▼ The study involved three separate research efforts in which 10 Holstein calves and 85 purebred Hampshire and Suffolk ewe and ram lambs were employed. The objectives of the experiments were to characterize appetite, describe its inheritance and evaluate its associations with animal performance. In the calf experiment, appetite was determined by recording the time required by each calf to completely suckle 3 pints of milk from a standardized feeding bottle. The average suckling speed was 112 seconds with a standard deviation of 38.4 seconds. The maximum and minimum suckling speeds were 298 and 60 seconds, respectively and the coefficient of variation was 34%. A repeatability value of 0.33 was obtained for suckling speed indicating that it might be a moderately heritable trait. There was no significant correlation between MPPA (Most Probable Producing Ability) for appetite and any of the performance characteristics tested. In the lamb experiment, 44 lambs averaging 134.7 lbs. in weight and 10 1/2 months of age and 41 lambs averaging 95 lbs. in weight and 5 1/2 months of age were used in winter of 1974 and summer of 1975, respectively to study the association between appetite and animal performance. Appetite was determined by weighing the feed remaining at the end of 2 one-half hour intervals after the presentation of daily allocation of fresh feed. Appetite was expressed as pounds of feed consumed in 30 or 60 minutes (FC 30, FC 60), the proportion of the 24-hour feed consumption eaten in 30 and 60 minutes, (FC 30/FC 24, FC 60/FC 24), the amount of feed consumed in 30 and 60 minutes per unit of metabolic weight, also called eating rate index (ERI 30, ERI 60), and the average daily feed consumption for the entire experimental period per unit of metabolic weight. The appetite measures were found to be significantly associated with one another (P< .01), suggesting the possibility they are estimates of the same biological characteristic. The repeatability values for appetite ranged from 0.18 to 0.51 in the winter trial and from 0.33 to 0.59 in the summer trial. These values indicated that appetite might be a moderately to highly heritable trait. The males had higher repeatability values in both lamb trials. In the winter trial the males ate significantly faster than the females and also gained more. The difference was 13.6 lbs. and 0.41 lb. at 60 minutes. In the summer trial the males still gained more but there was no significant difference in eating rate. The difference was 9.8 lbs. and 0.10 lb. at 60 minutes. This probably suggests metabolic differences associated with age and sex in relation to appetite, at different stages of the growth cycle. There were significant correlations between total weight gain, average daily gain, and feed efficiency and most of the appetite estimates in the winter trial. None of the above performance characteristics was significantly associated with appetite estimates in the summer lambs. ▼ Between Nov. 2013 and Apr.2014, a serological study was conducted on small ruminants to determine the prevalence of brucellosis, identify risk factors and its public health implication. A total of 1190 blood samples collected from sheep and goat populations in Chifra and Ewa woredas (876 caprine and 314 ovine), 155 (13%) tested positive using mRBPT. Further testing of the positive reactors with CFT confirmed that 147(12.35%) samples were positive for brucella infection. The results revealed that among the risk factors considered in the analysis, species, sex, age, parity number and flock size had statistically significant effect on seropositivity (p<0.05). Woreda had no statistically significant effect on seropositivity (p>0.05). The individual animal level seroprevalence was significantly higher in goats (14.4%) than in sheep (6.7%) populations. The analysis revealed that goats were more than 2 times (OR=2.34) at risk of getting infected with brucella than sheep. The seroprevalence was also significantly higher in female animals (13.8%) than in males (6.5%) with the likelihood of female animals to get infected with brucellosis is higher than that of males. Similarly, Brucella seroprevalence was also significantly increased with age; the odds of seropositivity in older animals are 2.36 times higher than that of younger animals. Seroprevalence rate of brucellosis was also significantly varied with parity, with the likelihood of infection being 2.038 times higher in animals with 3 to 4 parities than otherwise. Regarding flock size individual animal sero-prevalence was higher in larger flocks than in the smaller ones (OR=0.68). Almost all the respondents in the studied areas were not aware of brucellosis as a disease affecting different species of livestock (91.1%) although all of them recognized the existence of abortion among small ruminant flocks and most of them handle abortion materials with bare hand without protecting themselves (82.2%). The habit of drinking raw milk is practiced by the majority of the respondents 45 (100%) while there is no habit of consuming raw meat. Poor awareness of the zoonotic importance of brucellosis and the practices of consuming raw milk and handling potentially infectious materials using bare hands pose a serious danger to small ruminant owners. Finally, this study demonstrated that small ruminant brucellosis is highly prevalent in the study districts. Hence, it is suggested that there is a need for planning and implementation of joint programs by stakeholders in prevention and control of the disease as well as raising public health awareness. ▼ In the U.S., gastrointestinal fermentation from cattle is estimated to account for approximately 25% of total anthropogenic related methane (CH4) emissions. In addition, 2-8% of gross energy consumed by cattle is lost in the form of CH4, representing an energetic cost to the animal. Thereby, in order to decrease greenhouse gas emissions (GHG) and improve the efficiency of cattle production additional research on gastrointestinal CH4 emissions from cattle is needed. In ruminants carbohydrate (CHO) catabolism and nitrogen (N) utilization have a tremendous impact on ruminal methanogenesis. However, the impact of purified carbohydrates in the presence of a variety of N sources on rates of CH4 and VFA production remains unknown. In order to determine these rates for use in predictive models of the ruminal fermentation, we formulated a fractional rate equation to fit the rate of CH4 production and measured the concentration of CH4 and VFA and using purified CHO with a variety of N sources in two in vitro mixed ruminal microorganism fermentation studies. In both studies, a CHO treatment × incubation time (IT) effect was observed for both VFA and CH4 (P < 0.01). There was also an N × IT interaction for CH4 production at 24 h in Study 2, where nitrogen free and NH3 treatments produced greater concentrations of CH4 than treatments with amino acids (P < 0.01). A nonlinear equation for the conversion of carbohydrates to CH4 was able to fit starch treatments in Study 1 and glucose treatments in Study 2. Overall, this study demonstrated different fermentation patterns among all CHO and N sources and was the first step in determining rates for in vitro CH4 production. Although cattle contribute with high amounts of anthropologic GHG, they are not the only methanogenic producing food source in the U.S. Rice and wild ruminants (e.g. bison, elk, and deer) are also methanogenic producing food sources. The objective of this final study was to compare the efficiency of beef and milk production to pre-settlement wild ruminants and rice production on a kilogram of CH4 emitted to kilogram human-edible protein production basis. Bison had the highest ratio of 13.93 kg CH4: Protein, followed by elk (12.50) deer (6.66) and beef (2.47). Overall, wild ruminants emitted 296 to 564 percent more CH4 per kilogram of human-edible protein produced than current beef cattle production systems. Rice yielded the second lowest CH4 to human-edible protein ratio (0.83), followed by dairy cattle milk production (0.50). We believe, this analysis provides insight on the efficiency of methanogenic food sources that may aid in the development of regulatory guidelines of CH4 production. ▼ La peste des petits ruminants est une maladie infectieuse animale très contagieuse. Largement répandue en Afrique, au Moyen Orient, et en Asie, elle fait des ravages dans les élevages ovins et caprins. Les petits ruminants représentent une ressource nutritionnelle et économique essentielle dans les pays en développement, notamment pour les communautés rurales les plus pauvres. En Afrique sub-saharienne, l'impact de la maladie est d'autant plus élevé que les mouvements des animaux sont compliqués à contrôler (transhumance, commerce illégal, zones de conflits, etc.) et les contrôles sanitaires difficiles à organiser. Une coalition internationale a décidé de prendre en main le contrôle de cette maladie ravageuse en élaborant avec l'OIE et la FAO une stratégie mondiale de contrôle progressif et d'éradication de la maladie.La stratégie est basée sur la vaccination de masse mais les protocoles de vaccination ont principalement été élaborés sur des bases empiriques. Après une campagne de vaccination, la dynamique démographique entraînant le renouvellement du cheptel, la proportion d'individus immunisés au sein de la population (taux d'immunité) est amenée à diminuer au cours du temps (entrées d'animaux non vaccinés, e.g. les naissances, et sorties d'animaux vaccinés, e.g. les ventes). La décroissance du taux d'immunité permet d'évaluer l'efficacité de la vaccination à moyen terme. Si le taux d'immunité est assez élevé, le virus n'a plus d'hôte pour se propager. Le renouvellement du cheptel est délicat à estimer et varie d'un système d'élevage à l'autre. Ce manuscrit présente une méthode d'optimisation des protocoles de vaccination pour obtenir la meilleure couverture immunitaire possible.Un modèle dynamique de prédiction du taux d'immunité dans des élevages de petits ruminants d'Afrique sub-saharienne au cours d'un programme de vaccination pluri-annuel a été développé en utilisant la théorie des modèles démographiques matriciels. Cet outil a été utilisé pour évaluer différents protocoles proposés pour les zones sahéliennes arides / semi-arides et les zones sub-humides / humides. Les paramètres des modèles ont été estimés à partir des données disponibles et d'une revue exhaustive de la littérature.Des indicateurs synthétiques de l'efficacité des protocoles ont été calculés (persistance du niveau protecteur, taux d'immunité moyen, etc.), puis comparés.L'étude a confirmé la pertinence des protocoles proposés par l'OIE et la FAO, apportant des précisions pour les divers scénarios. Les couvertures vaccinales atteintes doivent être très élevées (>80%) pour permettre la protection d'un troupeau pendant toute la durée du programme. En zone sahélienne, les troupeaux doivent être vaccinés au plus tôt après la saison des pluies pour optimiser la portée de la vaccination. ▼ L’amélioration génétique des caprins laitiers est basée, à ce jour, sur la mesure de la quantité et de la qualité de la production laitière des femelles en ferme, à intervalles de 4 à 5 semaines au cours de la lactation, selon des protocoles stricts. L’évaluation génétique de la quantité de lait repose sur l’estimation de la quantité de lait totale produite par lactation. Cette sélection à partir de la quantité totale de lait à la lactation a tendance à sélectionner des animaux avec une production au pic de lactation de plus en plus élevée. Une production importante en début de lactation peut être à l’origine, pour les femelles, de problèmes métaboliques. De plus, en caprins laitiers, dans un contexte de production saisonnée, une production laitière qui se maintient après le pic, i.e. persistante, permettrait une production laitière plus étalée, en lien avec les attentes du marché. Il y a donc un intérêt zootechnique et économique de vouloir sélectionner des chèvres laitières plus persistantes. Dans notre étude, l’approche consiste à modéliser la forme de la courbe de lactation à partir des informations recueillies lors de chaque contrôle en ferme. Les modèles permettant l’analyse de telles données longitudinales sont généralement appelés modèles de contrôles élémentaires. L’un des principaux intérêts, est une meilleure prise en compte d’effets d’environnement, affectant la production le jour du contrôle, avec un effet troupeau-jour de contrôle ne dépendant que des animaux présents lors du contrôle. Le deuxième avantage de ce type de modèle réside dans la modélisation de la plupart des effets génétiques et d’environnement sous forme de courbes ; il serait donc possible de sélectionner les animaux avec la meilleure valeur génétique pour la persistance. La mise au point de ces modèles nécessite l’étude préalable des effets d’environnement affectant la production laitière au cours du temps. Suite à une analyse descriptive détaillée des courbes de lactation des deux principales races caprines françaises (Alpine et Saanen), nous avons montré qu’il existait une variabilité de la forme de courbes de lactation, et en particulier que le mois de mise bas était impliqué dans les différentes formes de courbe. Nous avons ensuite proposé un modèle de régression aléatoire, proche de celui développé chez les bovins laitiers français. La modélisation proposée permet d’obtenir directement deux index génétiques : un premier correspondant à la valeur génétique de l’animal pour la quantité totale de lait au cours de la lactation et un deuxième correspondant à une valeur génétique de la persistance laitière de l’animal, et cela sans corrélation entre les deux. Le modèle développé permet de prendre en compte de façon disjointe les chèvres en primipares et les chèvres en multipares. Nous avons également étudié les corrélations entre index de différents caractères, au cours de la lactation ainsi que les corrélations entre persistance et fertilité à l’IA ou entre persistance et longévité. ▼ Ruminant utilisation of poor quality feeds is governed by rates of degradation and passage through the rumen. Firstly, the passage rate of feed material determines the degree of bypass nutrients and the efficiency of synthesis of microbial protein in the rumen, making modelling of passage rate important. Secondly, diurnal feeding behaviours are not normally used in predicting feed intake although their influences are vital in understanding the dynamics of intake. Lastly, critical to rumen kinetics studies lies in understanding the dynamics of rumen fill levels post meal termination. The objectives of the study were to: (1) develop digesta passage rate prediction models for climatically, nutritionally and genetically diverse classes of ruminant herbivores; (2) ascertain the effects of diet quality on diurnal feeding behaviour in sheep and goats; and (3) determine the influence of diet quality on passage rates and, the extent and trend of solid digesta disappearance after meal termination. Artificial neural networks were used to develop prediction models for liquid and solid passage rates. Studies that reported fractional passage rates, class and body mass of ruminants were included in the dataset. Factors that affect rates of passage included animal and feed factors. The database was composed of observations of domestic and wild ruminants of variable body mass (1.5 to 1238 kg) from 74 studies and 17 ruminant species from different climatic regions. Observations were randomly divided into 2 data subsets: 75% for training and 25% for validation. Developed models accounted for 66 and 82% of the variation in prediction of passage rates for solid and liquid, respectively. On validation using an independent dataset, these models attained 42 and 64% of precision in predicting passage rates for solid and liquid, respectively. The effects of tropical roughage and diet quality on dry matter intake, duration and number of daytime and night time eating bouts, idling sessions and ruminating activities in small ruminants were investigated. Roughage quality was improved by urea treatment of veld hay, while diet quality was improved by supplementing with lucerne hay, sunflower meal, lespedeza, fish meal and sunflower meal. Day-time (0600-1800 h) and night time (1800-0600 h) feeding behaviour activities of sheep and goats were recorded. Generally, roughage and diet quality, and time of the day had significant effects on time spent ruminating and eating. Intake rates (g/bout and g/min) were not affected by diet and roughage qualities. Generally, goats and sheep fed on roughage alone ruminate at night and eat more during the day but those fed roughage and supplemented with lucerne hay spent more time ruminating than eating. Time spent eating and ruminating had positive correlations to feed intake. Intake rates (g/min and g/bout) had strong positive correlations to intake, which were significant. O objectivo deste trabalho é descrever as principais aplicações da laparoscopia em ruminantes e o uso desta técnica na correcção do deslocamento abomasal, de forma regular, na clínica de animais de pecuária. A área da endoscopia tem tido uma evolução notável nas últimas décadas, para a qual têm também contribuído os avanços verificados a nível tecnológico. Em laparoscopia de ruminantes são geralmente usados laparoscópios rígidos e as principais vantagens desta cirurgia incluem o facto de ser mini-invasiva, a redução da duração do procedimento, poucas complicações e tempo de recuperação pós-cirúrgico mais curto. As principais desvantagens incluem o preço do equipamento, a curva de aprendizagem e a perda de sensação táctil em comparação com a cirurgia aberta. São indicados neste trabalho os tipos de equipamento comummente usados e sugestões de cuidados pré e pós cirúrgicos. A anatomia abdominal observada através do laparoscópio e possíveis abordagens cirúrgicas são também referidas. No que diz respeito ao deslocamento abomasal, para além de várias técnicas disponíveis para a sua correcção, é descrita a patogenia da doença, os factores de risco, a influência de doenças concomitantes e é apresentado um conjunto de casos observados na Clínica para Ruminantes (medicina interna e cirurgia) em Gießen submetidos a cirurgia laparoscópica segundo o método de Janowitz. A abomasopéxia por meio de colocação de toggle abomasal, seja pelo método de Janowitz, Christiansen ou Newman, Anderson e Silveira, são técnicas que podem ser praticadas com facilidade na clínica de campo.

ABSTRACT - Laparoscopy in ruminants: using a mini-invasive procedure for correction of the leftsided displaced abomasum. - The aim of this dissertation is to describe the main applications of laparoscopy in ruminants and the use of this method for the correction of the displaced abomasum, in a regular basis, in farm animals’ practice. The evolution on the endoscopy field during the past decades was remarkable, for what is also important the great advances verified in technology area. Rigid laparoscopes are commonly used in ruminants’ laparoscopy. Some advantages that are related with this kind of surgery are the mini-invasiveness, the reduction of the surgical time, the few complications and a recovery period faster than with conventional surgery. The main drawbacks are the price of the equipment, the learning curve and the lost of the tactile sensation comparing with open surgery. In this paper, there is a review of the equipment commonly used and some suggestions for pre and post-surgical care. The laparoscopic abdominal anatomy and some possible approaches are also referred. Cowdria ruminantium (Cowdria) is an obligate intracellular pathogen that causes heartwater in ruminants. Cellular immunity and the type I cytokine IFN-γ have been implicated in protective immunity to heartwater. The aim of this thesis was to identify proteins of the Welgevonden isolate of Cowdria that induce lymphocyte proliferation and IFN-γ production. Differential centrifugation was found to be the simplest and most efficient method of Cowdria purification. Cowdria organisms were fractionated into their constituent proteins of between 11 and 168 kDa by continuous flow electrophoresis. The resulting fractions were tested for their ability to stimulate lymphocyte proliferation in vitro. In an attempt to simulate the natural infective process, peripheral blood mononuclear cells (PBMC) were obtained from two cattle rendered immune by infection and treatment and assayed in proliferation assays with the proteins fractions. In a parallel study, four cattle were immunised with inactivated Cowdria to determine whether their lymphocytes responded similarly. Cowdria-specific proliferation was detected for only a brief period after immunisation by infection with live organisms. This response was only detected again two to three years later. In contrast, PBMC from animals immunised with inactivated organisms were continuously responsive for at least three years. Only Cowdria proteins with molecular masses of 11, 12, 14 to 17 and 19 to 23 kDa induced proliferative responses in PBMC obtained from all six animals. Cell surface phenotypic analysis of Cowdria specific T-cell lines indicated that CD4⁺ lymphocytes were enriched over time with a concomitant increase in antigen-specific proliferation and IFN-γ production. Proteins of molecular masses 13 to 18 kDa induced CD4⁺ lymphocyte proliferation and IFN-γ production by T-cell lines from all the animals tested. Antibodies raised in a chicken and in rabbits to these low molecular weight proteins had low titres and specificity. Two-dimensional electrophoresis indicated that proteins within a single molecular weight range comprised several components with different pIs, revealing the complexity of the Cowdria proteome. This complicates the search for potentially protective antigens. Nevertheless, since they cause proliferation and IFN-γ production by lymphocytes from immunised cattle, these low molecular weight proteins merit further investigation as potential vaccine antigens.

▼ To improve the ability to predict amino acid flows in ruminants, which will enhance the efficiency of use of feed nitrogen (N), methods to better predict metabolizable protein (MP) and amino acid supply need to be developed. The objectives of this study were (1) to evaluate and improve the efficiency of precipitable true protein assay to understand the peptide profile that is chelated to better predict the supply of those components to support the MP supply of the cow; (2) to evaluate Streptomyces griseus assay as indicator for AA profile of MP supply; and (3) evaluate variation with published total digestion assays and make improvements to better estimate intestinal protein digestion of feeds for ruminants. Throughout, methods were evaluated or developed with commercial labs in mind so values could routinely be analyzed for use in the CNCPS. The efficiency of the precipitated true protein (PTP) assay was improved with tungstic acid (TA) or stabilized TA filtered on 1 or 6 [MICRO SIGN]m filters (P < 0.05) but was time consuming as were ultrafiltration devices. Nonprotein N as assayed contains substantial quantities of amino acids and peptides. The Strep. griseus assay will predict rumen degradation of N but will not predict AA supply or digest carbohydrates and fiber. An in vitro (IV) assay to estimate intestinal protein digestion for ruminants was developed using an enzyme mix of trypsin, chymotrypsin, lipase and amylase at activities found in sheep digesta to replace pancreatin, reduced sample loss by eliminating the use of bags by employing Erlenmeyer flasks, utilized small pore size filter papers to further improve recoveries of undigested feed N and corrects for microbial contamination with corn silage ND. Acid detergent insoluble (ADI) N is digestible; thus, undigested N from the IV intestinal assay more accurately represents the protein C fraction than does ADIN given the more physiological conditions of rumen incubation and enzyme exposure. The IV assay provides a method, adoptable by commercial labs, to evaluate intestinal digestibility of ruminant feeds to better predict the MP and amino acid supply for input into the CNCPS. ▼ A cross-sectional study design was employed between November 2013 and April 2014 to determine antibody/immunity level of vaccinated sheep and goats at herd level, to determine the sero-prevalence and risk factors of PPR in non-vaccinated areas and to investigate incidence and recent circulation of PPR virus in field samples collected from small ruminants of eastern Amhara region bordering Afar, Ethiopia. A total of 969 serum samples were collected from sheep and goats in the study districts. Multistage sampling, with four hierarchical stages, was used as sampling strategy. Peasant Association /villages and individual animal were selected by random sampling. A total of 32 samples, comprising 18 swab samples and 14 heparinized whole blood samples were collected from the suspected animals for the presence of PPR viral antigen. 28 samples were examined for the presence of PPRV RNA by one step RT-PCR assays. Anti-mortem samples were collected from active cases with the aim of virus isolation. In addition five years retrospective data on PPR outbreak was used in the present study. Questionnaire survey was also conducted to assess the association of seroprevalence of PPR and possible risk factors. Proportions were calculated for seroprevalence visa-vis fixed factors that included animal species, sex and age, districts and village. Univariable analysis for the proportions was carried out using Chi-square analysis in Epi Info software. A confidence limit of less than 5% was used to indicate a significant level. All variables with P < 0.05 (two-sided) in the univariable analysis were further tested by multivariable logistic regression model to assess their effect on PPR seropositivity. Clinical signs of PPR were observed in both sheep and goats, in both sex and all agegroups, in the all study districts. The clinical signs included high fever, ocular and nasal discharge, few abortions, respiratory distress and diarrhea. The disease outbreak was severe in Habru district with morbidity, mortality and case fatality rates of 21.9%, 8.4% and 38.4 % in small ruminants, respectively. The overall seroprevalence of PPR virus antibody was 28.1%, 64.5% and 56.5% in unvaccinated, vaccinated and unknown vaccination status of small ruminants, respectively. The multivariable logistic regression model revealed residing in Rayakobo district, adult age, communal grazing and recent introduction of new animals as risk factors for PPR seropositivity in sheep and goats. There was statistically significant (p= 0.000) differences in the level of seroconversion among the vaccinated districts with similar vaccine response of different age groups, sex, and species involved. In unvaccinated population around 28.1% and only 24.5% were with PI > 50% and 76%, respectively, whereas in vaccinated population around 64.5% and 58.8% of animals were with PI > 50% and 76%, respectively. ▼ The current research was done to evaluate the nutritive values and the ruminal degradation of dry matter (DM) and crude protein (CP) from three non-conventional oil cakes, viz: amarula (Sclerocarya birrea) (AOC), macadamia (Integrifolia) (MOC) and baobab (Adansonia digitate L.) (BOC). The oil cakes were collected from biodiesel producers in Limpopo Province, transported to the ARC-Animal Production campus, where proximate and ruminal nutrient degradation analysis were conducted. Triplicates samples from each oil cake were analyzed for the nutritive values, mineral and amino acids contents. Three rumen cannulated mid-lactating (days in milk; DIM: 180±5) Holstein cows weighing 667±43 kg body weight were allocated to determine the in situ ruminal dry matter (DM) and crude protein (CP) degradation. The cows were offered a totally mixed ration (TMR) (60 concentrate: 40 forage ratio) that was compounded according to their daily nutrient requirements, and were milking was done twice per day at 12 hrs intervals. The three oil cake samples were ground using a 2-mm screen after which sub-samples (6.5 g) were put in 10 x 20 cm; 50 μm pore size polyester bags to achieve 15 mg/cm² (ratio of the sample size to surface area). The bags were then fistulated in each cow’s rumen in triplicate for a period of 2, 4, 8, 16, 24, or 48 hrs. After being incubated, the bags were removed from the rumen and washed with cold (4°C) water in 20-L buckets. Following immersing in cold water, the bags were machine washed until clean water was obtained. The bags were then dried at 60 °C in an oven for 48 hrs. The dried bags were individually weighed, and the content of each bag were removed and stored into glass vial until analysis. The remaining two duplicate sets of each sample were rinsed using cold water in order to determine solubility at 0 hrs. The AOC had higher (P<0.05) ether extract (EE) and CP content than both BOC and MOC. Macadamia oilcake (MOC) and BOC had higher (P<0.05) fractions of fibre (NDF, ADF and ADL) compared to the AOC. The AOC had greater (P<0.05) content of essential amino acids than in the BOC and MOC. Additionally, AOC had a high (P<0.05) phosphorus, but low calcium and potassium concentration. While AOC had high effective degradability of DM, it also had high water soluble as well as DM and CP rapidly degradable fractions. Effective degradation of CP was higher in AOC and BOC than in MOC. However, BOC had a high insoluble but degradable fraction of CP. ▼ The mechanisms of how conceptus-derived interferon tau (IFNT) induces interferon stimulated genes (ISGs) and cell survival genes through endocrine action that contributes to resistance of the Corpus Lutum (CL) to prostaglandin Fα (PGF) and the maintenance of early pregnancy in sheep were examined. Using microarray screens, several genes were identified in the CL to be significantly up-regulated [ISG15 and myxovirus (influenza virus) Resistance 1; MX1], maintained [interleukin 6; IL-6, Pentraxin, long 3; PTX3, luteinizing hormone receptor; LHR, and vascular endothelial growth factor; VEGF] or down-regulated [serpin peptidase inhibitor, clade E; SERPINE1, thrombospondin 1; THBS1] in response to pregnancy and luteolysis. These studies in the CL were expanded to other tissues (endometrium, liver, uterine vein tissue) and bodily fluids (blood and histotroph) to identify other endocrine actions of IFNT and other conceptus secretory products that could be used to explain mechanisms of establishment and maintenance of pregnancy and to be used as blood markers for pregnancy status. The extensive examination of key genes described herein that were differentially regulated during Days 12-15 of the estrous cycle and pregnancy in ewes is novel. The experiments are the first to examine extensive temporal regulation of key genes associated with luteolysis such as estrogen receptor (ESR1), oxytocin receptor (OXTR), prostaglandin transporter (SCLO2A1) and caspase 3 (CASP3); and luteotrophic-cell survival genes such as Phosphoinositide-3-Kinase/V-Akt Murine Thymoma Viral Oncogene (PI3K/AKT) and ISGS. To identify novel markers of pregnancy status and establishment of pregnancy, global mass spectroscopy was used for analysis of the proteome and metabolome. From this extensive analysis, 14 proteins/metabolites were more extensively examined: Acetyl-carnitine, Carnitine, Ecdysteroids, N-acetyldileucine, Valine, Collagen, Type I, Alpha 1 (COL1A1), Collagen, Type I, Alpha2, (COL1A2) Annexin A1, Annexin A2, Annexin A5, IFNT, Trophoblast Kunitz Domain Protein 1 (TDKP1) Profilin 1 (PFN1) and S100 Calcium-Binding Protein A11 (S100a11). The critical roles for these metabolites and proteins are discussed in context of the establishment and survival of a pregnancy. To confirm that IFNT has an endocrine role during early pregnancy, a highly specific [no cross-reaction with Interferon alpha, beta and gamma (IFNA, IFNB or IFNG)] and sensitive (23.95 pg/ml in serum) IFNT radioimmunoassay was validated herein. IFNT could be detected using this refined assay in ewes from Days 13-16 of pregnancy in histotroph, in uterine vein serum by Day 15-16 of pregnancy and as early as Day 19 of pregnancy in tail blood from pregnant dairy cows. Through these studies it was determined that Day 14 is a very pivotal day for the establishment of pregnancy in ewes. Detection of IFNT in tail blood of pregnant cows on Day 19 provides further evidence for the endocrine action and systemic release of IFNT during pregnancy in ruminants. ▼ Three experiments were conducted to evaluate the effects of increasing the tetany risk ratio on blood serum Mg levels, nutrient digestion, mineral balance, and Mg supplementation source and feeding behavior. Experiments 1 and 2 were randomized complete block designs, with 24 wethers were maintained in individual metabolism crates and assigned to four treatments (6 wethers/treatment). Experiment 1 compared tetany risk ratios (TRR) of 1.6, 2.3, 2.9, or 3.5. Dry matter, NDF, and N digestibilities were not different between treatments. Nitrogen retention decreased (P < 0.05) as the tetany risk ratio increased. No treatment differences were recorded in Mg, Ca, K, or P balance. The TRR 2.3 treatment had the greatest decrease (P < 0.05) in serum Mg after 15d. Experiment 2 compared tetany risk ratios of 1.5, 2.6, 1.5 plus MgO, or 1.5 plus MgCl. The TRR 2.6 treatment showed increased (P < 0.05) DM, NDF, and N digestibilities when compared to all other treatments. Nitrogen digestibility decreased (P < 0.05) with the addition of supplemental Mg. No treatment differences in Mg, Ca, K, or P balance were recorded. The TRR 2.6 treatment had the greatest decrease (P < 0.05) in serum Mg after 5 d. Experiment 3 was a cafeteria study using a switchback design, 23 Angus heifers were weighed and randomly assigned to one of two locations (11 heifers in drylot, and 12 heifers on pasture) containing two mineral supplements (0.0% Mg and 10.0% Mg). The groups were rotated between locations after 15 d for 30 d of measurements. Individual mineral consumption (grams/d), feeder attendance (trips/d), and feeding duration (seconds/d) were measured using a GrowSafe® individual feeding system. Heifers consumed 119% more (P < 0.01) 0.0% Mg supplement each day than the 10.0% Mg. Heifers made almost twice as many (P < 0.01) trips to the feeder, and spent an additional 91.3 s consuming the 0.0% Mg than the 10.0% Mg. Total mineral intakes were 87.3 % greater (P < 0.01) when supplemented on pasture compared to drylot. During the first 15 d, total mineral intakes were higher (P < 0.01) than for the second 15 d. ▼ Enteric methane produced by ruminants is a dietary inefficiency that contributes to global warming. Feeding diets containing starch to ruminants has been reported to decrease enteric methane emissions. The feeding of methane mitigating agents such as supplemental fat, nitrate and 3-nitrooxypropanol (3- NOP) to ruminants has also been reported to decrease enteric methane emissions, but the degree of methane mitigation may depend upon the basal diet offered to the animal. Furthermore, data are lacking on the net impact of 3-NOP and nitrate on whole farm greenhouse gas (GHG) emissions across different production systems. The aim of this thesis was to quantify the productivity and mitigation potential of these methane mitigating agents when they were included in different diets, and to model the effects of feeding 3-NOP and nitrate on whole farm GHG emissions and on the economics of dairy and beef farms. Initially, the methane mitigating agents were tested in vitro, they decreased methane production (MP) and their methane mitigating efficacies were not affected by basal diet. In the following in vivo experiment feeding supplemental fat to dairy cows decreased methane yield in cows fed either wheat or corn based diets. A modelling study predicted that both 3-NOP and nitrate would decrease whole farm GHG emissions; however, 3-NOP had a greater effect on enteric methane, and thus whole farm emissions, than nitrate. The research of this thesis indicates that the antimethanogenic responses to fat, nitrate and 3-NOP are not affected by substrate type, suggesting that these compounds could be effective in ruminants fed wheat grain, which is routinely fed to ruminants in Australia. Feeding 3-NOP could be economical for beef and dairy farms, depending on the cost of 3-NOP. It is concluded that 3-NOP could make an important contribution to reducing whole farm GHG emissions; however, either a production benefit would have to be demonstrated or a carbon offset method would have to be in place to incentivize its use in the livestock industries.

▼ Schmallenberg virus (SBV) is a novel orthobunyavirus in the Simbu serogroup, genus Orthobunyavirus, family Peribunyaviridae, and order Bunyavirales. The virus emerged in late 2011 near the German/Dutch border region and was mostly associated with a mild transient disease of sheep and cattle. It is transmitted by biting midges (Culicoides species) and causes abortions, stillbirths, and congenital defects in naïve pregnant sheep and cattle. SBV has spread throughout most of Europe. However, to date, the initial introduction route of the virus to Europe is poorly understood. Consequently, SBV poses a threat to other countries like the US, where competent insect vector species and susceptible livestock populations exist and extensive trade with Europe is conducted. To this end, research work was conducted with the following three aims: (1) development of a large animal model for SBV infection; (2) subunit vaccine development for SBV; and (3) development and evaluation of Schmallenberg disease diagnostics. This dissertation contains four chapters. The first chapter provides a literature review on SBV emergence, taxonomy, replication, transmission, pathogenesis, diagnosis, and control. The remaining three chapters contain the research conducted to address the above-mentioned three study aims. In chapter two, the clinical, virological, and serological responses in two ruminant models, i.e., cattle and sheep, are presented. Infectious serum was shown to be the best inoculum in both species when compared to infectious cell culture supernatant and brain homogenate. The virological and serological responses to SBV were more apparent in cattle than in sheep. Thus, cattle are recommended as the better SBV infection model for the evaluation of vaccines and diagnostics. Chapter three presents the immunogenicity and efficacy of a baculovirus-expressed subunit vaccine composed of SBV glycoproteins C and N. After vaccination, SBV Gc-specific antibody response was detected in all vaccinated animals. Neither of the vaccines conferred protection against SBV challenge infection. Therefore, future studies should focus on better understanding of the difference in post-translational modifications on Gc protein in different expression systems and subsequent conformational and stability conditions that are crucial for its immunogenicity. Using the different reagents (sera, recombinant proteins, and tissues) generated in the SBV animal model development and vaccine experiments, we tried to develop and evaluate serological and molecular diagnostic assays for SBV. Among the different SBV proteins evaluated for SBV antibody detection, N and Gc were the most reactive antigens, followed by Gn. In the indirect ELISA experiments, the SBV-infected cell lysate was the least reactive. Among the three commercial nucleic acid extraction kits, the GeneReach total RNA extraction kit yielded approximately 10× more SBV in vitro transcribed RNA than the Qiagen and Applied Biosystems extraction kits. ▼ The zoonotic disease leptospirosis was first recognized in 1886 as heamoragic fibrile disease of man. The cause, Leptospira spp., family Leptospiraceae and order Sprirochetales, was isolated a few years later from rodents and other animals. During the 100 years of fascinating research, became clear that the pathogen is very important not only for Public Health, but also the productivity of food producing animals. Animal (wild and domestic) infections maintain pathogenic serovars in nature, if climatic factors (humidity and temperature) are favorable causing new infection to other animals and accidental infections to man entering a contaminated environment. Between these animals are also small ruminants.Seroprevalence of small ruminats could reach 60%, if the environmental factors favor the pathogen, and the worldwide serotypes identified serologically are Autumnalis, Castellonis, Gripotyphosa, Icterohaemorrhagiae, Hardjo, Sejroe, Sermani and others. In Greece, very little was known about leptospirosis in general until 2002, when an attempt was made to research the infection. This attempt showed that seroprevalence was about 24.1% among small ruminant abortion cases compared to 11% from non-cases. These findings showed that the consequences from this infection are completely unknown in Greece, thus unknown is also the contribution of food producing animals to human cases.For the above reasons, we decided to study the pathogenicity of Leptospira spp in small ruminants, because these animal species produce the majority of food products in Greece. For this purpose, Peloponissos (Southern Greece) was chosen as the area of concern for the following reasons: 1.KE.E.L.P.NO has recorded prefectures of Ilia and Messinia among those recording the highest number of human clinical leptospirosis in Greece2.The prefectures belong to the Veterinary Laboratory of Tripolis, with which we had good working relationship and 3.In Peloponissos we had good relationship with many other veterinarians of the State Veterinary Agencies. ▼ The purpose of this investigation was to provide direct experimental evidence for or against the suggestion that amino acids and B vitamins are among the nutrients absorbed from the rumen of the ruminant. The investigation was subdivided into two parts: (1) six ruminal fistula experiments and (2) nine blood-sampling experiments. The goat (Capra) was the experimental animal. In the ruminal fistula experiments, solutions of amino acids, B vitamins, and usually propionic acid were added to an empty rumen for 120 or 150 minutes. Propionic acid, a substance known to be absorbed from the rumen, was included in most of the solutions to provide a positive control for ruminal absorption. In addition, in all but the first two experiments, polyethylene glycol was added to the solutions in order to provide a marker substance for differential water movement into or out of the rumen. Controls for the experiments included the taking of a sample from the solution in the rumen at zero time and incubating the sample for the duration of the experiment in a water-bath held at 37°C. The ruminal and control solutions were sampled at periodic intervals during the experiment. The results demonstrated not only marked decreases in the concentrations of propionic acid in the ruminal solutions but also, in most cases, marked decreases in the concentrations of the amino acids and the B vitamins assayed: tryptophan, methionine, tyrosine, glycine, lysine, riboflavin, nicotinic acid, and pantothenic acid. In contrast, the concentrations of these constituents in the control solutions usually demonstrated either no decrease or a small increase. In the first three blood-sampling experiments, the concentrations of amino acids and nicotinic acid in the plasma draining the rumen were compared with the concentrations in the peripheral plasma. The results demonstrated that seldom were the concentrations of an amino acid or nicotinic acid higher in the plasma draining the rumen than in the peripheral plasma. The results did suggest, however, that there had been slight increases in the plasma concentrations of the amino acids with time. Accordingly, the next six blood-sampling experiments were designed to determine if time-related increases in the blood concentration of alpha amino nitrogen occurred after the addition of amino acids to the rumen. Blood samples were removed periodically from anesthetized goats, both before and after the addition of a solution of amino acids to an empty rumen. The results demonstrated time-related increases in the blood concentrations of alpha amino nitrogen. These results and those obtained for the ruminal fistula experiments are interpreted as supporting the suggestion that amino acids and B vitamins are absorbed from the rumen.
