▼ This study examined the prevalence, trends, and predictors of non-evidenced based
atypical antipsychotic use in commercially insured children with mental health conditions between 2003 and 2009. A national administrative claims database was used to identify a sample of 150,272 children with a mental health condition who received a prescription for an
atypical antipsychotic medication. Generalized estimating equation models were developed to identify patient factors associated with non-evidenced based use. Non-evidenced based use was common in this sample (34%-62% of subjects depending on the specific medication). Adjustment reaction, anxiety disorder, attention-deficit disorder, conduct disorder, depression, eating disorders, obsessive-compulsive disorder, autism spectrum disorders, and tic disorder were all significant predictors of non-evidenced based use. Further studies that evaluate the temporal relationship between the diagnosis, prescription, and presence of an inpatient admission as well as provider factors affecting non-evidenced based use of these drugs, should be considered.

▼ Nontuberculous mycobacteria (NTM) have been found to be widely dispersed in the environment, being considered potentially pathogenic for humans and animals, and there are rare reports of their human to human transmission. Potential transmission modes of NTM to humans and animals are water and aerosols. Hospitalized patients with NTM for the period 2003 – 2013 and drinking water samples from their residence areas for the period 2010-2013 were studied. Same methods as those used for the isolation and identification of M. tuberculosis used for the isolation and identification of NTM, both in clinical specimens as well as in drinking water samples. Two different chemometric methods, Cluster Analysis and Factor Analysis, were first used in the literature to analyze the data matrix consisted of the parameters: NTM in patients, NTM in drinking water, patient gender, source of the biological sample and sampling area of drinking water. Totally, 367 hospitalized patients who lived at 30 different localities in the Prefecture of Larissa (central Greece) were tested positive for NTM. The source of the NTM isolates was respiratory specimens from pulmonary clinic (40.9%), samples from gastrointestinal clinic (3.0%); pathology clinic (45.5%); and rheumatology clinic (3.3%), while the remaining of the samples came from other clinics. In the 383 NTM isolates from the clinical specimens, the most frequently isolated NTM species were M. fortuitum (n=118, 30.8%), M. gordonae (n=87, 22.7%), M. peregrinum (n=46, 12.0%), M. chelonae (n=11, 2.9%); M. avium (n=8, 2.1%), M. intracellulare (n=7, 1.8%), while 88 (23.0%) of these isolated were not identified. Noted that M. tuberculosis isolated with NTM in 8 patients and 2 NTM were isolated together in 15 patients while 3 NTM together were isolated in 1 patient. Additionally, 3360 drinking water samples were collected from the 30 localities and analyzed for the period 2010 to 2013, and 11.2% of them found NTM positive. Cluster Analysis and Factor Analysis results show that the NTM found in patients and those found in the drinking water samples correlated to each other. No other association was found between the parameter “NTM” and the other studied parameters of drinking water (physicochemical, biological and microbiological quality parameters of drinking water). These results provide some indications that drinking water could be linked with NTM cases in humans.

▼ The plastoglobule (PG) is an enigmatic lipoprotein particle in photosynthetic and nonphotosynthetic plastids in plants. In chloroplasts, PGs are attached to the thylakoid membrane, providing a dynamic reservoir for prenyl- and neutral-lipids and a small set of proteins. Identification of the PG proteome suggests an active role for PGs in chloroplast metabolic processes. However, the majority of the proteins of the PG have not been characterized, making a comprehensive understanding of PG function difficult. The objective of this dissertation was therefore to clarify the functional role of the PG in chloroplasts. The PG proteome from Arabidopsis thaliana leaf chloroplasts was determined by mass spectrometry of isolated PGs and quantitative comparison with the proteomes of unfractionated leaves, thylakoids and stroma. This refined PG proteome consisted of 30 proteins, including 6 ABC1K
atypical kinases and 7 fibrillins together comprising more than 70% of the PG protein mass. Compared to previous PG proteome analyses, several proteins were excluded and six new PG proteins were identified, including an M48 metallopeptidase and two additional ABC1K
atypical kinases, confirmed by immunoblotting. A genome-wide co-expression network for the PG genes was constructed from mRNA expression data revealing a network with four distinct modules that each contain at least one ABC1K and/or fibrillin gene. Each module showed clear enrichment in specific functions, including chlorophyll degradation/senescence, isoprenoid biosynthesis, plastid proteolysis and plastid redox and kinase regulators of electron flow. A new testable model for the PGs is presented, in which sets of genes are associated with specific PG functions. The PG is proposed to serve as a thylakoid membrane microdomain, recruit ing low-abundant proteins into physical proximity facilitating specific metabolic reactions and physiological responses, comparable to lipid rafts of plasma membranes. Extensive phylogenetic evidence for the ABC1K family showed their ancient origin prior to the archaea-bacteria split and organization into three primary clades characterized by evolutionary origin and sub-cellular localization; ABC1Ks appear to have been introduced into photosynthetic eukaryote genomes through both plastid and mitochondrial endosymbiosis. Hypothesized functions of the PG-localized ABC1K1 and ABC1K3 as regulators of chloroplast prenyl-lipid metabolism are investigated. It was found that A. thaliana T-DNA insertion single (abc1k1 and abc1k3) and double (k1k3) mutants have a dose dependent, conditional light stress phenotype manifested as leaf degreening and necrosis; the minimal threshold for the visible phenotype is lower in the double mutant than either single mutant, suggesting additive or synergistic roles for each gene in the adaptation to excess excitation energy. ▼ The purpose of this study is to compare the experiences of men and women working in child care/early education fields. Men are in the minority in these occupations, so this thesis asks if men and women have different experiences at work through assessing if they have different perceptions of job growth, their relationships with supervisors, coworkers, and/or parents, and what each gender perceives the main challenges to be. If differences between men’s and women’s experiences are discovered, this study aims to understand if they are positive or negative for men and how they might impact job satisfaction. In-depth interviews with five men and four women were conducted and transcribed. Results show that men have higher perceptions of future job growth, but conversely experience fewer on-the-job friendships, feelings of isolation, and perceive their relationships with their charges’ parents to be weaker than their female counterparts. In turn, average job satisfaction was rated slightly lower for men than women in these fields. These somewhat contradictory findings indicate that men may experience both positive and negative outcomes of being the minority gender in these occupations.

▼ Atypical hemolytic uremic syndrome (aHUS) is a rare disease characterized by hemolytic anemia, thrombocytopenia, and renal impairment. Complement dysregulation is associated with most cases, especially deficiency in the regulator complement factor H (CFH). Recessive mutations resulting in loss of the sialyltransferase activity of ST3GAL1 were identified in three siblings with aHUS with normal systemic complement laboratory indices. To test whether ST3GAL1 deficiency causes complement activation by reducing available CFH binding sites, complement deposition was assessed by flow cytometry on endothelial cells with decreased sialic acid content. Treatment with neuraminidase, which cleaves sialic acids, and genetic knockout of ST3GAL1 did not cause complement activation. Global blockade of sialyltransferases, which previously caused kidney failure in mice, did not induce a phenotype in rats. Glycoproteins lacking sialic acid residues in ST3GAL1 knockout endothelial cells were isolated and identified using a pull-down approach followed by mass spectrometry, providing targets for future study.

▼ Cystic Fibrosis (CF) is a life threatening autosomal recessive disorder caused by a mutation in the Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) gene, leading to irregular secretions and inflammation in tubular organs. Disease manifestations of CF are heterogeneous in severity and can be present in the sinopulmonary, hepatic, gastrointestinal, and genitourinary tract. Since the 1960’s, physicians and scientists have described a less severe form of CF known as atypical CF, usually seen in adults. Patients with atypical CF tend to have one severe CF mutation on one chromosome, and one less common, mild CF mutation on their other chromosome; or have one severe mutation on one chromosome and an abnormal number of trinucleotide repeats in the CFTR gene on their other chromosome. Today, of the approximately 1000 patients diagnosed with CF per year in the United States, roughly 10% are diagnosed with the atypical presentation of the disease as adults. Patients suffering from atypical CF typically have only one organ system that is dysfunctional, and their clinical symptoms may be less severe than those of a classical case where there are two severe CF mutations. Common symptoms include idiopathic bronchiectasis, chronic sinusitis, congenital bilateral absence of the vas deferens (CBAVD), and idiopathic pancreatitis. Unlike patients suffering from the classical presentation of the disease, most are pancreatic sufficient – however the possibility of pancreatic insufficiency still exists. Patients with atypical CF represent a diagnostic challenge for physicians due to the mild, slowly progressing array of clinical symptoms, the general lack of knowledge about atypical CF, and the general association of CF as a childhood disease. Increasing physician awareness of the adult population with CF is a paramount in improving the diagnosis, care and treatment of patients with atypical CF. Missed diagnoses can result in hospital admissions and morbidity that may have been avoidable. The goal of this thesis is to describe the causes of CF, the common symptoms seen in both CF and atypical CF, the proper diagnosis of atypical CF, and to identify the therapies, both current and in development, used to treat atypical CF.

▼ This study aims to discover whether there is a correlation between atypical clinical presentations of Alzheimer’s disease (AD) and atypical distribution patterns of AD pathology. To provide a measure of the atypical clinical presentations, we obtained standardized neuropsychological test scores for a group of 345 subjects of the Boston University Alzheimer’s Disease Center cohort that had received a clinical or pathological diagnosis of AD. Each of the neuropsychological test scores included in our analyses was classified into one of five cognitive domains, according to the primary domain each test assesses: memory, executive function, attention, visuospatial function, and language. From these test scores, global cognitive performance scores and individual domain performance scores were calculated for a subset of 53 subjects that had brain tissue slides available for pathological analysis. Difference scores were computed for each domain, providing a within-subject comparison of performance between each individual cognitive domain and overall cognitive performance. For these same 53 subjects, tissue slides from six brain regions were obtained and digitally scanned. Neurofibrillary tangle (NFT) quantification was performed for all tissue slides using a computer algorithm modified to recognize AT8 staining patterns. NFT densities were then calculated for five general brain regions (frontal, parietal, temporal, limbic and occipital). In addition, a global NFT density score was computed for each subject, averaging NFT densities across all regions. From these densities, difference scores were calculated for each brain region individually, providing a measure of how each region’s NFT density compares to the overall brain NFT density. Multiple linear regressions analyses were performed with five pairs of cognitive domain difference scores and region NFT density difference scores: memory difference scores and limbic difference scores, executive function difference scores and frontal difference scores, attention difference scores and parietal difference scores, visuospatial difference scores and occipital difference scores, and language difference scores and occipital difference scores. Though we expected to observe significant negative correlations between each of the five difference score pairs, the only statistically significant correlation observed was between memory difference scores and limbic difference scores (β= -0.361, p<0.05). These results suggest that poorer performance in memory-related neuropsychological tests, when compared to global cognitive performance, can predict higher NFT densities in limbic regions when compared to the overall brain pathology. Although no other difference score pairs showed any statistically significant correlations, many study limitations, including small sample size and simplifications in analysis, should be addressed in the future to provide better understanding of these atypical presentations of AD and their underlying pathologies.

▼ Autism Spectrum Disorder (ASD) is characterized by pervasive delays in socialization, communication, and repetitive behaviors and restricted interests. While there is a growing body of evidence on the etiology of ASD, there are a limited number of studies examining factors which may impact the differentiation of ASD compared to other developmental disabilities. Additionally, few studies have examined factors which may predict level of ASD symptom severity. The primary aim of this study was to investigate whether premature birth occurs more commonly in infants and toddlers (17-37 months) with ASD than those with atypical development. A secondary aim of this study was to investigate whether length of gestation predicts scores on the Baby and Infant Screen for Children with aUtIsm Traits (BISCUIT), Part 1, a measure of overall impairment, in participants with ASD. Participants were separated into two groups (i.e., ASD, atypical development), and compared on the basis of parent/caregiver reported incidence of premature birth and average weeks of gestation. Additional analyses included within group comparisons for the ASD diagnostic group by separating individuals who were born prematurely and full term and analyzing their total scores on the BISCUIT-Part 1 were. Differences in overall level of functioning were assessed. Results of the current study indicate that infants and toddlers with atypical development are more likely to be born prematurely; however, average weeks of gestation did not significantly differ between the groups. Follow up analyses of participants with ASD revealed that premature birth was not a predictor of level of severity. Implications of these findings are discussed.

atypical antipsychotics helped improve the management of positive symptoms, many patients are still suffering from negative and cognitive symptoms and side effects. Serotonin-2A receptors have been implicated in the mechanism of action of
atypical antipsychotics.
Atypical antipsychotics such as olanzapine and clozapine are antagonists of 5-HT2A/Gαq/11 pathway, but activate the 5-HT2A/JAK2/STAT3 pathway. Studying the functional selectivity of
atypical antipsychotics at the 5-HT2A receptor may lead to a better understanding of the role that 5-HT2A receptors play in the therapeutic effect of
atypical antipsychotics. This dissertation investigates five analogs of olanzapine and the 5-HT2A receptor antagonist, ketanserin, for functional selectivity at the 5-HT2A receptor. A calcium mobilization assay was used to investigate the 5-HT2A/Gαq/11 pathway and immunoblotting was used to investigate the 5-HT2A/JAK2/STAT3. We identified three analogs that induced calcium mobilization and two analogs that prevented 5-HT2A receptor agonist induced calcium mobilization. We also found that four analogs induced phosphorylation of JAK2 and one analog that prevented 5-HT2A receptor agonist induced phosphorylation of JAK2. In the future, functionally selective analogs can be used to study the 5-HT2A/Gαq/11 and 5-HT2A/JAK2/STAT3 pathways separately in order to gain a better understanding of the role each individual pathway plays in the therapeutic effect of
atypical antipsychotics.

Partindo do case-study da Autoeuropa, pretendemos explorar o que a nós nos parece ser o futuro da negociação coletiva em Portugal, ou seja, uma maior intervenção das comissões de trabalhadores na contratação coletiva.
This dissertation explores the issue of collective bargaining. The Portuguese labor law has been through a crisis, which arouses increasing interest for the diffusion of new and diverse forms of atypical employment relationships. Based on the case study of Autoeuropa, we want to explore what seems to be the future of collective bargaining in Portugal, that is, a greater involvement of workers committees in collective bargaining.

O tremor epizoótico (TE) é uma encefalopatia espongiforme transmissível (EET) que ocorre naturalmente em ovinos, caprinos e muflões. É uma doença conhecida há mais de 250 anos. Caracteriza-se pela acumulação da isoforma patológica- proteína priónica resistente (PrPres ou PrPsc) de uma proteína fisiológica do hospedeiro- proteína priónica celular (PrPc) no sistema nervoso central, conduzindo à progressiva neurodegeneração e morte. O tremor epizoótico ou scrapie, representa o protótipo das doenças priónicas. Existem dois tipos de scrapie classificados até à data, o scrapie clássico e o scrapie atípico/Nor98. A suscetibilidade a qualquer dos tipos de scrapie é determinada por polimorfismos do gene priónico.
O scrapie clássico é uma doença infecciosa sendo que o scrapie atípico/Nor98 é visto como uma doença de ocorrência esporádica e dependente da mudança espontânea de conformação da proteína priónica. O tipo de scrapie mais prevalente em Portugal é o scrapie atípico/Nor98. Assim, elaborou-se o presente estudo retrospetivo com o intuito de determinar o potencial de ocorrência de scrapie no nosso país entre 1995 e 2002, examinando, pela técnica de imunohistoquímica (IHQ), amostras de tecido nervoso de pequenos ruminantes arquivadas no Laboratório de Patologia do Instituto Nacional de Investigação Agrária e Veterinária (INIAV), e contribuir para a caracterização epidemiológica desta doença no nosso país.
ABSTRACT - Retrospective study on scrapie in Portugal - Scrapie is naturally occurring transmissible spongiform encephalopathy (TSE) in sheep, goat and mufflons almost worldwide and it is known for about 250 years. It is characterized by the accumulation of an abnormal isoform- resistant prion protein (PrPres or PrPsc) of host encoded prion protein- cellular prion protein (PrPc) in the central nervous system leading to progressive neurodegeneration and death. Scrapie represents the prototype of the so-called prion diseases. It is observed to date as two types, classical and atypical/Nor98 scrapie. The susceptibility to both types is modulated by polymorphisms of the prion gene.
Whereas classical scrapie is clearly a naturally occurring transmissible disease, atypical scrapie seems to be caused by spontaneous misfolding of prion protein. The most prevalent type of scrapie in Portugal is the atypical/Nor 98 strain.
This retrospective study intends to find out the potential occurrence of scrapie in Portugal before 2002, examining through imunohistochemistry (IHC) samples of small ruminants archived from 1995 to 2002 in the Pathology laboratory from INIAV (Instituto Nacional de Investigação Agrária e Veterinária – The National Institute for Agricultural and Veterinary Research), contributing to the epidemiological characterization of the disease in this country.

▼ Atkinson proposed a concept of âflexible firmâ in 1984, and distinguished workers into three parts: core workers, peripheral workers and external workers. In 1997, Handy considered that organization would be composed of core work team, temporary employees, and flexible workers in the future. Therefore, Work in the organization would be divided into two parts: core work and non-core work. Core workers are responsible for core work, and non-core work are outsourced or distributed to other contractors. Then,
atypical employment rises and develops. The use of this kind of employment will become a trend, and companies can save money this way.
The purposes of this thesis are: (1) to understand the situation of
atypical employment in the Bureau of National Health Insurance (the NHI); (2) to find out the reasons of unionization for temporary workers in the NHI; (3) to understand the restraints on unionization of
atypical employees; (4) to provide suggestion to government and government-run companies.
According to the results of interviews and analysis, the reason that the NHI continues to employ temporary employees is to stabilize business. The NHI can continue to employ temporary employees since these temporary employees are not included into Labor Standards Law. Three reasons that temporary employees of the NHI can unionize are: (1) work time and worksites of temporary employees of the NHI are fixed; (2) insurance industries are covered by Labor Standards Law, and temporary employees of the NHI are included into Labor Standards Law; (3) organizational system of the NHI is a government-run financial and insurance organization, so it is not limited by the fourth clause of Labor Union Law.

▼ ãIn recent years, the industry environment has been affected by the globalization and the dramatic changes of industrial competition, thus the enterprises, considering cost, flexibility and speed, show a yearly rising employment rate of foreign workers in Taiwan and some of them even reach the top limit. In addition, they gradually stop employing full-time or long-term workers, but start to employ the outsourced and dispatched personnel. As a result, the traditional employment relationships are confronted with challenges. According to the survey issued by CHEER magazine in April, 2004, more than 12% enterprises possess 10~20% contract-based employees of their total staff, and in the future more than 35% enterprise will continue to enlarge the scale of contract-based employees. Hence, the long-term official employment relationship is no longer the only choice, the enterprises tend to use multiple and flexible employment modes.
ããFrom 2008 to now, after experienced the financial crisis, have the enterprises added the application of flexible manpower employment strategies, or examined and adjusted their current flexible employment strategies? Therefore, the study further explores the current situation of enterprisesâ application of flexible employment strategies.
ããThis study hopes to utilize the interview to collect the enterprisesâ current situation of flexible manpower employment, application reasons and effectiveness, as well as the relevant management regulations and practical operations derived from the application of flexible manpower employment in the enterprises, so as to explore the influences of flexible manpower employment strategies on the enterprises. Finally, it can be served as references for the enterprises of different fields to utilize the flexible manpower employment and the government to draft relative policies and labor laws in the future.
ããFrom the interview data, the flexible manpower mainly takes charge of the peripheral work like administrative support and elementary manufacturing or service works. However, it is extraordinary that the enterprises related to credit cards of financial service sector often employ the dispatched manpower, or assign the core business to the
atypical employees.
ããAs for the reasons for applying flexible employment, taking cost into consideration, serving as bridge to employ the regular workers are the primary. The benefits of flexible employment strategies are mainly shown in the cost reduction in welfare, training and recruiting and the flexibility of personnel application. During the financial crisis of 2008 when the case company conducted the organizational adjustment and downsizing, it primarily controlled and downsized the dispatched manpower by means of non-renewal of contracts upon labor termination or early termination of contract. Background: Diagnosing adipocytic tumors can be challenging because it is often difficult to morphologically distinguish between benign, intermediate and malignant adipocytic tumors, and other sarcomas that are histologically similar. Recently, a number of tumor-specific chromosome translocations and associated fusion genes have been identified in adipocytic tumors and atypical lipomatous tumors/well-differentiated liposarcomas (ALT/WDL), which have a supernumerary ring and/or giant chromosome marker with amplified sequences of the MDM2 and CDK4 genes. The purpose of this study was to investigate whether quantitative real-time polymerase chain reaction (PCR) could be used to amplify MDM2 and CDK4 from total RNA samples obtained from core-needle biopsy sections for the diagnosis of ALT/WDL. Methods: A series of lipoma (n = 124) and ALT/WDL (n = 44) cases were analyzed for cytogenetic analysis and lipoma fusion genes, as well as for MDM2 and CDK4 expression by real-time PCR. Moreover, the expression of MDM2 and CDK4 in whole tissue sections was compared with that in core-needle biopsy sections of the same tumor in order to determine whether real-time PCR could be used to distinguish ALT/WDL from lipoma at the preoperative stage. Results: In whole tissue sections, the medians for MDM2 and CDK4 expression in ALT/WDL were higher than those in the lipomas (P < 0.05). Moreover, karyotype subdivisions with rings and/or giant chromosomes had higher MDM2 and CDK4 expression levels compared to karyotypes with 12q13-15 rearrangements, other abnormal karyotypes, and normal karyotypes (P < 0.05). On the other hand, MDM2 and CDK4 expression levels in core-needle biopsy sections were similar to those in whole-tissue sections (MDM2: P = 0.6, CDK4: P = 0.8, Wilcoxon signed-rank test). Conclusion: Quantitative real-time PCR of total RNA can be used to evaluate the MDM2 and CDK4 expression levels in core-needle biopsies and may be useful for distinguishing ALT/WDL from adipocytic tumors. Thus, total RNA from core-needle biopsy sections may have potential as a routine diagnostic tool for other tumors where gene overexpression is a feature of the tumor.

▼ The present prospective study was undertaken to determine (a) the predominant bacterial and atypical pathogens in acute exacerbations of chronic obstructive pulmonary disease (COPD) using all three detection approaches ;serology, culture and polymerase chain reaction( PCR), (b) their implications in the inflammatory profile and functional impairment, and (c) the parameters predicting detection of a potentially pathogenic microorganism in sputum cultures.Patients’ data were collected for statistical evaluation. Conventional culture revealed at least one potentially pathogenic microorganism in 52 patients. The most common bacterial pathogens were Haemophilus influenzae (23.9% -22 patients in 17 cases as a single pathogen and 6 cases in combination with other pathogens) and Pseudomonas aeruginosa 14.1%, 13 patients , - in 10 cases as a single pathogen and in three cases in combination with other pathogens).PCR for atypical pathogens was positive in eight patients (five for M. pneumoniae and three for C. pneumoniae), six of which also yielded at least one bacterial pathogen in conventional culture (Table 3). These eight patients had no significant differences, regarding fever, peripheral neutrophilia, ESR, CRP, post FEV1%, FEV1/FVC%, duration of hospitalization, or final outcome, when compared with patients that yielded bacterial pathogens only.An infection due to Chlamydophila pneumoniae or Mycoplasma pneumoniae (using PCR and/or serology) was diagnosed in four and two patients, respectively, but PCR missed the pathogen detection in two of the C. pneumoniae cases. In addition, PCR detected atypical pathogen DNA in four more patients (one with C. pneumoniae and three with M. pneumoniae), which were attributed to carrier status. No patients were diagnosed with Legionella spp. infection. Acinetobacter baumannii and Pseudomonas aeruginosa positive cultures were associated with prolonged hospitalization and more severe airway obstruction of the patients (p=0.030 and 0.031, respectively). Multiple linear regression analysis revealed that increased WBC and CRP were independently associated with a positive culture for bacterial pathogen (p=0.014 and 0.001, respectively).

▼ La trypanosomose humaine africaine, maladie tropicale négligée sévit en Afrique subsaharienne dans les zones rurales reculées avec un accès difficile aux soins de santé. Les traitements disponibles sont coûteux et nécessitent d'importantes ressources humaines et logistiques. En dépit du manque des personnels de santé qualifiés dans les zones d'endémie, la population fait souvent recours à la médecine traditionnelle. Ainsi la recherche de nouvelles molécules actives semble nécessaire pour contribuer à l'effort de recherches d'alternatives thérapeutiques contre la THA. In vitro des plantes médicinales de la République Centrafricaine de la famille des combretaceae ont présenté une activité trypanocide sur T. brucei brucei.Plusieurs cas d'infection humaine par des trypanosomes parasites de l'animal ont été également rapportés. Le nombre de ces infections est probablement sous-estimé. Certaines sont généralement guéris d'une manière spontanée, d'autres nécessitent une prise en charge thérapeutique. Des médicaments conventionnels disponibles contre la THA ainsi que le fexinidazole en phase d'essai clinique et la combinaison NECT se sont révélés efficaces contre le T. lewisi (parasite naturel de rat, à l'origine de la trypanosomose humaine atypique) in vitro en comparaison avec T. brucei gambiense et in vivo chez le rat. Enfin, le taux d'arginase sérique, enzyme associée au THA est élevé chez les patients trypanosomés par rapport aux sujets sains du foyer. Ce taux est revenu à la normale après un traitement efficace. Ainsi, l'évaluation de l'arginase sérique peut donc être considérée comme un bio marqueur du suivi de l'efficacité du traitement.
The Human African Trypanosomiasis is a neglected tropical disease prevalent in sub-Saharan Africa mainly in remote rural with poor access to health care. Available treatments are expensive and require significant human and logistical resources. Despite the lack of skilled health workers in endemic areas, people often fall back on to traditional medicine. Therefore investigating for new molecules seems necessary to contribute to the effort of research for alternatives therapies against HAT. In vitro, medicinal plants from the combretaceae family in Central African Republic showed trypanocidal activity on T. brucei brucei.Several cases of human infection with trypanosome parasites of animals have also been reported. The number of infections is probably underestimated. Some are usually cured in a spontaneous way, others require therapeutic care. Conventional drugs available against the THA, NECT combination and fexinidazole in clinical trial phase proved to be effective against T. lewisi (natural parasite of rats, causing atypical human Trypanosomosis) in vitro compared with T. brucei gambiense and in vivo in rats. Finally, the level of serum arginase, enzyme associated with the THA, is high in Trypanosomosis patients compared to healthy subjects from the same area. ▼ Chemokines constitute a family of almost 50 small secreted cytokines, recognized by about 20 different 7TM spanning G protein coupled receptors (GPCRs), that activating pertussis toxin sensitive G proteins induce cell migration. These receptor are abundantly expressed by leukocytes and, controlling cell migration, they dictate leukocyte positioning during homeostatic patrolling within peripheral tissues, their maintenance in bone marrow during maturation and in addition mediate their recruitment to inflamed tissues. Upon inflammation in fact a number of chemokines are produced or activated by inflammatory mediators and diffuse within the tissue, generating a chemical gradient along which leukocytes migrate to reach the center of inflammation to contain and remove the insulting factor. This system needs an extremely tight control, since its dysregulation has been demonstrated to be at the basis of different inflammatory diseases, auto-immunity and has also been linked to cancer development. In particular, in this thesis we focused our attention on two regulatory system: post-translational modifications of chemokines, mediated by enzymes specifically released upon inflammation also by immune cells, and on the activity of
atypical chemokine receptors, a subfamily of chemokine receptors that despite high structural homology and similar binding properties compared to conventional chemokine receptors, are unable to drive chemotaxis but act instead as key regulators of the chemokine system activity. In detail we looked at the ability of these regulatory mechanisms to modulate chemokine signaling properties generating a biased signaling, an emerging feature of GPCR pharmacology that describes the ability of a given receptor to elicit different or even opposite functional activities depending on the ability of different agonists to stabilize different receptor’s active structural conformation, resulting in different phenotypes mediated by the same receptor. In the chemokine system biased signaling has already been described to occur on different receptors upon binding of their different ligands, therefore during our investigation on chemokine regulatory system signaling we maintained our focus on the ability of these systems to bias chemokine signaling properties in order to better understand how this regulation occurs.
To this point we assessed the ability of differently post-translationally modified chemokines to elicit signaling activities on different receptors by measuring in HEK293 cells their potential in inhibiting adenylyl cyclase, a proximal downstream signal of Gα inhibitory proteins activation, and in inducing β-arrestin recruitment to the receptors in energy transfer-based assays. We also compared signaling properties of an
atypical chemokine receptor to the ones elicited by a conventional receptor analyzing the phosphoproteome modifications occurring constitutively and after stimulation with the same agonist.

Breast cancer is the most prevalent cancer in women in the western world including Australia. Early detection and accurate diagnosis of new breast lesions is essential for appropriate medical management. Fine needle aspiration (FNA) is a cytological investigative tool commonly used to provide the initial pathological diagnosis of breast lesions. An atypical cytology report (C3) is an ambiguous or equivocal result. This uncertainty creates a dilemma and a more invasive investigative procedure such as core biopsy or incisional biopsy may be required, which comes at greater cost and anxiety to the patient. The aims of this project were to understand the true nature of C3, to determine the underlying causes of C3 and to devise a strategic approach to minimise its use without compromising the other cytological categories. The practical aims were to create a greater understanding of the issue and to produce a collective uniform approach to reporting atypical breast cytology cases thereby refining its use. The results of a blind rescreen of 256 consecutive C3 cases were statistically analysed. From these results, a cytomorphological approach to assess the risk of malignancy was developed and tested against a validation set of 230 subsequent C3 cases. Various strategies have been developed to reduce the incidence of the C3 category. Extrinsic factors can be easily reduced by greater involvement by cytology staff in the FNA procedure. Intrinsic factors can be understood and considered when allocating cases into C3. Specific diagnoses, such as papillary neoplasm can direct more appropriate definitive management. The cytologists in our institution have gained greater awareness of the atypical category by actively participating in the project and by having access to teaching resources and examples. The benefits manifest as financial, medical and social enhancements. It is hoped that some of these approaches will be taken up by other institutions in Australia and internationally.

▼ L’Eye Tracking est une méthode de mesure précise et non invasive des mouvements oculaires permettant d’accéder aux paramètres neurophysiologiques du système oculomoteur. L’analyse des mouvements oculaires est un outil qui contribue au diagnostic des syndromes parkinsoniens. En Guadeloupe, les syndromes parkinsoniens atypiques, sont présents en proportion anormalement élevée. Ils représentent environ 70% des syndromes parkinsoniens. Notre travail a été de réaliser des normes oculomotrices des sujets sains et de caractériser les syndromes parkinsoniens sur le plan oculomoteur à partir des normes oculomotrices réalisées. Les normes oculomotrices des sujets sains antillais différents de celles des témoins d’origine tchèque. Ces différences suggèrent la possibilité d’une variabilité des standards oculomoteurs entre deux populations de sujets sains. Nos résultats témoignent de la nécessité de constituer des groupes témoins spécifiques à la population étudiée lors des études cliniques et les travaux de recherche sur les mouvements oculaires. Ils soulèvent également la question de l’influence de facteurs génétiques et/ou environnementaux sur les paramètres oculomoteurs. Les patients porteurs d’une maladie de Parkinson originaires et résidants aux Antilles présentent des anomalies oculomotrices plus fréquentes qu’attendues avec notamment un taux élevé d’erreurs aux antisaccades. Cette caractéristique ne semble pas corrélée aux performances cognitives. Chez les parkinsoniens atypiques l’altération parfois sévère de l’ensemble des paramètres oculomoteurs est en accord avec les données cliniques et d’imagerie cérébrale et témoignent de lésions diffuses corticales fronto-pariétales et sous corticales.
Eye Tracking is a precise and non-invasive method of measuring eye movements that provides access to the neurophysiological parameters of the oculomotor system. Eye movement analysis is a tool that contributes to the diagnosis of parkinsonian syndromes. In Guadeloupe, atypical parkinsonian syndromes are present in an abnormally high proportion. They represent about 70% of Parkinson's syndromes. Our work has been to achieve oculomotor standards of healthy subjects and characterize the parkinsonian syndromes on the oculomotor level from oculomotor standards. The oculomotor standards of Caraibbean healthy subjects differ from those of Czech origin. These differences suggest the possibility of variability of oculomotor standards between two populations of healthy subjects. Our results demonstrate the need to establish control groups specific to the population studied in clinical studies and research work on eye movements. They also raise the question of the influence of genetic and / or environmental factors on oculomotor parameters. Patients with Parkinson's disease coming from and residing in the Caribbean have more frequent oculomotor abnormalities than expected with a high percentage of error in antisaccade task. ▼ Objective This retrospective longitudinal study investigated the relationship between readiness to change (The Transtheoretical Model of Stages of Change), parenting (e.g., parenting style, practices, and stress), and the use of
atypical antipsychotic medication on body mass index (BMI) in adolescents with obesity, most of whom were Hispanic. Methods Data were obtained from an existing clinical database of 41 adolescents between 12-18 years of age (Mean age 14.35 years, 39% male, 4.9% White, 19.5% Black, 63.4% Hispanic, 12.2% Other) who participated in outpatient behavioral weight management group therapy, that included motivational interviewing, once a week for a period of seven weeks. Multilevel Regression Modeling analysis was employed to examine within and between participant differences in BMI. Readiness to change was measured with the use of baseline self-report ratings on the University of Rhode Island Change Assessment (URICA). Parent ratings on the Parenting Relationship Questionnaire-Child and Adolescent (PRQ-CA) were obtained as a measure of parenting style, practices, and stress. Medication status (e.g., the use of
atypical antipsychotic medication) was collected for each individual. Results Although used by only two subjects
atypical antipsychotic medication was significantly related to higher BMI at baseline. Parenting confidence was significantly related to lower BMI at baseline. From week to week, there was not a significant fixed effect for rate of change in BMI within or between individuals; however, random effects for rate of change between individuals were present. Mean baseline BMI was lower for individuals who presented in the action stage of change than those who presented in the precontempation/contemplation stages. Conclusion Understanding the influence of individual readiness to change, parenting confidence, and the use of
atypical antipsychotic medication on adolescent weight status, may improve comprehensive assessment and interventions in the clinical treatment for adolescent obesity. Implications for professionals are discussed and directions for future research are highlighted. Furthermore, given the diverse demographic of the sample used in this study (63.4% Hispanic, 19.5% Black); multicultural considerations for adolescents with obesity are discussed.

▼ Delta-like homologue 1 (Dlk1) and Dlk2 encode vertebrate-specific transmembrane proteins belonging to the Jagged/Delta/Serrate family of Notch ligands. Murine Dlk1 is widely expressed during embryonic development and targeted deletion results in defects in numerous developmental processes, such as adipogenesis, haematopoiesis, neurogenesis and skeletal muscle formation. However, the mechanisms by which DLK1 regulates these processes remains unclear. The purpose of this project is to examine the function of these genes using zebrafish as an in vivo model, allowing insight to the ancestral functions of these genes. We have strong evolutionary evidence that dlk2 is the ancestral version of the gene from which dlk1 is derived; therefore, the thesis focuses primarily on the role of dlk2 in the zebrafish system. I initially examine the expression of zebrafish dlk1 and dlk2 during embryonic development and in the adult brain, determining similarities and differences between mouse and zebrafish. In particular, dlk1 and dlk2 in the fish exhibit a pattern that is more reminiscent of Dlk2 in the mouse. This developmental expression pattern is essential for the interpretation of the modulation of Dlk2 in later chapters, and is aided by the generation of a mammalian Dlk2 antibody that cross-reacts with zebrafish. We obtained a dlk2 mutant and used this line to examine the role of the DLK2 protein in development and in the adult brain. I demonstrate that, in the absence of DLK2, a population of neural precursor cells appear to over-proliferate early in zebrafish development. Later, by larval stages, these cells are absent, suggesting a premature activation and subsequent depletion of the progenitor cell pool in the mutant, reminiscent of the Dlk1 mutant in mouse. Associated with this phenotype are larval behavioral defects in motor response. In this thesis, it will be shown that in the adult dlk2 mutant zebrafish, the radial glial cell population in the telencephalon is completely depleted. These radial glial cells are thought to be responsible for adult neural regeneration in zebrafish, and our characterization of a mutant completely lacking this cell population provides a rich model to further examine and understand the functions of this well-studied but poorly understood cell population. These findings have both functional and evolutionary implications for the relative roles of these two vertebrate specific atypical Notch ligands.

atipični i anaplastični meningeomi, koji po klasifikaciji SZO
pripadaju tumorima gradusa II odnosno gradusa III. Atipični tumori
čine od 4,7% do 7,2%, a anaplastični od 1% do 2,8% svih meningeoma.
Do sada je poznato nekoliko činjenica važnih za prognozu ishoda
lečenja od kojih su tri najvažnija, a to su: histološki gradus,
ekstenzivnost hirurške resekcije i primena postoperativne zračne
terapije. Dijagnoza atipičnih i anaplastičnih (malignih) meningeoma
se postavlja procenom određenih histopatoloških parametara koji su
dati u klasifikaciji tumora SZO. Broj mitoza /10 polja velikog
uveličanja (PVU) je najznačajniji histološki kriterijum. Za
dijagnozu atipičnih meningeoma potrebno je da se u tumoru nađe ≥ 4
mitoze/10 polja velikog uveličanja ili da su prisutne bar tri od
sledećih histopatoloških karakteristika: povećana celularnost,
prisustvo sitnih ćelija sa visokim nukleo/citoplazmatskim odnosom u
korist jedra, difuzan tip rasta, prisustvo istaknutih jedaraca u
jedru i fokusi sitnih ili opsežnih nekroza. Za dijagnozu
anaplastičnih meningeoma potrebno je da se u tumoru nađe ≥ 20
mitoza/10 polja velikog uveličanja. Pored toga ovi tumori pokazuju
i jasne citološke karakteristike maligniteta i mogu imati izgled
sličan kao kod sarkoma, karcinoma i melanoma...

▼ Animal cells, as distinct as epithelia and migratory cells, have cell polarity that is defined by a common set of molecules. The Par complex polarizes the cortex of animal cells through the activity of
atypical protein kinase C (aPKC). In this work, I aimed to determine the mechanism of aPKC substrate polarization and identify common characteristics of aPKC substrates that are polarized by phosphorylation. I found that several diverse Par-polarized proteins contain short highly basic and hydrophobic motifs that overlap with their aPKC phosphorylation sites. These Phospho-Regulated Basic and Hydrophobic (PRBH) motifs mediate plasma membrane localization by electrostatics-based phospholipid binding when unphosphorylated but are displaced into the cytoplasm when phosphorylated. To assess whether the Par complex polarizes other proteins by this mechanism, I developed an algorithm to identify potential PRBH motifs and score these linear motifs for basic and hydrophobic character, as well as the quality and number of aPKC phosphorylation sites. Using this algorithm, I identified numerous putative PRBH candidates in the fruit fly proteome and performed two screens of these candidates for Par-polarized proteins. The first screen focused on determining whether aPKC regulates cortical targeting of proteins that are reported to be polarized. This screen identified the Rho GAP crossveinless-c (cv-c) to be a novel aPKC substrate and found that aPKC is sufficient to polarize cv-c in a reconstituted polarity assay. The second screen characterized the localization of putative PRBH motif-containing proteins in vivo. This screen identified a previously uncharacterized protein, CG6454, to be basolateral in epithelia; however, ex vivo experiments found it to have a Ca2+-dependent and aPKC-independent membrane targeting mechanism. Overall this work identified a common mechanism for Par substrate polarization and used knowledge of this mechanism to identify a novel Par effector.

Att undersöka hur stor bedövningseffekt som kan uppnås efter injektion med aktiv (lidokain) eller in-aktiv substans (koksaltlösning) samt att undersöka om deltagarna korrekt kan identifiera vilken injektion de fått, när de vet att det är en 50/50 chans att de har fått lidokain eller koksalt.
Material och metod
20 friska frivilliga deltagare randomiserades in i två grupper. En grupp fick injektion med aktiv substans och en med in-aktiv substans. Deltagarna informerades om att chansen att få aktiv eller in-aktiv substans var lika, 50 %. Deltagarna utsattes för ett specifikt smärtsamt stimulus före och efter injektionen. De graderade sedan sin smärtintensitet på en 0-10 NRS-skala. Följande dag tillfrågades deltagarna vilken substans de trodde de hade fått.
Students T-test användes för att beräkna skillnaden i smärtintensitet före och efter injektion i båda grupperna. Fischer’s exakta test användes för att beräkna kvalitativa data. P <0,05 ansågs vara statistiskt signifikant.
Resultat
Det var en statistiskt signifikant skillnad mellan den aktiva (NRS 2,9) och den in-aktiva (NRS 0,0) gruppen gällande förändringen i smärtintensitet före injektion och efter injektion. Alla deltagare kunde korrekt identifiera vilken injektion de fått.
Slutsats
Ingen bedövningseffekt kunde mätas efter injektion med inaktiv substans hos friska individer när deltagarna visste att det var en 50/50 möjlighet att de skulle få den aktiva substansen. Alla individer kunde korrekt avgöra om de fått injektion med aktivt bedövningsmedel eller in-aktivt koksalt.
Aim
To investigate the amount of anesthetic effect that can be achieved following injection with active (lidocaine) or non-active (saline) agent and to examine if the participants correctly can identify which injection they received when there is a 50/50 chance that they have received lidocaine or saline.
Materials and Method
20 healthy volunteers were randomized in two groups. One group got injection with active agent and one with non-active agent. The participants were instructed that chances of receiving active versus non-active agent were equal, 50 %. The participants were exposed to a specific painful stimulus before and after the injection and they had to rate their pain score on an 0-10 NRS-scale. The following day, the participants were asked what agent they thought they had received.
Students T-test was used to calculate the difference in pain intensity between pre- and post-injection in both groups. Fisher's exact test was used to calculate qualitative variables. P<0.05 was considered statistically significant.
Results
There was a statistically significant difference between the active (NRS 2.9) and the non-active (NRS 0.0) group regarding the change in pain intensity rating from pre-injection to post-injection. All participants could correctly identify which injection they received.

▼ The purpose of this study was to examine the potential impact of client weight and ethnicity on counselors’ recognition and appraisal of eating disorder symptoms. Previous research has demonstrated weight-based bias in healthcare practitioners, including individual therapists. In addition, previous research has illustrated that racial and ethnic stereotypes may contribute to misdiagnosis/underdiagnosis of disordered eating in women of color. This study randomly assigned participants to one of six different vignette conditions. The vignettes varied only by the weight (low, high) and ethnicity (White, Black, Hispanic) of the client they describe. All vignettes featured a young woman who engages in restrictive eating behaviors and over-exercising, and who has lost 25% of her body weight over a short period of time. Participants were asked to label and rate the severity of her presenting problem, as well as rate the frequency and severity of the client’s symptoms. It was hypothesized that participants would rate the client’s eating disordered behaviors as less severe when the client is not significantly underweight, despite the presence of drastic, medically unsafe weight loss. Demographic information of participants was collected in order to identify any potential relationship between demographic factors and patterns of decision-making. Results indicated that the weight of the client in the vignette appeared to influence participants’ clinical evaluation of the client. Participants were more likely to recommend a medical follow-up when the client was lower weight. Participants’ responses on an Anorexia symptom subscale indicated that they rated the behaviors as more frequent in the lower weight condition, and their rating of the overall severity of her presenting problem was higher in the lower weight condition. No differences were observed based on race, or on interaction of race and weight. Implications for theory, research, and practice are discussed.

▼ Autism Spectrum Disorders (ASDs) are characterized by pervasive impairments in repetitive behaviors or interests, communication, and socialization. As the onset of these features occurs at a very young age, early detection is of the utmost importance. In an attempt to better clarify the behavioral presentation of communication and socialization deficits to aid in early assessment and intervention, impairments in these areas were examined among infants and toddlers (17-37 months) with Autistic Disorder (AD), Pervasive Developmental Disorder – Not Otherwise Specified (PDD-NOS), and non-ASD related developmental delay. The Baby and Infant Screen for Children with aUtIsm Traits-Part1 (BISCUIT-Part1) and the Battelle Developmental Inventory, 2nd Edition (BDI-2) were utilized to examine communication and socialization levels, respectively, among these groups. All groups significantly differed on level of socialization impairment with the Autism group displaying the greatest impairment and the non-ASD related developmental delay group evincing the least impairment. In regards to communication deficits, the non-ASD related developmentally delayed group differed significantly in comparison to the Autism and PDD-NOS groups; however, no significant differences were found between children with AD and PDD-NOS. While communication and socialization impairments were found to significantly correlate for all participants with the exception of those with PDD-NOS, these correlations were not found to significantly differ from one another across groups. A regression analysis examining which communication items on the BISCUIT-Part1 predicted socialization impairment on the BDI-2 found that the two significant predictors were use of language in conversation with others and communicates effectively (e.g., using words, gestures or sign language). The implications, limitations, and future directions of these results are discussed.

▼ Background: Myocardial infarction affects people around the world. Proportionally more women than men die from the condition and those women who have the greatest risk are the ones who live an unhealthy lifestyle. This includes individuals with poor dietary habits and low levels of physical activity as well as those with additional medical diagnoses such as hypertension, diabetes and psychological disorders. Aim: The aim of the study was to describe women's experiences after undergoing a myocardial infarction. Method: A method to contribute to evidencebased practise based on analysis of qualitative research was performed. Ten articles were analysed and resulted in four themes and ten subthemes. Result: Findings were: Women experience atypical symptoms before a myocardial infarction occurs. Women also need more support as they may be worried about leaving their everyday lives and letting go of everything around them. The results also highlight the recovery being difficult for the women who prioritize the family before their own health. Conclusion: It is important that all women obtain a permanent health care contact, as most of the women find that recovery is difficult. Relapsing is a possibility making it imperative that women become aware of the symptoms of myocardial infarction and how they appear.

▼ Background: Standard of care for management of ADH identified on percutaneous biopsy is surgical excision. Research efforts have focused on utilizing strict multidisciplinary review to identify patients at lowest risk for upgrade that may benefit from omission of surgery. However, having an ipsilateral breast cancer, in addition to the site of ADH, has been an exclusion criterion for consideration of observation over excision of the site of ADH. Methods: This retrospective analysis examined patients who had both a breast cancer and an additional site of ADH in the same breast diagnosed on percutaneous biopsy, who underwent surgical excision of both areas at our institution from 2008-2018. Imaging characteristics (size of cancer, size of ADH, distance between cancer and ADH, percutaneous biopsy technique) and pathologic features (histologic subtype of cancer, cancer grade and prognostic markers, extent of ADH, presence of necrosis or micropapillary features in the ADH) were reviewed from the biopsy, as well as the final surgical pathology at the site of ADH excision to determine features associated with ADH upgrade. Results: Sixty-two women had biopsy proven ADH and a breast cancer at separate sites in the same breast over the 10-year study period. The overall upgrade rate at the site of ADH was 17.7% (n=11), with 9 cases to ductal carcinoma in situ (DCIS) and 2 to invasive breast cancer (IBC). The most common associations with upgrade were the presence of ipsilateral DCIS over IBC (p=0.034), using ultrasound guidance for biopsy of ADH (p=0.019), and the presence of cell necrosis in the ADH (p=0.039). Neither the radiographic size of the ADH nor the distance of the ADH from the ipsilateral cancer were associated with upgrade. The group at lowest risk for upgrade had stereotactic biopsy of the site of ADH and no necrosis associated with ADH, which resulted in 0% upgrade rate. Conclusion: When an ipsilateral breast cancer is present, the upgrade rate at the site of ADH is on par with reported contemporary ADH upgrade rates without ipsilateral breast cancer present. Similar to those studies of ADH alone, upgrade was significantly associated with biopsy modality and presence of ADH necrosis. When considering the ipsilateral breast cancer, upgrade rate for ADH was not affected by the size of the cancer, size of atypia, or distance between the ipsilateral malignancy and atypia. This suggests that omission of surgical excision for ADH in patients with concurrent breast cancer may be appropriate, like those with isolated ADH, when a select low risk subset undergoing rigorous multidisciplinary review can be identified.

▼ Mitogen-activated protein kinases (MAPKs) are Ser/Thr
kinases that regulate diverse cellular processes. Extracellular
signal-regulated kinase 3 (ERK3) is an
atypical MAPK that has
recently gained interest for its roles in cancer. ERK3 promotes
cancer cell migration, invasion and drug resistance, and its
expression is upregulated in multiple cancers. However, little is
known about the molecular mechanisms that regulate ERK3’s
expression and kinase activity. Here, we have identified the
oncogenic polycomb group protein BMI1 as a positive regulator of
ERK3 level in cancer cells. Mechanistically, BMI1 upregulates ERK3
expression by inhibiting the expression of the tumor suppressor
microRNA (miRNA) let-7i, which directly targets ERK3 mRNA. ERK3
then acts as an important downstream mediator of BMI1 in promoting
cancer cell migration. Importantly, ERK3 protein level is
positively correlated with BMI1 level in head and neck tumor
specimens of human patients. In addition, we have investigated the
roles of the activation loop phosphorylation and the unique
elongated C-terminus extension in regulating ERK3’s kinase activity
and functions in cancer cells. Our study revealed that mutating the
activation loop phosphorylation site S189 or truncating the
C-terminus extension greatly decreases ERK3’s in vitro kinase
activity towards the oncogenic protein steroid receptor
co-activator 3 (SRC-3), an important downstream target for ERK3
signaling in cancer. While the reduced activity of ERK3 S189A
mutant can be partly attributed to its decreased interaction with
the substrate SRC-3, the importance of the C-terminus extension in
kinase activity possibly stems from its intramolecular interaction
with the kinase domain of ERK3. In agreement with their decreased
kinase activity towards SRC-3, the S189A mutant and the C-terminus
deletion mutants have greatly diminished ability to stimulate
migration and invasion of lung cancer cells as compared to wild
type ERK3. Furthermore, we have identified a kinase-independent
role of ERK3 in promoting cancer cell migration and invasion. Taken
together, the findings of our study provide important insights into
the molecular regulation of ERK3 signaling in cancer, which can
accelerate the progress in developing anti-cancer therapeutic
strategies through the inhibition of ERK3 signaling.
