▼ Infecções causadas por tripanossomatídeos causam milhares de mortes anualmente, além de levarem a redução da capacidade produtiva, com elevada morbidade na população acometida. A eficácia terapêutica é limitada na maioria dos casos, sendo o benzonidazol o único fármaco aprovado para uso do tratamento do Trypanosoma cruzi, sendo ativo somente na fase aguda da Doença de Chagas, com grande efeitos colaterais. No caso das leishmanioses, as substâncias de tratamento existentes causam toxicidade renal e cardíaca, além de induzirem a resistência e apresentarem eficácia insuficiente. Observando todo o contexto torna-se necessário a busca por novas substâncias que sejam mais eficazes e menos tóxicas. Assim, o trabalho representa uma contribuição para a busca de novas moléculas bioativas para o tratamento da doença de Chagas e leishmaniose a partir da padronização e realização de ensaios celulares usando a forma epimastigota da cepa Y do T. cruzi e
promastigota para Leishmania chagasi e L. amazonensis. A padronização do método colorimétrico de MTT foi realizada a partir da comparação com o método tradicional de contagem por microscopia usando câmara de Neubauer, seguido do estudo de viabilidade por citometria de fluxo, como método confirmatório e, finalmente a padronização do ensaio de ciclo celular usando os fármacos de referência benzonidazol (T. cruzi) e anfotericina B (Leishmania spp.). Os compostos testados neste estudo são proveniente de classes distintas de inibidores de alvos macromoleculares (cisteíno proteases, DHODH, GAPDH, quinases) e foram planejados e/ou selecionados pelo Grupo de Química Medicinal (NEQUIMED). Dentre todas as substâncias, inibidores de quinases apresentaram maior potencial para estudos subsequentes, sendo o T. cruzi o parasito mais sensível, onde um grande número de substâncias apresentou atividade nos estudos de triagem. Para uma delas (Neq0474) foi realizado o ensaio de dose-resposta, com EC50 =
53 µmol/L. L. chagasi apresentou a maior resistência dentre todos os parasitos estudados neste trabalho, enquanto que L. amazonensis foi sensível para Neq0438. Algumas substâncias estudadas apresentaram potencial para estudos mais aprofundados visando identificar novas alternativas terapêuticas para estas doenças parasitárias.
Infections caused by trypanosomatides lead to thousands of deaths annually, besides the reduction of the quality of life and working capability, with high morbidity to the patients. The therapy efficacy is limited in most cases, being benznidazole the only approved drug in Brazil, which works only in the acute phase of the Chagas Disease with severe collateral effects. For leishmaniasis, the drugs cause renal and cardiac toxicity and trigger resistance. ▼ In today's world, programming teachers perform tedious tasks, which are time consuming; for instance, preparing and marking daily assignments, preparing and marking programming projects, preparing and marking short exams, etc. These tasks distract programming teachers from fulfilling their key role - teaching. Therefore, using automated marking approach with ability to communicate with students is highly desirable. Despite the existing approaches for automated student marking, there is still a need for more improvement. An automated program marking approach is proposed in this study based on a proposal by iMarking®. This approach automates the process of marking and assignments submission and facilitates the communication between teachers and students by designing and implementing a web-based application. In addition, the proposed approach adopts Optical Character Recognition (OCR) to extract the text from images to be evaluated using novel evaluation metrics. The novel evaluation metrics are formulated based on observation and experiment and aim to calculate the matching similarity and mismatching percentage of the submitted student answers when compared with the optimal answers. Evaluation results from a sample of 100 different programming questions show that the proposed approach is efficient in automatically marking the student answers with 100% accuracy. Furthermore, it is found to be time saving - approximately 197 seconds for marking ten questions - which is in line with the objective of creating a more efficient system for teachers.

▼ Immune surveillance requires efficient trafficking of leukocytes throughout the body. To achieve this, leukocytes have evolved to be highly migratory and responsive to environmental cues, which provide guidance for proper tissue distribution. The translation of external environmental cues to intracellular physical changes in leukocytes requires a cascade of receptors, signal transducers, and mechanical effectors. My doctoral research focused on using
T-cells as a model to study the unique cellular process of how signal transducers interact with and regulate mechanical effectors in fast migrating immune cells. Specifically, it is known that the signal transducer Mst1 kinase is required for
T-cell polarization, adhesion, and active migration, but the underlying mechanisms remain poorly understood. I have demonstrated that Mst1 regulates two mechanical effectors, the molecular motor Myosin-IIA and the cytoskeleton regulatory protein
L-plastin, through the process of phosphorylation. Theregulation of Myosin-IIA enables it to generate contractile force inside a migrating
T-cell, maintaining the shape and proper adhesion of the cell to extracellular matrix, both being requirements for successful migration. The regulation of
L-plastin enables it to activate integrin adhesion molecules as well as to properly organize lamellipodial actin. In addition, I have identified novel adhesion structures in
T-cells called microadhesions, which potentially provide traction force to migrating
T-cells. Overall, my research has identified a novel pathway acting between a signal transducer and two mechanical effectors in
T-cell migration.

▼ This research has been undertaken in response to an average 50% attrition rate from a technology enhanced distance-learning course in applied art and design. It studies the participants’ lived experience over a 12-week period from enrolment to the end of the first of twelve modules. The commercial provider recruited 23 mature, female participants from 8 different countries for this City & Guilds certificated course. Data was collected utilizing Interpretive Phenomenological Research methodology (IPA) via two open-ended telephone interviews with participants. The twin roles of the study author as researcher and director of the educational provider were reviewed and a methodology for disaggregation proposed. ATLASti software was used to aid data coding and retrieval. Four key themes emerged from the data; Confidence, Persistence, Support and Timing, which were each subsequently divided into 3 sub categories. Literature searches moved from education to psychology as the study progressed. Implications for further discussion encompass pre-course testing, provider conflict of interest, innovative methods of support and increased legislation. Findings centre around the commencement process and the need for provider instigated advice at the beginning of the course experience, as well as the limitation of design content during the first module and the accuracy of pre-course information and the participant’s engagements and understanding of it. The study also found a lack of correlation between early tutor/student support and retention and several planning activities undertaken by a sub-section of the participants, which appeared to be successful. This study has also contributed to the literature on retention, highlighting the participant’s personal experiences rather than the quantitative retention data more usually associated with retention studies and has demonstrated that interventions intended to decrease course attrition levels may be required at an earlier stage than previously thought and also across a wider subject range than previously recognised.

Nous étudions ce problème dans deux familles degraphes dynamiques, les graphes dynamiques périodiquement variables (PV-graphes)et les graphes dynamiques T-intervalle-connexes. Les résultats obtenus dans cette thèseaméliorent des résultats existants et donnent des bornes optimales sur le problèmeétudié.
In this thesis, we study the complexity of the problem of exploration by a mobileagent in dynamic graphs. A mobile entity (called agent) moving in a dynamic graph hasto traverse/visit each of its vertices at least once. This fundamental problem in computatingby mobile agents has been well-studied in static graphs since the original paper ofClaude Shannon. However, for highly dynamic graphs, only the case of periodic dynamicgraphs has been studied. We study this problem in two families of dynamic graphs,periodically-varying graphs (PV-graphs) and T-interval-connected dynamic graphs. Theobtained results improve the existing results and give optimal bounds on the studiedproblems.

▼ Lymphocytes require antigenic encounter to activate and proliferate, eventually clearing the source of antigenic challenge. The peripheral lymph nodes (PLN) are the primary sites of antigenic encounter and thus the ability of lymphocytes to migrate to this tissue is a requirement for mounting effective immune responses. The process of lymphocyte migration to the PLN is known as the "adhesion cascade". Specifically, lymphocytes are captured from the blood through the adhesion molecule,
L-selectin, followed by CC chemokine receptor 7 (CCR7)-mediated integrin activation, which ultimately results in cell transmigration into the PLN. Because the PLN is the site where antigenic encounter is most likely, we hypothesized that migration may induce pro-proliferative signaling, mediated by
L-selectin and conducted through CCR7. To study the potential "priming" effects of adhesion to antigen-mediated lymphocyte proliferation, spleen and PLN lymphocytes were subjected to in vitro transmigration and proliferation assays. Subsequently, multi-color flow cytometry analysis was used to examine treatment effects on the
T cell population. Results showed that lymphocyte proliferation was significantly increased when
L-selectin and CCR7 were activated simultaneously, compared to CCR7 activation alone.
Specifically, bulk tissue populations, as well as the CD4+ and CD8+
T cell subsets showed enhanced proliferation from both spleen and PLN sources at early and late time points. Additionally, to discern whether this enhanced proliferation may be a by-product of decreased apoptosis, an apoptosis assay was also performed at the same time points. Interestingly,
L-selectin engagement resulted in protection from apoptosis for all
T cell populations 72 hour following activation. However, this protective effect was not observed at the 36 hour time point. Taken together, these results suggest a role for
L-selectin in enhancing
T cell proliferation, at least in part, through an anti-apoptotic effect. This research will add to a better understanding of the regulation of lymphocyte proliferation and increase our knowledge of
L-selectin signaling function.

This study is a critical assessment and analysis of Xhosa poetry that addresses social and political concerns with particular reference to the poetry of Qangule (1970), Mtuze (1982, 1990) and Xozwa (1994 ). Chapter one introduces the study by presenting its aim, scope, method of research, theory, definition of concepts and significance of the study. Chapter 2 focuses on the theoretical framework of the study. It discusses the theory of Marxism, which is used as a basis of this study, by explaining the concept of Marxism, its historical background, assumptions, methodology and Marxism and poetry. Chapter 3 deals with social concerns that are addressed by Mtuze (1982 and 1990) and Xozwa (1994 ). These social concerns are divorce and alcohol usage. The discussion in this chapter focuses on how divorce affects members of the family concerned, especially children of divorced parents. It also looks at how alcohol is used in society. While the discussion in this chapter acknowledges moderate use of alcohol, it focuses more on how this substance may also be abused. It looks at how alcohol abuse affects individuals and society economically and how dangerous it is to the health and lives of people. Chapter 4 focuses on political concerns as addressed by Qangule (1970) and Mtuze (1982). It assesses and analyses poems that address the oppression of black people during the apartheid regime in South Africa. Concerns like citizenship and deprivation of freedom, labour conditions and language are discussed in this chapter. Chapter 5 is a concluding statement which gives a summary of the study, evaluation of Xhosa poetry with regard to addressing the concerns discussed in the study and recommendations regarding future research.

▼ The aim of this project is to determine whether a computer application can be used to develop phonemic awareness in the early primary classroom, which is a key component of phonics. This thesis explores the evolution of the strategy for teaching literacy in the UK which shows phonics to be a key component of that strategy. However, government reports which inform the direction of the literacy strategy call for more empirical study in all areas of literacy teaching; this thesis documents such an empirical study. This research project creates a phonics based computer application designed specifically for young children aged 5 to 6 years (year 1 in UK primary schools). The timing and level of content presented by the computer application activities are grounded in appropriate academic theory. A significant component of the work is the development of interface design guidelines for children’s applications. These guidelines are then used to inform the development of the phonics-based computer application. A Randomised Controlled Trial (RCT) is designed to determine the application’s effectiveness in developing the phonemic awareness skills of young children in a classroom setting. In order to control experimental bias resulting from problems with the usability of the computer interface, the usability of the application’s interface is evaluated in the classroom by year 1 children before the application is used in a pragmatic RCT. The results of the final usability evaluation found no usability issues and the application was wholly intuitive to the children in the evaluation groups. The results from the RCT (N=266) show no statistically significant improvement in the learning rate of phonemic awareness by the intervention group using the computer program compared to the traditional teacher-delivered paper-based method used with the control group, even though the computer program was designed carefully for this age range. The results did suggest however, that the intervention group developed at the same rate as the control group which implies that the computer program could be used to support teachers by reducing the amount of resource-intensive phonics tuition required by children in this age range.

▼ Argument diagramming can scaffold the process of argumentation but only a few studies have investigated its impact on the quality of argumentative writing. This research contributed to this direction with two studies.
An exploratory study investigated the impact of argument diagramming, applied as a paper-based or a computer-based method, on the quality of argumentative text. The computer method increased the refutations and overall quality of essays. The study highlights the significance of writers’ argumentative ability for interpreting improvement.
A qualitative study looked into the impact of argument diagramming on the process of writing cognition through analysis of online process data, diagrams and essays of sixteen undergraduate students. Writers with myside bias schema used the method to increase counterarguments and refutations. Writers at lower level of pseudo-integration adopted more advanced strategies like weighing, and writers at middle level of pseudo-integration formed positions with qualifications. Needs at higher levels of argumentative ability are not met. The support of writing planning processes through argument diagramming affects mainly the semantic aspects of the text while the support of linearization processes affects mainly the rhetorical aspects.
The analysis of interviews revealed that interacting with argument diagramming can improve awareness of argumentation schema, hence, a writer can progress from unaware, to aware-and-lost and aware-but-oriented. Improvement is signified as being sensitised to limitations, gaining knowledge of writing processes and the ability to self-regulate.

▼ Low-level classroom disruption (LLCD) is the fundamental behavioural issue in primary schools across England. Typically defined as surface-level behaviours (Esturgó-Deu & Sala-Roca, 2010), LLCD includes talking unnecessarily, fidgeting, distracting others, rocking on the chair and daydreaming (Ofsted, 2014). Educational literature has extensively referenced LLCD, making inferences about the potential antecedents, from within the classroom to the wider contexts (home and societal factors). However, and contradicting this, LLCD is viewed as a concept controllable by effective teachers at classroom level. Thus, research is typically classroom based, and centred round the management and control of LLCD. To date no psychological research has investigated the bio-ecological antecedents of LLCD. This mixed methods study pioneers this line of enquiry. By applying the Person, Process, Context Time Model of Development (Bronfenbrenner, 1985) processes that influence behaviour were considered. Key Stage Two pupils aged 8-11 years (N=274) from 3 schools in England, provided quantitative data at two time points (with a year lag between) recording: gender, peer pressure, executive function, global self-worth, appropriate conduct, home chaos, screen time, sleep, television in bedroom, and extra-curricular activity. A sub-sample of these pupils' parents (N=58) reported on their own personal screen time use, parenting practices and the family's socioeconomic status. Semi-structured interviews with members of teaching staff (N=8) provided an in-depth account of the lived experience of LLCD in the classroom providing evidence of the impact LLCD on staff and pupils. Results show a significant increase to the presentation of LLCD across the two time points for the whole pupil sample, with male pupils displaying significantly higher levels of LLCD than the female pupils at both times. Findings also indicated at both time points that higher screen time use in the home context was directly associated with increased LLCD in the school context for the whole pupil sample. For the male pupils only this association was partially mediated through increase in proneness to boredom. The repeated measures investigations found the relationship between screen time and LLCD to then be converse, with increases in LLCD significantly related to higher screen time for the male pupils, suggesting a cyclical reciprocal pattern of influence. Repeated measures analysis also suggested converse gender differences between the pupils' self-perceived appropriate conduct and LLCD. For the male pupils a significant result was found indicating that a lower self-perception of appropriate conduct was associated with a higher presentation of LLCD whereas, for the female pupils a higher perception of their own appropriate conduct was associated with a lower presentation of LLCD. The semi-structured interviews with teaching staff (N=8) supported the Ofsted (2014) report of LLCD having a negative impact on both the teaching and learning that takes place in the classroom.…

▼ Argument diagramming can scaffold the process of argumentation but only a few studies have investigated its impact on the quality of argumentative writing. This research contributed to this direction with two studies. An exploratory study investigated the impact of argument diagramming, applied as a paper-based or a computer-based method, on the quality of argumentative text. The computer method increased the refutations and overall quality of essays. The study highlights the significance of writers’ argumentative ability for interpreting improvement. A qualitative study looked into the impact of argument diagramming on the process of writing cognition through analysis of online process data, diagrams and essays of sixteen undergraduate students. Writers with myside bias schema used the method to increase counterarguments and refutations. Writers at lower level of pseudo-integration adopted more advanced strategies like weighing, and writers at middle level of pseudo-integration formed positions with qualifications. Needs at higher levels of argumentative ability are not met. The support of writing planning processes through argument diagramming affects mainly the semantic aspects of the text while the support of linearization processes affects mainly the rhetorical aspects. The analysis of interviews revealed that interacting with argument diagramming can improve awareness of argumentation schema, hence, a writer can progress from unaware, to aware-and-lost and aware-but-oriented. Improvement is signified as being sensitised to limitations, gaining knowledge of writing processes and the ability to self-regulate.

▼ Insecticide resistance is a global problem. Insecticide resistance management is very important, considering the time, effort, and cost of discovering and developing a new insecticide. There are diverse resistance mechanisms, but enhanced detoxification through overexpression of cytochrome P450s and target site insensitivity through mutation in insecticide binding site are the two most common mechanisms. The xenobiotic detoxification is divided into three successive phases (I, II and III), which ensures the metabolism and excretion of the detrimental toxins. Each phase comprises of a specific group of metabolizing enzymes such as P450s (phase I), GSTs (phase II) and ABC transporters (phase III). The major goal of my research was to understand the molecular mechanism of insecticide resistance in two economically important coleopteran pests, Leptinotarsa decemlineata and Tribolium castaneum. The transcriptional regulation of the P450 genes mediating insecticide resistance in L. decemlineata (imidacloprid-resistant) and T. castaneum (deltamethrin-resistant) were studied and the xenobiotic trans and cis-elements identified. RNA interference (RNAi), and reporter assays revealed that the cytochrome P450 genes involved in insecticide resistance are regulated by transcription factor Cap n Collar ‘CncC’ and muscle aponeurosis fibromatosis ‘Maf’ belonging to the b-ZIP transcription factor family. Site-directed mutagenesis was employed to identify the binding site for CncC and Maf. Sequencing of RNA isolated from CncC knockdown T. castaneum identified genes regulated by CncC and involved in insecticide detoxification. RNAi and insecticide bioassays confirmed the function of select phase II (glutathione-S-transferases) and phase III (ABC transporters) identified by RNA sequencing. Overall, these data revealed that the xenobiotic transcription factor CncC is the master regulator of multiple genes that are involved in insecticide resistance.

▼ ZnT-1 expression in the pre-implantation mouse embryo and its effect on calcium influx Pre-implantation embryos develop into 9 stages over the first 5 days post-fertilisation. Calcium influx from the external environment via calcium channels, including the L-type and T-type calcium channels, is critical for embryonic gene activation and cell proliferation. In cardiomyocytes these channels are regulated by the ubiquitously expressed zinc-transporter protein ZnT-1. When plasma membrane bound, ZnT-1 facilitates zinc-efflux. Free cellular zinc regulates ZnT-1 expression, with an increase in zinc inducing transcription. In this study, ZnT-1 mRNA and protein expression were investigated in pre-implantation embryo stages using qPCR and immunofluorescence. Embryos were cultured in vitro in zinc-supplemented media and compared to embryos cultured in the absence of zinc and to in vivo developed embryos. ZnT-1 mRNA was expressed at all stages and the presence of zinc increased mRNA expression a the late 2-cell stage only. There was no difference in expression between in vivo developed and cultured embryos. ZnT-1 protein was expressed from the early 2-cell stage onwards; not affected by zinc culture and localized to the plasma membrane at the late 2-cell stage only. Calcium imaging was performed to examine whether ZnT-1 membrane localization altered calcium influx. Experiments on early and late 2-cell embryos showed that there was no difference in calcium influx when ZnT-1 was localized to the plasma membrane. In summary ZnT-1 transcription was induced by zinc at the late 2-cell stage. Protein expression was not affected by zinc culture but was developmentally regulated, localizing to the plasma-membrane at the late 2-cell stage without effect on calcium influx.

▼ Grâce au partenariat Tradition and Transition among
Labrador Inuit unissant le Gouvernement Nunatsiavut et l’Université
Mémorial de Terre-Neuve, la fouille exhaustive du site Double Mer
Point (GbBo-02) a pu être effectuée. Elle nous a permis d’étudier
l’impact des interactions culturelles au Labrador sur la vie
quotidienne des occupants de l’Habitation 1 durant la période de
contact, soit entre XVIIe et le XIXe siècle; un sujet d’étude
encore sous-exploité par l’archéologie. Des idées théoriques tirées
des Cultural Contact Studies et de l’archéologie postcoloniale ont
ainsi pu être appliquées au cadre contextuel du Labrador par la
réalisation de ce mémoire. Notre réflexion théorique portant sur
l’étude des assemblages de la période de contact nous a mené à
développer une méthodologie de recherche combinant l’aspect
culturel d’un objet et son mode de fabrication. L’Habitation 1, une
maison semi-souterraine en tourbe datée entre la fin du XVIIIe
siècle et le début du XIXe siècle, a pu être interprétée à partir
des données (matérielles, zooarchéologiques, architecturales et
environnementales) collectées lors des fouilles en 2015. La
datation du site, les phases d’occupation et les aires d’activités
ont été déterminées grâce à diverses analyses. L’Habitation 1 a
dévoilé une occupation plus complexe que la plupart des maisons
semi-souterraines de cette période. Elle a témoigné de plusieurs
phases d’occupation dont la première serait dorsétienne, tandis que
la dernière démontre l’intégration d’un Européen à une communauté
inuite. L’application de la méthodologie développée a quant à elle
permis d’identifier des transferts culturels directs, d’observer
l’intensification de traits culturels intrinsèques, d’étudier
l’influence d’une nouvelle matière sur le quotidien d’une
communauté et d’évaluer l’impact de ces changements à long terme.
Ce mémoire participe à la reconstitution de l’histoire culturelle
du Labrador et espère encourager la réalisation de recherches
portant sur les interactions culturelles et l’étude de la culture
matérielle de la période de contact.
Through the Tradition and Transition among Labrador
Inuit partnership between the Nunatsiavut Government and the
Memorial University of Newfoundland, the extensive archaeological
excavation undertaken at Double Mer Point (GbBo-02) allowed us to
study the impact of cultural interactions in Labrador on the daily
life of the inhabitants of House 1 during the contact period,
between the 17th and 19th centuries, a subject still
under-exploited by archeology. Theoretical ideas from cultural
contact studies and postcolonial archeology have thus been applied
to Labrador through this research. Our theoretical reflection about
the study of material culture from the contact period led to the
development of a methodology combining the cultural aspects of
objects and their manufacture. House 1, a semi-subterranean sod
house dated from the late 18th and early 19th century, was

▼ The annual and seasonal water use efficiency of six pasture combinations were calculated from the ‘MaxClover’ grazing experiment at Lincoln University, Canterbury. Pastures had been established for seven years and were grazed by best mangement practises for each species combination. Measurements from this study are from individual plots of six replicates of cocksfoot (CF)/Subterranean clover (Sub), CF/Balansa clover (Bal), CF/White clover (Wc), CF/Caucasian clover (Cc), ryegrass (RG)/Wc and lucerne (Luc).
Dry matter measurements of yield, botanical composition and herbage quality were assessed from 1 July 2009 to 30 June 2010. Lucerne had the highest total annual yield of 12880 kg DM/ha/y, followed by CF/Sub at 9460 kg DM/ha/y. All other pastures produced 6600 to 8360 kg DM/ha/y. Soil water measurements showed that all pastures had a plant available water content of 316 ± 24 mm. All pastures used 655 ± 22 mm/y of water. Lucerne had the highest annual water use efficiency (WUE) of 23 kg DM/ha/mm/y. The annual WUE of CF/Sub was 16 kg DM/ha/y and the lowest was 11 kg DM/ha/mm/y from RG/Wc and CF/Wc pastures.
The CF/Sub and CF/Bal pastures had the highest total annual legume (sown and volunteer white clover) content of all grass based pastures at 18%. Because of yield differences CF/Sub had the highest annual nitrogen yield of 216 kg N/ha of the grass based pastures although it was less than half that from lucerne (462 kg N/ha). RG/Wc pastures had the highest proportion of weeds which represented 62% of total dry matter in Year 8 (2009/10).
For dryland farmers spring is the crucial period for production before growth slows when soil water becomes limiting. For the spring period (1/7/2009 to 29/1/2010) lucerne produced 11120 kg DM/ha, which was the highest yield of all pastures follwed by CF/Sub (1/7/2009 to 16/11/2009) which produced 6020 kg DM/ha. When dry matter production was regressed against thermal time, CF/Sub pastures grew at 5.7 ± 0.08 kg DM/° Cd between 1/7/2009 and 16/11/2009 compared with lucerne which grew at 4.2 ± 0.46 kg DM/° Cd between 1/7/2009 and 29/1/2010 which was an extra 74 days of linear spring growth. WUE during the initial period for lucerne (1/7/09 to 29/1/10) was 22.3 ± 0.5 kg DM/ha/mm (R2=1.00). The CF/Sub pastures produced 20 ± 1.5 kg DM/ha/mm (R2=0.98) from 1/7/09 to 16/11/09 from 306 mm of water used. The lowest WUE was 11.7 ± 0.21 kg DM/ha/mm by CF/Wc pastures.
Insect pests within CF/Sub, CF/Wc, RG/Wc and lucerne pastures were quantified in winter. Pests included Argentine stem weevil, clover root weevil, Sitona weevil and grass grub. Argentine stem weevil populations were highest in CF/Sub pastures (63 weevils m⁻²) whereas clover root weevil populations were highest in CF/Wc (5 weevils m⁻²). Grass grub was present in all grass based pastures and was the highest in RG/Wc with 208 grubs m⁻². Present pest levels were not at damaging thresholds.

▼ The lungs are constantly exposed to particulate matter in the external environment, and thus are vulnerable to exposure against microorganisms transmissible via inhalation. As a result, pulmonary diseases such as pneumonia are an important source of morbidity and mortality worldwide. Legionella pneumophila is a Gram negative bacillus and one of the leading causes of community acquired pneumonia. L. pneumophila is an accidental pathogen to humans where upon inhalation, the bacteria establish a replicative niche within alveolar macrophages of the lung. This triggers a robust host immune response that operates to eradicate the bacteria. However, despite the importance of an inflammatory response being important for L. pneumophila clearance from the lung, we do not fully understand the immune mechanisms operating between the onset of infection and bacterial clearance. This Thesis aims to investigate the molecular and cellular pathways in the lung in response to L. pneumophila infection, and define a network of cellular behavior cooperating to efficiently clear bacteria from the lung.
Chapter 3 investigates the role of plasmacytoid DC (pDC) in response to L. pneumophila infection. It is shown that pDC upregulate their expression of CD8 that is associated with cellular activation. However, despite the cells trafficking to the lung and displaying an activated phenotype, the role of pDCs was not important for the overall clearance mechanisms of L. pneumophila from the lung.
To understand the key cell types interacting with L. pneumophila during infection, a broad appraisal of the key phagocytic cell types in the lung during infection was made. Chapter 4 outlines that alveolar macrophages, despite being a replicative niche for L. pneumophila, were rapidly depleted in the lung after infection. This occurred alongside a strong infiltration of neutrophils and monocyte-derived cells (MCs) that actively engulfed bacteria for intracellular degradation.
Previous work has shown that the IL12-INFg axis is important for pulmonary clearance of L. pneumophila. Despite this, the key cell types contributing to their production, as well as a mechanism for how these cytokines function is not well understood. Chapter 5 shows that monocytic and dendritic cells are the sole contributors of IL12-related cytokines in the lung during infection. MCs were a significant contributor of IL12 that was important for acute L. pneumophila clearance. As demonstrated in Chapter 6, IL12, in turn, stimulated the production of IFNg by lung-infiltrating NK cells, as well as memory T cells in a non-cognate manner. Lymphoid cell-derived IFNg specifically stimulated the intracellular bactericidal activity of MCs to assist with clearance of L. pneumophila from the lung.

le nord du Québec et du Labrador connait présentement un déclin
rapide. Cependant, la démographie, l’utilisation de l’espace et la
sélection de l’habitat en relation avec le déclin de cette
population sont méconnues. À l’aide de 35 caribous adultes munis de
colliers satellites, nous avons évalué la sélection des habitats à
deux échelles spatiales, la survie, et l’effet de la chasse ainsi
que du chevauchement avec la population voisine de caribous
migrateurs de la Rivière-George. Nous avons démontré que la
sélection de l’habitat est similaire entre les deux échelles
spatiales étudiées, mais variable selon les saisons. Nos résultats
suggèrent que la sélection est déterminée par l’accessibilité et la
disponibilité des ressources alimentaires en hiver et durant la pré
mise-bas, l’évitement des insectes en été, et par la présence de
plantes graminoïdes lors du rut. Bien que la prédation soit connue
pour être le principal facteur limitant les populations et la
sélection chez le caribou lors de la mise-bas, aucun comportement
anti-prédateur n’a été détecté chez les femelles lors de cette
saison. Nos résultats démontrent que le déclin est principalement
attribuable à une faible survie des femelles adultes due à la
chasse et à la prédation. Nous avons observé que l’utilisation de
l’aire de distribution du caribou des monts Torngat par le troupeau
Rivière-George a grandement diminué durant les 25 dernières années
en raison de leur déclin. Ceci pourrait causer une diminution du
flux génique et des possibilités d’immigration entre les deux
troupeaux. Nos modèles démographiques suggèrent que le caribou des
monts Torngat continuerait de décliner rapidement si aucune mesure
de gestion n’était prise de façon à augmenter la survie des
adultes.
The Torngat Mountains caribou herd in northeastern
Québec-Labrador is rapidly declining. Few studies have examined the
causes of this decline and little is known about the habitat
selection, survival and space use of this herd. Using 35 caribou
equipped with satellite collars between 2011 and 2015, we evaluated
the Torngat Mountains caribou habitat selection at two different
spatial and temporal scales, survival, and effect of harvest on
future population trends. We also evaluated the Torngat Mountains
caribou space use in relationship with its neighbor, the
Rivière-George migratory caribou. We found little variation in
habitat selection between the two spatial scales studied. However,
there was variation in factors limiting habitat selection among
different seasons. Habitat selection was primarily driven by
resource availability and abundance during winter and pre-calving,
by insect avoidance in summer and by the presence of graminoids
during the rut. Although predation is known to be the main limiting
factor of caribou populations and habitat selection by female
caribou during calving, we found no evidence of predator avoidance

T (Treg) cells mediate tumor immune evasion by suppressing anti-tumor effector
T cell responses in peripheral lymphoid tissues and within the tumor. While elevated Treg cell numbers have been shown to correlate with increased tumor growth, mechanisms that regulate their distribution within secondary lymphoid tissue and tumor tissue are not well understood.
L-selectin, an adhesion molecule constitutively expressed on all classes of leukocytes, functions early in the adhesion cascade and regulates the migration of lymphocytes to lymph nodes through high endothelial venules. In addition,
L-selectin can mediate migration of lymphocytes to sites of inflammation by binding to ligands present on inflamed endothelium. Treg cells express high levels of
L-selectin and require
L-selectin for entry into resting lymph nodes. However, the role of
L-selectin in regulating Treg cell distribution and migration into tumors and lymph nodes during chronic inflammation, such as cancer, has not been examined. Therefore, we investigated the role of
L-selectin in regulating Treg cell rolling and adhesion to an endothelial cell monolayer in vitro as well as the distribution and migratory patterns of Treg cells using the murine 4T1 breast cancer model. Importantly, ɑ
4ɑ
7 integrin/VCAM-1 interactions were found to significantly contribute to Treg cell adhesion to endothelial cell monolayers under shear stress only in the presence of
L-selectin function. In vivo, Treg cell populations preferentially accumulated in tumors and tumor-draining lymph nodes during progression of disease, and increased at higher rates than conventional CD4
+ T cell populations as tumors progressed. Furthermore, Treg cells preferentially migrated to tumors and tumor-draining lymph nodes in a
L-selectin-dependent manner, thereby promoting immune suppressive environments. These studies provide further insight into the mechanisms of
L-selectin function in regulating Treg cell distribution during chronic inflammation such as cancer, and may lead to a better understanding of tumor immune evasion and provide new targets for immunotherapeutic strategies.

▼ This is the first study in the field of Information and Communication Technology (ICT) integration into Early Childhood Education (ECE) to call upon a blended theoretical framework of Bronfenbrenner’s ecological systems theory, Fullan’s concepts of educational change and complexity theory. In drawing the collected data together within the framework of Bronfenbrenner’s ecological systems theory, a range of linear factors that influence practitioners’ use of ICT in the playroom at the micro-level (teachers’ pedagogical beliefs, confidence, technological pedagogical knowledge); meso-level (local school policy, leadership, support) and macro-level (national ECE curriculum and national ICT policy) were identified. Currently, structured research into ICT integration in ECE is extremely limited in Saudi Arabia. This thesis addresses this substantial knowledge gap in the practice of ICT integration in Saudi Arabian ECE settings through a collective case study approach of Saudi Arabian programs. According to the literature, Scotland, for some time, has been at the forefront of developing strategies for the integration of ICT into early years. Policy in Scotland has also been supported by a range of literature, studies and reference to ICT use in the curriculum; that have assisted practitioners in making important pedagogical decisions for using ICT in the playroom. For these reasons, Scotland is included in this research as an example that can provide some insights for improvement in the Saudi Arabian context. Six case studies were used to address the study’s research questions: four in Jeddah city, Saudi Arabia and two in Scotland (located between Glasgow City Council and East Dunbartonshire). As part of the research’s endorsement of a holistic approach, the researcher triangulated multiple research methods (questionnaire, semi-structured interview, playroom observation and documentary analysis) to investigate the status of ICT use in preschool settings and factors that influenced teachers’ ICT practices. The target community was made up of practitioners in ECE settings, including head teachers and practitioners from both private and public preschools. Research findings suggest that practitioners in both locations hold a positive perspective of the importance of ICT integration into ECE. However, enthusiasm and positive attitudes do not always lead to high levels of ICT integration. In Saudi Arabia in particular, much of the integration is achieved in an informational, teacher-centred/traditionalist manner, rather than encouraging child-centred, constructivist approaches. The results revealed that teachers’ pedagogical beliefs and their relationship to teaching practices strongly influenced integration practices. Furthermore, school characteristics were equally, if not more, influential upon integration levels. ▼ Despite the enormous body of literature studying the typing errors of adults, children's typing errors remain an understudied area. It is well known in the field of Child-Computer Interaction that children are not 'little adults'. This means findings regarding how adults make typing mistakes cannot simply be transferred into how children make typing errors, without first understanding the differences. To understand how children differ from adults in the way they make typing mistakes, typing data were gathered from both children and adults. It was important that the data collected from the contrasting participant groups were comparable. Various methods of collecting typing data from adults were reviewed for suitability with children. Several issues were identified that could create a bias towards the adults. To resolve these issues, new tools and methods were designed, such as a new phrase set, a new data collector and new computer experience questionnaires. Additionally, there was a lack of an analysis method of typing data suitable for use with both children and adults. A new categorisation method was defined based on typing errors made by both children and adults. This categorisation method was then adapted into a Java program, which dramatically reduced the time required to carry out typing categorisation. Finally, in a large study, typing data collected from 231 primary school children, aged between 7 and 10 years, and 229 undergraduate computing students were analysed. Grouping the typing errors according to the context in which they occurred allowed for a much more detailed analysis than was possible with error rates. The analysis showed children have a set of errors they made frequently that adults rarely made. These errors that are specific to children suggest that differences exist between the ways the two groups make typing errors. This finding means that children's typing errors should be studied in their own right.

▼ This is the first study in the field of Information and Communication Technology (ICT) integration into Early Childhood Education (ECE) to call upon a blended theoretical framework of Bronfenbrenner’s ecological systems theory, Fullan’s concepts of educational change and complexity theory. In drawing the collected data together within the framework of Bronfenbrenner’s ecological systems theory, a range of linear factors that influence practitioners’ use of ICT in the playroom at the micro-level (teachers’ pedagogical beliefs, confidence, technological pedagogical knowledge); meso-level (local school policy, leadership, support) and macro-level (national ECE curriculum and national ICT policy) were identified.
Currently, structured research into ICT integration in ECE is extremely limited in Saudi Arabia. This thesis addresses this substantial knowledge gap in the practice of ICT integration in Saudi Arabian ECE settings through a collective case study approach of Saudi Arabian programs. According to the literature, Scotland, for some time, has been at the forefront of developing strategies for the integration of ICT into early years. Policy in Scotland has also been supported by a range of literature, studies and reference to ICT use in the curriculum; that have assisted practitioners in making important pedagogical decisions for using ICT in the playroom. For these reasons, Scotland is included in this research as an example that can provide some insights for improvement in the Saudi Arabian context.
Six case studies were used to address the study’s research questions: four in Jeddah city, Saudi Arabia and two in Scotland (located between Glasgow City Council and East Dunbartonshire). As part of the research’s endorsement of a holistic approach, the researcher triangulated multiple research methods (questionnaire, semi-structured interview, playroom observation and documentary analysis) to investigate the status of ICT use in preschool settings and factors that influenced teachers’ ICT practices. The target community was made up of practitioners in ECE settings, including head teachers and practitioners from both private and public preschools.
Research findings suggest that practitioners in both locations hold a positive perspective of the importance of ICT integration into ECE. However, enthusiasm and positive attitudes do not always lead to high levels of ICT integration. In Saudi Arabia in particular, much of the integration is achieved in an informational, teacher-centred/traditionalist manner, rather than encouraging child-centred, constructivist approaches. The results revealed that teachers’ pedagogical beliefs and their relationship to teaching practices strongly influenced integration practices. Furthermore, school characteristics were equally, if not more, influential upon integration levels. In general, in the Saudi ECE sector, the journey to ICT integration is at an initial, unstructured stage and observed attempts at integration are largely the result of practitioners’ individual efforts.

▼ This thesis addresses the issue as to how to model a student’s motivation when using a learning environment. Currently, this is considered as an important topic since a) motivation is regarded as key to successful learning by educationalists, b) those building computer-based learning environments are attempting to design in features that will assist in motivating learners while using their systems, c) researchers in the areas of intelligent learning environments (ILEs) and intelligent tutoring systems (ITSs) are looking for methods to detect a student’s motivation during their interaction with an adaptive system, and d) researchers in ITSs are looking for strategies that a learning environment could use to motivate a student or to scaffold his/her motivation. A well-founded model that partially explains the way in which motivation changes during an interaction with an ILE is potentially very valuable to these different goals. In particular, the model should prove to be of great benefit when creating learning environments that take into account the motivational aspects of the learners since it represents the process of how they are motivated through the use of an ILE. In this thesis the focus is on the construction of a learner’s motivational structure for an ILE grounded in the context of an educational game. The thesis reports the development of a qualitative model of the motivation of learners during their interaction with the ILE. One of the main issues is the specification of the context as the motivational structure of learners cannot be assumed to be the same in different contexts. A preliminary causal model showing the relationships between a learner’s motivational characteristics and features of the ILE was developed. Two computer-based research instruments were then built: a game prototype aiming to teach some concepts in databases (Alex’s Adventure) and a computer program (MoRes). Alex’s Adventure was developed in such a way that it included all features of the ILE presented in the model whereas the other parts of the model were implemented through MoRes. Six detailed case studies were performed in order to validate the preliminary model. The validation resulted in changes to the preliminary model which are believed to produce an improved model. The analysis also resulted in a number of key points and condition–action rules which are considered to be of use for the implementation of future adaptive computer systems that could manage and support learners. The thesis makes an original contribution to the Artificial Intelligence in Education (AIED) community, particularly in terms of the method of case study analysis and the new model of motivation.

▼ The Institution of Engineers Malaysia (IEM) and the Board of Engineers Malaysia (BEM) stress the need for Malaysian engineering graduates to be able to integrate sustainable development knowledge, skills and values in their professional practice. The 2012 Engineering Accreditation Council (EAC) manual outlines 12 outcomes that students of Malaysian institutions of higher learning offering engineering programmes are expected to develop upon completion of their studies. Of the 12 outcomes, three are explicitly linked to sustainable development. While institutions of higher learning are required to develop the prescribed skill set using outcome based approaches to learning, integration measures are not specifically stipulated. 30 hypothetical competences were developed as a means of addressing the issue of integrating sustainable development outcomes within the undergraduate engineering programme curriculum. Using a private higher learning institution offering engineering programmes as a case study, this study set out to explore the views of the institution’s stakeholders, i.e. the final year undergraduate engineering students, academicians, university management and engineers, as well as education for sustainable development experts and ESD practitioners on (a) the extent to which sustainable development and education for sustainable development is incorporated within the engineering curriculum of the higher learning institution (b) the inclusion of sustainable development programme and module learning outcomes within the undergraduate engineering curriculum and how it can be included within the curriculum, and (c) the additional components needed for the EESD framework for undergraduate engineering education in Malaysia. Respondents’ perspectives were sought through a triangulation mixed methods approach, through surveys, interviews and analysis of documents. Findings of the study were then used to develop the proposed (a) guidelines to incorporate sustainable development competences holistically within undergraduate engineering programme outcomes and common module learning outcomes, and the (b) Whole Institution EESD Framework.

▼ This dissertation presents a thesis in the form of three analyses to support creative decision making in the design of learner-centred, technology-enhanced education. The thesis was developed through use and improvement in practice over three decades. The dissertation also describes the action research methodological approach & methods employed and explores a framework based on concepts of design, learner, technology as a foundation for framing the thesis. Finally it evidences the claim for an original contribution to knowledge through an annotated selection from a portfolio of practice. The experience gained through increasingly responsible work rôles, in which the author was expected to guide other designers, shaped the articulation of the three analyses which were made for evaluating designs as a springboard for iterative improvement. These analyses are: • an 'expressive constructivist' model of learning; • an account of how technology can support such learning; • a learner-centred breakdown of questions to focus on progress in education more holistically. This thesis arose from growing tacit and experiential understanding and has been informed by a theoretical and conceptual framework based on academic literature. It is argued that the successful application of these analyses in the practice cited has generated the validity of the thesis, and to a limited extent, reliability.

▼ L’objectif de ce projet de recherche est de
développer un modèle d’évaporation de l’eau d’une rivière en milieu
boréal. Un site expérimental sur la rivière Natashquan dans la
région de la Côte-Nord au Québec a été sélectionné pour y installer
une station hydrométéorologique faisant un suivi des variables
environnementales locales. Cette rivière est marquante dans le
paysage de la Minganie par la superficie de 15 930 km2 de son
bassin versant et son débit moyen annuel de 349 m3/s. Ce puissant
cours d’eau est une des dernières rivières naturelles de la
Côte-Nord n’étant pas harnachée par un barrage. Une campagne
constituée de 5 sorties de mesures intensives sur le terrain a été
effectuée afin d’observer l’évaporation de l’eau de la rivière par
la méthode des mini-lysimètres flottants. Ces derniers s’inspirent
du processus derrière le bac évaporatoire de classe A pour obtenir
une estimation de l’évaporation in-situ en ayant un petit bac
évaporatoire portatif à la surface de l’eau de la rivière. Un
modèle d’évaporation par transfert de masse a été développé avec
les données récoltées et 8 fonctions de vent de la littérature ont
été testées. Puis, un modèle d’évaporation par ensemble de réseaux
de neurones a été retenu, car il offrait une meilleure performance
que les approches classiques. Ce réseau de neurones ayant comme
variables d’entrée la tension de vapeur à saturation, le
rayonnement incident d’ondes longues, la vitesse du vent et la
tension de vapeur de l’air, permet d’explorer une non linéarité
entre les variables qui n’est pas accessible à l’approche de
transfert de masse. Ce modèle met en évidence le manque de
recherche sur les processus gouvernant l’évaporation fluviale dans
le cadre de rivière de grande envergure et particulièrement
l’évaporation des rivières du milieu boréal.
The objective of this study was to develop a river
evaporation model for the boreal biome. A hydrometeorological
station was installed on the bank of the Natashquan river to
collect environmental data. The Natashquan river is major
hydrographic component of the region with its 15 930 km2 watershed
and its 349 m3/s mean annual flow. This powerful watercourse is one
of the only remaining significant river that has not yet been
dammed in the Côte-Nord region. Five intensive field campaigns were
carried out targeting different conditions during the summers of
2018 and 2019. River water evaporation was observed using the
floating minipan method. This method, inspired by the process
behind the class A evaporation pan, allows small portable
evaporation pans to be deployed on the water surface and to monitor
in-situ evaporation in more holistic conditions. Observations were
used to calibrate the wind function of a mass-transfer evaporation
model and compare it with different wind functions taken from the
literature. A stacked neural network was selected as the river
model based on a higher performance compared to the mass transfer

A alface (Lactuca sativa L.) ? sens?vel ?s condi??es clim?ticas, principalmente ? alta temperatura, modificando a sua arquitetura, peso, qualidade e, principalmente a produ??o. A a??o da temperatura do ar ? determinante nas mudan?as dos est?dios de desenvolvimento dos vegetais, interferindo tanto na emiss?o de folhas quanto na mudan?a dos
est?dios fenol?gicos, al?m de provocar altera??es morfol?gicas e fisiol?gicas. A faixa de temperatura de 15 a 24 ?C ? considerada adequada ao desenvolvimento da alface, entretanto, quando cultivada em regi?es de temperatura e luminosidade elevadas, esta hortali?a pode deixar de desenvolver todo o seu potencial gen?tico. A aduba??o pode ser utilizada para incrementar a produtividade na cultura da alface. No entanto, os efeitos das condi??es clim?ticas juntamente com a aduba??o e suas influ?ncias no crescimento cultura requerem maior aprofundamento no estudo, pois, o ambiente de cultivo, juntamente com a aduba??o e o componente gen?tico da planta, s?o os grandes respons?veis pelas mudan?as no crescimento, flora??o e senesc?ncia. Assim, objetivou-se verificar o comportamento e padr?o de crescimento de plantas de alface cv. Regina sob diferentes temperaturas e estrat?gias de aduba??es. O estudo foi conduzido em sala de crescimento, em DIC com esquema fatorial 4x6, sendo: quatro
temperaturas m?dias constantes para o cultivo da alface (12 ?C; 20 ?C; 23 ?C e 28 ?C) e seis aduba??es (controle, convencional mineral, Geofert - organomineral peletizado, Organomineral UFVJM, esterco de curral curtido e cama de codorna compostada). As plantas foram avaliadas quanto ao crescimento, p?s-colheita e perda de massa fresca em cada ciclo. Nas condi??es do experimento, os tratamentos cama de codorna, esterco de curral e organomineral Geofert promoveram maior crescimento ?s plantas de alface cv. Regina, sendo que essas estrat?gias de aduba??o podem permitir melhor desempenho produtivo das plantas at? mesmo em temperaturas de estresse. Os adubos org?nicos e Geofert s?o alternativas eficientes para o crescimento de plantas de alface cv. Regina e conferem ?s plantas menor perda de massa fresca, quando comparado ? aduba??o mineral convencional.
