L'algorithme FastICA est l'un des algorithmes les plus populaires dans le domaine de l'analyse en composantes indépendantes (ICA). Il existe deux versions de FastICA: Celle qui correspond au cas où l'échantillon est de taille infinie, et celle qui traite de la situation concrète, où seul un échantillon de taille finie est disponible. Dans cette thèse, nous avons fait une étude détaillée des vitesses de convergence de l'algorithme FastICA dans le cas où la taille de l'échantillon est finie ou infinie, et nous avons établi cinq critères pour le choix des fonctions de non-linéarité. Dans les trois premiers chapitres, nous avons introduit le problème de l'ICA et revisité les résultats existants. Dans le Chapitre 4, nous avons étudié la convergence du FastICA empirique et le lien entre la limite de FastICA empirique et les points critiques de la fonction de contraste empirique. Dans le Chapitre 5, nous avons utilisé la technique du M-estimateur pour obtenir la normalité asymptotique et la matrice de covariance asymptotique de l'estimateur FastICA. Ceci nous a permis aussi de déduire quatre critères pour choisir les fonctions de non-linéarité. Un cinquième critère de choix de non-linéarité a été étudié dans le chapitre 6. Ce critère est basé sur une étude fine de la vitesse de convergence de FastICA empirique. Nous avons illustré chaque chapitre par des résultats numériques qui valident nos résultats théoriques.

The FastICA algorithm is one of the most popular algorithms in the domain of Independent Component Analysis (ICA). There exist two versions of FastICA: the one that corresponds to the ideal case that the sample size is infinite, and the one that deal with the practical situation, where a sample of finite size is available. In this thesis, we made a detailed study of the rate of convergence of the FastICA algorithm of both versions, and we established five criteria for the choice of the non-linearity function. In the first three chapters, we introduced the problem of ICA and revisited the classical results. In Chapitre 4, we studied the convergence of empirical FastICA and the link between the limit of empirical FastICA and the critical points of the empirical contrast function. In Chapter 5, we used the technique of M-estimator to obtain the asymptotic normality and the asymptotic covariance matrix of the FastICA estimator. This allowed us to derive four criteria to choose the non-linearity function. A fifth criterion for the choice of the non-linearity function was studied in Chapter 6. This criterion is based on the rate of convergence of the empirical FastICA algorithm. At the end of each chapter, we provided numerical simulations that validate our theoretical results.

▼ Background: Workers in a platinum group metals (PGMs) refinery are potentially exposed to various precious metals (iridium, osmium, palladium, platinum, rhodium and ruthenium) and their metal-salt compounds which may cause rhinitis, asthma, contact urticaria and conjunctivitis. Some cases revealed that sensitisation occurred in employees where it was not possible to detect any airborne soluble platinum or where the respiratory soluble platinum exposure was below the occupational exposure limit. It is unclear whether respiratory exposure or a combination of respiratory and dermal exposure may be involved in sensitisation and the possible elicitation of skin symptoms. Objectives: To determine if dermal exposure to PGMs took place during the refining process and in the administration area by using a removal method and to compare dermal exposure on the different anatomical areas and in two different working areas, Areas A and B for each of the PGMs. Methods: Dermal exposure samples were collected with a removal method using GhostwipesTM. The samples were collected from the palm of the hands, the wrists and the necks of the workers, before the shift started, before tea time, before lunch time and after the shift ended. The skin wipes were analysed for the PGMs (iridium, osmium, palladium, platinum, ruthenium and rhodium) according to Methods for the Determination of Hazardous Substances (MDHS) method 46/2, using Inductively Coupled Plasma-Mass Spectrometry. Results: No published data is available on occupational dermal exposure to PGMs in a precious metals refinery. This study proved that dermal exposure to PGMs in the refinery took place and was quantified. The PGM dermal exposure results in general, were very low (measured in nano grams), with platinum having the overall highest exposure. Exposure also occurred the most frequently during the last two intervals of the day, before lunch time and at the end of the shift. Exposure on all three the anatomical areas that were tested in the study, varied much with the palm of the hands having the highest exposure levels. There were also variations in exposure between areas A and B due to the fact that the processes in these two areas differ. Conclusions: It was confirmed that dermal exposure to PGMs took place at the precious metals refinery. The highest exposure took place before lunch time and towards the end of the shift. The metal to which the workers were exposed the most was platinum and the production area where the workers had the highest exposure to most of the metals was Area B.

▼ Vel is a universal antigen present on the red blood cells (RBCs) membrane, which defines the Vel-blood group system. The identification of Vel-encoding gene, SMIM1, suggested that this is a regulator of erythropoiesis. Its role in RBCs and potential role in other cells, however, remains largely unknown. Here, I aimed to characterise the role of SMIM1 in haematopoietic lineages and other organs by using two main approaches (in vivo and in vitro) and three models (human volunteers, mouse and cell lines). First, using available datasets, I showed that SMIM1 is expressed in megakaryocytes (MKs), platelets, neutrophils and naïve, memory and class-switched B-lymphocytes. I further demonstrated that SMIM1 localisation and the type of multimers it forms in some of those cells are cell type dependent. Second, I assessed the effect of the absence of SMIM1 in qualitative and quantitative blood traits in two human cohorts (Vel-negative blood donors recruited in this study and Vel-negative/weak individuals of the 400,000 UK Biobank study) and in a Smim1 mutant mouse model. Small but significant alterations were observed in RBC, platelets and neutrophils counts in Vel-negative individuals and Smim1 mutant mice, however, the directionality of these changes and their significance was model specific. Third, I investigated the role of SMIM1 in platelets formation and function. I showed that SMIM1 is phosphorylated during platelet activation in humans and its ablation in male mouse platelets leads to a reduction of P-selectin surface expression on resting platelets. These findings suggest a role for SMIM1 during platelet activation. Furthermore, I described a novel metabolic phenotype both in human and mouse SMIM1 knockouts. I showed that SMIM1 absence leads to an increase in body weight (and consequently in body mass index (BMI)) in male Vel-negative donors and female Smim1-/- mice. Altogether, these findings provide new insights into the role of SMIM1 and the biological processes it is involved, which may have the potential to reveal unknown clinical phenotypes related with SMIM1 ablation and to inform their prevention and treatment.

La polymérase d'A.R.N. la protéine obligatoire (le RbpA) est l'activateur transcriptional global (mondial) de l'espèce actinomycetes qui est essentielle pour la croissance et qu'augmente la tolérance de bactéries aux antibiotiques. RbpA de la tuberculose Mycobacterium (Mtb) stimule spécifiquement la transcription par la polymérase d'A.R.N. (RNAP) contenant σA ou les sous-unités σB, mais aucune de la 11 autre alternative σ des facteurs. Il a été rapporté que le fonctionnement de RbpA est dépendant de promoteur et c'est indispensable pour le déroulement de promoteur du promoteur sigAP constitutif.Pour déchiffrer la nature de spécificité de promoteur de RbpA, nous avons utilisé des essais biochimiques, mutagensis et des approches de génomique. Nous avons identifié ce remplacement 'TG' le motif entre-14 à-17 positions (postes) dans le promoteur sauvage-type sigAP fait la transcription indépendante de RbpA. Aussi, nous avons montré que la capacité d'augmentations de RbpA de RNAP pour fondre le promoteur sigAP aux températures sous-optimales et stabilise des complexes de promoteur. Mutational l'analyse de résidus d'acide aminé H166 et E169 dans la région σB 3.0 (σR3.0) a démontré une implication de σR3.0 dans la stabilisation RbpA-servie-d'intermédiaire de complexes de promoteur RNAP. Plus loin, la substitution à RbpA au résidu d'acide aminé R79 a affecté la stabilité complexe de promoteur, tandis que les substitutions à RbpA resdiues K73, K74 a affecté l'initiation de transcription. Cependant, aucun des mutants RbpA n'a étudié l'ouverture ici affectée de l'ADN de promoteur par RNAP. Les rôles différentiels joués par ces résidus RbpA dans la stabilisation de complexe de promoteur et l'initiation de transcription ensemble avec l'effet produit par les mutations σB suggèrent l'implication de R3.0 σB dans l'action de RbpA. Ensuite, nous avons exécuté la large de génome cartographie des gènes RbpA-dépendants du σB regulon en utilisant une Analyse de Puce à ADN de Finale(d'Écoulement) in vitro (des ROMS). L'analyse ROM a montré la preuve claire de 15 augmentation de pli du nombre de gènes activés par σB-RNAP en présence de RbpA. L'analyse de bio-informatique de 140 gènes contrôlés par la paire de RbpA-σB nous a permis d'identifier la signature caractéristique dans le-10 consensus ('TANNNT') spécifique à la sous-unité σB.Notre étude sur l'impact d'échelle de génome de RbpA, ensemble avec le déchiffrement du mécanisme moléculaire d'action de RbpA, souligne une importance de l'interaction entre σR3.0 et RbpA dans la transcription Mtb. Basé sur nos résultats nous proposons que RbpA puisse jouer un rôle du remplacement(remplaçant) fonctionnel pour-10 motifs prolongés(étendus) dans l'espèce mycobacterium.

▼ “A Narratological Analysis of D. M. Thomas’s The White Hotel (1981)” originated within a seminar on British Postmodernist Literature during the first Master’s Degree in “British and North-American Culture and Literature” (2001-04) at the Universidade da Madeira set up by the Department of English and German Studies. This dissertation seeks to present a narratological analysis of Thomas’s novel. The White Hotel stands as a paradigmatic example of the kind of literature that has dominated the British literary scene in the past three decades, commonly referred to as postmodernist fiction, owing to its formal craftsmanship (multiplicity of narrative voices and perspectives, mixing of differing genres and text types, inclusion of embedded narratives) alongside the handling of what are deemed as postmodernist topoi (the distinction between truth and lies, history and fantasy, fact and fiction, the questioning of the nature of aesthetic representation, the role the author and the reader hold in the narrative process, the instability of the linguistic sign, the notion of originality and the moral responsibility the author has towards his/her work), The narratological approach carried out in this research reveals that Thomas’s text constitutes an aesthetic endeavour to challenge the teleological drive that is inherent in any narrative, i. e., the inevitable progression towards a reassuring end. Hence, the subversion of narrative telling, which is a recurrent feature in Thomas’s remaining literary output, mirrors the contemporary distrust in totalising, hierarchised and allencompassing narratives. In its handling of historical events, namely of the Holocaust, The White Hotel invites us to reassess the most profound beliefs we were taught to take for granted: progress, reality and truth. In their place the novel proposes a more flexible conception of both the world and art, especially of literary fiction. In other terms, the world appears as a brutal chaotic place the subject is forced to adjust to. Accordingly, the literary work is deemed hybrid, fragmented and open. So as to put forth the above-mentioned issues, this research work is structured in three main chapters. The initial chapter – “What is Postmodernism?” – advances a scrutiny not only of the seminal but also of more recent studies on postmodernist literary criticism. Following this, in Chapter II – “Postmodernist British Fiction” – a brief overview of postmodernist British fiction is carried out, focusing on the fictional works that, in my opinion, are fundamental for the periodising of British postmodernism. In addition, I felt the need to include a section – “D. M. Thomas as a Postmodernist Novelist” – in which the author’s remaining literary output is briefly examined. Finally, Chapter III – “A Narratological Analysis of The White Hotel” – proposes a narratological analysis of the novel according to the particular Genettian analytical model. This work analyses an existing mobile-finance scheme at Portuguese PT Inovação, targeting users that do not have a bank account, and using the USSD communication channel to process financial transactions between three parties: the User, an Agent that represents, or acts on behalf of, an institution, but not necessarily a bank or a financial one, and the Financial Transaction Manager (FTM) that manages the Agent network, the Users and the transactions made. We start by analyzing USSD communications: by itself it is not a secure communications channel, but it is available at every GSM device, allows for instant messaging services and is inter-operable, i.e. is not telecom dependent. Besides, it can run on commodity mobile phones, and requires practically no software download. From the user point of view, it resembles a normal text message and requires no special communications contract with the telecom operator other than the one that allows for sending text messages. It presents some security issues, namely, no authentication, no confidentiality, no integrity. We demonstrate that these issues can be solved through the use of end-to-end secure protocols on top of USSD in addition to other security mechanisms. PT Inovação’s m-finance scheme already implements a set of operations and financial transactions. We analyze the system’s threat model and we propose a solution that will protect a specific communication path, namely, between the Agent and the FTM. We suggest the implementation of SSL/TLS over USSD, a lightweight version that we call USSL/UTLS. We demonstrate that it is feasible to implement such security mechanism on a USSD communication channel, and that it provides end-to-end security over the network communication path, at least if the devices present some processing capabilities. We propose some possible implementation paths, and conduct a brief performance analysis.
