Abstract: MAGP-1 is a glycoprotein present in the elastic fibers, functioning as part of the microfibrils components. Studies have shown that MAGP-1 expression is critical for vascular development and is associated with decreased interaction between integrins and the extracellular matrix in zebra fish. Also MAGP-1 interacts with von Willebrand factor and the active form of TGF-? and BMP. In mice lacking MAGP-1, thrombus formation is delayed, increasing the occlusion time of carotid artery despite normal blood coagulation in vitro. MAGP-containing microfibrils may play a role in hemostasis and thrombosis. In this work, we evaluated the function of MAGP-1 and its relation to TGF-? in the arterial thrombosis process. For this purpose, we obtained and analyzed a mutated MAGP1 (VG1) protein, its participation in the arterial thrombosis formation and its interaction with important proteins in the coagulation process. We analyzed thrombus formation time in wild type and MAGP-1-deficency mice. The potential participation of TGF-? in this process was accessed when we treated both wild type and MAGP-1-/- mice with losartan and captopril. Besides, we evaluated the gelatinolytic activity in the arterial walls. The MAGP-1 mutation avoided its thrombus formation time recovery, although the mutated MGP-1 still interacting with vWF and fibronectin. Losartan and captopril were able to recover the thrombus formation time without changing blood pressure, aPTT and PT, platelet aggregation and adhesion, but increased gelatinases activity. Abstract: Endothelial progenitor cells (EPC) present pluripotent characteristics proliferating and differentiating into mature endothelial cells. These cells are mainly characterized by the expression of CD34, VEGFR2, CD31, internalization of AcLDL and formation in vitro of vessel like structures. They can be mobilized from bone marrow to the sites of vascular injury promoting vascular re-endothelialization. They represent 0.1% of bone marrow and 0.01% of peripheral blood cells and can be culture for cell therapy use. In this work, we analyze the role of late EPC grown for 30 days in two different culture media on the arterial thrombus resolution induced by FeCl3. MMC (mononuclear cells) were isolated from mice bone marrow and cultured in EGM2® (commercial medium) or DMEM-M1 (DMEM plus VEGF (10ng/mL), IGF (2ng/mL) and bFGF (2 ng/mL)). Cells showed positive expression for CD34, CD133, VEGFR2, CD31 and VE-cadherin, AcLDL internalization and vessel-like structures formation ability. We performed in vivo thrombosis analysis using in C57BL/6 mice. The carotid artery was isolated and lesioned with FeCl3 and the animals received CPE¿s injections 15 min before, 1 hour and 24 hours after injury. The vessels were analyzed 0, 3 and 7 days after injury in CTR (no cells), EGM2 (EGM2® cells) and M1 (DMEM-M1 cells) groups. Vessel histological analysis showed a reduction of the thrombus area 7 days after injury. EPCs injected were found in the lumen and adventitia. By intravital microscopy, we observed the dynamics of thrombus formation, with embolization in M1 and EGM2 groups. Occlusive thrombus formation time was prolonged in MI and EGM groups when compared to CTR and injected with HUVEC and fibroblasts groups. EPCs were able to inhibit platelet aggregation in vitro, increased the expression of eNOS and decrease SDF-1 levels in circulating plasma after 7 days. MMP-2 and 9 activities increased in M1 and EGM groups when compared to CTR, with an activity higher in M1 than in EGM2 group. We conclude that the DMEM-M1 medium was able to generate a viable and efficient population of EPC comparable to the commercial medium for cell therapy. M1 and EGM2 cells extend occlusion time and promote accelerated thrombus resolution after arterial lesion, improving the vessel with increased eNOS expression. Thrombus resolution may be related to increased MMPs activity in the thrombotic matrix. Decreased SDF-1 levels may indicate that EPC are signaling for regeneration of the endothelium and decreased mobilization from the bone marrow. Abstract: Cardiovascular diseases are one of the leading causes of death in the world nowadays. Myocardial infarction and stroke are the main causes of these deaths, and arterial thrombosis is one of their principal underlying diseases. Arterial thrombosis is characterized by the obstruction of the artery lumen by the formation of a clot, which occurs in response to an endothelial injury or activation. After the formation of the thrombus, remodeling of the vessel occurs, which may result in the exacerbated proliferation of smooth muscle cells (SMC) with consequent formation of the neointimal layer and restenosis of the vessel. The main treatments for thrombosis involve the action of anticoagulants such as unfractionated heparin (UFH) and low molecular weight heparin (LMWH), these are glycosaminoglycans (GAGs) that inhibit the coagulation cascade but have some limitations in their use. Thus, it is necessary to study alternative treatments, such as the use of Dermatan sulfate (DS), which acts as an anticoagulant and has been studied as an alternative anticoagulant treatment. The objective of our study is to evaluate and compare the effect of UFH, LMWH and DS on smooth muscle cell (SMC) proliferation and migration in vitro, and in arterial thrombosis and neointimal layer in mice. For this purpose, SMC were isolated and their proliferation and migration capacity were tested in vitro. In the in vivo analyzes, carotid thrombosis was induced with a metal probe and the animals were treated with UFH, LMWH or DS with 4 intravenous injections, 15 minutes, 12, 24 and 48 hours after injury. The animals were sacrificed 3 and 21 days after surgery for analysis of the thrombus and neointimal layer. As results, we observed that all GAGs decreased SMC proliferation in vitro and there was no difference in cell migration after 24 hours of treatments. In animals submitted to arterial injury, we observed a decrease in thrombus area in all treated groups when compared to controls, with LMWH and DS with similar values. We observed a decrease in the presence of Ly6G e CD11b positive cells, that are the main markers of inflammatory cells in the UFH and DS. For the adhesion molecule ICAM-1, the group treated with DS showed a decrease in the expression of this protein. DS also decreased the expression of latent and active MMP-2 and UFH decreased active MMP-2. In the formation of the neointimal layer, 21 after the injury, there was a significant decrease in the groups treated with UFH and DS compared to the control and LMWH groups. We conclude that all GAGs have reduced the thrombus area, and the treatments with UFH and DS, decreased the inflammatory cells and expression of active MMP-2, 3 days after the injury and reduced the neointimal layer.
