Micro-scale processing (MSP) techniques are miniaturized version of upstream and downstream conventional unit operations that are designed to accelerate the pace of bioprocess design and development. Previous ‘dead end’ filtration studies have demonstrated the usefulness of this concept for membrane filtration processes. However, these experiments were performed without stirring which is the most common strategy to control the effects of concentration polarization and fouling on filtration performance. In this work, the pressure-driven stirred conditions of a conventional stirred-cell module were integrated with a 96-well filter plate to develop a high throughput technique called ‘stirred-well filtration’ (SWF). The design allowed for up to eight constant flux filtration experiments to be conducted at once using a multi-rack programmable syringe pump and a magnetic lateral tumble stirrer. An array of pressure transducers was used to monitor the transmembrane pressure (TMP) in each well. The protein sieving behavior and fouling propensity of Omega™ ultrafiltration membranes were assessed via a combination of hydraulic permeability measurements and protein sieving tests in constant filtrate flux mode. The TMP profile during filtration of bovine serum albumin (BSA) solution was strongly dependent on the stirring conditions – for example the maximum TMP in the stirred wells were an average of 7.5, 3.8, and 2.6 times lower than those in the unstirred wells at filtrate fluxes of 12, 36, and 60 LMH (5, 15, and 25 μL/min) respectively. The consistency of the data across different wells for the same stirring condition was very good. To demonstrate the effectiveness of the SWF technique, the eight tests for a simple 2² factorial design-of-experiments (DOE) test with duplicates was run to evaluate the effect of solution pH and salt concentration on protein filtration. The combination of SWF with statistical methods such as DOE is shown to be an effective strategy for high-throughput optimization of membrane filtration processes.

Membrane filtration processes are widely utilized across different industrial sectors for biological and environmental separations. Examples of the former are sterile filtration and protein fractionation via microfiltration (MF) and ultrafiltration (UF) while drinking water treatment, tertiary treatment of wastewater, water reuse and desalination via MF, UF, nanofiltration (NF) and reverse-osmosis (RO) are examples of the latter. A common misconception is that the performance of membrane separation is solely dependent on the membrane pore size, whereas a multitude of parameters including solution conditions, solute concentration, presence of specific ions, hydrodynamic conditions, membrane structure and surface properties can significantly influence the separation performance and the membrane’s fouling propensity. The conventional approach for studying filtration performance is to use a single lab- or pilot-scale module and perform numerous experiments in a sequential manner which is both time-consuming and requires large amounts of material. Alternatively, high-throughput (HT) techniques, defined as the miniaturized version of conventional unit operations which allow for multiple experiments to be run in parallel and require a small amount of sample, can be employed. There is a growing interest in the use of HT techniques to speed up the testing and optimization of membrane-based separations. In this work, different HT screening approaches are developed and utilized for the evaluation and optimization of filtration performance using flat-sheet and hollow-fiber (HF) membranes used in biological and environmental separations. The effects of various process factors were evaluated on the separation of different biomolecules by combining a HT filtration method using flat-sheet UF membranes and design-of-experiments methods. Additionally, a novel HT platform was introduced for multi-modal (constant transmembrane pressure vs. constant flux) testing of flat-sheet membranes used in bio-separations. Furthermore, the first-ever HT modules for parallel testing of HF membranes were developed for rapid fouling tests as well as extended filtration evaluation experiments. The usefulness of the modules was demonstrated by evaluating the filtration performance of different foulants under various operating conditions as well as running surface modification experiments. The techniques described herein can be employed for rapid determination of the optimal combination of conditions that result in the best filtration performance for different membrane separation applications and thus eliminate the need to perform numerous conventional lab-scale tests. Overall, more than 250 filtration tests and 350 hydraulic permeability measurements were performed and analyzed using the HT platforms developed in this thesis.

▼ Collector well designs are necessary to maximize groundwater uptake and riverbank filtration without negatively impacting an aquifer. Unfortunately, there is a lack of information and research regarding the implementation of collector well design parameters. In the past, collector well installation was too costly, but recent advances in well technology have made collector wells more cost effective. This research will contribute a set of guidelines to optimize riverbank filtration and groundwater remediation. This study models the hydrodynamics surrounding collector well configurations in riverbank filtration and groundwater remediation. Visual Modflow® was utilized to run a variety of numerical models to test four areas: flux along the laterals of a collector well, collector well interactions with a river, collector well yield, and collector well remediation capability. The two design parameters investigated were lateral length (25 m, 50 m, and 100 m) and number of laterals (3 and 4). The lateral flux tests confirm flux increases towards the terminal end of each lateral and pumping rate is the controlling factor in flux amount obtained along the laterals. The analysis of the flux-river interaction shows the main factor in determining flux amount is the initial river geometry, followed by the pumping rate, regional background flow, and collector well design, respectively. The models suggest that the 4-lateral collector well design is more effective than the 3-lateral design and in addition, 100 meter length laterals provide the highest amount of yield with the least amount of drawdown. The remediation tests investigate the application of vertical well equations to evaluate collector well designs in two areas: minimum pumping rate to capture line source of particles and first arrival time of particles. The remediation models show 100 meter length laterals provide both the lowest pumping rate and the highest residence time with the surrounding aquifer for maximum remediation. Ultimately, these models provide basic design guidelines and explain which designs are most effective, depending on the collector well purpose. ▼ Die Redoxbedingungen und insbesondere der Sauerstoffgehalt im Grundwasser haben nicht nur einen wesentlichen Einfluss auf die Qualität des daraus gewonnenen Trinkwassers, sie beeinflussen auch in erheblichem Maße die Leistungsfähigkeit und Lebenserwartung der Förderbrunnen. Daher sind ein prozessbasiertes Verständnis und eine quantitative Analyse der in Grundwasserleitern stattfindenden Sauerstoffströme grundlegend, um neben dem Rückhalt von Schadstoffen, auch die Wahrscheinlichkeit von Brunnenalterungsprozessen zu ermitteln und vorherzusagen. Gerade die Brunnenalterung ist für die Betreiber von Brunnenanlagen ein zentrales Thema und deshalb von großer Bedeutung für Praxis und Forschung. Unlösliche Eisenverbindungen, auch bekannt als Verockerung, vermindern durch ihre Ablagerung in den Brunnenfiltern, aber auch im angrenzenden Grundwasserleiter, die Produktivität der Brunnen in erheblichem Maße. Die notwendige Instandhaltung und der Neubau von Brunnenanlagen verursachen erhebliche Kosten für die Betreiber. Produktivitätsabnahmen durch Verockerungen werden hauptsächlich bei solchen Brunnen beobachtet, die Grundwasser aus Grundwasserleitern mit einer ausgeprägten Redoxzonierung fördern. Mehrere wissenschaftliche Studien haben bereits ein Zusammenspiel von hydrochemischen und mikrobiologischen Prozessen als Hauptursache für die Entstehung von Eisenausfällungen in Brunnen identifiziert. Diese bilden sich, sobald Sauerstoff und Eisen in gelöster Form im Wasser vorhanden sind. Beide Stoffe treten gemeinsam im Wasser auf, wenn sich Grundwässer mit unterschiedlichen Redoxbedingungen mischen. Es ist nachgewiesen, dass die Entnahme von Grundwasser mittels Brunnen solche Mischungsprozesse verstärkt auftreten lässt. Insbesondere häufige und ausgeprägte Schwankungen der Grundwasseroberfläche und Maßnahmen zur künstlichen Grundwasseranreicherung können größere Mengen an Sauerstoff ins Grundwasser eintragen. Die Auswirkungen unterschiedlicher Grundwasserbewirtschaftungsstrategien auf die Quellen und den Transport von Sauerstoff im Grundwasser wurden jedoch bisher nicht explizit betrachtet. In der hier vorgestellten Arbeit werden die Sauerstoffströme ausgehend von statistischen, modellbasierten und labortechnischen Verfahren sowie im Geländemaßstab beschrieben und quantifiziert. Die Bedeutung der Sauerstoffströme für die Leistungsentwicklung von Brunnen wird basierend auf den Ergebnissen für verschiedene Bewirtschaftungsszenarien und verschiedene hydrogeologische Randbedingungen bewertet. Die Prozesse wurden beispielhaft für die quartären Grundwasserleiter Berlins untersucht. Diese bilden die wichtigste Trinkwasserressource für die Bundeshauptstadt. Die Analyse von Stamm- und Betriebsdaten der Brunnen, sowie von Instandhaltungsdaten zeigt die Anfälligkeit der Brunnen für Alterungsprozesse und verdeutlicht wie wichtig detailliertere Untersuchungen zu diesem Thema sind. Cette thèse présente des calculs d’algèbre homologique dans la catégorie des modules instables. Dans le premier chapitre, on rappelle des généralités sur cette catégorie, parmi elles le théorème de la caractérisation de la filtration de Krull grâce au foncteur de Lannes. En utilisant la théorie des représentations, Schwartz a démontré ce théorème dans les années 90s. Au cours du chapitre, on donne une preuve élémentaire de ce théorème. Une méthode efficace pour construire des résolutions à partir des suites exactes dans cette catégorie y est introduite ainsi. On l’appelle pseudo-hyper résolution. La catégorie des foncteurs polynomiaux stricts homogènes de degré fini a été reliée avec la catégorie des modules instables d’après les travaux de Hai en 2010. Le foncteur connectant ces deux catégories est nommé d’après lui. On montre dans le deuxième chapitre que ce foncteur est pleinement fidèle, permettant de considérer la catégorie des foncteurs polynomiaux stricts homogènes de degré fini comme une sous-catégorie pleine de la catégorie des modules instables. Le chapitre 3 est consacré pour étudier les résolutions injectives minimales des cohomologies de sphères. On montre que le morphisme à la Bockstein permet de déterminer une grande partie de ces résolutions. Dans le dernier chapitre, on étudie l’action de la torsion de Frobenius sur les groupes d’extensions. Ceci amène à étudier la résolution injective minimale du module F(1), étant générateur projectif monogène engendré par un élément de degré 1. De l’information partielle de la partie nilpotente de cette résolution est donnée à la fin du chapitre.

The aim of this work is to study injective resolutions of certain objects in the category U of unstable modules. More precisely, we concentrate on the minimal injective resolution of the module F(1) as well as ones of the cohomology of spheres ΣnF2.For that purpose, the pseudo-hyper resolution is introduced, allowing to construct an explicit resolution of an unstable module from an acyclic sequence admitting this module as its first homology. This construction and the hyper resolution do look alike but are not identical. In our situation, we consider the sequence without splitting it into short exact sequences to avoid unnecessary concerns about the differentials. Placing the resolutions of each term in the sequence together, we obtain a fake double complex. Fortunately, the injectivity of modules in this double complex allow us to insert enough differentials that make the total sequence a complex. This complex is indeed the resolution for the considered module. To deal with the particular cases of ΣnF2, the pseudo-hyper resolution wil be translated into the algorithm BG. Together with this procedure, the Bockstein sequence gives a simple description on a large part of the minimal injective resolution of ΣnF2.The minimal injective resolution of F(1) is too much to deal with. Riverbank filtration (RBF) is an accepted method of treatment for water supply from rivers which markedly improves the source water quality. Pumping water from wells adjacent to water body induces surface water to infiltrate through the aquifer attenuating contaminants present in the source water. This study used field data from an operating RBF system located along the Rhine River in Düsseldorf, Germany, and carried out focused research on the fate of dissolved organic carbon and on transport of E. coli. Furthermore, a novel application of RBF and aquifer storage and recovery (ASR) was considered in a study to address seasonal water scarcity in the Albany region of the Georgia in US. A reactive transport model (PHT3D) was developed to study the fate of dissolved organic carbon incorporating transient boundary conditions. Modeling residence times showed that high floods in the river reduced the travel time to the RBF well to 8 days, while low flows increased it to about 60 days. Aerobic processes with some partial denitrification occured in the aquifer. The temporal changes in the breakthrough of dissolved oxygen were best reproduced when the temperature dependency of the biogeochemical processes was explicitly considered. The results showed that seasonal temperature changes superimposed by changes in residence time strongly affected the extent of redox reactions along the flow path. Microbial transport is modeled in groundwater using the advection-dispersion transport equation adding on the processes of attachment, detachment and inactivation processes. The rates defined by these processes are velocity dependent and cannot be used directly for a transient RBF model. An approach to handle such transient conditions is developed using colloid filtration theory and the concepts of single collector contact efficiency and verified on a benchmark 1-D study. The model is extended to the Rhine River RBF system. The transport of E-coli and coliforms was successfully modeled. The RBF-ASR study showed the viability of obtaining excess river flows through RBF and storing it a deeper aquifer to be recovered later. Geochemical changes were also investigated. The overall study contributes to our understanding of the various processes involved in RBF and subsurface flows.

▼ A better understanding and control of internal combustion engine pollutants require more insightful investigation of gasoline oxidation chemistry. An oxidation study has been done on n-heptane, iso-octane, their binary mixtures (Primary Reference Fuel, (PRF)), and nine hydrocarbon mixtures which represent the second generation of gasoline surrogates (multi-component surrogates). This study aims to develop a better understanding of the combustion reaction by studying the oxidation reaction of different fuels inside a jet-stirred reactor and numerically simulating the reaction using different models under the following conditions: pressure 1 bar, temperature 500-1050K, residence time 1.0 and 2.0s, and two fuel-to-oxygen ratios (ϕ=0.5 and 1.0). Intermediate and product species mole fractions versus temperature profiles were experimentally measured using a GC (gas chromatograph). The experiment was performed within the high and low-temperature regions, where the high-temperature oxidation showed similar behavior for different compositions but the low-temperature oxidation showed significant dependence on the composition of the surrogates. Additionally, the effect of octane number on oxidation chemistry has been investigated and it was found that the low octane number surrogates were more reactive than high octane number surrogates during the low temperature regime. Furthermore, Kinetic analysis was conducted to provide insightful understanding of different factors of fuel reactivity. In addition, the pyrolysis of two TPRF, (Toluene primary reference fuels) mixtures (TPRF70 and TPRF97.5), representing low octane (research octane number 70) and high octane (research octane number 97.5) gasoline, was also studied in jet-stirred reactor coupled with gas chromatography (GC) analysis to investigate the formation of soot and polycyclic aromatic hydrocarbons (PAH) formation. Cette thèse porte sur les modèles à densité pour les temps de défaut et leur application au risque de crédit et de contrepartie. La première partie est une contribution théorique à l'étude des projections sur différentes filtrations de la densité de Radon-Nikodym, sous la forme d'exponentielle de Doléans-Dade, intervenant lors de changements de mesure. Le résultat principal est la caractérisation des changements de mesure qui préservent l'immersion, obtenue par application de nos formules de projection. La deuxième partie a pour objet une dynamisation informationnelle du modèle de copule gaussienne statique appliqué à un portefeuille de crédit, pouvant être vue comme un modèle à densité permettant de traîter de la couverture des CDO par CDS ou bien du risque de contrepartie sur les dérivés de crédit. Les principales contributions sont l'introduction de la perspective dynamique , qui permet de donner une justification théorique aux bump-sensibilités de copule gaussienne utilisées par les praticiens, et l'application aux calculs de CVA sur un CDS.

This thesis is about density models of default times and applications to credit and counterparty risk. The rest part is a theoretical contribution to the study of projections on different filtrations of the Radon-Nikodym density of a measure change. The main result is a characterization of the measure changes preserving immersion in a density setup, obtained by application of our projection formulas. The second part is about an informational dynamization of the Gaussian copula model of portfolio credit risk, resulting in a density model of default times suitable to del with dynamic issues such as hedging of CDO through CDS or counterparty risk on credit derivatives. Here the main contributions are the introduction of the dynamic perspective, which allows one to give a theoretical justification to the Gaussian copula bump sensitivities used by practioners, and the application to CVA computations on CDS.

Cette thèse se compose de quatre parties indépendantes. Le fil conducteur de celle-ci est le grossissement de filtration. Dans la première partie, nous présentons des résultats classiques de grossissement de filtration en temps discret. Nous étudions quelques exemples dans le cadre du grossissement initial de filtration. Dans le cadre du grossissement progressif nous donnons des conditions pour obtenir la propriété d'immersion des martingales. Nous donnons également diverses caractérisations des pseudo temps d'arrêt et nous énonçons des propriétés pour les temps honnêtes.Dans la deuxième partie, nous nous intéressons à la détermination du prix de produits à annuités variables dans le cadre de l'assurance vie. Pour cela nous considérons deux modèles, dans ces deux modèles nous considérons que le marché est incomplet et nous adoptons l'approche par prix d'indifférence. Dans le premier modèle nous supposons que l'assuré procède à des retraits aléatoires et nous calculons la prime d'indifférence par des méthodes standards en contrôle stochastique. Nous sommes conduits à résoudre des équations différentielles stochastiques rétrogrades (EDSR) avec un saut. Nous fournissons un théorème de vérification et nous donnons les stratégies optimales associées à nos problèmes de contrôle. De ceux-ci, nous tirons une méthode de calcul pour obtenir la prime d'indifférence. Dans le second modèle nous proposons la même approche que dans le premier modèle mais nous supposons que l'assuré effectue des retraits qui correspondent au pire cas pour l'assureur. Nous sommes alors amenés à traiter un problème de max-min.Dans la troisième partie, nous étudions la relation des solutions d'EDSR dans deux filtrations différentes. Nous étudions alors la relation entre ces deux solutions. Nous appliquons ces résultats pour obtenir le prix d'indifférence dans les deux filtrations, c'est-à-dire le prix auquel un agent aurait le même niveau d'utilité attendue en utilisant des informations supplémentaires.Dans la quatrième partie, nous considérons des équations différentielles stochastiques rétrogrades avancées (EDSRAs) avec un saut. Nous étudions l'existence et l'unicité d'une solution à ces EDSRAs. Pour cela nous utilisons la décomposition des processus à sauts liée au grossissement progressif de filtration pour nous ramener à l'étude d'EDSRAs browniennes avant et après le temps de saut.

▼ Today’s oil and gas industry is faced with several geographic and economic challenges that have significantly increased the pressure on companies engaged in oil and gas exploration and production. Technical as well as economic challenges like the highly volatile crude oil prices, global competition for depleting resources and pressure from shareholders for return on investment are threatening to the industry. In the quest to address these challenges, operators are continuously seeking advanced technology that could increase production, improve recovery, and minimize cost. Although advanced technology such as 3D and 4D seismic downhole sensors have significantly improved the amount of accessible realtime information, the amount of data is often massive and too complex to accurately analyze. Within the past decade, significant advances in drilling and completion techniques have been made to enable more active monitoring and control of production wells. Smart well technology, also known as Intelligent Well Completions (IWC), is one of such technologies that integrates permanent downhole sensors with surface-controlled downhole flow control valves, enabling operators to monitor, evaluate, and actively manage production (or injection) in real time. All of this is achieved without any well interventions, thus completely eliminating the risk and economic losses associated with well intervention. A comprehensive review of smart well technology, as well as real-world case studies will be presented. A case study simulation is performed to evaluate the additional value that is derived by adopting smart well technology. The simulation results clearly indicate that adopting smart well technology significantly reduced field water cut, accelerated the productions time and improved the Net Present Value (NPV) of the project. Finally, a workflow is presented which can be used to assess to applicability of a given field with multiple producing wells. ▼ Crystallization is a key unit operation in the fine chemical and pharmaceutical industries, many of which employ batch stirred vessels for crystallization. Although using stirred vessels for crystallization has advantages such as better mixing and faster cooling, one of the disadvantages is that due to the presence of mechanical parts in the vessel such as baffles, impeller etc., crystals break up while stirring and generate unwanted secondary nucleation. This process contributes to a wide crystal size distribution with a smaller than desired mean crystal size. For studying crystal breakage phenomenon, experimentalists choose to use nonsolvents for crystal breakage experiments to isolate breakage from simultaneously occurring phenomena such as Ostwald-ripening, aging and agglomeration. Although performing experiments in non-solvents eliminates other phenomena and helps isolate breakage, the results can not always be correlated to saturated solutions due to density and viscosity differences between the two conditions. In this research, the effects of Ostwald ripening, aging and agglomeration on the crystal size and shape distributions are quantitatively measured. Micro and macro scale experiments were performed in both non-solvents and saturated solutions and the results were compared to determine the effects. Both in situ focused beam reflectance method (FBRM) and off-line analyses were performed to characterize the crystal size distributions. Results from experiments show that there is significant difference between the breakage behavior of crystals in non-solvents and in saturated solutions, mplying significant impacts of Ostwald ripening, aging, agglomeration and dissolution in saturated solutions. Calculations using Zwietering correlation also show that the difference between the viscosities and densities in the two systems may also be a contributing factor to the difference in the breakage profiles. It was also found that growth rates of crystals can differ when they are subjected to stress and strain. In macroscale experiments, dissolution was found to have a significant impact on the crystal size distribution. Abrasion was found to be the dominating fracture mechanism for most systems. Extent of breakage and morphological changes were found to be dependent on stirring rates, suspension density, shape and hardness of crystals and the type of system.
