▼ As important enzymes in mitochondrial nucleotide salvage pathway, thymidine kinase 2 (TK2) and deoxyguanosine kinase (dGK) are expressed constitutively in almost all cells. These two enzymes catalyze the initial rate-limiting phosphorylation of pyrimidine and purine deoxynucleosides, respectively, providing DNA precursors for mitochondrial DNA (mtDNA) replication.
Inherited genetic defects in TK2 have been associated with infantile myopathic form of mtDNA depletion syndrome (MDS). In study I, two mutations, R225W and a novel T230A, in TK2 are identified as a new genetic cause of adult-onset autosomal recessive progressive external ophthalmoplegia (arPEO) and the kinetic and structural effects of the two mutations on enzyme function have been characterized.
Nucleoside analogs are widely used in anti-viral and anti-cancer chemotherapy, but they can cause severe side-effect such as mtDNA depletion. In study II, the potential mechanism underlying pyrimidine nucleoside analogs-associated mitochondrial toxicities was investigated, and showed that thymidine analogs had opposite effects on dThd and dCyd phosphorylation and thus can inhibit dThd salvage, leading to imbalanced dTTP and dCTP pools. It was found that the mechanism is most likely due to that TK2 normally exists in an inactive form with bound dTTP.
The redox regulation of TK2 and dGK was studied in study III and IV. The activity of both enzymes was sensitive to the cellular redox status. Under oxidative stress, both TK2 and dGK can be reversibly S-glutathionylated by GSSG. The modification of the conserved Cys189 in TK2 was responsible for a partial inactivation and selective degradation of TK2 in mitochondria, most likely via the AAA⁺ Lon protease. The oxidative effect of nucleoside analogs was also evaluated. Treatment with 3’-azido- 2’,3’-dideoxythymidine (AZT) and 2’,3’-dideoxyinosine (ddI) led to degradation of mitochondrial TK2 and dGK, whereas uridine and guanosine supplementations to AZT respective ddI treatments prevented both proteins from degradation.

▼ Wiele spośród analogów nukleozydów znalazło zastosowanie w medycynie w leczeniu chorób wirusowych i nowotworowych. Wśród nich szczególną grupę stanowią 2’,3’-didehydro-2’,3’-dideoksynukleozydy wykorzystywane w terapii choroby AIDS.
W 2001 Sharpless przedstawił nowatorskie podejście w syntezie organicznej, oparte na obserwacji procesów zachodzących w żywych organizmach – click chemistry. Sztandarowym przykładem reakcji typu ”click” jest 1,
3-dipolarna cykloaddycja Huisgena katalizowana jonami miedzi na +I stopniu utlenienia pomiędzy terminalnymi alkinami i organicznymi azydkami.
W ramach niniejszej pracy doktorskiej zastosowano metody tzw. chemii ”click” do syntezy czterech serii analogów 5’-fosforanów nukleozydów. Jako substraty w katalizowanej jonami miedzi(I) cykloaddycji Huisgena wykorzystano 5’-azydonukleozydy oraz maskowane analogi grup fosforanowych zwierające terminalne wiązanie potrójne (dietylopropargilofosfonianian oraz amidofosforany, pochodne L-aminokwasów). Otrzymano również serię nowych koniugatów nukleozydowo-alkaloidowych. Nukleozydy zwierające terminalne wiązanie potrójne oraz 9-azydoepichinina zostały poddane reakcji cykloaddycji Huisgena w obecności jonów miedzi(I).
Dodatkowo dla wybranych koniugatów nukleozydowo-chininowych oraz analogów 5’-fosforanów nukleozydów wyznaczono wartości parametru IC.50

dariniai, pasižymintys įvairiu biologiniu aktyvumu, pvz., efektyvia
sąveika su opioidiniais receptoriais, veiksmingu citotoksiniu
veikimu ir centrinės nervų sistemos apsauginiu efektu. Neseniai
nustatyta, kad jie dalyvauja triptazės inhibavime, oksitocino
receptoriaus antagonizme ir plazminogeno aktyvatoriaus inhibavime.
1,4-Dipakeistų 2,5-piperazindionų sintezė atlikta, vykdant
4-benziloksianilino acilinimą su chloracto rūgšties chloranhidridu
ir susidariusio produkto ciklizaciją. Vykdant
1,4-diacetil-2,5-piperazindiono aldolinę kondensaciją su
2-chlor-3-chinolinkarbaldehidu susintetintas simetrinis
3,6-di(2’-chlor-3’-chinoliliden)-2,5-piperazindionas. Reakcijos
buvo sekamos plonasluoksnės chromatografijos metodu. Susintetintų
junginių struktūra patvirtinta IR, 1H ir 13C BMR spektrų
duomenimis.
2,5-Piperazinediones are cyclic dipeptide
derivatives, which show a multidute of interesting biological
activities, e.g., efficient interactions with opioid receptors,
potent cytotoxic effects and neuroprotective effects. Recently,
they have also been associated with tryptase inhibition, oxytocin
receptor antagonism and plasminogen activator inhibition. Synthesis
of 1,4-disubstituted 2,5-piperazindione was carried out by
acetylation of 4-benzyloxyaniline with chloroacetyl chloride and
product’s cyclization. Condensation of
1,4-diacetylpiperazine-2,5-dione with
2-chloroquinoline-3-carbaldehyde has been applied to the synthesis
symmetrical 3,6-diylidenepiperazine-2,5-dione. The purity of the
obtained compounds was controlled by thin layer chromatography. The
structure was proved by the data of IR , 1H and 13C NMR spectra
analysis.

▼ Leucemia mielóide aguda (LMA) é uma doença de caráter multifatorial e heterogênea. Alterações somáticas no código genético recorrentes na LMA são causadas principalmente por agentes químicos e físicos nocivos ao DNA, associados a falhas nos sistemas de reparo. Pesquisas realizadas na última década sugerem que metilação aberrante e polimorfismos genéticos em genes supressores tumorais e genes do metabolismo de radicais livres podem estar envolvidos na leucemogênese. O presente estudo teve como objetivo investigar a associação entre polimorfismos nos genes TP53 codon 72, GSTT1, GSTM1 e hipermetilação da região promotora do gene BRCA1 com a leucemia mielóide aguda. Duzentos e vinte e três pacientes adultos diagnosticados com LMA e 202 indivíduos adultos são (controles) foram genotipados para polimorfismos no TP53, GSTT1 e GSTM1 e 55 pacientes com AML de novo foram avaliados para metilação no BRCA1. As frequências observadas para o TP53 foram: 33,6% ARG/ARG, 44,4% ARG/PRO e 22% PRO/PRO nos pacientes e 38,
3% ARG/ARG, 48,5 ARG/PRO e 12,8% PRO/PRO nos controles. O alelo PRO foi associado à susceptibilidade ao desenvolvimento de LMA no modelo genético recessivo (p=0.0198), no co-dominante (p=0.0291) e no alélico (p=0.0438). O polimorfismo do GSTT1 foi observado em 23% dos pacientes e em 27% dos controles (p=0,297). O polimorfismo do GSTM1 foi observado em 45% dos pacientes e em 43% dos controles (p=0,573) e a co-ocorrência dos dois polimorfismos foi observada em 12% dos casos e 8,5% dos controles (p=0.286). Nenhuma associação foi encontrada entre os polimorfismos nos genes GSTs e LMA. O gene BRCA1 não foi encontrado hipermetilado em nenhum dos 55 pacientes analisados. Os resultados do presente estudo apontam que o polimorfismo do TP53 parece favorecer a leucemogênese. Em contrapartida, os polimorfismos no GSTT1, GSTM1 e metilação do BRCA1 parecem não ter relevância na leucemia mielóide aguda.

▼ Pseudomonas putida F1 is unable to grow on styrene, a common industrial pollutant, despite degrading it through the toluene degradation (tod) pathway. This dissertation investigates the biochemical and genetic aspects of styrene degradation by P. putida F1 and identifies substrate-level catechol-
2,
3-dioxygenase (C23O) inactivation as the factor which prevents growth on styrene. Novel adaptations to growth on styrene were investigated and found to mitigate C23O inactivation through management of
3-vinylcatechol production and consumption. Previous studies of P. putida F1 identified TodF, a hydrolase responsible for the degradation of styrene meta-fission product (MFP), as putatively responsible for F1's inability to grow on styrene. Through kinetic and growth analyses, we demonstrated that TodF is capable of styrene MFP degradation and thus not responsible for prohibiting growth on styrene. Preliminary genetic analysis of styrene adapted mutants strengthened this conclusion. It was found that cultures of F1 exposed to styrene accumulated 3vinylcatechol in the media, suggestive of inhibited C23O activity. Specifically, micromolar concentrations of
3-vinylcatechol were found to inactivate TodE during catalysis. Analysis of cells growing on styrene suggested that inactivation of TodE and the subsequent accumulation of
3-vinylcatechol resulted in toxicity and cell death. Over-expression of TodE or an inactivation resistant C23O on a plasmid was able to prevent catechol accumulation and confer the ability to grow on styrene. We characterized a spontaneous F1 mutant, designated SF1, which was capable of growth on styrene and did not accumulate
3-vinylcatechol. Resting cell assays demonstrated that the activity of toluene dioxygenase (TDO), the multicomponent enzyme responsible for the production of
3-vinylcatechol from styrene, was reduced in SF1. DNA sequence analysis of the tod operon revealed a single base pair mutation in todA (C479T), a gene encoding the reductase component of TDO. Replacement of the wild-type todA allele in F1 with todAC479T from SF1 reduced TDO activity, prevented
3-vinylcatechol accumulation, and conferred the ability to grow on styrene. Collectively, our data reveals a unique adaptation where slowing down the rate of vinylcatechol production via decreased TDO activity leads to reduced C23O inactivation and permits growth on styrene.

Partial least squares regression was used to model the concentration of IBMP in berries at 50 days after anthesis (DAA, accumulation) and log-fold decrease in IBMP concentration from 50 DAA to harvest (degradation) as a function o f viticultural and environmental data collected from multiple Cabernet franc vineyards (10 in 2008 and 8 in 2009). The most important predictors for modeling IBMP accumulation were factors associated with vine vigor, and higher vigor was positively correlated with IBMP accumulation. IBMP degradation could not be satisfactorily modeled across multiple sites, but within sites, factors associated with vine vigor, crop to vine size, and fruit maturity were important predictors. In the warmer growing season (2008), IBMP concentrations at 50 DAA (range 2008 = 103 to 239 pg/g; range 2009 = 12 to 87 pg/g) were significantly higher than 2009 at all 8 sites. IBMP degradation was less in the cooler growing season, but harvest concentrations (range 2008 = 1 to 13 pg/g; range 2009 5 to 14 pg/g) were not significantly different between years at 5 out of 8 sites as a result of lower preveraison accumulation. Basal leaf removal treatments imposed preveraison reduced IBMP concentrations in Cabernet franc (34 to 88%) and Merlo t (38 to 52%) berries at harvest, but postveraison treatments were not efficacious. Shoot tipping and chlormequat treatments applied to Cabernet franc vines during the preveraison period altered vine growth and canopy density, but did not affect IBMP concentrations in berries at harvest. Clonal selection was evaluated as a potential tool to manage IBMP, but the Cabernet franc clones under study (1, 214, 312, 327) did not possess distinct characteristics that consistently resulted in differential vine growth and IBMP concentrations. These experiments suggest that high IBMP concentrations are likely to occur in vigorous sites where high preveraison temperatures are followed by poor ripening conditions. Preveraison basal leaf removal and managing for vine ba lance are potential strategies to control IBMP levels.

▼ Sorghum contains many phenolic compounds which have potential antioxidant, anti-inflammatory, and chemopreventive properties as well as natural colorants in foods. Phenolic compounds of stalks, sheaths, leaves, glumes and grains from tan (ARTx631/RTx436), red (Tx2911) and purple (Tx3362) sorghum plants were characterized by UPLC-MS/MS. Antiproliferative properties of selected sorghum extracts were evaluated using HT-29 colon cancer cells and absorption of their polyphenolics was determined by a Caco-
2 in vitro model system.
Phenolic acids, flavones,
3-deoxyanthocyanidins and chalcones were found in all plant components. Phenolic acids were predominant in the stalks, sheaths and grains of all sorghum types. Flavone glycosides were predominant in leaves, sheaths and stalks while flavone aglycones in glumes and grains.
3-Deoxyanthocyanidins and chalcones were mostly found in sheaths, leaves, glumes and grains of Tx2911 (red) and Tx3362 (purple) genotypes. Sorghum leaves showed high levels of flavone glycosides while glumes had high levels of flavones aglycones.
Glume extract of ATx631/RTx436 (tan) and Tx2911 genotypes had the strongest antiproliferation activity (IC_(50) = 85-178 µg/ml), these extracts had also the highest levels of flavone aglycones (19.6-49.8 mg/g). Absorption of flavones (30.4-42.

3-deoxyantocyanidins (1.4-11.
3%), while absorption of methoxylated
3-deoxyanthocyanidins (11.
3%) was higher than non-methoxylated
3-deoxyanthocyanidins (1.4 – 1.6 %). Flavones had high absorption compared to other sorghum phenolics suggesting that sorghum flavone aglycones are more bioavailable than other sorghum phenolic compounds. Consequently, sorghum glumes could be used as used as a source of phytochemicals to increase value of sorghum crop.

▼ The design of supramolecular systems with controlled arrangement of chromophores, electron/energy donors and acceptors remains a challenge in artificial photosynthesis. Biological systems have been known to accomplish the intricacy presented simply by self-assembly. With this inspiration, biomimetic structures that utilize self-assembly to tether donors and acceptors together have been developed and studied. Using oligopeptides as the donor-acceptor bridging linkers and metal-ligand interactions as the only driving force for self-assembly, a variety of compounds with unique photophysical properties could be synthesized. Study of these compounds would provide useful information to fine tune the structures to create long-lived charge separated state and promote efficient solar energy conversion. This thesis describes the synthesis, characterization and application of a series of oligopeptide derivatized [Ru(bpy)
3]
2+ complexes for artificial photosynthesis. Factors affecting photophysical properties are investigated and the first example of using this motif to photocatalyze chemical reactions is thoroughly studied.
A series of [Ru(bpy)
3]
2+ compounds linked to 1-
3 Pd2+ by oligoaminoethylglycine(aeg) were synthesized for use in photocatalytically dimerizing α-methylstyrene . This is the first example using the aeg chain between a photosensitizer and a reaction center. The products had faster reaction rates and better selectivity than conjugated linkers described in the literature. Catalytic efficiency when linking [Ru(bpy)
3]
2+ with 1,
2 and
3 Pd2+ centers was also compared and it was found the complex with three Pd centers had the lowest catalytic activity because of lower chemistry stability. The excited state quenching mechanism is likely to be an electron transfer process based on solvent study, variable temperature study and control experiments using added sacrificial electron donor.
A series of [Ru(bpy)32+] hairpin structures were designed to study impact of side chain change of different oligopeptide linkers (e.g. aminoethyglycine (gly), aminoethyvaline (val), aminoethyleucine (leu) and aminoethyphenylalanine (phe) ) on the emission photodynamics of [Ru(bpy)
3]
2+-[M(bpy)
2]2+ complexes (M = Cu, Pd or Zn). For the Ru-Cu and Ru-Pd complexes, the non-radiative decay rate decreases and the energy barrier for non-radiative decay process increases as the steric bulk of the side chain increases. However, no trend was observed for the Ru-Zn complexes. Solvent study showed all the Ru-Cu and Ru-Pd complexes have a negative trend of non-radiative decay rates vs. Pekar factor but no trend was observed for the Ru-Zn complexes, which indicates Ru-Cu and Ru-Pd have an electron transfer non-radiative decay mechanism and is different from the mechanism of Ru-Zn. The impact of side chain change on the non-radiative decay of Ru-Cu and Ru-Pd is probably due to bulky side chains restrict the necessary geometry change that accompanies the electron transfer.

Type 2 diabetes (T2D) is the fastest growing chronic health condition, driven by obesity-related insulin resistance (IR). Obesity accounts for 50% of the total diabetes burden in Australia and worldwide. As the global prevalence of obesity increases, diabetes-related health care costs will continue to rise. Interventions that prevent or delay the onset of diabetes can potentially reduce the overall diabetes burden and associated health care costs. Weight loss is clinically effective in ameliorating IR and reducing T2D risk, however long-term weight loss remains difficult to achieve and maintain, and research into adjunct therapies are warranted. Previous studies reported a sex-dependent association between long-chain omega-3 polyunsaturated fatty acids (LCn-3PUFA) eicosapentaenoic acid (EPA; c20:5n-3) and docosahexaenoic acid (DHA; c22:6n-3) and T2D. Sex hormones are known to regulate synthesis of DHA from α-linolenic acid (ALA; c18:3n-3), though precise mechanisms through which they do so are not well understood. Sex hormones also affect T2D risk differentially in men and women. Interactions between sex hormones and LCn-3PUFA may in part explain some of the differential T2D risk in men and women. Therefore, this thesis aimed to investigate the interactions between sex hormones and LCn-3PUFA for the prevention of obesity-associated IR and subsequent development of T2D. A systematic review and meta-analysis was undertaken to review the existing evidence to determine whether currently available literature supported the prevailing hypothesis, that LCn-3PUFA reduce IR in a sex-dependent manner. The literature was searched to identify randomised controlled trials (RCT) which used a dietary or supplemental n3-PUFA intervention and included measures of IR or insulin sensitivity as an outcome. Thirty-two studies including n=1848 participants were included, with two studies conducted in men, seven in women and the rest in mixed population. Meta-analysis showed that interventions with LCn-3PUFA in females ≥6 weeks duration displayed a reduction in IR, with no change in IR in studies in male populations. However, few intervention studies were conducted in males and significant heterogeneity was present that could not be explained in our analysis, limiting conclusions that could be drawn. Furthermore, there were no studies in which men and women were exposed to an identical intervention protocol, necessitating development of a double-blind RCT. A cross-sectional study of n=2,092 older (55-85 years) Australians assessed relationships between LCn-3PUFA status and diabetes, with sub-group analyses used to investigate sex- and weight- related interactions. There was a significant three-way interaction between sex, weight status and LCn-3PUFA status in the prediction of T2D, therefore sub-group analysis was performed based on this. ▼ Four kinds of new compounds, namely, methyl-3-hydroxy-2-naphthoate, ethyl-3-hydroxy-2-naphthoate, n-propyl-3-hydroxy-2-naphthoate and i-propyl-3-hydroxy-2-naphthoate, have been generated, which were structurally characterized by UV-Vis spectra, IR spectra and photoluminescent spectra. These compounds reveal a one-dimensional (1D) structure, which will be further extended to a three-dimensional (3D) supramolecular framework via a variety of hydrogen bonds, π⋯π interactions and C-H⋯π interactions and so on. The photoluminescent properties of compounds 1-4 were investigated in the solid state at room temperature and meanwhile, in ethanol solution. While the UV-Vis spectra of compounds 1-4 was carried out in ethanol solution.
This paper further studies and analyzes the structures and properties by changing its different substituents.

A vancomicina (van) é empiricamente prescrita para o tratamento de infecções por estafilococos em pacientes submetidos à hemodiálise (HD). O monitoramento das concentrações séricas mínimas (CSM) de van é utilizado como o mais preciso e prático método para se estimar sua eficácia, sendo recomendada a manutenção das CSM entre 10 e 20 µg/mL. O objetivo deste trabalho foi investigar as CSM de van de pacientes sob HD, em um período de dois anos e correlacioná-los com a concentração inibitória mínima (CIM) deste glicopeptídeo para Staphylococcus spp recuperados de bacteremias nestes mesmos indivíduos. As amostras recuperadas (N=80) foram testadas quanto à CIM (diluição em ágar ou caldo) de van, gentamicina (gen), oxacilina (oxa),
tetraciclina (tet), rifampicina (rif), ciprofloxacino (cip), penicilina G (pen) e daptomicina (dap). Foram coletadas 101 amostras de soro de 22 pacientes em regime de administração de 1g a cada 4-5 dias, mais gen (1,5-2,0 mg/Kg) ao final de cada HD. As amostras foram coletadas antes de cada sessão de HD, nos tempos de 0, 48, 72, 96 e 120 horas após a infusão do fármaco. As CSM de van foram obtidas pelo imunoensaio de micropartículas quimioluminescentes (CMIA). Doseamento microbiológico de van foi realizado por meio da técnica pourplate com ágar sais mínimos. S. aureus foi a espécie mais frequentemente isolada (24%), seguida de Staphylococcus spp coagulase negativa (ScoN) (15%). Maiores taxas de resistência ocorreram para oxa (17% e 58%), pen (85% e 84%) e cip (26% e 45%) em S. aures e ScoN, respectivamente. Das amostras de sangue coletadas, 77,8% exibiram concentrações séricas de van <10 µg/mL e considerando apenas as coletadas imediatamente antes da próxima dose de van
(CSM), 90,6% também continham concentrações inferiores àquelas recomendadas para efetiva atividade antiestafilocócica. Os valores de concentrações séricas de van obtidas pelo bioensaio apresentaram ótima correlação com o CMIA (padrão ouro) (R2 = 0,96), com valores estatísticamente similares (p<0,05) quando aplicado o fator de correção. A prevalência de Staphylococcus spp resistente à oxa sustenta o uso empírico de van, contudo, a alta taxa de isolados de S. aureus que exibiram CIM de van de 1 µg/mL (59,5%) associada às baixas CSM de van a que são submetidos a maioria dos pacientes indica chance de falha terapêutica e seleção de cepas resistentes. Assim, uma abordagem terapêutica individualizada e monitoramento da droga devem ser considerados. Para tanto foi validado o ensaio microbiológico que, pelo baixo custo e simplicidade da técnica facilita a implementação do monitoramento de van na rotina de laboratórios clínicos de pequeno porte.

This present research proposes a Brain-Computer Interface (BCI) architecture adapted to motor mental tasks and music imagery. For that purpose the statistical properties of the electroencephalographic signal (EEG) were studied, such as its probability distribution function, stationarity, correlation and signal-to-noise ratio (SNR), in order to obtain a minimal empirical and well-founded parameter system for online classification. Stationarity tests were used to estimate the length of the time windows and a minimum length of 1.28 s was obtained. Four algorithms for artifact reduction were tested: threshold analysis, EEG filtering and two Independent Component Analysis (ICA) algorithms. This analysis concluded that the
algorithm fastICA is suitable for online artifact removal. The feature extraction used the Power Spectral Density (PSD) and three methods were tested for automatic selection of features in order to have a training step independent of the mental task paradigm, with the best performance obtained with the Kullback-Leibler symmetric divergence method. For the classification, the Linear Discriminant Analysis (LDA) was used and a step of reclassification is suggested. A study of four motor mental tasks and a non-motor related mental task is performed by comparing their periodograms, Event-Related desynchronization/synchronization (ERD/ERS) and SNR. The mental tasks are the imagination of either movement of right and left hands, both feet, rotation of a cube and sound imagery. The EEG SNR was estimated by a comparison with the correlation between the ongoing average and the final ERD/ERS curve, in which we concluded that the mental task of sound imagery would need approximately five times
more epochs than the motor-related mental tasks. The ERD/ERS could be measured even for frequencies near 100 Hz, but in absolute amplitudes, the energy variation at 100 Hz was one thousand times smaller than for 10 Hz, which implies that there is a small probability of online detection for BCI applications in high frequency. Thus, most of the usable information for online processing and BCIs corresponds to the α/µ band (low frequency). Finally, the ERD/ERS scalp maps show that the main difference between the sound imagery task and the motor-related mentaltasks is the absence of ERD at the µ band, in the central electrodes, and the presence of ERD at the αband in the temporal and lateral-frontal electrodes, which correspond tothe auditory cortex, the Wernickes area and the Brocas area.

A presente pesquisa teve como objetivo simular e avaliar os impactos de variáveis ambientais (ferro e UV-C) sobre o crescimento e produção de MCY de M.aeruginosa (NPLJ-4). Para tanto, tal cepa foi cultivada sob condições controladas. No ensaio do ferro, não houve diferença significativa de densidade, biovolume e clorofila-a entre as diferentes concentrações de ferro, enquanto que taxa de crescimento, tempo de duplicação e concentração de toxina apresentaram tal diferença. Foram observadas mais divisões celulares (G) a uma menor taxa nas culturas com maior teor de Fe, causando aumento de densidade e biomassa (viceversa). As divisões reduziram-se a uma taxa maior até que o Fe ficasse escasso (10,4, 1 e 0,5 μM, respectivamente).
Culturas com 0,5 μM registraram: maior taxa,menor tempo de duplicação, menor densidade e biovolume. Quanto a toxina, células da fase log (6º ao 14º dia) e estacionária (16º ao 35º dia) influenciaram nos altos valores de MCY-LR total das culturas com Fe. Os teores aumentaram do 10º para o 20º dia e caíram no 30º dia nas culturas com 4 e 10 μM Fe. Ademais, o tratamento com 1 μM Fe obteve maior densidade, biovolume, picos de maior área e maiores concentrações de MCY-LR total em relação as culturas com 0,5, 4, e 10 μM Fe,respectivamente. Assim, o crescimento de MA nem sempre está atrelado aos maiores níveis de Fe e uma única célula pode ser responsável por produção de grande quantidade de toxina. Já no experimento de simulação de exposição à radiação UV-C, obteve-se remoção completa da MCY-LR total em meio ASM-1 com floração de M. aeruginosa, sendo que mais que 50% da toxina foi degradada nas 2 primeiras horas de exposição. Os valores de MCY-LR total,
densidade, biovolume e clorofila-a declinaram à medida que o tempo de exposição à UV-C aumentava. Ademais, não ocorreu produção de microcistina LA e RR em nenhum dos experimentos.

Um mercado florestal cada vez mais competitivo e atrelado às demandas por multiprodutos da madeira favorece o estudo de métodos de otimização que busquem maximizar a receita dos empreendimentos florestais. Comparativamente, pouco se sabe a respeito da eficiência e da eficácia entre os diferentes métodos de solução aplicáveis ao Problema de Otimização do Sortimento Florestal (POSF). A dificuldade em encontrar os sortimentos ótimos é atribuída ao crescimento exponencial do número de padrões de corte a serem analisados em função do número de produtos comercializados e das dimensões dendrométricas dos fustes traçados. É necessário, portanto, alguma técnica que otimize o sistema de busca. Nesse sentido, o objetivo desse trabalho foi
modelar matematicamente o POSF e comparar por meio de três estudos de caso diferentes métodos de solução propostos para resolver o problema. Os dados correspondentes ao estudo de caso 1 constituem-se de 408 árvores de Pinus taeda L. oriundas de Santa Catarina. Os dados do estudo de caso 2 correspondem a 197 árvores de Eucalyptus sp. oriundas do sul da Bahia. Já o estudo de caso 3 é composto por 42.974 árvores de Eucalyptus saligna oriundas do Paraná. Os métodos de otimização implementados em nível de árvore individual no cenário bucking-to-value alvos de análise dos estudos de caso 1 e 2 foram: (1) Programação Dinâmica (PD), (2) estratégia gulosa, (3) busca exaustiva, (4) Heurística de Construção por Partes (HCP) e as meta-heurísticas (5) Greedy Randomized Adaptive Search Procedure (GRASP) e (6) Iterated Local Search (ILS). Os resultados desses algoritmos foram confrontados no estudo de caso 1 com os já conhecidos na literatura do Algoritmo Genético (AG) e do Simulated Annealing
(SA). No estudo de caso 2 o valor de comercialização das toras foi determinado em função de sua classe de qualidade. Quatro classes de qualidade foram definidas segundo a quantidade de nós existentes. Embora a PD seja o método de solução mais indicado para resolver o POSF em nível de árvore individual, a HCP, o GRASP e o ILS alcançaram excelentes resultados chegando, respectivamente, a 99,99%; 99,93% e 99,01% da solução ótima no estudo de caso 1 e a 99,98%; 99,97% e 99,84% da solução ótima no estudo de caso 2. Por outro lado, no cenário bucking-to-demand, alvo de análise do estudo de caso 3, foi avaliado um sistema de otimização multinível implementado por meio dos seguintes métodos de solução híbridos: (1) PD + Programação Linear Inteira (PLI) executada via CPLEX, (2) PD + Heurística de Busca Intensiva (HBI), (3) GRASP + HBI, (4) ILS + HBI e (5) HCP + HBI. Nesse caso, a PD + HBI foi capaz de chegar a 99,77% da solução ótima em menos da metade do tempo gasto pela PD + PLI para obter
a solução exata do problema.

O principal problema encontrado quando da aplicação de técnicas de sistemas de informações geográficas e sensoriamento remoto para a predição de incêndios florestais é a necessidade de integrar diferentes fontes de dados. Os métodos aplicados, geralmente são baseados em técnicas de regressão ou em coeficientes que dependem de conhecimentos dos especialistas. Esta pesquisa objetivou testar a capacidade da árvore de classificação e regressão (CART) em avaliar o risco de incêndios florestais no estado do Espírito Santo. A análise CART é uma técnica estatística não paramétrica que gera regras de decisão na forma de uma árvore binária, para um processo de classificação ou de regressão. O produto MCD45A1 de área de queima, relativo a um período de 16
anos (2000-2015), foi utilizado para, a partir dos pontos centrais da célula de grade, obter um mapa de ocorrência de incêndio por meio de uma abordagem de densidade Kernel. O mapa resultante foi então utilizado como variável de entrada para a análise CART com variáveis de influência de incêndios usados como preditores. Um total de 12 variáveis preditoras foram determinadas de diversas bases de dados, abrangendo aspectos ambientais, físicos e socioeconômicos. As regras induzidas pelo processo de regressão permitiram a definição de diferentes níveis de risco, expressa em 35 unidades de gestão, utilizado para a produção de um mapa de predição de fogo. De acordo com os resultados, as áreas de maiores riscos no estado são representadas pela Região Nordeste, Vale do Rio Doce e Sudeste (Costa Sul). Os resultados do processo de regressão (r=0,94 e r²=0,88), a capacidade de análise do algoritmo CART para destacar as relações hierárquicas entre as variáveis preditoras e a interpretabilidade

A compactação de diferentes materiais lignocelulósicos é uma das alternativas para melhorar os problemas relacionados à produção energética e diminuir o uso de fontes derivadas de combustíveis fósseis. Diante do exposto, teve-se por objetivo avaliar a potencialidade energética de pellets produzidos com diferentes biomassas, tais como madeira de eucalipto de um clone do híbrido de Eucalyptus grandis x Eucalyptus urophylla, bagaço de cana-de-açúcar (Saccharun officinarum L.) e capim-elefante (Pennisetum purpureum Schum). Para a produção dos pellets utilizou-se diferentes composições. A umidade e granulometria das partículas foram determinadas de acordo com as características de cada material. Determinou-se a densidade a granel e
energética; os teores de extrativos totais, lignina total, cinzas e poder calorífico superior das composições e dos pellets. Para os pellets além das análises supracitadas, determinou-se ainda, a durabilidade mecânica, teor de finos, compressão diametral, diâmetro, comprimento e densidade unitária. O processo de peletização das biomassas resultou em aumento do poder calorífico superior, densidade a granel e energética e redução da umidade dos combustíveis produzidos. Os pellets produzidos com duas biomassas obtiveram melhor desempenho do que aqueles produzidos com apenas uma biomassa. De modo geral, o desempenho dos pellets produzidos foi superior ao pellet comercial. Os pellets com 50% de capim-elefante e 50% de madeira de eucalipto se destacaram, com relação ao poder calorífico superior, densidade a granel e energética, durabilidade mecânica e densidade unitária. O uso da água de forma racional tem se tornado cada vez mais importante nos últimos anos, devido à má distribuição das chuvas e ao aumento da demanda por água, como na produção agrícola e florestal. A evapotranspiração é uma importante variável do ciclo hidrológico e uma das principais componentes do balanço hídrico no solo. O uso de equações simplificadas é uma alternativa potencial para estimar a evapotranspiração de referência quando os dados meteorológicos são limitados. O objetivo deste trabalho foi aplicar e testar diferentes métodos para estimar a evapotranspiração de referência (ET0) para o estado do Espírito Santo, com limitação de dados meteorológicos, utilizando o método de Hargreaves e Samani em diferentes condições
meteorológicas, adotando o método de Penman-Monteith FAO-56 como referência. Calibrou-se a ET0 pela equação de Hargreaves e Samani utilizando regressão linear, e ajustou-se o coeficiente de Hargreaves e Samani (CH) através dos métodos de Vanderlinden et al. (2004) e de Martí et al. (2015). Foram realizados diferentes ajustes por regressão linear, considerando-se todas as estações meteorológicas utilizadas neste estudo (ajuste geral), diferentes tipos de clima (ajuste por clima), os períodos seco e chuvoso do ano (ajuste período seco e ajuste período chuvoso), classes de amplitude térmica (ajuste por classes), e o tipo de clima combinado com classes de amplitude térmica (ajuste por clima e classes). Estimou-se, também, para fins de comparação, a ET0 pelos métodos de Hargreaves e Samani original (HS) e Penman-Monteith com limitação de dados climáticos (PML). Em geral, o erro absoluto médio (MAE) dos métodos de HS, PML, Vanderlinden et al. (2004), Martí et al. (2015), ajuste geral,
ajuste por classes, ajuste por clima, ajuste por período e ajuste por clima e classes, foram de 0,68, 1,46, 0,81, 0,77, 0,53, 0,51, 0,51, 0,51 e 0,49 mm dia-1, respectivamente. Nos períodos seco e chuvoso separadamente, os erros (MAE) foram de 0,41 e 0,61 mm dia-1, respectivamente. O ajuste por classes de amplitude térmica proporcionou melhores estimativas da ET0 em dias mais secos, nos quais necessitam-se de melhores estimativas para o manejo de irrigação na agricultura. O método de PML obteve o pior desempenho entre os métodos testados, não sendo recomendado para estimar a evapotranspiração no estado. Os ajustes por regressão linear obtiveram desempenho superior aos ajustes do CH, no qual melhoraram as estimativas da ET0 em até 30%. Com limitação de dados meteorológicos, o método do ajuste geral é considerado como o mais recomendado dentre os métodos testados, devido a sua simplicidade de aplicação. Para estimar a ET0 entre os meses de abril e setembro no estado, recomenda-se
utilizar o ajuste do período seco.

O conhecimento a respeito do crescimento da árvore e da produção florestal, assim como as estimativas dessas variáveis, são essenciais para viabilizar o planejamento, gerenciamento e controle de uma floresta. A modelagem da distribuição de diâmetros é amplamente utilizada para esse fim, que além de permitir a descrição atual do povoamento, permite a projeção da produção e auxiliam na tomada de decisão. Dessa forma, esse trabalho tem como objetivo propor um sistema de projeção da distribuição de diâmetros para povoamentos de Schizolobium parahyba var. amazonicum (Huber ex Ducke) Barneby (paricá) sob diferentes espaçamentos. Os dados utilizados nessa pesquisa foram provenientes de povoamentos homogêneos de paricá,
localizados no estado do Pará, plantados sob os espaçamentos 3x2, 3x3, 3x4, 4x4 e 5x5 m. Ajustou-se as funções Weibull 2 e 3 parâmetros, Weibull 2 parâmetros com parâmetro de locação fixo no DAP mínimo, Weibull 3 parâmetros truncada à direita, Hiperbólica 2 e 3 parâmetros, Log-logística 2 e 3 parâmetros e Logística-generalizada. Após o ajuste das fdps, foi aplicado o teste de Kolmogorov-Smirnov para a avaliação da aderências aos dados, a 5% de significância, assim como a análise gráfica dos dados observados e estimados. As funções avaliadas obtiveram aderência aos dados observados superior a 98 %, e de acordo análise gráfica descreveu com eficiência os dados de campo nos cinco espaçamentos estudados, indicando que todas podem ser empregadas para a modelagem da distribuição de diâmetros de paricá. Na projeção dos diâmetros utilizou-se a função Weibull para a recuperação dos parâmetros por ser flexível e de fácil utilização. Foram ajustados sete modelos de regressão, para cada um dos
espaçamentos. Os modelos foram avaliados por meio do coeficiente de determinação ajustado ( ), viés (V%) e da raiz do quadrado médio do erro (RQME%). O parâmetro de forma não apresentou boa estimativa, principalmente para os espaçamentos 3x4 e 5x5 m. Foi possível obter sistemas de equações para a projeção dos parâmetros das funções em quatro dos espaçamentos estudados, exceto para o espaçamento 5x5 m. Nos casos em que a prevenção não é suficiente para evitar a deflagração de um incêndio florestal, a rápida detecção do mesmo constitui fator decisivo para minimizar os possíveis danos provocados pelo fogo, bem como a redução dos custos associadas ao seu combate. Mediante tal contexto,com esta pesquisa objetivou-sedesenvolvimento de um modelo de risco baseado em banco de dados e avaliar, juntamente com outras três metodologias de riscos de incêndios florestais, a que melhor representa os incêndios florestais em áreas de florestas plantadas na costa centro-norte do estado do Espírito Santo e costa sul da Bahia, contemplando as seguintes etapas metodológicas: a) Determinação das áreas com clima homogêneo; b) Estatística dos incêndios
florestais; c) Cálculo e espacialização dos índices de risco de incêndios florestais: Fire Weather Index (FWI); Fórmula de Monte Alegre Alterada (FMA+) e Risco de Fogo do Instituto Nacional de Pesquisas Espaciais (RF); d) desenvolver um modelo para cálculo do risco de incêndios florestais baseado em banco de dados(RIF-Database); e, f) Escolhados modelos de riscos, a partir dos incêndios florestais catalogados pela empresa FIBRIA Celulose S.A.A metodologia adotada mostrou-se eficiente para o agrupamento de estações meteorológicas em três subzonas climáticas homogêneas para a área de estudo.Os resultados evidenciam que há duas épocas de ocorrência de incêndios florestais na área estudada, sendo que para a primeira época, a subzona 1é de dezembro a março, para a subzona 2 é de janeiro a março, e para a subzona 3 está nos meses de janeiro e fevereiro. A segunda época, para todas as subzonas, é entre os meses de agosto a outubro. O modelo FWI apresentou os melhores resultados para a
subzona 1, o modelo RIF-Database apresentou resultados excelentes, é o recomendado para as subzonas 2 e 3. O modelo FWI é visto como o modelo mais próspero para a área de estudo, uma vez que, encontrou valores superiores, para uma subzona, ao modelo desenvolvido por meio da base de dados da área de estudo, entretanto, será necessário um estudo para a calibração dos seus parâmetros. O paricá (Schizolobium amazonicum Huber ex Ducke) apresenta-se como excelente alternativa para uso em plantios de povoamentos florestais puros, sistemas agroflorestais e projetos de recuperação de áreas degradadas. A madeira é utilizada para vários fins, destacando-se o emprego na fabricação de lâminas e compensados. No entanto, apesar da sua importância econômica e do seu uso potencial, a espécie ainda necessita ser estudada em termos silviculturais. Diante disso, o presente estudo teve como objetivo avaliar as respostas morfológicas, fisiológicas, anatômicas e adaptativas de mudas de Shizolobium amazonicum frente a diferentes níveis de sombreamento e de demandas atmosféricas. O experimento foi conduzido em dois ambientes com diferentes
demandas atmosféricas (baixa e alta) com os vasos dispostos em três bancadas de metal de 3,0 x 1,2 x 1,0 m de comprimento, largura e altura, respectivamente. Em cada ambiente, duas bancadas foram sombreadas por meio de estruturas de 1,5 m de altura com tela de nylon, cor preta, com níveis de sombreamento (intenso e moderado) e uma bancada permaneceu sem proteção (pleno sol), proporcionando desta forma 3 diferentes níveis de radiação fotossinteticamente ativa. Empregou-se delineamento experimental inteiramente casualizado (DIC), com 6 tratamentos, em esquema fatorial 2x3 com 6 repetições, sendo 2 níveis de demanda atmosférica e 3 níveis de radiação solar, com um total de 36 plantas. Após 94 dias submetidas aos tratamentos, o crescimento das mudas foi caracterizado por meio das medidas de variáveis morfológicas, fisiológicas, anatômicas e índice de plasticidade fenotípica. As médias das variáveis foram submetidos à análise de variância e posteriormente comparadas pelo teste Tukey (P
≤ 0,05). As análises foram realizadas utilizando-se o programa estatístico Assistat 7.7 beta. Na espécie em estudo, o sombreamento exerceu maior influência sobre a maioria das variáveis analisadas (massa seca total, razão de massa foliar, razão de massa caulinar, razão de massa radicular, diâmetro do coleto, área foliar específica, razão de área foliar, clorofila total, densidade estomática e comprimento do limbo), contudo, a demanda atmosférica foi um fator limitante para as variáveis altura, comprimento de entrenós, área foliar total e área foliar unitária. De acordo com os dados observados de massa seca total e índice de plasticidade fenotípica, a espécie mostrou preferência por ambiente de demanda atmosférica alta (temperatura média de 25,2°C e déficit de pressão de vapor dágua (DPV) médio de 1,1 Kpa) em condição de sombreamento moderado e pleno sol. Na última década, várias rotas de reutilização de resíduos de embalagens cartonadas têm sido propostas na literatura. Dentre os processos de recuperação, a pirólise de resíduos cartonados vem se destacando devido à possibilidade de obtenção de produtos com elevado valor agregado, como os óleos parafínicos e barras de alumínio. Recentemente, o leito de jorro tem sido investigado como um reator promissor para a pirólise de sólidos, devido sua alta eficiência energética. Entretanto para que a pirólise ocorra de forma adequada é imprescindível a realização de um procedimento totalmente investigativo da dinâmica do escoamento de ar em meio aos resíduos no leito de jorro cônico. Assim, este estudo tem como objetivo analisar o comportamento fluidodinâmico
do compósito PEBD/AL (em forma de pellet, ds = 2,58 mm) e as misturas de compósito PEBD/AL e areia (ds = 2,58 mm), em um leito de jorro cônico. Os dados experimentais de queda de pressão são medidos por um transdutor de pressão diferencial. Estes são endereçados a uma interface A/D, da National Instruments. A comunicação USB permite que os dados sejam processados em um computador pelo programa de aquisição de dados desenvolvido em linguagem de programação gráfica utilizando o Labview 10.0. Os dados de velocidade do ar são obtidos pelo inversor de frequência que acompanha a unidade experimental. O modelo Euleriano é utilizado nas simulações para descrever o fluxo de ar-sólido em leito de jorro cônico. As equações do modelo são abordadas utilizando a técnica de dinâmica dos fluidos computacional com o método de volumes finitos, utilizando uma malha estruturada bidimensional para discretização do domínio. Os resultados obtidos com a inserção da turbulência (k- ɛ, disperso) ao
modelo subestimam os valores experimentais, onde os dados de velocidade de mínimo jorro (Vmj), queda de pressão de mínimo jorro (ΔPmj) e queda de pressão máxima no leito (ΔPmax) apresentaram desvio percentual máximo igual a -13,5; -9,5 e -23% respectivamente. Qualitativamente, os perfis de porosidade são fisicamente consistentes e estão de acordo com os relatados na literatura. Palavras-chave: Pirólise. Energia. Fluidodinâmica computacional. Leucemia mielóide aguda (LMA) é uma doença de caráter multifatorial e heterogênea. Alterações somáticas no código genético recorrentes na LMA são causadas principalmente por agentes químicos e físicos nocivos ao DNA, associados a falhas nos sistemas de reparo. Pesquisas realizadas na última década sugerem que metilação aberrante e polimorfismos genéticos em genes supressores tumorais e genes do metabolismo de radicais livres podem estar envolvidos na leucemogênese. O presente estudo teve como objetivo investigar a associação entre polimorfismos nos genes TP53 codon 72, GSTT1, GSTM1 e hipermetilação da região promotora do gene BRCA1 com a leucemia mielóide aguda. Duzentos e vinte e três pacientes adultos diagnosticados com LMA e 202 indivíduos adultos são (controles) foram genotipados para polimorfismos no TP53, GSTT1 e GSTM1 e 55 pacientes com AML de novo foram avaliados para metilação no BRCA1. As frequências observadas para o TP53 foram:
33,6% ARG/ARG, 44,4% ARG/PRO e 22% PRO/PRO nos pacientes e 38,3% ARG/ARG, 48,5 ARG/PRO e 12,8% PRO/PRO nos controles. O alelo PRO foi associado à susceptibilidade ao desenvolvimento de LMA no modelo genético recessivo (p=0.0198), no co-dominante (p=0.0291) e no alélico (p=0.0438). O polimorfismo do GSTT1 foi observado em 23% dos pacientes e em 27% dos controles (p=0,297). O polimorfismo do GSTM1 foi observado em 45% dos pacientes e em 43% dos controles (p=0,573) e a co-ocorrência dos dois polimorfismos foi observada em 12% dos casos e 8,5% dos controles (p=0.286). Nenhuma associação foi encontrada entre os polimorfismos nos genes GSTs e LMA. O gene BRCA1 não foi encontrado hipermetilado em nenhum dos 55 pacientes analisados. Os resultados do presente estudo apontam que o polimorfismo do TP53 parece favorecer a leucemogênese. Em contrapartida, os polimorfismos no GSTT1, GSTM1 e metilação do BRCA1 parecem não ter relevância na leucemia mielóide aguda.

▼ The formation of thick collages in firing zone furnace of clinker in cement plants, called clinker ring, is a phenomenon that creates economical and environmental disorders. To remove the clinker ring it is necessary to cool up the oven for 24 hours cutting it with a breaking machine. This process is dangerous and highly cost-consuming, since the furnace fails when producing for about five days and, after removal, it is necessary to heat the furnace with expensive fuel oil. The causes of formation of rings are divided into chemical and operational aspects, and there are several possible causes within these two groups. This work is focused on the formation of rings due to the chemical appearance. So, this work aimed to study the formation of clinker rings and propose improvements in order to minimize and / or eliminate the formation of these rings, to obtain a process with improved operational stability, greater sustainability, economy of fossil fuels and gains in reducing CO2 emissions, the main responsable of the greenhouse effect. The chemical behavior during the formation of clinker rings in the cement factory Camargo Correa Cements, in the Ijaci-MG city during the furnace stop form February 3rd to February 10th 2007, has been studied. Clinker samples were collected four days before stopping furnace due to the ring formation, and four days after starting the furnace without the ring formation, which were subjected to analysis of the reflected light optical microscopy, chemical analysis by x-ray fluorescence (XRF) with calculations of the fitness factor for collage formation (AW), burning factor (BF), percentage of liquid phase, silica module (MS) and alumina module (MA). Collages ring samples are also been collected. These samples were submitted to wet chemical analysis and x-ray diffraction (XRD). In addition, fuel samples were collected and submitted to analysis of lower calorific value (LCV), ash content and particle size in 170-mesh sieve. The results showed that, with regard to the chemical aspect, the ring formation is related to the low viscosity of the liquid phase of clinker during formation which, in turn, is linked to the relationship between the aluminum oxide (Al2O3) and iron oxide (Fe2O3), also known as MA, whose ideal values were those exceeding 1.80. The values found in AW, BF and percentage of liquid phase, described in the literature, were not found as efficient parameters for the non-formation of ring. The XRD and wet chemical analysis of the ring showed that the ring formed is a clinker one and not sulfate. Through the analysis of microscopy it was possible to verify that the furnace was operating in an oxidizing environment. The analysis of fuel samples suggest that operates with a fuel with a less than 2.5%.

Todavia, apÃs colhida, a casca da lichia perde rapidamente sua cor vermelha. O escurecimento da casca tem sido atribuÃdo à rÃpida degradaÃÃo da antocianina. Dada essa alta perecibilidade, o controle do escurecimento à fundamental para o aumento na vida Ãtil pÃs-colheita, visando ao mercado interno e à exportaÃÃo de frutas. Avaliou-se o comportamento pÃs-colheita de lichias quanto ao escurecimento da casca e Ãs modificaÃÃes relacionadas com a qualidade da polpa de frutos submetidos a diferentes embalagens: bandeja recoberta com filme de polietileno de baixa densidade com e sem perfuraÃÃes e frutos recobertos com pelÃcula de fÃcula de mandioca (3%). Os frutos foram mantidos em temperatura ambiente (25,1ÂCÂ1,5ÂC; 69,3%Â8,62%UR) e armazenados por seis dias. Foram realizadas anÃlises fÃsicas, quÃmicas e bioquÃmicas, no dia 0 e a cada 2 dias, atà o sexto dia de armazenamento. A embalagem recoberta com filme de polietileno perfurado mostrou-se eficiente na conservaÃÃo da cor vermelha da casca de lichias durante seis dias de armazenamento, em temperatura ambiente, sem alterar os parÃmetros nutricionais analisados na polpa.
Litchi is a subtropical fruit of high commercial value, due to its characteristic sweet acidic taste, excellent aroma, high nutritive value and attractive deep bright red color of its peel. However, harvested litchi fruit rapidly lose its bright red skin colour. Peel browning of harvested litchi has largely been attributed to the rapid degradation of the anthocyanin pigments. Due to it high perishibility, browning control is a fundamental process to the increase of post-harvest useful life aiming the home market and fruit exportation. This study evaluated the postharvest behavior of litchi fruits regarding pericarp browning and changes on litchi fruits pulp quality when applied differents packings: plastic trays covered with plastic film or hole plastic film and fruits covered with cassava starch film (3%). Litchi fruits were stored for six days at room temperature (25,1ÂCÂ1,5; 69,3%UR). Physical, chemical and biochemical analyses were performed on day 0 and on every two days till the end of the storage period. Packing covered with hole plastic film maintained the deep bright red color of litchi fruit peel during the six days of storage, on room temperature, whitout pulp nutritive value alteration.

▼ Die heutzutage rasche Expansion der Informationstechnologien, deren Eindringen in allen Bereichen des Lebens, Erhebung und Verarbeitung personenbezogener Daten durch den Einsatz von neuen Technologien macht es unumgänglich, den Schutz von personenbezogenen Daten auf internationaler und regionaler Ebene zu gewährleisten. Daher möchte ich eine Abschlussarbeit im Bereich des Datenschutzgesetzes verfassen. Mein wichtigstes Ziel ist es daher, eine umfassende und detaillierte Analyse der Daten-Schutz-Systeme, entsprechende Gesetze und Regulierungsansätze in der AR und der EU durchzuführen.
Das Anliegen dieser Arbeit ist die Beurteilung der Angemessenheit des Datenschutzes in Aserbaidschan.
Rapid expansion of information technologies nowadays, their penetration in all spheres of life, collection and processing of personal data through the use of new technologies makes inevitable to ensure the protection of personal data on the international and regional level. Therefore, I would like to write a thesis in the sphere of data protection law. To be more certain, my main intention is to make a comprehensive and detailed analysis of data protection systems, respective laws and regulatory approaches in AR and EU.
The main purpose of this thesis is to assess the adequacy and sufficiency of data protection in Azerbaijan.

▼ We evaluated attraction of cerambycid beetle species to blends of known cerambycid
pheromones to determine whether such blends could be used as effective trap lures for detecting
and monitoring multiple species simultaneously. Pheromone-baited traps captured 1,358
cerambycid beetles, of which 1,101 (81.1%) belonged to three species in the subfamily
Cerambycinae: Neoclytus acuminatus (F.), Neoclytus mucronatus (F.), and Xylotrechus colonus
(F.). Beetles of these species were significantly attracted to synthetic blends that contained their
pheromone components (isomers of
3-hydroxy-
2-hexanone and/or
2,
3-hexanediol), despite the
presence of pheromone components of different species, including other isomers of
2,
3-
hexanediol, (E/Z)-6,10-dimethyl-5,9-undecadien-
2-yl acetate, and citral. In some cases attraction
was partially inhibited by the pheromone components of heterospecific species, whereas for N.
acuminatus, attraction was completely inhibited when blends contained (2R*,3S*)-hexanediol,
the racemic mixture of diastereomers of its pheromone, (2S,3S)-hexanediol. Among the
remaining beetles captured were three species in the subfamily Lamiinae: Astyleiopus variegatus
(Haldeman), Graphisurus fasciatus (Degeer), and Lepturges angulatus (LeConte). All three
lamiine species were previously known to be attracted to (E/Z)-6,10-dimethyl-5,9-undecadien-
2-
yl acetate, and were captured in significant numbers by blends containing that compound. Our
results suggest that different types of cerambycid pheromones can be combined to create
effective multi-species lures for use in surveillance programs that target exotic cerambycid
species.

▼ The extrinsic labelling of biomolecules for identification and/or quantification is a common methodology used in the lifesciences. This labelling, for example, can be carried out with (i) radioactive markers or (ii) compounds with absorption and/or fluorescence properties. With the advent of confocal microscopy and live cell imaging, fluorescence offers the tools to visualise and quantify the chemical and biochemical processes in living cells. The subject of this thesis concerns the synthesis and investigation of the fluorescence of a new class of organic fluorophores; the 2,5-dihydro-1,2,3-triazines.
A series of fluorescent 2,5-dihydro-1,2,3-triazines were synthesized via an interesting Huisgen 1,3-dipolar cycloaddition between 2,4,5-triphenyl-1,2,3-triazolium-1-arylaminide 1,3-dipoles and terminal alkyne propiolates. The initial cycloadduct undergoes a sigmatropic rearrangement followed by a ring expansion to generate the 2,5-dihydro-1,2,3-triazine fluorophore. The synthesis of 2,5-dihydro-1,2,3-triazine fluorophores with different structural changes, and the affect these changes had on the photophysics of the fluorophores was investigated. This was undertaken with a view to better understanding the fluorescence of this new class of fluorophore and to developing a fluorophore capable of conjugating to biomolecules. Several modifications were explored including (i) substituting the imine phenyl and N2 phenyl rings with heavy atoms, (ii) changing the methyl ester to a methyl ketone or 4-substituted phenyl ester, (iii) varying the substituent on the imine carbon and (iv) removing the phenylimine group.
In collaboration with Dr. Alan G. Ryder at NUI Galway and Prof. Luke A. Burke at Rutgers University, New Jersey; a full fluorescence charaterisation was performed building on preliminary photophysical studies. Steady state ultraviolet absorption, fluorescence spectroscopy and time domain lifetime measurements were conducted in order to elucidated a model of fluorescence for the 2,5-dihydro-1,2,3-triazines. These measurements in combination with TD-DFT calculations (relative energies, dipole moments, oscillator strength and calculated transition wavelength), courtesy of Prof. Luke A. Burke, allowed us
to generate a photophysical model that explains the fluorescence of the 2,5-dihydro-1,2,3-triazines. It was found that three structures may exist for the 2,5-dihydro-1,2,3-triazines: g0 (optimized for S0), g1 (optimized for S1) and, g2 (optimized for S2) and that the positioning of the imine and carbonyl ester on the triazine is very important in dictating the fluorescent and electronic properties. It was found that the phenylimine group is essential for the long lived fluorescence of the 2,5-dihydro-1,2,3-triazines. Furthermore, we propose that in order to maintain fluorescence the phenylimine group must have substituents small enough to allow the imine group to remain in plane with the carbonyl group. ▼ Engineered biocatalysts have become increasingly sought after as replacements for individual chemical synthetic steps. However, their implementation in multistep sequences as biosynthetic pathways has proven to be a significant challenge, in part due to a lack of broadly applicable methods for their assembly. One strategy for construction of a de novo biosynthetic pathway can be inspired by the theory of retrograde evolution, where pathway enzymes are recruited and optimized in the reverse order of biosynthesis. Herein, this process, termed ‘bioretrosynthesis’, is demonstrated through the construction and evolution of a pathway for in vitro production of the antiretroviral nucleoside analog didanosine (
2,
3 dideoxyinosine). Using methods in structure guided mutagenesis and directed evolution, we have evolved phosphopentomutase to accept
2,
3-dideoxyribose 5 phosphate with a 700-fold change in substrate selectivity and
3-fold improved turnover in cell lysate as a retrograde extension from an engineered purine nucleoside phosphorylase. A second pathway extension via evolution of ribokinase resulted in a didanosine biosynthetic pathway with 9,500-fold change in nucleoside production selectivity. Surprisingly, the product of bioretrosynthesis in this step was not a retrograde extension from phosphopentomutase, but instead resulted in the discovery of a pathway shortening bypass via a new phosphorylation regiochemistry in the engineered ribokinase.

▼ This thesis is an investigation of ruthenium-catalyzed [
2+2] cycloaddition reactions of a
3-aza-
2-oxabicyclo[
2.2.1]hept-5-ene with unsymmetrical alkynes. Yields of up to 90% were obtained though regioselectivity was modest. Select cycloadducts could be separated and used to access a highly functionalized [
3.
2.0] bicyclic structure through reductive cleavage of the N-O bond. These ring-opened products displayed a chemical exchange phenomenon in 1D carbon NMR and required characterization by 2D NMR techniques. In addition, a haloalkynylation reaction was found to occur when 1-iodo-
2-phenylethyne and the
3-aza-
2-oxabicyclo[
2.2.1]hept-5-ene were submitted to the cycloaddition conditions. An effort was made to optimize the reaction between 1-iodo-
2-phenylethyne and norbornadiene in favour of the addition product.
