O Herpesvírus canino tipo 1 (CaHV-1) está associado a desordens reprodutivas e mortalidade neonatal, podendo ser encontrado em cães saudáveis assintomáticos ou associado a manifestações clínicas variadas como lesões vesiculares genitais, infecções oculares e doença respiratória. O objetivo deste estudo foi detectar a presença do CaHV-1, através da reação em cadeia de polimerase (PRC), em amostras de urina e em swab ocular e vaginal de cadelas na cidade de Uberlândia, Minas Gerais, Brasil. Foram utilizadas 20 cadelas com histórico ou sinais de desordens reprodutivas. Dois animais apresentaram resultado positivo na PCR (10%) nas amostras de urina e secreção ocular, respectivamente, um diagnosticado com piometra e outro saudável e histórico de natimortalidade. Todas as amostras de secreção vaginal foram negativas. No sequenciamento de DNA as amostras positivas apresentaram 100% de similaridade de nucleotídeos com outras quatro cepas de CaHV-1 já descritas e depositadas no Genbank. Os cães positivos na PCR no presente estudo provavelmente eram portadores de infecção latente. A detecção do DNA viral nas amostras avaliadas confirma a presença do CaHV-1 na população canina do município de Uberlândia. Trata-se da primeira detecção molecular do agente na região sudeste do Brasil.

Canine herpesvirus-1 (CaHV-1) is known to cause reproductive disorders and fatal infections in puppies, and may also be found in asymptomatic dogs or related to vaginal vesicular lesions, ocular infections and respiratory diseases. The aim of this study was investigate the ocurrence of CaHV-1 using the polymerase chain reaction (PCR) within urine samples, ocular and vaginal swab samples from Uberlândia, MG, Brazil. Twenty mature bitches, with at least one estrous and reproductive disorders history and/or symptoms were used. Two out of twenty dogs (10%) were positive in the urine and ocular swab samples, respectively, one with pyometra diagnosis and another one healthy with history of stillbirth. No positive results were detected within vaginal swab samples. Sequence analysis of the DNA polymerase gene of the positive samples indicated 100% identity with the sequence of the four CaHV-1 strains selected from Genbank. These findings suggest that positive dogs might be CaHV-1 latent carriers. Additionally, the results confirm the presence of CaHV-1 circulating within urban canine population of Uberlândia, MG. This is the first report that a CaHV-1 infection has been detected in the Southeast Brazil.

▼ Infectious disease transmission requires that epidemiologically relevant contact occurs between infectious and susceptible individuals. Thus, for mathematical models to accurately predict disease emergence and dynamics they must incorporate the contact patterns responsible for transmission. In this context, this thesis investigates how the level of contact detail included in an infectious disease model influences its predictions. Three models are considered. The first investigates infections spreading through territorial populations, with potential canine rabies spread in Australian wild dogs a case study. Two factors governing wild dog contacts are considered: geographic distance and heterogeneous wild dog behaviour. Not including spatial constraints results in a model that overestimates the probability of an epidemic and that fails to generate the outcome 'rate of spread'. Conversely, not incorporating heterogeneous dog behaviour results in a model that underestimates the probability an epidemic will occur. The second model investigates tick-borne pathogen spread between ticks and vertebrate hosts. Key features of tick feeding behaviour include: tick aggregation on hosts, co-aggregation of larval and nymphal ticks on the same hosts, and co-feeding. Co-aggregation increases R0. Models failing to incorporate tick co-aggregation will therefore underestimate the likelihood of pathogen emergence, especially in geographic regions and seasons where larval burden is high and for pathogens mainly transmitted during co-feeding. The third model investigates the effect of clustering (triangle and square contact patterns) on the spread of infection through social networks. Clustering reduces R0 and the magnitude of the reduction increases with higher transmission probabilities. Models that fail to incorporate clustering will overestimate the likelihood of disease establishment, especially for highly transmissible diseases. In conclusion, the three disease models collectively reveal model predictions are improved and additional outcomes are generated by the inclusion of realistic host contact patterns. These findings reinforce the value of incorporating biologically-faithful contact patterns into infectious disease models.

Si tratta di un problema multifattoriale che dipende da diversi aspetti della funzione riproduttiva. La qualità dell'ovocita, determinata durante la follicologenesi, è sicuramente cruciale per ottenere risultati riproduttivi ottimali. Per migliorare la fertilità femminile nel bovino è fondamentale definire i fattori e i meccanismi che determinano la qualità del gamete femminile, in quanto il declino della funzione riproduttiva in questa specie è dovuto principalmente alla scarsa qualità degli ovociti. In particolare, è importante conoscere quali proteine regolano la meiosi, la fecondabilità e lo sviluppo embrionale dell'ovocita. In questo progetto, abbiamo considerato principalmente il ruolo del Progesterone Receptor Membrane Component-1 (PGRMC1) sia nell'ovocita che nel compartimento follicolare, considerato che il dialogo tra compartimento germinale e compartimento somatico è uno dei fattori chiave coinvolti nell'acquisizione della competenza allo sviluppo del gamete femminile. Il PGRMC1 è una proteina multifunzionale ed altamente conservata in specie anche filogeneticamente distanti. Nei mammiferi è espressa in diversi sistemi, compresi gli organi riproduttivi, e localizza in diversi compartimenti subcellulari. Tuttavia, il suo ruolo e il preciso meccanismo d'azione in ogni sistema e/o compartimento subcellulare non sono ancora del tutto conosciuti. La parte più considerevole di questo progetto di dottorato riguarda gli studi effettuati sul ruolo del PGRMC1 nella divisione cellulare, data la sua localizzazione a livello di fuso mitotico e meiotico. Abbiamo dimostrato che la deplezione del PGRMC1 determina difetti nella divisione cellulare e la sua interazione con l'Aurora chinasi B (AURKB) indica che potrebbe svolgere la sua azione durante la citodieresi, l'ultima fase della divisione. Durante la maturazione meiotica, abbiamo valutato anche il suo ruolo nel mediare l'azione del progesterone (P4) confrontando il suo effetto con quello del recettore nucleare del progesterone (nPGR). L'inibizione di entrambi i tipi di recettore determina lo stesso effetto sull'organizzazione della piastra metafasica e sulla competenza allo sviluppo dell'ovocita, ma in fasi differenti. Inoltre, abbiamo testato l'ipotesi che il PGRMC1 possa modulare la funzione del nucleolo. Studi di immunofluorescenza hanno confermato la presenza del PGRMC1 nel nucleolo di cellule della granulosa (bGC) e negli ovociti di bovino, nonchè la colocalizzazione con la nucleolina, la proteina nucleolare più abbondante che svolge importanti funzioni in questo compartimento subcellulare. Inoltre, la down-regolazione del PGRMC1 determina uno spostamento della nucleolina dal nucleolo al nucleoplasma suggerendo un'associazione funzionale tra queste due proteine. ▼ There are several central and peripheral mechanisms that influence the GnRH secretion. For a long time it has been a mystery how these mechanisms influence the GnRH neurons. In recent years there have been exciting new discoveries in this field, especially with the discovery of kisspeptin and its receptor. Kisspeptin is a neuropeptide that seems to have an important role in the regulation of GnRH neurons in the hypothalamus in various mammalian species. It is, among other things, supposed to be involved in the onset of puberty and in the mediation of the positive feedback exhibited by estrogens on the GnRH neurons in females, which leads to the GnRH/LH surge. Although a lot of research has already been done, to our best knowledge, kisspeptin and its receptor have not yet been researched in the dog. Because the kisspeptin signaling has been called a major gatekeeper of the gonadotropic axis, it would be interesting to discover its role in the canine reproduction. To lessen the amount of dogs that are yearly euthanized all over the world because of overpopulation, it is important to find a non-surgical method of contraception that prevents estrous cyclicity. The kisspeptin signaling pathway could be a suitable target for developing an effective, safe, long-term or permanent non-surgical contraceptive for the bitch. The aims of the present study were to identify the canine KISS1 and KISS1R genes and to characterize the amino acid sequences of canine kisspeptin and the kisspeptin receptor. The identification and characterization were done in silico. The results show that the KiSS1 and the KiSS1R gene are present at the canine genome and a large homology is found between the canine peptides and the peptides found in other mammalian species. This gives a basis to believe that kisspeptin signaling is also important in GnRH secretion in the dog and that it may indeed be a suitable target for a non-surgical contraceptive. ▼ In vitro maturation of dog oocytes has always been the main obstacle preventing reproductive biologist from producing canine in vitro cultured embryos. The unsuccessful oocyte maturation in canine species originates from their unique physiological and biological specifications. Ovulation of dominant follicles in bitch (6-12 in each oestrous cycle) occurs at prophase I stage of oocyte nucleus and meiotic resumption develops during 3-5 days of oviductal transition. During this PhD thesis, studies were designed in order to speculate characteristics of canine oocyte maturation in vitro in terms of maturation media components, gas composition of the incubator and hormonal requirements. Level of oxidative stress during 72h (culture period) of in vitro maturation showed that 5%O2, 5% CO2 and 90% N2 composition improves meiotic resumption and reduces degeneration rate significantly compared to 5% CO2 in air. Utilization of caffeine as a non specific phosphodiesterase inhibitor at 10mM for 12h at the beginning of the 72h culture (12+60) also improved MII maturation rate (16.9% ± 2.4; P < 0.05). Among several hormonal treatments recombinant porcine Growth Hormone (PGH) at 100ng/ml and Melatonin (MTN) at 100nM concentrations had outstanding improvement over meiotic resumption (28.9% ±10.0 and 56.2% ±8.6 respectively; P < 0.05). Attempts were made to study developmental potentials of optimally matured oocytes by parthenogenetic activation (PA) and in vitro fertilization (IVF) using chilled semen. Partial digestion of the zona pellucida prior to IVF improved the cleavage rate at 48h 6.4% ± 0.3 and resulted in production of a single 8 cell embryo. Moreover; canine follicular cells were culture in order to characterize their primary culture morphology and steroidogenic responsiveness to physiological and pharmaceutical substances. Immunolocalization of aromatase (CYP19) positive cell clumps, presumptive oestrogen producing colonies, was identified. This primary culture also maintained its steroidogenic machinery up to 96h (measured by radioimmunoassay) with a significant increase in production of estradiol and progesterone after 72h compare to the start of the culture.

Koirilla esiintyvistä lisääntymisongelmia aiheuttavista infektiivisistä mikrobeista tyypillisimmät ovat Brucella canis ja canine herpes virus-1 (CHV-1). Näiden aiheuttamat ongelmat saattavat muodostua hyvinkin merkittäviksi yksittäisessä kennelissä alentaen syntyvien pentujen määrää huomattavasti vuositasolla. Useissa maissa tehtyjen tutkimusten perusteella on todettu herpeksen olevan enzoottinen virus koirapopulaatioissa. Brucella canista esiintyy harvemmin, mutta sen aiheuttamien vakavien lisääntymisongelmien takia vähäinenkin esiintyvyys on merkittävää. Suomessa ei ole aiemmin tutkittu Brucella canis –bakteerin tai CHV-1:n esiintyvyyttä paikallisessa koirapopulaatiossa. Tutkimuksen tarkoituksena oli kartoittaa näiden mikrobien esiintyvyys Suomessa ja niiden merkitys suomalaisten koirien lisääntymisongelmissa. Tutkimukseen pyydettiin lehti-ilmoituksella mukaan jalostuskoiria, joilla oli ollut lisääntymisongelmia. Näytteitä otettiin myös täysin terveistä koirista verrokkiryhmäksi. Näytteeksi otettiin verta lasiseen seerumiputkeen noin 8 ml. Näytteet kerättiin pääsääntöisesti kenneleissä paikan päällä. Yhteensä näytteitä saatiin 388 kpl. Brucella canis –vasta-aineet tutkittiin kaikista näytteistä EELA:ssa. Yhdestäkään näytteestä ei löytynyt positiiviseksi tulkittavia vasta-ainemääriä. Herpesvasta-ainetutkimusta varten valittiin 40 lisääntymisongelmista kärsivien kenneleiden näytettä sekä 41 näytettä verrokkiryhmäksi ongelmattomista kenneleistä. Herpesvasta-aineet tutkittiin Ruotsissa SVA:n laboratoriossa. Herpesvasta-aineita tutkimuksessa todettiin usealla koiralla. Seroprevalenssi ongelmakenneleiden ryhmässä oli 90%. Verrokkiryhmän kenneleissä seroprevalenssi oli selvästi matalampi, 24%. Vasta-ainetiitterin korkeus todettiin tilastollisesti merkitsevästi riippuvaksi kennelin ongelmastatuksesta ja koiran käytöstä jalostuksessa. Herpesvasta-aineita todettiin enemmän ongelmakennelien koirilla kuin ongelmattomien kenneleiden koirilla. Tutkimuksen johtopäätöksenä voidaan todeta herpeksen olevan yksi tärkeimmistä aiheuttajista kennelin lisääntymisongelmissa. Kennelin herpestilanteen selvittäminen tulisikin sisällyttää tutkimuksiin kartoitettaessa syitä kennelin lisääntymisongelmiin.
