As doenças denominadas como negligenciadas têm causado nos últimos anos uma preocupação muito acentuada na comunidade científica e para as autoridades de saúde, relacionada às suas terapêuticas, no sentido de que os medicamentos existentes não se apresentam totalmente eficazes, além de determinarem efeitos colaterais extremamente elevados. Nesse perfil se encaixam a doença de Chagas e as Leishmanioses, etiologias determinadas respectivamente por Trypanosoma cruzi e parasitas do gênero Leishmania. Como proposta de encontrarmos uma alternativa para o tratamento dessas parasitoses, avaliamos o potencial terapêutico de três estatinas (sinvastatina, pravastatina e mevastatina), comercialmente encontradas para o tratamento de níveis elevados de colesterol e triglicérides, baseado no princípio de que a rota bioquímica para a formação de colesterol é semelhante à do ergosterol, componente da membrana plasmática desses protozoários. Foram realizadas avaliações in vitro e in vivo das estatinas puras e de suas associações com o benzonidazol, medicamento de referência no tratamento da doença de Chagas e com a anfotericina B, medicamento de referência no tratamento das leishmanioses, partindo do pressuposto que a substituição do benzonidazol / anfotericina B ou a diminuição de suas doses em combinação com as estatinas, atuando como coadjuvantes, pudessem auxiliar no tratamento e diminuir os efeitos colaterais provocados pela medicação. Observou-se nos ensaios com T. cruzi in vitro boa atividade antiparasitária, com valores de porcentagem de lise celular mais altos ou comparados aos encontrados para o benzonidazol, já in vivo, apenas a mevastatina apresentou redução da parasitemia em relação ao controle positivo e às outras estatinas testadas. Nos ensaios com L. braziliensis e L. major não foi observado resultado significante, tanto in vitro como in vivo, apesar de em algumas situações encontrarmos resultados próximos ao controle positivo.

Over recent years, neglected diseases has been a problem to the scientific community and health associations due the absence of total efficacy of drugs, and by their potential side effects. Chagasd́isease and Leishmaniasis - both caused respectively by Trypanosoma cruzi and by parasites the genus Leishmania fit on this profile. Aiming to find an alternative treatment of these parasitosis we evaluated the potential therapeutic of three statins (simvastatin, pravatatin, and mevastatin) which are commercially employed for treatment of high levels of cholesterol and triglycerides, based on the similarity of the biochemical route of cholesterol and ergosterol formation, being the ergosterol a component of plasmatic membrane of these protozoa. Les maladies tropicales négligées causées par les protozoaires kinétoplastidés du genre Leishmania et Trypanosoma représentent une menace pour près d’un demi-milliard de personnes en zone intertropicale, entrainant jusqu’à 50 000 décès par an. Parmi les molécules en développement clinique pour traiter ces pathologies, le fexinidazole est une prodrogue appartenant à la famille des 5-nitroimidazoles et qui exerce son action anti-infectieuse via une étape de bioactivation catalysée par des nitroréductases (NTR) parasitaires, enzymes dont le co-facteur est une flavine. Afin d’identifier de nouveaux nitrohétérocycles antiparasitaires substrats des NTR, une petite chimiothèque d’imidazo[1,2-a]pyridines synthétisées au laboratoire a subi un criblage in vitro ayant conduit à l’identification d’une molécule Hit, à la fois active sur Leishmania donovani et Trypanosoma brucei brucei. Ce composé a servi de point de départ à un travail de pharmacomodulation, dans un premier temps en position 8 du cycle imidazo[1,2-a]pyridine : l’introduction de groupements variés à l’aide de réactions de couplage pallado-catalysées de Suzuki-Miyaura, Sonogashira et Buchwald-Hartwig ou des réactions de SNAr, a permis de mettre en lumière plusieurs composés « tête de série » au profil biologique nettement amélioré. Dans un second temps, le travail de pharmacomodulation entrepris a été étendu aux positions 2, 3 et 6 du cycle imidazo[1,2-a]pyridine en vue de compléter les données de relations structure-activité, d’étudier en particulier l’impact du potentiel rédox et d’optimiser les paramètres physico-chimiques et pharmacocinétiques in vitro des meilleurs composés.

The kinetoplastids of the Leishmania and Trypanosoma genus are the causative agents of neglected tropical diseases that threaten nearly half a billion people in the intertropical zone, resulting in 50 000 deaths per year. Among the molecules in clinical development to treat these pathologies, fexinidazole is a prodrug belonging to the 5-nitroimidazoles family, which exerts its anti-infectious action via a bioactivation step catalyzed by parasitic nitroreductases (NTR), enzymes whose cofactor is a flavin. In order to identify novel nitroheterocycles as parasitic NTR substrates, a small chemical library of imidazo[1,2-a]pyridines synthesized by our laboratory was screened in vitro, leading to the identification of a Hit molecule active both on Leishmania donovani and Trypanosoma brucei brucei. This compound served as a starting point for a pharmacomodulation work, initially in position 8 of the imidazo[1,2-a]pyridine ring: the introduction of various chemical groups using the pallado-catalyzed coupling reactions of Suzuki-Miyaura, Sonogashira and Buchwald-Hartwig, or SNAr reactions, highlighted several "lead" compounds with a significantly improved biological profile. ▼ Les perturbations anthropiques en Amazonie liées au déboisement de la forêt tropicale conduisent à une mosaïque de paysages constituée de végétations secondaires (forêts secondaires, palmeraies, jachères) et de pâturages. Ces modifications favorisent la prolifération de grands palmiers héliophiles invasifs de la famille Attalea spp., palmiers qui constituent l'écotope principal des espèces de Rhodnius, punaises hématophages vectrices de Trypanosoma cruzi, agent étiologique de la Maladie de Chagas en Amérique Latine. Cette étude a porté sur différentes unités de paysage de trois communautés rurales du bas Tapajós (Amazonie brésilienne) ayant une époque d'installation différente sur le territoire (25-75 ans). Six unités de paysage ont été définies sur le terrain et appliquée via une classification supervisée à une image SPOT 5, afin d'obtenir une cartographie du risque environnemental associé à la présence de palmiers dans la région. Sur les cent trente trois palmiers disséqués appartenant aux trois espèces Attalea maripa, A. phalerata et A.speciosa, 73 (54.88%) étaient infestés par R. robustus (742 insectes récoltés). Des diminutions significatives de densité de triatomes ont été observées chez A. maripa, dans la communauté la plus récemment établie (Araipá) et dans les unités de paysage les plus anthropisées. L'infection des insectes par T. cruzi et T. rangeli a été examinée à l'aide de méthodes moléculaires (mini exon SL_IR and sno-RNA-C11). Respectivement 123 (16.57%) et 69 (9.3%) insectes dans 31 (23.3%) et 17 (13.82%) palmiers ont été identifiés positifs à T cruzi et à T.rangeli. Aucune infection n'a été trouvée dans les insectes collectés à Araipá et aucune différence significative n'a été mise en évidence entre les différentes unités de paysage. Les souches de Trypanosoma cruzi identifiées à l'aide de 4 marqueurs moléculaires (mini exon SL-IR, GPI, HSP60 et D7-24α-rRNA) appartiennent à la lignée TcId et 10 (8.13%) individus présentent une infection mixte TcI-TcII. Vingt espèces d'hôtes réparties en trois classes (mammifères, oiseaux, sauropsidés) ont été identifiées comme sources alimentaires à partir du repas sanguin contenu dans le tube digestif des insectes, à l'aide d'amorces cytochrome b, spécifiques de vertébrés. Quatre-vingts et un pourcent des repas détectés ont été effectués sur des mammifères, hôtes potentiels de T.cruzi dont Tamandua tetradactyla, source alimentaire principale. Cet hôte a été clairement identifié comme réservoir de T.rangeli ainsi que suggéré pour T.cruzi. L'analyse phylogénique réalisée à l'aide de séquences de cytochrome b démontre que les individus de Rhodnius identifiés dans la région du Tapajos appartiennent au clade II, ce qui correspond à une extension de l'aire précédemment décrite pour ce clade. L'utilisation du marqueur mitochondrial cytochrome b a permis de mettre en évidence une structuration phylogénétique (haplogroupes) non retrouvée à l'aide des marqueurs microsatellites.
