The purpose of this study was to identify the levels of Sister ChromatidExchanges (SCEs), if there is any cytostaticity or cytotoxicity, and moreoverthe possible instability of the genetic material in patients with Chronic RenalFailure after culturing of their peripheral blood lymphocytes of their blood.This study was done in 32 patients with CRF (with an average age of56.2 years old) from whom 18 had also a cancer, and furthermore in 18healthy blood donors (with an average age of 44.6 years old) from whom noone was a smoker or under the influence of drugs or virus infections.The samples were peripheral lymphocytes blood cultures with heparin,using 11 drops of blood in 5 ml chromosomal medium. After that, there wasan incubation at 37οC for 72 hours. Cell proliferation was allowed for at least 2mitotic cycles with the presence of BrdU in a final concentration of 5μg/ml andmitomycin C (MMC) in a final concentration of 15ng/ml added at the beginningof the cell cultures. After 70 hours, colcemide was added in a finalconcentration of 0,5 μg/ml for two hours, and at the end of the incubationperiod the cells were collected and Giemsa dye was applied (using themodified method FPG).There was an evaluation of the Proliferation Rate Index (PRI) of thelymphocytes as an index of cytostaticity, and an evaluation of Mitotic Indices(MI) as an index of cytotoxicity.There was a significant increase of the frequency of SCEs, estimatedat 9,23 SCEs/cell, in comparison to the frequency of the healthy donors wherethe average amount was 4.61 SCEs/cell. The increase percentage of SCEslevel was calculated at 100.2% (p<0,05).Furthermore, a decrease was observed at the PRIs and at the MIs arks(ΜΔ) of the patients, in comparison to those of the healthy donors. Thereduction of PRIs touched the percentage of 2,8%, which is particularly lowand it indicates the existence of a cytostaticity, in comparison to the reductionpercentage that was observed at the case of MIs, that was found 64,4% and itindicates a high cytotoxicity.It is important also to emphasize that there wasn’t any significantdifference at the above results between the patients with CRF and thepatients with CRF and cancer.Finally, by adding MMC there was a significant statistical increase ofthe SCEs (p<0,05) at the patients with CRF in comparison to healthy donors.That indicates the increased chromosomal fragility in the genetic material ofthese patients, a fact that leads to precaution and medication usage withcaution in patients with CRF.

No presente trabalho foram estudadas quatro espécies de peixes pertencentes à família Pimelodidae, coletadas em duas bacias hidrográficas distintas. Pimelodus paranaensis e Pimelodus heraldoi coletados no rio Piquiri/PR, Parapimelodus nigribarbis e Pimelodus maculatus do lago Guaíba/RS. Os dados citogenéticos revelaram para as quatro espécies um número diplóide igual a 56 cromossomos, com diferenças em suas fórmulas cariotípicas: 22 m + 22 sm + 4 st + 8 a para P. paranaensis; 18 m + 24 sm + 6 st + 8 a para P. heraldoi; 20 m + 20 sm + 4 st + 12 a para Parapimelodus nigribarbis e 24 m + 20 sm + 6 st + 6 a para P. maculatus. As NORs foram localizadas na região terminal do braço longo em um par de cromossomos subtelocêntrico em todas as espécies, sendo observado em P. heraldoi e P. maculatus um heteromorfismo de tamanho desta região entre os cromossomos homólogos. Em relação ao padrão de distribuição da heterocromatina, P. paranaensis, P. heraldoi e Parapimelodus nigribarbis, apresentaram blocos heterocromáticos nas regiões terminais de vários cromossomos e em poucas regiões centroméricas. P. maculatus apresentou uma menor quantidade de heterocromatina, sendo observado um cromossomo portador de uma banda intersticial. A coloração com o fluorocromo CMA3, evidenciou regiões correspondentes as NORs, entretanto, em Pimelodus paranaensis, Pimelodus heraldoi, Parapimelodus nigribarbis também foram observadas marcações fluorescentes em outros pares de cromossomos. Através da coloração com o fluorocromo DAPI, foram identificadas apenas regiões DAPI negativas, sendo estas coincidentes com as regiões GC ricas. Os resultados de banda C + CMA3, apresentaram alguns cromossomos com marcações fluorescentes em P. paranaensis, P. heraldoi e Parapimelodus nigribarbis; em P. maculatus apenas a heterocromatina associada à NOR mostrou-se rica em bases GC. Por outro lado, com banda C + DAPI, vários cromossomos apresentaram bandas heterocromáticas DAPI positivas, inclusive a marcação intersticial no par 1 em Pimelodus maculatus, indicando que, nestas espécies, a composição da heterocromatina possui, principalmente, bases AT.

The present work was a cytogenetic study of four species of fish belonging to the family Pimelodidae that were collected from two different hydrographic basins. Pimelodus paranaensis and Pimelodus heraldoi were collected in the Piquiri River/PR, and Parapimelodus nigribarbis and Pimelodus maculatus were from Lake Guaíba/RS. The data revealed that the four species had the same diploid number of 56 chromosomes, with differences in their karyotype formula: 22 m + 22 sm + 4 st + 8 a for P. paranaensis; 18 m + 24 sm + 6 st + 8 a for P. heraldoi; 20 m + 20 sm + 4 st + 12 a for Parapimelodus nigribarbis; and 24 m + 20 sm + 6 st + 6 a for P. maculatus. The NORs were located in the terminal area of the long arm in a pair of st chromosomes in all of the species, where P. heraldoi and P. maculatus showed a size heteromorphism for this region between homologous chromosomes. ▼ Intracellular symbioses between cnidarians and dinoflagellates from the genus Symbiodinium are widespread throughout the marine environment. These associations are ecologically significant, especially in tropical waters where symbiotic interactions between corals and Symbiodinium culminate in the formation of limestone reefs. This thesis focuses on cellular and molecular aspects of the symbiosis, specifically the initiation of the symbiosis and characterization of a host gene, sym32, that is believed to function in the symbiosis. Sym32 was originally identified as a differentially expressed protein in symbiotic vs. aposymbiotic individuals of the sea anemone, Anthopleura elegantissima. Based on its deduced amino acid sequence, sym32 belongs to a family of cell adhesion proteins that play roles in cell recognition in a diverse array of organisms. Chapter 2 examines the process by which a new cnidarian host acquires its first symbionts. Larvae of the scleractinian coral Fungia scutaria, which are initially aposymbiotic, acquired symbionts while feeding. Symbionts that entered the larval gastric cavity with food were subsequently taken into host gastrodermal cells by phagocytosis. Chapter 3 describes immunolocalization of sym32 in A. elegantissima tentacles. In aposymbiotic tentacles, sym32 was localized to vesicles within the host gastrodermal cells. Symbiotic tentacles lacked sym32-containing vesicles. Instead, sym32 was present among the membranes that enclose the symbionts within host cells. Western blots of proteins from Symbiodinium revealed a 45/48kD doublet that cross-reacts with anti-sym32 antiserum. This suggests that homologous proteins are expressed in both host (32kD) and symbiont (45/48 kD). Chapter 4 describes the effects of environmental factors on expression of host sym32. Aposymbiotic and symbiotic anemones maintained in continual darkness for 3 weeks experienced a dramatic decline in sym32 protein levels, relative to anemones maintained on a 12:12 h light:dark cycle. This suggests that light plays a major role in regulating sym32. Exposure of anemones to elevated temperatures for 2 days in the dark caused a mild bleaching response (expulsion of symbionts from the host), but did not affect the levels of sym32 protein. Chapter 5 examines the role of sym32 during the infection process, using antibody interference techniques. F. scutaria larvae and symbionts incubated in sym32 antiserum during the infection process experienced a decline in infection rates. Further, symbionts that were incorporated into host gastroderm appeared to be degenerating in antiserum treatments, but appeared to be healthy in preimmune controls. ▼ Myeloproliferative neoplasms (MPN) are a group of clonal hematopoietic stem cell cancers with overlapping laboratory, cytogenetic, molecular, and clinical features. According to the World Health Organization (WHO, 2016), there are three classic Philadelphia chromosome negative MPN (Ph- MPN) entities: polycythemia vera (PV), essential thrombocythemia (ET) and primary myelofibrosis (PMF). Currently, the genomic characterization of these diseases is based on conventional cytogenetics and the mutational status of three driver genes (JAK2, CALR, and MPL). Although other non-driver mutations may be present in up to 40% of patients, their clinical and prognostic relevance, at present, is not entirely clear. Cytogenetic abnormalities are absent in 90% of patients with ET, 70% of patients with PV, and 50% of patients with PMF. Moreover, up to 10% of patients do not harbor mutations in any of the three driver genes. Chromosomal microarray (CMA) abnormalities have been identified in ~80% of MPN patients but the application of this technology for prognostic clinical purposes has not been widely used. The goal of this dissertation was to gain additional insight into the genetic basis of these diseases by integrating three genomic technologies; conventional cytogenetics, next-generation sequencing (NGS), and CMA, each with its advantages and limitations. To achieve this goal, three groups of patients were investigated: (1) patients with advanced forms of Ph- MPN who required therapeutic splenectomy, (2) patients that progressed from the chronic form to more advanced forms of Ph- MPN that were associated with specific chromosomal abnormalities, and (3) cytogenetically normal patients with Ph- MPN that represent early stages of their disease. The hypothesis in aim 1 was to determine whether the spleens of patients with advanced forms of Ph- MPN, such as myelofibrosis requiring therapeutic splenectomy, contain hematopoietic stem cells which harbor unique genomic aberrations that contribute to disease progression. The results of the integrated analysis showed a high concordance of genomic lesions between spleen and bone marrow cells suggesting a similar clonal architecture between these two tissues. In spite of the overall similarity, CMA identified additional novel genomic changes present in the spleen indicating that there are genomic differences between myelofibrosis hematopoietic cells that reside in the spleen and bone marrow. CMA results demonstrated for the first time recurrent gains within four genes: RHOC, RASA3, NCOR2, and TAF15, strongly suggesting higher genomic complexity of spleen cells. These candidate genes may play a role in the pathogenesis PMF and support the hypothesis that integrating genomic technologies is a valuable tool in the discovery of new genes that may be implicated in the genetic landscape of myelofibrosis. ▼ Sex-determining mechanisms (SDMs) set an individual’s sexual fate by its genotype (genotypic sex determination, GSD) or environmental factors like temperature (temperature-dependent sex determination, TSD). While sexual reproduction is one of the most taxonomically conserved traits, SDMs are incredibly diverse and evolve frequently. The vast diversity and complexity of SDMs harbored within vertebrates, including turtles still puzzles researchers, and our understanding of how SDMs evolve remains incomplete. This dissertation examines the cytogenetics underpinning of SDM evolution in turtles, exploring the evolution of sex chromosomes and genome organization via chromosomal rearrangements in turtles. In Chapter 2, I characterized the sex chromosome system of Emydura subglobosa turtles by the identification of sex-specific regions that revealed its type of heterogamety. Combining this new information with published data from other turtles, I examined the evolution of sex chromosomes in chelid turtles in a phylogenetic framework and uncovered a novel evolutionary trajectory from what had been previously proposed, which I found involves a Y-autosome fusion. Furthermore, these data also permitted dating this Y-autosome fusion in the Emydura lineage to at least 45 Mya. In Chapter 3, I examined the patterns of synteny of 10 target genes among 12 TSD and GSD focal turtle species to identify genomic regions whose evolution may be associated with transitions in turtle sex determination. I found no evidence to support the hypothesis that changes in the relative position of these gene regulators of sex determination/differentiation are associated with evolutionary transitions in turtle sex determination. However, I uncovered intrachromosomal rearrangements that altered the relative position of some genes that are involved in sexual development within a single chromosome (intrachromosomal rearrangements), fusion/fission events in other chromosomes (interchromosomal rearrangements), and cases of autosome to sex chromosome translocation of a pair of genes in a GSD turtle that render them potential candidate sex-determining genes. In Chapter 4, I used fluorescent in situ hybridization of bacterial artificial chromosome clones (BAC-FISH) from a painted turtle Chrysemys picta library to enhance the physical mapping of the genome assembly of the painted turtle whose contiguity was improved by BioNano optical mapping. This approach improved the C. picta assembly by ~242% compared to the previous assembly, and physical mapping permitted anchoring ~45% of the genome, including 20 genes related to the sex determination network of turtles and vertebrates. Here, I also detected a few discrepancies between BAC-FISH physical mapping and the new assembly, which highlight the importance of molecular cytogenetics data to identify and correct assembly errors. A subfamília Loricariinae apresenta 209 espécies distribuídos em 31 gêneros, sendo que até o momento apenas 15 espécies foram estudadas citogeneticamente. Frente à grande variabilidade interespecífica que é encontrada na macroestrutura cariotípica da subfamília Loricariinae, este grupo é um excelente material para estudos citogenéticos. No presente estudo foram analisadas cinco espécies da subfamília Loricariinae: Loricariichthys platymetopon e Rineloricaria pentamaculata, coletados na bacia do Rio Paranapanema; Loricariichthys anus, Rineloricaria cadeae e Rineloricaria strigilata, coletados no sistema hidrográfico do lago Guaíba/RS. Os exemplares foram submetidos a análises cariotípicas por meio da coloração convencional (Giemsa), impregnação por nitrato de prata, bandamento C e fluorocromos base-específicos Cromomicina A3 e DAPI. Na análise com Giemsa, Loricariichthys platymetopon e Loricariichthys anus apresentaram 2n=54 cromossomos, entretanto com diferentes fórmulas cariotípicas. Loricariichthys platymetopon apresentou 6m+18sm+4st+26a e L. anus 8m+16sm+4st+26a, sendo que nesta ultima espécie foi encontrado um polimorfismo estrutural no par 25, tanto para machos como para fêmeas. O gênero Rineloricaria, apresentou número diplóide, de 2n=56 cromossomos (2m+6sm+48a) para R. pentamaculata, 2n=62 cromossomos (2sm+2st+58a) para R. cadeae e 2n=70 (4sm+2st+64a) para R. strigilata. A impregnação pelo nitrato de prata detectou RONs simples para todas as espécies, sendo intersticial somente para L. anus. A coloração com CMA3 mostrouse correspondente a estas regiões, que apresentam-se negativas com DAPI. A heterocromatina mostrou-se distribuída na região pericentromérica da maioria dos cromossomos e também associada às RONs em todas as espécies analisadas. Os resultados obtidos no presente trabalho são importantes para uma melhor caracterização das espécies estudadas e podem auxiliar nos estudos relacionados a evolução cariotípica desta subfamília.

The subfamily Loricariinae has about 209 species and 31 genera, of which only 15 have been examined, distributed in five genera. The great interspecific variability found at the karyotypic macrostructure of this subfamily, make this group an excellent material for cytogenetics studies. At the present study, five species belonging to the subfamily Loricariinae were investigated: Loricariichthys platymetopon and Rineloricaria pentamaculata, from Paranapanema river basin/PR; Loricariichthys anus, Rineloricaria cadeae and Rineloricaria strigilata from hydrographic system of Guaíba lake/RS. The specimens were submitted into cytogenetics studies by convencional color (Giemsa), silver nitrate staining, Cbanding and Cromomicin A3 and DAPI fluorochromes. At Giemsa analysis, L. platymetopon and L. anus presented 2n=54 chromosomes, but with different karyotypic formulas. L. platymetopon showed 6m+18sm+4st+26a and L. anus 8m+16sm+4st+26a, at the last species has been found an structural polymorphism, for males and females. The main objective of the present study was to enhance the genetic variability in yield and other quantitative traits in chickpea (Cicer arietinum L.), a self pollinated crop. Due to lack of sufficient naturalvariability, conventional methods of plant breeding have a limited scope in the improvement of chickpea. Therefore, an attempt was made to evaluate genetic variability in different generations of single and combined treatments of gamma rays and ethylmethane sulphonate (EMS) in the two varieties, Pusa-256 and BG-1053 of chickpea. Various other aspects of this study were:(i) bio-physiological damages and cytological aberrations in M1 generation (ii) frequency and spectrum of chlorophyll and morphological mutations (iii) effectiveness and efficiency of the mutagens (iv) estimation of genetic parameters viz., genotypic coefficient of variation, heritability (broad-sense) and genetic advance in different quantitative traits (v) estimation of some mineral elements (iron, manganese, zinc and copper) compositions and physiological parameters (NRA, chlorophyll and carotenoid contents) in the high yielding mutant lines (vi) estimation of seed protein contents and protein profiling of the high yielding mutants isolated in M3 generation. The effect of gamma rays and EMS and their combination treatments were studied on seed germination, seedling height, NRA, chlorophyll and carotenoid contents, pollen fertility and plant survival at maturity in M1 generation. A linear ependence of seed germination, seedling height and pollen fertility on dose of the mutagens was observed in both the varieties of chickpea. Plant survival at maturity, NRA, chlorophyll and carotenoid contents decreased in all treatments of the mutagens. However, such parameters were dose independent. Combination treatments showed an enhanced effect on seed germination, seedling height and pollen fertility in comparison to individual mutagenic treatments.

O objetivo deste trabalho é identificar e avaliar o efeito genotóxico do carcinógeno 4NQO em células da medula óssea de camundongos C57BL/6 por meio da técnica de detecção de micronúcleos, bem como identificar a possível diferença neste efeito pela ausência constitutiva do gene da galectina-3. Camundongos C57BL/6 selvagens (WT) e knockout (KO) para o gene da galectina-3 foram tratados por via oral durante 16 semanas com a 4NQO e divididos em 2 subgrupos cada um, de acordo com o tempo de observação: WT16/KO16 - tratados durante 16 semanas, eutanasiados imediatamente após o término do tratamento e WT32/KO32 - tratados com 4NQO durante 16 semanas, eutanasiados 16 semanas após o término do tratamento. Após o término do tratamento de cada subgrupo, obedecendo à cronologia de observação do experimento estabelecida previamente, os animais sofreram eutanásia. Em seguida as línguas foram retiras e utilizadas para análises microscópicas e a medula óssea foi coletada dos fêmures e submetida à técnica do MN. Ao final do experimento, 100% dos camundongos dos grupos WT e KO de 16 e 32 semanas de observação apresentaram alterações no tecido epitelial lingual que puderam ser reconhecidas como displasia, carcinoma in situ ou carcinoma epidermóide. No grupo composto por animais WT tratados com a 4NQO foi encontrado maior quantidade de PCEs micronucleados comparado com o mesmo grupo constituído por animais KO, essa diferença foi estatisticamente significante. O presente estudo revelou que animais knockout para galectina-3 quando submetidos ao tratamento crônico via oral de solução carcinogênica apresenta menor número de micronúcleos, o que pode indicar uma menor susceptibilidade a expressão destes danos.

The purpose of the study was to observe the possible difference of the genotoxic effect caused by the chronic treatment with 4-nitroquinoline carcinogen (4NQO) in wild-type and knockout mice for the galectin-3 gene. Wild-type C57BL/6 mice (WT) and knockout mice (KO) for the galectin-3 gene were orally treated with 4NQO and killed at 16 (WT16/KO16) and 32 week (WT32/KO32). After the treatment of each sub-group the animals had suffered euthanasia, the tongue had been remove and used for microscopical analyses and the bone marrow was removed of femur and submitted to micronuclei (MN) test. To the end of the experiment, 100% of the groups WT and KO of 16 and 32 weeks had presented alterations in the tongue epithelial tissue that could have been recognized as displasia, carcinoma in situ or carcinoma. In the groups treated with 4NQO the number of micronucleated PCE was significantly higher in WT when compared with the animals knockout (KO). The present study revealed that knockout mice for galectin-3 when submitted to the chronic treatment with carcinogen presents minor number of micronuclei, what it can indicate a lesser susceptibility the expression of these damages.

A membrane receptor for corticosterone (CORT) has been described in brains of the roughskin newt (Taricha granulosa). This receptor is believed to be a G-protein coupled receptor because corticosterone binding is inhibited by guanyl nucleotides and enhanced by Mg²⁺. The studies described in this thesis use biochemical, pharmacological and molecular techniques to characterize the newt neuronal membrane glucocorticoid receptor (mGR) in pursuit of its molecular identification. The mGR was successfully solubilized from newt neuronal membranes and conditions were defined that maintained corticosterone binding activity for further study. The solubilized receptor was partially purified using standard chromatographic techniques and an immobilized ligand affinity resin (CORT-Sepharose). These chromatographic studies were combined with the use of a novel photoaffinity ligand (azido-CORT) to biochemically characterize the mGR protein, finding that it is an acidic glycoprotein with an apparent molecular weight of 63 kDa and an isoelectric point of approximately 5.0. Pharmacological studies with mGR showed that a subset of kappa opioid ligands displaced corticosterone from the receptor binding site with K[subscript i] values in the nanomolar to low-micromolar range. The interaction of mGR with kappa opioid ligands was specific because no mu-, delta-, or orphanin-specific opioid ligands were effective at displacing corticosterone from the receptor. These data suggest that the newt neuronal mGR may be a kappa-opioid like receptor. Finally, molecular studies were used to clone a novel newt brain protein, neuronal axonal protein 22 (NAP-22), that was identified in a protein differential display strategy designed to identify mGR. Studies with the cloned and expressed NAP-22 protein suggest that it is not the mGR but, instead, may be a mGR-associated protein. These studies provided new information about the biochemical and pharmacological properties of mGR, and may have discovered a protein that is associated with the newt neuronal mGR. ▼ Chromosome changes in the haematological malignancies, lymphomas and sarcomas are known to be important events in the evolution of these tumours as they can, for example, form fusion oncogenes or disrupt tumour suppressor genes. The recently described recurrent fusion genes in prostate and lung cancer proved to be iconic examples as they indicated that important gene fusions are found in the common epithelial cancers also. Breast cancers often display extensive structural and numerical chromosome aberration and have among the most complex karyotyes of all cancers. Genome rearrangements are potentially an important source of mutation in breast cancer but little is known about how they might contribute to this disease. My first aim was to carry out a structural survey of breast cancer cell line genomes in order to find genes that were disrupted by chromosome aberrations in “typical” breast cancers. I investigated three breast cancer cell lines, HCC1187, VP229 and VP267 using data from array painting, SNP6 array CGH, molecular cytogenetics and massively parallel paired end sequencing. I then used these structural genomic maps to predict fusion transcripts and demonstrated expression of five fusion transcripts in HCC1187, three in VP229 and four in VP267. Even though chromosome aberrations disrupt and fuse many genes in individual breast cancers, a major unknown is the relative importance and timing of genome rearrangements compared to sequence-level mutation. For example, chromosome instability might arise early and be essential to tumour suppressor loss and fusion gene formation or be a late event contributing little to cancer development. To address this question, I considered the evolution of these highly rearranged breast cancer karyotypes. The VP229 and VP267 cell lines were derived from the same patient before and after therapy-resistant relapse, so any chromosome aberration found in both cell lines was probably found in the common in vivo ancestor of the two cell lines. A large majority of structural variants detected by massively parallel paired end sequencing, including three fusion transcripts, were found in both cell lines, and therefore, in the common ancestor. This probably means that the bulk of genome rearrangement pre-dated the relapse. For HCC1187, I classified most of its mutations as earlier or later according to whether they occurred before or after a landmark event in the evolution of the genome - endoreduplication (duplication of its entire genome). Genome rearrangements and sequence-level mutations were fairly evenly divided between earlier and later, implying that genetic instability was relatively constant throughout the evolution of the tumour. Surprisingly, the great majority of inactivating mutations and expressed gene fusions happened earlier. The non-random timing of these events suggests many were selected.

▼ Chromosome microdissection is a technique that was first described in 1981 by Scalenge et al. Since then, several modifications have been made and it was recently applied to dissect abnormal parts of aberrant chromosomes, the origin of which was unknown, with the ultimate aim of identifying the origin of the dissected chromosomal material (Meltzer et al. 1992). The aim of this project was to develop the technique, so that it could be further used as a molecular cytogenetic tool in the delineation of chromosome aberrations encountered in the diagnostic workload of this department which remain unidentifiable by using standard cytogenetic techniques. The steps associated with the technique involved the microdissection of several copies of a specific chromosomal region, PCR amplification using a degenerate oligonucleotide primer (DOP primer with six 'N' degenerate bases), labelling of the amplified product with biotin-11-dUTP and finally, reverse chromosome painting by hybridising the labelled product on normal metaphase cells. The location of the hybridisation signal would then reveal the chromosomal origin of the microdissected DNA of interest. All parameters that could affect the efficiency of the DOP-PCR amplification, such as the primer concentration and annealing temperatures were evaluated by using low concentrations of at first, human genomic DNA and finally FACS isolated copies of chromosome 4 as a positive control. Satisfactory results meant that the values of the parameters tested were kept constant for all further attempts. Contamination was the main drawback in interpreting the PCR results. The nature of the DOP primer that could amplify literally any type of DNA template, in conjunction with the fact that the starting microdissected material did not exceed in total a few hundred femptograms (fg) (an average size band equals to 15-50fg, 1fg=10-9 mug) made the amplification extremely sensitive to extraneous contaminations. It was therefore necessary to sequentially introduce multiple specific precautions (i.e. barrier-pipette tips, autoclaving all solutions, preparing small aliquots of each of the PCR reagents to allow only a single use per experiment and performing all steps involved with the PCR inside a sterile safety cabinet) in order to prevent contamination. Several approaches to chromosome microdissection were investigated. Successfiil results were obtained by dissecting the fragment of interest and transferring it into a collection tube by gently touching the tip of the needle inside the collection drop, so as to release the DNA fragment without having to break the tip of the needle inside the tube. For this, it was very important to obtain the correct size of needles. Successfiil micro-FISH probes were produced by less than ten fragments, proving the efficiency of the technique.
