▼ Consumption of whole grains has been linked to reduced risk of colon cancer. This study determined
estrogenic activity of sorghum phenolic extracts of different phenolic profiles and identified possible
estrogenic compounds in sorghum in vitro, as well as evaluated the potential of
estrogenic sorghum phenolic extracts to prevent colon carcinogenesis in vivo. The thermal stability of sorghum 3-deoxyanthocyanins was also studied, to determine their suitability as functional food colorants.
White and TX430 (black) sorghum extracts showed
estrogenic activity in cell models predominantly expressing estrogen receptor-α (ERα) or ERβ at 5 and 10 µg/mL, respectively. The same treatments led to induction of apoptosis in cells expressing ERβ. The red TX2911 sorghum did not possess these activities. Compositional analysis revealed differences in flavones and flavanones. Flavones with estrogen-like properties, i.e. luteolin and apigenin, were detected in White and TX430 (black) sorghum extracts, but not in red TX2911 extract. Naringenin, a flavanone known to antagonize ERα signalling, was only detected in the red TX2911 extract. Additional experiments with sorghum extracts of distinct flavones/flavanone ratio, as well as with pure apigenin and naringenin, suggested that flavones are the more potent ERβ agonists in sorghum. On the other hand, 3-deoxyanthocyanins were probably not
estrogenic.
Estrogenic white and black sorghum phenolic extracts (fed at 1% level in the diet) reduced the number of azoxymethane induced colon premalignant lesion (aberrant crypt foci) by 39.3% and 14.7%, respectively, in ovariectomized mice. Further studies are needed to elucidate the protective mechanisms induced by these sorghum extracts.
Sorghum 3-deoxyanthocyanins retained good color stability after 30 minutes of heat treatment at 121 °C under pressure: More than 80% of color retained in pH 1 and 2 HCl and citric acid solutions, and 39-84% retained from pHs 3-7. Formic acid negatively affected the color stability at pH 1 and pH 2 due to its reducing capacity. Methoxylation decreased the thermal stability of 3-deoxyanthocyanins. The heat stability of 3-deoxyanthocyanins indicates good potential for food use.
Overall, the inherent
estrogenic activity of specific sorghum phenolic extracts is a likely mechanism for colon cancer prevention. Further studies are needed to assess physiologically relevant dietary level of sorghum phenolics for prevention of colon cancer, and effect of food processing on the
activity and bioavailability of the chemopreventive components.

▼ The first objective of this study was to investigate the removal efficiency of the endocrine disrupting chemicals(EDCs), including phenolic compounds such as nonylphenol(NP), octylphenol(OP) and bisphenol A(BPA), and natural and synthetic chemicals such as estrone(E1), 17β-estradiol(E2), estriol(E3), diethylstilbestrol(DES) and 17α-ethinylestradiol(EE2) of wastewater treated by the constructed wetland from the Wan-Dan embankment. The second objective was to assess the
estrogenic activity with different treatment units using the T47D-KBluc bioassay.
According to the T47D-KBluc assay, the estrogenicities in influent(In), grit chamber(GC), pre-aeration tank (AT), the contact aeration tank(CAT), aquatic plants area (PA) and effluent(Eff) were ranged from 1.18 to 30.4, 0.85 to 27.74, 0.46 to 3.32, 0.53 to 6.48, 0.48 to 13.44 and 0.52 to 21.16ng 17β-estradiol(E2) equivalents/L. The results of the different flows from the constructed wetland, 100CMD, 200CMD, 350CMD and 500CMD showed the mean removal efficiency of
estrogenic activity, 64%, 42%, 36% and 31%, respectively.
In chemical analysis, the samples were pre-concentrated by solid phase extraction, and analyzed for target compounds by LC-MS/MS method. According to the results, estrone, estradiol, ethynyl estradiol and diethylstilbestrol were not detected throughout the study period. The detection frequency of estriol, nonylphenol, octylphenol and bisphenol A were approximately 16%, 3%, 21% and 1%, respectively. The removal efficiency of estriol and octylphenol, showed was larger than 99% and 37%,respectively. Nonylphenol canât be removed in the system. Bisphenol A detected only in one sample located in the aquatic plants area.
Comparison of to the chemical analysis and bioassay, the concentration of
estrogenic activity ranged from ND to 0.46ng / L and 0.48 to 30.3ng / L, respectively. Bioassay could be used to detect known and unknown
estrogenic compounds, as a rapid and effective detection method.

▼ The aims of this study were to use the heterologously expressed laccase in Pichia pastoris as oxidative biocatalysts for the degradation of BPA and to find out natural mediators that could assist this laccase to degrade BPA efficiently and environmental friendly. In addition, removal of the estrogenic activity of BPA by the white rot fungus Physisporinus rivulosus cultures and the role of its laccase 2 in the BPA disapperance was also focused.
Firstly, the removal of BPA’s estrogenic activity by the white rot fungus Physisporinus rivulosus was confirmed in cultures both with and without the fungal mycelium. Next, the recombinant laccase 2 (rLac2) played a role in the BPA disapperance. At the similar laccase activity level, removal of BPA’s estrogenic activity was done more efficiently in the fungal cultures than in the cell-free enzymatic treatments. Metabolites present in the fungal cultures could possibly act as natural mediators that enhance the removal of BPA. In addition, combination of nine laccases present in the fungal cultures could possibibly enhance the degradation of BPA. Finally, none of the ten mediatiors used was found to act as an efficient rLac2 enhancer in degrading BPA.
Degradation of BPA was followed using a bioreporter system. The yeast Saccharomyces cerevisiae has been genetically modified in order to express the estrogen receptor alpha and produce a bioluminescent signal upon contact with estrogenic substances such as BPA. This bioreporter system has been further developed to specifically detect the estrogenic activity of bisphenol A and to be used in a high-throughput manner.

▼ Bisphenol A used in the production of polycarbonate plastics and epoxy resins is ubiquitous in the environment. The raw material is released to the environment during the manufacturing process and by leaching from consumer products. Recent studies are suggesting that low-dose amounts of Bisphenol A may have adverse health effects on humans. The possibility of removing Bisphenol A from natural water sources or from solvents used to extract the material from consumer products before they enter the market has been studied. The use of model compounds and related substances (4-isopropylphenol, 4-(t-butyl) phenol, and nitrophenols) have been used to study their removal from aqueous solutions using column chromatography and Octolig®, a commercially available material with polyethylenediamine moieties covalently bonded to high-surface area silica gel. The experimental results suggest that 2-nitrophenol and 4-nitrophenol can be successfully removed while 3-nitrophenol, 4-isopropylphenol, and 4-(t-butyl) phenol did not yield a high percent removal. A look at the pKa of the compounds provides an interesting explanation of the results. It is suggested that the compounds with a pKa of approximately 8.3 or higher would require the solution to be at a high pH for anion formation. The resulting pH of the solution would simultaneously deprotonate the ethylenediamine moieties of Octolig® rendering it incapable of removing the anions by ionic interaction.

▼ Nas últimas décadas, a ocorrência de atividade estrogênica e interferentes endócrinos (IEs) no ambiente aquático têm se tornado uma crescente preocupação. Dentre as diversas substâncias classificadas como IEs, destacam-se os fármacos, produtos de higiene e cuidados pessoais, hormônios naturais e sintéticos, produtos químicos industriais, praguicidas e muitos outros compostos que atingem o ambiente aquático por meio de descargas de esgoto doméstico, industrial ou de escoamento agrícola. Os objetivos deste estudo foram determinar a atividade estrogênica em amostras de águas superficiais, e avaliar seus efeitos biológicos no desenvolvimento de embriões de Danio rerio, a fim de propor faixas baseadas em valores de desencadeamento de efeitos para categorizar a atividade estrogênica. As amostras ambientais também foram analisadas por cromatografia líquida acoplada com a espectrometria de massas para identificar as substâncias que são suspeitas de causar
alteração endócrina. Os compostos analisados foram: praguicidas, hormônios, triclosan, bisfenol A, octilfenol, nonilfenol, e a cafeína como indicador de atividade antrópica. A atividade estrogênica foi medida pelo ensaio Bioluminescent Yeast Estrogen (BLYES), que fornece os resultados em equivalente de 17β-estradiol (EEQ). No entanto, este ensaio não é capaz de prover informações sobre os efeitos adversos em organismos aquáticos. Para observação de possíveis efeitos na biota, os embriões foram expostos a amostras de águas superficiais com resultados acima de 0,1 EEQ no BLYES. Os ensaios foram realizados de acordo com a OECD No. 236 (2013), verificando efeitos agudos como: ausência de batimento cardíaco, não formação de somitos, não desprendimento da cauda, e embrião coagulado. Malformações embrionárias tais como: redução do tamanho do organismo, edema cardíaco e vitelínico, curvatura da coluna vertebral, também foram avaliadas. As informações obtidas pelo ensaio com embriões
de Danio rerio foram adequadas para mostrar os efeitos da mistura de contaminantes em organismos não-alvo. A atividade estrogênica medida pelo BLYES ficou abaixo do limite de quantificação (0,1 EEQ) em 44,8% do total de 116 amostras analisadas, e a faixa de atividade estrogênica variou de 0,11 a 14,6 EEQ. Além disso, a presença de contaminantes mesmo que em concentrações baixas ressalta a necessidade de mais estudos para entender os efeitos dessas substâncias nos organismos aquáticos.

▼ Les perturbateurs endocriniens (PE) posent des risques pour la santé Humaine et pour la faune. L’utilisation de tests biologiques basés sur des mécanismes d’action spécifiques permet de caractériser le potentiel PE des substances chimiques dans le cadre de l'évaluation toxicologique, mais les capacités de biotransformation de ces modèles sont rarement prises en compte. Or, le métabolisme conditionne le devenir des xénobiotiques (détoxication vs. bioactivation) et donc in fine l'activité biologique mesurée. Dans ce travail, le devenir de deux contaminants œstrogéniques émergents (benzophénone-2, bisphénol S) a été comparé dans de nouveaux modèles in vitro et in vivo de poisson zèbre (cellules ZELH-zfERs, larve transgénique cyp19a1b-GFP) et des lignées humaines. Nos résultats démontrent que les modèles de poissons zèbres sont métaboliquement compétents et soulignent leur pertinence dans une approche intégrée in vivo/in vitro pour le criblage de l'activité PE des substances chimiques.
Endocrine disruptors (ED) are potentially harmful for Human beings and for wildlife species. Species-specific mechanism-based screening tests nowadays allow characterizing the ED potency of chemicals in the context of their toxicological assessment. However, the biotransformation capabilities of the biological models used are seldom taken into consideration, despite they ultimately condition the fate of the tested xenobiotics (detoxification vs. bioactivation), and, consequently, the biological response. In this work, we examined the fate of two emerging ED (benzophenone-2, bisphenol S) using novel in vitro and in vivo zebrafish models (ZELH-zfERs cells and cyp19a1b-GFP transgenic larvae) as well as human cell lines. Our results demonstrate that the zebrafish models are metabolically competent, and underline their relevance, accuracy and usefulness in an integrated in vivo/in vitro screening approach designed to assess ED's biological activity

▼ L’objectif de ce travail de thèse est d’évaluer le potentiel de nouveaux bio-essais in vitro et in vivo basés sur le poisson zèbre pour la biosurveillance de la contamination chimique de l’eau par les xeno-estrogènes. Pour cela, les bio-essais ont été appliqués pour évaluer les effets de polluants aquatiques environnementaux seuls, mais aussi en mélanges simples (reconstitués) et complexes (échantillons environnementaux). L’évaluation d’échantillons d’eau à travers les bioessais in vitro humain (MELN) et poisson zèbre (ZELH-zfERβ2) a montré des différences qualitatives et quantitatives de réponse, non expliquées par les molécules estrogéniques détectées. Afin de mieux comprendre ces différences, l’activité de polluants aquatiques a été caractérisée sur les différents modèles, individuellement et au sein de mélanges de 2 à 12 polluants, combinant molécules estrogéniques et non estrogéniques pour simuler des situations environnementales.Les résultats obtenus montrent que les bio-essais basés sur le poisson zèbre répondent différemment, et parfois de manière opposée, au modèle humain MELN aux mélanges reconstitués. Ces différences s’expliquent par des sensibilités différentes à certaines molécules entrainant des interactions avec la réponse aux xeno-oestrogènes spécifiques à chaque modèle biologique. Dans son ensemble, ce travail montre que les particularités biologiques de chaque bioessai peuvent influencer la réponse des xeno-estrogènes quand présents au sein de mélange avec d’autres polluants. Ces résultats sont discutés au regard de l’utilisation de bio-essais dans l’évaluation de la contamination chimique des masses d’eau.
This PhD thesis aims at assessing the potential of innovative in vitro and in vivo zebrafish based bioassays for biomonitoring of surface water contamination by xeno-estrogens. For this purpose, the bioassays were applied to assess the effect of environmentally relevant surface water pollutants, alone and in simple (artificial) and complex (environmental samples) mixtures. The screening of surface water samples in zebrafish- (ZELH-zfERβ2 cells) and human-based (MELNcells) bioassays revealed qualitative and quantitative differences which could not be entirely explained by the xeno-estrogens identified. To better understand the response of bioassays to complex environmental mixtures, the activity of environmentally relevant surface waterpollutants was characterized across the bioassays, alone and in 2 to 12-component mixtures combining estrogenic and non-estrogenic chemicals to simulate an environmental contamination.The results indicate that zebrafish-based bioassays have a different and even in some cases an opposite response to the simple mixtures compared with the human-based bioassay MELN. These differences are explained by different sensitivity to some pollutants leading to bioassay-specific interactions with estrogen receptor activation. ▼ Muitos oxidantes químicos reativos acarretam na ruptura de estruturas moleculares complexas de vários tipos de compostos orgânicos decompondo-as em estruturas mais simples e propiciando condições melhores para uma efetiva ação de micro-organismos na degradação biológica. A presença de hormônios, entre eles o 17β-estradiol, em estações de tratamento de esgoto e em águas subterrâneas e superficiais mostra a necessidade de uma avaliação dos processos de tratamento convencionais. O objetivo deste trabalho foi a análise e remoção de hormônios através de técnicas de cloração e ozonização de amostras reais de águas de saída de filtro de estações de tratamento de água (ETAs), operadas pelo Serviço Autônomo de Água e Esgoto (SAAE) de São Carlos (ETA Centro), que capta águas dos ribeirão Feijão e córrego Espraiado e pela Sociedade de Abastecimento de Água e Saneamento S/A (SANASA) de Campinas (ETAs 3 e 4), que capta águas do rio Atibaia. Foram
realizados ensaios contaminando-se as amostras com 17β-estradiol, que é o hormônio natural mais presente no meio ambiente, em concentração de 6.000 ng L-1, submetendo-se tratamento com dosagens em torno de 0,5 e 2,0 mg L-1 desses oxidantes, em tempos de contato de, respectivamente, 10 e 30 min. As amostras submetidas à contaminação e tratamento e as de controle (sem contaminação e tratamento) foram analisadas através da remoção do 17β-estradiol com a verificação da atividade estrogênica das amostras por ensaios de Sistema de Expressão de Estrogênio Induzida por Levedura Bioluminescente (BLYES), que apresentou-se como uma ferramenta simples. Os resultados apresentados neste trabalho demonstram que a oxidação por ozônio se mostrou mais eficiente do que aquela por cloro para a remoção da atividade estrogênica causada, única e exclusivamente, pelo 17β-estradiol para uma dosagem inicial desse hormônio relativamente alta (6.000 ng L-1). Todavia, em todos os
ensaios a concentração final da atividade estrogênica permaneceu acima do limite de quantificação desses hormônio, indicando que a remoção não foi completa, mesmo em condições favoráveis, isto é, matriz limpa, com padrões de potabilidade para os parâmetros físico-químicos.
Many reactive chemical oxidants cause the disruption of the complex molecular structures of various types of organic compounds decomposing them into simpler structures and providing better conditions for effective action of micro-organisms in the biological degradation. ▼ Estrogenic contamination of waterways is of world-wide concern due to the adverse effects observed in aquatic biota. Recently, wastes from agricultural activities have been
identified as likely sources of steroid estrogens released into the environment. Wastes from dairying activities are of particular concern in New Zealand. This project included development of analytical methods to measure free and conjugated estrogens,
measurement of estrogens from the source to receiving environments and an investigation of effluent treatment technologies.
The analytical method developed in this study was based on GC-MS measurement of free estrogens (17α-estradiol (17α-E2), 17β-estradiol (17β-E2) and estrone (E1)) and LC-IT-MS measurement of their sulfate-conjugates (17α-E2-3S, 17β-3S, E1-3S) in raw and treated farm dairy shed effluents (DSE). Effluents from farms in the Canterbury and Waikato Regions, two regions where dairy farming is the dominant land-use, were collected and analysed. All effluents demonstrated high concentrations of steroid estrogens, particularly 17α-E2 (median 760 ng/L). Estrogenic activity was also elevated, at up to 500 ng/L 17β-E2 equivalents using the E-Screen, an in vitro cell proliferation bioassay. Comparison to the chemical data indicated that for most samples, the highest proportion of estrogenic activity was derived from steroid estrogens naturally excreted by dairy cows. Conjugated estrogens were measured in several raw effluent samples, at similar concentrations to those of free estrogens, particularly E1.
Dairy effluent treatment systems reduced free estrogen concentrations by 63-99% and reduced estrogenic activity by up to 89%. In spite of high removal efficiencies, estrogens remained elevated in the treated effluents that are discharged into waterways. Steroid
estrogens and estrogenic activity were detected in streams and groundwater in areas impacted by dairy farming. Although concentrations were generally low, in two streams the concentrations were above levels regarded as safe for aquatic biota (<1 ng/L). The results demonstrate that dairy effluents are indeed a major source of estrogens to the environment and to waterways.

O presente trabalho teve como objetivo estudar quatro espécies relacionadas no Projeto BIOTAFAPESP, as quais têm se destacado quanto às atividades farmacológicas: Byrsonima fagifolia Niedenzu, Byrsonima crassa Niedenzu, Indigofera truxillensis Kunth e Indigofera suffruticosa Miller. Foram realizados ensaios de mutação gênica reversa utilizando-se as linhagens TA98, TA100, TA97a e TA102 de Salmonella typhimurium, com ausência e presença do sistema metabólico de ativação. Foram avaliados extratos, frações e algumas substâncias isoladas, a saber: a)
B. crassa: amentoflavona, substância isolada do extrato metanólico; b) B. fagifolia: extrato metanólico e clorofórmico, frações acetato e aquosa, ácido gálico, galato de metila, ácido quínico e o ácido 3,4- digaloilquínico; c) I. truxillensis e I.suffruticosa: extrato metanólico e clorofórmico, frações de alcalóides, flavonóides e de glicerolipídeos, além das substâncias índigo, indirubina e o kaempferol- 3,7-diraminosídeo. Foi também avaliada a isatina, molécula precursora dos alcalóides índigo e indirubina. As atividades anti-bacteriana, anti-Leishmania, citotóxica e fitoestrogênica foram avaliadas para substâncias isoladas. Os ensaios de atividade fitoestrogênica foram realizados através do método de e-screen, utilizando-se células tumorais MCF7. Os ensaios de atividade citotóxica (células MCF-7) foram realizados pela técnica de sulforadamina-B. Os ensaios anti- Leishmania (L. amazonensis) também foram relizados com as técnicas de MTT (sal de tetrazólio) e contagem manual em

▼ The presence of biologically-active
estrogenic endocrine disrupting compounds (EDCs) in treated effluents from biological wastewater treatment facilities has prompted wide-spread interest in the behavior of these contaminants during the activated sludge process. The yeast-estrogen screen (YES) was used to quantify the
estrogenic activity of samples taken from different areas of three wastewater treatment facilities. An
estrogenic mass-balance around these facilities revealed that the majority of influent
estrogenic activity was removed in the activated sludge process, but the main route for EDC discharge to the natural environment was via the treated effluent. The
estrogenic activity in the effluent from a membrane bioreactor (MBR) was lower compared to a fully aerobic activated sludge process using secondary clarification, suggesting that enhanced removal of particulate and colloidal material may improve EDC removal efficiency.
Colloidal material was obtained from settled mixed liquor suspended solids (MLSS) collected from a pilot MBR and a full-scale activated sludge process that included anoxic and aerobic zones. The MLSS was sized fractionated by filtration, and used to quantify the sorption coefficients for pyrene, 17β-estradiol (E2), and 17α-ethinylestradiol (EE2) by fluorescence quenching. The MLSS-derived colloidal organic carbon (COC) sorption coefficient (Kcoc) for pyrene ranged from (< 1 to 80) L/kgcoc, indicating a similar affinity for pyrene compared to natural organic matter. Kcoc coefficients for E2 ranged between (< 1 to 158) L/kgcoc for E2 and (< 1 to 228) L/kgcoc for EE2, and are the highest E2 and EE2 sorption coefficients reported in the literature to date. There was a strong correlation between the Kcoc coefficients and molar extinction coefficient at 280 nm (e280) for pyrene and E2, suggesting that the interaction of the π;-electrons is an important factor in determining overall sorption behavior. There was no such correlation for EE2. Based on the Kcoc coefficients and COC concentrations of the samples, between 1 and 50% of the aqueous E2 and EE2 concentrations were associated with colloidal material.
In a novel application of the YES bioassay, the bioavailability of colloid-associated E2 was quantified by comparing the EC50 values of the dose-response curves generated in the presence and absence of size fractionated COC. An increase in EC50 values as a function of COC concentration was attributed to a reduction in bioavailability of E2, suggesting that MLSS-derived COC can reduce, but not eliminate, the biological impact of EDCs. However, there was a high degree of variability in the EC50 values, and estimates of the colloid-associated E2 fraction based on the Kcoc-e280 correlation were unsuccessful in accurately predicting increases in EC50 values. Nevertheless, the YES bioassay may represent a powerful tool in determining the bioavailability of EDCs in complex environmental samples.

▼ A number of widely used industrial chemicals have been shown to possess endocrine-disrupting properties. In this thesis, a series of in vitro tests, and an in vivo reproductive performance test with fathead minnows, were used to clarify the extent of estrogenic activity exhibited by the phthalate esters - a class of compound hitherto referred to as 'estrogenic'. Using a recombinant yeast estrogen screen, I demonstrated that a small number of commercially available phthalates showed extremely weak estrogenic activity. The most potently estrogenic phthalate of those tested was BBP, which was approximately one million-fold less potent than 17B-estradiol. The phthalates which were estrogenic in the yeast screen were also mitogenic on estrogen-responsive human breast cancer cells (MCF-7 and ZR-75 cell lines). The most prolifically used phthalate, DEHP, was not estrogenic in any of these assays. The small number of metabolites of phthalate metabolites tested (including MBuP, MBzP, MEHP and MnOP) were also not estrogenic in the recombinant yeast assay. The ability of BBP (as the most potently estrogenic phthalate in vitro) to induce a vitellogenic response (an indicator of estrogen exposure) in fathead minnows (Pimephales promelas) exposed via the water was assessed. No induction of vitellogenin was observed, indicating that 100 Jlg BBP/L (a concentration higher than would normally be found in the environment) is not estrogenic to this species of fish under the conditions employed for this experiment. In the same study, fecundity of breeding pairs of fathead minnows was assessed; exposure to BBP was not found to affect reproductive performance in these fish. A possible alternative mechanism of action of the way in which the phthalates induce frequently reported reproductive disorders was observed. Some of the phthalates, and, notably, some of their metabolites, were demonstrated to act as anti-androgens in a recombinant yeast androgen assay. 4-Nonylphenol is another industrial chemical which is used in large volumes, and due to the nature of its use (mainly in detergent formulations), is discharged into water systems via sewage effluents. This chemical has been shown to be estrogenic to fish at the concentrations at which it has been detected in the environment. 4-Nonylphenol was tested for its ability to affect plasma and pituitary gonadotropin levels in female recrudescing rainbow trout (Oncorhynchus mykiss). Plasma and pituitary levels of FSH were suppressed in fish exposed to 10 and 100 Jlg 4-NP/L. In addition, FSH gene expression was reduced in these fish, and also in the fish exposed to 1 Jlg 4-NP/L. Pituitary LH content and gene expression of this hormone were suppressed in the fish exposed to 100-, and 10- and 100 Jlg 4-NP/L respectively. Gonadal development in vertebrates is regulated by FSH. Ovarian development ceased in the fish exposed to 100 JAg 4-NP/L, possibly as a result of the suppression of FSH synthesis and/or release in these fish.

Spektrofotometrijskim metodama ispitan je antioksidantni potencijal ekstrakata i etarskih ulja. Hepatoprotektivni efekat određen je na homogenatu i hemolizatu jetre laboratorijskih miševa. Budući da ova biljka ima visok sadržaj izoflavona, određena je estrogena, antiestrogena aktivnost, kao i citotoksičnost ekstrakata. Antimikrobna aktivnosta ekstrakata i etarskih ulja ispitana je na šest bakterijskih sojeva. Ekstrakti i etarska ulja su bili umereno aktivni u pogledu bioloških aktivnosti, osim u slučaju antimikrobne aktivnosti koju nije pokazao nijedan ispitivani uzorak.
The chemical characterization of <em>T. pratense</em> extracts was determined by LC-MS-MS, and the essential oils of the GC-MS analysis, wherein the various bioactive compounds were identified. Antioxidant potential of extracts and essential oils was tested by spectrophotometric methods. Hepatoprotective effect was determined in haemolysate and liver homogenate of laboratory mice. Estrogen, antiestrogen activity, as well as the cytotoxicity of the extracts were determined due to the high content of isoflavones. Antimicrobial activity of extracts and essential oils was investigatied on six bacterial strains. Essential oils and extracts were moderately active in terms of biological activity, except in the case of antimicrobial activity where none of the extracts were active against tested bacterial strains.

▼ Endocrine disrupting chemicals (EDCs) are ubiquitous
in the environment and their
presence in water bodies is
documented. They discharge into surface water (SW)
unmonitored,
posing a threat to both aquatic and terrestrial lives. This is a
challenge
as not all populations have access to treated drinking
water (TDW). The EDC
contaminated serves as a route of exposure,
together with ineffective treatment
plants. Given the complexity
of the endocrine system, EDCs may mimic or
antagonise natural
hormones or disrupt their synthesis, metabolism and excretion.
The
associated health effects include testicular dysgenesis syndrome,
metabolic
disorders and cancers. Policy and internationally
standardised test methods are
however sti ll limited. This study
therefore aimed to assess the suitability of two
assays used for
screening
estrogenic activity and one for androgenic
activity in
different water sources.
The study consisted of two phases. In
phase 1, water sample (tap, surface and
treated wastewater) were
collected from a catchment area in Pretoria. The samples
and a
spiked MilliQ laboratory water sample were extracted with solid
phase
extraction (SPE) and sent to Germany for distribution to
participating laboratories.
Samples (n=24) from six different
countries were received to test for androgenic
activity in the
MDA-kb2 reporter gene assay. In phase 2, SW and TDW samples
were
collected from April 2015 until March 2016. The samples were
filtered,
extracted using SPE and assayed with the YES assay,
T47D-KBluc reporter gene
assay for
estrogenic activity and MDA-kb2
reporter gene assay for androgenic
activity. In phase 1,
androgenic
activity was detected in 4 out of 24 (21%) samples and
ranged from 0.23 ± 0.040 ng/L to 0.008 ± 0.001 ng/L DHTEqs. In
phase 2,
estrogenic activity was detected in 16 out of 24 (67%) SW
samples in the T47DKBluc
reporter gene assay and ranged from 0.31
± 0.05 pg/L to 10.51 ± 5.74 pg/L
EEqs. It was below the detection
limit (dl) in the YES assay. Androgenic
activity was
detected in 4
out of 24 (17%) SW samples, ranging from 0.0033 ± 0.0050 ng/L to
0.090 ± 0.040 ng/L DHTEqs. Androgenic and
estrogenic activity was
higher i n pretreatment
samples compared to post-treatment in both
treatment plants. In phase 1, the MDA-kb2 reporter gene assay was
successfully applied to water
samples from different sources.
Androgenic
activity was highest in treated
wastewater. In phase 2,
treatment plants proved to be effective in removing
estrogens
detected in the SW samples, as the TDW samples were below the dl.
Estrogenic activity is within the ranges reported in other studies.
Positive samples
were below the 0.7 ng/L proposed trigger value
for health risk assessments.
Detected androgenic
activity was
lower in TDW samples compared to the SW
samples supplying the two
treatment plants indicating that they were both effective in
removing the androgenic
activity detected. Few studies have
reported androgenic

Some new compounds, 16- and 17-substituted 16,17-secoestratriene derivatives, as well as D-homoestratriene derivatives, were synthesized, starting from 3-benzyloxy-17-hydroxy-16,17-secoestra-1,3,5 (10)-triene-16-nitrile. The newly synthesized compounds were tested for their <em>in vivo </em>estrogenic and antiestrogenic activity, <em>in vitro </em>antiaromatase activity, antioxidative activity by DPPH and TBA tests, as well as antiproliferative activity against MCF-7 ATCC, MDA-MB-231, HT-29 i MRC-5 cell lines.

▼ The aim of the study was triple: (1) To determine whether dehydroepiandrosterone (DHEA) exerts hormonal activity in breast cancer cells (MCF-7), in endometrial adenocarcinoma cells (Ishikawa) as well as in prostate adenocarcinoma cells (LNCaP). We were also interested in the type of the receptor which is being involved in mediating DHEA’s activity. The results indicate that DHEA exhibits estrogenic activity in Ishikawa adenocarcinoma cells and in MCF-7 breast cancer cells while it is unable to induce proliferation in LNCaP prostate cells. DHEA activation of MCF-7 cell proliferation was mediated through ERα and required mainly conversion of DHEA to 5-androstenediol (5-diol). Given that 17β hydroxy-steroid-dehydrogenase type 1 (17β HSD-1), the enzyme that converts DHEA to 5-diol, is overexpressed in breast cancer cells, the use of DHEA as a dietary supplement is contraindicated in high risk subjects for breast cancer. (2) To study whether DHEA can modulate the anticancer activity of a chemopreventive factor (ebenfuran III), which is being uptaken by the diet. The results showed that estradiol, at physiological concentrations, was able to improve the anticancer activity of ebenfuran III in breast cancer cells, while DHEA, even at supraphysiological concentrations, was ineffective in this respect. (3) To study whether DHEA and its metabolites can protect the immortalized hippocampal HT22 cells from oxidative stress-induced cell death. The results showed that only E2 is able to protect the HT22 cells, in pharmacological concentrations, while DHEA is ineffective in this respect. We also showed that estradiol, at physiological concentrations, was able through ERα to improve the neuroprotective potency of a synthetic antioxidant (1,4 benzoxazine).
O σκοπός της διατριβής ήταν τριπλός: (1) Να διευκρινιστεί εάν η δεϋδροεπιανδροστερόνη (DHEA) έχει ορμονική δράση σε καρκινικά κύτταρα μαστού (MCF-7), μήτρας (Ishikawa) και προστάτη (LNCaP) και μέσω ποιου υποδοχέα εξασκεί τη δράση της. Τα αποτελέσματα έδειξαν ότι η DHEA παρουσιάζει οιστρογονική δράση σε καρκινικά κύτταρα μαστού και μήτρας ενώ δεν επάγει τον πολλαπλασιασμό στα κύτταρα αδενοκαρκινώματος του προστάτη. Ειδικά για την επαγωγή του πολλαπλασιασμού στα κύτταρα MCF-7, η δράση αυτή μεσολαβείται από τον υποδοχέα οιστρογόνων τύπου α (ERα) και οφείλεται κυρίως στο μεταβολισμό της DHEA σε 5-ανδροστενοδιόλη (5-diol). Δεδομένου ότι η 17β υδροξυ-στεροειδο-δεϋδρογενάση τύπου 1 (17β HSD-1), που είναι το ένζυμο που μετατρέπει την DHEA σε 5-diol, υπερεκφράζεται στον καρκινικό ιστό, η χρήση της DHEA αντενδείκνυται σε άτομα υψηλού κινδύνου για καρκίνο του μαστού. (2) Να μελετηθεί η δράση της DHEA σε συνδυασμό με ένα χημειοπροστατευτικό παράγοντα (την εμπενφουράνη ΙΙΙ) που προσλαμβάνεται με συγκεκριμένη διατροφή. Τα αποτελέσματα έδειξαν ότι η οιστραδιόλη σε φυσιολογικές συγκεντρώσεις ενισχύει την αντικαρκινική δράση της εμπενφουράνης ΙΙΙ σε καρκινικά κύτταρα του μαστού. Αντίθετα η DHEA, ακόμη και σε πολύ υψηλές συγκεντρώσεις, δεν βελτιώνει την αντικαρκινική δράση της εμπενφουράνης ΙΙΙ.…
