▼ This dissertation examines an important yet woefully understudied aspect of Hern�n Cort�s after the conquest of Mexico. The Marquisate of the Valley of Oaxaca was carefully constructed during his lifetime to be his lasting legacy in New Spain. The goal of this dissertation is to reexamine published primary sources in light of this new argument and integrate unknown archival material to trace the development of a lasting legacy by Cort�s and his direct heirs in Spanish colonial Mexico. Part one looks at Cort�s's life after the conquest of Mexico, giving particular attention to the themes of fame and honor and how these ideas guided his actions. The importance of land and property in and after the conquest is also highlighted. Part two is an examination of the marquisate, discussing the key features of the various landholdings and also their importance to the legacy Cort�s sought to construct. This analysis is extended into the second and third generation of the marquisate, an aspect of Cort�s's legacy mostly ignored by scholars, to trace the success or failure of various developments. In looking at Cort�s and the marquisate in its entirety (as compared to regional studies), it becomes clear that the marquisate was more important to Cort�s than previous historical literature indicates, as it was directly tied to his desire to construct a legacy in New Spain as well as his ideas of fame and honor. Cort�s utilized the marquisate not only for economic gain, but for political and social goals as well. Through showing that Cort�s was devoted to the development and growth of his marquisate, this research highlights the legacy Cort�s sought to construct in Mexico, and the impression he hoped to make on Mexico beyond the conquest. Cort�s controlled the largest estate in New Spain, and how he and his heirs managed that land would fundamentally shape New Spain and the history of Mexico. La courbe S-N est le moyen le plus courant d'analyse et de prédiction de la durée de vie d'un matériau, d'un composant ou d'une structure. Cependant, les modèles standards, qu'ils soient basés sur la théorie de la rupture ou sur des modèles probabilistes n'ajustent pas la courbe dans la totalité sans information sur la microstructure du matériau. Or, cette information provient d'analyses fractographiques souvent coûteuses et rarement disponibles dans le cadre d'une production industrielle. D'un autre côté, les modèles statistiques ne proposent pas d'interprétation matériau et ne peuvent pas être utilisées pour réaliser des prévisions. Les résultats d'un test de fatigue sont par ailleurs très dispersés, plus particulièrement pour les fortes durées de vie, lieu d'apparition d'un phénomène de bi-modalité. Ces constats sont la raison de la proposition d'un nouveau modèle probabiliste. Celui-ci est composé d'un modèle de mélange spécifique, prenant en compte l'approche apportée par la mécanique de la rupture sans nécessiter de d'information supplémentaire sur la microstructure du matériau. Il utilise le fait que la fatigue peut être vue comme la somme d'un amorçage de fissure suivi de sa propagation. Les paramètres du modèle sont estimés à l'aide d'un algorithme EM, où la phase de maximisation combine une méthode d'optimisation de Newton-Raphson et une intégration de type Monte-Carlo. Le modèle "amorçage-propagation" offre une représentation parcimonieuse des courbes S-N dont les paramètres peuvent être facilement interprétés par des ingénieurs matériau. Ce modèle a été testé à l'aide de simulations et appliqué à des données réelles (données sur l'Inconel 718). Ceci nous a permis de mettre en évidence le bon ajustement du modèle à nos données, et ce, pour toutes les déformations disponibles.

S-N curve is the main tool to analyze and predict fatigue lifetime of a material, component or structure. But, standard models based on mechanic of rupture theory or standard probabilistic models for analyzing S-N curves could not fit S-N curve on the whole range of cycles without microstructure information. This information is obtained from costly fractography investigation rarely available in the framework of industrial production. On the other hand, statistical models for fatigue lifetime do not need microstructure information but they could not be used to service life predictions because they have no material interpretation. Moreover, fatigue test results are widely scattered, especially for High Cycle Fatigue region where split S-N curves appear. This is the motivation to propose a new probabilistic model. This model is a specific mixture model based on a fracture mechanic approach, and does not require microstructure information. It makes use of the fact that the fatigue lifetime can be regarded as the sum of the crack initiation and propagation lifes. ▼ Tavunloppuisen /s/:n aspiraatio ja poisjättö on espanjan kielessä laajasti levinnyt ilmiö, jonka merkitys on kiistaton sekä kielen historian että dialektologian ja sosiolingvistiikan näkökulmasta. Tässä tutkielmassa perehdytään tavunloppuisen /s/:n heikentymiseen Extremaduran espanjassa, jossa ilmiö on yksi variantin tunnusomaisimmista piirteistä. Aikaisemmat tutkimukset osoittavat, että /s/:n aspiroituminen on yleisempää sanan sisällä kuin sen lopussa ja että sen poisjättö ja reaalistuminen sibilanttina ovat yleisempiä sanan lopussa kuin sen sisällä. Toisaalta myös äänneympäristö vaikuttaa tavunloppuisen /s/:n allofonien esiintymisfrekvenssiin: aspiraatio on yleisintä ennen konsonanttia, ja sibilanssin säilyminen ja frikatiivin poisjättö on yleisempää ennen vokaalia ja taukoa kuin ennen konsonanttia. Niissä konteksteissa, joissa /s/:n poisjättö ja reaalistuminen sibilanttina ovat yleisimpiä, konservatiivisimmat variantit suosivat sibilanssin säilymistä kun taas innovatiivisimmissa varianteissa dominoi poisjättö. On myös ehdotettu, että tavunloppuisen /s/:n reaalistuminen sibilanttina olisi yleisempää pohjoisessa Extremadurassa kuin autonomian eteläosissa, ja vastaavasti sibilantin poisjättö olisi yleisempää etelässä kuin pohjoisessa. Kyseisen tutkimuksen tulokset ovat suuntaa-antavia, mutta kielivät kielellisen, pohjois-etelä -suuntaisen jatkumon olemassaolosta. Tutkielman tavoitteena on vahvistaa kyseisen jatkumon olemassaolo sekä selvittää, noudattaako Extremaduran espanjan tavunloppuinen /s/ aiemmissa tutkimuksissa määriteltyjä universaaleja tendenssejä. Tutkimusaineisto koostuu kolmestatoista sosiolingvistisestä haastattelusta, jotka ovat osa Madridin Universidad Autónomassa kerättyä Corpus Oral y Sonoro del Español Rural – korpusta ja joiden äänitteet ja ortografiset transkriptiot ovat saatavilla korpuksen omalla internetsivustolla. Haastatteluista etsitään esimerkkejä tavunloppuisesta sibilantista erilaisissa kielellisissä konteksteissa. Esimerkit transkriboidaan foneemisesti, minkä jälkeen käsitellyn aineiston pohjalta suoritetaan laskelmia tavunloppuisen /s/:n reaalistumisesta. Tutkimus osoittaa, että sibilanssin säilyminen on yleisempää Pohjois-Extremadurassa kuin sen eteläosissa ja että frikatiivin poisjättö on yleisempää etelässä kuin pohjoisessa. Lisäksi tutkimuksen perusteella voidaan todeta, että tavunloppuinen /s/ reaalistuu Extremaduran espanjassa pääosin aikaisemmissa tutkimuksissa määriteltyjen tendenssien mukaisesti. Näin ollen Pohjois-Extremaduran alueella tutkitut variantit voidaan luokitella konservatiivisiksi ja eteläisessä Extremadurassa analysoidut variantit innovatiivisiksi varianteiksi.

Aujourd'hui, nos appareils électroniques sont de plus en plus densément composés de composants nanoélectroniques. En conséquence, la dissipation de chaleur produite dans ces circuits augmente également énormément, provoquant une déperdition d’énergie considérable, en pure perte. Les effets thermoélectriques entrent en jeu ici car ils permettent d'utiliser cette chaleur perdue pour produire un travail utile. Par conséquent, l’étude du transport thermique et de l’effet thermoélectrique dans les nanostructures revêt une importance significative du point de vue scientifique et technologique.Dans cette thèse, nous présentons nos études expérimentales du transport thermique et thermoélectrique dans différents types de dispositifs à un seul électron, où le flux électronique peut être contrôlé au niveau de l'électron unique. Tout d’abord, nous montrons la mesure du transport de chaleur contrôlé par la grille dans un transistor à un seul électron (SET), agissant comme un commutateur thermique entre deux réservoirs. Nous déterminons la conductance thermique à l’aide d’un bilan thermique en régime permanent prenant en compte les différents chemins du flux de chaleur. La comparaison de la conductance thermique du SET avec sa conductance électrique indique une forte violation de la loi de Wiedemann-Franz.Deuxièmement, nous étendons l’étude du transport thermique dans les dispositifs à un seul électron dans le régime de boîte quantique, où, outre les interactions de Coulomb, il faut également prendre en compte les différents niveaux électroniques discrets. Nous discutons du bilan thermique entre deux réservoirs de chaleur couplés par un seul niveau de point quantique, et de la dissipation des électrons tunnel dans les contacts. Cela produit des formes de diamant de Coulomb dans la carte de température électronique de la source, en fonction de la polarisation et de la tension de grille.Enfin, nous présentons la mesure du transport thermoélectrique dans une jonction à boîte quantique unique, du régime de couplage faible au régime de couplage fort Kondo. Nos expériences introduisent une nouvelle façon de mesurer le pouvoir thermoélectrique en réalisant une condition de circuit ouvert quasi-parfaite. Le pouvoir thermoélectrique dans une boîte faiblement couplée montre le comportement e-périodique avec la charge induite par la grille, alors qu’il montre une période distincte de 2e en présence de corrélation Kondo. L’étude de la dépendance thermique révèle que la résonance de Kondo n’est pas toujours au niveau de Fermi, mais qu’elle peut être légèrement décalée, en accord avec les prédictions théoriques.Cette étude ouvre la porte à l’étude de transistors à une boîte quantique unique dont les propriétés thermodynamiques sont régies par les lois de thermodynamique quantique.

Ces travaux centrés essentiellement sur la fonction thiohydroximate s’articulent selon trois thèmes : (1) les acides thiohydroxamiques, (2) la synthèse et (3) l’étude de réactivité des N-oxydes de thioimidate (TINOs), avec pour objectif l’étude d’une enzyme : la S-glucosyltransférase (S-UGT). La S-UGT est une enzyme clé dans la formation des glucosinolates. Les travaux menés ont tout d’abord été axés sur la synthèse d’acides thiohydroxamiques, substrats de la S-UGT, pour l’étude cinétique de l’enzyme. Dans ce but, nous avons envisagé différentes méthodes et avons pu obtenir les molécules ciblées, permettant ainsi d’obtenir des résultats préliminaires quant à la cinétique de l’enzyme. D’autre part, la synthèse d’iminosucres, inhibiteurs potentiels de l’enzyme, a été envisagée par une double fonctionnalisation d’un N-oxyde de thioimidate (TINO) sur charpente pyranose. Nous avons donc développé une méthode robuste d’accès aux TINOs à partir de différents pyranoses. Le couplage de Liebeskind-Srogl a permis d’accéder aux nitrones. Des additions-éliminations sur les TINOs nous ont donné accès à des N-oxydes d’amidine et à de nouveaux TINOs. Les réactions de réduction et de débenzylation des TINOs ont également été explorées. Des tentatives de cycloaddition 1,3-dipolaire sur une nitrone ont été effectués en vue d’obtenir à des isoxazolidines, précurseurs d’iminosucres Alpha, Alpha-disubstitués. Les résultats concernant la synthèse d’inhibiteurs de la S-UGT restent préliminaires. Toutefois, après déprotection, les différentes molécules synthétisées durant cette thèse pourront être envisagées comme inhibiteurs de glycosidases.

This thesis is articulating into three topics: (1) synthesis of thiohydroxamic acids, (2) the synthesis and (3) the study of the reactivity of thioimidate N-oxides (TINOs). The purpose of these works is the study of the enzyme: S-glycosyltransferase (S-UGT). The enzyme S-UGT is an essential enzyme for the glucosinolate formation.The starting point of these works was focused on the synthesis of thiohydroxamic acids. These products are the substrates of the enzyme S-UGT and could be used for the study of the enzyme kinetic. The second point is to use TINOs anchored on pyranose backbones as precursors of iminosugars which are potential inhibitors of the enzyme S-UGT. An efficient methodology was developed to access a large panel of TINOs from different pyranoses. Liebeskind Srogl coupling allowed us to obtain nitrones. Addition-elimination on TINOs gave us amidine N-oxides and new TINOs. Reduction and debenzylation reactions on TINOs were also explored. 1,3-Dipolar cycloaddition attempts on a nitrone were performed to obtain isoxazolidines which are precursors Alpha, Alpha-disubstituted iminosugars. The results recording the synthesis of S-UGT inhibitors remain preliminary. However, after deprotection, the different molecules, synthesised during this thesis, will be tested as glycosidase inhibitors.

Le phénomène de défaillance par fatigue peut apparaitre à des niveaux de contraintes éloignés du seuil de dimensionnement statique de mécanique des milieux continus, tout simplement du fait que l’endommagement suit un processus complexe et de longue haleine. Disposer d’outils performants de prévision de durée de vie en fatigue est donc une nécessité de conception pour le dimensionnement des machines, composants, structures, par natures soumises à des sollicitations variables dans le temps, liée à leur utilisation en service. Pour contribuer à cet objectif nous nous sommes attachés à une conception concernant les outils les plus fréquemment utilisés pour ces prévisions en fatigue : les critères multiaxiaux de fatigue et les lois d’endommagement et de cumul en fatigue. La première partie du travail effectué a consisté en une étude de validité et d’analyse de nouveaux critères de fatigue multiaxiaux proposés depuis une dizaine d’années. Cette validité est comparée vis-à-vis de celle des critères implantés dans l’industrie. L’approche intégrale dont le principe est l’établissement du caractère endommageant d’un cycle multiaxial par la contribution des sollicitations présentes sur tous les plans physiques et non un seul comme le cas de l’approche « plan critique », est la plus efficace en règle générale pour traduire correctement le comportement en fatigue sous chargement multiaxial. L’intégration d’une démarche probabiliste proposée permet d’associer la probabilité de survie aux résultats de calcul d’un critère. La seconde partie de cette thèse a conduit à proposer de nouveaux modèles différentiels d’endommagement permettant de retrouver, dans le cas d’un chargement d’amplitude constante, le formalisme classique des courbes S-N des matériaux (Wöhler, Basquin, Bastenaire et Stromeyer). L’influence de l’ordre d’apparition des cycles de contraintes, les valeurs moyennes, l’évolution non linéaire du dommage par fatigue sont ainsi prises en compte. Le problème de sensibilité du modèle et donc ses prévisions de durée de vie en fatigue uniaxiale d’amplitude variable, comme cela est rencontré dans le modèle de Chaboche, est ainsi écarté. La confrontation du modèle proposé pour les courbes S-N de Basquin et Bastenaire est prometteuse en terme de prévision de duré de vie. Une étude comparative par rapport à la loi de Miner, prise comme référence, avec celles proposée et de Mesmacque & Amrouche à donnée des résultats concluants pour les lois à évolution non linéaire.

Analizirana je morfološka i fitohemijska varijabilnost, kao i diferencijacija prirodnih populacija taksona roda Satureja (Lamiaceae) sa prostora Balkana. Morfološki karakteri su izmereni kod 366 individua iz 27 prirodnih populacija S. montana ssp. montana, S. montana ssp. variegata, S. montana ssp. pisidica, S. kitaibelii, S. subspicata ssp. subspicata, S. subspicata ssp. liburnica. Proučavano je 12 morfoloških karaktera (šest karaktera lista i šest čašice). Karakteri čašice su pokazali mnogo manju intrapopulacionu varijabilnost i bolje su odslikavali razlike između taksona, naročito podvrsta unutar vrste S. subspicata. Primenjene univarijante i multivarijantne statističke metode su pokazale različite stepene diferencijacije na različitim nivoima u zavisnosti od vrste. Diferencijacije populacija je pokazana kod S. kitaibelii i delimično kod S. subspicata. Dve podvrste S. subspicata se na osnovu morfoloških karaktera jasno odvajaju. U slučaju S. montana nisu uočene diferencijacije, ni na jednom nivou, zbog velike intrapopulacione varijabilnosti. Uočene su korelacije morfoloških karaktera i pojedinih bioklimatskih parametara, uglavnom vezanih za temperaturu i padavine. Takođe, kod S. subspicata postoji i pozitivna korelacija između morfoloških karaktera i geografskog regiona, što nije bio slučaj kod druge dve vrste. Određen je sastav i relativni udeo komponenti površinskih flavonoida izolovanih sa celih listova kod 108 individua iz 25 prirodnih populacija sa Balkana. Detektovano je 11, a identifikovano 10 jedinjenja. Kod populacija S. subspicata ssp. subspicata je bilo prisutno svih 11 jedinjenja, dok je u zavisnosti od populacije broj jedinjenja kod druge dve vrste varirao 5-10 (S. montana) i 7-10 (S. kitaibelii). U profilu S. montana u većini slučajeva dominiraju 5,6- diOH-7,3'4'-triMe flavon i cirzimaritin, kod S. kitaibelii ksantomikrol i cirzimaritin, a unutar S. subspicata ksantomikrol i cirzimaritin (ssp. liburnica), odnosno ksantomikrol i 8- metoksicirzilineol (ssp. subspicata). Udeo ksantomikrola je najveći kod populacija S. subspicata ssp. liburnica (skoro 60 %). Površinski flavonoidi, analizirani statističkim metodama, na celom setu podataka, pokazuju diferencijaciju između vrsta, uključujući i podvrste. Najviše se odvaja tipska podvrsta S. subspicata od svih ostalih taksona. Iako podvrsta S. montana ssp. montana pokazuje najveću varijabilnost, kontinentalne populacije ove podvrste se preklapaju sa populacijama S. kitaibelii s jedne, dok se primorske preklapaju sa populacijama S. montana ssp. variegata, s druge strane. Populacije S. subspicata ssp. liburnica se po sastavu flavonoida nalaze između ovih primorskih i kontinentalnih populacija. Uočena je korelacija sastava površinskih flavonoida i geografske distribucije populacija, jaka kod S. subspicata i S. kitaibelii, a slaba kod S. montana...

▼ The development of alternative techniques for structural rehabilitation is of critical importance to the safety and sustainability of concrete structures suffered from corrosion of steel. The structural strengthening (SS) and impressed current cathodic protection (ICCP) are conventional methods to improve the safety and sustainability of corroded concrete structures at different perspectives. The SS can recover the load-carrying capacity of deteriorated concrete structures, while the ICCP can retard the corrosion of steel in concrete. Currently, a new strategy to integrate ICCP with SS has been proposed that is termed as ICCP-SS intervention system. Fabric reinforced cementitious matrix (FRCM) composites have emerged as an alternative reinforcement material to fiber reinforced polymer (FRP) composites for structural strengthening and rehabilitation. The carbon-FRCM composite can be adopted as the dual-functional material in the ICCP-SS system in which carbon- FRCM plays the role of reinforcement material in the SS system and of anode material in the ICCP system. To this end, an experimental program was undertaken to propel the application and optimization of the ICCP-SS intervention system. This study aims to clarify the characterization of RC beams with externally bonded carbon- FRCM under static and cyclic loading. Two carbon-FRCM composites were considered for this research, one with multilayer carbon-FRCM (M/C-FRCM) intended for understanding the effectiveness of carbon-FRCM in strengthening beams and one with anodically polarized carbon- FRCM (AP/C-FRCM) for evaluating the mechanical degradation caused by anodic polarization in the ICCP on the strengthening characterization of RC beams. From the material perspective, the mechanical properties of both M/C-FRCM and AP/C-FRCM composites were also studied. The first study investigates the static performance of RC beams with externally bonded M/CFRCM. The static tensile tests of M/C-FRCM were conducted, followed by the four-point bending tests of beams with externally bonded M/C-FRCM under static loading. Nine M/C-FRCM specimens were prepared for tensile tests, and four beams were prepared for bending tests. The prediction model of apparent strain of carbon fabric (CF) mesh in the M/C-FRCM was proposed based on the global strain of carbon-FRCM in tension. The load-carrying capacity of beams with externally bonded M/C-FRCM improved that was largely dependent on the amount of CF mesh. An analytical study on the prediction formulas in flexure for RC beams with externally bonded carbon- FRCM was performed by using the model of apparent strain of CF mesh. Prediction results agreed well with the experimental results. The second study explores the fatigue performance of RC beams with externally bonded M/CFRCM. The cyclic tensile tests of M/C-FRCM were performed and followed by the four-point bending tests of beams with externally bonded M/C-FRCM under cyclic loading. ▼ Offshore oil and gas projects usually require the presence of more than one facilities in the same location. These facilities need to be connected with each other in order to enable the transfer of personnel among each other. As a result, bridges are implemented for such purposes with their ends being positioned at extensions of the two connected platforms, known as the bridge landings. Such a bridge should be able to follow the excitations that are imposed at its two ends from the response of the connected platforms due to the applied environmental loads. Thus, in its longitudinal direction, the bridge should be pinned-supported at one platform and sliding-supported at the other. Such a configuration enables the bridge to adapt to the continuously varying relative movement that is induced by the motion of the two connected platforms. This results in the generation of friction at the sliding end of the bridge. Similarly to any other offshore structure, a bridge landing should be able to withstand the maximum operating loads and its configuration should be checked against the different limit states. Although a jacket substructure is commonly analysed against the serviceability, ultimate and fatigue limit states, a bridge landing is checked against only the first two states. However, the generated friction at the sliding bridge supports results in varying stresses at the corresponding bridge landing. This indicates that the fatigue limit state should also be examined and thus investigation is required in order to highlight its significance in the design of such a structure. This is the motivation behind the certain thesis, which intends to clarify the sensitivity of a bridge landing into the varying dynamic load of the generated friction. In order to do so, a specific case is examined, with real information about the structure and the environmental details. The analysis comprises examining three limit states (SLS, ULS, FLS), concluding into the governing one for the case of the bridge landing. The structural analyses were performed using the SACS software, which enables performing all the SLS and ULS checks. Regarding fatigue, though, the whole analysis was conducted independently, using a simplified approach that enables to deal with the issue in a quick way. This comprises the base case approach, through which assumptions are made regarding the wave and friction main characteristics. After verifying the significance of the fatigue limit state in the design, an assessment of the base case approach follows. This is performed through the examination of the main sensitivity parameters that influence the simplified approach through which the fatigue assessment was conducted. The results of the sensitivity analyses are then incorporated in order to review the method and conclude into any possible improvements. Finally, enhancement of the structure is examined through four different ways, aiming to turn it to be sufficient against the fatigue requirements. This works describes synthesis, characterization and potential cytotoxity of O,O'- -dialkyl-(S,S)-ethylenediamine-N,N'-di-2-(4-methyl)-pentanoic acid, [(S,S)-H4eddl]Cl2, as well as their corresponding palladium(II), platinum(II) and platinum(IV) complexes. Esters (S,S)-R2eddl•2HCl (R = ethyl, n-propyl, n-butyl and n-pentyl) were obtained by instillation of thionyl chloride in an appropriate absolute alcohol, and then refluxing with addition of (S,S)-ethylenediamine-N,N'-di-2-(4-methyl)-pentanoic acid. All of the esters were obtained as dihydrochlorides: (S,S)-Et2eddl•2HCl, (S,S)-Pr2eddl•2HCl, (S,S)-Bu2eddl•2HCl, (S,S)-Pe2eddl•2HCl. Esters were characterized by elemental analysis, IR and NMR spectroscopy, and in the case of (S,S)-Pr2eddl•2HCl the structure was confirmed by X-ray structural analysis. Complexes of palladium(II): [PdCl2{(S,S)-Et2eddl}], [PdCl2{(S,S)-Pr2eddl}], [PdCl2{(S,S)-Bu2eddl}] and [PdCl2{(S,S)-Pe2eddl}] were obtained by reaction of potassium- -tetrachloridopalladate(II) with synthesized esters and characterized by elemental analysis, IR and NMR spectroscopy. Complexes of platinum(II): [PtCl2{(S,S)-Et2eddl}], [PtCl2{(S,S)-Pr2eddl}], [PtCl2{(S,S)-Bu2eddl}] and [PtCl2{(S,S)-Pe2eddl}] were obtained by reaction of potassium- -tetrachloridoplatinate(II) with these esters and characterized by elemental analysis, IR and NMR spectroscopy. Complexes of platinum(IV): [PtCl4{(S,S)-Et2eddl}], [PtCl4{(S,S)-Pr2eddl}], [PtCl4{(S,S)-Bu2eddl}] and [PtCl4{(S,S)-Pe2eddl}] were obtained by reaction of potassium- -tetrachloridoplatinate(II) with mentioned esters and characterized by elemental analysis, IR and NMR spectroscopy. DFT calculations were performed for the synthesized complexes of platinum(II) and platinum(IV) and it was found that the (R,R)-N,N’-configuration isomer is with the lowest energy and formation of only this isomer should be expected, which is consistent with NMR spectroscopy. Cytotoxic activity of synthesized compounds was determined against chronic lymphocyte leukemia cells (CLL) and human cell lines: SW480 colorectal cancer, breast MDA-MB-361 and MDA-MB-453, Jurkat T leukemia and K562 chronic myelogenous leukemia compared with activity of cisplatin, as a reference anticancer drug. All compounds showed higher antitumoral activity then activity cisplatin against CLL calls. It was found that the most active complexes is complexes platinum(II) with n-Bu group in the ester chain. Complexes of the platinum(IV) have an exellent citotoxic activity in the inhibition of Jurkat and K562 cell lines, with is very similar to that shown by activity of cisplatin.

Ce travail de recherche donc concerne l'étude du système ultrasonique de fatigue en torsion de type indirect pouvoir étudier le comportement des matériaux métalliques à haute résistance, essentiellement au domaine de fatigue gigacyclique. Il existe actuellement deux types de système ultrasonique de torsion : un système qualifié de "direct" dans lequel le convertisseur délivre une rotation permettant, de manière simple et directe, la mise en vibration de torsion du système ; un système qualifié de "indirect" qui utilise un convertisseur longitudinal et nécessite une transformation en rotation pour créer la vibration en résonance de torsion. Premièrement, la présente d'étude sur le système indirect permet de comprendre son comportement de fatigue et ses propriétés. Cette étude est présentée à partir des calculs analytiques et numériques afin de dimensionner et modéliser ce système. Ensuite, l'optimisation du système à l'aide de la méthode des éléments finis est réalisée et un système plus effectif et plus robuste est obtenu. Des comparaisons entre les résultats numériques et les résultats de calibration sont proposées et une interprétation des écarts constatés est proposée avec les nouvelles simulations numériques. Deuxièmement, une campagne expérimentale à 20 kHz au-delà de 10⁸ cycles est menée en utilisant le système optimisé sur les aciers 16MnCr5 et 50CrV4 du projet FreqTigue. La méthode de l'escalier est utilisé dans les essais afin de décrire la courbe S-N, d'estimer la résistance à la fatigue et d'obtenir la durée de vie en fatigue. Des comparaisons des courbes S-N à différentes fréquences et sollicitations sont présentées. Les surfaces de rupture des éprouvettes sont analysées pour tenter d'identifier les causes de la rupture.

This research work concerns the study of the torsional ultrasonic fatigue system of indirect type which is able to study the behavior of high-strength metallic materials, mainly in the domain of gigacyclic fatigue. With the developments of technique, there are currently two types of ultrasonic torsional system: the first one is described as "direct" and its converter delivers a rotation which allows the system vibrate in torsion; the second one is described as "indirect", which uses a longitudinal converter and requires the transformation of a translation into a rotation to create a desired torsional resonant vibration in the specimen. Firstly, the study of the indirect system could understand the fatigue behavior and the properties of this system. This study is presented with the analytical and numerical calculations in order to determine and model the indirect system. Then, the optimization of this system by using the finite element method is performed and a more efficient and robust system is developed. Some comparisons between the numerical results and calibration’s results are proposed and an interpretation of differences is proposed by using new numerical simulations. Les travaux rapportés dans ce mémoire concernent le développement de nouvelles réactions métallo-catalysées pour la création de liaison carbone-hétéroatomes ainsi que leurs applications à la synthèse d’inhibiteurs de la protéine de choc thermique 90.Au cours de ce travail, l’étude de la réactivité d’hétérocycles de type quinoxalinones et N-aminoazoles vis-à-vis du couplage de Buchwald-Hartwig a été réalisée. Des conditions ont ainsi pu être développées pour créer la liaison carbone-azote entre des 3-chloroquinoxalinones et des partenaires nucléophiles azotés (amides, azoles…) afin de construire une bibliothèque d’analogues du 6BrCaQ en série quinoxalinone. Par ailleurs, la création de la liaison carbone-azote de motifs N-aminoazoles avec des partenaires hétérocycliques halogénés (coumarines, quinoléines…) ou aromatiques halogénés a été étudiée. Celle-ci a permis l’accès à une chimiothèque d’analogues du 6BrCaQ ainsi que le développement du couplage permettant la mono- ou di-substitutions des motifs N-aminoazoles.Dans le dernier chapitre de ce manuscrit, la création de la liaison carbone-soufre en série glycosidique métallo-catalysés (Pd et Ni) entre des thioglycosides et divers aglycones électrophiles (halogénures (hétéro)aromatiques, vinyliques et acétyleniques) a été examiné. Tous les composés obtenus lors de ce travail sont en cours d’évaluation biologique.

The development of new metal-catalyzed reactions to form carbon-heteroatom bonds have been studied in order to access to hsp90 inhibitors. For this purpose, reactivity of various 3-chloroquinoxalinone have been explored towards Pd-catalyzed Buchwald-Hartwig reaction with nitrogen nucleophiles (amides, azoles…) which allow access to a serie of 6BrCaQ analogues. In the same objective, the reactivity of N-aminoazole moieties in a Buchwald-Hartwig cross-coupling reaction has been also performed. This methodology led to the synthesis of new 6BrCaQ analogues. Moreover, conditions have been defined to access mono- or di-arylated N-aminoazoles structures starting from aryl chlorides. Finally, reactivity studies on metal-catalyzed carbon-sulfur bond forming reaction between thioglycosides as new nucleophiles partners with various aglycon halides ((hetero)aromatics, alkenyls and alkynyls halides) have been performed. Thanks to a nickel- or a palladium-catalysis, we have been able to introduce these thiosugars on various electrophiles partners and complex molecules. Thioglycosylated 6BrCaQ has been thus obtained.Biological evaluations of new synthesized compounds are currently in progress.

▼ L’objectif de cette étude a été de développer et étudier des bioréacteurs anoxiques pour l’élimination du soufre des flux de déchets liquides, et d’évaluer l’intégration du processus pour le traitement simultané des flux gazeux contaminés au H2S et des eaux usées contenant du NO3-.Les expériences relatives à l’oxydation du soufre dans la phase liquide ont été évaluées dans deux bioréacteurs à croissance fixe différents, à savoir un réacteur à lit fluidisé (RLF) et un réacteur filtrant sur lit mobile (RFLM), inoculés par une bactérie ayant la capacité de réduire les nitrates et d’oxyder le soufre (SO-NR). Les bioréacteurs ont été évalués sous différent ratios molaires azote/soufre (N/S) en utilisant le S2O32- et le NO3- comme source d’énergie et accepteur d’électrons, respectivement. Les résultats ont révélé que le RLF et le RFLM sont parvenus à des capacités d’extraction (CE) du S2O32- supérieures à 98 %, et à extraire complètement le NO3- au ratio N/S de 0,5. En conditions de forte limitation en nitrate (ratio N/S de 0,1), la CEx du S2O32- dans le RFLM (37,8 %) était supérieure à celle observée dans le RLF (26,1 %). En conséquence, le RFLM a montré une meilleure résilience à la limitation en nitrate que le RLF puisque la CEx du S2O32- a été ramenée à 94 % en une journée après restauration du ratio N/S à 0,5, alors que le RLF a pris 3 jours pour obtenir une CEx de 80 % pour le S2O32-. Les modèles de réseau neuronal artificiel ont pu être utilisés pour prédire les performances du RLF et du RFLM, à savoir la CE du S2O32- et du NO3-ainsi que la production de sulfate. La biomasse SO-NR développée dans le RFLM a été utilisée pour traiter simultanément les flux de déchets contenant du H2S et du NO3- dans un biofiltre (BF) anoxique. Le BF anoxique a obtenu une capacité d’élimination (CEl) maximale du H2S de 19,2 g de S m-3 h-1 (CEx) pour un apport de 20,0 g S m-3 h-1 (~500 ppmv) en H2S et un ratio N/S d’environ 1,7, respectivement. Comme certaines eaux usées contenant du NO3-peuvent contenir des produits organiques, le RLF anoxique inoculé avec du Paracoccus versatus souche MAL 1HM19 a été développé pour l’extraction simultanée du H2S, du NO3- et du carbone organique contenus dans les flux de déchets. Les résultats ont montré des taux d’extraction respectifs de 16,7 g NO3 – N m-3 h-1 et 42,0 g d’acétate m-3 h-1, ce qui est supérieur aux valeurs observées dans le BF inoculé avec des autotrophes (11,1 g NO3 – N m-3 h-1 et 10,2 g d’acétate m-3 h-1). Le BF anoxique a été opéré sous différentes conditions transitoires (i.e. gaz divers et plusieurs vitesses de flux de ruissellement liquides, un apport intermittent en NO3- et apports élevés en H2S) afin d’évaluer l’impact des modifications sur les variables du processus qui se produisent généralement dans les applications pratiques. Les différentes conditions transitoires ont significativement affecté la CEl du H2S dans le BF anoxique. En appliquant des apports élevés en H2S, la CEx du H2S a été presque totalement rétabli à 100 % dans les 40 heures suivant la reprise de l’opération… Advisors/Committee Members: Lens, Piet N. L.
