▼ Waterlogging is a major environmental constraint severely limiting crop production both in Australia and worldwide. In Australia, the problem is especially acute in higher rainfall areas with poorly draining soils. With most Australian commercial barley cultivars being waterlogging sensitive, waterlogging tolerant genetic material has to be selected and used in the breeding programs. Of specific interest is Chinese germplasm collection. However, lack of appropriate screening methodology makes selection ofwaterlogging tolerant cultivars difficult. Also, waterlogging is a complex abiotic stress encoding a large number of physiological and biochemical mechanisms and complicated by many confounding factors. Accordingly, the aims of this project were three-fold: (1) to develop an efficient screening methodology for waterlogging tolerance in barley breeding programs; (2) to select some waterlogging tolerant cultivars among existing barley germplasm; and (3) to understand the physiological, morphological and anatomical mechanisms encoding waterlogging tolerance in barley. The project was an integral part ofGRDC-funded collaboration between Australia and China on barley genetics. Barley plant growth was adversely affected by waterlogging. As waterlogging stress developed, chlorophyll content, net C02 assimilation and maximal photochemical efficiency of PSil (Fv/Fm) decreased significantly. Among these, chlorophyll fluorescence of dark-adapted samples (Fv/Fm values) was found to be the most efficient screening parameter for large-scale programs for waterlogging tolerance. Among studied genotypes, the Chinese cultivar TX9425 was found to be the most waterlogging tolerant, with the least reduction of plant growth, chlorophyll content, chlorophyll fluorescence and photosynthetic parameters. Naso Nijo (a Japanese cultivar) and Franklin (an Australian cultivar) were found to be the most susceptible to waterlogging. The adverse effects in stressed plants were alleviated following 2 weeks recovery in all cultivars. These contrasting genotypes were used later in all physiological studies. Overall, this study suggests that substantial genetic potential exists to improve waterlogging tolerance in barley. Measuring chlorophyll fluorescence of darkadapted samples (Fv/Fm values) is recommended as an efficient screening tool for waterlogging tolerance in barley breeding program. Key features targeted by breeding should include both morphological (production of more adventitious roots and formation of larger aerenchyma area in adventitious roots) and physiological (high ability of 0 2 uptake and K+ acquisition in plant roots) traits, as well as the plant's ability to withstand soil-borne phytotoxins. Foliar application of nutrient c~ be used in practice to alleviate the adverse effects ofwaterlogging.

▼ A series of experiments were carried out from 1988 to 1990 at Thirlstane in NorthWestern Tasmania to determine the suitability of modern high yielding winter barley cultivars for dual purpose (winter grazing plus grain) management. The barley cultivar Ulandra (WU 3076) was used in the experiments. It has a prostrate growth habit and late flowering, characters which should make it suitable for use as a dualpurpose crop where growing points need to be protected during grazing. In the first experiment sown in late April 1988, four grazing treatments were used, (i) control, no grazing, (ii) early grazing (as soon as a reasonable amount of forage was available), (iii) late grazing Gust prior to the shoot apex reaching grazing height), (iv) early and late grazing (a combination of grazing treatments (ii) and (iii)). The grazing was carried out using Angus cattle to defoliate the crop as quickly as possible. In addition, small areas of each plot were mechanically defoliated, to assess whether cutting can simulate the effects of grazing. Immediately following the completion of the second grazing all treatments were split for nitrogen application of 0 or 50 kg N/ ha, to determine its effects in assisting recovery following grazing. The least amount of dry matter was removed by the early grazing and the most by the single late grazing. Nitrogen compensated for the loss of dry matter following early grazing, in that maximum dry matter and grain yield were brought up to the level. of the nil N control. Nitrogen had no effect on late or twice grazed treatments, where maximum dry matter and final grain yield were greatly reduced. Grazing reduced final plant height by 15-20 cm. As the control treatment lodged, particularly with N application, the reduction in plant height could be considered to be an advantage. Cutting resulted in lower grain yields than grazing due to the greater severity of its effects on the growing crop. In 1989 the experiment was repeated for a range of sowing times, late March, early April and late April. Wet weather in winter caused some problems with waterlogging, and as a consequence growth was reduced on all treatments. The late March sowing was the most affected as the worst of the water logging occurred at the time of grazing. The late April sowing was grazed in late August-early September during drier conditions and was therefore better able to recover from the effects of grazing. The early April sowing produced the largest amount of forage for all grazing treatments. The late March and early April sowings grew rapidly in late autumn and early spring before the onset of the colder weather and produced more leaf area than the late April sowing, although none of the leaf areas were as high as might normally be expected. The late April sowing grew slowly up to the beginning of the warmer weather in early spring and whilst leaf area was always smaller than the other sowings, the crop appeared better able to utilise it in terms of final grain yield. well below the…

▼ An investigation of Mn uptake by five-day-old excised barley roots revealed that a metabolically-mediated process was involved. In short-term experiments, the rate of Mn absorption was comparable to that of the macronutrient cations. Like other metabolically absorbed cations, the Mn absorption rate was a direct function of the ambient concentration and the pH. An evaluation of mutual effects between Mn and other cations revealed several specific regulatory effects. Of the alkali cations studied, Li alone had a pronounced stimulatory effect while Na, K and Rb markedly reduced the absorption of Mn. The alkaline earth cations also exerted widely differing effects. Calcium appeared to promote the absorption of Mn, whereas Mg had a highly inhibitive effect. The combination of both Ca and Mg was even more inhibitory. Strontium apparently was without effect and Ba had a moderately depressive effect. Other polyvalent cations which were effective inhibitors of Mn absorption were Fe⁺⁺, Zn, Cu, Al and La. In contrast, Fe⁺⁺⁺ was virtually without effect. Manganese effectively blocked the absorption of Li and Mg, but greatly enhanced that of Na, K and Rb. These diverse regulatory effects and many others reported in the literature are explained by the following hypothesis: the cationic environment at the extracellular surface of the membrane is believed to control the specificity of the ion carrier. By attaching to critical activation sites, cations induce conformational changes in the carrier which modify its selective transport properties. The accessibility or affinity of transport sites for a given ion would depend on the particular configuration of the carrier. This mechanism, together with the mutual competition between some ions for the same transport site and the cationic maintenance of the cellular membranes, would explain most, if not all, of the regulatory effects exerted by cations on the ion absorption process. ▼ Foliar applications of urea-ammonium nitrate (UAN) around anthesis increased seed protein content by about 40% in 1983 and 1984 in 'Scio' barley (Hordeum vulgare L.). In exploring the mode and the mechanism of this increase, agronomic and physiological studies were conducted. The late N applications did not affect yield or harvest index, but elevated the seed amino acid content and reduced the sugar content. The 40% increase of total seed protein was unevenly distributed to globulin-albumin, hordein, and glutelin fractions by 27, 67, and 40%, respectively. No new protein subunits were observed after the treatment as revealed by SDS-polyacrylamide gel electrophoresis of the protein fractions. The approximate molecular weights of the hordein subunits were 33, 35.5, 38, 40.5, 42, 44, 50, 52, 54, 57, 61, 72, and 79 kD. The physiological responses to repeated foliar-UAN sprays were monitored in young and mature leaves. The absorbed urea and ammonia were metabolized in 2 days and stimulated the endogenous activities of nitrate reductase (NR) and urease from 2- to 77-fold within hours with the higher activity of NR sustained for several days. The activities of glutamine synthetase and glutamate synthase were also increased but much less in extent. These enhanced activities and added N substrates resulted in an increase of leaf contents of amino acids, chlorophyll, soluble protein, and a small decrease of sugars. Eight hours after the UAN spray, when amino acid content was highest in sprayed-seedling leaves, the absolute weights of glutamine, asparagine, alanine, glutamate, serine, glycine, valine, and histidine were 2535, 511, 460, 373, 361, 240, 129, and 59 ng g⁻¹ fresh weight over the control, respectively. The highest increase in weight percent of the extract was found in glycine, which is different from the results of ammonia or nitrate treated leaves, indicating that the absorbed urea may enhance the photorespiratory synthesis of glycine or reduce the degradation of glycine due to the high concentration of ammonia produced by urease hydrolysis of the influxed urea. ▼ The focus of this study was to determine if S₁ family recurrent selection might be successfully used for barley improvement. Objectives were: 1) to identify suitable methods of evaluating grain yield in small plots, 2) to study the effects of intermating on mean values and variances of selected agronomic characters, and 3) to evaluate the response to one cycle of S₁ family selection for grain yield. Genetic materials included parents, random F₃ lines from four single cross populations, and random S₁ lines from the Cycle 0 (C-0) population which was developed by intermating the four single crosses for two generations. Heading date, plant height, and grain yield were measured on F₃ and S₁ lines grown in unreplicated small (0.7 m²) two row plots in two locations in 1979. The same traits were measured on bulk progenies of F₃ and S₁ lines grown in unreplicated large (6.0 m²) plots in three locations in 1980. Moving mean technique, which uses the mean grain yield of a number of adjacent plots as a covariable in an analysis of covariance, improved the precision of yield trials. Error mean square of replicated parents was reduced between 0 and 80 percent at different experimental sites, depending on the magnitude of field variation. Comparison of grain yield of parents grown in both large and small plots in the same year-location indicated that grain yield in small plots was affected by competition effects associated with plant height. Additional evidence for the inaccuracy of small plot yield data was supplied by the higher positive correlation between grain yield and plant height in small plots as compared to large plots. Breakup of favorably interacting associations of alleles by intermating was studied by comparing the mean of the four single crosses with the mean of the C-0 population. Two generations of intermating did not affect the mean values for plant height or grain yield, however the C-0 population headed significantly earlier due to the effect of rapid generation advancement in the intermating process. Two generations of intermating increased genetic variance for heading date and plant height, but no effect could be detected for grain yield. While some of the estimates of genetic variance for grain yield were negative, most were small and non-significant. Lack of genetic variation for grain yield is believed to be due to common parentage of the genetic materials in this study. Response to selection for grain yield was evaluated with Cycle 1 (C-1) S₁ lines developed by intermating the highest yielding 10% of C-0 S₁ lines. The C-1 population was significantly higher yielding than parents and significantly taller than parents and the C-0 population. Since little genetic variation for grain yield was found in this study, the response to selection for grain yield shown in small plot tests is believed to be a manifestation of competition effects associated with the increased plant height of the C-1 population. ▼ During barley germination, α-amylase and other hydrolases are synthesized in the aleurone layer and secreted into the starchy endosperm. As starch is degraded by amylases, lysophospholipids are released from inclusion complexes with the starch and undergo hydrolysis by a lysophospholipase (LPL) activity. Studies with embryo-free half seeds and isolated aleurone layers from barley (Hordeum vulgare L. cv Himalaya) show that LPL activity appears in the aleurone layer of imbibed half-seeds. Secretion, on the other hand, requires the continued presence of gibberellic acid (GA₃) and commences after a 10 to 14 hour lag period. Ca²⁺ alone has very little effect on the level of LPL activity in the aleurone layer or that secreted into the surrounding medium. However, 50 mM Ca²⁺ together with GA₃ dramatically increases the total activity. Density labeling experiments with deuterium oxide indicates that GA₃-stimulated LPL is synthesized de novo. Two forms of LPL activity are distinguished by ion exchange chromatography on carboxymethyl cellulose. A predominantly basic activity is observed in the starchy endosperm of germinated barley and the incubation medium of GA₃-stimulated half-seeds. In contrast a predominantly acidic activity appears in the incubation medium of GA₃-treated aleurone layers. The predominantly acidic activity from the medium of GA₃-stimulated aleurone layers is converted to the basic form in the presence of starchy endosperm, amylase-digests of barley starch or EDTA. The factor from starch is thermally stable. Divalent metal ions do not shift the basic activity to the acidic form. On gel filtration chromatography the acidic LPL elutes at a high molecular weight position (M[subscript r] ~ 160000) distinct from the basic activity (M[subscript r] 40000). The elution volume of the acidic activity treated with EDTA shifts to that of the basic activity. Anti-serum prepared against a purified basic LPL generates similar enzyme inactivation profiles against extracts of both the predominantly basic and acidic LPLs. ▼ Four, six-rowed, spring barley cultivars were used as parental material in this study. The parents were distinctly different in plant height. Washington-6124-62 is a standard height selection while Short Wocus and Minn. 66-102 are intermediate. The fourth parent was a dwarf selection obtained from India. The two intermediate cultivars and the dwarf cultivar were crossed to the tall parent, Wa-6124-62, with a portion of the F₁ progeny backcrossed to the shorter parent. In addition, the intermediate parent, Short Wocus, was crossed to Indian Dwarf. The following generations were available for study: parents, BC₁F₁, F₁, and F₂. The purpose of this study was to obtain information concerning the inheritance of plant height and to determine when selection for this trait could be made in the breeding program most effectively. To achieve this objective heritability and the mode of gene action were studied as well as number of genes segregating for plant height. The effect of temperature on plant height was also investigated in a growth chamber utilizing two temperature levels, 19° C and 30° C. Plant height was recorded on an individual plant basis at two sites that were distinctly different environmentally and in the growth chamber. The number of nodes were counted, and the length of internodes measured for genetic interpretation of culm length. Broad sense heritability values for plant height of each cross were estimated by using the variance of parents, F₁'s and F₂' s. Heritability values were quite high in the F₂ generation ranging from 50 to 90 percent, and suggest that plant phenotype gave a relatively good indication of the genotype for plant height. Height of the F₁ plants was between the mid-parent and tall parent indicating partial dominance for tall culm length. In earlier studies, many investigators found that additive gene action was the major component of genetic variability for plant height. Results obtained from this study suggest that the nature of gene action is a function of the parents used. Where large differences exist between parents, plant height appeared to be largely influenced by non-additive gene action. When the parents were similar in height the trait was controlled mainly by additive gene action. Fairly discrete height classes in the segregating populations indicated that plant height was controlled by only a few genetic factors in these populations. Ratios obtained from segregating populations suggested the presence of three recessive genetic factors for short height in the dwarf parent (Indian Dwarf). The tall parent, Wa-6124-62, and intermediate parent, Minn. 66-102, appeared to differ for plant height by one major genetic factor. Difference in height among the cultivars was mostly due to difference in internode length rather than the number of nodes. ▼ Four agronomically and genetically diverse spring barleys were used in a crossing program to study the inheritance of lysine in barley and to determine its possible association with certain agronomic and morphological characters. The cultivar Hiproly was used as the source of high protein and lysine. Characters measured included plant height, head type, awn type, lemma color, number of heads per plant, number of seeds per head, seed weight, and plant yield. Soluble protein content was determined by the Lowry colorimetric method. Lysine was analyzed by two methods: (1) enzyme hydrolysis and colorimetric determination with 2-chloro, 3, 5- dinitropyridine; (2) Udy dye-binding colorimetric method. The F₁, F₂, and F₃ progenies were classified for the Hiproly endosperm character by histochemical techniques using Udy Orange G dye as a staining agent. Fractionation of the soluble proteins into the Mendal- Osborn fraction was used to determine the effects of the Hiproly gene on kernel proteins. High lysine content in Hiproly was found to be inherited by one recessive gene. In addition several minor factors were indicated in all crosses. The recessive gene was associated with the recessive Hiproly endosperm gene with a recombination value of 22.45 percent. It is suggested that the gene designations for the two recessive genes be (lys) for the high lysine gene and (stb) for the starch binding endosperm gene. High lysine was negatively associated with protein content; however, the association was not strong enough to limit selection for high lysine, high protein lines. No associations were found between lysine and any of the morphological or agronomic characters studied. It was concluded that it is possible to breed high yielding, high protein and lysine barleys using Hiproly as a source of high protein and lysine. ▼ This study evaluated the effects of 5 different soaking treatments in conjunction with 5 varying irrigation intervals on the germination, growth and nutritional values of seed of Hordeum vulgare Sv13. The 5 different soaking times consisted of 1, 3, 8, 16 and 24 hours. The barley seed was first cleaned and then placed in a vessel containing 500 ml of distilled water with a 20 % solution of sodium hypochlorite (bleach) at room temperature. Thereafter the pre-soaked seeds were transferred to a perforated container, containing no medium and placed into a growing chamber equipped with drip irrigation. The seed was then irrigated with 1245 ml of water at 5 different intervals namely every 2, 4, 8 10 and 12 hours. The temperature of the hydroponic growing room was kept at a constant 23 °C using a hotoperiod of 16-hour day/ 8-hour darkness. The seed was allowed to germinate and grow for a period of 8 days before being harvested. The objectives of this study were to determine the most beneficial combination of soaking treatment in conjunction with the most beneficial irrigation interval on the germination rate of the seed allowing for radicle emergence and coleoptile production. It was also used to determine which combination of treatments was most beneficial to the growth and nutritional values of the seed post-harvest. Another objective was to ascertain the shortest soaking time for application in a small-scale, hydroponic growing unit as well as the frequency of irrigation required to grow seedlings, thereby determining the amount of water required to produce a seedling mat for a small-scale, subsistence farmer, with the emphasis being on water reduction. Each treatment was replicated 10 times and consisted of 500 grams of seed, which when placed into its container measured 2 centimetres in depth, totalling 25 treatments in all. Germination was measured by observing radicle emergence in the first 2 days of the growing period first after a 24-hour cycle and again after 48 hours. The numbers of leaves present at harvest after an 8-day growing period were also counted to determine germination rate of the seeds. Growth was determined by average leaf height as well as the tallest leaf on day 8 of the growing cycle. Root mat expansion was also measured, post-harvest, which was compared to the initial 2 cm planting depth of seed. Wet and dry weights of the plant material were measured post-harvest. Samples of the harvested material were also sent for nitrogen and protein analysis. It was discovered that most of the results favoured a shorter soaking time and an increase in irrigation frequency, bar a few exceptions. Most favoured a pre-soaking time of only 1 hour together with an irrigation frequency of between 2 and 4 hours. This shows that small-scale farmers would be able to reduce the time spent on soaking of their seed. ▼ With an increasing population it is important to increase crop yield. However, there is a low priority in breeding for increased tolerance to low input or marginal environments. A potential source of viable resilience and resource use efficiency traits are landraces local to areas of marginal land, such as the Scots Bere from the highlands and islands of Scotland. Bere barley is a deeply historically rooted landrace of barley which has been grown on predominately marginal land for the last half millennia, allowing them to yield well in these conditions. The project aim was to assess and genetically characterise traits associated with enhanced resistance/tolerance, and to identify contributing genomic regions. In this project the JHI spring barley collection, consisting of a number of Bere lines, was screened for biotic stress resistance to Rhynchosporium commune and abiotic stress resistance to the conditions of Mn deficiency and salt stress, additionally the interaction of these stresses was assessed. The results identify a number of Bere lines that show an increased resistance/tolerance to each of the three stresses, compared to elite cultivars. The Bere population, as a whole, showed an inherent enhanced Mn use efficiency correlating to increased accumulation of Mn in their shoot. These results suggest that Bere landraces have unique abilities to cope with stress. Interaction studies revealed complex line-specific interactions, along with an overall adverse effect of salt on rhynchosporium symptoms. Several genomic regions for Mn use efficiency, salt tolerance, and rhynchosporium resistance traits originating from the Bere lines were identified, along with potential candidate genes. Further examination and validation of these regions should be undertaken for future breeding for marginal lands. By introgression into elite cultivar backgrounds they may contribute biotic and abiotic stress-tolerance genes creating novel cultivars to efficiently and resiliently yield under low input and marginal environments.
