Le paludisme reste une des maladies infectieuses les plus importantes au monde. Récemment, le laboratoire de Dr E. Davioud-Charvet a conçu des 3-Benzyl-Ménadiones substituées (benzylMD) comme agents antipaludiques prometteurs. Les études sur le mode d'action ont mis en évidenceque ces molecules déstabilisent l'équilibre redox des érythrocytes infectés en agissant comme agent catalytique redox (redox-Cycler), une stratégie prometteur pour le développement de nouveaux agents antipaludiques. Le travail de thèse présenté a caractérisé l'activité in vitro et le mécanisme d'action de tête de série, la 3-[4-(trifluorométhyl)-Benzyl]-Ménadione 1c, ce qui représente une partie principale du développement des benzylMDs. Une deuxième partie explorait les relations structure-Activité de benzylMD dérivés. Dans l'ensemble, les résultats démontrent l'activité in vitro très prometteuse de la benzylMD 1 cet soutiennent l'amélioration de benzylMDs comme nouveaux candidats-Médicaments antipaludiques.

Malaria is still one of the most important infectious diseases worldwide. Previously, the laboratory of Dr. E. Davioud-Charvet presented the chemical design of very promising antimalarial agents, 3-[substituted-Benzyl]-Menadiones (benzylMD). Studies on the mode of action evidenced that these agents disturb the redox balance of the parasitized erythrocyte by acting as redox-Cyclers - a promising strategy for the development of new antimalarial agents. The presented PhD work characterized the in vitro potency and the mechanism of action of the lead agent, the 3-[4-(trifluoromethyl)benzyl]-Menadione 1 c, which represents an essential part of the lead optimization stage of the benzylMD drug development process. A second part of this work focused on the structure-Activity relationships benzylMD derivatives. Overall, the presented findings demonstrate the promising in vitro potency of lead benzylMD 1c and highly support the further development of benzylMDs as antimalarial drug candidates.

La plasmodione est une benzylmenadione (bMD) qui a été désignée comme inhibiteur subversif de flavoenzymes. Ces enzymes utilisent FAD comme co-facteur et sont impliquées dans de nombreux processus biologiques, et notamment dans le maintien de l’homéostasie redox par les systèmes thiorédoxine et glutathion. La plasmodione présente une forte activité contre P. falciparum in vitro, elle est actif sur tous les stades asexués, préférentiellement sur les stades anneaux, avec une vitesse d’élimination des parasites très rapide. Mon objectif a été de tester sa capacité à bloquer la transmission du parasite murin au moustique et de contribuer à déterminer son mode d’action. Pour ce faire, j’ai dans un premier temps mis au point de nouveaux protocoles. Mes résultats ont démontré que (i) la plasmodione réduit le développement du parasite in vivo et limite la transmission du parasite au moustique en agissant sur tous les stades parasitaires sexués infectieux pour le moustique ; (ii) qu’un dérivé de la même famille, plus soluble, est beaucoup plus efficace, notamment sur les stades sexués et la transmission ; (iii) que lorsqu’ils sont délivrés directement au moustique, il n’y a aucun effet du bleu de méthylène ou de la plasmodione sur la survie des moustique ; (iv) enfin en utilisant des parasites invalidés pour la GR, la GS, la γGCS ou la GluPho, mes résultats suggèrent que le modèle d’action proposé des bMDs est à revoir, et que d’autres flavoenzymes pourraient être la cible.

Plasmodione is a benzylmenadione, synthetized as a flavoenzyme inhibitor. These enzymes use FAD as a cofactor and are involved in numerous biologic processes, including the regulation of the redox equilibrium through thioredoxin and glutathione metabolisms. Plasmodione is very efficient in vitro against all stages of the human parasite P. falciparum, especially on ring stages, and is a fast killer. The aims of my PhD were to monitor the ability of Plasmodione to block parasite transmission to mosquitoes and to characterized its mode of action. For that, I had to implement new protocols. My results showed (i) that Plasmodione decrease both the parasite development in vivo and its transmission to mosquitoes while acting on every sexual stage infectious for the mosquito. (ii) A derivative from the same chemical family, more soluble, is more efficient than Plasmodione, especially on asexual stages and on transmission. (iii) When compounds are directly deliver into mosquitoes, neither methylene blue or Plasmodione have an effect on the survival. (iv) Finally, the use of knock out parasites for the GR, GS, γGCS or GluPho genes suggest that the proposed mode of action of the benzylmenadiones has to be corrected and other flavoenzymes could be targeted rather than GR.

▼ This thesis describes a detailed examination of parameters that control formation of substituted benzalhydantoins in a multistep process from aldehydes and 3-un/substituted hydantoins following modifications of the Wheeler and Hoffman process. The purpose of the study was to develop optimal reaction conditions for a planned, automated parallel synthesis of indoles, with the benzalhydantoins as intermediates. An initial comparative study, based on preparative yields, of the synthesis of benzalhydantoins from the condensation of substituted aldehydes and unsubstituted and 3-substituted hydantoins under the established Wheeler and Hoffman procedure was found imprecise. Consequently, an LC-MS analytical method was developed for reaction monitoring. Investigation of the roles of temperature, reaction time, and aldehyde and hydantoin substituents showed dependence on the latter two features and revealed formation of benzylhydantoin alcohols. The process of 3-allylation of benzalhydantoins was investigated as a model for the final step in an alternative preparation of 3-alkylbenzalhydantoins. Overall, an excess of reagent allyl chloride played an important role in optimizing yields, however yields were limited. A microwave-irradiation method gave more rapid conversions and allowed better comparison of reaction rates. A parallel investigation of the scope of tandem allylation-condensation reactions, which indicated that the alkylation of hydantoin and the condensation of aldehydes with hydantoin occurred simultaneously, was described. 3-Unsubstituted benzalhydantoins were obtained as major products, even after prolonged reaction times, suggesting that condensation of aldehydes with 3-substituted hydantoins was slower than with hydantoin itself. A systematic study of the effect of microwave acceleration on the initial condensation was described, and the revealed benzylhydantoin alcohols were found as major products, rather than benzalhydantoins. The alcohols were studied in one example in the solid state by single crystal x-ray crystallographic analysis and found to form as two racemic diastereoisomers that crystallized together in racemic pairs. Twenty-three synthetic (thio)hydantoins and benzalhydantoins were evaluated for antimicrobial activity against Candida albicans, Malassezia furfur, Escherichia coli and Staphylococcus aureus, by the paper disc diffusion method. 3-Allyl-4'-nitro- and 3-n-butyl-2'-bromo-4',5'-dimethoxybenzalhydantoin showed the most promise as drug leads. Five representative benzalhydantoins examined by x-ray crystallography showed disparate behaviour that could not be related to molecular structure nor physicochemical or biological properties. ▼ The fabrication of on-chip interconnects in the semi-conductor industry employs electrodeposition of copper for fabrication of lines and vertical interconnects (vias) on scales as small as a few tens of nanometers. A significant challenge is to control the distribution of current to assure void-free bottom up filling of these features, a process commonly referred to as superfilling. The presence of a combination of additives on the copper surface regulates the copper deposition rate, and in the proper concentrations produce a high deposition rate at the bottom of features and a reduced deposition rate on the planar wafer surface and upper side-walls of the features. It is known that the adsorption of the additives results in superfilling. However, there are few direct measurements of the coverage of the respective adsorbates and limited information is available for the surface chemistry of additives at various concentrations. The present study is built on the competitive adsorption model in which adsorbed benzyl viologen (BV) or polyethylene glycol (PEG) are displaced progressively from the interface by 3-mercapto-1-propane sulfonic acid (MPS) adsorption during copper electrodeposition. Adsorption of BV and interactions between BV and MPS as well as PEG and MPS were explored by chronoamperometry. During copper electrodeposition with simultaneous BV adsorption, the fractional surface coverage of BV was shown to be potential and temperature dependent. By fitting the data to the Langmuir model, it was shown the adsorption of BV on the copper electrode surface is a spontaneous exothermic process. During copper electrodeposition with displacement of BV by MPS, increased MPS concentration in the electrolyte increases the driving force for MPS adsorption and BV desorption. This potential and temperature dependent displacement was shown to be a spontaneous endothermic process. During copper electrodeposition with displacement of PEG by MPS, more negative potential, higher MPS concentration and higher temperature promotes MPS adsorption and desorption of PEG. ▼ In last few decades many folate analogs have been discovered and several compounds are successfully being used as anticancer agents. Methotrexate (MTX), pemetrexed (PMX), raltitrexed (RTX): are a few examples of classical antifolates that are currently in clinical use. Although these compounds are widely used, all of them show dose limiting toxicity due to their non-specific transport into normal cells as well as malignant cells. Currently, clinically used classical antifolates suffer from this disadvantage because there is no way to selectively transport them into malignant cells. Currently available antifolates are transported via reduced folate career (RFC) as well as Folate receptors (FRs) and/or Proton coupled folate transporter (PCFT). Because RFC is ubiquitous in the human body, the dose limiting toxicity is unavoidable for an antifolate if it is a substrate for RFC. To circumvent this challenge with antifolate chemotherapy, it is necessary to develop agents which are selective for the cancerous tissues. Exploiting the site specificity of FRs and/or the acidic pH specificity of PCFT, the goal of developing targeted antifolate agents could be achieved. This approach of selective transport into tumor cells via FRs and/or PCFT and over RFC targets inhibition of GARFTase dependent salvage pathway for purine biosynthesis inside the tumor. Based on this premise, five 5-substituted pyrrolo[2,3-d]pyrimidines have been designed, synthesized and characterized. Among them four are novel compounds and one is a previously reported compound. The previously reported compound has never been tested for FR/PCFT-GARFTase selectivity before, and therefore was of interest and was synthesized. To investigate the role of side chain aromatic group in the folate analogs, phenyl, fluorophenyl and furan groups have been placed in the linker region. The compounds comprise of different aliphatic linker chain length from 2C to 5C. To test selectivity, FR and PCFT transport have been studied. and GARFTase inhibitory assay will be conducted. ▼ High grade glioma continues to portend a dismal prognosis for patients, despite ongoing, albeit incremental, advances in surgery, irradiation and chemotherapy. This has necessitated an overhaul in the understanding of disease pathogenesis, and the search for a superior model for screening of small molecule therapeutic agents, especially in light of the poor translational outcomes generated from work on traditional, serum-fed, glioma cell lines. The cancer stem cell hypothesis has revolutionised neuro-oncological research, and the discovery of the glioma stem cell (and in particular, its characteristic in vitro gliomasphere-producing qualities) has led to new avenues of thinking, in terms of glioma proliferation, invasiveness and mechanisms of chemo- and radioresistance. Coupled with this has been further characterisation of the genetic constitution of high grade gliomas, which show Pi3k-related aberrations in up to 90% of such lesions, related mainly to EGFR amplification, EGFRvIII expression and PTEN mutation. This study has led to the production of sixteen gliomasphere-producing (or glioma stem cell) clones, from the collection of sixty-six individual specimens, which included lower grade lesions. It was confirmed that this set of stem cell lines exhibited all of the trademark characteristics of a tumour stem cell, characterised by a potential for self-renewal, pluripotency, proliferation, and tumour formation upon xenograft transplantation. Western blot analysis confirmed that thirteen of the sixteen reproducible stem cell clones demonstrated hyperphosphorylated Akt. This led to an investigation of suitable Pi3k inhibitors via a series of in vitro lactate dehydrogenase cytotoxicity assays, which showed that the N10-substituted phenoxazine, Akt inhibitor Akt X, was the only agent which demonstrated effective in vitro cytotoxic potential against glioma stem cells. Interestingly, Akt X was shown to be more efficacious against those glioma stem cell clones which failed to express PTEN protein on Western blot analysis, and this result was found to be statistically significant. Further in vitro LDH cytotoxicity assays revealed glioma stem cells were largely resistant to conventional irradiation and temozolomide, but this was reversed by Akt X, confirming this agent’s potential as a successful stem cell-specific agent in the clinical management of high grade glioma patients. Extensive exome sequencing analysis confirmed the presence of a number of genetic variants in the glioma stem cell clones, whose putative effect and expression concerns signalling pathways not commonly reported in conjunction with gliomagenesis in the past, including the WNT/-catenin and SWH pathways. ▼ Anthraquinone (AQ) has been used in electron transfer studies in DNA. The focus of this dissertation is the synthesis of conjugates between AQ derivatives and 2’-deoxyadenosine (dA), which can be used to induce adenine oxidation in DNA. Different AQ derivatives were attached to dA via ethynyl or ethanyl linkers. If incorporated into DNA, these short linkers should enable regiocontrol for electron transfer from adenine within the DNA duplex structure. The challenge in working with anthraquinone-2’-deoxynucleosides conjugates is that they and their intermediates are insoluble in water and only sparingly soluble in most organic solvents. A strategy used to overcome this problem was the use of either tert-butyldiphenylsilyl (TBDPS) or 4’,4-dimethoxytrityl (DMTr) 5’-protected deoxynucleosides as starting materials. A water-soluble, ethynyl-linked AQ-dA conjugate with a 3’-benzyl hydrogen phosphate was synthesized using DMTr protection. The DMTr group was not stable to the hydrogenation required to make the ethanyl-linked AQ-dA conjugate with 3’-benzyl hydrogen phosphate. Hence the latter was synthesized starting with the TBDPS protecting group. Both of these syntheses were based on the Pd coupling between ethynylanthraquinone and 8-bromodeoxyadenosine derivatives. New conjugates between AQ and A, in which the AQ moieties have been modified with formyl, trifluoroacetyl and methyl ester groups as electron withdrawing substituents were also synthesized. The synthesis of these AQ-dA conjugates was based on Pd coupling between bromoanthraquinone and 8-ethynyldeoxyadenosine derivatives. This route avoided the use of ethynylanthraquinone derivatives that had extremely low solubility and photoinstability. Other anthraquinones with electron withdrawing groups (which should provide enhanced driving force to enable respective AQ derivative to oxidize adenine) were synthesized as models. Cyclic voltammetry showed that the conjugate with the two ester groups and ethynyl linker was the most easily reduced of the derivatives synthesized. Conjugates between AQ and dU were also synthesized. Those conjugates can potentially be used to oxidize guanine or adenine or they can be used as a deep trap for an electron in reduced DNA. ▼ Homoprotocatechuate (HPCA) 2,3-dioxygenase (HPCD) is an Fe(II)-dependent extradiol-cleaving dioxygenase, which oxidatively cleaves the aromatic C(2)-C(3) bond of its catecholic substrate. Here we compare the reactivity of Fe-HPCD with its Mn(II)- and Co(II)-substituted analogues. While Mn-HPCD exhibits steady-state kinetic parameters comparable to those of Fe-HPCD, Co-HPCD exhibits significantly higher K_M^O2 and k_cat values. The high activity of Co-HPCD is surprising, given that cobalt has the highest standard M(III/II) redox potential of the three metals. These kinetic differences and the spectroscopic properties of Co-HPCD have proven to be useful in further exploring the unique O₂ activation mechanism associated with the extradiol dioxygenase family.</DISS_para> <DISS_para>Employing the electron-poor substrate analogue 4-nitrocatechol (4NC), which is expected to slow down the rate of catechol oxidation, we were able to trap and characterize the initial O₂-adduct in the single-turnover reaction of 4-nitrocatechol by Co-HPCD. This intermediate exhibits an S = 1/2 EPR signal typical of low-spin Co(III)−superoxide complexes. Both the formation and decay of the low-spin Co(III)−superoxide intermediate are slow compared to the analogous steps for turnover of 4NC by native high-spin Fe(II)-HPCD, which is likely to remain high-spin upon O₂ binding. Possible effects of the observed spin-state transition upon the rate of O₂ binding and catechol oxidation are discussed.</DISS_para> <DISS_para>Two transient intermediates were detected in the reaction of the [M-HPCD(4XC)] enzyme-substrate complexes (M = Mn or Co, and 4XC = 4-halocatechols, where X = F, Cl, and Br) with O₂. The first intermediate (Co4XlCInt1) exhibited an S = 1/2 EPR signal associated with an organic radical species. Based on the UV-Vis and EPR data, Co^4XCInt1 was assigned to a unique low-spin [Co(III)(4XSQ^*)(hydro)peroxo] species where the semiquinone radical is localized onto C4 of the ring. M^4XCInt2 was observed to have a high-spin metal(II) center by EPR and exhibit intense chromophores similar to the independently synthesized halogenated quinones (4XQ). Based on the UV-Vis and EPR data, M^4XCInt2 is assigned to a [M(II)(4XQ)(hydro)peroxo] species. The M^4XCInt2 species were further characterized by resonance Raman spectroscopy. Resonance enhanced vibrations between 1350-1450 cm^-1 suggest that M4XCInt2 is a metal-semiquinone species, conflicting with the initial assignment of these intermediates as a quinone species. Based on the EPR and resonance Raman data, M^4XCInt2 might be assigned to a [M(II)(SQ^*)O₂^*-] diradical species.

▼ In the present thesis, an effort has been undertaken in order to understand the photochemical reactivity of molecules under the influence of uv light. The ab-initio CASSCF method was used throughout this research, since it is currently the most appropriate method for the description of excited states and reaction paths providing very good equilibrium geometries both in the ground and excited states, and depicting at the same time the topography of the potential energy surfaces (PES) very satisfactorily. Multiconfigurational second-order perturbation theory (CASPT2 or CASMP2) is used for the accurate calculation of energies such as activation energies and heats of reactions. This research consists of two parts. o In the first part we study the photodissociation of the C-Si bond in the first excited triplet (T1) state of substituted allyl silanes of the general form CH2=CHC(R1R2)SiH3, where R1, R2 = H, Me, Et. It is found that the photodissociation reaction, CH2=CHC(R1R2)SiH3 + hν > CH2=CHC(•)R1R2 + •SiH3 produces free radicals in their ground states and that a linear relationship of the form ΔHr = b•D0 + a exists between the heat (ΔHr) of the above photoreaction and the bond dissociation energy, BDE (D0) of the C-Si bond in the S0 state. The parameters a and b are constants. As the number and size of the substituents (R1, R2) increases, the BDE and the heat of the bond dissociation reaction (ΔHr) in the T1 state were found to decrease, and thus the reaction becomes more exothermic. This suggests a weakening of the C-Si bond with increasing substitution. Calculations of the minimal energy path for the C-Si bond dissociation in the T1 state predict also a useful Evans-Polanyi linear relationship ( Ea = b1• ΔHr +a1) between activation energy Ea, which is an excited state property and D0, a ground state property. This enables us to estimate important reactivity parameters such as activation energy and rate constant of an excited state, based on an easily accessible ground state property such as the BDE. The kinetics of the photoreactions in the T1 state is also examined. o In the second part we examine the excited states S1, T1 and the photochemistry of benzyl silane (PhCH2SiH3). This molecule is considered to be the prototype aromatic organosilicon compound, and therefore conclusions based on its photochemistry concern the whole homologous series derived from it. In particular, it was found that in the T1 state the molecule dissociates exothermically (-11.07 kcal/mol) giving benzyl (PhCH2•) and silyl (•SiH3) radicals in their ground states. The S1 excited singlet potential energy surface was explored in great depth and the results show that benzyl silane has a very rich photochemistry. ▼ The formation of nitrogen-containing molecules is of great importance in synthetic organic chemistry owing to the abundance of alkaloids that exhibit biological activity. While the first two projects deal with expanding the synthetic tools available for synthesis of such compounds, the final project focuses on using existing methodology to create and test novel potential clinical agents. The use of (2-azaallyl)nitriles in double allylation reactions with allylmagnesium bromide to generate N, N-bis(3-butenyl)amines is described. The (2-azaallyl)nitriles function as synthetic equivalents of amine alpha,alpha'-dications, also called 2-azaallyl dications. This chemistry allows for the expeditious construction of highly substituted amines, resulting in the formation of two new carbon-carbon bonds adjacent to the nitrogen atom. The doubly allylated products are converted to 2,3,6,7-tetrahydroazepines by ring-closing metathesis. Many biologically-active natural products contain the azepine ring, so facile methods for its formation provide valuable synthetic tools. Also discussed is the formation of pyrrolizidines from N-(tri- n-butylstannyl)methylphthalimides. Through nucleophilic addition of an organometallic reagent, the phthalimides could be converted to cyclic carbinol amides, which were shown to undergo ionization and destannylation under acidic conditions to generate azomethine ylides. Inclusion of a suitable dipolarophile resulted in capture of the azomethine ylide to provide the pyrrolizidine. Various substituents on the phthalimide were tolerated by this methodology, and stabilized succinimide derivatives also underwent successful cycloadditions. Given the large number of biologically-active natural products that possess the pyrrolizidine ring structure, this chemistry could find wide synthetic application. Finally, the preparation of 2-(2,5-dihydropyrrol-1-yl)-1,4-quinones by the coupling of quinones with pyrrolines is discussed. The 2-(2,5-dihydropyrrol-1-y1)-1,4-quinones were designed to be masked alkylating agents, revealed upon exposure to light. The pyrroline of the 2-(2,5-dihydropyrrol-1-yl)-1,4-quinones is relatively unreactive toward nucleophiles, but upon photolysis, an internal oxidation/reduction reaction occurs, transforming the quinone into a hydroquinone and the pyrroline into a pyrrole. The newly-formed pyrrole is highly electrophilic and readily undergoes an alkylation reaction. Eventually, the presence of oxygen may reoxidize the hydroquinone to a quinone. The design of such photoactivatable alkylating agents is a worthwhile endeavor because it provides a handle for control of the alkyation, thereby making it more attractive as a potential clinical agent. ▼ Reaction of (Cp*IrHCl)2 (Cp* = η5-C5Me5) with 2-methylthiophene and 2,5-dimethylthiophene at 120°C in the presence of H2 results in the cleavage of the thiophene carbon-sulfur bond(s). In both cases the thiophenes are ring opened and hydrogenated resulting in dinuclear Ir complexes with bridging thiolates. The primary product in the reaction involving 2.5-dimethylthiophene is [Cp*IrCl]2(μ-H)(μ-S-2-hexyl). This product has been characterized and is present in diastereomeric pairs. Reaction with 2-methylthiophene produces a complex mixture consisting of five products. The product distribution consists of mono and di-substituted bridging thiolate products three of which have been structurally characterized by single-crystal X-ray diffraction. Independent synthesis of each of these products has been performed, and characterization of the reaction mixture has been accomplished by 1H and 13C NMR spectroscopies, as well as by ESI-MS and elemental analysis. Reaction with 2-acetylthiophene showed very similar reactivity; an X-ray structure confirmed the nature of the diastereomeric pairs present. The processes of C–C and C–S bond cleavage of 2-cyanothiophene have been studied with the homogeneous organometallic compound [Ni(dippe)H]2. Reaction at room temperature resulted in cleavage of the nitrile substituted C–S bond to give the Ni-metallacycle complex (dippe)Ni(K2-S,C–SCH=CHCH=C(CN)), which has been fully characterized by NMR spectroscopy and X-ray diffraction. Conversion to the C–CN cleavage product (dippe)Ni(CN)(2-thiophenyl) occurred when the solution was heated to 85°C. On closer inspection four other intermediates were observed by 31P NMR spectroscopy at -60°C. Structures for the intermediates were elucidated through a combination of independent synthesis, theoretical calculations, chemical characterization, and experimental precedent. A kinetic product (dippe)Ni(K2-S,C-SC(CN)=CHCH=CH) was formed from cleavage of the non-substituted C–S bond, as well as a Ni(0) η2-nitrile intermediate (dippe)Ni(η2-C,N-2-cyanothiophene), and a dinuclear mixed Ni(0)-Ni(II) product. The unusual lithium coordinated complex [(dippe)Rh(μ-H)]2[LiBEt3H] was synthesized and characterized by NMR spectroscopy and X-ray crystallography. The [(dippe)Rh(μ-H)]2 and LiBEt3H moieties are bound together through lithium. Bonding interactions occur between the lithium and the metal hydrides, as well as the lithium and the B-H bond of BEt3H. Formation of the complex is highly solvent dependent and may be part of a general bonding phenomenon between borohydrides and metal hydride complexes. The rhodium dimer [Rh(dippe)(μ-H)]2 reacted with 4-methyldibenzothiophene to form the C–S cleavage product [Rh2(dippe)2(μ-S-MeC12H8)(μ-H)], which has been fully characterized by NMR spectroscopy (1H, 31P, and 13C), elemental analysis, APCI-MS, and X-ray diffraction. ▼ A variety of calixarene analogues involving heteroatoms as the bridging units have been reported because the substitution of the carbon bridges with heteroatoms can impart novel properties and influence the conformer of the macrocycle. However, the chemistry of replacing the bridging methylene units of calixarenes derived from p-tert-butylphenol with free amino groups to prepare azacalixarene macrocycles is still in its infancy. Nitrogen bridges can alter the conformational properties of the macrocyclic array through changes in bond lengths and angles associated with the nitrogen heteroatom, which results in a cone conformation. Additionally, the nitrogen heteroatom also provides additional binding sites to the macrocycle and can participate in hydrogen bonding along the endo rim which may also influence conformational properties. Furthermore, the nitrogen heteroatom opens up new avenues to further functionalize the bridge position through amine alkylation or acylation chemistry. The synthetic methodology was developed for hexamethyl triazacalix[6]arene by preparing a functionalized linear oligomer followed by cyclization over seven reactions steps in 32.1% overall yield. Of the eight conformers possible for the hexamethyl triazacalix[6]arene, in solution the cone conformer is preferred due to the bifurcated hydrogen bonding by the amine-methoxy functional groups along the endo rim. Additionally, the cone-to-cone ring inversion can be observed by temperature-dependent 1H-NMR spectroscopy since the temperature dependent signal is the result of the conformational flexibility of calixarenes and indicates that the protons are exchanging environments rapidly on the NMR time scale at high temperature but are frozen at lower temperatures. 1H NMR spectra was collected at various temperatures beginning at room temperature (298 K) and stopping at -57 ???C (216 K). The rate of exchange for a given temperature was determined by the iterative parameter refinement of the line shape (1000 cycles and with a 92+ % overlap) and used in an Erying plot to determine the thermodynamic parameters. The enthalpy of activation (???H???) was determined to be 8.95 kcal/mol and the entropy of activation (???S???) was determined to be -4.3 cal/mol*K for the cone-to-cone inversion barrier. The Gibbs free energy of activation (???G???) for hexamethyl triazacalix[6]arene was calculated to 9.9 kcal/mol at coalescence. In comparison, the activation barrier for cone-to-cone inversion for the full carbon-bridged calix[6]arene is estimated to be below 9.3 kcal/mol. Advisors/Committee Members: Spence, John.
