▼ Studien undersöker invånare och verksammas upplevelser av ett, enligt en rapport frånunderrättelseenheten, utsatt bostadsområde i Västerås. För att besvara frågeställningen har vigenomfört 14 intervjuer varav 5 var semistrukturerade och resterande var ostrukturerade. Vihar även genomfört 6 observationer, där samtliga var icke deltagande observationer. Dåmålsättningen var att undersöka så förutsättningslöst som möjligt har vi använt oss av BarneyGlasers variant av grundad teori som metodologiskt angreppssätt. Tidigare forskning som vibelyser är delade i tre teman: Segregation, familjestrukturer och sysselsättning. En teorisom är grundad i data presenteras i resultatdelen och beskriver det som sker i Bäckby.Studiens teoretiska referensramar är Erving Goffmans teori om stigma, Howard. S. Beckersteori om avvikande samt Giorgio Agambens teori om undantag och belägring som fungerarsom en komplettering till förståelsen av hur det är i ett utsatt bostadsområde. Slutsatsernavisar att det är en handfull ungdomar som står för oron som sker i Bäckby och dess handlingarmöjliggörs på grund av misslyckade strukturella åtgärder. De förhållanden vi har funnitavgörande för trygghets upplevelse i Bäckby är inte endast att ungdomar behöversysselsättning, utan det behövs även en spridning av människor med utländsk bakgrund blandetniska svenskar och förebyggande åtgärder från externa aktörer.

synthesizing enzymes that
bypass the classical
penicillin-sensitive transpeptidation pathway by making 3-3
crosslinkages.
Until recently, there has been little physiological
data on the role of 3-3
crosslinks in cell envelope biology.
However, organisms with complex cell walls, such
as M.
tuberculosis, use L,D transpeptidation as the predominant form of
cross-linking,
indicating their importance. In E. coli and other
gram-negative bacteria, some of these
enzymes crosslink Braun’s
lipoprotein to the PG and incorporate non-canonical D-amino
acids
(NCDAA) into the PG, suggesting additional roles for these proteins
in cell
envelope stability. In this study, we constructed and
characterized Ldt mutants in E. coli
and M. smegmatis, as a model
organism for the pathogenic M. tuberculosis, to investigate
the
physiological role these enzymes play in cell envelope biology.
Of
the five LDts in E. coli, we show that loss of the Ldts involved
with coupling
the PG to Braun's lipoprotein resulted in the loss
of OM stability. The triple mutant strain
was sensitive to the
metal chelating agent EDTA, leaked periplasmic proteins, and was
resistant to the toxic effects of NCDAA. These data are consistent
with a role for these
enzymes in coupling the PG to the OM. Loss
of the Ldt enzymes involved with 3-3 crosslinkages
had no
discernable effect on the cell envelope. However, we detected an
effect
on cell growth only in the quintuple mutant bearing a DAP
auxotrophic mutation.
The genomes of M. tuberculosis and M.
smegmatis encode at least five Ldts. We
compared the translated
sequences of the Ldt genes in 18 mycobacterial genomes and
grouped
them into five classes. Of the single mutants, only one, ldtC
(Class 1), displayed
an increased susceptibility to imipenem,
which targets the Ldt enzymes. A triple mutant
missing ldtC and
the Class 2 genes exhibited a hypersusceptibility to imipenem,
ampicillin, lysozyme, and NCDAA. Consistent with previous findings,
we also show that
the catalytic activity of LdtC was dependent
upon an invariant cysteine in the active site.
This data suggests
that the enzymes represented by Class 1 and 2 may be key players
in
mycobacterial peptidoglycan physiology and that the phenotypic
differences displayed by
the mutant strains may reflect
differences in function of the mycobacterial Ldts classes.
In
addition, we constructed an Ldt null strain in M. smegmatis. This
strain
displayed a variety of growth defects such as a decreased
growth rate, and temperature
sensitivity at 42°C. We also show the
cell wall was affected in this mutant, as the strain
was
hypersensitivity to penicillins, cephalosporins, carbapenems,
vancomycin, and
lysozyme. Our data further suggests that the cell
envelope is altered as the Ldt null strain
was hypersusceptible to
rifampin, and ethambutol antibiotics and demonstrated enhanced
binding and uptake of Congo red.

▼ There is writing about how nineteenth-century churchgoers' views of other countries were formed by church periodicals. And there is writing about how opinions of the Cing and Japanese Empires were changed by the 'Cing-Japanese War of 1894-1895' ('First Sino-Japanese War'; 'Jiá-wǔ War'), and the 'Japanese-Táiwanese War of 1895'. But, there are no works making connections between these bodies of writing. This work makes such a connection through a comparison of writing about the wars in two church periodicals, the England-based Monthly Messenger and Gospel in China , and the Táiwan-based 'Dāi-lám Church News' ('Dāi-lám Hu-siá n Gào-ho ê -b e'). It makes the argument that the periodicals' writers and editors were on the side of the Japanese, as it seemed to them that Japanese success against the Cing Empire, and Japanese rule in Tái-wan, would make Western ways commoner in these places, which would be good for the Protestant Churches there. But the writers and editors had to give their opinions differently, as their readers were in different positions with respect to the wars. The Monthly Messenger's readers were in England; nothing the writers said about the war would make them any less safe, and so in both wars the periodical gave its support to the Japanese loudly and frequently. But the Church News' readers and writers were in Tái-wan. Openly supporting an attacking country could put them in danger, so the writers said nothing for or against any side in the first war, and were but quietly against the Táiwanese Republic in the second.

▼ Infecções causadas por tripanossomatídeos causam milhares de mortes anualmente, além de levarem a redução da capacidade produtiva, com elevada morbidade na população acometida. A eficácia terapêutica é limitada na maioria dos casos, sendo o benzonidazol o único fármaco aprovado para uso do tratamento do Trypanosoma cruzi, sendo ativo somente na fase aguda da Doença de Chagas, com grande efeitos colaterais. No caso das leishmanioses, as substâncias de tratamento existentes causam toxicidade renal e cardíaca, além de induzirem a resistência e apresentarem eficácia insuficiente. Observando todo o contexto torna-se necessário a busca por novas substâncias que sejam mais eficazes e menos tóxicas. Assim, o trabalho representa uma contribuição para a busca de novas moléculas bioativas para o tratamento da doença de Chagas e leishmaniose a partir da padronização e realização de ensaios celulares usando a forma epimastigota da cepa Y do T. cruzi e promastigota para Leishmania chagasi e L. amazonensis. A padronização do método colorimétrico de MTT foi realizada a partir da comparação com o método tradicional de contagem por microscopia usando câmara de Neubauer, seguido do estudo de viabilidade por citometria de fluxo, como método confirmatório e, finalmente a padronização do ensaio de ciclo celular usando os fármacos de referência benzonidazol (T. cruzi) e anfotericina B (Leishmania spp.). Os compostos testados neste estudo são proveniente de classes distintas de inibidores de alvos macromoleculares (cisteíno proteases, DHODH, GAPDH, quinases) e foram planejados e/ou selecionados pelo Grupo de Química Medicinal (NEQUIMED). Dentre todas as substâncias, inibidores de quinases apresentaram maior potencial para estudos subsequentes, sendo o T. cruzi o parasito mais sensível, onde um grande número de substâncias apresentou atividade nos estudos de triagem. Para uma delas (Neq0474) foi realizado o ensaio de dose-resposta, com EC50 = 53 µmol/L. L. chagasi apresentou a maior resistência dentre todos os parasitos estudados neste trabalho, enquanto que L. amazonensis foi sensível para Neq0438. Algumas substâncias estudadas apresentaram potencial para estudos mais aprofundados visando identificar novas alternativas terapêuticas para estas doenças parasitárias.
Infections caused by trypanosomatides lead to thousands of deaths annually, besides the reduction of the quality of life and working capability, with high morbidity to the patients. The therapy efficacy is limited in most cases, being benznidazole the only approved drug in Brazil, which works only in the acute phase of the Chagas Disease with severe collateral effects. For leishmaniasis, the drugs cause renal and cardiac toxicity and trigger resistance. In this context, novel efficacious and secure substantes are necessary to improve the current therapeutic strategies, which was the goal of this project by means of cell-based assays. ▼ In today's world, programming teachers perform tedious tasks, which are time consuming; for instance, preparing and marking daily assignments, preparing and marking programming projects, preparing and marking short exams, etc. These tasks distract programming teachers from fulfilling their key role - teaching. Therefore, using automated marking approach with ability to communicate with students is highly desirable. Despite the existing approaches for automated student marking, there is still a need for more improvement. An automated program marking approach is proposed in this study based on a proposal by iMarking®. This approach automates the process of marking and assignments submission and facilitates the communication between teachers and students by designing and implementing a web-based application. In addition, the proposed approach adopts Optical Character Recognition (OCR) to extract the text from images to be evaluated using novel evaluation metrics. The novel evaluation metrics are formulated based on observation and experiment and aim to calculate the matching similarity and mismatching percentage of the submitted student answers when compared with the optimal answers. Evaluation results from a sample of 100 different programming questions show that the proposed approach is efficient in automatically marking the student answers with 100% accuracy. Furthermore, it is found to be time saving - approximately 197 seconds for marking ten questions - which is in line with the objective of creating a more efficient system for teachers.

▼ Immune surveillance requires efficient trafficking of leukocytes throughout the body. To achieve this, leukocytes have evolved to be highly migratory and responsive to environmental cues, which provide guidance for proper tissue distribution. The translation of external environmental cues to intracellular physical changes in leukocytes requires a cascade of receptors, signal transducers, and mechanical effectors. My doctoral research focused on using
T-cells as a model to study the unique cellular process of how signal transducers interact with and regulate mechanical effectors in fast migrating immune cells. Specifically, it is known that the signal transducer Mst1 kinase is required for
T-cell polarization, adhesion, and active migration, but the underlying mechanisms remain poorly understood. I have demonstrated that Mst1 regulates two mechanical effectors, the molecular motor Myosin-IIA and the cytoskeleton regulatory protein
L-plastin, through the process of phosphorylation. Theregulation of Myosin-IIA enables it to generate contractile force inside a migrating
T-cell, maintaining the shape and proper adhesion of the cell to extracellular matrix, both being requirements for successful migration. The regulation of
L-plastin enables it to activate integrin adhesion molecules as well as to properly organize lamellipodial actin. In addition, I have identified novel adhesion structures in
T-cells called microadhesions, which potentially provide traction force to migrating
T-cells. Overall, my research has identified a novel pathway acting between a signal transducer and two mechanical effectors in
T-cell migration.

▼ This research has been undertaken in response to an average 50% attrition rate from a technology enhanced distance-learning course in applied art and design. It studies the participants’ lived experience over a 12-week period from enrolment to the end of the first of twelve modules. The commercial provider recruited 23 mature, female participants from 8 different countries for this City & Guilds certificated course. Data was collected utilizing Interpretive Phenomenological Research methodology (IPA) via two open-ended telephone interviews with participants. The twin roles of the study author as researcher and director of the educational provider were reviewed and a methodology for disaggregation proposed. ATLASti software was used to aid data coding and retrieval. Four key themes emerged from the data; Confidence, Persistence, Support and Timing, which were each subsequently divided into 3 sub categories. Literature searches moved from education to psychology as the study progressed. Implications for further discussion encompass pre-course testing, provider conflict of interest, innovative methods of support and increased legislation. Findings centre around the commencement process and the need for provider instigated advice at the beginning of the course experience, as well as the limitation of design content during the first module and the accuracy of pre-course information and the participant’s engagements and understanding of it. The study also found a lack of correlation between early tutor/student support and retention and several planning activities undertaken by a sub-section of the participants, which appeared to be successful. This study has also contributed to the literature on retention, highlighting the participant’s personal experiences rather than the quantitative retention data more usually associated with retention studies and has demonstrated that interventions intended to decrease course attrition levels may be required at an earlier stage than previously thought and also across a wider subject range than previously recognised.

▼ Mountain snowpacks act as seasonal reservoirs, providing a critical water resource to ~1.2 billion people globally. Regions with persistent snowpacks (e.g., mountain and polar environments) are responding quickly to climate change and are warming at faster rates than low-elevation temperate and equatorial regions. Since 1915, snow water equivalent (SWE) in the western U.S. snowpack has declined by 21% and snow covered area is contracting in the Rocky Mountains. Despite the clear importance of this resource and the identification of changes affecting it, no current remote sensing approach can accurately measure SWE at high spactiotemporal resolution.
L-band (1-2 GHz) Interferometric Synthetic Aperture
Radar (InSAR) is a promising approach for detecting changes in SWE at high spatiotemporal resolution in complex topography, but there are uncertainties regarding its performance, particularly when liquid water content (LWC) is present in the snowpack. LWC exhibits high spatial variability, causing spatially varying
radar velocity that introduces significant uncertainty in SWE-retrievals. The objectives of this thesis include: (1) examine the importance of slope, aspect, canopy cover, and air temperature in the development of LWC in a continental seasonal snowpack using 1 GHz ground-penetrating
radar (GPR), a proxy for
L-band InSAR, and (2) quantify the uncertainty in
L-band
radar SWE-retrievals in wet-snow. This research was performed at Cameron Pass, a high elevation pass (3120 m) located in north-central Colorado, over the course of multiple survey dates during the melt season of 2019. Transects were chosen which represent a range in slope, aspect and canopy cover. Slope and aspect were simplified using the northness index (NI). Canopy cover was quantified using the leaf area index (LAI). Positive degree days (PDD) was used to represent available melt-energy from air temperature. The spatiotemporal development of LWC was studied along the transects using GPR, probed depths, and snowpit measured density. A subset of this project substituted Terrestrial LiDAR Scans (TLS) for probed depths. Surveys (17 in total, up to 3 surveys per date) were performed on seven dates which began on5 April 2019, where LWC values were ~0 vol. %, and ended on 19 June 2019 where LWC values exceeded 10 vol. %. Point measurements of LWC were observed to change (ΔLWC) by +9 vol. % or -8 vol. % over the course of a single day, but median ΔLWC were ~0 vol. % or slightly negative. LAI was negatively correlated with LWC for 13 out of the 17 surveys. NI was negatively correlated with LWC for 10 out of the 17 surveys. Multi-variable linear regressions to estimate ΔLWC identified several statistically significant variables (p-value < 0.10): LAI, NI, ΔPDD, and NI x ΔPDD. Snow-on Terrestrial LiDAR Scans (TLS) were conducted twice during the melt season, and a snow-off scan was conducted in late summer. Les L-moments sontdevenus des outils classiques alternatifs aux moments centraux pour décrire les comportementsen dispersion, asymétrie, kurtosis d’une distribution univariée à queue lourde. Eneffet, contrairement aux moments centraux correspondants, ils sont bien définis dès quel’espérance de la distribution d’intérêt est finie. Les L-moments peuvent être vus comme laprojection de la fonction quantile sur une famille orthogonale de polynômes, récupérant lalinéarité inhérente aux quantiles. Nous estimerons dans un premier temps les paramètresde modèles semi paramétriques définis par des contraintes sur ces L-moments par des méthodesde minimisation de divergences.Nous proposons dans un second temps une généralisation des L-moments aux distributionsmultivariées qui passe par la définition d’un quantile multivarié défini comme untransport entre la distribution uniforme sur [0; 1]d et la distribution d’intérêt. The description and the estimation of univariate and multivariate models whose underlyingdistribution is heavy-tailed is a strategic challenge. ▼ La détection radar passive met à profit des émetteurs non-coopératifs, déjà présents dans l’environnement, qui transmettent des signaux de télécommunications, de type DVB-T dans l’étude présentée.Elle utilise les réflexions de ces signaux sur de potentielles cibles et les exploite comme échos radar au niveau d’un récepteur aéroporté.Ces nouveaux systèmes de détection, par nature discrets et économes en énergie et en allocation de fréquences, étendent la surveillance à la basse altitude.Si les différentes étapes des traitements classiques utilisés en radar passif terrestre (estimation du signal de référence, réjection, filtrage adapté, détection)demandent d’être réorientées sérieusement pour répondre aux contraintes liées à la réception aéroportée,il en va de même du récepteur qui doit satisfaire les exigences matérielles de la plateforme aérienne.Dans ce but, un système expérimental embarqué sur motoplanneur a été développé permettant d’acquérir des signaux réels indispensablesà la compréhension de l’impact de la propagation des signaux DVB-T.La méthode d’estimation du signal de référence proposée permet d’une part, de lutter contre les fluctuations du canal de propagation induites par les multi-trajetsen exploitant la diversité d’antenne et d’autre part, de prendre en compte les variations temporelles en s’appuyant sur la méthode BEM (Basis Expension Model).Ensuite, une analyse théorique sur la répartition du fouillis de sol est apportée.L’exploitation des signaux expérimentaux permet de la valider par une analyse dans le plan distance-Doppler et angle-Doppler.Une projection cartésienne permet de mettre en évidence des échos forts confrontés avec la vérité terrain.L’estimation du signal de référence et la connaissance de l’étalement du fouillis de sol sont les piliers fondamentaux de la détection car ces composantes représentent deux contributions à rejeter.Pour le signal de référence, une méthode classique de réjection où les coefficients du filtre sont estimés au sens des moindres carrés est mise en oeuvre.Un filtrage spatial orthogonal à la direction d’arrivée du signal de référence est ajouté afin de diminuer l’impact du bruit émis.Le large étalement en Doppler et en distance nous a conduit à rejeter le fouillis sur des périodes de corrélation plus courtes.Les travaux présentés apportent une compréhension fine de l’impact de la propagation sur les traitements de détection en radar passif aéroporté et offrent des perspectives engageantesquant à la détection de cibles de moyennes à grandes Surfaces Equivalentes Radar.

▼ Genomic era begins with development of sequencing methods. Genome sequencing is now cost-effective and fast, giving rise to increasing amounts of genomic data. However, the function of 1% of the deposited sequences have been experimentally characterized. There is no robust method of functional assignment for these sequences. Functional assignments are now performed using a variety of software tools to utilize the known biochemical data of characterized proteins to annotate similar sequences in genome databases. With this large scale automatic annotations of genome databases, annotations were transferred from homologs regardless of their reliability which result in propagation of errors and transfer misleading information to scientific community. Nevertheless, annotating the homologs within a superfamily is a valid approach. To this end, the enolase superfamily is an excellent model system for functional assignment. Structurally, this superfamily contain substrate specificity residues in the N-terminal capping domain and catalytic residues in the C-terminal barrel domain. These proteins with the common structural fold have the ability to abstract a proton α to carboxylate on the substrate before proceeding to dehydration, epimerization, deamination, racemization or cycloisomerization. There have been enough studies of this superfamily to provide valuable insight into the types of reactions performed based on catalytic residues and substrate specificity residues, establishing basis for functional characterization of unknown members.
In this thesis, I discuss my efforts on characterizing an enolase superfamily member:
D- glucarate dehydratase related protein (GlucDRP). GlucDRP share more than 60% sequence identity with GlucD, a well-characterized protein. I showed that there is a protein-protein interaction between GlucD and GlucDRP which form a detectable heterospecies. The kinetics of heterospecies, isolated from wild type E. coli, was equal to an average of the activity of a GlucD and a GlucDRP. Additionally, to determine what percent of GlucDRPs involves in the formation of the heterospecies, a hexahistidine-tag was introduces upstream of the GlucDRP gene in the E. coli chromosome and it was shown that more than half of expressed GlucDRP forms heterospecies. Finally, the three-dimensional crystal structure of the heterospecies was determined which confirmed the formation of α2/β2 tetramers of GlucD and GlucDRP.
At a BLAST e-value of 10-175, three clusters of GlucDRPs segregated from the authentic GlucD. Structural alignments of GlucD and GlucDRP proteins showed good superposition between these structures except in 100s loop, which interacts with the substrate and closes the active site upon entrance of the substrates. This structural differences may be responsible for differences in catalytic activity.

▼ Dentro das possibilidades terapÃuticas atuais para o tratamento de pacientes com problemas congÃnitos de metabolismo, a dieta constitui o pilar mais importante e a D-tagatose tem atraÃdo uma grande atenÃÃo nos Ãltimos anos devido a seus benefÃcios à saÃde humana, bem como à semelhanÃa de suas propriedades com a sacarose. Dentre as suas muitas aplicaÃÃes, ressalta-se o potencial em auxiliar no controle de peso, uma preocupaÃÃo crescente no Brasil, uma vez que a obesidade cresce a ritmos alarmantes. No entanto, a L-arabinose isomerase, enzima que catalisa isomerizaÃÃo de D-galactose em D-tagatose, ainda nÃo està disponÃvel comercialmente e, portanto, estudos visando à obtenÃÃo deste biocatalisador se fazem necessÃrios de maneira a viabilizar a implantaÃÃo do processo industrial. Portanto neste trabalho, estudou-se a obtenÃÃo da enzima L-arabinose isomerase utilizando uma cepa de Enterococcus faecium. A enzima produzida por fermentaÃÃo foi caracterizada e imobilizada em suportes a base de quitosana. Os resultados de estabilidade tÃrmica, operacional e de estocagem obtidos para a enzima imobilizada covalentemente sobre quitosana em meio alcalino (pH 10), confirmou a importÃncia do controle do pH durante a imobilizaÃÃo, uma vez que a formaÃÃo de uniÃes multipontuais à favorecida quando comparado ao pH 7,0. No entanto, baixas concentraÃÃes de proteÃna eram obtidas na etapa de fermentaÃÃo, portanto, estudou-se a produÃÃo da enzima L-AI de forma heterÃloga em Escherichia coli. As proteÃnas recombinantes expressas foram purificadas por cromatografia de afinidade em uma Ãnica etapa, e visualizadas em SDS-PAGE. O sucesso na construÃÃo do gene e na clonagem em vetores de expressÃo em E. coli resultou em quantidade satisfatÃria de expressÃo das proteÃnas recombinantes, pois apresentaram-se na forma solÃvel, facilmente purificadas e ativas, permitindo suas caracterizaÃÃes. Por ultracentrifugaÃÃo analÃtica foi possÃvel descobrir que a enzima L-AI recombinante tem uma tendÃncia para formaÃÃo de estruturas com maior tamanho (oligÃmeros). A seguir, suportes multifuncionais foram preparados para a imobilizaÃÃo da L-AI e observou-se uma rÃpida imobilizaÃÃo, apresentando um elevado rendimento de imobilizaÃÃo, superior a 75%. Devido à baixa estabilidade tÃrmica dos derivados, estudos futuros serÃo necessÃrios para a estabilizaÃÃo da estrutura quaternÃria desta enzima.

▼ In dit onderzoek wordt bekeken wat de invloed is van spelfouten in sollicitatiebrieven en sponsorbrieven op de tekstwaardering, overtuigingskracht en het imago van de zender. Uit eerdere wetenschappelijke onderzoeken en scripties weten we dat lezers van sollicitatiebrieven letten op spelfouten. Lezers van andere soorten brieven letten niet of minder op spelfouten. Vanwege een verschil in materiaal en vragenlijsten zijn de resultaten van eerdere onderzoeken echter niet goed te vergelijken. Daarom werden in dit onderzoek twee zoveel mogelijk op elkaar
lijkende brieven met zes spelfouten en zonder spelfouten opgesteld. In hoeverre is er een verschil in beoordeling tussen sollicitatie- en sponsorbrieven met spelfouten?
Het type spelfout dat in dit onderzoek centraal staat is een ten onrechte weggelaten –
t bij de stam van een werkwoord in de derde persoon enkelvoud (‘hij vind’). Er is een sollicitatiebrief en een sponsorbrief van vijf alinea’s lang geschreven met fictieve bedrijfsnamen en personages.
Van elke brief werden vier versies gemaakt: een foutloze versie van schrijver Anna de Jong, een foutloze versie van schrijver Peter van der Berg, een versie met zes spelfouten van schrijver Anna de Jong en een versie met zes spelfouten van schrijver Peter van der Berg. Proefpersonen
schreven na het lezen van één van de brieven ieder maximaal vijf gedachten op die ze hadden tijdens of direct na het lezen van de brief. Daarna beantwoordden zij vragen over de afhankelijke variabelen. Er deden 160 werknemers van organisaties in allerlei bedrijfstakken aan dit
onderzoek mee. Van alle proefpersonen was 45 procent man en 55 procent vrouw. De gemiddelde leeftijd van alle proefpersonen was 43 jaar.
Uit dit onderzoek blijkt dat spelfouten een negatieve invloed hebben op de tekstwaardering, overtuigingskracht en het imago van de zender. Deze invloed is bij splitsing van de proefpersonen op kennis van spelling het sterkst voor de groep met kennis van spelling. Proefpersonen waarderen een tekst met spelfouten minder positief dan een foutloze tekst. Zij vinden een brief met spelfouten ook minder overtuigend dan een foutloze brief. De meerderheid van de proefpersonen heeft bij brieven met en zonder spelfouten geen gedachten over de schrijver of deze in ieder geval niet opgeschreven. Wordt de schrijver wel genoemd, dan is dit vooral door lezers van een brief met spelfouten. Kenners van spelling vinden de schrijver van een brief met spelfouten minder deskundig, minder betrouwbaar en minder aantrekkelijk dan de schrijver van een foutloze brief. Het oordeel van de niet-kenners staat hier haaks op. Zij beoordelen het imago van de schrijver van een brief met spelfouten juist positiever dan het
imago van de schrijver van een foutloze brief. Alle proefpersonen vinden de sollicitatiebrief begrijpelijker dan de sponsorbrief. Het negatieve effect van spelfouten geldt voor kenners vooral voor de sponsorbrief en juist minder voor de sollicitatiebrief. Dit gaat tegen de algemene norm

▼ Recent developments in fusion technology enable the use of high-yield deuterium-tritium (
D-
T) based generators for a variety of applications related to nuclear engineering, medicine, security, and material analysis. We recently designed and set up a laboratory based on a
D-
T generator. The generator and ancillary equipment can be remotely monitored and operated. The generator will mainly be used to perform scatter-based experiments to characterize fast-neutron sensitive detectors. We simulated the facility using the Monte Carlo code MCNP to quantify the dose that facility operators and the general public will be exposed to during facility operations. Also, validation of the simulated results was conducted with experimental work involving a new type of Bonner sphere spectrometer. This facility will be available to test equipment and perform experiments in a well-characterized neutron field.

Tarkoituksena oli laatia Kurikan terveyskeskuksen laboratorion ja lääkärinvastaanoton käyttöön selkeä ja helppokäyttöinen perehdytysmateriaali valituille analyyseille. Opinnäytetyön tehtävänä oli selvittää vierianalytiikan tutkimusten Troponiini T:n sekä Fibriinin D-dimeerien laboratoriodiagnostiikka ja niiden merkitys potilaan hoidossa sekä laatia kirjallinen perehdytysmateriaali Cobas h 232 liuskanlukulaitteelle ja sen vieritesteille P-TnT ja P-FIDD.
Opinnäytetyössä on raportti- ja tuotososio eli se tehtiin toiminnallisena opinnäytetyönä. Opinnäytetyön raporttiosuus sisältää teoriatiedot perehdytyksestä ja kirjallisesta perehdytysmateriaalista, vierianalytiikasta, vierianalytiikan laadusta, vierianalytiikkaan perehdyttämisestä, Cobas h 232 liuskanlukulaitteesta, Troponiini T ja Fibriinin D-dimeerien diagnostisesta käytöstä. Raporttiosuudessa käydään läpi myös toiminnallinen opinnäytetyö, opinnäytetyön prosessi sekä kuvataan tuotos.
Työn tuotos on perehdytysmateriaali, jossa tuodaan esiin liuskanlukulaitteen toimintaperiaate, laadunvarmistus, potilasnäytteen määrittäminen sekä tulosten tarkastelu. Perehdytysmateriaalin avulla työntekijä saa kokonaisvaltaisen kuvan Cobas h 232, liuskanlukulaiteen käytöstä ja sen diagnostilaisesta käytöstä. Perehdytysmateriaali on suunniteltu niin laboratorion kuin lääkärinvastaanoton uudelle henkilöstölle ja opiskelijoille.
This thesis topic was given by JIK Federation of Municipalities, Kurikka health center laboratory. The purpose of this work is to establish orientation material regarding Cobas h 232 strip reader and the analyses of fibrin D-dimers and Troponin T. The target is to establish an easy-to-use material for Kurikka health care centre. This study was to investigate the Point of care testing studies Troponin T and fibrin D-dimer in laboratory diagnostics and their role in patient care, as well as to prepare a written orientation material regarding Cobas h 232 stripe reader.
The thesis is a report and has an output section, and it was carries out as a functional thesis. The report of the thesis contains information on the theory of induction and orientation material, Point of care testing, the quality, orientation of Point of care testing, the Cobas h 232 strip reader and how to and why use Troponin T and fibrin D-dimer analyses. The report section covers also the functional thesis, its process and the output description.
The work output of the orientation material, which is treated strip reader-principle of the working head, quality assurance, patient sample determination, as well as the results of the review. ▼ Ratvísi T-frumna til húðar er miðlað af mörgum þáttum. Má þar nefna sameindirnar CLA og CCR4 sem eru tjáðar á T-frumum og stjórna fari þeirra í gegnum æðaþekjuna og til húðar. Viðtakinn CCR10 kemur einnig við sögu og gerir T-frumum kleift að ferðast til yfirhúðar. Vítamínið D3 er myndað í yfirhúð. Það er forveri virka efnisins 1,25(OH)2D3. Sýnt hefur verið fram á að angafrumur geti myndað 1,25(OH)2D3 út frá D-vítamíni í húð og sýnt T-frumum. Markmið þessa verkefnis var að athuga hvort 1,25(OH)2D3 hvetji tjáningu CCR10 á T-frumum og stuðli þar með að fari þeirra til húðar.
Einkjarna blóðfrumur voru einangraðar úr blóði og ræktaðar í 12 daga með IL-2. Á 6. degi ræktunar var 1,25(OH)2D3 bætt út í helming ræktar. Eftir 12 daga voru framkvæmdar innan- og utanfrumulitanir og sýnin keyrð í frumuflæðisjá.
Marktæk aukning sást á tjáningu CCR10 í frumum ræktuðum með 1,25(OH)2D3 samanborið við viðmiðunarrækt. CCR4+ T-frumur sýndu mesta tjáningu á CCR10 hjá bæði CD4+ og CD8+ T-frumum. CCR10 tjáning fór hins vegar ekki saman við tjáningu CLA. Hugsanleg ástæða þessa er sú að T-frumurnar voru ræktaðar í 12 daga eftir ræsingu og tjáning CLA gæti verið lokið. Einnig var litað fyrir CD25, CD127 og α1 en það eru sameindir sem einkenna T-stýrifrumur. Niðurstöður gáfu til kynna að tjáning CCR10 virðist ekki vera tengd T-stýrifrumum. Til að skapa aðstæður sem líkjast þeim sem myndast í bólgum fékk hluti frumuræktarinnar IL-12. Þetta jók CCR10 tjáningu og hæsta tjáning á CCR10 mældist í ræktunum með IL-12 ásamt 1,25(OH)2D3.
Vegna óljósra ástæðna fengust ekki marktækar niðurstöður úr innanfrumurækt.
Hér hefur verið sýnt fram á að 1,25(OH)2D3 hvetur tjáningu CCR10 á T-frumum og hvetur þannig far þeirra til yfirhúðar Þær upplýsingar er hægt að nýta við áframhaldandi rannsóknir á ratvísi T-frumna til húðar.

▼ The purpose of this thesis us three-fold: 1. to research The Balcony by Jean Genet in order to organize a scholarly approach for directing a production of the play; 2. to record direction of the play; 3. to evaluate the research and direction and its effect on production. The thesis is a compilation of the director's research, analysis and experience with the production. Part One is the director's analysis of the script including: 1. historical considerations; 2. stylistic analysis; 3. character analysis and descriptions; 4. setting analysis; 5. justification of scriptual choice; and 6. directorial interpretation of the play for production. Part Two is the prompt book for the director's production of The Balcony which took place on November 29, 30 and December 1, 2 and 3, 1972 in the theatre of W. Raymond Taylor Drama and Speech Building at the University of North Carolina at Greensboro. The prompt book is the production script which includes notation on: 1. blocking; 2. composition; 3. details of characterization; 4. stage business; 5. pace, rhythm and tempo; and 6. technical cues. Interspersed within the prompt book are photographs and figures. Part Three is the director's evaluation of the production. ▼ Lymphocytes require antigenic encounter to activate and proliferate, eventually clearing the source of antigenic challenge. The peripheral lymph nodes (PLN) are the primary sites of antigenic encounter and thus the ability of lymphocytes to migrate to this tissue is a requirement for mounting effective immune responses. The process of lymphocyte migration to the PLN is known as the "adhesion cascade". Specifically, lymphocytes are captured from the blood through the adhesion molecule,
L-selectin, followed by CC chemokine receptor 7 (CCR7)-mediated integrin activation, which ultimately results in cell transmigration into the PLN. Because the PLN is the site where antigenic encounter is most likely, we hypothesized that migration may induce pro-proliferative signaling, mediated by
L-selectin and conducted through CCR7. To study the potential "priming" effects of adhesion to antigen-mediated lymphocyte proliferation, spleen and PLN lymphocytes were subjected to in vitro transmigration and proliferation assays. Subsequently, multi-color flow cytometry analysis was used to examine treatment effects on the
T cell population. Results showed that lymphocyte proliferation was significantly increased when
L-selectin and CCR7 were activated simultaneously, compared to CCR7 activation alone.
Specifically, bulk tissue populations, as well as the CD4+ and CD8+
T cell subsets showed enhanced proliferation from both spleen and PLN sources at early and late time points. Additionally, to discern whether this enhanced proliferation may be a by-product of decreased apoptosis, an apoptosis assay was also performed at the same time points. Interestingly,
L-selectin engagement resulted in protection from apoptosis for all
T cell populations 72 hour following activation. However, this protective effect was not observed at the 36 hour time point. Taken together, these results suggest a role for
L-selectin in enhancing
T cell proliferation, at least in part, through an anti-apoptotic effect. This research will add to a better understanding of the regulation of lymphocyte proliferation and increase our knowledge of
L-selectin signaling function.

▼ This study investigates the existence and character of tidal flow separation at the Sand Motor, a mega-scale sand nourishment at the Delfland coast. Tidal flow contracts at the tip of the Sand Motor, downstream the tip the flow diverges and flow separation could occur. In case of flow separation the diverging streamlines cannot follow the contours of the coastline and the main flow will detach from the coastline. Flow separation causes an eddy downstream of the tip. The spatial variation in tidal flow is important to investigate, because the presence of tidal flow separation could be a hazard for swimmer safety. An advantage of the presence of tidal flow separation is the possible increased mixing of nutrients. By a literature study the first expectations about the location and period of tidal flow separation are done. Comparable studies to tidal flow separation are done in the past, which are relevant for this research. Two important conditions for tidal flow separation are an adverse pressure gradient and a developed boundary layer. From the literature study can be concluded that the location of the eddy generated by tidal flow separation is on the leeside of the headland. During the second half of the flood or ebb period tidal flow separation was expected, because then the two conditions are present. The core of this research is the field measurement campaign using GPS-tracked drifters. In total 15 measurement sessions are performed during both flood and ebb periods. The drifters recorded flow convergence and offshore flow near the tip of the Sand Motor, which possibly indicated the presence of tidal flow separation. The observed flow patterns are subsequently processed and characterized. To characterize the measurement sessions, the environmental conditions are researched. By observing
radar images of the drifter sessions, the importance of the freshwater plume, discharged from the Rhine River, became visible. It interrupted tidal flow separation multiple times during drifter sessions. Also tidal flow separation and the associated eddy were observed in the
radar images. Interpretation of the measurements shows that the wind conditions and the freshwater plume are the most important environmental conditions for the appearance or absence of tidal flow separation. By observing the results of several flood measurement sessions, the eddy seems to grow in time and gradually extends further offshore. The importance of the timing of the drifter sessions, to measure the tidal flow separation is tested with the numerical model
D-Flow FM. The model is used to research the development in time of the flow separation and the associated eddy. Simulations show that the eddy travels from north to south during flood, which explains the measurements several flow patterns during flow separation. The simulations also show growth of the eddy between its generation and dissipation. ▼ SAR (Synthetic Aperture Radar) images have been mainly used to geological applications based on a more qualitative approach, which tends to subutilize the total content of geological information available in the data. With the advent of polarimetric SAR systems, quantitative analysis is becoming essential, particularly when focusing on the understanding of the energy/target interactions in the microwave spectrum. This investigation aimed to evaluate in both qualitative and quantitative ways the geological applicability of multipolarized L-band SAR data acquired by the SAR-R99B system (SIVAM-CENSIPAM) in the Lower Curaçá River Valley, Bahia State. The study area is characterized by a semi-arid environment, with a flat topographic terrain, showing shallow residual soils well correlated to the substratum and sparse caatinga vegetation. Geologically, the region is part of the São Francisco Craton, and comprises Paleoproterozoic granulites, gneisses, granites and a sequence of Cu-mineralized maficultramafic intrusives. Subordinately, metasedimentary rocks represented by schists, phyllites, marble of Neoproterozoic ages are also found. The methodology can be divided into two steps: (1) qualitative analysis based on photogeological interpretation of the multipolarized images and (2) quantitative analysis through statistical approaches (linear regression and separability) using as input backscattered responses (backscattering coefficient of Lhh, Lvv, Lhv, Lvv/Lhv and Lhh/Lhv) and surface roughness information (micro-topography). The investigation has allowed several findings: (1) the qualitative analysis showed that Lvv polarization presented an overall best performance in the discrimination of the main rock units and in the characterization of the main geological structures in the study area. The poorest performance was related to Lhh; (2) statistical analysis based on linear-regression indicated Lvv polarization as the most sensitive band to the micro-topographic variations, followed by Lhh, Lhv and ratio Lvv/Lhv. Furthermore, the ratio Lhh/Lhv presented the poorest correlation with surface roughness; (3) separability analysis also pointed out the Lvv polarization as the most important band in the discrimination of mafic-ultramafic responses related to Vertisols, followed by Lhv and Lhh; (4) moisture affecting the dielectric properties of soils and vertical structure of caatinga vegetation were important controls in the backscattered responses which favored the rock unit discrimination. Finally, the high spatial resolution of the SAR images was important in the detection of the geological structures in the area.

O presente trabalho visa comprovar a hibridação natural entre Digitalis thapsi L. (endemismo ibérico) e Digitalis purpurea L. por meio de técnicas morfológicas e moleculares.
Procedeu-se à caracterização morfológica através da microscopia ótica e eletrónica de varrimento e à análise molecular, através da técnica RAPD-PCR, sendo os resultados obtidos submetidos a tratamento estatístico usando para o efeito técnicas e programas informáticos adequados - SPSS e NTSYS-pc.
Os resultados da análise macroscópica mostraram, terem os exemplares híbridos, folha e corola morfologicamente semelhantes às de D. thapsi, e cálice floral similar ao de D. purpurea. A corola destes exemplares híbridos é de coloração intermédia entre as duas espécies (D. thapsi e D. purpurea), tendo também os dois tipos de máculas característicos destas espécies sido observados nestes.
A nível microscópico, os exemplares híbridos observados mostraram ter: a cutícula das células epidérmicas das folhas e flores e o tipo e forma dos tricomas secretores, semelhante aos observados em D. thapsi; e o tipo e forma dos tricomas tectores e número de camadas de células do parênquima lacunoso foliar semelhantes aos de D. purpurea. O parênquima em paliçada dos exemplares híbridos observados tem um número de camadas celulares intermédio entre as duas espécies (D. thapsi e D. purpurea).
A observação e tratamento dos dados moleculares obtidos por RAPD-PCR mostrou terem os exemplares híbridos e D. thapsi um perfil molecular muito semelhante, sendo evidente a predominância dos caracteres desta espécie (D. thapsi) nestes, permitindo assim concluir serem os exemplares observados um híbrido D. thapsi x D. purpurea.
The aim of this work was to confirm the natural hybridization between Digitalis thapsi (Iberian endemism) and Digitalis purpurea L. by means of morphological and molecular techniques.
Proceeded to the morphological characterization by optical microscopy and scanning electronic and molecular analysis by RAPD-PCR, and the results analyzed statistically using for this purpose appropriate techniques and software - SPSS and NTSYS-pc.
Macroscopic results revealed that the hybrid presents leaves and corolla similar to D. thapsi and floral cup similar to D. purpurea. Hybrid corolla appeared to be intermediate color and the two types of stains found in other species. The corolla of exemplary hybrid is intermediate in color between the two species (D. thapsi and D. purpurea), and also the two types of stains characteristic of these species were observed in these. At microscopic level, the samples proved to be hybrids observed: the cuticle of the epidermal cells of leaves and flowers and the type and form of secretory trichomes, similar to those observed in D. thapsi, and the type and shape of trichomes and number of layers of spongy parenchyma cells leaf similar to D. purpurea. ▼ This study integrated multi-temporal, multispectral optical and L-band synthetic aperture radar (SAR) imagery to classify agricultural crops throughout a single growing season in northeastern Ontario, Canada. Various optical and SAR band/date combinations were tested to identify optimal dates and datasets for crop classification at various phenological stages using both object-based decision tree rulesets and traditional per-pixel strategies. Object-based decision tree classification of 2 pairs of SPOT-5 optical and L-Band ALOS SAR imagery yielded crop identification accuracy results comparable with hierarchically masked per-pixel classification, with corn classes regularly achieving high classification accuracies (+90%). Regardless of classification approach, results indicate that at least one complimentary date of optical imagery be used in combination with an mid- season optical/ SAR imagery pair to optimally classify northeastern Ontario agricultural landscapes.

This study is a critical assessment and analysis of Xhosa poetry that addresses social and political concerns with particular reference to the poetry of Qangule (1970), Mtuze (1982, 1990) and Xozwa (1994 ). Chapter one introduces the study by presenting its aim, scope, method of research, theory, definition of concepts and significance of the study. Chapter 2 focuses on the theoretical framework of the study. It discusses the theory of Marxism, which is used as a basis of this study, by explaining the concept of Marxism, its historical background, assumptions, methodology and Marxism and poetry. Chapter 3 deals with social concerns that are addressed by Mtuze (1982 and 1990) and Xozwa (1994 ). These social concerns are divorce and alcohol usage. The discussion in this chapter focuses on how divorce affects members of the family concerned, especially children of divorced parents. It also looks at how alcohol is used in society. While the discussion in this chapter acknowledges moderate use of alcohol, it focuses more on how this substance may also be abused. It looks at how alcohol abuse affects individuals and society economically and how dangerous it is to the health and lives of people. Chapter 4 focuses on political concerns as addressed by Qangule (1970) and Mtuze (1982). It assesses and analyses poems that address the oppression of black people during the apartheid regime in South Africa. Concerns like citizenship and deprivation of freedom, labour conditions and language are discussed in this chapter. Chapter 5 is a concluding statement which gives a summary of the study, evaluation of Xhosa poetry with regard to addressing the concerns discussed in the study and recommendations regarding future research.

▼ The aim of this project is to determine whether a computer application can be used to develop phonemic awareness in the early primary classroom, which is a key component of phonics. This thesis explores the evolution of the strategy for teaching literacy in the UK which shows phonics to be a key component of that strategy. However, government reports which inform the direction of the literacy strategy call for more empirical study in all areas of literacy teaching; this thesis documents such an empirical study. This research project creates a phonics based computer application designed specifically for young children aged 5 to 6 years (year 1 in UK primary schools). The timing and level of content presented by the computer application activities are grounded in appropriate academic theory. A significant component of the work is the development of interface design guidelines for children’s applications. These guidelines are then used to inform the development of the phonics-based computer application. A Randomised Controlled Trial (RCT) is designed to determine the application’s effectiveness in developing the phonemic awareness skills of young children in a classroom setting. In order to control experimental bias resulting from problems with the usability of the computer interface, the usability of the application’s interface is evaluated in the classroom by year 1 children before the application is used in a pragmatic RCT. The results of the final usability evaluation found no usability issues and the application was wholly intuitive to the children in the evaluation groups. The results from the RCT (N=266) show no statistically significant improvement in the learning rate of phonemic awareness by the intervention group using the computer program compared to the traditional teacher-delivered paper-based method used with the control group, even though the computer program was designed carefully for this age range. The results did suggest however, that the intervention group developed at the same rate as the control group which implies that the computer program could be used to support teachers by reducing the amount of resource-intensive phonics tuition required by children in this age range.

▼ Argument diagramming can scaffold the process of argumentation but only a few studies have investigated its impact on the quality of argumentative writing. This research contributed to this direction with two studies.
An exploratory study investigated the impact of argument diagramming, applied as a paper-based or a computer-based method, on the quality of argumentative text. The computer method increased the refutations and overall quality of essays. The study highlights the significance of writers’ argumentative ability for interpreting improvement.
A qualitative study looked into the impact of argument diagramming on the process of writing cognition through analysis of online process data, diagrams and essays of sixteen undergraduate students. Writers with myside bias schema used the method to increase counterarguments and refutations. Writers at lower level of pseudo-integration adopted more advanced strategies like weighing, and writers at middle level of pseudo-integration formed positions with qualifications. Needs at higher levels of argumentative ability are not met. The support of writing planning processes through argument diagramming affects mainly the semantic aspects of the text while the support of linearization processes affects mainly the rhetorical aspects.
The analysis of interviews revealed that interacting with argument diagramming can improve awareness of argumentation schema, hence, a writer can progress from unaware, to aware-and-lost and aware-but-oriented. Improvement is signified as being sensitised to limitations, gaining knowledge of writing processes and the ability to self-regulate.
