▼ The mechanistic target of rapamycin complex 1 (mTORC1) serves as a molecular link between cellular growth conditions and anabolic processes that are fundamental to cell growth and proliferation. However, while it is established that the PI3K-Akt pathway regulate mTORC1 through a mechanism that involves spatial control of the TSC complex, it is unknown whether other growth factor signaling pathways do so. The goals of this dissertation were to characterize the mechanisms by which upstream growth factor signaling pathways converge on the TSC complex to activate mTORC1, and to determine the significance of these mechanisms in disease.
I identify that both the Ras-ERK and PKC pathways stimulate mTORC1 signaling through spatial regulation of the TSC complex. Growth stimulus specific to each pathway induces the dissociation of TSC2 from the lysosome, where it interacts with Rheb, and in serum-starved conditions, keeps Rheb in its GDP-bound form, unable to activate mTORC1. Using antibodies to specific TSC complex components, I further demonstrate that it is the intact TSC complex that dissociates from the lysosome upon growth stimuli treatment. Furthermore, suppression of both ERK and PKC signaling using multiple small molecule inhibitors specific to each pathway, reverses the growth stimuli-mediated TSC2 dissociation.
Given previous models on ERK-mediated TSC complex regulation of mTORC1 activity, I sought to gain more mechanistic details surrounding this regulation. I specifically show that signaling through EGF and PMA does not affect TSC complex stability, but rather regulate the complex through effects on lysosomal localization and Rheb binding. Finally, I discuss how the TSC complex spatial regulation mechanism may be useful in predicting sensitivity or resistance of cell models to targeted therapeutics.
Collectively these studies identify a possible general mechanism by which upstream growth factor-regulated kinases activate mTORC1 through the stimulated release of the TSC complex from lysosomal Rheb. Our results advance our understanding of how diverse upstream molecular inputs can affect the physiological and oncogenic activation of mTORC1, and will hopefully guide future work in studying resistance to targeted therapeutics.

▼ BACKGROUND: Tuberous sclerosis complex (
TSC) is a multisystem hamartomatous disease caused by inactivating mutations in the TSC1/TSC2 genes leading to hyperactivation of mammalian target of rapamycin complex 1 (mTORC1) in
TSC tumors. Novel therapeutic regimens and imaging biomarkers remain critical unmet needs in
TSC.
Mitochondrial fatty acid oxidation (FAO) is a major cellular source of acetyl-CoA. Acetyl-CoA is a central metabolite in lipid anabolism and catabolism. The purpose of this study was to identify novel metabolic therapeutic targets, particularly involving lipid metabolism, to achieve durable responses in
TSC.
METHODS AND RESULTS: Fluoroacetate (FACE), an acetate derivative, is a surrogate biomarker of mitochondrial activity. It accumulates in the mitochondria without being oxidized to CO2, and it is converted to fluorocitrate, irreversibly inhibiting the TCA (tricarboxylic acid) cycle enzyme aconitase. Micro-positron emission tomography (PET) imaging of subcutaneous xenografts of TSC2-deficient Eker rat uterine leiomyoma-derived ELT3 cells showed rapid uptake of [18F]FACE that was maintained after 72-hour treatment with mTORC1 inhibitor rapamycin. This result suggests that mitochondrial oxidative metabolism is sustained in the presence of rapamycin.
Consistent with this finding, treatment of TSC2-deficient cells with rapamycin led to a significant increase in FAO and a decrease in glucose oxidation in vitro as measured by a 14C-CO2 collection metabolic assay. Expression of the A isoform of carnitine palmitoyltransferase I (CPT1A; FAO rate-limiting enzyme) was selectively increased in TSC2-deficient cells. Pharmacological inhibition of CPT1A by ST1326 led to a decrease in FAO, as measured by a 14C-CO2 collection metabolic assay, and a decrease in mitochondrial ATP production, measured by the Seahorse analyzer.
CONCLUSIONS: This study proposes a role for FAO in
TSC tumor bioenergetics and for CPT1A as a potential therapeutic target in
TSC.

▼ La sclérose tubéreuse complexe (TSC) est une maladie génétique caractérisée par une croissance des hamartomes dans différents organes y compris le cerveau, les reins, les poumons, la peau et le cœur. Ces lésions sont des sources de morbidité et de mortalité chez les patients TSC, car ils peuvent provoquerl’ épilepsie, l’autisme, le retard de développement et l’insuffisance rénale et pulmonaire. Les causes connues de TSC sont la perte de la fonction et les mutations des gènes TSC1 et TSC2. La majorité des lésions TSC contient plusieurs types cellulaires de la lignée mésenchymateuse, comme dans le cas des angiomyolipomes, l’lymphangioleiomyomatose et les angiofibromes. Un type unique de cellules épithélioïdes périvasculaires nommé (PEC) est constamment présent dans les lésions de TSC mésenchymateuses, comme angiomyolipomes et lymphangioleiomyomatose, basant sur les caractérisations morphologiques et l'expression des marqueurs communs mélanocytaires et myogéniques. Par conséquent, ces lésions sont officiellement classées, ainsi que d'autres tumeurs, comme PEComes. Leur origine cellulaire et les mécanismes moléculaires impliqués dans la pathologie restent à élucider. Ici, nous avons généré un modèle souris mosaïque TSC1 knockout qui développe des lésions rénales mésenchymateuses récapitulant périvasculaire épithélioïde cellules tumorales humaines (Pecoma) observés chez les patients TSC. Nous avons identifié YAP, le co-activateur transcriptionnel de la voie Hippo, a été régulée d'une manière mTOR-dépendante dans les lésions rénales de notre TSC1 knockout souris et les échantillons de l’angiomyolipome humaines. L'inhibition de YAP avec des outils génétiques ou pharmacologiques atténue considérablement la prolifération et la survie des cellules nulles TSC1 in vivo et in vitro. En outre, l’accumulation de YAP dans les cellules déficientes TSC1 / TSC2 pourra être dû à la dégradation de la protéine altéré par le système de l’autophagosome / lysosome. Ainsi, la régulation de YAP par mTOR et l'autophagie est un nouveau mécanisme de contrôle de la croissance, l'activité de YAP correspondant à la disponibilité des éléments nutritifs dans les conditions de croissance permissives. Il pourra servir comme une cible thérapeutique potentielle pour TSC et d'autres maladies avec une activité de mTOR dérégulée.
The Tuberous Sclerosis Complex (TSC) is a genetic disease characterized by growth of hamartomas in different organs including brain, kidney, lung, skin, and heart. These lesions are sources of morbidity and mortality in patients with TSC, as they may cause intractable epilepsy, autism, developmental delay, renal and pulmonary failure. Known causes of TSC are loss of function mutations in TSC1 and TSC2 genes. ▼ Tuberous Sclerosis Complex (
TSC) is a multi-organ hamartomatous disease caused by loss of function mutations in either the TSC1 or TSC2 genes. Despite involvement of multiple organs such as the kidneys, lungs, and skin, neurological aspects are usually the most severe due to a very high prevalence of cognitive impairment, autism and epilepsy. The protein products of TSC1 and TSC2, hamartin and tuberin respectively, regulate the mTOR kinase signaling pathway. Current models of
TSC propose that hamartoma formation is secondary to a loss of heterozygosity at either the TSC1 or TSC2 loci, and subsequent hyperactivation of mTOR Complex 1 (mTORC1). In this dissertation I explore the underlying mechanisms of organ specific pathogenesis in
TSC.
In the first half of my dissertation, I demonstrate that loss of Tsc1 in the distal convoluted tubule of the kidney results in cystogenesis. Cyst formation in these kidneys is due to a mTORC1 but not mTORC2 dependent process. I then show that cystic changes in these kidneys may be due to ciliary defects.
While a loss of heterozygosity has clearly been reported in the kidney and other organ system, second hit mutations in neural lesions have only rarely been identified. Thus, to begin to define the role of the heterozygosity of TSC1 or TSC2 during the pathogenesis of
TSC in the brain, we generated induced pluripotent stem cells (iPSC) from patients with
TSC. Deep sequencing of these patents revealed that all of our patient derived lines are heterozygous for TSC2 mutations. I then provide evidence that these heterozygous iPSCs are abnormal with increased cell survival and enhanced maintenance of pluripotency. These changes may be due to slight changes in mTORC1 signaling.
The work presented in this dissertation increases our understanding of the tissue specific phenotypes and underlying mechanisms of
TSC pathogenesis. This research may lead to the identification of new therapeutic targets for
TSC and associated comorbidities.

▼ Tuberous Sclerosis Complex (
TSC) is an autosomal dominant syndrome characterized by epilepsy, intellectual disability, and autism. Recent studies have suggested that white matter abnormalities, including hypomyelination, contribute to the cognitive deficits in
TSC patients, but the mechanism has remained elusive. I used the neuron-specific Tsc1 knockout mice that display a marked decrease in myelin and show that oligodendrocytes are arrested at immature stages of development in vivo resulting in a reduction in the number of myelinating cells. I established an oligodendrocyte culture system and examined the effect of neuron-conditioned media and found that the Tsc1 mutant phenotype was replicable in vitro using medium collected from Tsc1 knockdown (
TSC-KD) neurons, confirming that a secreted signal is responsible for inhibiting differentiation of the oligodendrocytes. I took an unbiased genome-wide approach and identified Connective Tissue Growth Factor (CTGF) as a putative candidate for the secreted signal. I confirmed that CTGF was upregulated in Tsc1 mutant neurons and characterized its spatial and developmental expression pattern in our mouse model. In vitro, CTGF was sufficient to inhibit differentiation of oligodendrocytes. The addition of CTGF neutralizing antibody to the
TSC-KD neuronal media was able to reverse the suppression of oligodendrocyte maturation, strongly suggesting that CTGF is a major component of the oligodendrocyte inhibitory signal derived from
Tsc mutant neurons. Since
TSC mutation affects all cells, I investigated the role of
TSC in oligodendrocytes. In response to
TSC knockdown, oligodendrocytes demonstrate an upregulation of cellular stress marker. I also found a decrease in myelin protein genes, a finding that offers interesting implications for the role of
TSC in hypomyelination. Furthermore, I expanded my research into Zellweger disease, a syndrome that involves
TSC in its neuropathological manifestations including white matter deficits, and found that localization of
TSC to the peroxisome is a critical factor in neuron development. Together, this body of work developed new approaches in Tuberous Sclerosis research in the brain to investigate a previously under-appreciated aspect of
TSC pathology - myelination. I have demonstrated that the
TSC pathway has important roles in neuron-oligodendrocyte communication and emphasize the critical importance of neuron-derived signals in the establishment of myelination.

▼ Tuberous Sclerosis Complex (
TSC) is a highly variable autosomal dominant multisystem disorder characterized by the growth of benign tumors, epilepsy, and
TSC- associated neuropsychiatric disorders (TAND). There is a high level of clinical variability, even within the same family. While reproductive decisions always carry a level of uncertainty, individuals with highly variable genetic conditions like
TSC must consider both the chance of passing on the condition and the uncertain clinical presentation. There is currently no literature on factors influencing reproductive decisions of adults with
TSC. To address this gap in understanding, we conducted an exploratory mixed-methods survey utilizing an anonymous online questionnaire to assess study participants’: 1) familiarity with the symptoms of
TSC, 2) understanding of the risk of passing on
TSC, 3) perceived disease burden/quality of life, and 4) family planning considerations. A total of 175 individuals aged 18-45 who were diagnosed with
TSC were included in the final data set. Participants were highly familiar with symptoms of
TSC with an average symptom knowledge score of 86.64%. Cortical tubers, angiofibromas, angiomyolipomas, and seizures were recognized as symptoms of
TSC by more than 95% of participants. Lymphangioleiomyomatosis (LAM) was the least recognized symptom (75.29%), with females being statistically more likely to recognize the symptom than males. Most participants (85.96%) were aware of the 50% recurrence risk of
TSC. Perceived disease burden was low with 58.58% viewing themselves as mildly or very mildly affected. Our disease burden/quality of life instrument found that a majority of participants reported that they felt different from those around them, that they were frustrated by their symptoms, that their symptoms made them anxious, and that they thought about their
TSC at least some of the time. Sleep disturbance and pain caused by
TSC were also common. However, a majority of participants felt like they were in control of their lives, felt good about their social life, felt comfortable meeting new people, and felt they had the support they needed. Around 60% of our study population was considering having future children with the average desired number of children equaling 2.25. Reproductive methods being considered included traditional conception (52.94%), adoption (45.10%), donor gametes or embryos (17.64%), and preimplantation genetic diagnosis ([PGD], 50.00%). Thematic analysis showed desire for biological children, personal health, desire to not pass on
TSC, financial concerns, and fertility issues were major factors in choice of reproductive method. Interest in prenatal testing was high with 67.44% stating they would test a hypothetical future pregnancy. Thematic analysis showed being informed, considering termination of pregnancy, and accepting whatever happens as major themes for individuals’ interest or lack thereof in prenatal testing. ▼ This study analyses existing Knowledge Management frameworks and assesses
components necessary to develop and sustain an effective KM in organizations in the
Public Sector. There is a wide array of knowledge content that depends on specific
functions of government. The Public Sector can leverage efficiencies across all public
services through accessing the right information for making informed decisions and
eliminate duplication of effort.
Teachers Service Commission (TSC) has been mandated by the Kenya
Constitution to carry out the Teacher Management function for all public educational
institutions. Smooth information flow and communication is crucial for effective decision
making. Inadequate information access due to information storage challenges is a major
problem that hampers proper service delivery at all levels within the TSC. The objective of
this study was to recommend a knowledge-oriented organizational model for TSC, which
would efficiently manage its intellectual and knowledge assets and improve information
flow.
The methodology used, which was qualitative research, was done from document
review sourced from primary and secondary sources as well as similar organization best
practice in Knowledge Management. The Common KADS model was used to analyze the
knowledge structure by distinguishing specific knowledge types and roles. The real needs
of the Commission as a case study within the Public Sector were gathered through a
knowledge mapping exercise. The analytical methods used included grounded theory,
thematic narrative, participant observation (ethnography) and content analysis.
Results from findings revealed that information access, process based knowledge
(organization memory), structured knowledge sharing forums, clear rewarding
mechanisms, mentoring practice and commitment towards knowledge creation were
ranked highly among managers for effective decision making. The study therefore
proposes a KM relational framework/ model integrated with the decision making
framework as an implementation strategy. This will ensure an embedded knowledge
sharing culture‘ within TSC and the Public Sector for improved service delivery.

▼ The unique properties of diamond make it an excellent material for electronic and optical applications. It is particularly attractive for ultraviolet radiation dosimetry due to its intrinsic properties, which include biological tissue equivalence and visible blindness. Importantly, the advent of synthetic diamond, especially Chemical Vapour Deposition (CVD) diamond, has made it more economically viable for such applications. A thorough understanding of the electronic properties of diamond is needed before these applications can be fully explored. Consequently, this thesis investigates charge carrier trapping states in CVD polycrystalline diamond for the optimization of ultraviolet radiation dosimetry.
The technique of Thermally Stimulated Conductivity (TSC) is used to probe electrically active defects and is also applied for dosimetric read-out. A range of as-grown CVD polycrystalline diamond films are studied to determine attributes that favour dosimetric-related TSC. In doing so, we establish that dosimetric TSC in these films originate from defects at the grain boundaries with a correlation to high crystalline quality.
With this finding in hand, we then investigate the possibility of optimising diamond for dosimetry by controllably introducing extrinsic dosimetric defects using ion implantation. However, it is shown that these defects are not suitable for dosimetry and have a detrimental effect on the indigenous TSC signal. This study verifies the importance of crystalline quality on the indigenous dosimetric properties of CVD polycrystalline diamond.
The possibility of doping CVD diamond during growth is also investigated as a means for intentionally introducing extrinsic dosimetric defects. Sulphur is selected as the dopant based on the theoretical energy levels formed by this defect, and because the prospect of S doping in diamond remains an actively debated issue in the literature. We report for the first time defect levels extracted from TSC analysis of S-doped CVD diamond and find consistency with theoretical predications. In addition, the dominant TSC trap level in S-doped diamond shows promise for radiation dosimetry with certain properties exceeding many current radiation dosimeters.
The experimental results in this thesis lead to a deeper understanding of defect and trapping mechanisms in CVD polycrystalline diamond and establish attributes that favour TSC and related dosimetric properties in such films. This knowledge is fundamental to the realisation of diamond for ultraviolet dosimetry.

by the development of benign malformations and specific neurological and
psychiatric symptoms. It was first formally described by the neurologist Bourneville
in 1880, based on a female patient who presented with epilepsy, skin tags, facial
rash, and sclerotic areas in some of the cerebral convolutions on pathological
examination of the brain.
TSC is a relatively common genetic syndrome with a birth incidence of 1:60001.
Although all major organs are susceptible to TSC, the most frequently affected
are the brain, skin, kidneys, heart, lung and eye. None of the manifestations
associated with TSC is pathognomonic for the disorder and accordingly, diagnosis
is made based on the presence of combinations of major and minor disease
features, as revealed by clinical, radiological and pathological examination.

▼ When an enterprise has developed to a certain scale and anticipated an impressive expansion, business transformation and diversification are unavoidable decisions. How to measure targets or strategies of business transformation and diversification by quantifiable performance measurements or non-quantifiable performance evaluation systems and make adequate modifications would be crucial issues.
This study applied Balanced Scorecard ãKaplan and Norton, 1996ã to the Taiwan Sugar Corporation ãTSCã as a case study. Balanced Scorecard is a tool provides a strategy framework for implementation and control, and transforms companyâs visions and strategies into measurable indexes with four dimensions. In order to synchronize the Balanced Scorecard sheet and strategy map of
TSC, the study used statistics method to analyze questionnaires and technique of in-depth interviews. Started with analyzing the results of
TSC diversification, the study expanded cause-effect relationships of eight major strategic themes, and combined 13 strategic objectives and 45 strategic key performance indicators.
The study has two major findings:
1.The current management system and performance measurement of state-owned business are influenced significantly by the government agency. Furthermore, the state-owned business also emphasis too much on financial and internal business process performance indicators, thus customer needs and needs for learning are sacrificed. As the result, the transform mission and strategic target, which focus on value of customers, canât be connected.
2.Through planning and designing Balanced Scorecard for
TSC and transforming organizational strategic objectives into actionable objectives and key performance indicator, we found that Balanced Scorecard is an integrated strategic management tool. Using this tool, the state-owned business can effectively move from âbudget performance measurement systemâ to âperformance management strategy systemâ.
The establishment of Balanced Scorecard has brought
TSC the following substantial benefits:
1.Improving the unclear decision making mechanism
2.Establishing strategy focus organization
3.Reducing the gap of current achievement evaluation systems

▼ LAM is a rare and progressive disease characterized by widespread proliferation of abnormal smooth muscle-like cells (LAM cells). LAM cells cause cystic destruction of lung parenchyma, abdominal tumours (angiomyolipoma, AML) and infiltration of axial lymphatics in torax and abdomen (adenopathy and lymphangioleiomyoma). LAM occurs sporadically or in association with tuberous sclerosis complex (
TSC), an inherited disorder with variable penetrance, which results from mutations in TSC1 or TSC2 genes. TSC1 or TSC2 genes, encoding hamartin and tuberin respectively, regulate mammalian target of rapamycin (mTOR). LAM affects primarly women of child-bearing age and the mechanisms causing the disease are not yet clarified. To explain the multisystemic clinical manifestations of LAM an experimental model is needed to study the pathological mechanism causing LAM. It may help to explain how LAM cells migrate from tissue to tissue and to develop a pharmacological approach. We recently isolated and characterized α-actin positive smooth muscle cells from chylous of a patient affected by LAM/
TSC (LAM/
TSC cells). These circulating cells showed reactivity to HMB45 and CD44v6 antibodies, markers of
TSC and LAM, and bear a germline TSC2 mutation in exon 21. Like TSC2 smooth muscle cells previously isolated (TSC2-/- and TSC2-/meth ASM cells), LAM/
TSC cells from chylous required epidermal growth factor (EGF) to proliferate and the blockade of EGF receptor (EGFR) caused progressive cell death. To better study LAM pathogenesis we developed a procedure for a quick invasion of the respiratory system by endonasally administrating LAM/
TSC cells. LAM/
TSC cells were administrated in immunodeficient female nude mice (nu/nu Hsd: athymic nude mice, 3 weeks old) and after 26 weeks anti-EGFR antibody and rapamycin were intraperitoneally injected 2 times a week for 4 weeks. 30 weeks after endonasal administration LAM/
TSC cells were detected in lungs, lymph nodes and uterus. In lung parenchyma, LAM/
TSC cells proliferated and caused cystic destruction with emphysematous-like picture such as in LAM patient lungs. This lesion and the proliferating rate were reverted by anti-EGFR antibody, while rapamycin was less effective and caused hemoptysis. In lungs blood vessel number was increased and, using LYVE-1 antibody, a significant increase of lymphatic vessel density (LVD) was observed in animals that received LAM/
TSC cells suggesting a possible correlation between LAM/
TSC cells and lymphangiogenesis. LVD decreased following anti-EGFR antibody and rapamycin treatments. When treatments were stopped, hemoptysis caused by rapamycin was reverted. Anti-EGFR antibody was more effective than rapamycin in reducing lung injury caused by LAM/
TSC cells administration and in decreasing lymphangiogenesis. In some lung parenchyma noduli were detected. Such in lungs of LAM patients they were positive to human COX IV, ER, PR and phosphoS6. In lymph nodes, LAM/

▼ La sclérose tubéreuse de Bourneville (STB) est une maladie génétique et multi-systémique à transmission autosomique dominante due à des mutations d’un gène TSC1 ou TSC2 qui codent respectivement pour hamartine et tuberine ayant une action inhibitrice sur la voie de signalisation mTOR. L’épilepsie précoce et pharmacorésistante est la manifestation neurologique la plus fréquente et la plus délétère de la STB. Elle débute souvent dans la première année de vie par des spasmes infantiles qui évoluent avec l’âge et en absence de traitement vers des crises toniques ou tonico-cloniques. Bien que les crises soient supposées être générées dans des tubers corticaux, les mécanismes de l’épilepsie ne sont pas bien élucidés et le traitement reste souvent inefficace. Des études morphologiques ont montré une altération de l’expression ARNm des récepteurs au glutamate dans les cellules géantes et les neurones dysplasiques des tubers, mais leur implication fonctionnelle restait à montrer. Les différentes sous-unités NMDA ont une expression âge-dépendante et région-spécifique, les plus grands changements survenant au début de la vie quand l’épilepsie de la STB apparaît. Ce travail avait pour but d’étudier à l’aide de méthodes électrophysiologiques in vitro et in vivo l’expression fonctionnelle des sous-unités NMDA aberrantes et de déterminer leur rôle dans l’épileptogènese chez les souris hétérozygotes Tsc1+/- et sur le tissu humain STB post-opératoire. Nous avons pu démontrer que : i) Les souris hétérozygotes pour le gène Tsc1 sont spontanément épileptiques in vivo et in vitro dans une courte fenêtre dévelopmentale de P9 à P18. ii) Elles présentent une altération d’expression des récepteurs NMDA couche-spécifique et mTOR dépendante avec une surexpression des sous-unités GluN2C/D dans la couche 4 et 2/3 et GluN2B dans les couches 2/3. Cette expression anormale est prévenue par l’administration d’un inhibiteur de la voie mTOR, la rapamycine. iii) Les mêmes altérations d’expression des récepteurs NMDA, sont montrées sur les tissus post-opératoires, non seulement de tubers de STB mais aussi des dysplasies corticales focales (DCF), ces deux malformations ayant des similarités étiologiques et physiopathologiques. iv) La RT-PCR quantitative confirme une expression excessive de GluN2C dans le cortex de souris Tsc1+/- et sur le tissu humain des tubers et DCF. v) Les décharges épileptiques chez la souris Tsc1+/- sont générées dans la couche granulaire 4 du cortex avant de se propager vers les couches superficielles et les couches profondes, empruntant ainsi les microcircuits corticaux. vi) L’expression excessive de la sous-unité GluN2C dans le cortex contribue à l’hyperexcitabilité neuronale chez la souris Tsc1+/- et sur des tissus humains de tubers et de DCF puisque les crises et les décharges sont bloquées par les antagonistes sélectifs de GluN2C/D. vii) Les crises chez la souris Tsc1+/- suivent une séquence âge-dépendante évoluant du type «spasms-like» vers «tonic-clonic like», rappelant celle de l’épilepsie humaine, avec deux pics de haute…

▼ La sclérose tubéreuse de Bourneville (STB) est une maladie génétique et multi-systémique à transmission autosomique dominante due à des mutations d’un gène TSC1 ou TSC2 qui codent respectivement pour hamartine et tuberine ayant une action inhibitrice sur la voie de signalisation mTOR. ▼ The mammalian target of rapamycin (mTOR) protein kinase exists in two distinct complexes, mTOR complex 1 (mTORC1) and mTOR complex 2 (mTORC2). The mTORC1 signaling pathway is involved in numerous cellular processes, including cell growth and protein synthesis, as well as various physiological and pathophysiological conditions, including muscle hypertrophy, inflammation, cancer, and diabetes. Upstream of mTORC1 the tuberous sclerosis complex (
TSC), comprised of TSC1 and TSC2, functions to inhibit mTORC1 signaling. This inhibition can be relieved by hormonal and lipid inputs, which induce the phosphorylation of TSC2 resulting in the inhibition of the TSC1/2 complex. In fact, the activation of mTORC1 is tightly regulated by nutrients, hormones, and lipids, particularly the branched chain amino acid (AA) leucine, insulin, and phosphatidic acid (PA) respectively. While the insulin-mediated activation of mTORC1 is largely elucidated, the mechanism through which PA and AAs function is not currently understood. Additionally, although the individual effect of each agonist on mTORC1 signaling has been studied, further research is required to identify the relative contribution of each input.
Therefore, the first aim of this work was to identify the mechanism through which PA functions to stimulate mTORC1 activity. The present studies demonstrate that PA functions through a receptor based mechanism that stimulates the extracellular signal-regulated kinase (ERK) cascade. The inhibition of ERK signaling blocked the PA-mediated stimulation of mTORC1 signaling, clearly demonstrating that the primary mechanism utilized by PA to stimulate mTORC1 is activation of the ERK cascade. Additionally, AAs and PA signal to mTORC1 through parallel pathways, as mTORC1 signaling increased additively in response to leucine and PA treatments. This is significant since the literature suggests that AAs signal to mTORC1 through the production of PA.
The second aim examined mTORC1 signaling in response to the activation of two signaling cascades that are involved in muscle hypertrophy, the ERK and protein kinase B (PKB) pathways. It was demonstrated that ERK and PKB, also called Akt, increased mTORC1 signaling through parallel pathways. The inhibition of either pathway reduced mTORC1 signaling partially in response to lysophosphatidic acid (LPA) and insulin treatment, which activate ERK and Akt respectively. The inhibition of both pathways completely blocked mTORC1 signaling in response to either agonist. Further, in the absence of TSC2, neither ERK nor Akt regulated mTORC1 signaling. This suggests that a functional TSC1/2 complex is required for the ERK- and Akt-mediated regulation of mTORC1 signaling.
Overall, the data presented herein demonstrate that the combination of multiple stimulatory inputs increase mTORC1 signaling in an additive fashion. ▼ The major determinant of an organization success today in the changing
and turbulent environment is the successful implementation of policies. TSC
cannot be left out of this major effort. It must follow the critical path of success,
for its own good and for the future of Kenyan citizens who want to pursue
education and need to be taught by teachers. Clear and well defined policies must
be put in the right perspective. This may call for a comprehensive effort to
develop a policy manual, which will be used as the source of reference for all
TSC core activities, especially in deployment of teachers, which is the centre of
focus for this research.
The study sought to fulfill four objectives namely:
i) To establish whether TSC has policies on deployment of teachers
ii) To determine whether TSC has put in place adequate measures to
facilitate implementation of policies on deployment on teachers.
iii) To establish whether TSC has been implementing staffing policies on
deployment of teachers
iv) To establish the possible problems TSC has encountered in implementing the staffing policies on deployment of teachers
To realize these objectives, there were four research questions that guided this
study:
i) What are the staffing policies on deployment of teachers by TSC?
ii) What measures has TSC put in place to facilitate the implementation
of deployment policies?
iii) How has TSC been implementing staffing policies on deployment of
teachers?
iv) What problems has TSC encountered in the implementation of staffing
policies on deployment of teachers? Literature review was carried out under the themes, concept of the term
policy, problems of successful implementation of policies, norms and practices of
teacher utilization, imbalances in the deployment of teachers, some
recommendations for action in the deployment of teachers, problems of
implementation of policies in the education sector and fmally, policy
implementation approaches.
The concept of the term policy brought in various writers. Policy means
guidelines meant to encourage work towards stated goals. The problem of
successful implementation of policies gave an overview of what hinders
implementation of policies such as lack of skills to implement, fear of change and
the policy itself being poorly thought out. Norms and practices of teacher
utilization described the ideal pupil-teacher ratio. Imbalances in the deployment
of teachers looked at the urban schools which are overstaffed and compared them
with rural schools which are understaffed. Some recommendations for action on
deployment of teachers suggested ways in which teachers can be utilized more
effectively and motivated. Some recommendations included in-servicing of
teachers, bonding of teachers in disadvantaged areas, addressing teachers welfare
including housing, giving teachers remote area allowance and decentralization of
administrative control of teachers. Problems of implementation of policies in the

▼ The classification problem is a critical issue in data processing field, especially emph{time series classification} (
TSC) problem. In the
TSC problem, the calculation of the distance of two time series is the kernel issue.
One of the famous methods for the distance calculation is the emph{dynamic time warping} (DTW), based on the dynamic programming. However, the time complexity of DTW is O(n
2). When the data size is large, it takes too much time to calculate. In order to overcome time consuming problem, emph{dynamic time warping with window} (DTWW) combines the warping window into DTW calculation. This method reduces the computation time by restricting the number of possible solutions, so the answer of DTWW may not be the optimal solution. In this thesis, we present a method that expands the possible solutions in the minimum first order. Our method not only reduces the required computation time, but also gets the optimal answer.

▼ La sclérose tubéreuse de Bourneville (STB) est une maladie génétique et multi-systémique à transmission autosomique dominante due à des mutations d’un gène TSC1 ou TSC2 qui codent respectivement pour hamartine et tuberine ayant une action inhibitrice sur la voie de signalisation mTOR. L’épilepsie précoce et pharmacorésistante est la manifestation neurologique la plus fréquente et la plus délétère de la STB. Elle débute souvent dans la première année de vie par des spasmes infantiles qui évoluent avec l’âge et en absence de traitement vers des crises toniques ou tonico-cloniques. Bien que les crises soient supposées être générées dans des tubers corticaux, les mécanismes de l’épilepsie ne sont pas bien élucidés et le traitement reste souvent inefficace. Des études morphologiques ont montré une altération de l’expression ARNm des récepteurs au glutamate dans les cellules géantes et les neurones dysplasiques des tubers, mais leur implication fonctionnelle restait à montrer. Les différentes sous-unités NMDA ont une expression âge-dépendante et région-spécifique, les plus grands changements survenant au début de la vie quand l’épilepsie de la STB apparaît. Ce travail avait pour but d’étudier à l’aide de méthodes électrophysiologiques in vitro et in vivo l’expression fonctionnelle des sous-unités NMDA aberrantes et de déterminer leur rôle dans l’épileptogènese chez les souris hétérozygotes Tsc1+/- et sur le tissu humain STB post-opératoire. Nous avons pu démontrer que : i) Les souris hétérozygotes pour le gène Tsc1 sont spontanément épileptiques in vivo et in vitro dans une courte fenêtre dévelopmentale de P9 à P18. ii) Elles présentent une altération d’expression des récepteurs NMDA couche-spécifique et mTOR dépendante avec une surexpression des sous-unités GluN2C/D dans la couche 4 et 2/3 et GluN2B dans les couches 2/3. Cette expression anormale est prévenue par l’administration d’un inhibiteur de la voie mTOR, la rapamycine. iii) Les mêmes altérations d’expression des récepteurs NMDA, sont montrées sur les tissus post-opératoires, non seulement de tubers de STB mais aussi des dysplasies corticales focales (DCF), ces deux malformations ayant des similarités étiologiques et physiopathologiques. iv) La RT-PCR quantitative confirme une expression excessive de GluN2C dans le cortex de souris Tsc1+/- et sur le tissu humain des tubers et DCF. v) Les décharges épileptiques chez la souris Tsc1+/- sont générées dans la couche granulaire 4 du cortex avant de se propager vers les couches superficielles et les couches profondes, empruntant ainsi les microcircuits corticaux. vi) L’expression excessive de la sous-unité GluN2C dans le cortex contribue à l’hyperexcitabilité neuronale chez la souris Tsc1+/- et sur des tissus humains de tubers et de DCF puisque les crises et les décharges sont bloquées par les antagonistes sélectifs de GluN2C/D. vii) Les crises chez la souris Tsc1+/- suivent une séquence âge-dépendante évoluant du type «spasms-like» vers «tonic-clonic like», rappelant celle de l’épilepsie humaine, avec deux pics de haute…

▼ Eskom Holdings SOC Limited (referred to as Eskom), a public state owned company, is
required to continually demonstrate environmental duty of care and report on its environmental
performance. In furthering its commitment to sustainable development and in line with
environmental compliance of its activities, Eskom is planning to have the Distribution Division
certified against the International Organisation for Standardization (ISO) 14001:2004
environmental management system (EMS) standard by 2014. In general, management review
may quickly turn into a dry review of the items on the agenda, done only to show an auditor the
review was held. As such, the aim of this study was to assess the effectiveness of management
reviews as one of the key continual improvement elements of the ISO 14001:2004
environmental management system within Eskom Distribution: Western Region.
This research has been designed to take place in three stages. These stages are planning,
execution and publication, and preparing the dissertation for marking. The research
methodology included a literature study on various national and international articles and
journals on EMSs that relate to environmental performance. The databases of Eskom
Distribution: Western Region ‒ i.e. the management review presentation results for the 2008/09,
2009/10 and 2010/11 financial years, key significant environmental aspects for the Technical
Service Centres (TSCs) and Minor/Major Engineering Works (MEW), key performance
indicators (KPIs), Risk Audit System (RAS) results, and results of the external EMS audit
conducted from 23–25 April 2007 were used for this research. Questionnaires were sent to the
TSCs’ senior supervisors and Safety, Health, Environment and Quality (SHEQ) management.
An analysis of the questionnaires completed by the TSC senior supervisors showed that all the
participants have seen an improvement in environmental performance due to annual
management reviews, except TSC11, who have not seen management review results.
It can be concluded from the results of the SHEQ management questionnaires that an
improvement (continual improvement as per the ISO 14001 requirement) was observed. The
improvements were identified through improved processes, improved gap analysis, increased
environmental awareness, and addressing risks.
Most of the ISO 14001:2004 requirements for the management review for all financial years
covered in this study were met. In terms of the objectives, targets and KPIs, Eskom Distribution:
Western Region performed effectively and most of the outcomes were implemented according
to the recommendations. Further, based on the external audit conducted from 23 to 25 April 2007, it is clear that Eskom
Distribution: Western Region conformed to the minimum requirements of the ISO 14001:2004
EMS. Their management reviews also met all requirements of the ISO 14001:2004 relating to
management review. This was proven by minutes of previous meetings that covered all the

▼ Flexible AC Transmission Systems (FACTS) have the ability of voltage supportand increase transmission capacity. In order to specify a FACTS devicethat is performing according to expectations in a network, a set of studiesand network analyses must be performed. Part of these studies are done usingpower system analysis programs such as PSS®E, which is a planning toolsimulating large power systems in phasor domain using RMS values. Theseplanning tools are used for evaluating stability and reinforcement needs ina power system. The results play a vital role in investment decisions inthe power system. FACTS devices are modeled in PSS®E using a programminglanguage called FORTRAN. It is important to model FACTS devicesaccurately to avoid misleading results. In this Master thesis, STATCOMand Hybrid-STATCOM models are proposed and programmed accordingto ABB’s control strategy. The models are tested in PSS®E and verifiedagainst detailed models in PSCAD. Also, the models are compared againstother industry wide spread generic models.

▼ The objective of this thesis is to investigate the
heat transfer performance of a partially-confined FC-72 spray with
varying dissolved air concentrations. An experimental test rig
consisting of a spray chamber coupled to a fluid delivery loop
system was used to obtain temperature, pressure, and critical heat
flux (CHF) data. A downward facing nozzle within the spray chamber
allowed the FC-72 fluid to be sprayed onto an upward facing,
thick-film, resistor heater. The heater was mounted onto a glass
post, with a sump system to allow removal of excess fluid. Type-E
thermocouples were imbedded in the post to obtain temperature data.
The parametric ranges for experimental testing were as follows:

data were obtained for comparison of CHF with varying C while
controlling the spray chamber pressure. No significant variation in
heat transfer performance was observed. The applicability of
Henry's law to the current system was investigated, and air
concentration measurements using Henry's law were compared with
those obtained using a direct sample method. The resulting air
concentration measurements did not agree. An empirical mathematical
relationship allowing for determination of surface heat flux with
varying flow rate was also developed. The relationship was obtained
using test data at flow rates of V̇ = 6.31, 8.41, and 10.5
cm
3/s (6.0, 8.0, and 10.0 gph), and was
validated using experimental data obtained for flow rates of V̇ =
7.36 and 9.46 cm
3/s (7.0 and 9.0
gph).

▼ Tuberous sclerosis complex (
TSC) is a multi-system, neurocutaneous disorder with neuropsychiatric features known as
TSC-associated neuropsychiatric disorders (TAND). While 90% of individuals with
TSC have some TAND features, only 20% receive treatment, leading to a 70% treatment gap. This study evaluated perception of disease severity, presence of anxiety and depression, as well as the utilization and barriers towards mental health services among adults with
TSC. Disease severity had a moderate and low-moderate association with anxiety and depression, respectively. Regardless of past utilization, respondents had a positive outlook towards the use of mental health services with the major barrier being cost.

▼ Magistrsko delo obravnava parametrično analizo voziščne konstrukcije z numerično metodo - metodo končnih elementov. Delo je sestavljeno iz petih delov, in sicer je v samem začetku dela teoretični del, kjer je na kratko opisana zgodovina in prihodnost voziščne konstrukcije ter splošen opis voziščne konstrukcije sedanjosti. V nadaljevanju je predstavljeno dimenzioniranje voziščne konstrukcije po tehničnih specifikacijah za ceste - TSC. Našteti in opisani so vsi pogoji, priporočila ter zahteve, ki jih mora načrtovalec oz. projektant upoštevati pri načrtovanju in dimenzioniranju voziščne konstrukcije. Po teoretičnem delu sledi praktičen primer dimenzioniranja voziščne konstrukcije po TSC (empirični postopek dimenzioniranja voziščne konstrukcije), kjer obravnamo realen primer odseka ceste v občini Starše. V zadnjem delu je podana parametrična analiza voziščne konstrukcije, izvedene z metodo končnih elementov (MKE), sledi zaključek. Na podlagi parametrične analize smo prikazali vpliv dimenzij in lastnosti vgrajenih materialov ter nosilnost podlage na trajnost voziščnih konstrukcij. Rezultate analitične metode smo primerjali s tehničnimi specifikacijami za ceste. Pri analitični metodi smo uporabljali programsko orodje Everstress.
The master’s thesis discusses parametric analysis of the roadway structure by numerical method – the finite element method. The master’s thesis consists of five parts
at the very beginning there is a theoretical part, which briefly describes the history and future of the roadway structure, and a general description of the roadway structure in the present. Afterwards the dimensioning of roadway structure according to the technical specifications for roadways (TSC) is presented. We listed and described all conditions, recommendations and requirements which the project planner must apply when designing and dimensioning the roadway structure. Following the theoretical part there is a practical example of dimensioning roadway structure according to the TSC (empirical procedure of dimensioning of roadway structure), where we deal with a realistic example of a road section in the municipality Starše. In the last part a parametric analysis of the roadway structure by finite element method (MKE) is given and a conclusion follows. Based on the parametric analysis we have shown the impact of dimensions and properties of the built-in materials and the bearing capacity of the foundation to the durability of roadway structures. We compared the results of the analytical method to the TSC. We used Everstress software for the analytical method.

▼ Magistrsko delo predstavlja določitev brezdimenzijskih faktorjev armiranja voziščnih konstrukcij z geomrežo s pomočjo dveh ločenih metod za dimenzioniranje asfaltiranih in neasfaltiranih voziščnih konstrukcij, ojačanih z geomrežo. Predstavljena je metoda dimenzioniranja voziščnih konstrukcij po TSC in način, kako bi lahko pripadajoč brezdimenzijski faktor armiranja vključili v postopek dimenzioniranja po TSC. Opisani so postopki za pridobitev ključnih parametrov, ki so potrebni pri dimenzioniranju voziščnih konstrukcij.
Master’s thesis represents the determination of non-dimensional factors of road reinforcement determined with two separate methods for the roadway design of unpaved and paved roadway construction reinforced with geogrid. The design method for roadway construction by TSC is presented and it’s described how we could integrate non-dimensional factor in roadway design construction by TSC. Some procedures of mean parameters needed for roadway dimensioning are described.

▼ Definitionen av ett projekt är att det måste gå att mäta om ett projekt nått sitt mål eller inte. Ett projekt skall också ha en klar och tydligt fastställd slutpunkt, där projektgruppen upplöses och projektet avslutas. De metoder som valts att användas för datainsamling är intervjuer med projektledarna och kunderna. Även litteraturstudier har använts för att kartlägga redan existerande kunskap inom området samt för att bygga upp en teoretisk referensram. I denna studie beskrivs arbetsgången hos två projekt. Vid jämförande och analys av arbetsgången vid de båda projekten visade det sig att det fanns både likheter och skillnader. I resultatet redovisas de båda projektens arbetsgång, samt vad projektledarna och kunderna har för åsikter om respektive projekts genomförande och slutresultat. Slutsatsen som man kan dra av jämförandet av Projekt A och B med den teoretiska projektgången är att i många fall oberoende på storleken på ett projekt så genomförs kundorderprojekt på samma sätt.
The definition of a project is that the goal must be able to be measured after the project is finished. A project must have a clear ending point where the project group is dissolved. The methods that have been chosen for collecting material are interviews with the project managers and the customers. Litterature has also been studied for a theoretical view. In this study two projects are analyzed. At comparison between the projects, it shows that there are both similarities and differences between the way to work in a project. In the result it is described how the two projects where carried out and also what the project managers and the customers thought of the realization and end result. The conclusion that you can draw is that in many cases the realization are not depending of size of the project. The projects are carried out the same way independent of the projects size.

▼ Les protéines GILZ (Glucocorticoid-Induced Leucine Zipper) et TSC-22 (Transforming growth factor-beta Stimulated Clone-22) appartiennent à la famille de protéines TSC-22D (TSC-22 Domain). GILZ a été décrit précédemment comme étant induit au cours de la déprivation en interleukine-2 (IL-2) des lymphocytes de la lignée cellulaire CTLL-2, permettant ainsi de retarder leur apoptose. Le but de notre travail était de déterminer les rôles respectifs de GILZ et TSC-22 au cours de l’apoptose des cellules CTLL-2.Nos résultats ont permis de montrer que TSC-22 augmentait l’apoptose induite par la déprivation en IL-2 des cellules CTLL-2. Nous avons mis en évidence une augmentation de l’activation des caspases ainsi qu’une régulation positive de l’expression de BIM. Nous avons en outre montré que l’expression de GILZ, protéine anti-apoptotique, induite lors de la déprivation en IL-2, était régulée négativement en présence de TSC-22. Enfin, nous avons montré que l’expression de l’ARNm de gilz était régulée négativement par TSC-22, mais que la stabilité de son ARNm n’était pas modifiée.Notre travail a donc permis de montrer que TSC-22 accélère l’entrée en apoptose des lymphocytes T en régulant négativement l’expression de la protéine anti-apoptotique GILZ.
GILZ (Glucocorticoid-Induced Leucine Zipper) and TSC-22 (Transforming growth factor-beta Stimulated Clone-22) belong to the TSC-22D (TSC-22 Domain) family of proteins. GILZ has been previously shown to be induced upon interleukin-2 (IL-2) deprivation in the T-cell line CTLL-2, allowing cells to delay apoptosis. The aim of our study was to elucidate the respective roles of GILZ and TSC-22 during IL-2 deprivation-induced T-lymphocytes apoptosis.Our results demonstrated that TSC-22 increased CTLL-2 cells apoptosis induced upon IL-2 deprivation. We highlighted in TSC-22 expressing cells both an increase in caspases activation and BIM expression up-regulation. We also demonstrated that GILZ expression, an anti-apoptotic protein, known to be induced after IL-2 withdrawal, was down-regulated in the presence of TSC-22. Moreover, we showed that gilz mRNA expression was also significantly repressed, but gilz mRNA half-life was not modified.Altogether, these results suggest that, in T-cells, TSC-22 could behave as a repressor of GILZ expression, accelerating IL-2 deprivation-induced apoptosis.

▼ Este trabalho aborda o emprego do Compensador Estático de Reativos (SVC) em redes de baixa tensão com geração distribuída de energia, tendo como objetivo a compensação de tensão e de fator de potência. Os principais distúrbios de qualidade de energia elétrica em baixa tensão, bem como as normas e os limites operacionais para os principais indicadores foram revisados, visando confrontação com as situações resultantes da integração do SVC no sistema elétrico. O SVC possui operação estabelecida como carga reativa controlável em sistemas de transmissão de energia, fazendo parte dos condicionadores da tecnologia FACTS (Flexible AC Transmission Systems). A exploração desta tecnologia em outro nível de tensão é o objetivo deste trabalho. As análises foram realizadas através de simulações computacionais, desenvolvidas no ambiente MATLAB, contemplando o desenvolvimento de modelos relacionados com aplicações de RCT (Reator Controlado a Tiristor), FC (Capacitor Fixo), do CCT (Capacitor Chaveado a Tiristor) e de um sistema de geração distribuída de energia elétrica trifásico em uma rede de baixa tensão a quatro fios. A metodologia para o dimensionamento dos elementos passivos dos condicionadores é apresentada, bem como a análise dos valores de potência reativa, fator de potência, distorções harmônicas e a avaliação da necessidade de utilização ou não de filtros passivos. A teoria da potência conservativa (TPC) é a metodologia utilizada para definição das grandezas elétricas e fatores de conformidade. Por fim, as características operacionais de um SVC aplicado a uma rede de baixa tensão a quatro fios são exploradas através dos resultados de simulação, com o objetivo de demonstrar a manutenção de suas características operacionais estabelecidas, no nível de baixa tensão, e apontar aspectos diferenciados quanto à regulação de fator de potência e da forma de tensão providos pelo SVC, para servir como informação de confronto frente a outras tecnologias comumente utilizadas neste nível de tensão.
This paper discusses the use of Static Var Compensator (SVC) on low voltage grids with distributed generation with the aim of voltage and power factor compensation. The main electrical power quality disturbances at low voltage as well as the rules and operational limits for the main indicators were reviewed aiming to confront the situations resulting from SVC integration in the electrical system. SVC has established operation as controllable reactive load in power transmission systems, being part of FACTS (Flexible AC Transmission Systems) technology conditioners. The application of this technology in another level of tension is the objective of this work. ▼ The purpose of this study is to assess the effectiveness of information, education and communication (lEC) materials
as a communication strategy at the Teachers Service Commission (TSC). The TSC has since 1999 embarked on an
aggressive campaign to empower its employees. A public relations (PR) section was established to steer
communication activities. Acres of materials were produced to disseminate information. However, despite these
efforts, complaints about corruption, inefficiency, poor corporate image and insensitivity to customer needs still
abound. There seemed to be a disconnect in the communication process between the employer and the employees.
The study assesses whether the communication strategies in place have been effective.
Four objectives were investigated. These were: the level of awareness of the IEC materials amongst TSC employees;
their impact on the employees; the appropriateness of their content; the channels used and the prevailing feedback
mechanisms. The study was conducted using an exploratory survey. The participants were TSC employees sampled from Nairobi
and Meru districts. A total of 250 employees were selected through stratified and random sampling techniques. Data
were collected through a self- administered questionnaire and a total of 199 questionnaires were returned.
The study established that although TSC has used multifaceted corporate communication strategies to inform and
empower its employees to realize set goals and objectives, the staff are not aware of most of the strategies
introduced after 1999.TSC employees have not been empowered by the information disseminated in terms of giving
them motivation, job mobility or satisfaction, in addition to enhancing their perception of a good image of the
organization. The study also indicated that feedback mechanisms within TSC communication strategies are poor. On
the effectiveness of the channels used, memos/circulars were found to be the most popular with website/Email and
suggestion boxes being the most unpopular. TSC employees felt there was need for frequent contacts with their
employer. However, the findings indicated that a climate of fear and distrust exists between the employees and their
supervisors. Employees are not free to discuss problems and difficulties they have in their job for fear of
jeopardizing their positions or having it held against them. In view of these findings, the study recommended that TSC carries out an audit of its communication strategies to
identify gaps that require urgent interventions. An aggressive sensitization campaign should be carried out to inform
TSC staff of the existence of the various IEC materials and the need to embrace them to realize effective service.
