
Eventos ocorridos em estágios iniciais do desenvolvimento, tanto em humanos quanto em animais, influenciam o desenvolvimento do cérebro e podem ter consequências em longo prazo na idade adulta. Assim, o presente estudo foi realizado para avaliar a influência da estimulação tátil neonatal (ET), associada à
administração de uma dose subterapêutica de sertralina (SERT), sobre parâmetros comportamentais relacionados a depressão em ratos. A influência da associação ET-SERT sobre o eixo hipotálamo-pituitária-adrenal, assim como sobre o desenvolvimento de comportamentos de ansiedade foram também avaliados. Filhotes machos de ratos Wistar foram diariamente submetidos à ET durante 10 min, desde o dia pós-natal (DPN) 8 até o DPN14. No DPN50, os animais foram previamente expostos ao nado forçado. Após 24h, receberam a administração de SERT e, decorridos 30 minutos, foram novamente avaliados mediante o estresse do nado forçado. No DPN 51, os animais também foram avaliados no labirinto em cruz elevado (LCE). Os animais foram eutanasiados 24h após os testes comportamentais, seguindo-se as avaliações bioquímicas. A depressão é um transtorno psiquiátrico com alta incidência na população. O tratamento envolve fármacos que possuem uma série de efeitos adversos que, em muitos casos, diminuem a adesão do paciente ao tratamento. Isto tem incentivado a busca por novos fármacos e o reino vegetal se apresenta como uma alternativa. Os flavonoides são uma classe de metabólitos secundários encontrados em diferentes espécies de plantas. A
quercetina (3, 5, 7, 3’ – 4’ pentahidroxi flavona), um importante flavonoide, possui diversas atividades biológicas, entre elas: atividade anti-inflamatória, anti-agregante, anti-hipertensiva, antidiabética, ansiolítica e antidepressiva. Esta última atividade, no entanto, ainda não é bem caracterizada e este fato levou ao desenvolvimento do presente trabalho, o qual teve como objetivo avaliar o efeito antidepressivo-símile da quercetina, por meio de testes comportamentais e neuroquímicos. Foram utilizados camundongos Swiss machos (protocolo CEUA/UFPB n. 112/2016), provenientes da Unidade de Produção Animal da Universidade Federal da Paraíba – UPA/UFPB. Os animais foram divididos em grupos compostos por seis camundongos, tratados por via oral com salina, quercetina 10 ou 40 mg/kg e por via intraperitoneal com imipramina 30 mg/kg, e submetidos aos testes do campo aberto e nado forçado. Para investigação do possível mecanismo de ação, a quercetina foi administrada isoladamente (0,5
mg/kg, v.o.) e em associação aos antidepressivos bupropiona (0,1 mg/kg, i.p.), fluoxetina (0,1 mg/kg, i.p.) ou imipramina (1 mg/kg, i.p.) em animais submetidos ao nado forçado. Em adição foram realizadas análises neuroquímicas para determinar a concentração estriatal de monoaminas e a concentração hipocampal de nitrito e TBARs. Os resultados mostraram que a quercetina não alterou a atividade locomotora no teste do campo aberto; por outro lado, o composto reduziu de forma significativa e dose-dependente o tempo de imobilidade dos animais no nado forçado. Este efeito foi potencializado na presença de bupropiona e imipramina, mas não de fluoxetina. A quercetina (10 mg/kg, v.o.) não alterou a concentração hipocampal de nitrito e TBARs; por outro lado, o composto aumentou significativamente a concentração de noradrenalina, mas não de dopamina, no estriado de animais submetidos ao teste do nado forçado. ▼ Recentemente suspeitas de que mecanismos epigenéticos poderiam estar relacionados à fisiopatologia da depressão foram levantadas. Estudos recentes indicam que as alterações na transcrição gênica, induzidas por estresse ou por drogas antidepressivas, parecem envolver mecanismos epigenéticos. Nesse sentido, resultados preliminares de nosso grupo de pesquisa indicaram pioneiramente inibição global da metilação de DNA através da administração sistêmica do agente inibidor da DNA metiltransferase (DNMTs), 5-aza-2-deoxicitidina (5-azaD), induz efeito tipo-antidepressivo, dose-dependente, no modelo animal do nado forçado em ratos(Sales et al., 2011). O córtex pré-frontal medial ventral (CPFMv) é uma estrutura límbica intimamente relacionada com a neurobiologia da depressão. Evidências recentes indicam que o efeito tipo-antidepressivo aparece associado a aumento dos níveis da neurotrofina BDNF (brain derived neurotrophic factor) e de seu receptor TrkB no
CPFMv, sendo a sinalização intracelular mediada pela ativação da proteína m-TOR. Contudo, não há evidências de que esses mecanismos moleculares estariam envolvidos nos efeitos induzidos pelos inibidores da metilação do DNA. Sabe-se, no entanto, que tanto o BDNF quanto TrkB têm sua expressão regulada por metilação do DNA. Diante disso, o objetivo presente trabalho será investigar a participação da via BDNF-TrkB-mTOR do CPFMv no efeito antidepressivo induzido por inibidores da metilação de DNA. Para tanto, ratos tratados com inibidores da metilação de DNA (5-azaD ou RG-108), em dois momentos diferentes (imediatamente após o PT e 23horas após o PT) foram submetidos ao teste do nado forçado (FST). Outro grupo de animais recebeu uma injeção intra-CPMv de k252a ou Rapamicina, 40 minutos antes do teste e uma injeção de BDNF intra-CPFMv, 30 minutos antes do teste. Em outro experimento, grupos independentes de animais submetidos ao nado forçado foram tratados sistemicamente com RG-108 e
receberam injeção intra-CPFMv de K252a (antagonista de Trk) ou de rapamicina (inibidor da m-Tor), a fim de investigar se o efeito dessas drogas depende da via BDNF-TrkB-mTOR no CPFMv. Um grupo independente foi tratado com RG108 e CPFM desses animais foi dissecado para posterior análise da expressão de BDNF, TrkB e m-TOR, bem como da metilação de DNA. O tratamento com RG108 e 5azaD sistêmico reduziu o tempo de imobilidade dos animais submetidos ao nado forçado nos dois tempo de administração. A administração intra-CPFMv de BDNF promoveu efeito antidepressivo no FST, e esse efeito foi bloqueado pela administração de k252a ou Rapamicina no CPFMv. No mesmo sentido, o efeito antidepressivo do RG108 sistêmico foi bloqueado pela administração intra-CPFMv de k252a ou Rapamicina. Entretanto, a medida dos níveis de metilação global no CPFMv não apresentou alteração como tratamento com RG108, e também não mostrou alteração nos níveis de BDNF presente no CPF. O tratamento com RG108 não alterou a

▼ Phosphodiesterases (PDE) regulate secondary messengers
such as cyclic adenosine monophosphate (cAMP) and cyclic guanosine
monophosphate (cGMP) by hydrolyzing the phosphodiester bond. There
are over 100 PDE proteins that are categorized into 11 families.
Each protein family has a unique tissue distribution and binding
affinity for cAMP and/or cGMP. The modulation of different PDEs has
been used to treat several disorders: inflammation, erectile
dysfunction, and neurological disorders. Recently, PDE inhibitors
were implicated for therapeutic benefits in Alzheimer’s disease,
depression, Huntington’s disease, Parkinson’s disease,
schizophrenia, and substance abuse. PDE1B is found in the
caudate-putamen, nucleus accumbens, dentate gyrus, and substantia
nigra–areas linked to depression. PDE1B expression is also
increased after acute and chronic stress. Two ubiquitous Pde1b
knockout (KO) mouse models, both removing part of the catalytic
region, decreased immobility on two acute stress tests associated
with depression-like behavior; tail suspension
test (TST) and
forced swim
test (FST). The decreases in immobility suggest
resistance to depression-like behavior, and these effects were
additive when combined with two current antidepressants, fluoxetine
and bupropion. The resistance to induced immobility was seen when
PDE1B was knocked down during adolescence or earlier. Expression of
Pde1b mRNA and protein showed that Pde1b is localized to
dopaminergic and glutamatergic post-synaptic cells. Serotonin
transporter, dopamine transporter, and dopamine receptor D1a
(Drd1a) Cre drivers were tested and only Pde1b KO in Drd1a specific
cells was sufficient to reproduce the immobility phenotype in TST.
Intrastriatal injections of dopamine receptor, D1, agonist
(SKF38393) and antagonist (SCH23390) revealed an additive
immobility effect of Pde1b KO and dopamine receptor antagonism. The
D1 agonist reversed the genotype immobility phenotype. The results
suggest that reduction of PDE1B causes resistance to acute
stress-induced depression-like behaviors through the dopamine
pathways.

▼ Vergence eye movements alter the angle between the two visual axes, creating changes in binocular fixation distance. They are primarily stimulated by retinal image disparities, but can also be driven by inputs from ocular accommodation (accommodative-vergence) and perceived proximity (size) changes. Because of these diverse and complex sensory inputs, the neuro-motor substrates that sub-serve vergence control possess robust adaptive capabilities to manage the interactions with other oculomotor systems (accommodation). This adaptive plasticity in vergence allows for a high degree of precision in binocular alignment to be maintained throughout life in the face of constantly changing environmental demands.
The precise alignment of each eyes’ fovea is a fundamental requirement for stereopsis and the perception of depth in 3 dimensions. In a significant portion of the ophthalmic clinical population, the adaptive capacities of vergence are reduced or dysfunctional, leading to difficulties focusing clearly and comfortably at near distances such as books, computer screens and other hand-held devices. Furthermore, new wearable technologies such as virtual and augmented reality increase the demand on the adaptive capacities of vergence by drastically altering the congruency of the sensory inputs to vergence. Currently, our understanding of the mechanisms that underlie this adaptive control and their behavioral limits are limited. This knowledge gap has led to conjecture in the literature regarding proper rehabilitative therapies for clinical dysfunctions of vergence control and in the optimal environmental design parameters that should provide comfortable and compelling user experiences in wearable technologies like VR and AR.
The inward (convergence) and the outward (divergence) turning of the eyes in response to retinal disparities are controlled by two separate systems and demonstrate significant directional asymmetries in their reflexive response properties. In general, reflexive divergence responses tend to be slower and longer than their convergence counter-parts. It is unclear whether the adaptive mechanisms are influence by these reflexive asymmetries. It is also unknown whether similar directional differences exist in the different adaptive capacities possessed by vergence. The purpose of the following dissertation was to characterize the effects of stimulus direction on the adaptive behavior of disparity-driven vergence eye movements with an end goal aimed at improving rehabilitation therapies for clinical populations with vergence dysfunction and providing valuable insight for the design and future development of wearable technologies like virtual and augmented reality environments.

▼ Joint development of perception and behavior based on efficient coding theory has been proposed and proved to be an effective approach in the study of eye movements. Disparity vergence, one type of eye movement, is believed to achieve robustness and accuracy by the cooperation between fovea and periphery. Complicated realistic environments, where objects in different depths project conflicting disparity information on different regions of the retina, cannot be handled well by previous joint development models of vergence control, which combine information from fovea and periphery in parallel. Here, we propose a hierarchical approach to integrating foveal and peripheral vision for vergence control. The model consists of two levels. At the bottom level, three options receive input from the fovea, the inner periphery, the outer periphery of the retina. At the top level, a policy chooses one of the three options for vergence control. Our experimental results show that the hierarchical model for integration of fovea and periphery provides more accurate and robust vergence control than a previous model where the sensory representation from different scales was combined in parallel. In addition, the hierarchical model shows good performance in dealing with a wide range of situations, such as small objects moving in depth, a lack of content in the fovea, and occlusion. Furthermore, a periphery to fovea process is observed when applying the hierarchical model in vergence control, which is consistent with psychological evidence found by other researchers.
