▼ Selenium (Se) is an essential micronutrient of sheep. Supplementation is especially important in young lambs in order to prevent Se-deficiency signs. In the United States, the FDA regulates Se supplementation to ruminant diets at a level of 0.3 mg/kg Se (as fed basis). Questions still exist regarding what chemical form of Se is the most bioavailable supplement and what are the best supplementation rates for optimal productivity. Three interrelated experiments from the same study were conducted 1) to evaluate the effect of Se source and supplementation rate in ewes on Se status of their lambs at birth, 2) to examine the effect of Se source and supplementation rate on IgG concentrations in ewe colostrum and lamb serum, and 3) to evaluate the effects of Se source and supplementation rate on ewe reproductive performance and subsequent vitality and growth performance of their lambs.
For these experiments, 240 ewes were divided into 8 treatment groups and drenched weekly (at an amount equal to their summed daily intake) for one year, including during gestation and early lactation, with no Se (deficient); at recommended levels (0.3 mg/kg) with inorganic sodium selenite, sodium selenate, or organic Se-yeast (Se-Y); or at supranutritional levels (0.9 and 1.5 mg/kg) with sodium selenite or Se yeast. In the third experiment, 88 ewes continued the study into year two for an additional 28 weeks. Year two treatments included no Se (deficient) ewes (n = 25); ewes supplemented at recommended levels (0.3 mg/kg) with organic Se-Y (n = 20); ewes supplemented at supranutritional levels (0.9 and 1.5 mg/kg) with organic Se-Y (n = 18 and 27, respectively).
In the first experiment, which assessed the effects of Se source and supplementation rate in ewes on Se status of their offspring, we found that Se administered by weekly drenching of ewes during gestation and early lactation was effective at increasing Se concentrations in ewe colostrum and milk at 30 days in milk (DIM), and in improving the Se status of lambs (whole-blood and serum Se at birth, and skeletal-muscle Se at 14 days of age) (all P < 0.001).
Selenium concentrations in lacteal secretions and lambs were higher in ewes drenched with Se yeast compared to ewes drenched with inorganic Se sources (P < 0.01).
Selenium concentrations in lacteal secretions and in lambs increased linearly with supranutritional concentrations of Se yeast (P < 0.001), whereas Se concentrations did not differ in ewes drenched with 0.9 or 1.5 mg/kg of inorganic sodium selenite (P > 0.05). In summary, weekly oral drenching of ewes with Se yeast during gestation and early lactation was found to be an effective method for improving Se status of their newborn lambs.

▼ Studies were conducted to investigate the metabolism of
selenium (Se) , and the influences of Se on the metabolism of cadmium (Cd) and inorganic mercury (Hg) in rats, and in ram semen in
vitro. Se-deficient (-Se) or Se-adequate (+Se) rats were injected intraperitoneally with either ¹⁰⁹CdCl₂ (5 μCi/rat) or ²⁰³HgN0₃ (10 μCi/rat). Semen ejaculates from yearling Suffolk rams were used for the in vitro studies. Whole-body retention of Cd and Hg in rats was significantly
(P < 0.05) increased by Se. However, regardless of the Se status,
the predominant route of Cd and Hg excretion was feces. Data on
whole tissue Cd retention for both -Se and +Se rats gave the
following order of decreasing tissue Cd levels: liver > kidney >
testis > epididymis > seminal vesicles > prostate > brain. Hg
distribution in tissues followed a similar order except that Hg concentrations were highest in kidney. Se caused a significant
redistribution of Cd and Hg among the cytosolic proteins in some
tissues (e.g., testis) but not in others like brain and prostate
gland. Such diversions were usually from low (10,000 - 30,000)
molecular weight (MW) proteins to higher (>70,000) MW ones.
Ram sperm incorporated small amounts of Cd and Hg in vitro.
A non-thionein metal-binding protein (MW = 10,000) was purified
from the spermatozoa, and found to contain low amounts of cysteine
and some aromatic amino acids (phenylalanine and tyrosine). Evidence was obtained which suggests that spermatozoa from rats and
rams are capable of converting inorganic Se (as selenite) into
selenoamino acids both in vivo and in vi tro. Amino acid analyses
of sperm hydrolysates showed that selenocystine and selenomethio
nine were respectively the major chemical forms of Se in rat and
ram spermatozoa. Cd and Hg concentrations ranging from 10⁻⁶ to 10⁻² M were
shown to be injurious to ram sperm in vitro as indicated by the
depressed motility and reduced oxygen uptake. Equimolar concentrations of Se as selenite, selenocystine or selenomethionine
counteracted the toxicity of Cd or Hg at low concentrations (10⁻⁵
to 10⁻⁶ M). Incubation of ram spermatozoa with concentrations of
Se ranging from 0.08 to 2.0 ppm markedly improved sperm motility
and oxygen consumption.

▼ This study was conducted to investigate the distribution
of glutathione peroxidase (GSH-Px) and
selenium in
semen of the boar, bull, ram and stallion, and in the reproductive
tissues of the boar, bull and ram. The effect
of repeated noncryoprotective freeze-thawing on GSH-Px
activity was also studied. Both hydrogen peroxide (H₂0₂)
and tetra-butyl hydroperoxide (t-butyl 0₂) were used as
substrates for the enzyme.
Most of the GSH-Px activity in semen of the four species
examined was associated with seminal plasma. GSH-Px
activities in the spermatozoa of these species were comparatively
very low. There were species and tissue
differences in the distribution of GSH-Px in reproductive
tissues. Determined per mg protein, GSH-Px activity was
highest in the testis, whereas the Cowper's gland and
ampulla showed the least enzyme activity. In general,
GSH-Px levels in the reproductive tissues of the bull were higher than, or comparable to, those of the ram. The
reproductive tissues of the boar showed the least GSH-Px
activity.
The GSH-Px determined in this study showed a similar
magnitude of response to both H₂0₂ and t-butyl 0₂ and had
a high correlation with tissue and fluid
selenium levels.
This demonstrates that the GSH-Px found in semen and
reproductive tissues of the boar, bull and ram is a
selenium-dependent enzyme.
Repeated freezing (at -21°C) and thawing, in the
absence of appropriate buffers, led to a decline in seminal
plasma and reproductive tissue GSH-Px activities.
Selenium distribution in semen and reproductive
tissues differed among the species studies. On the basis
of
selenium concentration per billion sperm cells, boar
spermatozoa had the highest
selenium concentration followed
by those of the stallion, bull and ram in that order.
Bull seminal plasma, however, had more
selenium than that
of any of the other three species. The testis and
epididymis had the greatest concentration of
selenium
found in the reproductive tissues. Levels of
selenium
in semen and reproductive tissues were significantly higher
(P<0.05) than those in blood. The implications of these
findings, as well as the inter-relationship between
selenium, GSH-Px and animal reproduction, are discussed.

▼ Selenium (Se) is an essential trace mineral required in maintaining health in humans. Recent research has demonstrated that Se shows promise as an agent that can reduce the harmful side-effects of chemotherapy (CT) and radiotherapy (RT), while not compromising the effectiveness of treatment. Clinical evaluations using varying doses of Se have demonstrated a significant reduction in toxicity of CT and RT. Se supplementation appears to mediate its biological effects through four main mechanisms: Se treatment promotes the death of malignant cells, but not normal cells, through the differential effects on the endoplasmic reticulum stress response; it inhibits hypoxia-induced angiogenesis which is required by tumours for their blood supply; it enhances DNA repair in normal but not malignant cells; and it reverses cellular resistance to some cytotoxic drugs. The objectives of this thesis were to evaluate the biological characteristics of the differential impact of a Se compound, methylseleninic acid (MSA), on malignant and non-malignant human cells and how that modulates the effects of cisplatin chemotherapy and/or radiation. In parallel with this, similar evaluations were undertaken in patients receiving a Se compound or placebo in conjunction with cisplatin and radiation for head and neck squamous cell carcinoma (HNSCC). MSA induced differential effects in peripheral blood mononuclear cells (PBMC) obtained from healthy individuals compared to the monocytic leukaemia THP-1 cell-line. While MSA did not change PBMC viability, it reduced that of THP-1 cells in a concentration-dependent manner. Furthermore, MSA treatment enhanced the cytotoxicity of CT and RT toward THP-1 cells without compromising the survival of PBMC. This increased sensitivity in THP-1 cells is likely to be related to significantly reduced levels of intracellular glutathione (GSH) in response to MSA. In contrast, GSH levels in PBMC rose substantially in response to MSA. Assessment of ER stress response proteins revealed that pro-survival proteins (GRP78, phospho-eIF2α, and spliced XBP-1) were upregulated in PBMC in response to MSA treatment. The malignant THP-1 cells had high levels of pro-survival proteins at baseline, which increased to a variable degree, especially caspase-8, in response to MSA. Putative pharmacodynamic markers of biological activity of Se were evaluated in plasma samples and PBMC from HNSCC patients in a randomised, placebo-controlled, double-blind phase II trial. Patients were treated with a 7-week radical course of concurrent cisplatin and RT (CRT) and were randomised to receive either selenomethionine (SLM) or matching placebo for 11 weeks, starting a week prior to CRT treatment. Because the trial is not yet complete, treatment allocation remains blinded and thus the pharmacodynamic results according to treatment with SLM or placebo cannot be made at the time of writing this thesis. ▼ Selenomethionine and methionine have been postulated to be similarly
involved in protein synthesis. This thesis has evaluated this
hypothesis from a molecular and genetic approach. Selenomethionine
and methionine were incorporated into rabbit reticulocyte hemoglobin
at the same rates and in equal quantities. Each methionine demonstrated
competitive inhibition for its analog incorporation into the globin.
There were no significant differences in a or (3 chains for the
methionine or selenomethionine rates of incorporation. Ion-exchange
peptide analysis from a tryptic hydrolysate from this globin showed
that selenomethionine and methionine were probably incorporated into
the same peptides. However the peptides containing selenomethionine
were somewhat displaced from the peptides containing methionine.
When methyl labeled methionine and
selenium labeled selenomethionine
were incorporated into the globin, both their activities could be
reduced 56-76% with dialysis against sodium sulfite. It was concluded
that this sulfitolysis procedure was removing activity from the
methionine and selenomethionine residues in the globin.
There was no difference in selenomethionine or methionine
aminoacylation of tRNA using materials from normal or
selenium adapted
Escherichia coli B. Both methionine and selenomethionine were rapidly
bound in this system. Assay by plating on membrane filters demonstrated
a higher binding of selenomethionine than methionine. However, assays
by reverse phase chromatography showed that methionine and selenomethionine
were bound in equal amounts and to the same two tRNA species.
The two methionines were competitive inhibitors of each other for the
aminoacylation of tRNA.
Aminoacyl synthetase and tRNA were then prepared free of ribonuclease.
Both selenomethionine and methionine had about the same
optimum aminoacylation conditions: enzyme and tRNA concentration,
ATP/Mg ratios, and pH range. Both aminoacylations were completely
dependent upon added ATP. Sodium ion inhibited both aminoacylations,
but could be overcome by potassium ion. Selenomethionine was aminoacylated
faster and in preference to methionine, but showed no saturation
of tRNA when selenomethionine concentration was increased.
Aminoacylated selenomethionine- and methionine-tRNA could be deaminoacylated
at pH 8.8, and then reaminoacylated to the same degree. Reverse
phase column chromatography indicated that from this ribonuclease free
material, there was only one methionine accepting tRNA species, which
was identical in position and stoichiometry to the selenomethionine
accepting tRNA species.

▼ A series of compounds with the general formula
R3PX2 and R3PX4 (where R = o-tolyl, m-tolyl, p-tolyl, o-anisyl,
m-anisyl, p-anisyl, o-thioanisyl, p-thioanisyl, p-fluorophenyl,
p-chlorophenyl, mesityl and X = Cl, Br, I) have been synthesised
and studied by X-ray crystallography, multinuclear NMR spectroscopy
and Raman spectroscopy. A study of the conformational features of
the aryl groups in these structures was undertaken in order to
investigate the role of steric effects and crystal packing on P-X
and X-X bond lengths.Compounds with the general formula RSeX /
RSeX3 (where R = p-fluorophenyl, p-chlorophenyl, p-bromophenyl,
p-iodophenyl and X = Cl, Br, I) have been synthesised by reacting
the diselenides with halogens in the stoichiometric ratios. The
X-ray crystal structure of p-ClC6H4SeI.I2 is the first
crystallographically characterised example of a compound of
stoichiometry RSeI3, although it is better represented as an I2
adduct of a Se(II) iodide than a Se(IV) compound. In contrast,
(p-FC6H4)Se-Se(I)2(p-FC6H4) is an I2 adduct where the Se-Se bond
has not been cleaved by iodine. Both structures are stabilised by
weak Se•••I and I•••I interactions. p-FC6H4SeCl3 is an example of a
dimeric structure with bridging chlorines.[Ph4PX] and Ph3PX2 were
reacted with PhEX (where E = Se, Te and X = I, Br, Cl) to produce
salts containing [PhEX2]- and [PhEX4]- anions. Where possible all
the products were then analysed via NMR and Raman spectroscopy and
X-ray crystallography.The reactions of the ligand
tris-p-fluorophenyl phosphine with sulfur and
selenium are
presented along with the subsequent reactions of these compounds
with diiodine. All of the compounds are characterised via X-ray
crystallography.

Ezért fontos, hogy a tesztelő is képzett szakember legyen, aki ismeri a szakterületének a fogalmait és képes legyen használni az eszközeit. A témaválasztásom célja a tesztelés alapfogalmainak megismertetése illetve a manapság legelterjedtebb teszt eszközök bemutatása. A fejezeteken keresztül igyekeztem a teljes tesztelési folyamatot egészében bemutatni és a hozzá kapcsolódó fogalmakat részletesen ismertetni, valamint általam hasznosnak vélt programokat bemutatni.
A szakdolgozatom végén egy új – általam érdekesnek és hasznosnak talált - tesztelésre épülő programozás tanítási módszertant írtam le.

▼ In the present study the light dependent biosynthesis of spherical red elemental selenium Se(0) granules (average size of 220 nm) by the cyanobacterium Synechococcus leopoliensis when exposed to high selenite concentrations for 9 days was investigated. The particles were identified as Se(0) by energy dispersive x-ray spectroscopy (EDX) analysis and were localized between the periplasmic space and the cell surface through light and environmental scanning electron microscopy (ESEM). The amount of selenite reduced to Se(0) increased with increasing culture density and less selenite remained in the cultures accordingly. Selenite treated cultures lost their characteristic pigments and turned from green to orange by day 5 and a concomitant increase in the buoyant cell density was observed over the treatment period. The potential abiotic and biotic reactions that may have produced Se(0) in the cyanobacteria are discussed.

▼ The infrared and polarized Raman spectra of SeOBr₂ as a solid, melt, and in solution have been obtained. The frequencies that were found are consistent with Cs symmetry and a pyramidal molecule. Ionic dissociation, like in other SeOX₂ compounds, is evidenced by electrical conductance measurements. SeOBr₂ can act both as a Lewis acid or base. For example, a salt with the composition KSeOBr₃ has been prepared from KBr and SeOBr₂.
Ligand redistribution reactions of various SeOX₂ and SeOY₂ compounds, where X and Y are F, CI, Br, and SO₃F, were studied via vibrational spectroscopic and ¹⁹F nuclear magnetic resonance techniques. The presence of the mixed SeOXY compounds has been detected in all these systems basically in equilibrium with their parent compounds. None of the SeOXY compounds could be separated out of the mixtures.

▼ 1. The experimental evidence of the present work in selenium toxicology is such as to cast grave doubts on the authenticity of some aspects of the work of previous investigators in this field. The response of the laboratory animals, subjected to the various procedures practised by these investigators in attempts to produce a condition of chronic selenium poisoning, suggests that these animals are, in addition to being the victims of selenium poisoning, the victims of inanition and/or dehydration.
2. The presence of up to 15 p.p.m. sodium selenite (6.9 p.p.m. Se) in the drinking water is tolerated without any drastic reduction in food and water intake by laboratory rats, provided that this concentration is reached gradually. At this level of selenium intake the animals will remain for at least 25 weeks in good condition and will exhibit growth only slightly less rapid than that of control animals. Any attempts to raise the selenium content above this level results In a compensatory lowering of the water intake and a resultant reduction in food intake of the animals.
3. The results obtained in the attempts to induce chronic selenium poisoning by dietary methods were such as to cause these methods to be abandoned due to their unsuitability for the present project. A method involving the subcutaneous injection of sub-lethal doses of sodium selenite incorporated into an absorption delaying vehicle was evolved and, after testing, was deemed suitable for the purpose of inducing a sub-acute condition of selenium toxicity to be used as a means of assessing the value of chemotherapy.
4. Experimental data on the urinary excretion levels of selenium was made available by the development of a new colorimetric procedure for the quantitative determination of selenium in biological samples. The data obtained by this method revealed that the maximum quantity of selenium excreted in the urine of rats over a period as long as ten days after injection was about one third of the quantity originally injected. No particular advantage in the use of the absorption delaying vehicle was discernable by this method but since, under the limitations imposed by the quantities of urine available, the delay in hours would be undetectable the vehicle was retained in the experiments assessing the value of BAL (2, 3-dithiopropanol) as a chemotherapeutic agent.
5. The use of BAL as a chemotherapeutic agent for combatting selenium toxicity has a deleterious effect on the intoxicated animals. The subcutaneous injection of quantities of BAL in peanut oil at levels considered well within the limits of safety resulted in an intensification of the distress of selenium injected rats. Deaths were more frequent than in the case of non BAL-treated animals. The animals which survived the effects of both poison and treatment showed a slightly higher and a more prolonged excretion of selenium in the urine.

▼ The spark spectra of selenium have been photographed from the infra-red to the vacuum ultra-violet on a variety of spectrographs including a two meter vacuum grating spectrograph, a twenty-one foot concave grating, a Hilger Constant Deviation, a Hilger medium quartz and a Hilger large automatic glass-quartz prism spectrograph. Two light sources have been used: An electrodeless spark discharge and a spark in helium. About 2200 selenium lines have been measured in the region 10450 to 345 Angstroms. On the basis of these measurements, new levels have been found in Se III, Se IV, and Se V.
The most important achievement was the discovery of the deepest excited terms 4s4p³ ⁵S⁰₂ in Se III and 4s4p² ⁴p in Se IV. The chief extension of the analysis has been in Se IV.
A few interferometric measurements were made in Se II and Se III. Using an electrodeless discharge tube excited by a high frequency generator wavelengths of 38 lines in the arc spectrum of potassium have been determined interferometrically.

Scintillation detection techniques were employed to measure, the relative gamma ray intensities, gamma-gamma coincidences, and conversion electron-gamma coincidences. A decay scheme with five excited As⁷⁵ levels at 200, 265, 280, 305 and 402 kev. is proposed. Multipolarities are assigned to the transitions between these levels on the basis of conversion coefficients, determined from previously measured conversion intensities.

▼ Prokaryotes metabolize selenium in two significant ways. One way prokaryotes metabolize selenium is through incorporation into the 21st amino acid selenocysteine (Sec), which is inserted into proteins co-translationally. Additionally, many prokaryotes exploit the selenium oxyanions selenite and selenate as terminal electron acceptors in anaerobic respiration. Little is known about the evolution of selenoproteins in prokaryotes. While selenate respiration has been studied in several bacteria, nothing is known about the physiology of selenite respiration. I investigated both aspects of selenium metabolism in Bacillus selenitireducens MLS10 by annotating the selenoproteome of MLS10 and constructing phylogenies of these selenoproteins to investigate the evolution of selenoproteins in the bacilli, and by obtaining the protein profiles using SDS-PAGE, determined the cytochrome content using the pyridine hemochrome assay, and tested for enzyme activity in native gels using selenite-grown MLS10 cells. My research demonstrates that the bacilli exploit Sec residues to a far greater extent than has heretofore been appreciated, that the selenoproteins exploited by the bacilli are closely related to those of the clostridia, and provides evidence that the evolution of selenoproteins in Gram-positive bacteria and the d-Proteobacteria is characterized by extensive horizontal gene transfer. Finally, my research provides evidence that selenite respiration is a distinct, inducible respiratory pathway in MLS10, and suggests future directions for further testing of this hypothesis.

▼ Selenium is an essential trace element nutrient for
human life, but it can be extremely toxic if taken excess. Various
adsorbents have been applied for the removal of selenium from water
but performed poorly in the removal of Se (VI), which is more
bioavailable and more difficult to remove. Herein, we reported a
new method to prepare poly (amido amine) functionalized graphene
oxide (PAMAM-GO) with different generations (GO-G0 to GO-G4). The
resulting products were applied in selenium removal from simulated
water, showing an increasing capacity with increasing generation.
The 4th generation of PAMAM-GO (GO-G4) shows a removal capacity of
60.9 mg g-1 in Se (IV) and 77.9 mg g-1 in Se (VI) at pH 6. The
adsorption mechanism is attributed to the electrostatic interaction
between selenium oxyanions and the adsorbents, proved by the
effects of initial pH on removal capacity and zeta potential
measurements.

▼ The first chapter of this thesis describes the
development of a general route for the generation of selenium
stabilized carbanions via a method that bypasses acid catalyzed
formation of selenoacetals. Aldehydes are first converted into
α-hydroxystannanes by nucleophilic addition of Bu3SnLi. The
resulting hydroxyl is then converted to the corresponding phenyl
selenide by treatment with PhSeCN and Bu3P to give the
α-(phenylseleno)stannane. Upon treatment with BuLi, the
α-(phenylseleno)stannane undergoes preferential Sn/Li exchange to
give a selenium stabilized carbanion. This carbanion is condensed
with an aldehyde to give a β-hydroxy selenide, which can be
converted into an allylic alcohol upon oxidation and selenoxide
fragmentation. The formation of α-(phenylseleno)stannanes has been
applied to aldehydes containing straight and branched alkyl chains,
aromatic substituents, and acid sensitive functional groups. The
selenium stabilized carbanions have been condensed with aldehydes
containing straight and branched alkyl chains, aromatic
substituents, and α,β-unsaturation. The second chapter describes
synthetic studies towards two marine natural products,
marinopyrrole A and B. The synthesis began with the preparation of
the unprecedented N,C2 linked bispyrrole core through a Paal-Knorr
condensation onto the amino group of ethyl
3-aminopyrrole-2-carboxylate. Preparation of the densely
halogenated core of the marinopyrroles through selective
chlorination of the bispyrrole seems to be impossible and was not
pursued. Due to unexpected alterations to the electronic structure
of the pyrrole ring in the presence of electron withdrawing
substituents, preparation of a fully functionalized top ring was
unsuccessful. Blocking the 3' position of the bispyrrole with a
removable blocking group was also unsuccessful by an ortho
metallation approach. Preparation of the halogenated bispyrrole
starting with the fully functionalized bottom pyrrole seems
promising after a model intramolecular conjugate displacement
reaction had shown that it is possible to mount the precursor to
the top pyrrole on top of the bottom pyrrole. Work is in progress
to prepare the mountable precursor to the top ring.

▼ Selenium (Se) is an essential micronutrient involved in important metabolic functions for all vertebrate species. As Se is reported to have a narrow margin between essentiality and toxicity, there is growing concern surrounding the adverse effects of elevated Se exposure caused by anthropogenic activities. Recent studies have reported that elevated dietary exposure of fish to selenomethionine (Se-Met) can alter aerobic metabolic capacity, energetics and swimming performance. My thesis aimed to further investigate mechanisms of sublethal Se-Met toxicity, particularly potential underlying cardiovascular implications of chronic exposure to environmentally relevant concentrations of dietary Se-Met in adult zebrafish (Danio rerio) and juvenile (yearling) rainbow trout (Oncorhynchus mykiss). In my first experiment, adult zebrafish were fed either control food (1.1 μg Se/g dry mass [d.m.]) or Se-Met spiked food (10.3 or 28.8 μg Se/g d.m.) for 90 d at 5% body weight per day. In the second experiment, juvenile rainbow trout were fed either control food (1.3 μg Se/g d.m.) or Se-Met spiked food (6.4, 15.8 or 47.8 μg Se/g d.m.) for 60 d at 5% body weight per day. Following exposure, ultrahigh resolution B-mode and Doppler ultrasound was used to characterize cardiac function. Chronic dietary exposure to elevated Se-Met had opposing results in zebrafish when compared to the rainbow trout. Zebrafish exposed to the highest dietary concentrations of Se-Met (28.8 μg Se/g d.m.) had significantly reduced ventricular contractile rate, stroke volume, and cardiac output, while Se-Met exposed rainbow trout had significantly greater stroke volume, ejection fraction, and cardiac output.
Following ultasonography, energy stores were measured via whole body (zebrafish) and liver, heart and muscle (rainbow trout) glycogen and triglyceride concentrations. Zebrafish in the highest exposure group were observed to have greater whole body glycogen concentrations when compared to the control group, while rainbow trout exposed to Se-Met concentrations greater than 15.8 μg Se/g showed significant increases in both glycogen and triglycerides in liver relative to the control group. In addition, rainbow trout in the highest exposure group had significantly reduced capability of managing blood glucose levels as was evident after 48hrs in a glucose tolerance test.
Exposure to Se-Met significantly decreased mRNA expression of a key cardiac remodelling enzyme, matrix metalloproteinase 2 (MMP2), in adult zebrafish heart, however significantly increased it (MMP9) in rainbow trout heart. Selenomethionine significantly increased echodensity at the junction between atrium and ventricle in zebrafish, and these results combined with increased MMP2 expression are consistent with cardiac remodelling and fibrosis. ▼ Cancer of the breast is the most common cancer affecting women, with a 1 in 9 lifetime risk of developing breast cancer. Carriers of mutations in the breast cancer susceptibility gene, BRCA1, present with an increased lifetime risk of 80-95% of developing breast cancer. BRCA1 is a tumour-suppressor gene that is involved in DNA repair of double-strand breaks and transcriptional regulation.
Studies have shown that oral supplementation with
Selenium (Se) could potentially be an effective preventive agent against BRCA1-mutation carriers. Se is an essential trace mineral that plays critical roles in maintaining health in humans. Se functions by protecting lipid membranes, proteins and DNA from free radical damage and is involved in redox regulation in the cell. These functions are mediated through specific Se-dependent proteins termed selenoproteins, in which Se is incorporated.
Se supplementation may exert its benefits by enhancing the DNA damage repair response. The effects also appear to be dependent on selenoprotein genotypes and concentration of Se in the body/diet. It is possible that Se supplementation may be effective in reducing the risk of breast cancer in BRCA1 carriers, but the use of organic Se compounds is likely to be preferable, but the dose-response needs to be determined.
The objectives of this research thesis are to evaluate the dose-response of sodium selenite and methylseleninic acid (MSA) in two BRCA1-mutated cancer cell lines in vitro. The MTT assay was used to monitor cell activity, along with the comet assay to evaluate the DNA damage level using a dose-response curve of the two Se compounds on control and bleomycin-induced DNA damaged cells. In addition, DNA from each cell line was sequenced to confirm the presence of the published BRCA1 mutation.
The data presented within this thesis demonstrates that a commonly-used inorganic form of Se, sodium selenite, is more genotoxic than the organic Se compound, MSA, in malignant BRCA1-mutated cells. Sodium selenite also has more direct cytotoxicity in BRCA1-mutated cells than MSA, as demonstrated by the MTT assay. However the enhanced repair capacity from Se on DNA damage in cells demonstrated by others has not been reproduced. Preliminary data showed potential at much lower doses (2 μM) of Se. Interestingly, the volatile metabolites produced from MSA appear to reduce DNA damage, supportive of this hypothesis. In order to understand if the effects observed are linked specifically to the BRCA1 mutation or breast cancer in general, future research should include the investigation of the effects of Se on breast cancer using alternative breast cancer cell lines, as well as non-malignant cells.

▼ Selenium is an important element in soils throughout the western United States, and its presence has important consequences for the ecology of these seleniferous sites. Some plants in seleniferous areas have evolved mechanisms to hyperaccumulate Se to 0.1 - 1% of their dry weight. Other plants accumulate moderate Se levels between 0.01 - 0.1% of their dry weight. In the studies described in this thesis, facets of the evolution of Se accumulation and the associated ecology of Se hyperaccumulators are considered. First, I examined the effect of increasing Se on reproductive parameters of Se accumulators and hyperaccumulators. The reproductive parameters were measured through cross-pollinations of greenhouse-grown accumulator plants receiving different Se concentrations. In the accumulator Brassica juncea , increasing Se concentrations in plant tissues caused decreases in biomass, pollen germination, seed weight, seed production, and seed germination. In some cases, however, interactions of similar Se concentrations in both parents actually proved beneficial to reproduction. The hyperaccumulator Stanleya pinnata showed no effect of increased Se concentration on pollen germination. These data provide interesting insight into the potential reproductive cost of Se accumulation, and the apparent evolution of physiological tolerance mechanisms in hyperaccumulators to avoid these reproductive problems. To further investigate the effect of Se on reproduction, S. pinnata plants were taken to a field site with hives of the European honey bee (Apis mellifera) to examine the effect of Se in floral tissues on potential pollinators. The bees and other pollinators showed no preference for or against Se in the flower and foraged on both high- and low-Se plants equally. Because the bees showed no preference, the honey of bees in seleniferous sites was analyzed for the presence of Se, and there were small amounts (up to 2 mg kg-1 FW) of Se found in this honey. These findings are important for bee keepers in seleniferous areas, as they show no evidence of toxic Se levels in their honey and they may even market their honey as Se-enriched and beneficial for human health. The finding that bees do not discriminate between high and low-Se plants does warrant further studies on the potential health effects of the ingested Se on the pollinators and the movement of Se into the food chain. Next, to further investigate the ecology of Se hyperaccumulators, I examined the interactions of hyperaccumulator roots with root-associated nematodes.
Selenium hyperaccumulators S. pinnata and Astragalus bisulcatus growing in the field have root Se concentrations between 100 and 1,500 mg Se kg-1 DW, a toxic concentration to most above-ground herbivores. Therefore, it was expected that with increasing root Se concentrations, there would be reduced levels of nematodes associated with plants. ▼ A series of compounds with the general formula R3PX2 and R3PX4 (where R = o-tolyl, m-tolyl, p-tolyl, o-anisyl, m-anisyl, p-anisyl, o-thioanisyl, p-thioanisyl, p-fluorophenyl, p-chlorophenyl, mesityl and X = Cl, Br, I) have been synthesised and studied by X-ray crystallography, multinuclear NMR spectroscopy and Raman spectroscopy. A study of the conformational features of the aryl groups in these structures was undertaken in order to investigate the role of steric effects and crystal packing on P-X and X-X bond lengths. Compounds with the general formula RSeX / RSeX3 (where R = p-fluorophenyl, p-chlorophenyl, p-bromophenyl, p-iodophenyl and X = Cl, Br, I) have been synthesised by reacting the diselenides with halogens in the stoichiometric ratios. The X-ray crystal structure of p-ClC₆H₄SeI.I₂ is the first crystallographically characterised example of a compound of stoichiometry RSeI₃, although it is better represented as an I₂ adduct of a Se(II) iodide than a Se(IV) compound. In contrast, (p-FC₆H₄)Se-Se(I)₂(p-FC₆H₄) is an I₂ adduct where the Se-Se bond has not been cleaved by iodine. Both structures are stabilised by weak Se...I and I...I interactions. p-FC₆H₄SeCl₃ is an example of a dimeric structure with bridging chlorines. [Ph₄PX] and Ph₃PX₂ were reacted with PhEX (where E = Se, Te and X = I, Br, Cl) to produce salts containing [PhEX₂]- and [PhEX₄] anions. Where possible all the products were then analysed via NMR and Raman spectroscopy and X-ray crystallography. The reactions of the ligand tris-p-fluorophenyl phosphine with sulfur and selenium are presented along with the subsequent reactions of these compounds with diiodine. All of the compounds are characterised via X-ray crystallography.

Opinnäytetyön tavoitteena oli tutkia, miten käyttöliittymätestausautomaatiota voitaisiin hyödyntää sovelluskehitysprosessissa. Tarkoituksena oli rakentaa käyttöliittymätestausautomaatio HSY-mittaridata-projektille.
Käyttöliittymätestausautomaatio voidaan luokitella kuuluvaksi järjestelmätestaukseen. Testaus olisi ollut hyvä ottaa osaksi prosessia jo varhaisemmassa vaiheessa projektia parhaiden tulosten saamiseksi. Selenium WebDriver osoittautui erinomaiseksi frameworkiksi laajemman käyttöliittymätestausautomaation rakentamisessa. Google Chrome -selain valittiin sopivimmaksi testiselaimeksi sen tarjoaman headless-tilan ja mobiiliemuloinnin vuoksi. CI-palvelimella on mahdollista suorittaa testejä ilman näytönohjainta.
Raportointia ja seurantaa tulisi parantaa resurssien käyttämisen tehostamiseksi. Toisaalta frameworkin valinta tulisi suorittaa aina nykytila huomioiden frameworkien nopean kehityksen vuoksi. Erilaisten testi-frameworkien tarjoamiin ominaisuuksiin tulisi tutusta paremmin. Nykyaikaisten JS-frameworkien toimintaa tulisi selvittää enemmän, ja sen pohjalta laatia yleispätevämpiä ratkaisuja epävakauden korjaamiseksi.
The bachelor’s thesis was done for Cybercom Finland Oy. Their HSY-meter data project needed test automation to help with the software development process. The goal of this thesis was to research how to implement user interface test automation into a modern software development process. The purpose was to build user interface test automation for the meter data project.
User interface is categorized as system testing. It should have been implemented earlier in the project for the best results. Selenium WebDriver turned out to be an excellent framework for building the user interface test automation. Google Chrome was chosen as the best browser for testing because it supports both mobile emulation and headless-mode. It is possible to run user interface tests on a CI-server without graphics card.
Reporting and follow-up should be improved to allow for better planning and resource allocation. When choosing the framework for user interface testing, one should always do their own research. Frameworks evolve at such a rapid pace and sometimes it comes down to personal programming language preferences. Other test frameworks besides JUnit should be researched more. JavaScript based frontend frameworks require more research to enable more stable tests in the future.

Selenium is a chemical element that is an essential micronutrient associated with various aspects of human health. The biochemical activity of selenium is mediated by proteins, which incorporate it into the amino acid selenocysteine (Sec). There are 25 genes in humans encoding Sec-containing selenoproteins. The functionally characterized selenoproteins are oxidoreductase enzymes involved in cellular oxidation-reduction reactions. Most selenoproteins are expressed in the mammalian brain, and dietary selenium deficiency causes preferential retention in brain relative to other body organs. Further, dietary selenium deficiency and specific selenoproteins are associated with various brain diseases. Mouse models have been extensively used to study the function and handling of selenium in mammals. Genetic deletion of a Sec-rich protein in mice causes brain selenium deficiency, neurodegeneration and neurological impairment, and disruption of a phospholipid hydroperoxidase selenoenzyme causes rapid neurodegeneration. Therefore, selenoprotein expression and function promotes a healthy nervous system. However, selenium metabolism and the function of several selenoproteins in brain are not clearly defined. The overall purpose of this work is to clarify the function and utilization of selenium in the mammalian brain, to reveal implications for developmental and neurological diseases. The goal of these studies is to investigate changes in brain function and selenoprotein expression under conditions of altered selenium metabolism in mice. The research presented in this dissertation covers three major topics that are separated into chapters. To investigate selenium distribution in the brain, the neurological consequences of disrupting selenium transport and recycling in mice are assessed and compared. Disruption of selenium transport caused more profound neurological consequences than disruption of selenium recycling. To investigate potential functions of selenium in the nervous system, select selenoproteins were examined for cellular and subcellular expression in cells and brain tissue from transgenic and control mice. Select selenoproteins and synthesis factors were observed at synapses, suggesting localized expression and physiological relevance. To investigate a potential interaction between selenium and methamphetamine, expression profiling of selenoproteins in mouse brain after exposure to methamphetamine is described. Selenoprotein synthesis was adversely affected by methamphetamine administration in mice. These results confirm the importance of selenium in the mammalian brain.

▼ Two pilot-scale wetland treatment system cells (nutrient amended and unamended) were designed and constructed to reduce aqueous Se concentrations in simulated energy-derived water. Specific objectives of this study were: (i) measure and correlate hydrosoil conditions with Se concentrations vertically through the hydrosoil; (ii) investigate Se-accumulating biogeochemical processes (dissimilatory Se reduction and sorption) operating in the hydrosoil; and (iii) evaluate the effect of a nutrient amendment on hydrosoil conditions, Se accumulation, and Se-sequestering biogeochemical processes in the hydrosoil. Se accumulation (i.e. total Se concentration) and hydrosoil conditions were measured with depth in the hydrosoil. Se-sequestering biogeochemical processes were investigated by counting Se-reducing microbial colony forming units (CFUs) and identifying Se geochemical fractions at various depths in the hydrosoil. The detritus (0-21 cm in nutrient amended cell and 0-14 cm in unamended cell) contained greater Se concentrations (308-830 µg/g and 138-569 µg/g) and greater CFUs (2,700-22,000 CFUs/mL pore water and 9,300-15,000 CFUs/mL pore water) than the underlying sandy sediment. Correlation of organic matter content with Se concentration (r = 0.95; p<0.00001 in nutrient amended and r = 0.87; p<0.00002 in unamended) suggests organic matter influences Se-accumulating biogeochemical processes. In detritus, mean hydrosoil conditions (redox: -2 to -173 mV, pH: 6.20-6.46, and organic matter: 52-86%) were more favorable for sorption than for dissimilatory Se reduction; however, the majority of Se measured in the detritus was elemental (52.1%-58.0% in the nutrient amended cell and 21.1%-62.6% in the unamended cell) suggesting that dissimilatory Se reduction is the dominant biogeochemical process sequestering Se in the detritus. T-tests indicate significant difference in pH (t = 2.87, p = 0.0132) of the hydrosoil between the nutrient amended cell and unamended cell, but no significant differences (p<0.005) in redox potential, organic matter content, and Se concentration. Greater Se concentrations and percent of elemental Se in the nutrient amended cell than the unamended cell suggests that the nutrient amendment enhanced dissimilatory Se reduction and therefore Se accumulation in the hydrosoil.

▼ Selenium and arsenic are important metalloids in the food chain from nutritional and toxicological point of view. These two metalloids are potentially enriched through geogenic processes and anthropogenic activities and they could sometimes co-exist in nature and become available to plants thereby entering the food chain. While selenium is known as an essential element to humans, it could also be toxic. Arsenic on the other hand is a potentially toxic element posing serious health risks to livestock and humans. They have been found to neutralise each other’s effects in animals but their interactions in plants are not well understood. Speciation analysis, which is a set of activities leading to identification and quantification of different forms or species of elements present in an entity, is required for a holistic understanding of the mechanisms and interactions involved in the plants’ metabolism of contaminants and essential elements. Many techniques are currently being used for speciation of selenium and arsenic in plants and they sometimes give contradictory outcomes. The hyphenation of HPLC with MS and synchrotron techniques are the two most commonly used state of the art techniques for speciation of these metalloids. This research therefore sought to access, explore and/ or develop analytical methods appropriate for the speciation of selenium and arsenic in plants. Many selenium and arsenic species have been identified and reported in the literature using well established procedures. The presence of elemental selenium in plants has also been widely reported in plants but to our knowledge this presence has never been experimentally proven and fully quantified. Because this species is non toxic, its proven occurrence in plants will represent a potential detoxification mechanism. Therefore in this study, a method was specifically developed for identification and quantification of elemental selenium. In order to investigate the occurrence of elemental selenium in plants, the newly developed method was applied using Thunbergia alata as a model plant. Arsenic is known to activate the synthesis of PC using glutathione and the complexation of the activating arsenic ions with the synthesised PCs is a well established detoxification mechanism for arsenic. However, very little is known about the role of glutathione and PCs in selenium detoxification. In order to be able to gain better insight into the interaction between selenium and arsenic in plants, the role of glutathione and PCs in selenium metabolism was investigated using Arabidopsis thaliana as a model plant. Sensitivity tests and speciation analysis were carried out on Arabidopsis thaliana WT and the mutants’ one of which is deficient in GSH synthesis and the other deficient in PC synthesis using selenite and arsenate as toxicants. melt runoff. In aquatic invertebrates, Se concentrations exceeded
values known to cause reproductive impairment in fish and bird
predators. Se concentrations in all fish species exceeded known
thresholds for reproductive impairment in avian consumers and the
majority surpassed concentrations that would negatively affect
wildlife and human consumers. Se concentrations in some fish
species have significantly increased over the past 6-16 years. The
strongest predictors of fish Se concentrations were growth rate,
condition factor, age, weight, trophic position (within lakes) and
vegetation type (among lakes). These results suggest that
consumption advisories are desirable for several lakes in the Banff
and Kananaskis area, and that Se concentrations in fish from other
area lakes should be investigated.

▼ To σελήνιο (Se) αποτελεί απαραίτητο ιχνοστοιχείο για τη ζωή ασκώντας τις δράσεις του ως απαραίτητο δομικό στοιχείο των σεληνοπρωτεινών (υπεροξειδάσες της γλουταθειόνης, σεληνοπρωτείνη Ρ, αναγωγάση της θειορεδοξίνης, αποϊωδιονάσες της ιωδοθυροξίνης κ.α.), οι οποίες με τη σειρά τους έχουν ενεργό ρόλο στους αντιοξειδωτικούς/αντιφλεγμονώδεις μηχανισμούς του οργανισμού, στη διατήρηση της οξειδοαναγωγικής κατάστασης των κυττάρων και στο μεταβολισμό των θυρεοειδικών ορμονών. Συνεπώς, η επαρκής πρόσληψη Se με τη διατροφή είναι ιδιαίτερα σημαντική για τη σωστή λειτουργία του οργανισμού. Ωστόσο, οι διατροφικοί παράγοντες που καθορίζουν τα επίπεδα Se στον ανθρώπινο οργανισμό δεν είναι πλήρως διευκρινισμένοι ενώ καμία μεγάλη επιδημιολογική μελέτη δεν έχει γίνει στον Ελληνικό πληθυσμό. Παράλληλα, αν και η αυξημένη πρόσληψη Se έχει συσχετιστεί θετικά με το κίνδυνο εμφάνισης συγκεκριμένων νοσημάτων (διαβήτης, υπέρταση), η σχέση του με παράγοντες κινδύνου για καρδιαγγειακά νοσήματα είναι πολύ λιγότερο μελετημένη και τα αποτελέσματα αντικρουόμενα. Αυτό εν μέρει οφείλεται στις πλειοτροπικές και πολλές φορές αντίθετες δράσεις των σεληνοπρωτεϊνών στους μηχανισμούς εμφάνισης καρδιαγγειακών νοσημάτων. Η συσχέτιση συνεπώς των επιπέδων ολικού Se με παράγοντες κινδύνου για καρδιαγγειακά νοσήματα δεν επαρκεί και είναι πλέον απαραίτητη η ανάπτυξη ισχυρών αναλυτικών μεθόδων, οι οποίες θα έχουν την ικανότητα να προσδιορίζουν ταυτόχρονα τις διάφορες σεληνομορφές στα βιολογικά δείγματα . Για τις ανάγκες της συγκεκριμένης διατριβής, χρησιμοποιήθηκε αναλυτική μέθοδος βασισμένη στη χρωματογραφία συγγένειας σε συνδυασμό με φασματομετρία μάζας επαγωγικά συζευγμένου πλάσματος (dAF-(LC)-ICP/MS). Στη συνέχεια της μελέτης, πραγματοποιήθηκε περαιτέρω διερεύνηση της συνύπαρξης μικρών οργανικών μορίων και της υπεροξειδάσης της γλουταθειόνης-3. Για το σκοπό αυτό έγινε σύζευξη χρωματογραφικής στήλης μοριακού αποκλεισμού στο σύστημα dAF-ICP-MS. Τα αποτελέσματα των αναλυτικών μεθόδων ανέδειξαν ότι το Se βρίσκεται με τη μορφή SelP( 54%), GPx-3 (26%), SelAlb (12%) και άγνωστης μορφή (8%) στον ορό αίματος. Από την εφαρμογή της μεθόδου σε ένα υποπληθυσμό της μελέτης «ΑΤΤΙΚΗ»(Ν=399) προέκυψε ότι ένα σημαντικό μέρος του Ελληνικού πληθυσμού, όπως αυτό τουλάχιστον εκφράζεται από τη μελέτη ΑΤΤΙΚΗ, έχει έλλειψη σεληνίου. Οι διατροφικοί παράγοντες που συσχετίζονται θετικά με τα επίπεδα Se είναι η κατανάλωση κόκκινου κρέατος και πουλερικών (κρεατοφαγική δίαιτα). Η ανάλυση των σχέσεων σεληνίου/πρωτεινών σεληνίου με παράγοντες καρδιαγγειακού κινδύνου έδειξε ότι υπάρχει σημαντική συσχέτιση του Se με τους περισσότερους κλασσικούς παράγοντες (ηλικία, παχυσαρκία, κάπνισμα, υπέρταση, γλυκαιμία, λιπιδαιμία, δείκτες οξειδωτικού στρες και φλεγμονής), η οποία διαμεσολαβείται από διαφορετικές πρωτεΐνες σεληνίου. Παράλληλα, διαφάνηκε ότι όχι μόνο το ολικό σελήνιο αλλά κυρίως η διαφορετική κατανομή μου στις πρωτεΐνες σεληνίου σχετίζονται με τους παράγοντες κινδύνου γεγονός που υποδεικνύει ότι η κατανομή αυτή αποτελεί καλύτερο δείκτη της κατάστασης Se ενός ανθρώπου και ίσως αποδίδει…

selenium and its alloy are important materials for flat panel digital X-ray detectors. The performance of the X-ray detector is directly related to the carrier mobility and carrier-trapping lifetime of amorphous
selenium and its alloy. An experiment was conducted to examine the dose rate effects on the carrier-trapping lifetime reduction of amorphous
selenium alloy due to X-ray irradiation. In addition, an experiment was conducted to investigate the temperature effects on X-ray induced carrier-trapping lifetime decrease and relaxation process. X-ray induced capacitance changes have been investigated as well. The interrupted field time of flight (IFTOF) techniques are used in order to acquire the carrier-trapping lifetime. Several amorphous
selenium alloy films are irradiated under X-ray. The composition of films are a-Se: 0.3% As: 5ppm Cl and the thickness of the films are about 150 μm. Different dose rates, which range from 0.2 Gy/s up to 2 Gy/s, are applied during the irradiation. The experiments are done for both electrons and holes. The dose rate difference does not result in a significant change in the X-ray induced carrier-trapping lifetime decrease. The same set of samples is studied under three different temperatures, 10 ℃, 23.5 ℃ and 35.5 ℃. The X-ray induced hole-trapping lifetime changes at different temperatures are investigated. After irradiation, the hole-trapping lifetime begins to recovery gradually. The temperature during the relaxation process remains unchanged. The change of hole-trapping lifetime is measured by IFTOF measurement. At higher temperature, upon receiving a certain amount of X-ray dose, the hole-trapping lifetime decreases more than it at lower temperature; however, it also recoveries more quickly at higher temperature. The relaxation process at different temperature also gives activation energy for holes. The X-ray irradiation does not result in any capacitance changes for a-Se films.
