▼ Health seeking behaviour of people around the globe will be affected by different socio-cultural and economical factors. People in Ethiopia, particularly tribal people have long history of using medicinal plants as a major component of their traditional medical practices over many centuries to satisfy their health care needs. The transfer of this sort of knowledge from generation to generation is mainly effected by words of mouth. Cross-sectional survey was conducted to document ethnopharmacological information of the Hamer semi pastoralists by using semi structured questionnaires administered to eight traditional healers/key informants and one thousand six hundred household (HH) respondents supplemented by eight focus group discussions (FGDs). The majority (84%) of HH respondents were males, 94.4% of whom suffered illnesses in the last two weeks recall period and had taken action. The prevalence of the use of traditional medicine among the Hamer community was found to be very common as indicated by FGD participants, key informants and HH respondents. HH respondents reported that traditional medical practices are their first choice when family members get sick. Females’ preference of traditional medical practices was higher than males. The main reasons for the preference of the community members to traditional medical practices include its effectiveness, low cost and ease of availability. A total of sixty medicinal plants were reported and collected along with their local names, indication(s), parts used and method of preparations. The health seeking behavior of the Hamer ethnic group is affected by different socioeconomic and cultural factors. There is also a strong indication for traditional medical practices and use of plant materials to treat various ailments and health problems among the study population. Therefore, due attention should be given to these traditional medical practices. The ethnopharmacological survey part of this study revealed that Aloe otallensis Baker., an indigenous Ethiopian plant, was among the most widely used medicinal plants by the members of the Hamer Ethnic Group. The plant is used for the treatment of malaria and wound healing. Malaria, a diseases caused by protozoan parasites is a major obstacle to the socioeconomic xii development of several developing countries including Ethiopia. The phytoconstituents of A. otallensis studied by thin layer chromatography (TLC) and its antimalarial and antioxidant activities were assessed by in vivo four day Plasmodium berghei suppressive test and 1,1- diphenyl-2-picrylhydrazyl (DPPH) assay, respectively. The study revealed the presence of picataloside (2,8-O,O-di (β-D-glucopyranosyl)-1,2,8-trihydroxy-3-methylnaphtalene) in the exudate of A. otallensis, which was isolated from the plant for the first time. The study further showed that the leaf exudate of the plant possesses maximum antimalarial activity (60.7% suppression) at a dose of 300 mg/kg. Les résines végétales sont des sécrétions de végétaux qui ont été utilisées par l’homme de l’Antiquité à nos jours dans de nombreuses applications (pharmaceutique, cosmétique et artistique). Ces exsudats sont composés majoritairement de terpènes. L'identification et la quantification de l'ensemble de ces composés dans les extraits végétaux reste un défi du fait de leur très grande diversité structurale. L’objectif de ce travail a été de développer de nouvelles approches analytiques pour identifier et quantifier les composés terpéniques présents dans ce matériel végétal afin d’en assurer le contrôle qualité et la certification. Deux méthodes séparatives ont été sélectionnées: la TLC et la GC. Pour ces deux techniques on s’est intéressé à toutes les potentialités de leur couplage avec la spectrométrie de masse. Le développement en TLC-1D et TLC-2D a permis le « screening » rapide des résines végétales et la faisabilité du couplage avec le MALDI-TOF-MS a été mise en évidence pour l’identification des marqueurs majoritaires (acides triterpéniques). La GC a permis une caractérisation plus aboutie des résines en mettant en place une méthode d’analyse exhaustive des terpènes des plus volatils au non-volatils. L’optimisation des différentes étapes de la méthodologie GC-MS s’est effectuée en se basant sur la méthode des plans d’expérience ainsi que sur des analyses statistiques tels que l’ACP et la CAH. Dans un souci d’apporter des éléments plus précis pour distinguer les résines les plus proches, la quantification de leurs marqueurs majoritaires a été établie après une validation complète de la méthode GC. L’ensemble de ce travail a permis de développer des outils pour une caractérisation rapide des extraits de résines permettant de différencier les espèces même les plus proches.

Resins are hydrocarbon secretions of many plants and well known for their protective benefits. They have been used as raw materials for a wide range of applications (pharmaceutic, cosmetic and artistic). Plant resins are complex mixtures of organic substances mainly terpenoid compounds which constitute the most abundant and structurally diverse group of plant secondary metabolites. The chemical characterization of this material results in long and difficult separation due to the wide range of polarity and volatility of its constituents. The aim of this work was to develop new analytical approaches to improve the identification of resins certifying their origin and ensuring the quality control. For that purpose two analytical methods were selected: TLC and GC approaches hyphenated to mass spectrometry. TLC-1D and TLC-2D allow a rapid screening and first visual differences of resins. The innovating TLC coupling to MALDI-TOF-MS gives a clear identification of major markers (triterpenic acids). CD4 count is the major surrogate marker for defining the best time to start antiretroviral therapy in HIV infected patients, as well as for defining the duration of prophylaxis against opportunistic infection. Its high cost, however, limits the use of this technique in resource-limited settings. Total lymphocyte count (TLC) has been evaluated as a substitute marker for CD4 count. The objective of this study is to evaluate the capability of TLC to be used as a substitute marker for CD4 count in order to detect patients who need prophylaxis against opportunistic infection (CD4 <200 cells/mm3), and those with CD4 <350 cells/mm3 (Brazilian limit to define AIDS). We evaluated TLC and CD4 of 1174 HIV-infected patients, in HUPES, from May/2003 to September/2004. CD4 counts were performed by flow cytometry, and TLC by an automated hematological counter. A total of 1510 subjects were evaluated. The CD4 count range was 4 to 2531 cells/mm3), and the TLC range 300 to 6200 cells/mm3. The best TLC limit to estimate a CD4 <200 cells/mm3 was ≤ 1700 cells/mm3 (SE= 76.3%; SPE= 65.2%, NPV= 93.1%), but the PPV was only 31.1%. The same findings were observed for the limit of <350 cells/mm3, but we find a worst sensitivity (SE= 59.4%, SPE= 96.6%, PPV= 57.3% and NPV= 79.4%). Altogether, these results indicates that although a statistical correlation exists between TLC and the CD4+ T cell count, the TLC is not a good predictor of the CD4+ T cell count. TLC would not be a safe marker for a CD4+ T cell count in HIV-infected patients, but could be used as an surrogate to monitoring the need to perform CD4 cell count.

A contagem de células T CD4+ é o principal marcador imunológico usado para definir o momento adequado para início da terapia antiretroviral e profilaxia para infecções oportunistas em pacientes portadores do vírus HIV. Seu alto custo limita o uso desta técnica em muitos locais no mundo. O objetivo deste estudo foi avaliar a utilidade da TLC em substituir a contagem de células T CD4+ como marcador imunológico para detectar os pacientes com necessidade de profilaxia contra infecções oportunistas (CD4 <200 cel/mm3), e aqueles com CD4 <350 cel/mm3 (limite brasileiro que define AIDS). Nós analisamos a TLC e contagem de células T CD4 de 1174 pacientes infectados pelo HIV provenientes do HUPES durante o período de maio/2003 a setembro/2004. A contagem de células T CD4+ foi realizada por citometria de fluxo e a contagem total de linfócitos por contador hematológico automatizado. Foram analisadas um total de 1510 amostras. A contagem de células T CD4+ teve valor mínimo de 4 cel/mm3 e valor máximo de 2531 cel/mm3, TLC teve mínimo de 300 cel/mm3 e máximo de 6200 cel/mm3. O melhor valor de TLC, capaz de predizer CD4 <200 cel/mm3, foi ≤ 1700 cel/mm3 (SE=76,3%; SPE=65,2%, NPV=93,1%), mas PPV de somente 31,1%. Tyrimo objektas ir metodai: Optimizuojant PC metodiką, analizuoti etaloniniai 0,2 mg/ml koncentracijos alprazolamo, kodeino, paracetamolio trichlormetaniniai tirpalai ir jų mišinys. Tirpiklių sistemoms buvo naudoti etanolis, trichlormetanas, eteris, 25% amonio hidroksidas, acetonas, izobutanolis. Dėmių ryškinimui naudota UV šviesos (254nm; 365nm) lempa arba Dragendorfo reagentas (modifikuotas pagal Munjė). Optimizuotos metodikos pritaikytos tiriant trichlormetaninius darbinius tirpalus, pagamintus iš vaistinių preparatų „Xanax“, „Paracetamolis Sanitas“ ir „Ultracod“. Siekiant pritaikyti ESC metodiką tiriamųjų junginių analizei, buvo tirti etaloniniai 0,1 mg/ml koncentracijos alprazolamo, kodeino ir paracetamolio metanoliniai tirpalai bei jų mišinys. Medžiagų atskyrimui ir identifikavimui naudotas chromatografas Waters 2695 su fotodiodų matricos detektoriumi Waters 996 (210 – 400 nm bangų ilgio diapazonas). Chromatografavimui naudoti metanolis, 3% acto rūgšties vandeninis tirpalas. Optimizuota ESC metodika pritaikyta tiriant metanolinius darbinius tirpalus, pagamintus iš vaistinių preparatų „Xanax“, „Ultracod“ ir „Solpadeine“. Aim: To optimize thin-layer chromatography and high-performance liquid chromatography methods for alprazolam, codeine, paracetamol and their mixture qualitative analysis. Object and methods: For TLC method optimization alprazolam, codeine, paracetamol and their mixture stock solutions (0,2 mg/ml) in trichlormetan were analysed. For mobile phase were used: ethanol, trichlormetan, ether, 25% ammonia hydroxide, acetone, isobutanol. For spots development were used UV light lamp (254nm; 365nm) or Dragendorff reagent (modified by Munje). Optimized methods were tried with pharmaceutical products “Xanax”, “Paracetamolis Sanitas” and “Ultracod” solutions. For HPLC method optimization alprazolam, codeine, paracetamol and their mixture stock solutions (0,1 mg/ml) in methanol were analysed. Chromatograph Waters 2695 with photo diode array detector Waters 996 (210-400 nm wave length) were used for qualitative determination. Analysis was made by using methanol and 3% acetic acid aqueous solution. Optimized method was applied in analysis of pharmaceutical products “Xanax”, “Ultracod” and “Solpadeine” solutions. Dissertação de Mestrado em Segurança Alimentar

A produção de mel em Portugal tem sido, geralmente, encarada como uma actividade de lazer, ainda que ultimamente tenham aparecido apicultores que se dedicam de forma profissional à apicultura possuindo já um número elevado de colmeias. A instalação que se vem verificando de uma situação do colapso das abelhas (desaparecimento das abelhas da colmeia) e a ameaça que poderá representar para a agricultura no que respeita à polinização, constituem uma preocupação a nível mundial. A preocupação dos consumidores no que respeita à segurança alimentar, particularmente relacionada com a presença quer de contaminantes químicos como microbiológicos esteve na base do desenvolvimento deste trabalho. Foi desenvolvido um processo de extracção para permitir que a Cromatografia de Camada Fina fosse usada como metodologia de rastreio de pesticidas no mel. Deveria ter sido possível detectar Amitraz ao nível de 0,2 μg/g (LMR 200 μg/kg), Cumafos 0,1 μg/g (LMR 100 μg/kg), Lindano e Metiocarbe 0,00001 μg/g (LMR 0,01 μg/kg) e então confirmar com uma técnica quantitativa. Mas o limite de detecção atingido não se conseguiu aproximar dos valores do Limite Máximo de Resíduo (LMR), sendo necessário prosseguir esta linha de investigação. Caracterizou-se a oferta de mel, enunciando que tipos de méis estão à disposição do consumidor no mercado da região de Lisboa. Analisou-se a rotulagem das embalagens de mel, com a finalidade de observar se a informação fornecida ao consumidor cumpria os requisitos legais. Do conjunto 27% das embalagens apresentaram-se como não conformes. O procedimento de extracção desenvolvido foi aplicado em 23 amostras de mel adquiridas no mercado da região de Lisboa. Procedeu-se também à análise microbiológica das mesmas amostras. Os contaminantes químicos pesquisados foram o lindano, cumafos, metiocarbe sulfóxido e amitraz, pois são utilizados pelos apicultores ou persistem no ambiente e têm sido detectados no mel. Detectou-se pesticidas em 5 das 23 amostras. Em relação aos contaminantes microbiológicos foi pesquisada a presença de Salmonella e de esporos de clostrídeos sulfito-redutores e efectuou-se a contagem de bolores e leveduras. Não foram detectados quaisquer dos microrganismos analisados em nenhuma das amostras de mel. Conclui-se que as amostras de mel analisadas apresentaram, uma elevada qualidade microbiológica.

▼ La plupart des bactéries ont un unique chromosome circulaire. Lors de la réplication de l’ADN, la circularité lie topologiquement les deux chromatides sœurs résultant de la réplication (caténanes et dimères). Ces liens topologiques doivent être résolus afin de permettre une bonne ségrégation de l’information génétique entre les deux cellules filles au cours de la division cellulaire. Les bactéries possèdent une machinerie très conservée: les recombinases à tyrosines XerC et XerD, capables de résoudre les dimères et une partie des caténanes, en catalysant un crossover au site spécifique dif situé dans la région Ter du chromosome. Lors de ce processus elles réalisent successivement deux échanges de brins. La réaction Xer est spatio-temporellement contrôlée par une protéine du divisome: FtsK. FtsK est une translocase qui pompe l’ADN à travers le septum de division. Lorsqu’elle rencontre une synapse constituée de deux sites dif chargés de XerC et XerD, elle active la catalyse de XerD pour initier le premier échange de brins. Dans un second temps XerC catalyse un second échange de brins indépendamment de FtsK. A ce jour le mécanisme d’activation de XerD n’est pas bien compris. Certains éléments mobiles résolvent leur états multimériques (tels que les plasmides) ou intègrent leur génome dans celui de leur hôte en détournant les recombinases XerCD. On parle d’IMEXs (integrative Mobile Element using Xer). Les éléments mobiles étudiés avant ma thèse utilisaient tous des voies de recombinaison initiées par la catalyse de XerC et ne nécessitant pas l’activation de XerD. Au cours de ma thèse j’ai étudié dans un premier temps le mécanisme d’intégration / excision d’une nouvelle classe d’IMEXs en utilisant comme modèle le phage TLCphi de Vibrio cholerae, la bactérie responsable du choléra. Par des approches de génétique j’ai démontré que TLCphi utilise une voie de recombinaison initiée par la catalyse de XerD et indépendante de FtsK. Mes travaux ont également montré que l’excision du phage participe à l’évolution des souches pandémiques de V.cholerae. Dans une seconde partie, j’ai identifié un facteur phagique qui permet à TLCphi de contourner le contrôle de FtsK sur l’activation de XerD. Ce facteur était une protéine de fonction inconnue présentant un domaine HTH et un domaine DUF3653. Ce dernier est retrouvé dans de nombreux IMEXs. Par des approches de biologie moléculaire j’ai étudié le mécanisme d’action de cette protéine. J’ai reproduit la réaction de recombinaison in vitro et démontré qu’elle active XerD en interagissant directement avec elle. Enfin dans un troisième temps, nous nous sommes intéressés aux disparités observées entre la recombinaison Xer chez E.coli et V.cholerae. En particulier, la recombinaison Xer semble agir seulement sur les dimères chez E.coli alors qu’elle est active également sur les monomères chez V.cholerae. Nous avons démontré que ces divergences de comportement ne viennent pas des Xer elles-mêmes, ni de leurs propriétés d'activations par FtsK. The aim of this work was to optimize instrumental analysis methods designed for separation and identification of bromazepam, clorazepate and nimesulide from their mixture samples. In order to achieve our goal we have set ourselves these tasks – to optimize thin-layer chromatography method for separation and identification of bromazepam, clorazepate, nimesulide mixture components; to optimize the thin-layer chromatographic method for the quantitative determination of bromazepam, clorazepate and nimesulide; to optimize separation and identification of components from bromazepam, clorazepate, nimesulide mixture using high-performance liquid chromatography method and to adapt and develop methodology design for plasma sample analysis.

Les enzymes et leur diversité d’actions sont appréciées dans des domaines d’applications variés allant del’agroalimentaire à la thérapeutique. Ainsi, une attention toute particulière est portée à leur étude afin d’améliorer uneaction (contre le vieillissement de la peau, antivirale, anticancéreuse…) ou un procédé de synthèse. Ce projet derecherche s’inscrit dans une démarche de développement d’outils analytiques basés sur la spectrométrie de masse,permettant le suivi rapide et sensible d’interactions enzyme-ligand.Dans une première étude, l’approche TLC couplée à une détection par UV a été évaluée pour la déterminationde constantes enzymatiques de l’enzyme invertase. Cette approche couplée à un MALDI/TOF MS a permis d’identifierdes substrats spécifiques de l’invertase au sein d’extraits de plantes. Pour preuve de concept, l’interactioncellobiohydrolase II–ligand est présentée dans le cadre de l’identification d’inhibiteur par TLC-MALDI/TOF et TLCENALDIMS.En seconde étude, nos travaux ont porté sur la caractérisation directe de différentes enzymes kinases, puis auxsuivis des réactions de phosphorylation de nucléosides /tides endogènes. Ces études, basées sur des approches « offline» (Flow Injection Analysis, FIA) et « on-line » (Frontal Affinity Chromatography, FAC) couplées à unspectromètre de masse haute résolution, ont été réalisées au moyen de ces kinases libres et immobilisées. Dans le cadrede la recherche de nouveaux candidats médicamenteux antiviraux, le suivi d’une phosphorylation spécifique desmolécules de synthèse, au regard de souches humaine ou virale de kinase, a également été évalué par ces deuxméthodologies.

Enzymes are very appreciated and useful in various application fields from agri-business to therapeutic due to theirdiversity of actions. Therefore, their action mechanisms are widely studied in order to enhance an action (anti-aging ofskin, antiviral, antitumorous) or a synthesis process. This research project is part of the approach to propose analyticaltools based on mass spectrometry, allowing rapid and sensitive follow-up of enzyme-ligand interactions.In a first study, the Thin-Layer Chromatography (TLC) approach coupled with UV detection was evaluated forthe determination of invertase kinetic constants. This approach coupled with a MALDI / TOF-MS led to theidentification of invertase substrates in plant extracts. As a proof of concept, the cellobiohydrolase II - ligand interactionwas presented in the framework of the identification of inhibitor by TLC-MALDI / TOF and TLC-ENALDI MS.In the second study, our work aimed at developing a direct method for the determination of kinetic parametersof kinases and following-up the phosphorylation reactions of endogenous nucleosides / tides. ▼ Toxigenic Aspergillus species produce mycotoxins that are carcinogenic, hepatotoxic and teratogenic immunosuppressing agents in both human and animals. Kenya frequently experiences outbreaks of aflatoxicosis with the worst occurring in 2004, which resulted in 125 deaths. This study sought to find possible reasons for frequent aflatoxicosis outbreaks in Kenya by isolating Aspergillus flavus strains from maize kernels sampled from different climatic regions of Kenya. Using diagonal transect random sampling, maize kernels were collected from Makueni, Homa Bay, Nandi, and Kisumu regions. The genetic diversity and variation among the isolates was examined by characterising the strains according to morphology, phenotype, vegetative compatible groups and molecular systematics. Selected atoxigenic and aflatoxigenic A. flavus isolates were also further analysed for aflatoxin production potential using quantitative real-time PCR and various bioanalytical techniques. The influence of the maize lines grown in Kisumu, Homa Bay, Nandi and Makueni region on A. flavus infection and aflatoxin production was also examined and served as the basis for an in vitro biocontrol assay. Out of 37 isolates identified, nitrate non-utilizing auxotroph’s complementation test revealed 20 vegetative compatibility groups. These groups were further designated using the prefix ʻʻKVCGʼʼ, where ʻʻKʼʼ represented Kenya and consequently assigned numbers 1 to 20 based on our findings. KVCG14 and KVCG15 had highest distribution frequency (n = 13; 10.8 %). The distribution of the L, S and S/L- morphotypes across the regions were 57 % (n = 21); 7 % (n = 3) and 36 % (n = 13) respectively. The phylogenetic analysis exhibited high diversity of A. flavus isolates from Makueni. ITS1 and ITS2 markers did not reveal significant information within intraspecies speciation of A. flavus. Furthermore, a unique isolate (KSM015) was identified that had characteristics of S-morphotype, but produced both aflatoxins B and G. Coconut agar medium (CAM) assay, TLC, HPLC and LCMS/MS analyses confirmed the presence or absence of aflatoxins in selected toxigenic and atoxigenic isolates. qPCR analysis revealed aflP, aflS, aflR and aflO transcripts as the most upregulated genes across the tested isolates whereas false detection of aflD gene transcript was observed in both induced and uninduced A. flavus isolates. Diversity Index (H) analyses ranged from 0.11 (Nandi samples) to 0.32 (Kisumu samples). Heterokaryon compatibility ranged from 33 % (for the Makueni samples, n = 3) to 67 % (Nandi samples, n = 6). The KDV1 maize line was more sensitive to A. flavus infection in comparison to GAF4. We also tested the biocontrol of atoxigenic isolates to inhibit toxin production by aflatoxigenic strains on infected maize kernels. It was shown that the atoxigenic strain (KSMO12) could inhibit the aflatoxigenic strain (KSM014) depending on the atoxigenic concentration during infection. ▼ ABSTRACT In 2013 the Iowa Teacher Leadership and Compensation planning grant (TLC) committed to providing 150 million dollars annually toward rewarding effective teachers with leadership opportunities and higher pay, attracting promising new teachers with competitive starting salaries, and fostering greater collaboration for all teachers to learn from one another. One-quarter of each district’s teaching staff is now assuming a leadership role by assisting their colleagues in collecting meaningful student data, analyzing student achievement results, and utilizing the findings in order to improve teacher instruction (State of Iowa Department of Education, 2013). These leadership responsibilities had typically been attended to by the building principal, whose job description over the past decade has been shifting away from a traditional managerial position toward an instructional leadership role. While both principal and teacher leadership roles are defined by the Iowa Standards for School Leaders and the Iowa Teaching Standards, it remains unclear as to how these roles are enacted on a daily basis within the school. This study sought to gain a deeper understanding of principals’ viewpoints regarding the changes brought about by Teacher Leadership Initiative (TLI), and to understand how principals’ perceptions of their support from central office administration, including from the superintendent and central office administrators influenced the principals’ understanding of their roles as they navigated this change in leadership structure. Qualitative, semi-structured interviews with 12 secondary principals from Iowa schools were conducted on two separate occasions over a period of two months. Hall’s Concerns-Based Adoption Model (CBAM) served as the conceptual framework for this study as the researcher examined how principals managed change within their positions in order to enlighten and elucidate both the academic and mainstream reader on the complexities of the daily interactions among district-level staff, principals, and teacher leaders within various school contexts. Findings revealed that principals faced a spectrum of emotion as they implemented the teacher leadership initiative. Principals reported feeling optimistic and hopeful about the possibilities that teacher leadership holds for improved teaching and learning. However, they also expressed a variety of concerns as they worked through the steps of implementation. At times, principals experienced personal concerns such as anxiety, jealousy and isolation. Other times, principals felt unsure they had the necessary knowledge and skills to do the new work brought forth by teacher leadership. Principals reported feeling frustrated that there was never enough time in a day to complete their work. ▼ The general goal of this thesis was to explore the chemical ecology and macromorphological variability in lichens that colonize soils in five locations in Manitoba with respect to the carbon-nutrient balance hypothesis (CNBH). The CNB hypothesize states that when a plant or lichen is growing in nutrient-poor conditions, the excess carbon may be shunted into biosynthesis of carbon-based secondary metabolites such as polyketides. Three specific goals were further examined in four chapters to investigate:1) the distribution of secondary metabolites and species assemblages with respect to soil characteristics (Ch. 2), which resulted in 2) a report on Cladonia magyarica and C. humilis in Manitoba, describing the habitat, and providing morphological and chemical comparisons to distinguish five species (Ch. 3), 3) the phenotype in lichen species showing plasticity with respect to soil type, temperature and precipitation (Ch. 4), and 4) the effect of temperature and moisture on the quantity of three secondary metabolites (atranorin, fumarprotocetraric acid, and usnic acid) produced by three lichen species (Ch. 5). Field collections of lichens, soil features and environmental data were made in five locations. Secondary metabolites were determined using Thin Layer Chromatography (TLC) and High-Performance Liquid Chromatography (HPLC). Morphological and fecundity characteristics were podetium height, cup diameter, squamule diameter, apothecium diameter, number of apothecia on cup and presence of apothecia. Soil characteristics were pH, organic matter, sand content, and sand grain shapes. The results showed that usnic acid and atranorin supported the CNB hypothesis. Species as Cladonia magyarica and C. humulis were reported for the first time in Manitoba. The CNB hypothesis was not supported for all lichen species, which may have been complicated by variability in biotic and abiotic conditions. Temperature and moisture affected the stability of secondary metabolites and partial degradation occurred. This thesis forms a foundation for further studies on the relationship between secondary metabolites with biotic and abiotic factors. ▼ Chokeberry consumption has been increasing due to research exposing great potential of health-promoting compounds. However, chokeberries are highly astringent and are typically consumed in processed forms in which product heating is applied. Processing chokeberries has been reported to degrade bioactive compounds, thus limiting the potential for consumers to obtain their health-promoting benefits. The purpose of this research was to examine the effects of chokeberry juice processing and storage on anthocyanin, flavonol, proanthocyanidin, hydroxycinamic acid, and polymeric pigment content, as well as percent polymeric color. In addition, new analytical methodologies were explored to better understand possible outcomes of polymeric pigments due to juice processing and storage. Chokeberries were processed into nonclarified juice with sampling at each stage of processing. Levels of anthocyanins, flavonols, proanthocyanidins, hydroxycinamic acids, and percent polymeric color were also analyzed each month throughout a 6 month storage period at ambient temperature. It was determined that anthocyanins readily degrade during juice processing, especially with heat applications during blanching and pasteurization. Comparatively, other compounds, such as proanthocyanidins were better retained during processing than anthocyanins, with significant levels remaining in the presscake. After pasteurization, lower levels of anthocyanins (7%), flavonols (52%), proanthocyanidins (55%), and hydroxycinamic acids (63%) remained in the juice. Alternatively, polymeric color values increased to 29% throughout processing. During juice storage, polyphenolic levels continued to decrease over 6 months while percent polymeric color values increased further to 44.5%. Little change occurred in levels of total flavonols (447.8 to 406 mg/100g), proanthocyanidins (19.7 to 16.5 mg/100g), and hydroxycinamic acids (72.7 to 48.9 mg/100g) over 6 months of storage. After observing a 55% reduction in anthocyanins due to blanching frozen chokeberries, standard juice processing was altered by removing the blanch step and its effect on polyphenolics was evaluated. The effect of two different juice processes (with and without blanching) on anthocyanin, proanthocyanidin, flavonol, and hydroxycinnamic acid contents as well as percent polymeric color was evaluated at each stage of processing. Juice pasteurization times and temperatures were also evaluated to develop a statistical model that would predict optimal anthocyanin retention. In comparison of the two processes, there were no significant differences in anthocyanin content after pasteurization; however, samples receiving no blanch had higher levels of anthocyanins after enzyme treatment. Pasteurized juice samples receiving a blanch treatment had 37% and 16% higher levels of total proanthocyanidins and flavonols, respectively than pasteurized juice receiving no blanch treatment. La composition en métabolites des organismes vivants évolue lorsqu’ils sont soumis à des modifications environnementales et diffère d’une espèce à l’autre. L’objectif de cette thèse a été de déployer des stratégies de couplages chromatographiques avec la spectrométrie de masse et de traitement des données MS pour répondre aux différentes problématiques rencontrées en termes de caractérisation, quantification et comparaison du contenu moléculaire de milieux complexes.Ces études ont permis en premier lieu, de développer des méthodes de déréplication par TLC/MS pour identifier rapidement les molécules connues et évaluer la composition des milieux d’intérêt. Afin d’améliorer et de standardiser la comparaison de différents échantillons des traitements d’images des plaques TLC ont été effectués et des études statistiques ont été appliquées. De plus, des couplages de la TLC avec la MS ont été développés pour pouvoir identifier différentes familles moléculaires de faible masse (flavonoïdes, stéroïdes). Dans un second temps, une méthodologie a été mise en place en UHPLC/HRMS pour caractériser la présence de 30 flavonoïdes C et O-glycosylés dans des échantillons de soies de maïs de différentes variétés et aider à la sélection de variétés efficaces pour lutter contre l’infestation de ravageurs. Des approches de représentation visuelle du contenu moléculaire ont été utilisées pour cibler les molécules d’intérêt, puis elles ont été quantifiées par UHPLC/MRM MS. Les différentes empreintes chromatographiques ont été comparées en utilisant des traitements statistiques afin de mettre en évidence les différences entre ces variétés et identifier les molécules permettant cette distinction.

The metabolite composition of living organisms changes when they are subjected to environmental modification and differs from one species to another. The aim of this PhD thesis was to deploy different chromatographic coupling strategies with mass spectrometry and MS data processing in order to respond in the most relevant way to the various problems encountered in terms of characterization, quantification and comparison of the molecular content of complex samples.Firstly, these studies led to the development of TLC/MS dereplication methods to rapidly identify known molecules and to evaluate the composition of samples. In order to improve and standardize the comparison of different samples, image processing of TLC plates were performed and statistical studies were applied. In addition, the hyphenation TLC/MS have been developed to identify different low molecular mass families (flavonoids, steroids). In a second way, a methodology was implemented in UHPLC/HRMS to characterize the presence of C and O-glycosyl flavonoids in corn silk samples of five different varieties and help in the varietal selection to fight against pest infestation. Magistrsko delo prikazuje študijo različnih tehnik izolacije in separacije kanabinoidov iz industrijske konoplje, njihovo dekarboksilacijo ter antioksidativne lastnosti. Kanabinoide iz industrijske konoplje smo izolirali z različnimi ekstrakcijskimi metodami in primerjali rezultate vsebnosti posameznih kanabinoidov, ki so bile določene s pomočjo tekočinske kromatografije (LC-MS). Pričakovano smo večjo vsebnost kislinskih kanabinoidov zabeležili v ekstraktih, ki so bili pridobljeni pri nižjih temperaturah (maceracija), medtem ko so pri višjih temperaturah bili ekstrakti podvrženi procesu dekarboksilacije ter posledično vsebovali več nekislinskih kanabinoidov. Posamezne kanabinoide iz pripravljenih ekstraktov smo ločevali s tankoplastno kromatografijo sklopljeno z masnim spektrometrom (TLC-MS). Preverili smo vpliv pomembnih kromatografskih parametrov (relativna vlažnost, temperatura, mobilna faza) na učinkovitost ločbe. Ugotovili smo, da je ločba posameznih kanabinoidov izjemno zahtevna. Zanimal nas je tudi vpliv temperature, časa in oblike, v kateri se kanabinoidi nahajajo, na proces dekarboksilacije kislinskih kanabinoidov. Opazili smo, da hitrost dekarboksilacije s temperaturo in časom močno naraste. Kanabinoidi v ekstraktih so za popolno dekarboksilacijo kislinskih kanabinoidov potrebovali manj časa kot kanabinoidi v rastlinskem materialu. Antioksidativne lastnosti posameznih ekstraktov in kanabinoidov smo določili s pomočjo DPPH in β-karotenske metode, ter jih primerjali z dvema pogosto uporabljenima antioksidantoma - askorbinsko kislino ter BHT. Izsledki obeh metod so potrdili izjemen antioksidativni potencial kanabinoidov v ekstraktih, ki so pokazali primerljive rezultate kot askorbinska kislina. Posamezni standardi kanabinoidov so antioksidativno manj učinkoviti, kar nakazuje na pomembnost sinergističnega učinka kanabinoidov.

The master's thesis presents a study of various techniques for the isolation and separation of cannabinoids from industrial hemp, their decarboxylation and antioxidant properties. Cannabinoids from industrial hemp were isolated using various extraction methods.The amount of individual cannabinoids was determined by liquid chromatography (LC-MS). As expected, a higher content of acidic cannabinoids was reported in extracts obtained at lower temperatures (maceration), while at higher temperatures the extracts were subjected to a decarboxylation process and consequently contained more non-acidic cannabinoids. Individual cannabinoids from the prepared extracts were separated with thin layer chromatography coupled with mass spectrometry (TLC-MS). The influence of important chromatographic parameters (relative humidity, temperature, mobile phase) on the separation efficiency has been studied. The separation of individual cannabinoids has proven to be extremely challenging. ▼ O própolis é uma substância resinosa, obtida pelas abelhas Apis mellifera, considerado como um produto “antibiótico natural” que desempenha um papel importante na defesa da colmeia, protegendo-a de microrganismos, fungos, bactérias e vírus. Este produto possui na sua composição uma grande variedade de compostos, salientando-se os compostos fenólicos, aos quais se atribuem forte poder antioxidante e atividade antimicrobiana. Neste trabalho usaram-se duas amostras de própolis de origem geográfica diferente: Bornes e Lousã. Estudou-se a composição polínica, os teores totais de compostos fenólicos e flavonoides, bem como, as propriedades antioxidantes na amostra de Bornes. Da análise do extrato de própolis por TLC (cromatografia em camada fina), efetuaram-se estudos da atividade antimicrobiana usando-se extratos de manchas removidas da placa de TLC, selecionadas por terem grande concentração de compostos (manchas mais escuras). Os extratos das manchas separadas na placa de TLC foram analisados por HPLC e MS. A amostra de Lousã foi analisada no seu conteúdo em teores de compostos fenólicos totais e flavonoides totais. A análise polínica da amostra de própolis de Bornes mostrou que nesta predominavam os pólens Erica sp. e Castanea sp. com valores de abundância de 41% e 32% , respetivamente. A análise do perfil químico de amostras de própolis de Bornes e Lousã baseou-se nos teores de compostos fenólicos totais e flavonoides totais. Os teores de compostos fenólicos totais foram determinados pelo método de Folin-Ciocalteu, utilizando como padrão o ácido gálico. Os resultados obtidos foram de 91,5 ± 0,7 mg/L e de 117 ± 2 mg/L para as amostras de Bornes e Lousã, respetivamente. O doseamento de flavonoides totais baseou-se no método espetrofotométrico usando o reagente cloreto de alumínio e como padrão, a quercetina. Os resultados obtidos foram de 23,4 ± 0,8 mg/L e de 32,9 ± 0,1 mg/L para as amostras de Bornes e Lousã, respetivamente. A atividade antioxidante foi avaliada pelo método do poder redutor e pelo efeito bloqueador de radicais livres de DPPH (2,2-difenil-1-picrilhidrazil) para a amostra de própolis de Bornes. Para o método do poder redutor os resultados evidenciaram valores de EC50 de 0,33 ± 0,02 mg/mL, e para o método de DPPH demonstraram valores mais baixos de EC50, 0,072 ± 0,003 mg/mL. O extrato de própolis de Bornes foi analisado por TLC (sílica gel 60 com fluorescência a 254 nm) usando a técnica bidimensional com o objetivo de obter uma primeira separação de compostos para posteriormente serem identificados por HPLC-DAD e MS. A separação dos compostos fenólicos na placa de TLC foi visualizada usando reagentes ou radiação UV 254 nm. Os reagentes usados na visualização das manchas referentes aos compostos fenólicos separados na placa de TLC, foram: cloreto de alumínio, cloreto de ferro, Folin-Ciocalteu, DPPH e vanilina.
