▼ Methods to control thatch layers in mature turfgrasses have relied on sand-topdressing and/or mechanical removal. Mechanical removal of thatch through vertical mowing and aerification is effective but disruptive to playing surfaces compared to sand topdressing. As a result, many turf managers have opted to implement sand-topdressing regiments as their primary method for reducing thatch buildup. This research was conducted to 1) determine the effect of cultivar on thatch decomposition and 2) examine the effect of sand topdressing on thatch microenvironment and decomposition. Sand topdressing treatments consisting of sterilized or non-sterilized sand applied at 0.4 cm 14 d-1 or as a single application at 1.2 cm and an untreated control to two hybrid bermudagrasses (Cynodon dactylon L.), ‘Tifway’ and ‘Celebration’ from May to September in 2008 and 2009. At the initiation of the experiment, Celebration had twice the thatch layer of ‘Tifway’. The only treatment that reduced thatch was sand applied every 14 d-1 reduced thatch 21% to 34% and 20% to 30% for ‘Tifway’ and’ Celebration’, respectively. In contrast, a single sand topdressing application led to net accumulations of 20% to 30% compared to accumulations of 29% to 38% increases in untreated plots. Sand applied more frequently resulted in higher microorganism populations and had no detrimental effect on turfgrass quality. Routine sand applications increased thatch relative humidity (RH) compared to untreated controls for both hybrid-bermudagrass cultivars. Laboratory experiments were conducted to examine the effect of temperature, 20 C and 30 C, and RH (80%, 90%, 95%, >99%) on ‘Tifway’ and ‘Celebration’ thatch decomposition. Increasing temperature and RH resulted in 189% to 397% increase in microbial degradation. Failure to provide adequate moisture reduced microbial activity and led to declines of 170% to 243% in decomposition when thatch was subjected to drying conditions. Because thatch tissue composition and response to changes in temperature and RH were similar between cultivars, newer more vigorous hybrid-bermudagrass such as ‘Celebration’ will require a frequent sand topdressing regiment in conjunction with mechanical removal for acceptable thatch control.

▼ While proton exchange membrane (PEM) fuel cells have been the focus of development in the past, anion exchange membranes (AEM) have the potential to dramatically lower the cost of fuel cells by utilizing non-noble catalysts and a variety of fuel sources. Although chemical degradation typically dominates membrane failure pathways in a fuel cell, mechanical breakdown due to humidity cycling is a common occurrence. This thesis aims to understand the mechanical properties of anion exchange membranes under fuel cell operating conditions. A humidity delivery system was developed for the TA Instruments ARES-G2 rheometer to allow for testing at a range of temperatures (30-100°C) and relative humidity conditions (0-95% RH). A modified Sentmanat Extensional Rheometer (SER) was used to perform tensile-like testing using less than 5% of material needed for a traditional tensile tester. These tools established metrics for a robust membrane through mechanical characterization across temperatures and humidities. A pentablock AEM with a balance of stiff and elastic blocks was shown to have adequate conductivity (up to 60 mS/cm at 90°C and 95%RH), low water uptake (<25%), and good mechanical integrity under dry and hydrated conditions, showing potential for being durable under hydration and mechanical stresses. Complementing the destructive tensile testing, a “water stress” test was explored to measure the tension and durability under hygral cycles. Membranes with a low (<5 MPa) and near constant water stress absorb and desorb water reversibly. The materials that performed poorly in the water stress tests also had elongation <50% under dry conditions and swelled with water. Membranes performing well in the water stress test also had an elongation to break 10 times that of its in-plane water swelling (in liquid water). Ion exchange membranes need to be able to mechanically stretch in the elastic region well above the in-plane swelling with water to withstand hygral stresses in an electrochemical device. By identifying a relationship between the mechanical and hygral stretching to predict durability in a working device, this thesis advanced the understanding of mechanical performance under relevant temperature and humidity conditions, which is critical to the development of durable anion exchange membranes. ▼ Les matériaux cimentaires tels que le béton, le ciment et le plâtre sont largement utilisés dans la construction, les matières premières dont ils sont faits étant abondantes sur Terre. Cette tendance ne devrait pas changer dans les prochaines décennies. Mais ces matériaux subissent l'impact du fluage. Le fluage des matériaux cimentaires est une problématique complexe. D'une part, dans les matériaux cimentaires, le fluage est souvent couplé avec d'autres phénomènes tels que le séchage, l'hydratation et la fissuration, et peut être influencé par différents paramètres comme la température, le niveau de contrainte, la teneur en eau et la formulation. D'autre part, la mesure du fluage par un test macroscopique traditionnelle du fluage requiert du temps (il est recommandé de réaliser l'essai de fluage du béton sur plusieurs mois afin de donner une caractérisation fiable du fluage à long terme) et s'avère fastidieuse, puisque les paramètres expérimentaux doivent être bien contrôlés sur de longues périodes de temps. Cette thèse étudie la micro indentation à l'échelle de la pâte de ciment ou du plâtre pour évaluer les propriétés de fluage propre à long terme des matériaux cimentaires, en comparant les fonctions de fluage obtenues par des tests de micro indentation de quelques minutes avec celles obtenues par des expériences macroscopiques de fluage réalisées pendant de longues années. Pour la pâte de ciment, la comparaison a été faite à l'échelle du béton à l'aide d'une certaine homogénéisation. L'étude a validé le fait que un test de micro indentation de quelques minutes peut fournir une mesure des propriétés à long terme de matériaux cimentaires. Une fois validée la technique d'indentation, nous avons étudié l'effet de la microstructure (c'est-à-dire la distribution des phases) et celui de l'eau sur le fluage propre à long terme des matériaux cimentaires. L'effet de la microstructure a été étudiée sur des matériaux tels que des pâtes de C3S et de C2S ainsi que sur des compacts de C-S-H synthétique, de portlandite (CH) et leurs mélanges préparés par compression de poudres. Une attention particulière a été consacrée à créer des compacts avec de grandes fractions volumiques de phase cristalline. Pour tous les échantillons examinés, nous avons identifié le bon modèle micromécanique qui permette de prédire les résultats. Le choix du modèle micromécanique concorde avec les observations microstructurales. L'effet de l'humidité relative a été étudié par le conditionnement et l'indentation de certains de ces matériaux (par exemple la pâte de C3S, de compact de C-S-H et de compact de CH) dans différentes humidités relatives allant de 11% à 94%. L'humidité relative a eu un effet significatif sur le fluage : pour tous les matériaux testés, une plus grande humidité a conduit à un fluage plus important. Le béton est un matériau hétérogène complexe dont les déformations comportent une partie différée qui est affectée par un grand nombre de facteurs tels que la température, l'humidité relative et l'évolution de la microstructure. La prise en compte des déformations différées et en particulier du fluage est indispensable dans le calcul des ouvrages en béton tels que ceux destinés à stocker des déchets radioactifs. Ce travail de thèse a pour objectifs : (1) de développer un modèle de fluage simple et robuste pour le béton en faisant appel à la micromécanique et en tenant compte de l'endommagement et des effets thermiques et hydriques ; (2) d'implanter numériquement le modèle développé dans un code de calcul par éléments finis de façon à pouvoir simuler le comportement d'éléments de structure simples en béton. Pour atteindre ce double objectif, le travail est scindé en trois parties. Dans la première partie, le matériau cimentaire est à l'échelle microscopique supposé être constitué d'une matrice viscoélastique linéaire caractérisée par un modèle de Maxwell généralisé et de phases particulaires représentant les granulats élastiques et les pores. Le schéma micromécanique de Mori-Tanaka, la transformée de Laplace-Carson et son inversion sont alors utilisés pour obtenir dans l'espace temporel des estimations analytiques ou numériques de ses paramètres mécaniques et hydromécaniques. Ensuite, le modèle micromécanique de fluage obtenu est couplé au modèle d'endommagement de Mazars via le concept de pseudo-déformations introduit par Schapery. Les paramètres intervenant dans le modèle viscoélastique endommageable ainsi établi sont systématiquement identifiés à l'aide de données expérimentales. Enfin, la prise en compte des effets de la température et de l'humidité relative dans le modèle viscoélastique endommageable est basée sur la méthode du temps équivalent ; l'efficacité de cette approche est démontrée et discutée dans le cas de chargements simples de fluage

Concrete is a complex heterogeneous material whose deformations include a delayed part that is affected by a number of factors such as temperature, relative humidity and microstructure evolution. Taking into account differed deformations and in particular creep is essential in the computation of concrete structures such as those dedicated to radioactive waste storage. The present work aims: (1) at elaborating a simple and robust model of creep for concrete by using micromechanics and accounting for the effects of damage, temperature and relative humidity; (2) at numerically implementing the creep model developed in a finite element code so as to simulate the behavior of simple structural elements in concrete. To achieve this twofold objective, the present work is partitioned into three parts. Darbo metodai - Empirinio tyrimo metodas paremtas duomenų rinkimu medieną sveriant, atrenkant mėginius ir juos džiovinant. Kiekybinis palyginimas - statistinis duomenų apdorojimas paremtas dispersine ir regresine analize ir taikant koreliacinius ryšius. Visą empirinę medžiagą sudarė 180 medvežių imtis su spygliuočių, lapuočių ir smulkintoji mediena. Tiriama medienos tankio ir drėgnio koreliaciniai ryšiai, taip pat šių parametrų kintamumas nuo medžių rūšies, meteorologinių sąlygų: juos kertant ir sandėliuojant. Darbo rezultatai. In this Master's paper an analysis is carried out to calculate the probability of estimating the volume of timber using the method of weighing. Work -study : Logs and flaked wood used for producing wood cutting panelling The aim of the work: Analysing the changes in thickness of logs and flaked wood used for producing wood cutting panelling and converting its weight into bulk. Methods of work: Empiric method based on weighing the timber, sorting out the samples and drying them. Quantitative analysis - processing the statistic data based on dispersive and regressive analysis and using correlation relations. The empiric analysis covered 180 trucks of softwood, deciduous wood and cutting.The correlation between the thickness and the humidity has been analysed. It has been also analysed how the thickness and humidity change in different species of trees, meteorological conditions, cutting down the trees and storing them. The results of the work: It has been decided that the greatest influence on the thickness of the wood has the density of the wood. ▼ As nowadays the economic activities around the world interact frequently and closely, marine shipping is an important approach for international exchange of capitals, goods, and services. However, the associated air pollution is also becoming a serious environmental issue of concern.Kaohsiung harbor is the largest international port in southern Taiwan and plays an important role in the national economic development, whereas its long-term emissions of a variety of air pollutants such as total suspended particle (TSP) cargo loading and unloading is notorious environmental problem. The main sources for the air pollution in this areainclude the loading activities on a number of piers, while the materials to handle typically are composed of iron waste (scrap), coke, silica sand, ore, limestone and cement. The objective of this study is to investigate the feasibility of usingneural network system to predict the TSP concentrations from different piers and to study the important factors that influence the TSP emissions. The TSP concentrations analyzed from 2010 to 2012 were used as the database to study. The variables included material density, relative humidity, wind direction and wind speed, as the dependent variable was the TSP concentration. In the results of developing the neural network for learning,the mode 1, which separate the inputdata (for learning) and the check data (for demonstrating) by their sizes, helped the neural network generate more accurate results. The percentage of the relative error was the lowest (13%) when the hidden layer was three. By optimizing proper factors, the neural network established in this study was capable to provide reasonable predictions for the possible TSP concentrations on the piers in Kaohsiung Harbor by knowing the information including the material density, relative humidity, wind direction and wind speedoneach harbor. The major factors affecting the TSP concentration in the harbor includedthe material density and relative humidity. With properly known atmospheric characteristics such as those considered in this study, the neural network investigated in this study provides a preliminary result that can employed to decide whether it is necessary to strengthen the precautionary measures or to perform other strategies to curb the TSP concentrations in a harbor area such as Kaohsiung Harbor in this studyã Advisors/Committee Members: Wei-Hsiang Chen (committee member), Chun-Hu Chen (chair), Shu-Kuang Ning (chair).

Healthcare systems including some low-resource settings have begun implementing these technologies providing the convenience and reduction of large laboratory set-ups. Low-cost is one of the main driving components when it comes to point-of-care diagnostics. Paper-based microfluidics has generated a great amount of interest for the development of low-cost diagnostic and self-contained analytical devices. Satisfying the World Health Organization’s (WHO) recommended ASSURED criteria; Affordable, Sensitive, Specific, User-friendly, Rapid and robust, Equipment free, Deliverable, paper-based microfluidics have made point-of-care testing more accessible. Applications range from healthcare, food safety, and environmental monitoring, among others. What has in part attracted attention is the low-cost, ease-of-use, and adaptability of these paper devices. Compared to conventional microfluidic devices, the paper-based counterparts are able to utilize paper’s inherent wicking property to eliminate the external pumping needed to drive the fluid. Channels are easily formed by either selectively removing sections of the paper substrate or by pattering channel boundaries with a hydrophobic material. In spite of the benefits and advantages described above, paper-based microfluidic technologies often lack the necessary sensitivity and sophistication available in conventional microfluidic devices. In order to be a competitive alternative, paper-based microfluidics require improvement and novel development of feasible detection methods. These methods will likely require increasingly complex chemistry and control of reagents. Thus, understanding imbibition as well as obtaining precise, accurate, and consistent fluid handling within the paper device will be crucial.Although considerable knowledge exists on techniques to manipulate fluid within the paper channel, what is lacking are studies on how non-laboratory conditions (e.g. relative humidity) influence fluid flow. This presentation aims to address this gap with particular focus on the effects of relative humidity and channel width. A series of controlled imbibition experiments is reported using cellulose papers commonly used in the field of paper-based microfluidics. We show that both the imposed relative humidity and the channel width have critical design considerations in paper-based devices. Additionally, we compare three models, the Lucas-Washburn model, the Fries et al. (2008) model which incorporates evaporation, and a newly developed water saturation model that incorporates evaporation as well as residual water in the paper. We assess their accuracy in representing the experimental data and systematically evaluate the importance of evaporation and water saturation under a wide range of relative humidity conditions. ▼ Approximately 20% of fruit and vegetable production is lost annually due to post-harvest spoilage. Technologies exist that reduce the spoilage during transport and storage, such as ethylene-permeable bags and controlled environment crispers. Although methods exist for controlling environmental conditions to a closer tolerance, these are currently cost-prohibitive. This research focused on the direct relationship between produce spoilage and tightly controlled humidity and temperature settings, with the goal of generating new information capable of driving innovation in the field. The objectives of this research were to store a model fresh fruit (strawberries) and a vegetable (romaine lettuce) in an environmental chamber capable of maintaining and reporting specified humidity and temperature conditions, and quantifying spoilage effects at various temperature and humidity levels. Ultimately, the goal is to provide the industry with a better understanding of the intersection between controllable food storage conditions, microbial spoilage, and food safety. Nine spoilage tests were completed at 7yC, 5yC and 3yC at relative humidities of 95%, 75% and 50% (see Figure 1). The overall results can be seen in Table 2 and the summary is as follows: Romaine lettuce heads lasted the longest, and stayed the freshest at high humidity (~95%), with temperature having a lesser impact. If humidity control is not possible, aseptically separating leaves and storing them individually in sterile, sealable bags will enhance organoleptic quality at low temperatures (~3yC), although this is probably not possible in practice. Strawberries stored at low temperature and humidity (~3yC and ~50%) resisted molding the longest, and lost firmness after 1-2 weeks. To balance the preservation of freshness and firmness, storage at low temperature and high humidity (~3yC and ~95%) was found to be ideal. Initial microbial count had important impacts on mold growth rate. Either high mold inoculum or metabiotic interactions between spoilage bacteria and molds may be responsible for these effects.

▼ Despite formidable advances in virology and medicine in recent decades, we know remarkably little about the dynamics of the influenza virus in the environment during transmission between hosts. There is still controversy over the relative importance of various transmission routes, and the seasonality of influenza remains unexplained. This work focuses on developing new knowledge about influenza transmission via the airborne route and the virus' inter-host dynamics in droplets and aerosols. We measured airborne concentrations of influenza A viruses (IAVs) and size distributions of their carrier aerosols in a health center, a daycare center, and airplanes. Results indicate that the majority of viruses are associated with aerosols smaller than 2.5 µm and that concentrations are sufficient to induce infection. We further modeled the fate and transport of IAV-laden droplets expelled from a cough into a room, as a function of relative humidity (RH) and droplet size. The model shows that airborne concentrations of infectious IAV vary with RH through its influence on virus inactivation and droplet size, which shrinks due to evaporation. IAVs associated with large droplets are removed mostly by settling, while those associated with aerosols smaller than 5 µm are removed mainly by ventilation and inactivation. To investigate the relationship between RH and influenza transmission further, we measured the viability of IAV in droplets at varying RHs. Results suggest that there exist three regimes defined by RH: physiological conditions (~100% RH) with high viability, concentrated conditions (~50% to ~99% RH) with lower viability, and dry conditions (<~50% RH) with high viability. A droplet's extent of evaporation, which is determined by RH, affects solute concentrations in the droplet, and these appear to influence viability. This research considerably advances the current understanding of the dynamics of the influenza virus while it is airborne and provides an explanation for influenza's seasonality. Increased influenza activity in winter in temperate regions could be due to greater potential for IAV carrier aerosols to remain airborne and higher viability of IAV at low RH. ▼ Roxithromycin is a semi-synthetic, macrolide antibiotic, derived from erythromycin A. It acts as a bacteriostatic drug at low concentrations and a bactericidal drug at high concentrations. It binds to the 50S subunit of the 70S ribosome, which causes the reversible inhibition of RNA-dependent bacterial protein synthesis. It is well known that active pharmaceutical ingredients (APIs) may exist in numerous solid states. Differences in the solid state significantly influence the physical and chemical properties of an API. The in vivo performance of a dosage form will also be influenced by the solid state properties of a given pharmaceutical active. The amorphous characteristics of APIs have a significant impact on their performance and thus offer the potential for exciting new pharmaceuticals. Whilst amorphous forms of poorly soluble APIs are more soluble than their crystalline counterparts, they tend to be physically unstable, which makes their formulation into solid dosage forms quite challenging. Roxithromycin has only 50% oral bioavailability due to its poor aqueous solubility and for this reason, its potential for optimal therapeutic effect are limited. Poor solubility is thus an important obstacle in formulation development. During this study, amorphous forms of roxithromycin were prepared via quench cooling, and desolvation of chloroform- and ethyl acetate solvates. These amorphous forms were characterised by means of several techniques, whilst their solubilities and stabilities were also investigated. The outcomes of the solubility studies illustrated the complexity of this API and its amorphous forms with regards to their interactions with water. Solubility studies confirmed the superior solubility of the roxithromycin glass (prepared through quench cooling) and amorphous forms (desolvation of solvates) over the roxithromycin monohydrate in water. The solubility in water improved in the order of roxithromycin monohydrate < roxithromycin glass < roxithromycin glass powder < amorphous chloroform desolvate. The roxithromycin monohydrate, as well as the amorphous forms of roxithromycin demonstrated stability over a one-month period of exposure 40°C and relative humidity (RH) of 75%. The roxithromycin glass powder tended to revert to the more stable crystalline monohydrate after week 3 of stability testing. The roxithromycin glass at lower temperatures of 25°C and 30°C (both at 75% RH) tended to transform into the more crystalline form at week 4 of the study. These transformations were, however, not as significant as during the 40°C / 75% RH study. The conclusion could therefore be made that this transformation into the crystalline form was more temperature – than moisture dependant. At a higher temperature (at identical humidity conditions), the transformation into the crystalline form was much faster. So far, freeze drying is regarded as the most suitable method to preserve the functional properties in blueberries. The high operation cost being the drawback of this method, alternative air drying and pre-treatment techniques were investigated aiming at finding a more cost effective method that can give high quality products comparable to freeze dried ones.The study examined the effect of temperature and relative humidity (RH) on the drying kinetics as well as on phenolics, anthocyanin content and antioxidant activity of the blueberries. Low temperature drying was performed at 30 °C and 15% RH using a heat pump dryer. High temperature drying was performed at 60, 70 and 80 °C with RH of 10, 15 and 20% using a cabinet dryer. The effects of pre-treatments including osmotic dehydration, mechanical abrasive wax removal and blanching were also investigated.The results showed lower functional properties of all air dried blueberries compared with the freeze dried fruit. A significant reduction of antioxidant content and activity was observed following low temperature drying in comparison with high temperature drying due to a prolonged drying time. However, the wax abrasive and blanching pre-treatments aided in reducing degradation of the phenolic compounds and preserving antioxidant activity by reducing drying time and inactivating enzymes that are responsible for anthocyanin degradation, respectively. Osmotic dehydration, in contrast, caused a greater loss of beneficial compounds and activities after dipping in a sucrose solution.The analysis by HPLC and LC/MS showed high concentration of anthocyanin and a phenolic compound namely, chlorogenic acid, in fresh southern highbush (Star, Sharpe, Crunchy) and rabbiteye blueberries (Climax, Brightwell and Powderblue). Theanthocyanins detected in all blueberry cultivars were monoglycosides of 5 anthocyanidins including delphinidin, cyanidin, petunidin, peonidin and malvidin. Among these, rabbiteye species were more abundant in malvidin followed by cyanidin derivatives whereas southern highbush species contained high concentration of malvidin and delphinidin. Peonidin glycosides had the lowest proportion within total anthocyanins in all blueberry cultivars. After drying, the levels of individual anthocyanins decreased in different manner depending on the cultivars. However, the proportion of each of them remained similar to that in the fresh berries.The blueberries dried using abrasive and blanching pre-treatment were also selected to be tested for their anti-proliferative properties (against cancer cells) given their high antioxidant content. ▼ The development of a control-oriented dynamic relative humidity model for a polymer electrolyte membrane (PEM) fuel cell stack is presented. This model is integrated with a first law based thermal model, which tracks energy flow within four defined control volumes in the fuel cell; the cathode channel, anode channel, coolant channel, and fuel cell stack body. Energy and mass conservation equations are developed for each control volume. On top of mass conservation, electro-drag and osmosis models were also implemented within the model to account for the major modes of vapor transfer through the membrane between the anode and cathode. Requisite alterations to the thermal model as well as mass flow rate calculations are also discussed. Initially, the model utilized a single lumped control volume for the calculation of all values each channel (anode and cathode). This lumped value method is computationally inexpensive, and makes the model optimal for control design. However, investigation of the mass-based Biot number showed the need for greater granularity along the length of the channels to properly capture the relative humidity dynamics. In order to improve the resolution of the model, while still minimizing the computation expense, the model was subdivided into a series of lumped value models. The cathode channel was the point of focus as it is the major concern from a controls perspective. This method captures the proper trends found in far more complex CFD models, while still maintaining a quick calculation time. Different levels are subdivision (3 and 6 submodels) are investigated, and the differences discussed. Particularly, temperature range, relative humidity range, the effect on the modeled voltage, and calculation time are compared. This control-oriented model is low order and based on lumped parameters, which makes the computational expense low.
