▼ Banana bunchy top is regarded as the most important viral disease of banana, causing significant yield losses worldwide. The disease is caused by Banana bunchy top virus (BBTV), which is a circular ssDNA virus belonging to the genus Babuvirus in the family Nanoviridae. There are currently few effective control strategies for this and other ssDNA viruses. “In Plant Activation” (InPAct) is a novel technology being developed at QUT for ssDNA virus-activated suicide gene expression. The technology exploits the rolling circle replication mechanism of ssDNA viruses and is based on a unique “split” gene design such that suicide gene expression is only activated in the presence of the viral Rep. This PhD project aimed to develop a BBTV-based InPAct system as a suicide gene strategy to control BBTV. The BBTV-based InPAct vector design requires a BBTV intergenic region (IR) to be embedded within an intron in the gene expression cassette. To ensure that the BBTV IR would not interfere with intron splicing, a TEST vector was initially generated that contained the entire BBTV IR embedded within an intron in a β-glucuronidase (GUS) expression vector. Transient GUS assays in banana embryogenic cell suspensions indicated that cryptic intron splice sites were present within the IR. Transcript analysis revealed two cryptic intron splice sites in the Domain III sequence of the CR-M within the IR. Removal of the CR-M from the TEST vector resulted in an enhancement of GUS expression suggesting that the cryptic intron splice sites had been removed. An InPAct GUS vector was subsequently generated that contained the modified BBTV IR, with the CR-M (minus Domain III) repositioned within the InPAct cassette. Using transient histochemical and fluorometric GUS assays in banana embryogenic cells, the InPAct GUS vector was shown to be activated in the presence of the BBTV Rep. However, the presence of both BBTV Rep and Clink was shown to have a deleterious effect on GUS expression suggesting that these proteins were cytotoxic at the levels expressed. Analysis of replication of the InPAct vectors by Southern hybridisation revealed low levels of InPAct cassette-based episomal DNA released from the vector through the nicking/ligation activity of BBTV Rep. However, Rep-mediated episomal replicons, indicative of rolling circle replication of the released circularised cassettes, were not observed. The inability of the InPAct cassette to be replicated was further investigated. To examine whether the absence of Domain III of the CR-M was responsible, a suite of modified BBTV-based InPAct GUS vectors was constructed that contained the CR-M with the inclusion of Domain III, the CR-M with the inclusion of Domain III and additional upstream IR sequence, or no CR-M. Analysis of replication by Southern hybridisation revealed that neither the presence of Domain III, nor the entire CR-M, had an effect on replication levels. ▼ Bananas are an important subsistence and export crop with over 130 tonnes of banana produced annually. Domesticated banana are triploid and sterile and it is thought that wild diploid bananas were domesticated between 7,000 and 10,000 years ago somewhere in the region of Indonesia, Philippines, New Guinea or the Southeast Asia peninsula. Triploid bananas were subsequently moved around the world through vegetative propagules and more recently through tissue culture. A number of diseases are associated with bananas, including banana bunchy top disease (BBTD) and banana streak disease (BSD), both caused by DNA viruses. BBTD is caused by Banana bunchy top virus (BBTV), a single-stranded DNA (ssDNA) virus (genus Babuvirus; family Nanoviridae). Characteristic symptoms of BBTD are severe stunting of the plant and bunching of the leaves, with a green dot-dash streak on the underside of the leaves. Both viruses can cause severe crop loss and are of concern to banana growing regions. BSD is caused by a number of banana-infecting badnaviruses species (Badnavirus; Caulimoviridae), all of which contain a double-stranded DNA (dsDNA) genome. Characteristic symptoms of BSD include chlorotic and necrotic streaks on the leaves, which are not always seen across all leaves, or even across an entire leaf. BBTV is a multi-component virus which is transmitted by the banana aphid Pentalonia nigronervosa. It is the type member of the babuviruses, which contains two other accepted species, Cardamom bushy dwarf virus (CBDV) and Abaca bunchy top virus (ABTV), which infect large cardamom and Musa spp, respectively. It is generally accepted that the genome of BBTV consists of six different components, with each component individually encapsidated. BBTV is able to evolve through recombination, reassortment and mutation. The large numbers of sequenced BBTV components in Chapter Two and Three (927), and publically available component sequences, have allowed for in-depth analyses into the diversity and evolution of this important plant pathogen. Our analysis shows that both reassortment and recombination play a significant role in the evolution of BBTV. Additionally, we found high genetic diversity in two geographic regions, the Southeast Asian / Far East region and the Indian subcontinent. Phylogenetic analysis of all full genomes, with recombinant regions and reassorted components removed, identified that the BBTV genomes circulating in the majority of countries have likely originated from single founder populations. All nanoviruses contain two common regions which are common across species. The common region stem-loop (CR-SL) is involved in initiation of replication and is recognised by the replication-associated protein (Rep), encoded in the DNA-R component. Understanding the basic of transmission characteristics of any vector born pathogen is the critical if the disease system is to be understood, further it is of up most importance for the creation of any control strategy. It is critical to understand ail aspects of a plant pathogenic virus's biology and how it changes in each environment in order to control its spread. Temperature and insect stage can profoundly change key aspects in viral transmission (TBA). My work on the basic biology of the virus/vector interactions expands on the work that was started over 80 years ago. In Chapter 2, I present work on temporal aspects of BBTV transmission in relation to its vector and host plant I specifically sought to determine how long it took for P. nigronervosa to transmit BBTV after acquisition (Le. the latent period within the insect). To answer this question I conducted studies utilizing classical transmission experiments. The latent period within the plant is the second component of this chapter. The goal was to determine the earliest date a plant begins to act as a source of the virus after inoculation. There have been no studies to determine the vector latent period of any viruses in the family Nanoviridae. The latent period within plant is a neglected area of viral research, specifically with BBTV. The focus of Chapter 3 is to determine how temperature and aphid life stage affect BBTV transmission. I sought to objectively determine how these factors quantitatively affect transmission. The first experiment was an investigation to determine if there are any differences in BBTV transmission by nymph and adults and to determine the impact of temperature on viral transmission efficiency. I tested the effect of three different temperatures (20°, 25°, 30°C) on acquisition and inoculation of BBTV by adult aphids as compared with third instar nymphs. Other experiments were conducted to determine if temperature has a different effect on BBTV acquisition and inoculation efficiency by aphids and what the effect of plant access period on transmission efficiency by adults. Throughout this work, two major components of virus transmission were analyzed. What the latent period of BBTV is within the plant and its aphid vector was determined. This will help improve IPM sampling plan. The second part of my research focused on determining how temperature and the aphid's life stage effects BBTV transmission. I expect both components of my work to improve current knowledge on the biology of circulative, non-propagative plant viruses by aphid vectors.

▼ Banana bunchy top virus (BBTV) is a circular, single-stranded (css) DNA virus that belongs to the genus Babuvirus in the family Nanoviridae. BBTV is responsible for the most devastating virus disease of banana known as "bunchy top", for which conventional control measures are generally ineffective. Genetically engineered resistance appears to be the most promising strategy to generate BBTV-resistant bananas but the success of this strategy is largely dependent upon the molecular characterisation of the target virus and knowledge of the virus life cycle, particularly the replication strategy. This PhD study was aimed at the molecular characterisation of the intergenic regions of BBTV, in order to complement the molecular information currently available and to potentially contribute to the development of transgenic resistance strategies against BBTV in banana. Three putative iterative sequences (iterons; GGGAC) previously identified in the BBTV intergenic regions were initially characterised. In order to determine their role in the binding of the master BBTV replication initiation protein (M-Rep), the putative iterons (F1 and F2 in the virion sense, and R in the complementary sense) were independently mutated in a BBTV DNA-6 greater-than-genome-length clone (1.1 mer). The DNA-6 1.1 mers (native and mutants) and the M-Rep-encoding component (DNA-1) were co-bombarded into banana (Musa spp. cv."Lady finger") embryogenic suspension cells and transient replication was evaluated by Southern hybridisation. Analysis of the DNA-6 replicative forms showed a significant decrease of approximately 41% for the F1 iteron mutant and 61% for the R iteron mutant in comparison with native levels. However, the mutation in the F2 iteron caused the most dramatic effect, decreasing replication to levels barely detectable by Southern hybridisation. These results suggest that the three iterons all play a role in BBTV replication, most likely as recognition and binding sites for the M-Rep, but that the F2 iteron appears to be the most important in replication. Following the observation that all BBTV isolates sequenced to date have identical iteron sequences, the extent to which the M-Rep would recognise, bind and initiate replication of heterologous components from geographically diverse BBTV isolates (the South Pacific and the Asian groups) was evaluated. Cross replication assays revealed that heterologous M-Reps from Fiji, Hawaii (South Pacific group) and Vietnam (Asian group) were able to initiate replication of the coat protein-encoding component (DNA-3) from the Australian BBTV isolate (South Pacific group). However, replication of DNA-3 from the Vietnamese isolate was not initiated by heterologous M-Reps from the two South Pacific isolates tested (Australia and Hawaii). These results suggest that a broad-range transgenic resistance strategy based on replication using Australian BBTV intergenic regions may be successful as this region will be recognised by the M-Reps from both Asian and South Pacific BBTV isolates. ▼ One approach to reducing the yield losses caused by banana viral diseases is the use of genetic engineering and pathogen-derived resistance strategies to generate resistant cultivars. The development of transgenic virus resistance requires an efficient banana transformation method, particularly for commercially important 'Cavendish' type cultivars such as 'Grand Nain'. Prior to this study, only two examples of the stable transformation of banana had been reported, both of which demonstrated the principle of transformation but did not characterise transgenic plants in terms of the efficiency at which individual transgenic lines were generated, relative activities of promoters in stably transformed plants, and the stability of transgene expression. The aim of this study was to develop more efficient transformation methods for banana, assess the activity of some commonly used and also novel promoters in stably transformed plants, and transform banana with genes that could potentially confer resistance to banana bunchy top nanovirus (BBTV) and banana bract mosaic potyvirus (BBrMV). A regeneration system using immature male flowers as the explant was established. The frequency of somatic embryogenesis in male flower explants was influenced by the season in which the inflorescences were harvested. Further, the media requirements of various banana cultivars in respect to the 2,4-D concentration in the initiation media also differed. Following the optimisation of these and other parameters, embryogenic cell suspensions of several banana (Musa spp.) cultivars including 'Grand Nain' (AAA), 'Williams' (AAA), 'SH-3362' (AA), 'Goldfinger' (AAAB) and 'Bluggoe' (ABB) were successfully generated. Highly efficient transformation methods were developed for both 'Bluggoe' and 'Grand Nain'; this is the first report of microprojectile bombardment transformation of the commercially important 'Grand Nain' cultivar. Following bombardment of embryogenic suspension cells, regeneration was monitored from single transfom1ed cells to whole plants using a reporter gene encoding the green fluorescent protein (gfp). Selection with kanamycin enabled the regeneration of a greater number of plants than with geneticin, while still preventing the regeneration of non-transformed plants. Southern hybridisation confirmed the neomycin phosphotransferase gene (npt II) was stably integrated into the banana genome and that multiple transgenic lines were derived from single bombardments. The activity, stability and tissue specificity of the cauliflower mosaic virus 358 (CaMV 35S) and maize polyubiquitin-1 (Ubi-1) promoters were examined. In stably transformed banana, the Ubi-1 promoter provided approximately six-fold higher p-glucuronidase (GUS) activity than the CaMV 35S promoter, and both promoters remained active in glasshouse grown plants for the six months they were observed. ▼ Banana bunchy top virus (BBTV) causes one of the most devastating diseases of banana. Transgenic virus resistance is now considered one of the most promising strategies to control BBTV. Pathogen-derived resistance (PDR) strategies have been applied successfully to generate plants that are resistant to numerous different viruses, primarily against those viruses with RNA genomes. BBTV is a circular, single-stranded (css) DNA virus of the family Nanoviridae, which is closely related to the family Geminiviridae. Although there are some successful examples of PDR against geminiviruses, PDR against the nanoviruses has not been reported. Therefore, the aim of this thesis was to investigate the potential of BBTV genes to interfere with virus replication when used as transgenes for engineering banana plants resistance to BBTV. The replication initiation protein (Rep) of nanoviruses is the only viral protein essential for viral replication and represents an ideal target for PDR. Therefore, this thesis focused on the effect of wild-type or mutated Rep genes from BBTV satellite DNAs or the BBTV integral genome on the replication of BBTV in banana embryogenic cell suspensions. A new Rep-encoding satellite DNA, designated BBTV DNA-S4, was isolated from a Vietnamese BBTV isolate and characterised. When the effect of DNA-S4 on the replication of BBTV was examined, it was found that DNA-S4 enhanced the replication of BBTV. When the replicative capabilities of DNA-S4 and the previously characterised Rep-encoding BBTV satellite, DNA-S1, were compared, it was found that the amount of DNA-S4 accumulated to higher levels than DNA-S1. The interaction between BBTV and DNA-S1 was also examined. It was found that over-expression of the Rep encoded by DNA-S1 using ubi1 maize polyubiquitin promoter enhanced replication of BBTV. However, when the Rep-encoded by DNA-S1 was expressed by the native S1 promoter (in plasmid pBT1.1-S1), it suppressed the replication of BBTV. Based on this result, the use of DNA-S1 as a possible transgene to generate PDR against BBTV was investigated. The roles of the Rep-encoding and U5 genes of BBTV DNA-R, and the effects of over-expression of these two genes on BBTV replication were also investigated. Three mutants of BBTV DNA-R were constructed; plasmid pUbi-RepOnly-nos contained the ubi1 promoter driving Rep expression from DNA-R, plasmid pUbi-IntOnly-nos contained the ubi1 promoter driving expression of the DNA-R internal gene product (U5), while plasmid pUbi-R.ORF-nos contained the ubi1 promoter driving the expression of both Rep and the internal U5 gene product. The replication of BBTV was found to be significantly suppressed by pUbi-RepOnly-nos, weakly suppressed by pUbi-IntOnly-nos, but strongly enhanced by pUbi-R.ORF-nos. The effect of mutations in three conserved residues within the BBTV Rep on BBTV replication was also assessed. Le génome du bananier (Musa sp.) est envahi par un nombre important de séquences de Banana streak virus (BSV), virus à ADN double brin de la famille des Caulimoviridae qui n'a aucune étape d'intégration au génome hôte au cours de son cycle de multiplication. La majorité de ces intégrations eBSV (endogenous BSV) est défective mais certaines sont restées fonctionnelles et peuvent être à l'origine de particules virales suite à des stress. L'objectif de ce travail de thèse est de préciser si les eBSV sont maintenus ou non dans le génome de Musa balbisiana des bananiers et d'étudier les conséquences évolutives que cela engendre. Nous avons tout d'abord caractérisé les eBSV pour trois espèces de BSV (Banana streak goldfinger virus (BSGFV), Banana streak obino l'ewai virus (BSOLV), Banana streak imove virus (BSImV) présents dans le génome du bananier modèle M. balbisiana cv Pisang Klutuk Wulung (PKW). Nous avons montré que les intégrations eBSGFV et eBSOLV étaient di-alléliques avec un seul allèle fonctionnel à chaque fois, contrairement à eBSImV qui est mono-allélique et pour lequel nous n'avons pas pu identifier l'allèle à l'origine de l'infection. Leur contexte génomique d'intégration diffère avec une co-localisation d'eBSGFV et d'eBSOLV sur le chromosome 1 et d'eBSImV sur le chromosome 2. Ces résultats nous ont permis de développer les outils moléculaires nécessaires à la caractérisation de ces trois eBSV dans la diversité de M. balbisiana. Cette caractérisation a révélé la diversité de structures des eBSV et éclairé une partie encore inconnue de la phylogénie de l'espèce M. balbisiana. Dans un second temps nous avons étudié les mécanismes de régulation des eBSV. Ce travail a porté sur les mécanismes d'ARN interférent pouvant expliquer le maintien des eBSV dans le génome des bananiers. Cette analyse révèle que les eBSV sont effectivement sous contrôle d'un mécanisme de type ARNi et la forte production de petits ARNs de 24nt ciblant les eBSV suggère qu'il s'agit d'un silencing au niveau transcriptionnel (TGS). En parallèle, nous avons aussi recherché les mécanismes mis en place par les bananiers non-porteurs d'eBSV en cas d'infection afin de connaître les défenses constitutives des bananiers face à une attaque virale BSV. Nous avons, sur la base de ces résultats, proposé un modèle de régulation des eBSV et des BSV et discuté de l'impact que ces mécanismes auraient pu avoir sur l'évolution des eBSV. L'ensemble des données de ce travail ont permis de préciser les étapes évolutives qu'ont connues les eBSV dans le génome du bananier, expliquant le maintien que l'on observe aujourd'hui.

The nuclear genome of banana plants is invaded by numerous viral sequences of banana streak virus (BSV), a DNA virus belonging to the family Caulimoviridae which does not require integration for its replication. Banana bunchy top virus (BBTV), a plant DNA virus in the family Nanoviridae, is transmitted by the banana aphid, Pentalonia nigronervosa, in a circulative nonpropagative manner. Information available on the interactions between BBTV and its vector is very limited. The goal of this work is to foster a basic understanding of BBTV-aphid interaction that will allow the generation of new options for the management of the disease. I have developed an immunofluorescence assay that allowed us to specifically localize BBTV into the anterior midgut (stomach) and into specific cells forming the principal salivary glands within the aphid vector. In order to examine the translocation and retention of BBTV into the aphid vector, I used timecourse experiments and transmission assays coupled with real-time PCR and immunofluorescence assays on dissected aphid tissues. These results indicate that BBTV is internalized into the anterior midgut in which it accumulates and is retained at concentrations higher than either in the haemolymph or in the principal salivary glands. I document that the anterior midgut and principal salivary glands can have direct contact within the aphid body, suggesting a possible haemolymph-independent translocation path. Through additional experiments, I have analyzed the interaction between GroEL proteins produced by the endosymbiotic bacterium Buchnera aphidicola, harbored within the aphid, and BBTV viral particles. Interaction assays were performed using immunocapture PCR, dot-blot, and far-western blot assays. However, I failed to obtain evidence of BBTV-GroEL interaction. Furthermore, I have used transmission assays, real-time PCR and immunofluorescence to analyze the localization, accumulation, and transmission efficiency of BBTV across 4 different lineages of Pentalonia, including Pentalonia caladii, a species colonizing heliconias, ginger and taro. Immunofluorescence assays showed similar BBTV translocation pattern among the 4 Pentalonia lineages. The results obtained showed for the first time that P. caladii is a competent vector of BBTV. Finally, I used fluorescent markers to study the process of cellular internalization in guts and salivary glands of P. nigronervosa. The data obtained suggests that BBTV does not use receptor-mediated endocytosis as described for aphid-trasnmitted luteoviruses. Therefore, I discuss the possible alternative mechanisms of virus internalization.

Une étude de prévalence des espèces BSOLV, BSGFV et BSIMV a montré que les espèces BSOLV et BSGFV sont très présentes dans les variétés hybrides interspécifiques MxH et FHIA-21 et que le niveau de prévalence de l’espèce BSGFV est significativement plus élevé chez FHIA-21 que chez MxH. Des résultats complémentaires suggèrent que la transmission des espèces virales BSOLV et BSGFV chez MxH et FHIA-21 résulte principalement de l’activation d’allèles eBSV infectieux plutôt que d’une transmission vectorielle par cochenille. La cinétique d’activation des allèles infectieux OL1 et GF7 dans les variétés MxH et FHIA-21 a été suivie durant 15 mois en conditions de culture au champ. Cette étude montre que les allèles infectieux OL1 et GF7 ne sont pas exprimés au même niveau ni dans les variétés MxH et FHIA-21ni pour une même variété (FHIA-21) et que le niveau d’activation dépend du mode de multiplication du matériel végétal. Des résultats préliminaires suggèrent que les infections par les espèces BSOLV et BSGFV n’ont que peu d’incidence sur la production fruitière chez les variétés MxH et FHIA. Globalement, cette thèse apporte une contribution significative à l’évaluation et à la gestion du risque BSV associé à la culture à grande échelle de variétés interspécifiques de bananier, notamment par la mise au point d’approches d’évaluation de ce risque transposables à d’autres variétés.

This thesis focuses on the risk of spreading Banana streak viruses in the Dominican Republic through the large scale cultivation of hybrid varieties Macho x Hembra and FHIA-21, which both harbor infectious eBSVs. An unprecedented survey showed that BSOLV and BSGFV are widespread in both varieties, with BSGFV being the most prevalent species, and that BSIMV is not present. BSGFV prevalence level was significantly higher in FHIA-21 than in MxH. Analyses of molecular taxonomical data of the natural mealybug vectors of BSVs and eBSV patterns of MxH and FHIA-21 were carried out and pointed to a marginal role of mealybugs in the transmission of BSGFV and BSOLV in the Dominican Republic in MxH and FHIA-21.The kinetics of activation of infectious eBSOLV and eBSGFV in MxH and FHIA-21 was monitored under field conditions. Results collected over 15 months showed that infectious alleles OL1 (BSOLV) and GF7 (eBSGFV) are differentially expressed in MxH and FHIA-21and that the mode of multiplication of the planting material influences expression levels. Preliminary results also suggest that BSV infection does not have a major effect on fruit production. Overall, this thesis contributes significantly to the development and implementation of appropriate strategies for evaluating and mitigating the risks of spreading BSVs that are associated with the cultivation of banana interspecific hybrids.

SANTRAUKA Potato mop-top virus, priklausantis Pomovirus genčiai ir Tobacco rattle virus, priklausantis Tobravirus genčiai – tai dirvožemiu plintantys virusai. Tai patogenai, kurie plačiai paplitę pasaulyje. Pastaruoju metu vis labiau susidomėta šiais virusais, mat jie sukelia daug žalos daugeliui augalų, tame tarpe ir labai svarbių kultūrinių augalų. Su šiais patogenais kovoti yra labai sudėtinga, taigi iškyla labai rimta problema, kurią bandoma išspręsti. Todėl šiame darbe ir buvo siekiama susipažinti su šiais virusais, jų pernešėjais bei padaryti pirmuosius žingsnius, ieškant būdus kovai su šais patogenais. PMTV – tai patogenas, kuriam nebūdingas gausus šeimininkų ratas. Jis infekuoja tik nedaugelį augalų rūšių. Tai viena iš savybių, kuria skiriasi nuo TRV. Pastarasis priešingai, pasižymi didele augalų-šeimininkų gausa. Šie virusai labai panašūs tuom, kad daugelyje augalų jie sukelia panašius simptomus bei pažeidimus. Net indikatoriniuose augaluose jų sukeltos žaizdos yra labai panašios. Taigi, susiduriama su problema, mat identifikuoti juos vizualiai yra labai sudėtinga. Tik N. benthamiana indikatorinis augalas gali padėti atskirti šiuos virusus: PMTV jame sukelia sisteminę reakciją, sisteminė mozaika išplinta po visą augalą, o TRV sukelia tik vietines žaizdas. Šiame baigiamajame darbe buvo siekiama identifikuoti PMTV bei TRV molekuliniais metodais. Identifikacijoje buvo pritaikytas imunofermentinis metodas – ELISA. SUMMARY Potato mop-top virus (genus Pomovirus) and Tobacco rattle virus (genus Tobravirus) – soil born viruses. These pathogenes are distributed worldwide and can significantly reduce the quality and yield of potato and other horticultural plants. So in recent years more and more people are interesting in these viruses. These viruses are responsible for economic losses in potato crops. To control the spread of viruses is vary difficult, so nowadays it is a serious problem, which everybody wants to solve it. Virus control depends on vector management, so the main purpose of this article was to know more about these viruses and thier vectors and to do first steps to finding ways of viruses’ suitable control measures. PMTV has narrow range of hosts. On the contrary TRV has a very wide natural host range. So it is one of the differences from these soil borne viruses. However diagnosis is further complicated by the fact that TRV and PMTV are very similar viruses, even they can cause similar symptoms to indicator plants. So to diagnose reliable these viruses by visual symptoms are very difficult. ▼ The C4 protein of Beet curly top virus (BCTV-B [US:Log:76]) induces hyperplasia in infected phloem tissue and tumorigenic growths in transgenic plants. The protein offers an excellent model for studying cell cycle control, cell differentiation and plant development. Expression of the C4 transgene, under an inducible promoter, in Arabidopsis thaliana plants (line IPC4-28) lead to extensive cell division in all tissues types examined, and radically altered tissue layer organization and organ development. Induced seedlings failed to develop shoot and root meristems, vascular tissue, true leaves and lateral roots. Exogenous application of brassinosteroid and abscisic acid weakly rescued the C4-induced phenotype, while induced seedlings were hypersensitive to gibberellic acid and kinetin. The C4 protein interacts in planta with three closely related members of the Arabidopsis SHAGGY-like protein kinase (AtSK21, AtSK22 and AtSK23) family, which are negative regulators of brassinosteroid (BR) hormone signaling. AtSK21 is known to hyperphosphorylate BES1 and BZR1 transcription factors, thereby inhibiting their function. The expression of C4 in IPC4-28 plants resulted in dephosphorylation of the BES1 transcription factor and changes in the expression levels of BES1 and BZR1 target genes. These changes were similar to those that occur when AtSK21 is inactivated by BR hormone treatment. Using several approaches a putative phosphorylation site (Ser at residue 49) on C4 is shown to be required for the C4/AtSK interaction. Mutation of Ser49 to Ala abolishes the C4/AtSK interaction, loss of the C4-expression phenotype, and failure to induce changes in the BR signaling pathway. Substitution of Ser49 by Thr restores both the C4/AtSK interaction and the effects of C4 on plant development and BR signaling. An active kinase is also required for the interaction, as AtSK kinase dead mutants fail to interact with C4. Taken together this indicates that C4 is a substrate for AtSK kinase activity and functions, at least in part, through disruption of the BR signaling pathway.

▼ Sugarcane bacilliform virus (SCBV) is an economically important pathogen of sugarcane in Australia which limits access to foreign sugarcane germplasm. Although SCBV is present in the major cane growing regions worldwide, very little is known about its variability, virulence and the yield losses resulting from infection. The limited information on SCBV has resulted in quarantine measures being introduced to protect the Australian sugarcane industry, with a major consequence being restricted access to imported sugarcane germplasm for breeding programs. Foreign sugarcane germplasm plays an important role in breeding of new commercial varieties for the Australian sugar industry and is essential for the long term productivity, profitability and sustainability of the sugar industry. This study was aimed at characterising Australian isolates of SCBV to enable the development of reliable and robust molecular and/or antibody-based diagnostic tests which could be used to not only assess the impact of SCBV on the Australian sugarcane industry, but could also be used to screen imported sugarcane germplasm for the virus. SCBV virions (SCBV-IM) were purified from the sugarcane accession "Ireng Maleng" and the dsDNA genome was cloned and sequenced. The genome of SCBV-IM comprised 7687 bp with an organisation typical of other badnaviruses. When the entire nucleotide sequence of SCBV-IM was compared to that of the Moroccan SCBV isolate (SCBV-Mo), less than 75% similarity was present. Within the coding regions, ORF I, ORF II and ORF III had 83%, 71% and 73% nucleotide similarity to SCBV-Mo, respectively. At the amino acid level, ORFs I, II and III from SCBV-IM showed 91%, 84% and 85% similarity to the equivalent regions in SCBV-Mo, respectively. To further investigate the level of sequence variability within Australian SCBV isolates, virions were purified from three further sugarcane accessions and a 220 bp fragment of the reverse transcriptase-coding region was amplified. Five clones from each sub-population were selected and sequenced. Analysis of these sequences revealed considerable variability in the virus population with variability within one plant as great as it was between isolates. However, since the use of specific primers could also be selecting for a sub-population of SCBV sequences, it was possible that the variability may actually be greater than that reported. These results indicated that SCBV isolates are complex and variable and may represent a continuum of genetic variability. High molecular weight DNA species larger than the SCBV 7.6 kbp unit-length genome were found in DNA extracted from purified SCBV-IM virions. We confirmed that these high molecular weight nucleic acids were virus-specific and open circular in conformation. Using field inversion gel electrophoresis (FIGE), the SCBV-IM DNA was separated into four discrete bands with sizes ranging from between 1 to 4 genome copies. ▼ Abstract: The green banana biomass is a component that can be industrially applied to a wide variety of foods because do not interfere in the sensory attributes of other ingredients present and has functional properties, especially the presence of resistant starch. This use can be expanded, including for export, if this product be produced in powder form. This research presents a feasibility study of drying green banana biomass in a non-commercial unit (experimental) of spray dryer with rotary atomizer. The variables selected in the experimental procedure were: the type of atomizer (type A, hole in a rectangular shape with semicircles at the ends and the total area of 162.88 mm2 and type B, hole in a circular shape and a total area of 254.47 mm2); the suspension concentration; the atomizer rotation; the biomass temperature and the biomass flow. The chamber temperature was set at 140ºC with air at 0.013 kg/s. It was defined as response the mass and the relative humidity of the product. These variables had two levels of variation, which corresponded to the completion of 32 trials. Among the variables used in the tests, the suspension concentration was the most significant to responses of mass and humidity, the others showed low effects. The test number 13 presented the best conditions of responses (9.17% and 12.656 g), was obtained with the atomizer type B, 50% concentration, atomizer rotation of 27,000 rpm, biomass flow rate of 40 mL/min and biomass temperature of 30°C. For this sample was realized a microscopic analysis, the granules size do not exceeded 500 ?m, therefore can be used in various food preparations. From the 13 test, the diameter calculation of the drying chamber for a production of 25 kg / h resulted in 5.14 m. O Brasil é o quarto produtor mundial de banana, com uma produção anual de 6,99 milhões de toneladas. É uma fruta de grande importância econômica e social, visto que é cultivada de Norte a Sul do país, garantindo emprego, renda e alimento para milhões de brasileiros durante o ano inteiro. No Paraná é a terceira fruta mais produzida no estado, com área de 9.900 ha. A maior parte da produção brasileira é destinada ao mercado interno, visto que é a segunda fruta mais consumida no país, e também devido à baixa qualidade da maior parte do produto. Essa baixa qualidade é o reflexo da falta de tecnologia das condições em que é cultivada, desde a condução na lavoura até a colheita. Uma tecnologia já utilizada em outras culturas mas pouco difundida entre os produtores de banana é o uso de plantas de cobertura para proteção do solo contra a erosão. Esse manejo é importante principalmente na implantação do bananal, pois até o início da produção decorre um tempo de cerca de 13 meses no qual o solo fica descoberto, exposto a erosão. Outra tecnologia importante para a qualidade das frutas é o ensacamento dos cachos logo após a sua formação, permanecendo como proteção até a colheita. Apesar dessa técnica ter vantagens já comprovadas em outras condições, para o estado do Paraná não existem dados relativos ao uso do ensacamento dos cachos. Assim, o trabalho foi dividido em dois projetos, ambos realizados no norte do Paraná. O objetivo do primeiro foi avaliar os efeitos da utilização de adubação verde de inverno, no estabelecimento de um pomar de bananeiras; e o segundo foi avaliar o efeito do ensacamento de cachos de banana, e o seu custo para o agricultor.

Brazil is the fourth largest producer of bananas, with an annual production of 6.99 million tons. Banana is a fruit of great economic and social importance, since it is grown from North to South of the country, generating jobs, income and food for millions of Brazilians, throughout the year. It is the third most produced fruit of the state of Paraná, with an area of 9,900 ha. Most of the Brazilian s production is destined for the domestic market, since it is the second most consumed fruit in the country, and also due to the low quality of most of the product. Such a poor quality is due to the lack of technology of the conditions in which it is grown, from the planting to the harvest. A technology which has already been used in other crops, but it is still not well known among banana producers, is the use of cover crops for soil protection against erosion. This management is particularly important for the implementation of the banana crops, because until the beginning of production, follows a period of about 13 months in which the ground is bare, exposed to erosion. Another important technology for the quality of fruit is the bagging of bunches soon after its formation, protecting until the harvest. Dentre as principais frutas produzidas no Paraná destaca-se a banana, que apresenta o desenvolvimento maior durante o período quente e úmido do ano e menor no período frio e seco. Com isso, a oferta do produto é maior durante os meses mais quentes, resultando em valores menores pagos pela produção, contrariamente ao que ocorre nos meses mais frios. O objetivo deste trabalho foi avaliar a influência da época de emissão de rebentos sobre o desenvolvimento da fase vegetativa e o vigor da bananeira Grande Naine. O trabalho foi realizado em um pomar comercial, localizado no município de Ibiporã, PR. Os tratamentos foram determinados pela época de emissão dos rebentos, mensalmente, ao longo do ano, totalizando 12 épocas. A área foi monitorada mensalmente, com avaliação de altura da planta e perímetro do pseudocaule. O delineamento experimental foi o inteiramente casualizado, com 12 tratamentos (meses do ano), com uma planta por parcela e 15 repetições. Os rebentos emitidos em agosto, setembro, outubro e novembro apresentaram desenvolvimento vegetativo mais rápido do que os demais, sendo que aos cinco meses de idade não diferiram estatisticamente entre si em altura e perímetro do pseudocaule. Aos dez meses de idade os rebentos emitidos em agosto e setembro foram superiores aos dos demais tratamentos para altura da planta, e os tratamentos agosto, setembro, outubro, novembro e dezembro foram superiores ao demais para a variável perímetro de pseudocaule. Nas condições em que este trabalho foi desenvolvido, conclui-se que a época de emissão dos rebentos influencia a duração da fase vegetativa de desenvolvimento de bananeira Grande Naine e o vigor das plantas.

Banana is among the main fruits produced in Paraná State. The plant development is higher during the hot and humid season and lower during cold and dry season. The fruit offering is higher during the warmer months, resulting in lower values paid by production, contrary to what occurs in the colder months. The aim of this study was to evaluate the influence of time of emergence of shoots on the vegetative development and on the plant vigor of Grand Nainebanana plants. The work was performed in a commercial Grand Naine banana orchard, in the municipality of Ibiporã (PR). The treatments were determined by the time of shoot emergence on a monthly basis throughout the year, totaling 12 times. The area was monitored monthly, with evaluation of plant height, and pseudostem perimeter. The experimental design was completely randomized, with 12 treatments (months of the year), one plant per plot and 15 replications. Plants that have emerged in August, September, October and November showed a faster development than the others, and at the age of five months did not differ significantly in plant height and pseudostem perimeter, but at the age of ten months the plants emerged in August and September were higher than those from other treatments for plant height; and plants from treatments August, September, October, November and December were higher than the others for…
