▼ Purpose: Sperm nuclear proteins and DNA integrity have been implicated in infertility and treatment failures. High stallion to stallion variability is observed in sperm cryopreservation protocols. The cells are destroyed with harsh chemicals prior to using biochemical assays to test sperm DNA quality. The feasibility of using Raman spectrometry in combination with a laser trap for non-destructive micromanipulation and characterization of DNA damage in motile stallion and human sperm is experimentally investigated in this thesis. Methods: Live stallion sperms were subjected to controlled cellular damage: (a) four grades of chemically induced oxidative stress using Xanthine – Xanthine Oxidase (b) three grades of osmotic stress using PBS and (c) membrane damage using thermal shock. Live human sperm DNA disintegration with time and oxidative stress were explored on fresh, cryopreserved and swim-up categories. The specimens ranged from sub-fertile patients to fertile donors in a limited study. Post-treatment sperms resuspended in sperm media, placed on a quartz coverslip were trapped with a 785 nm, 25 mW laser, using a 1.4 NA, 60X, water immersion microscope objective. A Raman spectrum of a trapped cell was acquired for 20 – 30 seconds. The spectra from 20 – 40 cells from each specimen were analysed in the 630 cm-1 – 1630 cm-1 region using statistical variance and PCA. Results: The Raman spectra from trapped motile sperm head contain intense peaks that did not require smoothing prior to analysis. PCA of the Raman spectra could not resolve the different grades of applied osmotic and oxidative stress in stallion cells. PCA showed high variability between specimens from the same stallion and between stallions, with distinct clustering by ejaculate. Membrane damage study and spectra from extended trapping also showed distinct specimen to specimen difference within and between stallions. Specimen to specimen variability is observed in motility and viability tests on 1000s of stallion cells using CASA and flow cytometry. Human sperms showed some clustering by category, time, stress and motility and appeared more sensitive to the tests than stallion sperms. Conclusions: Raman spectra originate from the dense region of the trapped sperm head and resemble the fingerprint of dense calf thymus DNA. The cells show species specific response to the applied stress/damage. Stallion sperms show high variability between ejaculates that could not be differentiated by stallions. Human cells appear more sensitive to the applied processes. LTRS of live sperms needs further detailed research, cross correlated with other established complementary techniques, to identify spectral bands that are most sensitive to the various grades of induced DNA and membrane damage.

A espectroscopia Raman é uma das tecnologias ópticas que tem ganho cada vez mais destaque na oncologia. Esta tecnologia fornece a composição molecular dos mais variados tipos de amostras, inclusive biofluidos, células e tecidos. Na tentativa de avaliar sua aplicação em lesões bucais, o objetivo deste trabalho foi obter um perfil espectral dos distúrbios epiteliais nãodisplásicos, displásicos, e do carcinoma espinocelular bucal, bem como verificar a possibilidade diagnosticar estas alterações pelos espectros. Inicialmente, foram usados 44 amostras de mucosa bucal em blocos de parafina, com diagnóstico de hiperplasia epitelial, hiperparaceratose, displasia epitelial e carcinoma espinocelular bucal. Da porção epitelial de cada uma destas amostras foram obtidos 10 espectros Raman. A média dos espectros de cada alteração gerou um perfil espectral correspondente à sua composição química. Os resultados desta análise apresentaram picos ou bandas espectrais referentes a moléculas usualmente encontradas nos tecidos biológicos: proteínas, lipídios e DNA. Em seguida, os dados espectrais foram submetidos a análise dos componentes principais (PCA) e análise discriminante linear (LDA), onde foi encontrada uma eficiência de 86,36% no uso da Espectroscopia Raman para o diagnóstico de alterações epiteliais. A validade do método foi comprovada pela Curva Roc resultando em 91% de sensibilidade e 96,9% de especificidade. Em um segundo momento, com a finalidade de avaliar a influência da fixação com formalina nas análises espectrais, a superfície epitelial de uma amostra de tecido bucal fresca ex vivo foi submetida a análise pela Espectroscopia Raman e armazenada em formalina tamponada a 10% por 24h para posteriormente se obter outro espectro. A análise comparativa entre os perfis espectrais de uma mesma amostra antes e após a fixação, demonstraram diferenças em relação aos picos de proteínas e DNA, que podem modificar o perfil espectral da alteração analisada e, portanto, gerar algum erro de classificação. De maneira geral, pelos resultados desta pesquisa, observou-se que a Espectroscopia Raman é uma tecnologia eficiente para identificar a composição molecular de amostras de tecido, seja imediatamente após a excisão cirúrgica, fixação em formalina ou emblocadas em parafina. Entretanto, deve-se considerar que estas a diferentes formas de condicionamento influenciarão diretamente no resultado dos espectros.

Raman spectroscopy (RS) is one of the optical technologies that has garnered a great deal of interest in oncology. This technology provides the molecular composition of various types of samples, including biofluids, cells and tissues. ▼ Advanced composites play important roles in the materials sciences, military, space and commercial applications. The desirable load transfer and mechanical strength of reinforced polymers are crucial for developing advanced composites. Owing to their excellent mechanical properties derived from the sp2 bonding structure and the nanoscale size, nano-carbons are attractive materials used for nanoscale reinforcement of polymer composites. This dissertation describes a novel method to develop polymer composites using near infrared (NIR) photon-assisted polymerization and nanoscale reinforcement. We used multi-walled carbon nanotubes (MWNTs), reduced graphene oxide (RGO), and graphene nanoplatelets (GNPs) to make polymer composites, and explored in-situ NIR photon assisted heating of these nano carbons to enhance polymerization of the nano-carbon/polymer interface, thus achieving significant load transfer and improved mechanical properties. To specify, nano-carbon was dispersed into the polymer matrix by shear or evaporation mixing method to attain a uniform distribution in the prepared thin film composite. The thin film was exposed to NIR light during polymerization instead of conventional oven based heating. NIR was effectively absorbed by nano-carbons and also atoms from polymer molecule; the induced photo-thermal heat was transferred into the polymer matrix to induce polymerization of the composite and the covalent bonding between nano-carbons and polymer matrix at the interface. Scanning electron microscope (SEM), Raman spectroscopy, and RSA were used to evaluate the load transfer and mechanical strength of the polymerized composite samples. Investigating first the nanotube/polymer composites synergized by NIR photon-assisted polymerization, large Raman shifts (20 cm-1 wavenumber for up to 80% strains) of the 2D band were recorded for the NIR light polymerized samples and an increase in Young‘s modulus by ~130% was measured for the 1 wt. MWNT/poly(dimethylsiloxane) (PDMS) composites. While at first it was thought that NIR radiation during polymerization heated the nano-carbons inside resulting in strengthening of the nano-carbon/polymer interface, it was seen after further experimentation with graphene reinforcements that other light induced bonding effects apart from heat were also responsible. Raman spectroscopy revealed that mixing graphene in polymer has a profound effect on the G, D and 2D bands. Investigating G bands for pure RGO and GNPs and comparing them with their polymer counterparts showed large shifts in the G band indicating lattice compression. The comparison of the NIR polymerization with the conventional oven based polymerization for both RGO and GNPs revealed large changes in wavenumbers and indicated increased load transfers for the NIR photon-assisted polymerization method. The Full Width Half Maximum (FWHM) data of the NIR treated samples exhibited smaller change at large strains compared to conventionally polymerized samples indicating the minimum slippage in the former. The role of occlusion on Tempormandibular Disorders (TMD) is still unclear, mainly regarding muscular function, therefore non invasive and reversible treatment, as lasertherapy, may be a valuable instrument in this function. The aim of this study was to evaluate the lasertherapy action on masseter muscle submitted to occlusal alteration. Forty Wistar rats were randomly divided in four groups: 10 for control group, 10 for occlusal alteration group (CCO), 10 for CCO + lasertherapy group (l830nm, 4J/cm2) and 10 for CCO + muscle relaxant group (2,5 mg/Kg Dantrolene). Rats underwent unilateral amputation of the left inferior and superior molar cusps to simulate an occlusal wear situation. The rats of control group had no occlusal wear. Half of the animals of each group was sacrificed in 14 days after the occlusal consuming and half 30 days after the occlusal consuming. The masseter muscles ipsilateral to the amputated molars were excised and processed for light microscopy, electron microscopy, and evaluation of calcium phosphate precipitate by Ramam spectroscopy. The light microscopy did not show differences between the groups. The electron microscopy was able to detect a degree of intracellular damage in muscle fibers of CCO group: swollen mitochondria with disrupted cristae and cleared matrix, signs of hypercontraction of I bands and myofibril disorganization. A decrease in Raman intensity for calcium phosphate precipitate was detected in 30 days laser group. Based on the results, there were alterations in muscular fibers of ipsilateral masseter with occlusal unilateral wear in Wistar rats, although the lasertherapy and pharmacotherapy played few action on treatment of masseter muscle in the studied conditions.

A ação da oclusão sobre as Disfunções Temporomandibulares (DTM) ainda é indefinida, principalmente em relação à função muscular. Por isso, terapêuticas não invasivas e reversíveis, como o laser, são terapias válidas e aceitas na literatura. O objetivo desse estudo é avaliar possíveis alterações no músculo masseter de ratos submetidos à laserterapia após alteração oclusal. Foram utilizados 40 ratos Wistar, divididos aleatoriamente em quatro grupos, sendo 10 para o grupo controle, 10 para o grupo Desgaste Oclusal (DE), 10 para o grupo DE + Laserterapia (l830nm) com dose de 4J/cm2 e 10 para o grupo DE + Relaxante Muscular (Dantrolene 2,5 mg/Kg). As cúspides superiores e inferiores dos molares esquerdos dos ratos foram desgastadas com broca diamantada sob refrigeração de solução salina. Os ratos do grupo controle não tiveram desgaste oclusal. Metade dos ratos de cada grupo foi sacrificada aos 14 dias e a outra metade 30 dias após o desgaste oclusal. Após o período experimental, o músculo masseter foi removido do lado ipsilateral ao desgaste oclusal em cada animal e, então, processado para microscopia de luz, microscopia eletrônica de transmissão e mensuração da intensidade Raman para o fosfato de cálcio precipitado nas fibras musculares. This study evaluated the effect of prolonged exposure times on immediate (IM) and 6-month (6M) adhesive properties: degree of conversion within hybrid layer (DC), silver nitrate uptake (SNU) and resin-dentin bond strenght (μTBS) for three one-step self-etch adhesive systems (Adper Easy Bond [EB], Clearfil S3 Bond [CS3] and Go [GO]).Ninety caries-free extracted molars were included in this study being 45 for μTBS and SNU (n=5) and 45 for DC (n=5). The dentin surfaces were bonded according to the manufacturers instructions, but they were light-cured for 10, 20, and 40 s at 600mW/cm2. Bonded teeth for the μTBS test were sectioned in order to obtain stick-like specimens (0.8mm2) and tested under tensile stress 0.5mm/min). Two bonded sticks from each tooth were analyzed by scanning electron microscopy for SNU. The DC within the hybrid layer were evaluated with micro-Raman spectroscopy. All tests were performed IM and at 6M. Data were analyzed by a threeway repeated measures ANOVA and Tukeys test (alpha=0.05).The prolonged exposure times increased the DC () [10s- 67.319.5; 20s- 84.6 12.3 e 40s- 82.9 13.1] for all adhesives and decreased the SNU ()[10s- 24.8 13.2; 20s- 13.3 7.5 e 40s- 13.5 9.3] significantly for all adhesives; however significant improvements in the μTBS (MPa) [10s- 17.3 3.7; 20s- 31.9 4.9 e 40s- 25.0 3.7] and stable bond strengths after water storage [IM- 25.2 7.3; 6M- 24.2 7.2] were observed only for CS3. EB [IM- 35.9 4.7; 6M- 28.0 6.0] and GO [IM- 25.6 5.0; 6M- 14,3 3,7] decreased the μTBS after 6 months. The prolonged exposure times improved the DC within the hybrid layer and reduced the SNU for all adhesives, however this did not prevent the degradation of the adhesive interface.

Este estudo avaliou o efeito dafotoativação prolongadaem tempo imediato (IM) e após 6 meses de armazenamento em água (6M) nas propriedades adesivas: grau de conversão no interior da camada híbrida (GC), nanoinfiltração (NI) e na resistência de união (RU) à dentina de três sistemas adesivos autocondicionantes simplificados (AdperEasyOne [EO], Clearfil S3 Bond [CS3] e Go [GO]).As superfícies dentinárias de 90 molares humanos foram expostas e os adesivos aplicados conforme recomendações dos fabricantes, porém a fotoativação realizada nos tempos10, 20 e 40 s a 600 mW/cm2 depois restaurados com resina composta. Para RU e NI, os dentes restaurados foram seccionados para obtenção de corpos-deprova (cps) na forma de palitos (0,8 mm2) e testados por microtração a 0,5 mm/min. O objetivo deste estudo foi avaliar os métodos de espectroscopia Raman e espectroscopia de absorção de infravermelho (FTIR) na identificação da degradação do trans-1,4-poliisopreno de cones de guta-percha sem uso, e o comportamento do material após diferentes períodos decorridos de sua fabricação por testes de indentação de nanodureza (IHT). Para a análise, foram selecionadas 27 amostras de cones de guta-percha sem uso, de 5 marcas comerciais vendidas no mercado brasileiro, cedidas por Cirurgiões-dentistas. As amostras foram avaliadas através de espectroscopia Raman, com base no sinal da ligação C=C, indicando a presença de trans-1,4-poliisopreno; FTIR, levando-se em consideração a ligação C=O, indicativa de oxidação do material; IHT, avaliando-se rigidez e módulo de elasticidade. Os dados foram analisados de maneira descritiva. Observam-se, nos espectros de Raman, amostras nas quais os picos relativos à ligação C=C foram identificados, bem como outras em que não foi percebido o referido pico, sem uma relação direta com os tempos decorridos da fabricação dos cones de guta-percha sem uso. Em relação ao FTIR, em todas as amostras analisadas foram observados picos relativos à ligação C=O, indicando haver algum grau de oxidação no material. Os testes de nanodureza, realizados em cones sem uso, mostraram não haver homogeneidade dos valores de dureza e módulo de elasticidade. A análise pelos métodos de espectroscopia Raman e FTIR em cones de guta-percha é dificultada pelos vários componentes presentes no material e a separação do polímero trans-1,4-poliisopreno pode evitar número excessivo de picos e facilitar a interpretação dos espectros. Espectroscopia Raman e FTIR são capazes de identificar a degradação do poliisopreno. A presença, ou não, da ligação C=C, nos espectros de Raman, não teve relação direta com a idade dos cones. Os cones de guta-percha sem uso apresentaram algum grau de degradação, devido à presença da oxigênio no polímero. As análises de nanodureza não foram conclusivas quanto ao comportamento do material relacionado ao envelhecimento.

The aim of the present study was to evaluate the methods Raman spectroscopy and infrared absorption spectroscopy (FTIR) to analyze the degradation of trans-1,4-polyisoprene polymer in gutta-percha points, before use. Mechanical properties were assessed through the instrumented hardness tests (IHT). Twenty-seven samples of gutta-percha cones collected before use, belonging to five different commercial brands sold in Brazil, were obtained from Dental Surgeons. The samples were evaluated by Raman spectroscopy, based on the sign of the C=C bond (representing the trans-1,4-polyisoprene polymer); FTIR, considering the C=O bond (indicating polymer oxidation), and instrumented hardness tests (IHT), to assess both rigidity and elasticity modulus. Resumo: O objetivo deste estudo foi avaliar a resistência de união e grau de conversão de um compósito a base de silorano em associação a sistemas adesivos autocondicionantes e convencionais. Para a realização do teste de microtração, foram utilizados 50 terceiros molares humanos sem raiz e sem esmalte oclusal e com a superfície dentinária exposta. As amostras foram submetidas ao desgaste da superfície dentinária com lixas de carbeto de silício de granulação 600 para a padronização da lama dentinária e divididas aleatoriamente em cinco grupos de acordo com o sistema adesivo aplicado (n=10): 3 sistemas autocondicionantes G1 - P90; G2 - Adper Easy One Bond; G3 - Adper SE Bond; e dois convencionais G4 - Adper Single Bond 2; G5 - Adper Scotchbond Multi-Uso Plus. Em seguida, foram aplicados os sistemas adesivos e a resina composta a base de silorano de acordo com as instruções do fabricante. Após o procedimento restaurador, as amostras foram seccionadas, obtendo-se palitos com largura de 0,7 - 1mm. Foram selecionados aleatoriamente 5 palitos de cada dente para a realização do teste de microtração e fraturados em Máquina de Ensaio Universal. Os valores obtidos em KgF foram convertidos para MPa. Para a avaliação do grau de conversão, através da espectroscopia FT-Raman, foram confeccionados 25 amostras em uma matriz metálica com orifício circular com 2mm de diâmetro e 1mm de altura. Os grupos utilizados neste teste foram os mesmos daqueles usados no teste de microtração. Os sistemas adesivos foram aplicados em uma tira de poliéster, fotoativado e posicionados sobre a resina composta a base de silorano. O conjunto foi fotoativado e seus espectros analisados através da espectroscopia FT-Raman. Os dados foram estatisticamente analisados por meio de análise de variância (ANOVA) one way e teste de Tukey com nível de significância de 0,05 (p?0,05). O sistema adesivo Adper Scotchbond Multi-Uso Plus apresentou os maiores valores de resistência de união (26,07 MPa), porém sem diferença estatística para os sistemas adesivos Adper Single Bond 2 (23,24 MPa) e P90 (20,83 MPa). O sistema adesivo Adper SE Bond apresentou o menor valor de resistência de união (9,65 MPa), não diferindo estatisticamente do sistema Adper Easy One Bond (13,73 MPa), porém com diferença em relação ao sistema adesivo P90. Com relação ao grau de conversão, a resina composta da face de contato com os sistemas adesivos Adper Scotchbond Multi-Uso Plus e o Adper Single Bond 2 apresentaram maiores valores, porém sem diferença significante entre si. Thin ceramic laminates have been widely requested by patients because it’s a reliable conservative aesthetic procedure, stable and presents adequate longevity. It is known that the color change of the cement below these thin restorations may result inappropriate final color. Thus, the purpose of this in vitro study was to evaluate the color agreement between pressable lithium disilicate ceramics with tryin pastes and corresponding resin cements, determination of the degree of conversion of resin cements using FT-Raman spectroscopic and color stability after artificial accelerated aging (AAA) for 300 hours. Eighty-four ceramic discs (0.3 mm, 0.5 mm, 1.0 mm thick) were prepared (IPS e.maxPress HT A2 – Ivoclar Vivadent) to simulate veneers. Evaluation the baseline color was done in the CIE-L*a*b* color space with reflectance spectrophotometer (control group). The try-in pastes were applied into the interlayer of the ceramic (simulation of selection color procedure) above silicon transparent matrix (0.1 mm thick) and the color complex were measured using a spectrophotometer (n=7), with no substrate. The same ceramic discs were used for cementation procedure with Variolink II (only base),after storage in distilled water, protected from light, at 37ºC for 24 hours, the specimens were mensured the color and submitted to FT-Raman spectroscopy to determinate the degree of conversion (DC). After that, the specimens were AAA for 300 hours and measured by spectrophotometer to calculate ΔE. Two-way ANOVA was used to analyze the data. Tukey’s post hoc for multiple comparisons and Spearman correlation test were performed (α = 0.05). There was no agreement between the try-in with resin cements, L* and b* values increased, increasing the value/opacity,yellowish of the complex. All groups exhibited values of ΔE>3.3, except resin color A3 group (1.0 mm ceramic thickness). Ceramic thicknesses (0.3 mm and 0.5 mm) did not affect the DC compared to the control (p> 0.05). Thus, DC was influenced by the color of cement, where the over saturated color showed higher conversion rate, DC decreased when ceramic disc thickness increased. No correlation between DC and ΔE was observed.

The aim of the present study was evaluate the adhesive properties of eroded dentin with different pH cycling using universal adhesive systems and to evaluate the application of collagen cross-linking agent or MMP inhibitors on the resin-eroded dentin interface, through of three in vitro studies. For the experiment 1, the teeth were randomly assigned into the experimental groups according to the combination of variables: dentin surface (sound dentin and eroded dentin) vs adhesive systems (10 universal adhesives) vs Adhesive strategy [etch-and-rinse (ER) and self-etch (SE)]. The adhesive systems were applied according to the manufacturer's instructions and the teeth were restored. They were sectioned to analysis of microtensile bond strength (μTBS) and nanoleakage (NL). The surfaces of 2 tooth were examined under scanning electron microscopy (SEM) to evaluate the surface characterization after erosive-cycling. For experiment 2, the teeth were randomly assigned into 18 experimental conditions according to the variables: Erosive cycling (eroded dentin with soft drink pH ≈ 2,7 and eroded dentin with citric acid pH 2.3) vs. adhesive systems [Prime & Bond Elect (PBE); Scotchbond Universal Adhesive (SBU); Tetric N-Bond Universal (TEU)] vs Treatment agents [6.5% primer containing proanthocyanidin (PA) and 0.1% riboflavin (RI)]. The surface was treated with primers and the adhesive systems were applied and restored. The samples were sectioned and tested for μTBS, NL and degree of conversion (DC). The surfaces of 2 tooth were examined under SEM to evaluate the surface characterization after erosive-cycling. For experiment 3, the teeth were randomly assigned into 18 experimental conditions according to the variables: Erosive cycling (eroded dentin with soft drink pH ≈ 2,7 and eroded eroded with citric acid pH 2.3) vs. Adhesive Systems (PBE; SBU; TEU) vs Acid pretreatment [control with 37% phosphoric acid;Phosphoric acid containing 2% chlorhexidine (CHXacid) and phosphoric acid containing 3% PA (PAacid)]. The teeth were acid-etched according to each experimental conditions, the adhesive systems were applied and the teeth were restored. The teeth were sectioned and tested for μTBS, NL and DC. Data were submitted to appropriate statistical analysis (α = 0.05). The μTBS values in eroded dentin were statistically lower than sound dentin, independent of the adhesive strategy (p < 0.01) The inclusion of primers containing collagen cross-linking agents in the eroded dentin adhesive protocol improved μTBS values (p <0.001),significantly reduced NL (p <0.004) for all the adhesives tested, as well as improved DC (p <0.03). This study evaluated the effect of prolonged exposure times on immediate (IM) and 6-month (6M) adhesive properties: degree of conversion within hybrid layer (DC), silver nitrate uptake (SNU) and resin-dentin bond strenght (μTBS) for three one-step self-etch adhesive systems (Adper Easy Bond [EB], Clearfil S3 Bond [CS3] and Go [GO]).Ninety caries-free extracted molars were included in this study being 45 for μTBS and SNU (n=5) and 45 for DC (n=5). The dentin surfaces were bonded according to the manufacturer’s instructions, but they were light-cured for 10, 20, and 40 s at 600mW/cm2. Bonded teeth for the μTBS test were sectioned in order to obtain stick-like specimens (0.8mm2) and tested under tensile stress 0.5mm/min). Two bonded sticks from each tooth were analyzed by scanning electron microscopy for SNU. The DC within the hybrid layer were evaluated with micro-Raman spectroscopy. All tests were performed IM and at 6M. Data were analyzed by a threeway repeated measures ANOVA and Tukey´s test (alpha=0.05).The prolonged exposure times increased the DC () [10s- 67.3±19.5; 20s- 84.6 ± 12.3 e 40s- 82.9 ±13.1] for all adhesives and decreased the SNU ()[10s- 24.8 ± 13.2; 20s- 13.3 ± 7.5 e 40s- 13.5 ± 9.3] significantly for all adhesives; however significant improvements in the μTBS (MPa) [10s- 17.3 ± 3.7; 20s- 31.9 ± 4.9 e 40s- 25.0 ± 3.7] and stable bond strengths after water storage [IM- 25.2 ± 7.3; 6M- 24.2 ± 7.2] were observed only for CS3. EB [IM- 35.9 ± 4.7; 6M- 28.0 ± 6.0] and GO [IM- 25.6 ± 5.0; 6M- 14,3 ± 3,7] decreased the μTBS after 6 months. The prolonged exposure times improved the DC within the hybrid layer and reduced the SNU for all adhesives, however this did not prevent the degradation of the adhesive interface.

Este estudo avaliou o efeito dafotoativação prolongadaem tempo imediato (IM) e após 6 meses de armazenamento em água (6M) nas propriedades adesivas: grau de conversão no interior da camada híbrida (GC), nanoinfiltração (NI) e na resistência de união (RU) à dentina de três sistemas adesivos autocondicionantes simplificados (AdperEasyOne [EO], Clearfil S3 Bond [CS3] e Go [GO]).As superfícies dentinárias de 90 molares humanos foram expostas e os adesivos aplicados conforme recomendações dos fabricantes, porém a fotoativação realizada nos tempos10, 20 e 40 s a 600 mW/cm2 depois restaurados com resina composta. ▼ Traditionally, environmental pollutants are sampled in the field and are subsequently analysed via laboratory-based analytical instruments. This is a time-consuming process that delays the identification of potentially hazardous materials that may be detrimental to the health of investigators, the general public and the environment. It also means that, currently, no or very limited information about a material is available to field-investigators. It was, therefore, necessary to develop new intelligence-led forensic protocols, whereby, materials are immediately identified in situ (at least in a preliminary fashion). This project aimed to determine if field-portable Raman and FTIR spectrometers were a viable option for the rapid in situ identification of pollutants, and if so, to what extent they could be applied. To achieve this, each portable spectrometer was evaluated and optimised to analyse a range of materials that are encountered at pollution incidents. For the Raman spectrometer, a number of parameters that affect the quality of a generated spectrum could be altered. Whilst a universally applicable set of optimal Raman operating parameters does not exist, a recommended starting point for analysis was established. When employed, this parameter set produces the highest-quality spectra for most samples. The findings of this project also resulted in the development of a ‘Standard Operating Procedure’ (SOP) for each portable instrument. The SOPs will provide investigators with an intelligence-led protocol for the rapid identification of pollutants at an incident scene. The SOPs can also improve on-site sampling practices and help direct any subsequent laboratory analyses. The employment of these portable spectrometers will also enable the most appropriate site clean-up method to be chosen and implemented in a timelier, more cost-efficient manner. Ultimately, it is hoped that the results described in this study will provide a strong basis for future research on portable Raman and FTIR spectrometers, and their applicability at environmental pollution incidents in Australia and beyond. ▼ Biological membrane structure is inherently complex, and understanding the structure of peptide chains inserted in membranes is vitally important for the development of effective diagnosis and treatments of diseases. Surface Enhanced Raman Spectroscopy (SERS) was used as a tool for studying lipid membrane structure in a natural fluid, room temperature environment. Gold nanostructures focus light to a molecular length scale at their surface, creating the possibility to visualize molecular structure. Optical excitation of gold nanoparticles at their size and shape-dependent plasmon resonant frequency induces strong oscillations of the nanoparticle’s free electron gas, leading to SERS – an enhancement of Raman scattering signals in a distance dependent manner at the molecular scale. This project utilizes gold nanorods, tuned to an excitation laser wavelength of 785 nm, as a substrate for lipid membranes so that their structure can be analyzed by SERS. The surfactant cetyltrimethylammonium bromide (CTAB) that stabilizes nanorods was exchanged with the biologically relevant lipid dioleoylphosphatidylcholine (DOPC), and the insertion of the peptide residue tryptophan into the lipid membrane was observed using SERS for confirmation. Full characterization of the lipid membranes was carried out, demonstrating a well ordered, gold supported bilayer. SERS spectra also contain information on molecular position and orientation relative to the surface, but are difficult to interpret quantitatively. An analysis method that combines SERS and unenhanced Raman spectra with theoretical calculations of the optical field and molecular polarizability is introduced. Together these reveal the molecular orientation and position of surfactant layers on gold nanorods and of tryptophan in phospholipid bilayers. This method offers a new approach to analyzing lipid membrane molecular structure under ambient conditions, with microscopic quantities, and without molecular labels. Este trabalho apresenta uma técnica para o desenvolvimento de um substrato sólido para a realização de espectroscopia Raman, aliada ao efeito SERS (Espalhamento Raman Intensificado pela Superfície), utilizando como analito o corante rodamina 6G. Ao longo do trabalho foi apresentada uma metodologia de síntese de nanopartículas de prata, estas utilizadas com o objetivo de amplificar o sinal do espalhamento Raman através do fenômeno conhecido como SERS, no qual nanopartículas metálicas em contato com o analito de prova causam uma intensificação do campo eletromagnético. Além disso foram estudados diversos parâmetros envolvidos no processo de análise em substrato sólido, como a membrana filtrante, as objetivas do microscópio, o processo de secagem, o tempo de integração, o pH da solução coloidal e a concentração de analito, a fim de se desenvolver uma metodologia eficiente. Para essas análises, além da espectroscopia SERS em substrato sólido, foram utilizadas técnicas de espectroscopia ultravioleta-visível e SERS em substrato líquido. Os resultados mostram que os espectros SERS obtidos em substrato sólido apresentaram intensificações em torno de 15x maiores que as intensificações obtidas em substrato líquido para uma mesma amostra. Além dos testes envolvendo a rodamina 6G como analito de prova foram também realizados testes com o herbicida glifosato. Para ambos analitos, as relações sinal/ruído nos espectros foram de 11,2 para 0,0021 mg/L de rodamina 6G e 3,8 para 3 mg/L de glifosato, evidenciando que tais concentrações estão acima do limite de detecção da técnica (relação sinal/ruído maior que 3) e podem ser efetivamente mensuradas.

This work presents a technique for the development of a solid substrate for Raman spectroscopy, coupled with the SERS (Surface Enhanced Raman Scattering) effect, using rhodamine 6G as a probe analyte. Throughout the work, a silver nanoparticles synthesis methodology was presented, which are used to amplify the Raman scattering signal through the phenomenon known as SERS, in which metallic nanoparticles in contact with the analyte cause an intensification of the electromagnetic field. Besides, several parameters involved in the solid substrate analysis process were studied, such as the filtering membrane, the microscope objectives, the drying process, the integration time, the colloidal solution pH and the analyte concentration, in order to develop an efficient methodology. For these analysis, besides the solid substrate SERS spectroscopy, ultraviolet-visible spectroscopy and liquid substrate SERS techniques were used. The results show that the SERS spectra obtained in solid substrate presented enhancements about 15x higher than the enhancements obtained in liquid substrate for the same sample. In addition to tests involving rhodamine 6G as a probe analyte, tests were also performed with glyphosate herbicide. ▼ An automatic classification of chemical compounds by computer processing of digitized spectral data is presented. The classification system is based on a branch of artificial intelligence known as supervised learning, and used binary linear classifiers to identify compounds as alcohols, esters, ethers, ketones or compounds containing double bonds. Each of the 1117 spectra in volume one of Sadtler's Standard Raman Spectra was coded using a scale from 0 to 9 in the range from 4000 to 200 cm^-1. One hundred and twelve readings were taken on each spectrum. These data were then examined using pattern recognition techniques, and several methods of combining infrared and Raman data from the dame compound were tested. The classification techniques were most successful when applied to concatenated infrared and Raman data. By taking the infrared data in the range from 500 to 1900 cm^-1 and concatenating them with Raman data from the same range, a vector representing each of the L.00 compounds in the data set was obtained. .The parallel polarized Raman was used in preference to the perpendicularly polarized spectrum when both were available, otherwise the nonpolarized spectrum was used. Using an iterative pattern recognition technique a vector was then calculated which would recognize compounds as members of a class or not members based on the sign of the dot product of the calculated vector and the vector representing the compound. When vectors were calculated using only half the data set, then tested for their ability to correctly classify the remaining compounds; it was found that they could correctly classify compounds more than 90% of the time. Vectors which were trained using the entire data set were helpful in determining characteristic group frequencies and each class treated is discussed. Several compounds for which observed frequencies have been assigned in the literature were tested to determine if the assignments could be supported by the trained vectors.
