Les graphes définis à partir de structures algébriques possèdent d’excellentes propriétés de symétries particulièrement intéressantes. L’exemple le plus flagrant est la notion de graphe de Cayley qui s’est révélée très riche non seulement du point de vue théorique mais aussi pratique par ses applications à de nombreux domaines incluant l’architecture des réseaux ou les machines parallèles. Néanmoins, la régularité des graphes de Cayley se révèle parfois être une limite étant donné qu’ils sont toujours sommet-transitifs et donc en particulier non pertinents pour générer des réseaux semiréguliers.Cette observation a motivé, en 2005, la définition d’une nouvelle classe de graphes définis à partir d’un groupe, appelés G-graphes. Ils possèdent aussi de nombreuses propriétés de régularité mais de manière moins restrictive.Cette thèse propose un nouveau regard sur cette classe de graphes par une approche plutôt orientée recherche opérationnelle alors que la grande majorité des études précédentes est dominée par des approches essentiellement algébriques. Nous-nous sommes alors intéressés à plusieurs questions :— La caractérisation des G-graphes : nous proposons des améliorations par rapport aux précédents résultats.— Identifier des classes de graphes comme des G-graphes grâce à des isomorphismes ou en utilisant le théorème de caractérisation.— Etudier la structure et les propriétés de ces graphes, en particulier pour de possibles applications aux réseaux : colorations semi-régulières, symétries et robustesse.— Une approche algorithmique pour la reconnaissance de cette classe avec notamment un premier exemple de cas polynomial lorsque le groupe est abélien.

Interactions between graph theory and group theory have already led to interesting results for both domains. Graphs defined from algebraic groups have highly symmetrical structure giving birth to interesting properties. The most famous example is Cayley graphs, which revealed to be particularly interesting both from a theoretical and a practical point of view due to their applications in several domains including network architecture or parallel machines. Nevertheless, the regularity of Cayley graphs is also a limit as they are always vertex-transitive and therefore not relevant to generate semi-regular networks. This observation motivated the definition, in 2005, of a new family of graphs defined from a group, called G-graphs. They also have many regular properties but are less restrictive. These graphs are in particular semi-regular k-partite, with a chromatic number k directly given in the group representation and they can be either transitive or not.This thesis proposes a new insight into this class of graphs using an approach based on operational research while most of previous studies have been so far dominated by algebraic approaches. ▼ Expansion of gene-specific tandem repeat sequences is the causative mutation of a growing list of neurological, neuromuscular and neurodegenerative diseases, of which there are currently over 40. The cause of the initial tandem repeat expansion mutations, the molecular mechanisms driving ongoing repeat tract instability and the causes of pathogenesis are not well understood for many of these diseases. The formation of alternative secondary nucleic acid structures in the DNA and RNA of expanded tandem repeat sequences is thought to drive repeat instability and pathogenesis by impairing normal DNA and RNA metabolic processes. My thesis involves characterization of three such alternative secondary structures that could potentially form in the DNA or RNA of disease-associated tandem repeat sequences and assessment of their processing and binding to candidate structure-specific proteins. In DNA, I assessed how the specific configuration of slipped-out 3-way junction structures formed in disease-associated (CAG)*(CTG) tandem repeats can influence protein binding and cleavage by structure-specific DNA repair proteins in vitro. Whereas previous studies focused upon slipped-DNA structures as a static entity, we show that slipped-DNA junctions exist in multiple interchanging conformations and the specific conformation of the junction can influence how it gets processed. These findings support that the junction conformation is an important modifier of disease-associated (CAG)*(CTG) instability. To further extend our understanding of nucleic acid structure in tandem repeat disease, I analyzed the structural and sequence determinants governing RNA:DNA hybrid formation and processing at various trinucleotide repeats. By using a well-established in vitro transcription assay, I demonstrated that stable R-loop formation occurs in all disease-associated trinucleotide repeats. I also identified novel double-R-loop structures formed when repeats are simultaneously bidirectionally transcribed, as occurs at many disease-associated trinucleotide repeat-containing genes. I went on to establish a novel R-loop processing assay using a cell-free extract system and demonstrated that R-loops and particularly double-R-loops can increase the instability of (CAG)*(CTG) repeats. Finally, I identified extremely stable RNA G-quadruplex structures in the amyotrophic lateral sclerosis and frontotemporal dementia (ALS-FTD)-associated (GGGGCC)n repeat in vitro, using RNA oligonucleotide models and assessed their ability to bind structure-specific RNA binding proteins. This led to the identification of the ASF/SF2 essential splicing factor as a candidate protein interactor that may have relevance to ALS-FTD. Taken together, my findings demonstrate unusual nucleic acid structure formation by disease-associated tandem repeats in the DNA, the RNA and during transcription when the DNA interacts with the RNA. ▼ This portfolio of exploration explores the role of transformative thinking and practice in a property entrepreneur’s response to the financial crisis which swept over Ireland from 2008. The complexity of this challenge and the mental capacity to meet its demands is at the core of the exploration. This inquiry emerged from the challenges the financial crisis presented to my values, beliefs, assumptions, and theories, i.e. the interpretive lens through which I make meaning of my experiences. Given the issue identified, this inquiry is grounded in aspects of theories of constructive developmental psychology, applied developmental science, and philosophy. Integrating and linking these elements to business practice is the applied element of the Portfolio. As the 2008 crisis unfolded I realised I was at the limits of my way of knowing. I came to understand that the underlying structure of a way of knowing is the ‘subject-object relationship’ i.e. what a way of knowing can reflect upon, look at, have perspective on, in other words, make object, as against what is it embedded in, attached to, identified within, or subject to. My goal became enhancing my awareness of how I made meaning and how new insights, which would transform a way of knowing, are created. The focus was on enhancing my practice. This Portfolio is structured into three essays. Essay One reported on my self-reflection and external evaluation out of which emerged my developmental goals. In Essay Two I undertake a reading for change programme in which different meaning making systems were confronted in order to challenge me as a meaning maker. Essay Three reported on my experiment which concerned the question whether it was possible for me as a property entrepreneur, and for others alike, to retain bank finance in the face of the overwhelming objective of the bank to deleverage their balance sheet of property loans. The output of my research can be grouped into General Developmental and Specific Business Implications. Firstly I address those who are interested in a transformational-based response to the challenges of operating in the property sector in Ireland during a crisis. I outline the apparatus of thought that I used to create insight, and thus transform how I thought, these are Awareness, Subject-object separation, Exploring other’s perspectives from the position of incompleteness, Dialectical thinking and Collingwood’s Questioning activity. Secondly I set out my learnings from the crisis and their impact on entrepreneurial behaviour and the business of property development. I identify ten key insights that have emerged from leading a property company through the crisis. Many of these are grounded in common sense, however, in my experience these were, to borrow Shakespeare’s words, “More honor'd in the breach than the observance” in pre Crisis Ireland. Finally I set out a four-step approach for forging a strategy. ▼ Leaf dimensions and pigments are the important traits in plants that play a key role in estimating light interception, absorption and food production. In predictive research, these parameters are a useful data source for devising crop management techniques such as cultivation, pruning and fertilisation. Destructive and non-destructive techniques are commonly used for estimating crop growth and nutrient status. Although, destructive methods are more accurate, these are expensive, laborious and impracticable for large fields. In contrast, various non-destructive techniques have been developed for predicting crop N requirements that are relatively fast and less expensive. However, lack of consistency in accurately predicting the true N levels of different crop species under variable environments require further exploration of this area. In the present study, a new and relatively more efficient technique has been proposed for measuring leaf dimensions, chlorophyll, and N and phosphorus (P) content. In the initial study, leaf images from a range of plant species were collected using a handheld portable digital scanner (Pico Life). Edge detection and filtering algorithms were applied to identify the leaf section of the image against the background. Data of forty leaves that vary in shape and size (from grasses to broad leaf plant species) were collected and processed using a new algorithm as well as the Li-Cor 3100. Data indicated high accuracy of the proposed algorithm for estimating leaf area, length, width and perimeter. It was verified by a strong correlation (R²=0.999) between leaf area measured by Li-Cor 3100 and by digital scanner. After successful application of the digital scanner for estimating leaf size and dimensions, the images collected by this scanner were used for predicting chlorophyll, P and N content of tomato, broccoli and lettuce leaves. The plants were grown under controlled conditions using nutrient solution and at early reproductive growth (after 8 weeks of growth) these were exposed to various N levels for seven weeks. Data on leaf chlorophyll and N content were collected through biochemical assays (LabChl). In addition, data were collected by the SPAD-502 and portable scanner. Images collected by the portable scanner were processed by averaging the R (red), G (green) and B (blue) values of all the leaf pixels. Based on the RGB values, a new algorithm was developed that estimates leaf chlorophyll content (ChlRGB). Despite slight variations in response to applied N levels in the three crops, the leaf chlorophyll and N content significantly increased with increasing N levels in nutrient solution in the studied crop species. Under N deficient conditions (N0), tomato and broccoli plants showed significantly lower leaf N content just 2 weeks after treatment (WAT), compared with N-treated plants (any N level) suggesting a rapid response of these crops to N deficiency. L’île déserte constitue un motif privilégié de la littérature d’aventures, une des marques de fabrique du genre en réalité, et il s’agit de comprendre l’intérêt littéraire qu’il revêt, en déterminant la spécificité de ce lieu et des œuvres qui s’y rattachent, afin de définir leurs codes et leurs motifs typiques ainsi que l’évolution de ces derniers dans l’histoire littéraire. Cette thèse s’intéresse en particulier à la mutation que traversent cet espace et ce genre au tournant du XIXe siècle, notamment chez Stevenson, Conrad et Wells. L’ambition de ce travail est ainsi de montrer que l’île est pour ces auteurs bien plus qu’un décor : elle constitue en outre une sorte de laboratoire littéraire, un lieu où s’élabore l’écriture utopique, encore à venir. En effet, bien que l’île déserte soit un espace littéraire surcodé et surdéterminé, il est également le lieu où tout semble possible tant en termes d’aventure que d’écriture : une sorte de brèche, hors de l’espace-temps, propice à la construction d’une autre réalité. Entre stabilité et flottement, utopie et réalité, l’île est à la fois un lieu d’ancrage, établi scientifiquement, mais aussi un lieu qui semble parfois flottant et indéterminé, qui apparaît et disparaît de la carte : un lieu de tension entre le réel et l’imaginaire, le moi et l’autre, le centre et la périphérie. Partant, cette thèse vise à évaluer dans quelle mesure la littérature d’aventures peut être déterminée par la spécificité de ce chronotope insulaire, et si réciproquement l’aventure donne également forme à l’île, créant de ce fait un sous-genre spécifique que nous appelons l’aventure insulaire. Il s’agit ainsi d’analyser l’impact de l’évolution de ce chronotope sur la poétique aventureuse de nos trois auteurs au tournant du siècle, par le biais d’une approche géocritique et géopoétique de leurs œuvres. Cette approche méthodologique originale a pour objet d’étudier les liens qui unissent espace et littérature, et permet ainsi d’élaborer une géographie littéraire et même une géopoétique de l’aventure insulaire, en montrant qu’il existe un certain isomorphisme entre espace insulaire et forme littéraire.

The desert island is one of the central motifs of the adventure novel, and the objective of this doctoral thesis is to understand why it is so crucial to the definition of this genre, by determining the specificity of this place and of the works that resort to it, in order to define their particular codes and motifs, as well as their evolution throughout literary history. It focuses in particular on the mutation undergone by this genre and this space at the turn of the 19th century, especially in the works of Stevenson, Conrad and Wells. It aims to show that the island is much more than a simple setting: that it actually constitutes a literary laboratory, where a utopic form of writing can be developed. Cette thèse se propose d’étudier le rapport entre fiction et raisonnement dans l’œuvre édouardienne (1901-1910) de l’écrivain, journaliste et polémiste anglais G.K. Chesterton (1874-1936). S’inscrivant dans le double cadre de la théorie de la fiction et de l’histoire des idées, ce travail repose sur l’hypothèse d’un usage singulier de la fiction dans la pensée chestertonienne, qui l’écarte de sa définition générique (comme d’un récit en prose, principalement destiné au divertissement) pour en faire un mode de raisonnement. Utilisée comme figure rhétorique, elle est également pensée comme un véhicule cognitif permettant de repousser la frontière du savoir, un moyen de compréhension du monde et un instrument de modélisation de l’idée, qui ne contredit pas le raisonnement mais le complète. La seconde hypothèse concerne le rôle central du contexte historique dans cet usage paradoxal de la fiction : le monde intellectuel britannique traverse, à la fin du XIXe siècle une crise épistémologique tandis que l’époque se caractérise progressivement par un âge d’or de la fiction de masse. L’ère édouardienne, située à la croisée de ces mouvements, apparaît donc comme un moment pivot. L’étude replace d’abord la formation intellectuelle et la naissance de la carrière de Chesterton dans le contexte de la vie littéraire de l’époque, afin de proposer une théorie de sa fiction d’examiner sa pratique dans ses articles, ses essais, ses études littéraires. Ses romances et ses récits de détection sont également envisagés puisque ces genres de la littérature de masse permettent à Chesterton de donner corps à sa pensée et de toucher un large public. En définissant des emplois originaux de la fiction, il s’agit d’examiner comment ils s’inscrivent dans une renégociation plus générale du rapport entre savoir et fiction à l’aube du XXe siècle.

This thesis explores the tension between fiction and reasoning in the Edwardian oeuvre of writer, journalist and polemicist G. K. Chesterton (1874-1936). Within the framework of fiction theory and the history of ideas, it is hypothesized that Chesterton diverts fiction from its most frequent use as a genre (a narrative in prose aiming at entertainment) to turn it into a mode. Used as a rhetorical figure, fiction is also devised as a cognitive vehicle that pushes back the frontiers of knowledge, a way of understanding the world and a conceptual modelling tool, which does not contradict but completes his reasoning. The second hypothesis of this work rests on the defining influence of the historical background on Chesterton’s imaginative use of fiction: the Edwardian intellectual world goes through an epistemological crisis while the ever-thickening stream of popular fiction turns the era into a Golden Age of storytelling. The era thus appears as a pivotal moment. Avec les récentes réformes, dont la loi Hôpital, Patient, Santé et Territoires (HPST) de Juin 2009, l’hôpital s’est transformé non seulement dans sa forme organisationnelle par la mise en oeuvre des pôles mais aussi dans son rapport à son environnement. Cette organisation polaire vise à rechercher une performance par une approche médico-économique. Ainsi les dispositifs de gestion ont été mis en place et cherchent à rendre compte de cette performance. Dans ce contexte, comment peuvent travailler des professionnels de champs aussi différents que celui de la gestion et celui du soin ? La gestion sous-entend une optimisation des moyens, alors que le service public défend en priorité des valeurs d’utilité sociale et d’attention aux usagers. Les acteurs du soin ont une forte identification professionnelle à cette mission d’utilité sociale. Dans ce travail, nous proposons d’identifier en quoi les décalages culturels au sein des pôles hospitaliers, participent à une perte de sens et de reconnaissance des agents à leur travail. Ces écarts culturels seront appréciés plus précisément au travers de l’appropriation des outils de gestion. Cette étude se base sur des entretiens réalisés au sein de différents hôpitaux (CHU et CH) et de secteur Médecine, Chirurgie, Obstétrique (MCO) et psychiatrie. Elle révèle lanécessité de pratiques managériales innovantes tournées notamment vers la gestion interculturelle afin que chaque acteur de ces nouvelles structures hospitalières se sente compris et intégré dans cette dynamique. L’étude proposée peut constituer un retour d’expérience intéressant pour mieux comprendre et améliorer l’utilisation des nouveaux outils de gestion. Des préconisations sont proposées et peuvent constituer des pistes de réflexion afin de répondre aux défis actuels de l’hôpital public.

With the latest reforms, among which the Hospital, Patient, Healthcare and Territory (HPST) Law, the French Hospital has changed not only the form of its organisation through the implementation of a polar organisation, but also its environmental concern. This polar organisation aims to strive for performance and efficiency via an managerial and medical approach. Thus, some management tools have been set up aimed in particular at giving feedback on such performance. In this context, how can professionals from such distinct fields as management and healthcare work? Management implies the optimization of resources when the public sector above all upholds values of social usefulness and patient care. Healthcare actors feel very much involved in this mission of social usefulness. In this work we propose to identify how cultural gaps within hospital poles contribute to a loss of meaning and acknowledgment among healthcare agents. Those cultural gaps will be examined more particularly through management tool appropriation. ▼ On 1 January 1901, the first day of the twentieth century, the six colonies of Australia federated in Sydney to become a single united country. On 9 May of the same year this symbolic inauguration became a practical reality when the first Federal Parliament was opened in the Melbourne Exhibition Building. Both these significant events were accompanied by equally significant processions through the two respective colonial, then state capital cities. Much has been written about the kind of country Australia was at the time of Federation and what manner of nation Australia consciously and subconsciously intended to become. However, our appreciation and understanding of these issues today tends to be determined, as do most historical studies, by an analysis of the written documents of the day, and subsequent written academic commentary. I would contend that such texts have, to some degree, always been restricted by the social and cultural limitations of the written documentary process, of those writing the work, and even the social and cultural limitations of those for whom such work is intended. Is it possible therefore, to analyse historical events by utilising other methodologies; other research materials and other approaches in collating evidence? And will such processes provide new insights, or simply re-iterate what has gone before? To this end I have adapted the techniques of Richard Holmes’ footstepping and Gilbert Ryle’s and Clifford Geertz’s thick description to undertake such an evaluation by retracing the routes of these two principal Federation parades and recording what additional evidence could be gained. This methodology has comprised a visual description and interpretation of the routes of the two Federation processions through the cities by referencing back to both the written record of the day through the press, and general historical commentary on these cities’ culture and society of the time. The primarily visual aspect of my analysis has been supplemented by a visual photographic documentation that provides both an historical record of the sites and locations along the parade routes as well as an added source of interpretation of the Federation process. Through this process, I have analysed the concept of national identity and the imagined Australian community as it was expressed at this specific period of Australia’s history. Finally, I have evaluated this methodology itself to determine the degree to which these techniques are a valid means of translating to the wider sphere of general historical research. Are footstepping and thick description effective processes for researching other previously inaccessible sources of historical evidence?

A presente dissertação tem como objetivo estabelecer um diálogo entre três obras da literatura britânica do século XIX: o romance Frankenstein (1818), da autora Mary W. Shelley; a novela O Médico e o Monstro (1886), de autoria de Robert Louis Stevenson; e o romance A Ilha do Dr. Moreau (1896), de H. G. Wells. Tal comparação será feita com base nas convenções advindas dos gêneros Gótico e Ficção científica, presentes nas obras. Como principal alicerce teórico para a definição de gêneros entendem-se as considerações de Tzvetan Todorov, que defende que os gêneros são inevitáveis como horizonte de interpretação, além de serem entidades em constante mudança numa cadeia de influências através da qual novos gêneros são criados a partir de outros pré-existentes. O presente trabalho parte desse pressuposto para determinar de que maneira os gêneros Gótico e Ficção científica estão presentes nas obras, observando como os traços do Gótico, ao se adaptarem através do tempo, deram lugar a convenções ainda semelhantes, mas que já apontavam para o que posteriormente seria considerado um novo gênero literário Primeiramente, são feitas considerações sobre conceitos de gênero textual/literário através do tempo, as quais mostram o quanto seu estudo permaneceu constante. A seguir são definidas certas convenções dos dois gêneros, assim como o modo como dialogam entre si. A segunda parte do trabalho analisa as duas primeiras obras em ordem cronológica, Frankenstein e O Médico e o Monstro, de maneira a perceber a predominância de convenções do Gótico – especialmente relacionadas ao conflito interior dos personagens, como o "duplo" – ao mesmo tempo que a emergência de temas da ciência, como os de criador/criatura e ambição científica. O último capítulo verifica como a primeira fase da Ficção científica de H. G. Wells em geral e A Ilha do Dr. Moreau em particular resgatam convenções dos dois gêneros supracitados, ao mesmo tempo servindo como consolidador das convenções do último. Conclui-se, portanto, que houve uma evolução que possibilitou a emergência de um novo gênero ligado ao contexto histórico das obras, o que legitima a consideração dos gêneros como entidades mais livres e não restritivas, que podem estar presentes em diversas obras ao mesmo tempo e ampliar seu horizonte de interpretação.

This thesis establishes a dialogue among three books from 19th century British literature: the novel Frankenstein (1818), by M. W. Shelley; the novella Strange Case of Dr. Jekyll and Mr. Hyde (1886), by Robert Louis Stevenson; and the novel The Island of Dr. Moreau (1896), by H. G. Wells. This comparison is made based on the specific Gothic and Science fiction conventions present in the books. The main theoretical support for the definition of genres employed here comes from Tzvetan Todorov.
