▼ Between Nov. 2013 and Apr.2014, a serological study was conducted on small ruminants to determine the prevalence of brucellosis, identify risk factors and its public health implication. A total of 1190 blood samples collected from sheep and goat populations in Chifra and Ewa woredas (876 caprine and 314 ovine), 155 (13%) tested positive using mRBPT. Further testing of the positive reactors with CFT confirmed that 147(12.35%) samples were positive for brucella infection. The results revealed that among the risk factors considered in the analysis, species, sex, age, parity number and flock size had statistically significant effect on seropositivity (p<0.05). Woreda had no statistically significant effect on seropositivity (p>0.05). The individual animal level seroprevalence was significantly higher in goats (14.4%) than in sheep (6.7%) populations. The analysis revealed that goats were more than 2 times (OR=2.34) at risk of getting infected with brucella than sheep. The seroprevalence was also significantly higher in female animals (13.8%) than in males (6.5%) with the likelihood of female animals to get infected with brucellosis is higher than that of males. Similarly, Brucella seroprevalence was also significantly increased with age; the odds of seropositivity in older animals are 2.36 times higher than that of younger animals. Seroprevalence rate of brucellosis was also significantly varied with parity, with the likelihood of infection being 2.038 times higher in animals with 3 to 4 parities than otherwise. Regarding flock size individual animal sero-prevalence was higher in larger flocks than in the smaller ones (OR=0.68). Almost all the respondents in the studied areas were not aware of brucellosis as a disease affecting different species of livestock (91.1%) although all of them recognized the existence of abortion among small ruminant flocks and most of them handle abortion materials with bare hand without protecting themselves (82.2%). The habit of drinking raw milk is practiced by the majority of the respondents 45 (100%) while there is no habit of consuming raw meat. Poor awareness of the zoonotic importance of brucellosis and the practices of consuming raw milk and handling potentially infectious materials using bare hands pose a serious danger to small ruminant owners. Finally, this study demonstrated that small ruminant brucellosis is highly prevalent in the study districts. Hence, it is suggested that there is a need for planning and implementation of joint programs by stakeholders in prevention and control of the disease as well as raising public health awareness. ▼ Brucellosis is a highly contagious zoonotic and devastating disease that affects households‟ potential to improve their well-being through trade in livestock and livestock commodities. Despite the disease being endemic in Zambia, there is inadequate information, on its socio-economic impact on the well-being of households rearing cattle. Therefore, a cross-sectional study was conducted in the Western and Southern provinces of Zambia to determine the impact of brucellosis on socio-economic wellbeing of cattle farmers at household level. The specific objectives of the study were to determine losses and costs associated with brucellosis in cattle; determine socioeconomic well-being levels at the household; evaluate the linkages between brucellosis and socio-economic well-being status; assess the extent to which the current health policies are implemented to support One Health approaches in brucellosis control; develop a framework for collaboration between veterinary and medical experts in controlling brucellosis and other zoonoses. Structured questionnaires, focus group discussions and key informant interviews were used to collect both quantitative and qualitative data in this research. Furthermore, both descriptive and inferential statistics were used to analyse quantitative data using IBM-SPSS 22® while qualitative data was analysed using Nvivo® and content analysis. All costs and monetary losses were estimated in Zambian Kwacha (ZMW) and the equivalent international currency (USD); the exchange rate was 1 USD to 11.45 ZMW. The overall total loss attributed to brucellosis-related calf mortality, in the studied households was 1,536,000 ZMW equivalent to USD 134,148.47; 77,700 ZMW equivalent to USD 6,786.02 was due to milk losses, 18,000 ZMW equivalent to USD 1,572.05 was due to mortality among seropositive cattle and 52,960 ZMW equivalent to USD 4,626.37 was due to veterinary interventions. Lack of money to pay for livestock health services was significantly associated with poor household socio-economic well-being (p = 0.003), while the level of education of household heads was associated with the highest positive brucellosis impact (p = 0.005) on household socio-economic well-being. Further, the results also show that there was a big difference in the levels of socio-economic well-being in the study areas with, 59.0% of households being classified as poor and only 2.0% of households were classified as rich. Consequently, the null hypothesis was rejected in favour of the alternative hypothesis, which stated that socio-economic well-being levels differ significantly where there is less impact of brucellosis in cattle and where such impact is higher(F = 11.268, p = <0.001). It was also found that more than two-thirds of the respondents from the Ministry of Health (MoH) and Ministry of Fisheries and Livestock (MFL) (73.1%) agreed that there was no policy support which directly facilitated One Health in terms of collaboration. ▼ Infectious diseases are an important consideration in the management of wildlife populations, and contact rate is a key parameter for understanding the epidemiology of such diseases. In the first section of this thesis, I review current issues and challenges that researchers face when designing animal contact studies and analyzing contact data. I examine how characteristics of methods for collecting contact data affect inferences that can be drawn about contact structures; describe applications of social network analysis of contact data to disease ecology and animal behavior, focusing on sampling issues and dynamic networks; suggest how new technologies can be used to answer important questions about variation in individual contact rates within populations; and propose a new statistical approach for analyzing contact data in a linear modeling framework. In the second section, I describe an experimental field study that used proximity loggers (a new technology for measuring contact rates) to understand transmission of Brucella abortus on elk feedgrounds in Wyoming. Brucellosis is a bacterial disease that causes abortions in elk and is transmitted by contact with infectious aborted fetuses. Supplemental feeding of elk on winter feedgrounds is believed to exacerbate B. abortus transmission by aggregating elk at high densities, increasing their chance of contacting infectious fetuses. I evaluated the effectiveness of a proposed low-density feeding strategy by comparing elk-fetus contact rates (as measured by proximity collars and video cameras) during high-density and low-density feeding treatments that provided the same total amount of food at different densities. Low-density feeding led to >50 percent reductions in the total number of contacts and the number of individuals contacting a fetus. Elk contacted fetuses and random control points equally, suggesting that elk were not attracted to fetuses but encountered them incidentally while feeding. The relationship between contact rate and disease prevalence is non-linear and simple disease models suggest that low-density feeding may result in dramatic reductions in brucellosis prevalence, though this depends on the amount of transmission that occurs on and off feedgrounds. ▼ Brucellosis is endemic in many livestock worldwide especially developing countries. The aims of this study were to estimate the seroprevalence of bovine brucellosis and risk factors associated with the seropositivity in rural and peri-urban buffaloes and cattle populations of Sindh. Firstly, a cross sectional study was conducted to estimate the seroprevalence of bovine brucellosis in cattle and buffaloes of Sindh province, Pakistan. Serum samples (2600) were tested using Rose Bengal Plate Test. The overall seroprevalence of brucellosis in Sindh province was 13.96% (95% C.I.; 11.55 - 16.37). Of the 917 herds tested, 232 or 25.30% herds (95%C.I.; 22.51-28.24) were positive for brucellosis. The adult animals were 2.05 (95% C.I.; 1.14-3.68, P= 0.02) times more likely to test positive for brucellosis. The animals in a peri-urban dairy production system were 2.07 times (95%C.I.; 1.09-3.90, P = 0.03) times more likely have brucellosis. The species or sex of animal did not appear to affect the risk of seropositivity in cattle or buffalo in this population. Secondly, a cross sectional survey was conducted to understand the structure and composition of farms, animal husbandry and management practices in peri-urban dairy colonies in Karachi and farmers’ awareness of zoonoses. The mean herd size was 93.58 animals and 88.01% of these animals were female buffaloes. Of 326 farms surveyed, only 37.42% were able to associate animals with transmission of diseases in human. The characteristics of peri-urban dairy farms in Karachi are discussed. Thirdly, the value of FTA® cards in detecting the Brucella DNA in milk samples was estimated by determining the detection limits of genus specific ERI PCR assay for FTA® cards and comparing the PCR results from whole sediments taken from culturing pooled milk samples with taking sediment on FTA® cards. The detection limits of this method ranged from 6.6 x 103 cfu/ml for B. abortus to 7.17 x 106 cfu/ml for B. suis. Assuming the results of ERI PCR for the whole sediment as gold standard (method 1), the method using sediment on FTA® cards as test samples (method 2) showed a diagnostic sensitivity of 81.44% (95% C.I.; 75.54-87.33) but a poor diagnostic specificity of 42.86% (95%C.I.; 16.95-68.78). The kappa value, κ, was 0.14 (p = 0.02) demonstrating a poor agreement between the two methods. Lastly, 181 bulk milk samples were used to estimate the herd level prevalence of bovine brucellosis in Landhi dairy colony, Karachi. The ERI PCR was used to test these samples. The herd prevalence was estimated as 92.26% (95% C.I.; 88.34-96.19). For each level (50 animals) increase in herd size, the risk of herd being brucellosis positive increases by 2.38 times. The herds that have a male animal for breeding are 0.09 times less likely to have brucellosis. ▼ A cross-sectional study was conducted in Lusaka Province of Zambia to estimate brucellosis seroprevalence and identify associated risk factors. This study was done between January 2007 and February 2008 in commercial cattle farms in 3 districts of Lusaka Province (Kafue, Chongwe and Lusaka districts) and one district in Central Province(Chibombo). Sera were collected from animals randomly selected from sampling herds and at the same time data such as sex, age and parity were also recorded. The collected sera were screened for Brucella antibodies using the Rose Bengal test (RBT) followed by confirmation on Competitive Enzyme Linked immunosorbent Assay (C-ELISA). Results were interpreted both in series and parallel. The risk factor identification was done using a pre-tested questionnaire which was administered simultaneously with the blood sampling. The factors assessed included types of cattle breeds, farm ownership, care responsibilities, marketing, feeding practices, herd size, stock density, disease control and proximity to other herds. Prevalence estimates, including the 95% confidence interval, were determined for each district and also for Lusaka province as a whole. The Fisher’s exact test was used to test for associations between Brucella positivity on the farm and the hypothesised risk factors. A total of 897 serum samples from Lusaka province (n=849) and Chibombo district (n=48) were screened. The estimated overall seroprevalence in Lusaka province was 9.4% (95% CI: 5.0% to 13.6%) while that for Chibombo district was 18.7% (95% CI: 7.5% to 29.9%) based on serial interpretation. The overall herd level prevalence for Lusaka Province was 40.6% (95% CI: 16.4% to 67.9 %) while that for Chibombo was 100%. The prevalences according to sex were oxen 0%; cows 8.1% (95% CI: 4.6% to 11.6%) and bulls 12.5% (95% CI: 3.8% to 21.1%). Prevalences according to age groups were 1 to 4 years 10.7% (95% CI: 4.9% to 16.9%); 4.5 to 5 years 4.2 % (95% CI: 0.0 to 7.8%); 5.5 to 7 years 6% (95% CI: 2.0% to 9.9%) and more than 7 years 9.9% (95% CI: 2.3% to 17.5%). At the animal level, seroprevalence varied according to sex with bulls having slightly higher odds of being seropositive (OR=1.7) compared to females, according to age groups, with the age category 1 to 4 years recording the highest seroprevalence. Chongwe district had a significantly higher prevalence (p < 0.001) than all the other districts in Lusaka province. A univariate analysis of risk factors showed that the method of acquiring a farm was the most important risk factor. However, other factor such as source of animals was significant at 95% confidence level. ▼ Brucellosis is a constraint to livestock production in Moçambique. Reproductive failure due to brucellosis has been reported in the country and abortions occur in cattle, sheep and goats on many of the farms in Maputo Province of Moçambique. Brucellosis is an infectious disease affecting mainly sexually mature animals and is caused by organisms of the genus Brucella. Brucella abortus is the species that mainly affects cattle throughout the world, while Brucella melitensis affects mainly goats and sheep. Control of bovine brucellosis using B. abortus S19 vaccine was undertaken before the civil war in Moçambique which started in 1978 and ended in 1992. During this period the vaccination status of animals was not known. The diagnosis of brucellosis is mainly based on serological surveys and conflicting results obtained previously emphasized the need for additional studies. The aims of this research were to determine the prevalence of brucellosis in cattle, goats and sheep due to B. abortus and B. melitensis in five districts of Maputo Province based on serology, and to assess the relationship between abortions and reproductive failure occurring in the study area by isolation of Brucella organisms or nucleic acid detection. A total of 971 adult beef and dairy cattle, 752 goats and 260 sheep from the study area were tested for Brucella antibodies using the Rose Bengal test (RBT), indirect ELISA and complement fixation test (CFT). Milk from 85 dairy cows and tissue samples from 32 cows and bulls from Gaza and Inhambane provinces slaughtered in Maputo municipal abattoir and Machava abattoir, and an aborted foetus were also examined. All serological test results for small stock were negative. An overall apparent prevalence of 14.0% (n=971) was found in beef and dairy cattle in the study area based on a 95% confidence interval. The highest prevalence was observed in Manhiça District (27.4% (n=180)) with a significant difference compared to other districts (p<0.05). The lowest prevalence was observed in Magude District (7.1% (n=241)). The prevalence in the remaining districts were 14.5% (n=138) in Matutuine, 8.7% (n=173) in Moamba and 14.6% (n=239) in Boane. Apparent prevalences of 15.5% (n=161) in small, 14.0% (n=578) in medium and 12.9% (n=232) in large livestock production sectors where found in the study area with no significant differences (p>0.05) between them. B. abortus was isolated from the spleen of an aborted foetus. A PCR amplification product of 600bp was generated from this isolate 584/05 and from the organs of five slaughtered cows using primers ISP1 and ISP2 for Brucella genus-specific sequences. The isolate 584/05 was confirmed as a B. abortus field strain when fragments of 498bp and 178bp were generated using a PCR assay for differentiation of B. abortus field strains and vaccine strains 19 and RB51. A correlation was made between the 584/05 isolate and serum from the cow that aborted. The results confirmed that the isolate was not a vaccine strain. ▼ The diagnostic performance of four serological assays for bovine brucellosis in African buffaloes, namely Rose-Bengal test (RBT), complement fixation test (CFT), indirect enzyme-linked immunosorbant assay (iELISA) and fluorescence polarisation assay (FPA) were evaluated and compared in a case-control study. The study followed the OIE assay validation pathway for validation of diagnostic tests applicable to wildlife species where there is a validated test available in a taxonomically closely related species. Two uninfected and four infected herds were recruited and an uninfected composite reference panel of 107 sera and infected composite reference panel of 93 were selected using composite reference standards. Diagnostic sensitivity (DSe) and diagnostic specificity (DSp) were calculated for individual tests and for different combinations of two tests in series and in parallel. Cut-off points were adjusted using receiver operating characteristics (ROC) analysis. Using these cut-off values, the index tests performed as follows: RBT DSe of 98.9% (95% CI 96.83% - 100%) and DSp of 98.1% (95% CI 95.6% - 100%), iELISA (cut-off >40.5%) DSe 98.9% (95% CI 94.2% - 100%) and DSp 100% (95% CI 96.6% - 100%), CFT (cut-off >0 iU/ml) DSe 74.2% (95% CI 64.1% - 82.7%) and DSp 100% (95% CI 96.6% - 100%) and FPA (cut-off >16 mP) DSe 97.9% (95% CI 94.2% - 99.7%) and DSp 100% (95% CI 96.6% - 100%). Based on performance index and area under the ROC curve, the iELISA performed best (198.9% and 1.0), followed closely by the FPA (197.9% and 0.989) and the RBT (197.0%). The CFT s lower performance (174.2%, and 0.871) was due to low DSe. Kappa values for test agreement between the index tests was above 0 for all combinations, and varied from unweighted Kappa of 0.685 (95% CI 0.608 0.762) between FPA and iELISA to 0.26 (0.136-0.383b) between CFT and RBT. Consideration of the indices for positive and negative test agreement between the index tests supported the differential specificity of tests for different immunoglobulin classes and higher in line with the findings in cattle. Positive predictive value in herd C and E were 100% for the iELISA, CFT and FPA, 97.3% in herd C and 98.4% in herd E for the RBT. Negative predictive values in herd C ranged from 89% for the CFT to 99.2% for the RBT and in herd E 73.1% for the CFT to 98.7% for the RBT. Overall repeatability was satisfactory, except for the FPA, which was considered the result of sample quality related to prolonged storage in a freezer. The index tests were all found fit for use to detect or confirm brucellosis in populations and individual animals. The values for DSe and DSp that were estimated will be of use in the interpretation of serological results and determination of diagnostic strategies in different circumstances. ▼ Brucellosis is one of the most common zoonotic diseases worldwide, with endemic disease areas in the Middle East, Mediterranean Basin, Central Asia, Africa, and Central and South America. Disease is caused by various species of the gram negative bacteria Brucella. Infection in humans results primarily from contact with infected livestock or consumption of contaminated livestock products; cattle, small ruminants, and swine are the primary reservoir hosts. Although the Brucella bacterium was discovered over a century ago, control of disease remains a major challenge in many areas worldwide. Research on this pathogen has mostly been conducted in mouse models, which are naturally resistant to infection. Little is known of the immune response of natural ruminant hosts to Brucella infection. Here we report an epidemiological study of brucellosis in Mongolia, as well as an experimental infection study of pregnant goats with two strains of B. melitensis – 16M, a fully virulent strain, and Rev. 1, a reduced virulence vaccine strain. Design of the experimental infection study was influenced by findings from field research in an endemic disease region. The objectives of the experimental challenge study were to characterize clinical disease, shedding, and tissue burdens in infected animals. The cellular immune response was then compared in animals infected with the two B. melitensis strains with the aim of identifying components of the protective response induced by the Rev. 1 vaccine strain and deficits in the immune response elicited by infection with virulent B. melitensis 16M. A fluorescence polarization assay was utilized to identify antibodies in milk samples and estimate the proportion of Brucella positive cattle, yak, and hybrids in three regions of Mongolia. Additionally, prevalence of brucellosis in herd owners was assessed via questionnaire. Information was also collected from herd owners regarding animal husbandry practices and herd health in order to identify individual-and herd-level characteristics that are predictive for brucellosis. The study indicates that brucellosis remains endemic in cattle, yak, and hybrids within Bulgan and Khuvsgul provinces of Mongolia despite a national control program. Herd level prevalence was determined to be 10.4% in the 77 herds tested. High levels of human disease were also reported. Results of the study indicate that the Mongolian brucellosis control program must be critically evaluated if the national goal of obtaining brucellosis-free status by 2021 is to be realized. In an experimental challenge study, pregnant does infected with B. melitensis 16M at midgestation had an 86% abortion rate, while no Rev. 1-infected does aborted. Fetal infection rate was 92% and 43% in kids of 16M- and Rev. 1-infected does, respectively. Widespread tissue colonization was noted in 100% of 16M-infected does, and all of these animals shed brucellae in milk and vaginal secretions. ▼ Introducción: El Callao que es la segunda provincia con mayor incidencia por brucelosis en el Perú. El Hospital Nacional “Daniel Alcides Carrión” (HNDAC) es hospital de referencia del Callao. Objetivos: Evaluar las características clínicas más frecuentes de la brucelosis en pacientes hospitalizados y ambulatorios en el HNDAC, evaluando según el tipo de brucelosis y sus complicaciones. Material y Métodos: Estudio retrospectivo descriptivo aplicado; abarca todo paciente con diagnóstico de brucelosis confirmada con serología y/o cultivo, atendidos en el HNDAC durante los años 1999-2000. Resultados: Se encontró 193 casos, 134 (69,4%) con brucelosis aguda, 56 (29%) con brucelosis subaguda, 03 (1,6%) con brucelosis crónica. Hubo 66 (34,2%) casos hospitalizados y 127 (65,8%) ambulatorios. Las manifestaciones clínicas más frecuentes en hospitalizados: fiebre (95,5%), hiporexia (78,8%), astenia (63,6%), artralgias (57,6%), nauseas (47%), vómitos (40,9%), palidez (31,8%), petequias (7,6%) y como complicaciones: anemia (83,3%), plaquetopenia (39,4%), leucopenia (36,4%), osteoarticular (16,7%), hepática (34,9%), la totalidad de púrpura, pancitopenia, orquitis, uveítis, colitis requirieron hospitalización. En ambulatorios: fiebre (77,2%), astenia (48,8%), cefalea (42,5%), anemia (50,4%), plaquetopenia (41,7%), alteración hepática (11,8%), osteoarticular (7,9%). En brucelosis aguda, se hospitalizaron 36,6%, se encontró fiebre (91,8%), astenia (60,4%), hiporexia(53,7%), anemia (66,4%), plaquetopenia (37,3%) leucopenia (27,6%), alteración hepática (20,9%), osteoarticular (5,2%), parotiditis (1,5%), pancitopenia (1,5%). En brucelosis subaguda se hospitalizaron 30,4%, se encontró: fiebre (67,9%), artralgias (42,9%), astenia (39,3%), hiporexia (32,1%), lumbalgia (28,6%), petequias(7,1%), epistaxis (7,1%), anemia(53,6%), plaquetopenia (50%), púrpura (8,9%), complicación osteoarticular (21,6%), hepática (16,1%). En brucelosis crónica: artralgias (66,7%), hiporexia (33,3%), lumbalgia (33,3%), complicación osteoarticular (66,7%), hepática (33,3%), ningún hospitalizado. Tuvieron control posterior 146 pacientes (75,6%). Se encontraron 27 (14,0%) recaídas, todos con sintomatología de menos de 02 meses, sobresale: fiebre (66,7%), artralgias (40,7%), plaquetopenia(55,6%), artritis periférica (11%) y artritis mixta (3,7%). Se especificó el tratamiento recibido en 72 pacientes (37.3%) de estos 17,3% fueron ambulatorios y 75,8% hospitalizados y se especificó el alta sólo en 04 casos (2,1%). Conclusiones: Los pacientes hospitalizados presentan mayor número de manifestaciones clínicas y de complicaciones. La brucelosis aguda es la forma clínica más frecuente, se presentan síntomas de compromiso general y menor complicación sobresale la hemática y hepática. La brucelosis subaguda tiene más signos hemorragíparos y mayor número de complicación especialmente hematológica y osteoarticular. En la brucelosis crónica sobresale las artralgias, todos con alguna complicación. ▼ Brucellosis is a disease of socio-economic and zoonotic importance worldwide. In animals it is associated with the ingestion of feed that is contaminated with cyetic material from aborting herd-mates, while in humans it is associated with the consumption of unpasteurised milk and dairy products from infected animals. It may also be acquired from contact with infected material of animal origin by farmers, veterinarians, and abattoir and laboratory workers. Brucellosis was first reported in South Africa in the late nineteenth century. It is still present in the country today, with reported annual losses of at least R 300 million, and a national annual incidence of 5 000 cases in humans. The global incidence of human brucellosis is about half a million infections annually. As the incidence of human brucellosis is directly associated with prevalence in animals, control of animal brucellosis is emphasised. Veterinary control is compromised by the chronic nature and the variable incubation period of the disease, with an estimated up to 15% of cattle in infected herds aborting before sero-conversion. Latency, which involves about 5% of calves born from infected dams, is also problematic as these infected animals often test seronegative, only to seroconvert in the peri-parturient period, thus allowing opportunity for disease spread within and between herds before diagnosis is made. In addition, the currently used serological tests are at times unable to distinguish brucellosis from cross-reacting antibodies from other infections or brucellosis vaccines. x It was the objective of this study to investigate, under South African conditions, the value of the brucellin skin test (BST) in improving the sensitivity and specificity of the currently used serological tests. It has proved a valuable additional test in diagnosing early and latent infections as well as in differentiating brucellosis from cross-reacting organisms in unvaccinated cattle in Europe. The study also evaluated the benefit of replacing some of the currently used serological assays with the fluorescence polarisation assay (FPA). The FPA, a rapid and homogenous serological test with only a few operational steps, has been validated and is in current use in Canada. The study was carried out in Mpumalanga Province, on herds selected to reflect prevailing South African farming conditions. These herds were divided into certified Brucella abortus-negative herds (608 head) for the estimation of BST specificity, and confirmed B. abortus-infected herds (845 head) for the estimation of BST sensitivity. The results obtained indicated the BST had a specificity of 99.18%, and a relative sensitivity of 42.86%. However, 65.38% of BST-positive animals were negative on serology. ▼ The automated complement fixation test (CFT) and the brucellosis card test (BCT) have been widely used as official tests in the New Zealand Bovine Brucellosis Eradication Scheme. During the course of the eradication programme it was observed that a significant proportion of cattle reacted to the BCT yet remained negative to the CFT and this often occurred on more than one occasion for any particular animal. Twenty cows, from reactor herds, that had been BCT+/CFT- on at least three successive occasions were slaughtered. Despite extensive sampling, attempts at isolating Brucella abortus organisms from tissues of these animals were unsuccessful. Serum from one cow was found to be positive to a wide range of serological tests and it also caused a strong prozone reaction in the CFT, which could easily have been overlooked. The possibility that the automated CFT, which is essentially a one dilution test, was unable to detect such prozoning sera was investigated. It was shown that providing a suitable choice of antigen concentration was made, such sera would be detected by the automated test. Brucella - specific IgG1, IgG2, and IgM levels in Prozoning and non prozoning sera were measured using the single radial immunodiffusion test. It was shown that serum containing a high proportion of specific IgG2 was likely to exhibit prozoning and that various degrees of prozoning could be induced by varying the ratio of specific IgG1 to specific IgG2. Cattle, previously sensitized by calfhood Br. abortus strain 19 vaccination, were experimentally inoculated with killed Br. abortus. It was shown that although serum agglutination test (SAT) and CFT titres appeared for a short period, titres to the BCT in some cattle tended to remain longer thus allowing an animal to be BCT+/CFT-. An analysis of herd testing data indicated that BCT+/CFT-animals were more likely to exist in infected herds than in non-infected herds. In heavily infected herds up to 16% of CFT- animals were BCT+ whereas in non-infected or very lightly infected herds less than 4% were CFT-/BCT+. It was concluded that in sensitized cattle at least exposure to the organism without true infection is capable of stimulating antibody which is detected by the BCT, but not necessarily able to provoke positive CFT titres. The performance of the Auto-Analyzer adaptation of the CFT as used in the New Zealand eradication scheme was assessed. Various prozoning sera from known infected animals were tested and the effect of varying antigen concentrations on these and other sera was noted. Significant differences in antigen concentration required for optimal complement fixation were detected. Prozoning sera required more antigen than non-prozoning sera and even sera that did not exhibit prozoning had varying optimal antigen requirements. By using I125 labelled bovine gama-globulin the dilution gradient of serum within the Auto-Analyzer system was estimated. O concelho de Nordeste, Ilha de S. Miguel - Açores, tem como principal actividade económica a pecuária de bovinos. Informações do Centro de Saúde de Nordeste indicavam que em 2005 as zoonoses registadas naquele concelho foram a brucelose e a leptospirose, podendo ambas ser transmitidas ao Homem a partir de bovinos infectados e de produtos de origem animal, nomeadamente, leite e lacticínios, entre outras fontes. Definiram-se como principais objectivos deste estudo a determinação da seroprevalência de algumas zoonoses de origem bacteriana responsáveis por abortos em vacas (brucelose, leptospirose e clamidiose) nas explorações agrícolas leiteiras do concelho e o estudo das características do sistema de produção que possam favorecer a sua disseminação e manutenção. Foram colhidos soros de 118 explorações leiteiras, seleccionadas aleatoriamente de uma forma proporcional ao número de efectivos existentes em 3 classes de tamanho, entre Novembro de 2006 e Maio de 2007. Para a brucelose (B. abortus), o método de diagnóstico usado foram os testes oficiais do Rosa de Bengala e da Fixação de Complemento. A seroprevalência das explorações foi de 0,85% (1/118) (IC95% de >0 a 2,16%), e a animal foi de 0,18% (4/2252) (IC95% de 0,05 a 0,31%). Para a leptospirose, as 2153 amostras de soro de vacas não vacinadas foram testadas pelo método do Teste de Micro Aglutinação (TAM) na diluição mínima de 1:100, e 393 foram positivas (18,3%; IC95% 17,7 a 19,5%). A prevalência de explorações seropositivas foi de 61,1% (IC95% de 53,1 a 68,0%). Foram testados 13 serovares, e identificados 9, mas não foi possível fazer o estudo da seroprevalência do serovar Hardjo, por este requerer diluições mais baixas, embora fosse suspeito de estar presente em infecção endémica. O serogrupo Sejroe foi o mais importante por apresentar a seguinte seroprevalência de serovares: Saxkoebing 67,9% e Wolffi 0,5%. O segundo serogrupo mais frequente foi o Icterohaemorrhagiae com os serovares Icterohaemorrhagiae, 14,8% e Copenhageni, 8,4%. Depois foram os serogrupos: Autumnalis (serovar Autumnalis 7,1%); Ballum (serovar Arborea 7,1%); Australis (serovar Bratislava 5,6%); Tarassovi (serovar Tarassovi 1,8%) e Canicola (serovar Canicola 1,8%). A coaglutinação com associação de L. ballum arborea e L. autumnalis autumnalis foi a que apresentou maior frequência, seguida da associação L. icterohaemorrhagiae icterohaemorrhagiae e L. icterohaemorrhagiae copenhageni. Para a clamidiose, foi usado o método de ELISA indireto para a pesquisa de anticorpos da Chlamydophila abortus e a seroprevalência das explorações foi de 82,9% (IC95% de 76,7 a 89,1%) e a animal foi de 14,0% (IC95% de 12,9 a 15,1%). Em virtude da importância econômica da Leptospira spp. e Brucella abortus para a saúde pública, e da ausência de dados referentes à soroprevalência da leptospirose e brucelose para bovinos de corte consumidos no Estado, neste trabalho objetivou-se estimar a prevalência de anticorpos anti-Leptospira spp. e anti-Brucella abortus em bovinos abatidos em matadouro público no Estado de Pernambuco. 412 amostras de sangue de bovinos foram colhidas após venossecção dos grandes vasos do pescoço, durante a sangria, e encaminhados ao Laboratório de Bacteriologia do Curso de Medicina Veterinária da Universidade Federal Rural de Pernambuco para obtenção do soro. Estes foram processados no Laboratório Agropecuário Nacional de Pernambuco, de acordo com metodologia oficial. Para a detecção de anticorpos anti-Leptospira spp. foi utilizada a Técnica de Soroaglutinação Microscópica (SAM). As amostras foram diluídas 1:50 e submetidas à triagem com 27 sorovares de Leptospira spp., sendo positivas aquelas com título igual ou superior a 100 com 50% ou mais de Leptospira spp. aglutinadas. A triagem dos anticorpos anti-Brucella abortus foi realizada com o Antígeno Acidificado Tamponado (AAT), a fim de identificar possíveis amostras reagentes ao Teste de Fixação do Complemento (TFC). A análise estatística das amostras foi realizada através da análise descritiva das mesmas. Para Leptospira spp., das 412 amostras testadas, 55 foram positivas (MAT >1:100), resultando em uma soroprevalência de 13,3% (95% IC = 10,1% e 16,6%). Animais soropositivos foram provenientes de 20 dos 24 municípios de origem, sendo muitos sorovares de Leptospira spp. identificados. Dos sorovares pesquisados, 16 foram detectados: Shermani (25,5%), Wolffi (14,5%), Hebdomadis (10,9%), Grippotyphosa (9,1%), Canicola (7,3%), Saxkoebing (5,5%), Bratislava (5,5%), Copenhageni (3,6%), Hardjo (3,6%), Djasiman (3,6%), Icterohaemorrhagiae RGA (1,8), Icterohaemorrhagiae ICTERO n 1 (1,8), Serjoe (1,8%), Panama (1,8%), Pyrogenes (1,8%) e Patoc (1,8%). Totalizando as amostras analisadas, 0 (0%) foi reagente ao TFC. Diante dos resultados, conclui-se que apesar de não haver bovinos brucélicos, os animais avaliados representam risco à saúde pública, devido à possibilidade das pessoas adquirirem a leptospirose através deles.

Em virtude da importância econômica da Leptospira spp. e Brucella abortus para a saúde pública, e da ausência de dados referentes à soroprevalência da leptospirose e brucelose para bovinos de corte consumidos no Estado, neste trabalho objetivou-se estimar a prevalência de anticorpos anti-Leptospira spp. e anti-Brucella abortus em bovinos abatidos em matadouro público no Estado de Pernambuco. 412 amostras de sangue de bovinos foram colhidas após venossecção dos grandes vasos do pescoço, durante a sangria, e encaminhados ao Laboratório de Bacteriologia do Curso de Medicina Veterinária da Universidade Federal Rural de Pernambuco para obtenção do soro. Estes foram processados no Laboratório Agropecuário Nacional de Pernambuco, de acordo com metodologia oficial. Para a detecção de anticorpos anti-Leptospira spp. foi utilizada a Técnica de Soroaglutinação Microscópica (SAM). As amostras foram diluídas 1:50 e submetidas à triagem com 27 sorovares de Leptospira spp., sendo positivas aquelas com título igual ou superior a 100 com 50% ou mais de Leptospira spp. aglutinadas. A triagem dos anticorpos anti-Brucella abortus foi realizada com o Antígeno Acidificado Tamponado (AAT), a fim de identificar possíveis amostras reagentes ao Teste de Fixação do Complemento (TFC). A análise estatística das amostras foi realizada através da análise descritiva das mesmas. Para Leptospira spp., das 412 amostras testadas, 55 foram positivas (MAT >1:100), resultando em uma soroprevalência de 13,3% (95% IC = 10,1% e 16,6%). Animais soropositivos foram provenientes de 20 dos 24 municípios de origem, sendo muitos sorovares de Leptospira spp. identificados. Dos sorovares pesquisados, 16 foram detectados: Shermani (25,5%), Wolffi (14,5%), Hebdomadis (10,9%), Grippotyphosa (9,1%), Canicola (7,3%), Saxkoebing (5,5%), Bratislava (5,5%), Copenhageni (3,6%), Hardjo (3,6%), Djasiman (3,6%), Icterohaemorrhagiae RGA (1,8), Icterohaemorrhagiae ICTERO n 1 (1,8), Serjoe (1,8%), Panama (1,8%), Pyrogenes (1,8%) e Patoc (1,8%). Totalizando as amostras analisadas, 0 (0%) foi reagente ao TFC. Diante dos resultados, conclui-se que apesar de não haver bovinos brucélicos, os animais avaliados representam risco à saúde pública, devido à possibilidade das pessoas adquirirem a leptospirose através deles.

As leishmanioses são antropozoonoses de caráter crônico e distribuição mundial e apresentam uma incidência estimada de 2 milhões de novos casos por ano, entre leishmaniose visceral (LV) e leishmaniose tegumentar (LT). Os cães são bastante susceptíveis à infecção por LV e devido à sua estreita relação com o homem, são considerados os reservatórios de maior importância para a transmissão da doença ao ser humano. As leishmanioses ocorrem de forma endêmica nas regiões Norte, Nordeste, CentroOeste e Sudeste, fazendo-se necessária a realização de inquéritos sorológicos, visando conhecer a situação epidemiológica da doença nas áreas com transmissão ativa ou com potencial de transmissão. A brucelose é uma zoonose de distribuição mundial que causa infecção sistêmica no homem e diminuição da eficiência reprodutiva e aborto em espécies domésticas e silvestres. No Brasil, a brucelose devido a Brucella abortus é a infecção brucélica mais prevalente, seguida da B. suis e B. canis. Devido ao caráter zoonótico e a perdas reprodutivas é importante diagnosticar animais infectados para interromper a transmissão da doença. A partir do exposto, realizou-se um inquérito sorológico em humanos e em cães habitantes de aldeias indígenas da região amazônica do Brasil, com o objetivo de identificar áreas endêmicas para Leishmania spp. e Brucella spp. O presente estudo analisou amostras de soro e sangue de todos os indígenas e cães habitantes da Terra Indígena Tapirapé, assim como de cães da Terra Indígena Karajá, através dos testes sorológicos ELISA e RIFI para a pesquisa de leishmaniose e dos testes sorológicos IDGA e AAT, além da PCR para a pesquisa de brucelose. Os resultados obtidos para os humanos apresentaram prevalências de 0,4% e 0,9% para LT e brucelose, respectivamente. Todos os humanos foram negativos para LV. Para os cães das aldeias Tapirapé os resultados de prevalência foram 1,8%, 0,9% e 6,1% para LV e LT e brucelose, respectivamente. Os cães das aldeias Karajá apresentaram resultados negativos para LV e prevalências de 0,9% e 5,2% para LT e brucelose, respectivamente. Os resultados obtidos neste estudo indicam baixa ocorrência de infecção por Leishmania spp. e ocorrência moderada de infecção por Brucella spp. Embora as leishmanioses se apresentem pouco disseminadas nas áreas estudadas, requerem atenção, assim como a brucelose, para a tomada de medidas de controle e profilaxia.

A pecuária é um setor do agronegócio que vêm se destacando na economia nacional. Com a intenção de amparar esta importante cadeia produtiva, a atuação dos serviços de fomento e defesa sanitária animal tornam-se cada vez mais necessárias. A compreensão integral do espaço sobre o qual estes serviços exercem sua influência, tornaria mais efetiva as estratégias de atuação frente aos problemas enfrentados. Em 2001, o Ministério da Agricultura Pecuária e Abastecimento realizou um estudo soroepidemiológico visando conhecer a situação epidemiológica da brucelose bovina em 14 Unidades Federativas. O trabalho incluiu um questionário epidemiológico aplicado na intenção de avaliar os possíveis fatores de risco para a doença nos rebanhos. O mesmo possibilitou a realização de uma análise descritiva da pecuária em diversas regiões. Os resultados demonstraram que a cadeia produtiva no país é altamente heterogênea, revelando diferenças significativas mesmo dentro dos Estados. De maneira geral, as frequências encontradas a respeito de investimento em tecnologia, intensificação dos manejos, utilização da inseminação artificial, utilização de raças especializadas, e participação de assistência veterinária, demonstrou-se crescente à medida que se avança em direção às Unidades Federativas da região Sul. A comercialização de reprodutores demonstrou-se intensa e realizada diretamente entre as propriedades. O comércio informal do leite e da carne bovina ainda corresponde por uma parcela significativa do mercado. Tendo em vista a escassez de projetos com este foco, reflete-se a respeito da necessidade de outros projetos deste tipo, considerando que os cenários em que as criações estão inseridas, deveriam ser tomados como ponto de partida na elaboração de estratégias de atuação frente aos problemas encontrados no setor.

Livestock is an agribusiness sector highlighted in the current national economy. With the intention of supporting this important activity, the performance of animal health promotion and defense services become increasingly necessary. With a full understanding of the environment on which these services exert its influence, strategies actions to minimize the problems faced become more effective. In 2001, the Ministry of Livestock Agriculture conducted a seroepidemiological study aiming to explore the epidemiological situation of bovine brucellosis in 14 Federal Units. The study included an epidemiological questionnaire which objective was to assess the possible risk factors for disease in livestock. It also allowed the development of a descriptive analysis of livestock breeding and raising in various regions. The results showed that the productive chain in Brazil is highly heterogeneous, revealing significant differences even within the same state. In general, frequencies of investment in technology, management practices, use of artificial insemination, use of specialized breeds, and veterinary care, increases as one moves toward the Federal Units located in the South of Brazil.
