▼ min A (retinol) is essential for life; however, little is known about how it istransported into cells. Ninety-five percent of retinol found in blood is bound to SerumRetinol Binding Protein (RBP4), suggesting that RBP4 is important for transport ofretinol. Importantly, RBP4 levels are linked to metabolic disorders such as metabolicsyndrome and type 2 diabetes. A better understanding of vitamin A transport couldprovide potential targets for the treatment of these disorders. Our lab recently identified apotential RBP4 receptor protein, Retinol Binding Protein Receptor 2 (RBPR2), alsoknown as Liver Stra6 Homologue (LSH). Cell culture models have revealed that LSHbinds RBP4 with high affinity. We hypothesized that mice with a whole body deletion ofLSH will show a reduction in retinol and retinyl esters in serum, hepatic tissue and fat.Our experimental model consisted of transgenic mice with total body deletion ofLSH and wild type litter mates, as control. Mice were sacrificed at 7 weeks and tissuesstored at -80ºC. Total body deletion of the LSH gene was confirmed by Taqmanquantitative PCR. Retinol and retinyl ester levels in liver, fat and serum were assessedusing High Performance Liquid Chromatography.Though no statistically significant differences were found between the retinol orretinyl ester levels in the livers or fat pads of mice with a total body deletion of LSH andwild type mice, there was a significant difference in serum retinol and retinyl ester levelsbetween female knock out mice and female wild type. No significant differences betweenthe two genotypes were observed in the serum of male mice. Results of this study suggest that LSH is not solely responsible for retinoidtransport into cells, or whole body retinol levels, though it may play a subtler role inretinol homeostasis.

▼ A ingestão de quantidades adequadas de alimentos, incluindo aqueles ricos em vitaminas, é necessária para uma vida saudável. A carência de vitamina A tem sido caracterizada como um problema de saúde pública nos países em desenvolvimento, entretanto, uma alta ingestão de vitamina A pode resultar em efeitos tóxicos e teratogênicos. Altas concentrações de vitamina A tem sido observadas nos fígados de animais. O objetivo deste trabalho foi avaliar os níveis de retinol em fígados de frango e verificar o efeito da estocagem sob condições de congelamento sobre esses níveis. Foram utilizados 64 fígados de duas linhagens de frango, Cobb e Ross, provenientes de quatro diferentes propriedades. Examinamos 32 fígados de cada linhagem, 8 amostras de cada granja. As amostras de fígado foram homogeneizadas individualmente, em seguida foram retiradas de cada amostra 4 alíquotas. Uma das alíquotas foi analisada imediatamente após o abate (T0), as demais foram analisadas após 30, 60 e 90 dias de armazenamento a 18 °C (T30, T60 e T90, respectivamente). A dosagem de retinol no fígado foi realizada através da Cromatografia Líquida de Alta Eficiência (CLAE). Os níveis de retinol variaram significativamente de acordo com a linhagem. O valor médio de retinol nas amostras frescas da linhagem Cobb e Ross foi de 6678,0 1337,4 e 8324,1 1158,5 g/100g, respectivamente. Valores de 4258 918,7 e 4650,5 1391,7 μg/100g foram encontrados após 90 dias de estocagem para a linhagem Cobb e Ross, respectivamente. O congelamento do fígado causou uma redução significativa nos seus níveis de retinol, ocasionando uma perda de até 44% com relação aos fígados frescos. A redução nos níveis de retinol ocorreu a partir de 30 dias de estocagem. Mesmo com as perdas decorrentes do congelamento, a ingestão de uma típica porção de 100 g de fígado, independentemente da linhagem do frango analisada, ultrapassa todas as recomendações de consumo e o limite máximo de ingestão tolerável de vitamina A (3000 g/dia) para adultos
The intake of adequate quantities of food, including those rich in vitamins, is necessary for a healthy life. The lack of vitamin A has been characterized as a public health problem in developing countries, however, a high intake of vitamin A can result in toxic and teratogenics effects. High concentrations of vitamin A have been observed in the livers of animals. The objective of this study was to assess the levels of retinol in chicken livers and verify the effect of frozen storage on these levels. 64 livers from two chicken strains, Cobb and Ross, were used, came from four different farms. ▼ As vitaminas A e E são reconhecidamente importantes nos estágios iniciais de vida, sendo os recém-nascidos dependentes da adequação nutricional do leite materno para suprir suas necessidades. Estas vitaminas compartilham vias de transporte para os tecidos e possíveis efeitos antagônicos têm sido observados em animais após a suplementação com vitamina A. Este trabalho teve como objetivo verificar o efeito da suplementação materna com megadose de vitamina A (200.000 UI) no pós-parto imediato sobre a concentração de alfa-tocoferol no colostro. Parturientes saudáveis atendidas em uma maternidade pública natalense foram recrutadas para o estudo e divididas em dois grupos: controle (n = 37) e suplementado (n = 36). Amostras de sangue e leite colostro foram coletadas até 12 horas pós-parto. Neste momento, nas mulheres do grupo suplementado foi administrada a cápsula de vitamina A. Decorrido 24 horas após a primeira coleta foi obtida a 2 amostra de colostro nos dois grupos para análise de retinol e alfa-tocoferol no leite. A concentração média de retinol de 50,7 14,4 μg/dL (Media Desvio Padrão) e de alfa-tocoferol de 1217,4 959 μg/dL no soro das mulheres indicaram estado nutricional adequado nas vitaminas A e E. A suplementação com retinol palmitato resultou em aumento não somente na concentração do retinol no leite colostro do grupo suplementado (p= 0,002), mas também na concentração do alfa-tocoferol (p = 0,04), alterando de 1456,6 1095,8 μg/dL para 1804,3 1432,0 μg/dL (leite 0 e 24 respectivamente) em relação aos valores no grupo controle, 984,6 750,0 μg/dL e 1175,0 730,8 μg/dL. As parturientes apresentaram diferentes respostas à suplementação, influenciadas pelos níveis basais de retinol no colostro. Aquelas com níveis prévios baixos de retinol no colostro (<60 μg/dL) apresentaram aumento na concentração de alfa-tocoferol no leite, enquanto que, as que tinham níveis adequados (>60 μg/dL), apresentaram redução após a suplementação. A suplementação com retinol palmitato é uma medida importante na prevenção da deficiência de vitamina A, porém somente quando considerada a real necessidade de suplementação materna, visto que o excesso de vitamina pode propiciar interações desfavoráveis entre nutrientes essenciais para o grupo materno-infantil

Retinol dosage in the liver was determined by High Performance Liquid Chromatography (HPLC). The levels of
retinol varied significantly according to the strain. The mean
retinol value in the fresh samples was 6678.0 ± 1337.7 and 8324.1 ± 1158.5 µg/100g in the Cobb and Ross strain, respectively. Values of 4258 ± 918.7 ± 1391.7 and 4650.5 ± 1391.7 μg/100g were found after 90 days of storage for Cobb and Ross strain, respectively. The liver freezing caused a significant reduction in their levels of
retinol, causing a loss of up to 44% with respect to fresh livers. The reduction in
retinol levels occurred from 30 days of storage. Even with the losses from the frozen, the ingestion of a typical portion of 100 g of liver, regardless the chicken strain analyzed, surpass all recommendations of consumption and the maximum tolerable intake of vitamin A (3000 μg/day) for adults

▼ The vitamins A and E are recognizably important in the initial stages of life and the newborn depends on nutritional adequacy of breast milk to meet their needs. These vitamins share routes of transport to the tissues and antagonistic effects have been observed in animals after supplementation with vitamin A. This study aimed to verify the effect of maternal supplementation with vitamin A megadose (200,000 UI) in the immediate post-partum on the concentration of alpha-tocopherol in colostrum. Healthy parturient women attended at a public maternity natalensis were recruited for the study and divided into two groups: control (n = 37) and supplemented (n = 36). Blood samples of colostrum and milk were collected until 12 hours after delivery. The women of the supplemented group was administered a retynil palmitate capsule and 24 hours after the first collection was obtained the 2nd sample of colostrum in two groups for analysis of
retinol and alpha-tocopherol in milk. The mean
retinol concentration of 50,7 ± 14,4 μg/dL (Mean ± standard deviation) and alpha-tocopherol of 1217.4 ± 959 mg/dL in the serum indicate the nutritional status biochemical appropriate. Supplementation with retynil palmitate resulted in increase not only
retinol levels in the colostrum of the supplemented group (p = 0.002), but also the concentration of alpha-tocopherol (p = 0.04), changing from 1456.6 ± 1095.8 mg/dL to 1804.3 ± 1432.0 mg/dL (milk 0 and 24 respectively) compared to values in the control group, 984.6 ± 750.0 mg/dL and 1175.0 ± 730.8 mg/dL. The women had different responses to supplementation, influenced by baseline levels of
retinol in colostrum. Those with previous by low levels of
retinol in colostrum (<60 mg/dL) had increased the concentration of alpha-tocopherol in milk, whereas those with adequate levels (> 60 mg/dL), showed a reduction after supplementation. Supplementation with
retinol palmitate is an important intervention in situations of high risk for vitamin A deficiency, when considering the need to maternal supplementation, since the excess vitamin can offer unfavorable interactions between nutrients essential for the mother-child group

▼ Retinol or vitamin A is crucial for many biological processes in the body such as proliferation, differentiation and reproduction. It is mainly stored in the liver and can be broken down to retinoic acid inside the cell. It travels in the circulatory system attached to Retinol Binding Protein (RBP) and its transport into the cell is mediated by the RBP receptor STRA6. Inside the cell, retinol binds to cellular Retinol Binding Protein (CRBP) and can be converted to retinoic acid, which can then act as a regulator of transcription. High levels of RBP have been implicated in type 2 diabetes. Looking for possible interacting partners for proteins such as RBP, can further elucidate the function of the protein and its possible role in type 2 diabetes. The Membrane Yeast Two Hybrid (MYTH) system allows for the study of possible interacting partners for membrane proteins. Previous studies using (MYTH) looked at possible interacting partners for RBP and CRBP. Two hits for RBP were G-protein coupled receptors (GPCRs) RAIG2 and RAIG3. One hit for CRBP was the TRPC4 ion channel. Retinoic acid inducible gene (RAIG) is a recently discovered family C GPCR and the TRPC4 channel is a membrane ion channel responsible for calcium and sodium ion influxes into the cell. An RBP-RAIG interaction could implicate the RAIG receptor in type 2 diabetes. A CRBP-TRPC4 interaction could activate the ion channel causing a calcium influx and a possible mechanism of insulin resistance in the cell, the genesis of which is still very controversial. This thesis involves performing a series of pull down assays using a novel oil-based assay to determine if the proteins in the yeast two hybrid system are indeed interacting.

A presente dissertação é o corolário de um conjunto de estudos desenvolvidos com o objectivo de contribuir para um melhor conhecimento dos efeitos da administração pré-natal de vitamina A sobre alguns dos mais importantes factores envolvidos na formação da rede vascular do pulmão. O estudo realizado está inserido no âmbito dos trabalhos que têm vindo a ser efectuados há mais de duas décadas no Instituto de Histologia e Embriologia da faculdade de Medicina de Coimbra, visando esclarecer os processos subjacentes à morfogénese, histogénese e histofisiologia pulmonares.
O murganho, Mus musculus, estirpe ICR (CD-1®), foi utilizado como modelo experimental, tendo sido analisados os aspectos morfológicos da presença do VEGF, dos seus receptores (VEGFR), do Tie-2, da MMP-1, da MMP-9 e do TIMP-1 no pulmão de fetos e de recém-nascidos cuja gestação decorreu sob o efeito da vitamina.
A. Nos mesmos animais, foi ainda realizado um estudo quantitativo da concentração pulmonar do VEGF e do Tie-2, a determinação da concentração plasmática do VEGF, bem como o estudo dos níveis de expressão genética do VEGFR-1 e do VEGFR-2. Em virtude da existência de dimorfismo sexual no pulmão adulto, foi considerado pertinente avaliar cada um dos parâmetros com separação por sexo
Para um melhor enquadramento do trabalho experimental, e uma vez apresentados os objectivos que se pretenderam alcançar, procedeu-se à organização do presente trabalho em seis capítulos. Assim, no capítulo I, são revistos, de forma sucinta, o actual estado do conhecimento da morfogénese pulmonar e os aspectos mais relevantes da formação e organização do compartimento vascular do pulmão, procurando integrá-los na descrição dos períodos de desenvolvimento do órgão. No capítulo II são expostos os principais fundamentos teóricos que estiveram na base da investigação efectuada, constituindo esta o contributo pessoal de natureza experimental do presente trabalho, explanada do terceiro ao sexto capítulo.
O capítulo III é dedicado ao estudo morfológico e quantitativo da presença do VEGF no pulmão fetal, desde o 15º dia de gestação até ao dia do nascimento, em animais a cujas mães foi administrada, por via subcutânea, uma dose única de vitamina A (45 000 UI, 150 µl). A determinação da concentração plasmática do VEGF foi igualmente apurada. Em paralelo foram estudados animais provenientes de gestantes às quais foi administrado o volume equivalente de placebo (grupos controlo).
O estudo morfológico e semiquantitativo da presença do VEGF nos diferentes períodos do desenvolvimento pulmonar foi efectuado por imuno-histoquímica. Para a quantificação da concentração deste factor de crescimento no pulmão e no plasma, recorreu-se à metodologia de ELISA. A análise morfológica e a determinação da concentração pulmonar revelaram que, ao 15º dia de gestação, o VEGF está presente em maior quantidade no pulmão de fetos controlo. ▼ Retinal dehydrogenase (RALDH) is the key enzyme that regulates the production of retinoic acid (RA), an important mediator of immune responses in the gut. RA is the biologically active metabolite of vitamin A. Vitamin A is at the same time processed and metabolized in small intestine. RA produced by intestinal epithelial cells is known to be essential for the induction of characteristic mucosal immune responses by innate immune cells in the intestinal environment. The expression of RALDH enzymes is tissue-specific, with certain tissues expressing higher concentrations than others, and with variations between the different isoforms of the enzyme. RALDH levels and RA production by the innate immune cells in the gut have been shown to be regulated by dietary vitamin A. However the expression of RALDH enzymes in the intestinal epithelium does not decrease with different levels of vitamin A in the diet. In the case of RALDH3 it is even up-regulated in mice fed vitamin A deficient diets. It is believed that this is due to a compensatory mechanism. In this review we look at the genetic and environmental factors that can affect the expression of this enzyme in small intestinal epithelial cells. From this perspective we point at the possible indirect effects that these factors could have on the innate immune cells in the gut through its effects on the production on RA by intestinal epithelial cells.

de vitamina A (400.000 UI) nas concentrações de
retinol sérico de puérperas durante os seis
primeiros meses pós-parto bem como descrever a freqüência de deficiência de
retinol sérico
nas mulheres antes da suplementação de vitamina A. Trata-se de um ensaio clínico
randomizado, controlado, triplo-cego, de base hospitalar, com 312 mulheres atendidas no
período pós-parto imediato e acompanhadas durante os seis primeiros meses, de dois serviços
públicos de saúde na cidade do Recife, região nordeste do Brasil, no período de agosto de
2007 a março de 2009. Todas as mulheres receberam uma cápsula contendo 200.000 UI de
vitamina A no pós-parto imediato e no 10º dia foram randomizadas em dois grupos de
suplementação: um grupo recebeu uma segunda cápsula de vitamina A (200.000UI) e o outro
grupo recebeu o placebo de vitamina A. Além dos dados socioeconômicos, demográficos,
antropométricos, clínicos e obstétricos foram coletadas amostras de sangue para as análises
das concentrações de
retinol sérico, PCR e hemoglobina. A pesquisa foi registrada sob o
número NCT00742937. 215 mulheres completaram o estudo (111 no grupo de 400.000 UI de
vitamina A e 104 no grupo de 200.000 UI de vitamina A). Não foi observado nenhum evento
adverso com a administração das cápsulas de vitamina A. Não houve diferença
estatisticamente significante nas concentrações de
retinol sérico entre os grupos de
suplementação aos 2 meses (2,13 vs 2,03 mmol/L), 4 meses (2,20 vs 2,24 mmol/L) e 6 meses
(2,29 vs 2,31 mmol/L). Observou-se um declínio da prevalência de concentrações inadequadas
de
retinol sérico de 21,1% para 2,35% ao longo dos seis meses após a suplementação. A
associação das concentrações de
retinol sérico com as variáveis independentes foi testada pela
Razão de Prevalência e apenas o número de gestações apresentou associação estatisticamente
significante com a deficiência de vitamina A (DVA). Os resultados revelam elevada
prevalência concentrações inadequadas de
retinol sérico na população estudada, apontando a
necessidade de desenvolver ações voltadas para a promoção da educação nutricional além da
realização de estudos populacionais para confirmar os fatores de risco que contribuem para o
desenvolvimento da DVA. Considerando nenhum efeito adicional observado e que a
população estudada tem um nível de deficiência de vitamina A considerada leve,nossos
resultados reforçam a manutenção da estratégia de prevenção preconizada pela OMS

▼ Chronic liver disease is a major health burden worldwide. Major determinants of this condition are viral infections, alcohol abuse and obesity. Genetic background modulates the effect of damaging agents on the liver. The genetic variant rs738409 in the patatin-like phospholipase domain-containing 3 (PNPLA3) gene associates with increased susceptibility to the entire spectrum of chronic liver disease, and in particular with non-alcoholic fatty liver disease. The variant results in an isoleucine to methionine substitution at position 148 (I148M) of the amino acidic sequence and was first associated with increased hepatocyte fat content. Despite the strength of the genetic association, the mechanisms causing liver fat accumulation and hepatocyte damage are not yet understood.
In this thesis, we tested the following hypotheses: 1) PNPLA3 is involved in hepatic very low density lipoprotein secretion 2) the protein acts as a glycerolipid hydrolase and the 148M mutation is a loss of function 3) PNPLA3 has a specific role in retinol metabolism in hepatic stellate cells. We tested the first hypothesis measuring VLDL secretion in a cohort of 55 individuals genotyped for the I148M variant and we found that carriers of the 148M allele secret less VLDL for a given amount of liver fat. We confirmed this result in vitro by measuring APOB secretion in cell lines stably overexpressing the 148I or the 148M PNPLA3. We tested the second hypothesis performing enzymatic activity assays using purified 148I and 148M recombinant proteins. The wild type protein had glycerolipid hydrolase activity and the 148M mutation induced a loss of function. Finally, we tested the third hypothesis assessing the effect of PNPLA3 up- and down-regulation on hepatic stellate cell retinyl palmitate content and retinol release. We found that PNPLA3 insulin–mediated up-regulation induces retinol release from hepatic stellate cells and that this effect is abolished by PNPLA3 silencing. We confirmed this finding by looking at human circulating levels of RBP4, a reliable marker of retinol plasma levels, in 146 individuals genotyped for the I148M variant. We found carriers of the M allele to have lower RBP4 plasma levels, confirming the role of PNPLA3 in retinol metabolism.
In conclusion, we identified two possible mechanisms underlying the susceptibility to chronic liver disease in carriers of the PNPLA3 mutation: 1) reduced intracellular triglyceride mobilization leading to hepatocyte damage 2) impaired hepatic stellate cell retinol metabolism causing abnormal response of hepatocytes to damaging agents.

▼ min A is essential for normal embryonic development and vision. Retinol binding protein (RBP) and its receptor, STRA6, are vital for the maintenance of intracellular stores of vitamin A. Recently, elevated serum RBP concentration has been implicated as a contributing factor to the development of insulin resistance and type II diabetes. However, conflicting opinions exist as to how increased RBP levels can cause insulin resistance. Some suggest it is as a result of the activation of macrophages in adipose tissue and the secretion of cytokines. Others suggest it is as a result of RBP induced STRA6 phosphorylation, and the activation of the JAK/STAT signalling pathway. Regardless of the mechanism, reducing circulating levels of RBP may be a novel strategy for the treatment of type II diabetes. Several small molecules have been designed to promote renal clearance of RBP, thus lowering serum levels. In order to consolidate the theories surrounding RBP induced insulin resistance, a proteomic study was devised to determine the direct effect of RBP on muscle cells, since the muscle is the main target of insulin induced glucose uptake. Results suggest that RBP may be affecting the enzymes involved in glucose storage and glycogen catabolism. Artificial methods aimed at reducing serum RBP levels may act by preventing RBP induced glycogen disruption. In a related study, it was noted that small molecules aimed at reducing circulating RBP levels had a direct effect on muscle cells to stimulate glucose uptake. This phenomenon occurred independently of the predicted mechanism of action. A second proteomic study was conducted to determine the direct mechanism of action of the compounds in muscle cells. The molecules appear to stimulate the influx of glucose by reducing the ATP yield from oxidative phosphorylation and enhancing the utilisation of alternate energy stores.
The C-terminal region of STRA6 appears to be a large SH2 motif-containing intracellular segment which may be capable of forming an independently folding domain. As such it may represent the site of interaction with other proteins in the system. Therefore, it was cloned, expressed and characterised. The secondary structure of the domain was shown to be largely α-helical and a model was constructed. Possible functional roles for this region were investigated.

Myopia is a refractive error characterized by an abnormal increase in eye length. The prevalence of myopia is increasing in a global manner and the increase in humans affected by myopia comes along with an increasing number of patients affected with high myopia (more than -6D). High myopia is associated with serious complications including retinal detachment, glaucoma and posterior staphyloma. Ocular development depends on visual experience. Myopia can be induced in animal eyes by rearing them with negative spectacle lenses which displaces the focal point behind the retina. The eye compensates for this imposed defocus by elongating more rapidly. Such visual manipulations enable the exploration of the so far poorly understood underlying biological mechanisms of myopia development. Myopia development is associated with transcription changes and physical changes in ocular tissues. This thesis focusses on the exploration of mRNA expression changes in the guinea pig retina that accompany changes in axial elongation. Therefore, myopic and non-myopic guinea pig retinas were compared via the high throughput method of RNA sequencing (RNA-seq). RNA-seq was used to compare normal retinal samples from untreated age-matched guinea pigs with those from guinea pigs that developed myopia over 4 or 11 days, and with samples originating from eyes that had recovered for 48 hours from the induced myopia. The RNA-seq results were analysed with 4 different downstream analysis tools (Voom, DESeq2, EdgeR and Cuffdiff) to determine differentially expressed genes (DEG’s). The results from the tools Cuffdiff were used for pathway analysis. Expression analysis with Cuffdiff resulted in 137 DEG’s after 4 days of myopia development, 98 DEG’s after 11 days of myopia development and 47 DEG’s after 48h of recovery from myopia development. Retinol binding protein 1 was significantly upregulated according to the corrected p-value in all 3 comparisons. Several of the DEG’s (FAM107A, Col4A4, ApoE, MMP-2, and RBP1) were successfully validated using quantitative real time polymerase chain reaction (qPCR). Some of the DEG’s after induced myopia which were determined by Cuffdiff are well known to play a role in scleral remodelling during myopia development: Bone morphogenetic protein 2, Metalloproteinase 1 (MMP-2) and collagens. MMP-2 was significantly upregulated after 11 days of myopia development, BMP-2 after 4 days of myopia development and a number of collagens were upregulated in both conditions. Pathway analysis revealed that the pathways RAR activation and Retinol biosynthesis were among the top 10 associated pathways after 4 days of myopia development and after 48h of recovery from myopia development. retinol have more
retinol and secretory immunoglobulin A in colostrum. 190 healthy puerperal women from a Brazilian public maternity were recruited and divided according to the cutoff point for serum
retinol (30 μg/dL). A number of the women was supplemented with 200000 UI (60 mg) of retinyl palmitate in the immediate postpartum. Serum and colostrum were collected on the 1st day postpartum and colostrum again on the following day.
Retinol (serum and colostrum) was analyzed by HPLC and SIgA (colostrum) by turbidimetry. The mothers presented with adequate biochemical indicators of nutritional status, according to serum
retinol (44.6 μg/dL). There were significant differences (p= 0.0017 and p= 0.043, respectively) in
retinol and SIgA levels in the colostrum of mothers with serum
retinol > 30 μg/dL and < 30 μg/dL. The concentration of SIgA in the colostrum of non-supplemented mothers on the 1st day postpartum was 822.6 mg/dL, decreasing after 24 hours to 343.7 mg/dL. Supplemented mothers showed levels of SIgA in colostrum of 498.9 mg/dL on the 2nd day postpartum (p= 0.00006). The colostrum of women with good vitamin A nutritional status had more
retinol and SIgA. Additionally, maternal supplementation increases the levels of SIgA in colostrum. The higher levels of SIgA on the 1st day postpartum showed the importance of early breastfeeding, given that it provides considerable immunological benefits to newborn infants

▼ Mães com bom estado nutricional em vitamina A na gestação e lactação terão melhores condições em nutrir e proteger o neonato através do leite materno. Nossa hipótese é que as mulheres com mais retinol no soro possuirão mais retinol e imunoglobulina A secretora no colostro. Foram recrutadas 190 puérperas saudáveis em uma maternidade pública brasileira, que foram divididas segundo o ponto de corte para retinol sérico (30 μg/dL). 118 delas foram suplementadas com uma dose de 200000 UI (60 mg) de palmitato de retinila no pós-parto imediato. Foram coletados soro e colostro no 1 dia pós-parto e colostro novamente no dia seguinte. O retinol (soro e colostro) foi analisado por cromatografia líquida de alta eficiência e a SIgA (colostro) por turbidimetria. As mães apresentaram estado nutricional bioquímico adequado segundo retinol sérico (44,6 μg/dL). Houve diferenças significativas (p= 0,0017 e p= 0,043, respectivamente) nos níveis de retinol e SIgA no colostro de mães com retinol sérico >30 μg/dL e <30 μg/dL. A concentração de SIgA no colostro das mães não suplementadas, no 1dia pós-parto, foi de 822,6 mg/dL, decrescendo, após 24 horas, para 343,7 mg/dL. As mães suplementadas apresentaram níveis de SIgA no colostro de 498,9 mg/dL no 2 dia pós-parto (p= 0,00006). O colostro de mulheres com bom estado nutricional em vitamina A possuem mais retinol e SIgA. Além disso, a suplementação materna aumenta os níveis de SIgA no colostro. ▼ Mothers with good vitamin A nutritional status during gestation and lactation are better able to nourish and protect their infant with maternal milk. Our hypothesis is that women with more serum retinol have more retinol and secretory immunoglobulin A in colostrum. 190 healthy puerperal women from a Brazilian public maternity were recruited and divided according to the cutoff point for serum retinol (30 μg/dL). A number of the women was supplemented with 200000 UI (60 mg) of retinyl palmitate in the immediate postpartum. Serum and colostrum were collected on the 1st day postpartum and colostrum again on the following day. Retinol (serum and colostrum) was analyzed by HPLC and SIgA (colostrum) by turbidimetry. The mothers presented with adequate biochemical indicators of nutritional status, according to serum retinol (44.6 μg/dL). There were significant differences (p= 0.0017 and p= 0.043, respectively) in retinol and SIgA levels in the colostrum of mothers with serum retinol >30 μg/dL and <30 μg/dL. The concentration of SIgA in the colostrum of non-supplemented mothers on the 1st day postpartum was 822.6 mg/dL, decreasing after 24 hours to 343.7 mg/dL. Supplemented mothers showed levels of SIgA in colostrum of 498.9 mg/dL on the 2nd day postpartum (p= 0.00006). The colostrum of women with good vitamin A nutritional status had more retinol and SIgA. Additionally, maternal supplementation increases the levels of SIgA in colostrum. The higher levels of SIgA on the 1st day postpartum showed the importance of early breastfeeding, given that it provides considerable immunological benefits to newborn infants
Mães com bom estado nutricional em vitamina A na gestação e lactação terão melhores condições em nutrir e proteger o neonato através do leite materno. Nossa hipótese é que as mulheres com mais retinol no soro possuirão mais retinol e imunoglobulina A secretora no colostro. Foram recrutadas 190 puérperas saudáveis em uma maternidade pública brasileira, que foram divididas segundo o ponto de corte para retinol sérico (30 μg/dL). 118 delas foram suplementadas com uma dose de 200000 UI (60 mg) de palmitato de retinila no pós-parto imediato. Foram coletados soro e colostro no 1 dia pós-parto e colostro novamente no dia seguinte. O retinol (soro e colostro) foi analisado por cromatografia líquida de alta eficiência e a SIgA (colostro) por turbidimetria. As mães apresentaram estado nutricional bioquímico adequado segundo retinol sérico (44,6 μg/dL). Houve diferenças significativas (p= 0,0017 e p= 0,043, respectivamente) nos níveis de retinol e SIgA no colostro de mães com retinol sérico >30 μg/dL e <30 μg/dL. ▼ A deficiência de vitamina A é um sério problema de saúde pública, e causa a morte e cegueira em crianças nos países em desenvolvimento. A fortificação de alimentos como ovos, pode ser uma importante fonte de vitaminas para o controle da deficiência. Foram utilizadas 60 codornas Coturnix coturnix japonica em delineamento experimental inteiramente ao acaso, com duração de sete semanas. As aves foram distribuídas em cinco tratamentos com quatro repetições cada. O objetivo foi avaliar a influência de diferentes níveis de retinol palmitato (2000UI, 4000UI, 8000UI e 16000UI) em codornas sobre os níveis de retinol na gema dos ovos. O método utilizado para dosar retinol na gema dos ovos de codornas foi a Cromatografia Líquida de Alta Eficiência (CLAE) e o método enzimático para quantificar a concentração de colesterol. O peso e a produção de ovos foram significativamente alterados pela suplementação oral com retinol nas aves. Os resultados mostraram um aumento progressivo na incorporação de retinol na gema do ovo em resposta à suplementação, atingindo valores 384% superiores aos valores de controle. Ao término das suplementações foi observada uma diminuição significativa nas concentrações de retinol na gema dos ovos, sendo as suplementações com 8000UI e 16000UI as mais duradouras e mesmo após três semanas continuaram com os níveis de retinol elevados. O conteúdo de colesterol nos ovos não foi significativamente alterado. O consumo de um ovo enriquecido com 16000 UI de retinol palmitato no presente estudo, por dia, provavelmente atenderia em torno de 10 e 7,3% das recomendações diárias deste micronutriente para crianças na faixa etária de 1 a 3 anos, e 4 a 8 anos, respectivamente. min A deficiency is a serious public health problem in developing countries, and it causes death and blindness among children in the developing countries. The fortification of food could be an important source of vitamins to control deficiency. 60 Coturnix coturnix japonica quails were used in a randomized design with duration of seven weeks. The birds were assigned into five treatments with four repetitions. The objective was to evaluate the influence of the supplementation with different levels of retinyl palmitate (2,000 IU, 4,000 IU, 8,000IU and 16,000 IU) in quails under the levels of retinyl in egg yolks. The method used to dose retinyl in yolks of quail eggs was High Performance Liquid Chromatography and the enzymatic method to quantify the cholesterol concentration. The weight and production of eggs was significantly modified by the supplementation with retinyl in the birds. The results showed a gradual increase in the incorporation of retinyl in the egg yolk as a response to the supplementation, reaching values 384% higher than the control values. ▼ Fatores como a tendência à diminuição dos níveis de retinol sérico das gestantes, barreira placentária e aumento da demanda de retinol, fazem com que puérperas e lactentes representem grupos de risco de deficiência em vitamina A, nutriente que participa de processos vitais, como a diferenciação, proliferação celular e apoptose. O objetivo do presente trabalho foi avaliar o efeito da suplementação de retinol palmitato (200.000 UI) sobre os níveis de retinol no leite de puérperas da Maternidade Escola Januário Cicco (MEJC), intervenção que vem sendo adotada pelo Ministério da Saúde desde 2002. O experimento do tipo longitudinal teve como participantes 106 puérperas (68 do grupo suplementado e 38 do grupo controle). A Cromatografia Líquida de Alta Eficiência (CLAE) foi o método utilizado para dosar retinol das amostras de leite e soro; e o crematócrito para determinar os níveis de gordura no leite. A quantificação do retinol apresentou médias de 99,0 64,4 ug/dl para o colostro; 160,1 94,4 ug/dl para o colostro 6 horas após suplementação; 68,9 33,S ug/dl para o leite de transição e 30,6 15,2 ug/dl para o leite maduro do grupo suplementado. As diferenças foram significativas entre todas as médias. O grupo controle também demonstrou médias de retinol significativamente diferentes, sendo estas superiores no colostro 88,6 62,1 ug/dl, reduzindo para 61,9 30,1 ug/dl no leite de transição e 32,9 17,6 ug/dl no leite maduro. Nenhuma diferença significativa foi observada quando as médias do retinol referentes aos três tipos de leite do grupo suplementado foram comparadas com as respectivas médias do grupo controle. A prevalência encontrada no soro (35,1% e 81,1 % para o ponto de corte 20 ug/dl e 30 ug/dl, respectivamente) e no leite (51,4%) revelou a deficiência de vitamina A como problema de saúde pública. A gordura do leite colostro, transição e maduro variou de forma semelhante no grupo suplementado (1,92 0,96; 3,25 1,27 e 3,31 1,36 gramas) e controle (1,87 1,14; 3,25 1,31 e 3,36 1,67 gramas), ocorrendo diferença entre colostroltransição e colostro/maduro. Não houve diferença entre os grupos. O seguinte trabalho demonstrou que a suplementação utilizando 200.000 UI não foi capaz de elevar os níveis de retinol no leite até o momento esperado e, provavelmente, não foi fornecida em quantidade suficiente para satisfazer as demandas das mães com reservas hepáticas mais espoliadas. ▼ The caprine milk is a product of high biological value and high digestibility. Due to these characteristics it is quite used by newly born children that are not breastfed or that are intolerant to the bovine milk. The vitamin deficiency is a public health problem in underdeveloped areas as the Northeast of Brazil and where areas the caprine ones adapt very well. The present study was led to analyze the influence of the feeding in the vitamin levels in the caprine milk. The animals used were the races Saanen and Murciana, divided in three groups. The first group with 38 animals of race Saanen and the second with 30 animals of race Murciana were, fed with concentrated and voluminous. A third group with 20 animals of the race Saanen was fed exclusively with voluminous. The four group was added with 10. 000 UI of retinol palmitato, administered directly, like capsule, in the mouth of animal. Parallel it was verified the level of retinol of milk in the beginning and final of the sucked, in the goats of the second group (race Murciana n =30). The retinol of caprine milk was determined through the system of liquid cromatografia of high efficiency (HPLC). The retinol levels in the studied groups were respectively: first (38. 5 12. 7 μg/100ml), second (40. 5 9. 7 μg/100ml); third, with 20 animals of race Saanen fed exclusively with voluminous (23. 1 6. 7 μg/100ml) and in the group a, suplementation with 10. 000 UI of retinol palmitato (43,7 18,8 μg/100ml) before, and (61,9 26,9 μg/100ml) after the supplementation. It was not found significant difference between the averages from animals of the first and second group, that were fed with the same concentrate diet and voluminous, showing that the retinol levels in the milk of these two races are equivalent. Already in the animals of the first and third group that they were fed with different diets, in those which diet was just voluminous, a drastic reduction was verified in the retinol levels. In relation to the retinol of the milk in different moments from the same sucked, it was observed in the beginning of the breast-feeding (22. 6 9. 8 μg/100ml) and at the end of the sucked (49. 6 14. 7μg/100ml), being the difference between the averages, statistically significant (p <0,0001). Already in the animals that were supplemented, a significant increase was observed in the retinol concentration, being obtained a medium response of 41,85

▼ Retinol dehydrogenases (RDH) convert retinol into retinal, the intermediate in the biosynthesis of retinoic acid. All-trans-retinoic acid (atRA) regulates gene transcription and/or translation through retinoic acid receptors (RARs) and PPARδ (1). To test function of Rdh1, an efficient (Vmax/Km) and widely distributed RDH (2), our lab created Rdh1 knockout (KO) mice (3). Initial study of Rdh1-KO mice determined that when fed a low or vitamin A-deficient (VAD) diet, Rdh1-KO mice gain 33% more weight than wild-type (WT). However, when fed a chow diet (22+ IU vitamin A/g diet – i.e. copious), KO mice appeared identical to WT. Our continued work on the Rdh1-KO mouse reveals additional insight into the role of Rdh1 in both retinoic acid synthesis and energy regulation. Both a 10% increase in length and 33% increase in adiposity contribute to weight gain in VAD-fed KO animals. Based on measurements of adipocyte size, increased adiposity results from hyperplasia of white adipose tissue. Rdh1-KO mice, fed a diet with the recommended amount of vitamin A (4 IU/g), also increase in weight and adiposity, but not length. Interestingly, high fat diet feeding fails to exacerbate weight or adiposity. Increased weight contributes to a decline in Rdh1-KO mouse health. Whereas young animals respond normally to tests of glucose and insulin tolerance, older Rdh1-KO mice become insulin resistant and glucose intolerant, with 2.5-fold higher insulin levels. These changes in Rdh1-KO glucose metabolism cannot be attributed to changes in pancreatic 9-cis-retinoic acid (9cRA) (4). As expected in heavier animals, leptin levels also increase 1.8-fold in Rdh1-KO mice. In addition, loss of Rdh1 reduces circulating thyroid hormones T4 and T3 by 12% and 17%, respectively, though TSH levels remain unchanged. These data demonstrate that atRA, generated by Rdh1, contributes to control of body weight, and does so to a physiologically relevant extent. Increased weight results from either increased caloric intake or decreased energy expenditure. Overall, Rdh1-KO mice move just as much as WT, and their food intake increases only in proportion to body weight. Normalized to body weight, we observe no change to energy expenditure by indirect calorimetry. Thermogenesis, the generation of heat for body temperature maintenance, also contributes to energy expenditure. Rdh1-KO mice respond normally to cold challenge and β3-adrenergic stimulation, but appear reduced in body temperature, especially when stressed by fasting. Consistent with these observations, the brown adipose tissue (BAT) of Rdh1-KO mice expresses normal levels of the β2- and β3-adrenergic receptors and lower levels of Gbpar1, a bile acid receptor also involved in thermogenesis (5).…

▼ Avaliou-se a eventual atividade quimiopreventiva por parte da vitamina A (VA) quando administrada a ratas Sprague-Dawley durante a etapa de pós-iniciação da carcinogênese mamária induzida pelo 7-12 dimetilbenz(a)antraceno (DMBA). Com exceção de 10 animais que constituíram um grupo controle à parte de ratas consideradas normais [grupo normal (N)], e que não foram submetidas a qualquer procedimento experimental durante todo o estudo, 40 ratas com 50 dias de idade receberam o agente carcinogênico DMBA, por meio de entubação gástrica na dose de 60 mg/kg/peso corpóreo. Após 2 semanas, as ratas iniciadas receberam durante 12 semanas consecutivas, por entubação gástrica, 0,25 mL/100 g de peso corpóreo de óleo de milho (grupo OM; controle, n=20) e 2,5 mg/100 g de peso corpóreo de vitamina A (grupo VA, n=20), sendo, então, eutanasiadas. Não houve diferenças (p>0,05) entre os grupos OM e VA quanto à latência de aparecimento da primeira neoplasia mamária, incidência, multiplicidade e peso médio das neoplasias mamárias, todas classificadas como malignas. Em comparação ao grupo OM, o grupo VA apresentou maior concentração (p<0,05) de retinol no tecido mamário neoplásico e hepático, além de maiores concentrações hepáticas (p<0,05) de palmitato de retinila. Não se detectou palmitato de retinila em neoplasias mamárias de ambos os grupos. Não houve diferença (p>0,05) entre neoplasias mamárias do grupo OM e o tecido mamário do grupo N quanto à metilação global do DNA. Em comparação a neoplasias mamárias do grupo OM, neoplasias do grupo VA apresentaram menor (p<0,05) metilação global do DNA. Conclui-se que a VA não apresentou atividades quimiopreventivas e seu metabolismo encontra-se alterado em neoplasias mamárias.
The potential chemopreventive activity of vitamin A (VA) was evaluated when administered to Sprague-Dawley rats during the stage of post-initiation of mammary carcinogenesis induced by 7-12 dimethylbenz(a)anthracene (DMBA). Except for 10 animals that constituted a separate control group of normal rats [normal group (N)], that were not subjected to any experimental procedure throughout the study, 40 rats with 50 days of age received the carcinogen DMBA by gavage at the dose of 60 mg/kg/body weight. After 2 weeks, the rats received for 12 consecutive weeks, by gavage, 0.25 mL/100 g body weight of corn oil (OM group, control, n = 20) or 2.5 mg/100 g body weight of vitamin A (VA group, n = 20), being then euthanized. There were no differences (p> 0.05) between the OM and VA groups regarding the latency of onset of first breast neoplasm, incidence, multiplicity and average weight of breast tumors, all classified as malignant. Compared to the OM group, the VA group had a higher concentration (p <0.05) of retinol in neoplastic breast tissue and liver as well as higher concentrations (p <0.05) of retinyl palmitate in the liver. Retinyl palmitate was not detected in breast tumors of both groups. There was no difference (p> 0.05) between breast tumours of OM group and mammary tissue of N group regarding global DNA methylation.…

▼ Mothers with good vitamin A nutritional status during gestation and lactation are better able to nourish and protect their infant with maternal milk. Our hypothesis is that women with more serum
retinol have more
retinol and secretory immunoglobulin A in colostrum. 190 healthy puerperal women from a Brazilian public maternity were recruited and divided according to the cutoff point for serum
retinol (30 μg/dL). A number of the women was supplemented with 200000 UI (60 mg) of retinyl palmitate in the immediate postpartum. Serum and colostrum were collected on the 1st day postpartum and colostrum again on the following day.
Retinol (serum and colostrum) was analyzed by HPLC and SIgA (colostrum) by turbidimetry. The mothers presented with adequate biochemical indicators of nutritional status, according to serum
retinol (44.6 μg/dL). There were significant differences (p= 0.0017 and p= 0.043, respectively) in
retinol and SIgA levels in the colostrum of mothers with serum
retinol > 30 μg/dL and < 30 μg/dL. The concentration of SIgA in the colostrum of non-supplemented mothers on the 1st day postpartum was 822.6 mg/dL, decreasing after 24 hours to 343.7 mg/dL. Supplemented mothers showed levels of SIgA in colostrum of 498.9 mg/dL on the 2nd day postpartum (p= 0.00006). The colostrum of women with good vitamin A nutritional status had more
retinol and SIgA. Additionally, maternal supplementation increases the levels of SIgA in colostrum. The higher levels of SIgA on the 1st day postpartum showed the importance of early breastfeeding, given that it provides considerable immunological benefits to newborn infants

▼ Vitamin A is important in many essential body processes and its deficiency results in serious consequences for human health. Breast milk is the only source of this vitamin for children that are exclusively breastfed. Analysis of vitamin A in mother s milk
is important because its concentration is related to maternal vitamin A status and to its ingestion by the mother during pregnancy. The aim of the present study was to assess the effect of maternal supplementation with retynil palmitate on the concentration of colostrum
retinol under fasting and postprandial conditions. A total of 149 nursing mothers were recruited at the Januário Cicco Maternity School (Natal, Brazil) and allocated to two groups: Comparison (n = 69) and Test (n = 80). Blood and colostrum (in fasting and postprandial conditions) samples were collected up to 24hs after delivery. Serum
retinol and colostrum levels were analyzed by high-performance liquid chromatography. The serum
retinol level of 41.6 ± 12.7μg/dL (mean ± standard deviation) indicates adequate biochemical nutritional status. Colostrum
retinol level was not influenced by serum
retinol levels under any of the conditions established. In the colostrum, the
retinol concentration in the unsupplemented test group was 67.3 ± 37.7 μg/dL under fasting and 80.3 ± 35.1 μg/dL under postprandial conditions (p<0.05), showing an increase of 19.3%. In the supplemented test group the values were 102.6 ±
57.3 μg/dL and 133.4 ± 78.3 μg/dL under fasting and postprandial, respectively (p<0.05), representing an increase of 30%. Considering that under fasting conditions most of the vitamin A transported to the milk originates in the
retinol binding protein (RBP), the postprandial increase in colostrum
retinol suggests a different transport mechanism of
retinol to maternal milk from that performed by RBP. This situation becomes more evident under supplementation conditions.

▼ Vitamin A deficiency is a serious public health problem in developing countries, and it causes death and blindness among children in the developing countries. The fortification of food could be an important source of vitamins to control deficiency. 60 Coturnix coturnix japonica quails were used in a randomized design with duration of seven weeks. The birds were assigned into five treatments with four repetitions. The objective was to evaluate the influence of the supplementation with different levels of retinyl palmitate (2,000 IU, 4,000 IU, 8,000IU and 16,000 IU) in quails under the levels of retinyl in egg yolks. The method used to dose retinyl in yolks of quail eggs was High Performance Liquid Chromatography and the enzymatic method to quantify the cholesterol concentration. The weight and production of eggs was significantly modified by the supplementation with retinyl in the birds. The results showed a gradual increase in the incorporation of retinyl in the egg yolk as a response to the supplementation, reaching values 384% higher than the control values. By the end of the supplementations a significant reduction in the concentrations of retinyl in the eggs yolk was observed. The most lasting supplementations were with 8,000 IU and 16,000 IU which lasted for three weeks. The cholesterol content in eggs was not significantly modified. The consumption of one egg enriched with 16000UI of
retinol palmitate in the present study, by day, would probably reach 10 and 7,3% of the daily recommendations of this micronutrient for children of 1 to 3 years of age, and for 4 to 8 years, respectively. ▼ A insuficiência renal crônica (IRC) é uma lenta e progressiva diminuição da função renal sendo um dos principais problemas de saúde pública do nosso país. A hemodiálise é o tratamento padrão aos quais estes pacientes são submetidos. Apesar de aumentar a sobrevida do paciente, ele é incapaz de corrigir totalmente os distúrbios metabólicos. Esse procedimento acaba amplificando o estresse oxidativo muito comum em pacientes com IRC. Esses distúrbios somados ao estresse oxidativo são os principais fatores que podem desencadear o desenvolvimento de doenças vasculares e, sabe-se que estas são as principais causadoras de morbidade e mortalidade nestes pacientes. Por outro lado, é importante estudar a condição micronutricional nestes pacientes, relacionada a vitaminas e carotenóides, uma vez que, substâncias exógenas potencialmente antioxidantes poderiam exercer um papel protetor contra as doenças vasculares. Por isto, a avaliação conjunta dos níveis de algumas vitaminas, carotenóides, parâmetros do estresse oxidativo e ainda parâmetros bioquímicos em pacientes com IRC submetidos ao tratamento de hemodiálise são relevantes. Neste estudo foram quantificados os níveis sangüíneos de vitamina A (
retinol), vitamina E (- e - tocoferol), carotenóides (licopeno, luteína, zeaxantina, -criptoxantina, - e - caroteno); biomarcadores do estresse oxidativo, como glutationa reduzida (GSH), malondialdeído (MDA), atividade das enzimas -aminolevulinato desidratase (ALA-D), glutationa peroxidase (GPx), catalase (CAT) e superóxido dismutase (SOD), e alguns parâmetros bioquímicos (uréia, creatinina; perfil lipídico: colesterol total, colesterol LDL, colesterol HDL e triglicerídeos e suas relações) em pacientes com IRC submetidos ao tratamento de hemodiálise (HD) (n=29) e em um grupo de indivíduos saudáveis (grupo controle; n=20). Os níveis de
retinol foram significativamente aumentados nos pacientes HD. Dos carotenóides, apenas o licopeno e o - caroteno apresentaram diminuição significativa nos pacientes. Os níveis de MDA plasmático e GSH eritrocitária e as atividades das enzimas SOD e CAT estavam aumentados nos HD; diferentemente da GPx e da ALA-D que estavam diminuídas. Em relação aos parâmetros bioquímicos, os níveis de uréia e creatinina estavam aumentados, assim como os parâmetros do perfil lipídico (colesterol total, LDL e triglicerídeos), o colesterol HDL estava diminuído. A relação colesterol total/colesterol HDL e colesterol LDL/colesterol HDL também conhecidos como índice de risco coronariano estavam aumentados cerca de três vezes nos pacientes hemodialisados. Ainda, o
retinol apresentou correlação negativa com a enzima ALA-D e correlações positivas com o MDA, SOD e CAT. ▼ Vitamin A is important in many essential body processes and its deficiency results in serious consequences for human health. Breast milk is the only source of this vitamin for children that are exclusively breastfed. Analysis of vitamin A in mother s milk
is important because its concentration is related to maternal vitamin A status and to its ingestion by the mother during pregnancy. The aim of the present study was to assess the effect of maternal supplementation with retynil palmitate on the concentration of colostrum
retinol under fasting and postprandial conditions. A total of 149 nursing mothers were recruited at the Januário Cicco Maternity School (Natal, Brazil) and allocated to two groups: Comparison (n = 69) and Test (n = 80). Blood and colostrum (in fasting and postprandial conditions) samples were collected up to 24hs after delivery. Serum
retinol and colostrum levels were analyzed by high-performance liquid chromatography. The serum
retinol level of 41.6 ± 12.7μg/dL (mean ± standard deviation) indicates adequate biochemical nutritional status. Colostrum
retinol level was not influenced by serum
retinol levels under any of the conditions established. In the colostrum, the
retinol concentration in the unsupplemented test group was 67.3 ± 37.7 μg/dL under fasting and 80.3 ± 35.1 μg/dL under postprandial conditions (p<0.05), showing an increase of 19.3%. In the supplemented test group the values were 102.6 ±
57.3 μg/dL and 133.4 ± 78.3 μg/dL under fasting and postprandial, respectively (p<0.05), representing an increase of 30%. Considering that under fasting conditions most of the vitamin A transported to the milk originates in the
retinol binding protein (RBP), the postprandial increase in colostrum
retinol suggests a different transport mechanism of
retinol to maternal milk from that performed by RBP. This situation becomes more evident under supplementation conditions.

▼ Mothers with good vitamin A nutritional status during gestation and lactation are better able to nourish and protect their infant with maternal milk. Our hypothesis is that women with more serum
retinol have more
retinol and secretory immunoglobulin A in colostrum. 190 healthy puerperal women from a Brazilian public maternity were recruited and divided according to the cutoff point for serum
retinol (30 μg/dL). A number of the women was supplemented with 200000 UI (60 mg) of retinyl palmitate in the immediate postpartum. Serum and colostrum were collected on the 1st day postpartum and colostrum again on the following day.
Retinol (serum and colostrum) was analyzed by HPLC and SIgA (colostrum) by turbidimetry. The mothers presented with adequate biochemical indicators of nutritional status, according to serum
retinol (44.6 μg/dL). There were significant differences (p= 0.0017 and p= 0.043, respectively) in
retinol and SIgA levels in the colostrum of mothers with serum
retinol > 30 μg/dL and < 30 μg/dL. The concentration of SIgA in the colostrum of non-supplemented mothers on the 1st day postpartum was 822.6 mg/dL, decreasing after 24 hours to 343.7 mg/dL. Supplemented mothers showed levels of SIgA in colostrum of 498.9 mg/dL on the 2nd day postpartum (p= 0.00006). The colostrum of women with good vitamin A nutritional status had more
retinol and SIgA. Additionally, maternal supplementation increases the levels of SIgA in colostrum. The higher levels of SIgA on the 1st day postpartum showed the importance of early breastfeeding, given that it provides considerable immunological benefits to newborn infants

▼ min A deficiency (VAD) is a nutritional deficiency that can be caused by
insufficient intake of food sources of vitamin A, or due to problems in their absorption,
transport or use. This is a problem of great importance because of the deleterious
consequences that require the body, such as compromising the immune system,
increasing susceptibility to infections, damage caused in the process of growth and
development, and affect vision and may cause from night blindness to irreversible
blindness nutritional. Children under five years are one of the largest groups of
biological vulnerability, particularly in post-weaning. It is therefore of fundamental
importance to establish the prevalence of vitamin A deficiency in this group to enable
appropriate information to the planning of measures to prevent and control this
situation. The objective was to estimate the prevalence of vitamin A deficiency in
children from 0 to 59 months of the semiarid region of Alagoas. For that, it was
determined the serum retinol by high performance liquid chromatography resolution on
a probability sample comprised 652 children. The prevalence of inadequate plasma
retinol levels (<20μg/dL) was 44.8%. There was no significant difference in mean
retinol by age. There was a statistically significant difference (p = 0009) in relation to
gender, with higher rates of inadequacy among girls (55.4%). The World Health
Organization states that a VAD prevalence of less than 20% characterized the situation
as serious public health problem. Thus, this condition is more than set for the semiarid
region of Alagoas, in that more than twice the cutoff point. Considering the existence of
a national program established by the Ministry of Health directed the control of VAD,
these results demonstrate the need for this to be properly assessed as to its operation and
effectiveness.

A deficiência de vitamina A (DVA) é uma carência nutricional que pode ser causada
pela ingestão insuficiente de alimentos fontes de vitamina A, ou devido a problemas em
sua absorção, transporte ou utilização. Trata-se de um problema de grande importância
devido às conseqüências deletérias que impõe ao organismo humano, tais como o
comprometendo do sistema imune, aumentando a susceptibilidade às infecções, os
prejuizos causados no processo de crescimento e desenvolvimento, além de afetar a
visão, podendo causar desde a cegueira noturna até a cegueira nutricional irreversível.
As crianças menores de cinco anos constituem um dos grupos biológicos de maior
vulnerabilidade, sobretudo no período pós-desmame. Assim, é de fundamental
importância estabelecer a prevalência de hipovitaminose A nesse grupo de modo a
possibilitar informações adequadas para o planejamento de medidas de prevenção e
