▼ Hypothalamus and adipose tissue are essential for regulating energy homeostasis in central and peripheral body sites, respectively. Disruption of energy homeostasis can lead to diseases like anorexia and obesity in humans and reduced productivity in animals. Therefore, integrating knowledge in hypothalamic appetite regulation and adipose tissue metabolism is essential to maintain energy homeostasis in both humans and animals. The aim of this dissertation was to elucidate molecular and cellular mechanisms of energy homeostasis in birds. We first determined adipose tissue physiological changes in chickens during the first two weeks post-hatch from lines selected for low (LWS) and high (HWS) body weight. These chickens have been selected for juvenile body weight for over 60 generations. The LWS are lean and anorexic, while HWS eat compulsively and develop obesity and metabolic syndrome. Such characteristics make the body weight line chickens good animal models to study physiological changes under anorexia and obesity. We found that LWS was more dependent on yolk reserves and subcutaneous fat mobilization for growth from hatch to day 4 post-hatch, with replenishment of the fat reservoir by increases in cell number. By contrast, HWS was more dependent on feed for growth and maintained depot mass through increased cell number and cell size. From day 4 to 14 post-hatch, HWS accumulated fat throughout the body, with less fat breakdown as compared to LWS. There was greater expression of cellular precursor and proliferation markers in LWS, with more dying cells in their abdominal fat on day 14 post-hatch, suggesting that programmed cell death is responsible for the lack of fat cell development in LWS. Exposure to thermal and nutritional stressors at hatch impaired growth by reducing yolk utilization and lowering body weight, lean and fat masses in LWS. There were many molecular changes in the hypothalamus, including changes in DNA that led to increased activation of corticotropin-releasing factor (CRF), a signaling molecule that is known to regulate the body's stress and appetite responses. Stress exposure increased global DNA methylation and DNA methyltransferase activity in the arcuate nucleus of the hypothalamus in LWS. Moreover, there was less methylation at the core binding site of methyl-CpG-binding domain protein 2 (MBD2), a protein that binds to methylated DNA to repress gene expression, in the CRF gene, in stressed LWS. In response to stress, there was decreased binding of MBD2 to the promoter region of CRF, which may explain increased expression of CRF in the paraventricular nucleus of LWS. These results demonstrate that early-life stressful events can cause epigenetic changes (like DNA methylation) that lead to alterations in physiology and behavior that persist to later in life. We next determined effects of nutritional status on adipose tissue physiology in Japanese quail, which have undergone less artificial selection than chickens and are more representative of a wilder-type bird. ▼ ENGLISH ABSTRACT: The South African Ostrich industry was established more than 100 years ago. Despite appreciable fluctuations and modifications in demand, the industry perseveres, with meat and leather as the current primary driving force. Low and variable early chick survival is globally considered as a major constraint in the industry. In spite of obvious welfare and production implications, research findings concerning ostrich chick survival are scant. Four studies involving chick survival to 24 weeks post hatch were thus performed. South African Black ostrich (Struthio camelus var. domesticus) data obtained from a commercial pair-bred ostrich flock maintained at the Oudtshoorn Experimental Farm, South Africa comprising 10418 records were utilized to run a series of single- and multiple-trait binomial analyses using either linear models (ASREML) or threshold models (Monte Carlo Markov Chain methods, employing Gibbs sampling software) respectively. Suitable fixed and random effect models together with variance and (co)variance components were derived from these analyses. Ostrich chick survival to 24 weeks post hatch was low (28%) with a large environmental component. Preincubation storage time only affected 0 to 3 week survival, while incubator had a significant effect on cumulative survival traits to 24 weeks as well as on 13 to 24 week survival. Female chicks outperformed males at 0 to 12 weeks, 0 to 24 weeks, 4 to 12 weeks and 13 to 24 weeks with regard to survival. Hen age was significant for all traits except survival during the first week as well as for survival from 13 to 24 weeks post hatch. All analyses exhibited comparable low heritability estimates for all survival traits except for 0 to 1 week survival and 13 to 24 week survival (h2 = 0.12 and 0.10; m2 = 0.08 and 0.07 respectively) which demonstrated higher additive genetic and maternal components when threshold models, using Gibbs sampling algorithms, were employed. Neither moisture loss nor day of external pipping exhibited notable additive genetic correlations with any of the estimated survival traits. Day-old chick weight demonstrated a low correlation with early chick survival (rg = 0.24 ± 0.19) with heavier chicks tending to have a higher survival probability. The effects of rearing method on early chick survival and growth were also investigated. Comparisons between chicks reared and fostered by adult ostriches in a semiextensive environment and chicks imprinted onto humans under an intensive system and between regular human handling in an intensive rearing system and the standard farm protocol of an intensive rearing system were made. No difference in the early chick survival to 3 weeks post hatch of chicks reared by foster parents and those chicks imprinted onto humans was demonstrated. At later ages, those chicks reared by foster parents outperformed human imprinted chicks. O objetivo deste trabalho foi avaliar a utilização da levedura hidrolisada como fonte de nucleotídeos para frangos de corte. As aves foram suplementadas com diferentes níveis de inclusão nas dietas iniciais de 1 a 14 dias de idade. Foram utilizados um total de 576 pintos da linhagem Ross 708, os quais foram distribuídos em 6 tratamentos com 8 repetições (12 aves por box). Os tratamentos consistiram da inclusão de 0, 2, 4, 6, 8 e 10 kg de levedura hidrolisada/tonelada de ração. A levedura hidrolisada foi adicionada na dieta no lugar do material inerte da ração. A dieta inicial foi fornecida na forma triturada enquanto as dietas de crescimento, final e de retirada foram fornecidas na forma de pellets. Durante todo o experimento o acesso à água e ração foi ad libitum. Todas as dietas foram feitas à base de milho, farelo de soja e gordura de frango e foram formuladas para atender as exigências nutricionais recomendadas pelo manual de recomendações nutricionais Ross 708. Aos 42 dias, as aves alimentadas com 1% de levedura hidrolisada tiveram maior peso corporal e ganho de peso quando comparadas aos demais tratamentos (P<0,05). Não houveram diferenças estatísticas entre os tratamentos para as variáveis mortalidade e densidade de vilos. Contudo aves que não foram suplementadas com levedura hidrolisada (tratamento controle) apresentaram menor profundidade de cripta e a suplementação de 1% de levedura resultou em maior altura de vilos. Aves as quais receberam dietas com 0,2% de inclusão de levedura hidrolisada apresentaram menor rendimento de peito que as aves que receberam os demais níveis de levedura, mas foram iguais as aves do tratamento controle. Contudo, o rendimento de carcaça, sassami e gordura abdominal não foram influenciados pelos tratamentos experimentais. Estes resultados demostraram a eficácia da utilização de levedura hidrolisada na dieta de frangos de corte no período de 1 a 14 dias sobre as características de produção.

The objective of this work was to evaluate hydrolyzed yeast utilization as nucleotides source to broilers. Birds were supplemented with different inclusion levels on starter diets from 1 to 14 days of age. A total of five hundred seventy six Ross 708 chicks were allotted to 6 experimental treatments with 8 replications (12 broilers per pen). Birds were randomly distributed in following treatments: 0, 2, 4, 6, 8 and 10 kg hydrolyzed yeast/ton of feed. Hydrolyzed yeast was added to the test diet in place of filler. Starter diets were supplied in crumbled form while grower, finisher, and withdrawal were supplied in pellet form. Throughout experiment water and feed were supplied ad libitum. All diets were based on corn, soybean meal and poultry fat, and were formulated to achieve nutritional requirements from recommendations guide for Ross x Ross 708 broilers. At 42 d chicks fed 1% hydrolyzed yeast demonstrated higher body weight and body weight gain over birds fed other treatments (P<0.05). Mortality and villous density did not differ among treatments. Salmonella do grupo paratifóide é responsável por toxi-infecção alimentar no homem, veiculada por alimentos contaminados. Este estudo determinou o perfil genotípico de nove amostras de S. Typhimurium, a sua patogenicidade, assim como sua capacidade de colonização e invasão em aves SPF. Verificou-se pela análise dos genes: agfA, avrA, cdtB, fimA, fliC, invA, iroN, lpfC, mgtC, pefA, sefC, sifA, sipB, sipC, sitC, slyA, sopB, sopE1, sptP ou spvC em amostras de Salmonella Typhimurium que todas foram negativas para os genes sopE1 ou spvC. O gene cdtB estava presente em apenas uma amostra (11,11%) e o gene pefA em duas amostras (22,22%). O gene sefC foi encontrado em três amostras (33,33%). Em oito amostras (88,88%) os genes agfA, fimA, lpfC ou sptP estiveram presentes. Os genes avrA, fliC, invA, iroN, mgtC, sifA, sipB, sipC, sitC ou slyA ou sopB estiveram presentes em 100% das amostras analisadas. Determinou-se quatro perfis genotípicos. No ensaio de patogenicidade observou-se que as amostras inoculadas por via oral, não causaram mortalidade de pintinhos SPF de um dia de idade, com a exceção da amostra SA 633 e SA 006 que apresentaram 30 e 10%, respectivamente. No entanto, observou-se que a infecção por via subcutânea provocou uma maior mortalidade de pintinhos, sendo que as amostras SA 003, SA 004, SA 005 e a amostra vacinal mostraram somente 10% de mortalidade, a amostra SA 002 30%, as amostras SA 632 e SA 634 70% e a amostra SA 633 100%. A amostra SA 006 não provocou nenhuma mortalidade. A amostra mais patogênica pela via subcutânea foi a SA 633. O ensaio de colonização foi realizado em pintinhos SPF, com as amostras SA 002, SA 003, SA 004, SA 005, SA 006, SA 632, SA 633, SA 634 e amostra vacinal. Verificou-se ausência de invasão no fígado e baço 24 horas pós- infecção, exceto para as amostras SA 632 (30%) e amostra vacinal (20%). Após sete dias da infecção houve invasão em dois pintinhos com a amostra SA 002 e um pintinho com as amostras SA 004 e SA 005. Apenas na amostra SA 632 constatou-se colonização em ceco após 24 horas em 10% das amostras e após 7 dias em 70% pós-infecção. Concluiu-se que entre as amostras de Salmonella Typhimurium analisadas existiam diferentes perfis genotípicos baseando-se na presença ou ausência de genes de virulência, e que a amostra vacinal assemelha-se a amostras de S. Typhimurium estudadas quanto a presença dos genes. Os resultados do teste de patogenicidade das amostras de ST indicaram que a via de inoculação modifica a patogenicidade de uma mesma amostra e que a mortalidade após a inoculação pela via subcutânea é sempre superior que pela via oral.

▼ The principal objective of this investigation was to evaluate the effects of mycotoxin contaminated diets (deoxynivalenol (DON)), aflatoxin B1 (AFB1), and fumonisin (FUM)), with or without the use of a commercially available deactivating compound (DC), in young pullets and replacement laying hens on performance, reproductive, serological, and histopathological parameters. In trial 1, experimental treatments consisted of control, low toxin (1 µg DON/g + 1 µg AFB1/g), and high toxin (2 µg DON/g + 2 µg AFB1/g) diets. Pullets fed the high toxin diet had reduced (P<0.05) body weights compared to control and low toxin diets at d 14, 35, 49, 56, and 63. At d 21 and 28, there was a significant interaction observed between mycotoxin and DC inclusion in body weights. Following necropsies (d 35 and 65), relative liver weights and histopathological liver tissue damage were increased (P<0.05) in pullets fed high toxin diets when compared to control and low toxin diets. Relative kidney weights were increased (P<0.05) due to high toxin diet at d 65. Expected negative effects of toxin administration on titer development were not observed. The only interaction observed between mycotoxin administration and DC inclusion in trial 1 was on body weights. In trial 2, experimental treatments consisted of control, DON (9 µg/g) challenge, AFB1 (2 µg/g) + FUM (54 µg/g) challenge, and a mixed challenge (6 µg DON/g, 1 µg AFB1/g, and 27 µg FUM/g). All mycotoxin diets fed to hens negatively influenced (P<0.05) feed efficiency for the trial period spanning weeks 6 through 10 when compared to control diets. Egg production was not affected (P>0.05) by all mycotoxin diets weeks 6 through 10. Relative weights of the liver and kidney were increased (P<0.05) by AFB1+FUM challenge weeks 4 and 9 compared to control diet. The data reported in this study demonstrate that dietary DON and / or AFB1+FUM influence some performance, reproductive, histopathological, and egg quality traits, but by and large, replacement layer pullets seem to be relatively resistant to the mycotoxins evaluated in this trial at the described levels of administration. ▼ Appetite regulation is important for survival across all vertebrate species and the hypothalamus is the regulatory center for control of feeding behavior. Thus, studying the functions of the hypothalamus on appetite regulation provide novel insight into the eating disorders, such as obesity and anorexia, a worldwide health issue. Also, such information is relevant for improving productivity in the modern chicken industry. The objective of this dissertation research was to determine the hypothalamic mechanisms underlying appetite regulation in birds. In Experiment 1, the Virginia lines of chickens were used to elucidate the mechanisms underlying stress-induced anorexia. These chickens have been selected for low (LWS) or high (HWS) body weight at 56 days of age and have different severities of anorexia and obesity, respectively. Chicks were subjected to a combination of thermal and nutritional stress after hatch. The results suggested the two lines displayed distinct appetite-associated gene expression profiles in response to stress in the hypothalamus. In particular, stress-induced anorexia in the LWS may result from potent feeding-inhibitory factor corticotropin-releasing factor (CRF). Thus, in Experiments 2 and 3, we attempted to determine the mechanisms of CRF's inhibitory effect on food intake in chickens and Japanese quail. We administered CRF by intracerebroventricular (ICV) injection and the hypothalamus was collected 1 hour later for molecular analyses. Results showed that CRF exerted a similar inhibitory effect on food intake in these two bird species. However, the inhibitory effect of CRF was primarily associated with a dampened neuropeptide Y (NPY) system which is a potent stimulatory factor for feeding behavior in chickens, whereas it may involve activated CRF and melanocortin systems in quail. In Experiments 4 and 5, we used the same experimental design as for CRF studies to determine the hypothalamic mechanisms of the inhibitory effects of neuropeptide K (NPK) and adrenomedullin (AM) in Japanese quail. Results from Experiment 4 showed that the feeding-inhibitory effect of NPK was associated with a group of increased feeding-inhibitory factors such as CRF and cocaine and amphetamine-regulated transcript (CART) and decreased feeding-stimulatory factors, such as NPY and agouti-related peptide (AgRP) in the hypothalamus. In Experiment 5, AM increased gene expression of CART and proopiomelanocortin (POMC). Overall, these experiments suggested the roles of the hypothalamus in stress or exogenous neuropeptide-induced anorexia in birds and may provide insights on understanding appetite regulation from evolutionary, agricultural, and biomedical perspectives. Destaca-se a Salmonella como um dos mais importantes patógenos veiculados por alimentos, devido ao fato de estar amplamente distribuído na natureza, possuir um grande número de reservatórios, apresentar sorotipos inespecíficos quanto aos hospedeiros e apresentar cepas multiresistentes aos antimicrobianos. Salmonella Enteritidis tornou-se um grande problema de saúde pública no Brasil, a partir da década de 80, quando foi associada a produtos avícolas. Desde 1962, a Salmonella Heidelberg tem sido isolada de aves e produtos derivados e aves no Brasil e tem reconhecida importância para saúde humana e em poedeiras comerciais na América do Norte. O presente trabalho teve por objetivo o estudo de diferentes cepas de Salmonella Heidelberg (SH) em comparação com Salmonella Enteritidis (SE) em pintinhos e frangos de corte no que diz respeito à capacidade de invasibilidade de órgãos, persistência de excreção fecal e alterações na morfometria intestinal. Para o estudo in vivo, 65 isolados de SH foram avaliados quanto à presença dos genes de virulência invA, lpfA e agfA e diferenciados por eletroforese em campo pulsado (PFGE). Outro objetivo foi testar uma mistura de ácidos orgânicos e óleos essenciais no controle de excreção fecal de SE. Duas amostras de SH não relacionadas pelo método de PFGE (SH23 e SH35) e uma cepa de SE padrão foram selecionadas para a inoculação das aves. As três cepas promoveram alteração na medida de vilosidades e criptas dos pintinhos, frente a grupo controle. A cepa SH35 promoveu maior reação inflamatória nos cecos. Ambas as cepas testadas foram capaz de atingir o fígado de pintinhos às 6 horas pós-inoculação. Tanto a SE como a SH demonstram persistência e intermitência de excreção fecal em frangos de corte até os 21dias pós-inoculação. A mistura de ácidos e óleos essenciais foi eficaz na redução da excreção fecal de SE, porém somente com o uso contínuo da mistura na ração. As cepas apresentaram comportamentos diferentes quando inoculadas em frangos de corte e devido à persistência destas nos intestinos dos frangos, prejuízo para as aves podem ocorrer, enquanto a subseqüente contaminação de carcaças, representa risco para a saúde humana.

Salmonella, one of the most important food borne pathogen due to the widely distribution in nature, large number of reservoirs, to show unspecific host serotypes and provide multiresistant strains to antibiotics. Salmonella Enteritidis became a big public health problem in Brasil, since 80’s, when it was correlated with poultry products. Since 1962, Salmonella Heidelberg has been isolated from birds and poultry products in Brazil, with importance in human health and layers in North America. A study was carried out to evaluate the use of Saccharomyces cerevisiae cell wall (SCCW) as an anti-mycotoxin additive in broiler chicks diets slaughtered at 22 day age on performance, carcass characteristics and organ weights. It was used a randomized complete block design with four treatments in a factorial 2 (with aflatoxin, without aflatoxin) x 2 (with adsorbent, without adsorbent). For the performance parameters evaluated, there was a significant difference only on feed intake, when the SCCW was added to the rations. There was no interaction between aflatoxin and SCCW factors in the diet on the performance variables studied. The addition of SCCW to the chicks diets stimulated the feed intake during 1-21 days of age. However, that did not reflect a significant increase in weight gain, or a change in feed conversion. There was a significant interaction between factors aflatoxin and SCCW on the weight post-fasting, cold carcass weight and thigh+upper weight. For the absolute hot carcass weights, breast, back and wings and carcass yields and relative weights of the cuts it was not observed a significant interaction between factors. The growth performance of broilers in the period 1-21 days old was not influenced by the levels of aflatoxin and SCCW factors used in experimental diets. Nevertheless for body weight after fasting, the cold carcass weight and absolute weight of the thigh + drumstick, the SCCW acted as an anti-mycotoxin additive by minimizing the aflatoxin toxicity.

Objetivou-se com este experimento avaliar a utilização da parede celular de Saccharomyces cerevisiae (PCSC) como aditivo antimicotoxina em rações destinadas a frangos de corte abatidos aos 22 dias de idade sobre o desempenho, características de carcaça e pesos de órgãos. Foi adotado um delineamento experimental em blocos casualizados, com 4 tratamentos dispostos em um esquema fatorial 2 (sem aflatoxina; com aflatoxina) x 2 (sem adsorvente; com adsorvente). Para os parâmetros de desempenho avaliados, observou-se diferença significativa apenas sobre o consumo alimentar, quando a PCSC foi adicionada às rações. Não houve interação entre os fatores aflatoxina e PCSC nas rações sobre as variáveis de desempenho estudadas. A adição de PCSC à ração dos frangos estimulou o consumo de ração no período de 1 a 21 dias de idade, entretanto, isso não se refletiu em aumento significativo no ganho de peso dos frangos ou alteração na conversão alimentar. Houve interação significativa entre os fatores aflatoxina e PCSC sobre o peso vivo pós-jejum, carcaça resfriada e o peso da coxa+sobrecoxa. Para os pesos absolutos de carcaça quente, peito, dorso e asas, rendimento das carcaças e pesos relativos dos cortes não se observou interação significativa entre os fatores estudados. O desempenho zootécnico de frangos de corte no período de 1 a 21 dias de idade não foi influenciado pelos níveis dos fatores aflatoxina e PCSC utilizados nas rações experimentais. ▼ At hatch, the skeleton of the chick is a well formed miniature of that in the adult bird; this is the end result of 21 days of development in the egg. In order to build a strong and healthy skeletal frame, the embryo relies on trace minerals (TM) deposited in the egg by the hen. The temperature at which the embryo grows is also a key factor influencing skeletal development. The overall purpose of this research was to understand the effects of maternal trace mineral nutrition on embryonic and post-hatch bone development and to investigate the relationship between temperature and bone characteristics. Mineral content in the egg and embryonic and post-hatch bone characteristics of embryos and chicks from Young (32 week), Mid (45 week) and Old (59 week) hens were not influenced when hens were supplemented with low levels of organic (OTM) copper, zinc and manganese relative to TM sulfates (ITM) at industry levels (Control). High ITM levels increased bone strength at hatch relative to Control but not relative to OTM; at hatch OTM widen bones from Young hens relative to all diets. Therefore, an opportunity exists for industry to reduce TM levels by supplementing OTM. As hens aged, the yolk Zn and Cu content increased and embryos from Young hens had reduced proportion of calcified tibia and femur relative to those from Older hens at day 20th of incubation and weaker bones at hatch. In another study, an incubator temperature of 36.0⁰C applied from the 15th day of incubation until hatch increased bone strength relative to 37⁰C. High eggshell temperature is negatively associated with bone calcification and strength. If the stronger bones at placement in the barn increased chick mobility then water consumption and access to nutrients important for post-hatch bone growth could be increased and this might decrease future bone problems. In summary, considerable maternal age and incubator temperature variation existed on skeletal growth of the progeny, demonstrating that there may be opportunities to use maternal nutrition and hatchery management to increase skeletal health in chicks at hatch, especially those from young flocks.
