▼ Multiple sclerosis (MS) is an immune-mediated demyelinating disease of the central nervous system (CNS) which has a complex aetiology that involves the interplay of both genetic and environmental factors. Although more than 200 susceptibility loci have been identified through genome-wide association studies, our understanding of the molecular mechanisms underlying these associations remains limited. My predecessor in the laboratory established that the MS associated variants rs9282641 and rs4810485 respectively influence the expression of CD86 and CD40 in freshly collected (ex-vivo) B cells. In my research, I sought to extend this work by investigating the impact of these variants in the context of activation in an effort to refine understanding of the molecular mechanisms underlying the effects of these genotypes on the risk of developing MS. In the first phase of my work I processed venous blood samples from 108 healthy volunteers, who were recruited from the Cambridge BioResource (CBR) to enable a balanced representation of each possible genotype. I isolated peripheral blood mononuclear cells (PBMCs) from these samples and then activated these with CD40L-transfected L cells. In line with the ex vivo results, I found that the expression of CD86 and CD40 remained associated with the genotypes of the respective MS associated variants. I then went on to investigate the effects of these stimulated cells on co-cultured naïve T cells by studying a second cohort of 28 volunteers recruited in pairs from the CBR; the individuals in each pair carrying either the AA or the GG genotype at rs9282641. In these experiments I co-cultured naïve T cells with purified B cells that had been activated with CD40L-transfected L cells. In these 12-day co-cultures, the T cells from subjects homozygous for the risk allele (GG) at rs9282641 demonstrated significantly enhanced proliferation and displayed significantly higher IFN-γ and IL-17 production, and lower IL-10 production. In my PhD I have therefore shown that the altered expression of co-stimulatory molecules on B cells induced by MS risk variants results in enhanced proliferation of pro-inflammatory T cells.

▼ Childhood behavior problems are pervasive with 50% of non-referred families citing noncompliance and behavior problems as an issue (Achenbach & Edelbrock, 1981). Many behavioral parent trainings (BPTs) treat these behaviors at an early age. Recently, adaptions to BPTs include group formats increasing accessibility and decreasing cost, especially for rural families with limited resources (Niec, Barnett, Prewett, & Stanley Chatham, 2016). Beyond BPTs, Alvord, Zucker, and Johnson Grados (2011) developed the Resilience Builder Program to enhance children’s social, emotional, and behavioral skills through a cognitive behavioral framework. The Resilience Builder Program improves anxious and depressive symptoms and reduces behavior problems in children (Watson, Rich, Sanchez, O’Brien, & Alvord, 2013). Although researchers (Borden, Schultz, Herman, & Brooks, 2010) theorized about the suitability for combining BPTs and resilience training, no such study examining the combination of these interventions exists to date. The current study sought to examine the effectiveness of a group treatment combining BPT and resilience training on reducing parental stress and child externalizing behaviors and increasing children’s resilience. A six-week group treatment format consisting of a parent training only group (e.g., Standard Group) and a parent training plus resilience group (e.g., Resilience Group) was utilized to determine the change in child externalizing behaviors, parental stress, and resilience. Multiple 2 (Group Type: Standard; Resilience) X 3 (Time: pre-; mid-; post) factorial ANOVAs were used to analyze the data. Results demonstrated no significant interactions between Group Type (Standard; Resilience) X Time (pre-; mid-; post) for parent stress, children’s resilience, or children’s externalizing behaviors. Significant main effects of Time were found across groups demonstrating a significant decrease in parental stress and children’s externalizing behaviors, and a significant increase in children’s resilience. However, when child age was included as a covariate, these effects did not hold. While there are limitations based on sample size (N = 15) and a lack of control group, there appears to be promising support for using shortened, group-based interventions in the treatment of externalizing behaviors among children. These results indicate BPT alone may be effective in increasing childhood resilience. Future research should aim to address limitations. Advisors/Committee Members: Dorthie Cross, Jeff Klibert.

▼ The material-point method (MPM) is a continuum-based numerical tool to simulate problems that involve large deformations. Within MPM, a continuum is discretized by defining a set of Lagrangian particles, called material points, which store all relevant material properties. Themethod adopts an Eulerian background grid, where the equations of motion are solved at every time step. The solution on the background grid is used to subsequently update all material-point properties, such as displacement, velocity, and stress. In this way, MPM incorporates both Eulerian and Lagrangian descriptions. Similarly to other combined Eulerian-Lagrangian techniques, MPM attempts to avoid the numerical difficulties arising from nonlinear convective terms associated with an Eulerian problem formulation, while preventing grid distortion, typically encounteredwithin meshbased Lagrangian formulations. Over the years,MPM has been successfully applied to many complex problems from engineering and computer graphics. Despite its impressive performance for these applications, the method still suffers from several numerical shortcomings, such as stability issues, inaccurate mapping of the material-point data, and unphysical oscillations that arise when material points travel from one element to another, the so-called grid crossing errors. This dissertation provides an overview of the existing literature that addresses these drawbacks, and introduces new mathematical techniques that improve the performance of MPM. Previous studies have indicated that the use of higher-order B-spline basis functions within MPM mitigates the grid-crossing errors, thereby improving the accuracy of the method. This thesis combines the B-spline approach, known as BSMPM, with an alternative technique to project the information from material points to the background grid. The mapping technique is based on cubic-spline interpolation and Gauss quadrature. The numerical results show that the proposed approach further increases the accuracy of the method and leads to higher-order convergence. Moreover, the extension of BSMPM to unstructured grids using Powell-Sabin splines is discussed. After that, this dissertation compares MPM to the optimal transportation meshfree (OTM) method. Both MPM and the OTM method have been developed to efficiently solve partial differential equations that arise from the conservation laws in continuum mechanics. However, the methods are derived in a different fashion and have been studied independently of one another. This thesis provides a direct step-by-step comparison of the MPM and OTM algorithms. Based on this comparison, the conditions, under which the two approaches can be related to each other, are derived, thereby bridging the gap between the MPM and OTM communities. In addition, the thesis introduces a novel unified approach that combines the design principles from BSMPM and the OTM method. ▼ Le facteur de transcription (FT) c-Myc est l‘un des oncogènes les plus éminents et est impliqué dans le développement, la progression et le maintien de plus de 70 % des tumeurs cancéreuses. Malgré ce fait, aucun inhibiteur de c-Myc n’est disponible en clinique. L’activité transcriptionnelle de c-Myc requiert la dimérisation de son domaine b HLH-LZ avec celui de Max, son partenaire obligatoire, afin de lier des séquences d’ADN cibles nommées E-Box. De son côté, Max forme aussi des homodimères capables de lier ces mêmes séquences, sans toutefois avoir un rôle actif bien défini dans le contrôle transcriptionnel. Récemment, plusieurs altérations dans le gène de Max ont été associées à diverses tumeurs cancéreuses. C’est le cas de ΔMax, un variant d’épissage alternatif, qui amplifie l’activité oncogénique de c-Myc chez des glioblastomes arborant la mutation EGFRVIII. Cependant, son mécanisme d’action demeure inconnu. Par une caractérisation structurale à l’aide de méthodes biophysiques, nous sommes les premiers à faire la démonstration que la perte de la capacité de ΔMax à former un homodimère lié à l’ADN non-spécifique est responsable de son activité pro-oncogénique, suggérant un rôle de suppresseur de tumeur pour Max. Longtemps connu comme un FT spécifique, un nouveau paradigme émergent décrit plutôt c-Myc comme un amplificateur général de la transcription, envahissant les promoteurs et amplificateurs de gènes déjà actifs. Les cellules cancéreuses développent alors une dépendance à l’activité transcriptionnelle de c-Myc que nous avons exploitée afin de développer un agent thérapeutique ciblant spécifiquement c-Myc. En effet, nous avons mis au point un b-HLH-LZ, basé sur celui de Mad1 (Mad*KR), capable de séquestrer Max et de compétitionner avec c-Myc pour la liaison à l’ADN spécifique et non-spécifique. Mad*KR est en mesure de pénétrer spontanément dans des cellules saines et cancéreuses et d’inhiber spécifiquement la prolifération de ces dernières. Mad*KR a également la capacité d’induire spécifiquement l’apoptose des cellules cancéreuses et d’inhiber la transcription de gènes. Un autre b-HLH-LZ pénétrant, appelé Omomyc, a déjà prouvé son utilité thérapeutique dans des études pré-cliniques dans un modèle murin. Cependant, son mécanisme d’action (MOA) était débattu dans la littérature. Dans ce contexte, nous avons contribué à l’élucidation du MOA d’Omomyc. Brièvement, nous avons démontré qu’Omomyc peut lier l’ADN sous forme d’homodimères et d’hétérodimères avec Max. De plus, nous avons montré que les hétérodimères Omomyc/c Myc ne sont pas en mesure de lier l’ADN. Collectivement, ces travaux ont permis de pousser l’utilisation des b-HLH-LZ du réseau Myc/Max/Mad plus près d’une thérapie polypeptidique pour traiter des lésions cancéreuses multiples et pourraient avoir un impact significatif dans notre lutte contre le cancer. ▼ Technology and psychological treatments have increasingly been used together to increase the reach of psychotherapy and potentially reduce treatment costs. This research focused on how technology has been used to deliver or facilitate treatments focused on behavioral parent training. Behavioral parent training is a research-supported method of improving parenting skills and child behavior. We first reviewed any existing research on the topic, and found that treatments that used technology to teach parenting skills were generally successful at improving parent and child behavior. The review also identified many research questions that have yet to be answered about the cost of such interventions, how they work with diverse groups of people, and what makes someone likely to stay with the treatment. The next study in this research project tested a shortened version of a technology-based treatment adapted from a group-based manual that was created for Spanish-speaking families. The program was called Padres Preparados Online (Prepared Parents Online), and it taught three parenting skills on a system that was available online or using an app. Parent coaching, typically carried out in in-person groups or on the phone, was also conducted online. Parents uploaded videos of themselves to an online system and the therapist would record and post video, audio, and text coaching comments to support parents in strengthening the skills they were learning. Results showed that parents and children improved in a variety of ways, ranging from decreased problematic child behavior to decreased parenting stress. This study demonstrated that technology can be used to deliver a parenting program to Latinx families, and helped the study team identify limitations and questions for future research. ▼ The SysML/KAOS method allows to model system requirements through goal hierarchies. B System is a formal method used to construct, verify and validate system specifications. A B System model consists of a structural part (abstract and enumerated sets, constants with their associated properties, and variables with their associated invariant) and a behavioral part (events). Correspondence links are established in previous work between SysML/KAOS and B System to produce a formal specification from requirements models. This specification serves as a basis for formal verification and validation tasks to detect and correct inconsistencies. However, it is required to manually provide the structural part of the B System specification. This thesis aims at enriching SysML/KAOS with a language that allows to model the application domain of the system and which would be compatible with the requirements modeling language. This includes the definition of the domain modeling language and of mechanisms for leveraging domain modeling to provide the structural part of the B System specification obtained from requirements models. The defined language uses ontologies to allow the formal representation of system domain. Moreover, the established correspondence links and rules, formally verified, allow both propagation and backpropagation of additions and deletions, between domain models and B System specifications. An important part of the thesis is also devoted to assessment of the SysML/KAOS method on case studies. Furthermore, since the systems naturally break down into subsystems (enabling the distribution of work between several agents: hardware, software and human), SysML/KAOS goal models allow the capture of assignments of requirements to subsystems responsible of their achievement. This thesis therefore describes the mechanisms required to formally guarantee that each requirement assigned to a subsystem will be well achieved by the subsystem, within the constraints defined by the high-level system and subsystem specifications. The SysML/KAOS method, thus enriched, has been implemented within an opensource tool using the model federation platform Openflexo, and has been evaluated on various industrial scale case studies. It enables the formal verification of requirements and facilitates their validation by the various stakeholders, including those with less or no expertise in formal methods. However, both the specification of the body of B System events and domain model logical formulas (that give B System properties and invariants) and the formal assessment (verification and validation) of the specification can only be manual. They are time consuming and require experts in formal methods. But this is the price to pay to achieve a formal verification and validation of requirements. ▼ In 2015, 224 infants in the United States were diagnosed with end-stage renal disease (ESRD). Infants with ESRD often require renal replacement therapy (RRT), commonly known as dialysis, for treatment. Although they are a minute subset of children, like other children with medical complexity (CMC), infants who require renal replacement therapy (RRT) are medically complex and need high levels of professional and home care. Although treatments are proving more promising, infants with ESRD have historically poor outcomes. Medical decision making for parents of infants with end stage renal disease can be a process fraught with conflict, uncertainty, discord, and stress. Questions of which treatment to pursue or whether or not to forego life-sustaining treatment, weigh heavily. Knowing which decision to make is not a simple task. To aid in decision making, healthcare professionals have promoted Shared Decision Making (SDM) as a model, however, the vast majority of individuals who become patients, still are not supported with the SDM model. The SDM model and others have not been adequately studied from the parent and patient perspective. Parents of infants with ESRD and other CMC, are affected emotionally, financially, physically and relationally by the everyday parenting and care of their child. Decision making is another task that increases the burden to caregivers. This dissertation will provide an explanation of the experience of parents making decisions for infants with ESRD, the meaning made for mothers within that experience, situating both in current research. The dissertation the provides the implications of experience and meaning findings – to social work practice, policy, and education. Article 1 describes the experience of mothers, resulting in four essential themes: (a) information and communication are important, (b) I want to give my baby every chance, (c) it's hard to know for certain what to do, and (d) mothers and fathers take different roles in care giving and decision making. Article 2 provides an explanation of the meaning that mothers of infants with ESRD make of their decision making and caregiving experience: 1) Information has its limits, and 2) Mothers pull the load. Understanding the broader experience and meaning of decision-making for parents, and specifically, mothers of infants with CMC, may increase the effectiveness of decision-making techniques, interventions, and tools developed for parent caregivers in the future. In addition this dissertation and the articles included in it provide a close look at author reflexivity in qualitative study as well as a template for studying the broad experience of parent decision makers that includes beginning with experience, adding meaning in order to reveal a practical purpose and meaningful implications for social workers, social work reserachers and other healthcare professionals. ▼ Parents of children recovering from hematopoietic stem cell transplant (HSCT) experience significant distress due to unpredictable and potentially life-threatening complications. Distress is heightened by intensive caregiving parents provide the child during the first 100 days after HSCT. Management of distress and adaptation to caregiving responsibilities may be enhanced if parents find benefit in their experiences through posttraumatic growth (PTG), yet little is known about the role of PTG 100 days after pediatric HSCT and how parentsâ experiences after HSCT foster PTG. The purpose of the study was to describe parental PTG 100 days after childrenâs HSCT. The study utilized a mixed methods, cross-sectional design with parents, approximately 100 days after their children received HSCT. A total of 31 parent participants reported environmental, personal, and disease characteristics and completed semi-structured, one-on-one interviews. Among these participants, 24 also completed measures of distress, coping, rumination, and posttraumatic growth. Evidence of parental PTG was described in each of the 5 PTG dimensions (relating to others, new possibilities, personal strength, spiritual change and appreciation of life). PTG was positively associated with parental distress, disengagement coping, and rumination measures of involuntary engagement and disengagement (r = .44-.47, p < .05). Appreciation of life demonstrated the strongest associations with distress and rumination (r = .53-.61, p < .01). Curvilinear relationships were observed for the association of distress, disengagement coping, and involuntary engagement with PTG (p < .05). Four major themes emerged from the interview data to describe parent experiences after HSCT: (a) psychosocial and healthcare contextual factors (b) cognitive, affective, and social support reactions to HSCT (c) problem-based, emotion-based, and cognitive coping strategies and (d) PTG. This study is the first to describe parental PTG 100 days after childrenâs HSCT. The results of this study increase understanding of how parentsâ experiences and caregiving responsibilities contribute to PTG. These findings may guide future research to understand how caregiving influences PTG and how nurses can influence adaptation to HSCT through PTG. Advisors/Committee Members: Dr. Mary C. ▼ Carbohydrates are among the most abundant molecules found on the cell surfaces of bacteria, parasites, and viruses. Apart from the conventional roles of carbohydrates as energy sources and structural polymers, carbohydrates are also associated with cancer metastasis, protein stabilization, pathogen infection and the immune response. Cells of our body have sensors made out of carbohydrates on outer surface of plasma membrane and acts as sensors and can detect many kinds of stimuli, and can signal the immune system to respond. Carbohydrate-protein molecular recognition processes have pivotal roles in infections and in immune response to pathogens. To date, several vaccines based on isolated capsular polysaccharides (CPSs) are marketed against infectious diseases. However, the use of isolated capsular polysaccharide poses several limitations, as natural sources are generally limited and the isolation is very challenging. Additionally, the isolated polysaccharides are heterogeneous and often contains impurities. Furthermore, limited protection of certain CPS antigens impairs the efficiency of vaccines. To overcome limitations associated with isolated polysaccharides, synthetic oligosaccharides present an effective alternative with great potential to understand glycan immunology and rationally design effective antigens. Consequently, characterization and reconstruction of carbohydrate epitopes with authentic composition has become one of the major target in glycoscience. To this end, strategies are needed to facilitate the streamlined design and generation of these antigens. This thesis concerns the development of an effective synthetic strategy to obtain Group B Streptococcus (GBS) type II oligosaccharide for vaccine development. GBS, a Gram-positive bacterium, inhabits the intestinal and genitourinary tract of 10‐30% of humans. GBS is one of the primary causes of bacterial infections among neonates and pregnant women, resulting in many severe diseases such as sepsis, meningitis, abortion, and so on. Type II GBS is one of the predominant GBS serotypes and is associated with about 15% of the invasive infections in adults and infants; therefore, represents an important human pathogen. The development of effective preventive vaccine against GBS is much needed to help pregnant women protect their newborns. This thesis describes the effective synthetic strategy to synthesize GBS type II oligosaccharide to be applied for vaccine development. Herein, we present a new and convenient synthesis of the repeating unit of GBS type II capsular polysaccharide. The structure of GBS type II was elucidated in 1983 and the repeating unit of GBS type II is a heptasaccharide composed of α-Neu5Ac (2-3)-ß-D-Gal-(1-4)- ß-D-GlcNAc-(1-3)-[-ß-D-Gal-(1-6)]-ß-D-Gal-(1-4)-ß-D-Gal-(1-3)-ß-D-Glc. ▼ Two rare and severe disorders of insulin action, namely insulin autoimmune syndrome (IAS) and type B insulin resistance (TB-IR), are caused by pathogenic antibodies against insulin or the insulin receptor, respectively. These may arise in isolation or may complicate management of pre-existing diabetes mellitus, and milder forms of the conditions are often suspected in patients with insulin-treated diabetes and labile glycaemic control. Antibody depletion can effectively treat either condition in many cases. This research aimed to target major limitations of existing diagnostics, specifically, that anti-insulin antibody (IA) testing alone does not establish whether antibodies alter insulin action to a clinically-significant degree, and that no clinically-accredited diagnostic test for TB-IR currently exists. An initial collaborative study examined the ability of a panel of commercial insulin assays to quantify ten different insulin preparations. Significant variability in performance of assays against animal-derived and insulin analogues was seen, with certain insulin analogues not detected at all, with important implications for the use of insulin immunoassays in insulin-treated patients. A suite of techniques for investigation of the clinical significance of IAs were then developed and assessed. In a study of five widely-used insulin immunoassays, dilution of IAS plasma led to increased insulin recovery, and polyethylene glycol (PEG) precipitation of IAS plasma decreased insulin recovery in the majority of assays. Gel filtration chromatography (GFC) discriminated high molecular weight and monomeric insulin, while ex vivo addition of exogenous insulin to plasma increased sensitivity of insulin immunocomplex detection. An observational study was performed of 7 patients, all ultimately diagnosed with IAS. IAs were measured using radioligand-binding assay and enzyme-linked immunosorbent assay (ELISA). Method comparison showed results to differ in rank order and relative magnitude. For one patient whose screening IA result was not grossly elevated using either IA assay method, immunosubtraction studies were consistent with the presence of an IgA, a class of antibody under-/not detected in the IA assays studied. Competitive radioligand-binding studies demonstrated IAs to have a range of affinities. 4 patients treated with individualised regimens of immunosuppressive therapy varied in clinical response, and 3 were managed conservatively. Plasma insulin and C-peptide measurements made using mass spectrometry demonstrated under-estimation of insulin and over-estimation of C-peptide concentration using immunoassay in IAS. An observational laboratory and clinical study was also undertaken of 30 insulin-treated patients with diabetes and unexplained labile glycaemia. Magistrsko delo obravnava izkušnje odraslih otrok dementnega starša, natančneje njihovo doživljanje psihološke izgube le-tega. Izvedenih je bilo devet polstrukturiranih intervjujev z odraslimi otroki dementnega, še živečega starša. Pridobljeni podatki so bili analizirani kvalitativno, in sicer z uporabo interpretativne fenomenološke analize. Na podlagi kodiranja podatkov so bile oblikovane štiri glavne teme (zaznane spremembe pri staršu, vloga skrbnika dementnega starša, doživljanje psihološke izgube dementnega starša, socialna opora) in trinajst podtem. Pokazalo se je, da je doživljanje psihološke izgube dementnega starša individualen proces, ki ga večinoma spremljajo neugodna občutja. Razumevanje in osmišljanje tega doživljanja se je med udeleženci razlikovalo. Psihološko izgubo so razumeli bodisi kot izgubo odnosa z dementnim staršem bodisi kot izgubo samega starša bodisi kot življenjsko izkušnjo. Prisotne so bile tudi druge individualne razlike v njihovih opisih. Žalovanje pred smrtjo se je večinoma pojavilo pri tistih udeležencih, katerih demenca starša je bila v naprednejši obliki, kar pa ni povzročilo večje prisotnosti neugodnih občutij. Pomemben dejavnik, ki so ga udeleženci izpostavili, je bil sprejemanje stanja. Raziskovanje tega področja je pomembno, saj je demenca stanje, ki ne preoblikuje zgolj življenja diagnosticiranega posameznika, temveč tudi življenja preostalih družinskih članov.

The master's thesis deals with the experiences of adult children of a parent with dementia, especially their experiences of psychological loss. Nine semi-structured interviews were conducted with the adult children of a demented - still living - parent. The obtained data were qualitatively analysed using interpretative phenomenological analysis. Based on the coding of the data, four main themes (Perceived changes in a parent, Caregiver's role of a parent with dementia, Experiencing psychological loss of a parent with dementia, Social support) and thirteen other subthemes were identified. Experiencing psychological loss of a parent with dementia is an individual process, mostly accompanied by unpleasant feelings. Understanding and making sense of this experience differed among participants. Psychological loss has been identified as a loss of a relationship with the parent, as a loss of the parent or as a life experience. Other individual differences in their descriptions were present as well. Pre-death grief mostly occurred with participants whose parental dementia progressed to a more advanced form. However, this did not cause greater presence of unpleasant feelings. An important factor that participants highlighted was acceptance of the condition. This area of research is important since dementia not only transforms lives of those who have been diagnosed with it but also lives of his family members.

▼ Morbidity and mortality are major concerns in graft-versus-host disease (GvHD), a response often seen after hematologic cell transplantation. Treatment of GvHD remains difficult and more knowledge is necessary. In this thesis we tried to contribute a small piece to the puzzle of GvHD by analyzing the possible relation between B cells and regulatory T cells (Tregs), from a primarily histological perspective. Humanized mice may serve as promising models in studies to GvHD. Designing the right model for the topic of interest is however subject to many choices. Based on our thorough review of the pearls and pitfalls from different mouse strains, transfer routes and sources of material, we decided that RAG2-/-γc-/- mice with intravenous transfer of huPBMC’s would be a sensible choice for our experiments. A thorough analysis was performed to evaluate the basic kinetics and pathology of our model. The induced GvH response in our model appeared to show many similarities with the sclerotic variant of human GvHD. Our analysis continued with an evaluation of our model with additional modifications: macrophage depletion and early B cell depletion. Macrophage depletion accelerated the induction and morbidity of the clinical GvH response and histologically increased the number of B-cells in the infiltrating cell signature, with only limited changes to the amount of Tregs. In early B cell depletion however, a clear improvement on survival was seen. Despite clinical improvement though, our histology only showed a significant effect on liver fibrosis and human TGF-β production within the liver. Other affected organs did not show signs of improvement or alterations. Our next exploration analyzed the effect of B cell activation on Tregs, using a combination of multiple in vitro studies with an in vivo study. Although based on our in vitro studies additional Tregs should temper the GvH reaction (even after B-cell activation), our in vivo study with myeloma was suprising. Clinical symptoms and survival of our mice did improve with additional Tregs, but no significant improvement was shown on a histological level. This contradiction between clinical symptoms and histology is known from human GvHD, as we continued to show with our last evaluation in this thesis, regarding human cutaneous GvHD. In this last analysis however, we’ve found that nowadays neither clinical grading nor histological grading seems to accurately predict a patients survival. With regard to the aim of these thesis, three major conclusions/considerations come forward. First, B cells and Tregs in GvHD have a complex relation, which is subject to the activation of B cells. Second, the lack of correlation between histology and clinical symptoms in GvHD might be explained by cytokines. Last but not least, we need to consider that Rituximab might serve best as a treatment for GvHD when used as an early B cell depletion therapy. Guatteria olivacea R. E. Fries ? uma esp?cie amaz?nica pertencente a fam?lia Annonaceae conhecida popularmente como ?araticum?, ?ata? e ?envireira?, que ocorre em floresta pluvial de terra firme com distribui??o no Brasil, particularmente nos estados do Amazonas e do Par?. De acordo com a literatura, n?o h? registros de estudos qu?micos e farmacol?gicos a seu respeito. Deste modo, neste trabalho foi realizado o estudo fitoqu?mico e de citotoxicidade in vitro dos extratos brutos, fra??es e subst?ncias isoladas das cascas e do ?leo essencial das folhas de G. olivacea. O estudo com as cascas resultou no isolamento e caracteriza??o de dez subst?ncias pertencentes ? classe dos alcaloides, sendo duas delas obtidas em mistura. Para o isolamento dos constituintes qu?micos foram empregadas t?cnicas cromatogr?ficas cl?ssicas (CC e CCDP) e para a identifica??o desses constituintes t?cnicas espectrosc?picas (RMN de 1H e 13C 1D/2D) e espectrom?tricas (EM). Os alcaloides isolados e identificados s?o pertencentes as subclasses dos aporfinos (asimilobina, puterina e discoguatina), oxoaporf?nos (liriodenina e oxoputerina), fenantrenos (atherosperminina, argentinina e N-?xido de atherosperminina) e tetraidroprotoberberinos (coripalmina e discretina). Todos os alcaloides identificados possuem relato no g?nero, por?m s?o in?ditos na esp?cie em estudo, sendo os alcaloides do tipo fenantreno de rara ocorr?ncia. Dentre todos os extratos (hex?nico e metan?lico) e fra??es das cascas, submetidos ao ensaio de atividade citot?xica in vitro, a fra??o alcalo?dica proveniente do extrato metan?lico foi a que apresentou maior atividade contra as linhagens de c?lulas tumorais avaliadas, com valores de CI50 iguais a 20,16 ?g.mL-1 para MCF-7 e 18,25 ?g.mL-1 para HepG2. Dentre os alcaloides isolados que foram avaliados, atherosperminina foi o que apresentou os melhores resultados contra as linhagens de c?lulas MCF-7, HCT116, HepG2 e HL-60 com valores de CI50 iguais a 41,10, 33,07, 42,10 e 33,79 ?mol.L-1, respectivamente. O ?leo essencial extra?do das folhas foi analisado por CG-DIC e CG-EM e apresentou como constituintes majorit?rios os sesquiterpenos germacreno D (17,65%), ?-atlantol (13,21%), ?xido de cariofileno (12,03%), espatulenol (11,26%) e (E)-cariofileno (7,26%). A avalia??o citot?xica desse ?leo exibiu atividade promissora frente as linhagens de c?lulas tumorais MCF-7, HCT116, HepG2, e HL-60 com valores de CI50 iguais a 27,22, 25,74, 34,95 e 16,11 ?g.mL-1, respectivamente. ▼ L’hépatite B est une maladie du foie qui pose un problème majeur de santé publique. Il n’existe à ce jour aucun traitement permettant de guérir complètement de l’infection, et de nouvelles thérapies ont besoin d’être développées. Étant donné son rôle clé dans le cycle de vie du virus de l’hépatite B (VHB), la protéine core qui forme la capside virale est aujourd’hui l’une des cibles avec le plus grand potentiel thérapeutique. Nos recherches sont focalisées sur la caractérisation des capsides du VHB dans différents états conformationnels en utilisant des techniques de biochimie et de RMN du solide, afin de révéler leur conformation précise sous différentes conditions, incluant l’interaction des capsides avec des antiviraux, et la relation entre la conformation de la capside et la maturation du virus. Un système d’expression bactérienne ainsi qu’un système acellulaire de synthèse de protéine à base de germes de blé ont été établis au laboratoire pour produire les capsides, et des protocoles pour désassembler puis réassembler les capsides en présence de différents types d’ARN ont été implémentés. Des échantillons de capsides formées dans E. coli et réassemblées in vitro ont été analysés par RMN. Les différentes formes de capsides observées incluent les protéines tronquées Cp140 et Cp149, la protéine entière Cp183, phosphorylée P-Cp183, et enfin des mutants. Dans un premier temps, nous avons préparé des échantillons pour l’attribution séquentielle de la protéine core par RMN du solide. L’utilisation de la détection carbone en RMN requiert plusieurs dizaines de milligrammes d’échantillon, qui ont pu être produits en utilisant l’expressions bactérienne en milieu minimum contenant des isotopes marqués. Les attributions séquentielles ont été réalisées sur la protéine tronquée Cp149, qui donne des spectres très similaires à Cp183. Cet échantillon a également été utilisé pour identifier les différences conformationnelles entre les 4 monomères de la capside, qui sont provoquées par la symétrie icosaédrale T=4. Ensuite, l’objet principal de cette thèse a été l’investigation et la comparaison d’une large variété de capsides, dans leur forme autoassemblée dans les bactéries E. coli, ainsi que dans leur forme réassemblée. Pour le réassemblage de la protéine entière, qui requiert la présence d’acides nucléiques, nous avons testé différents types d’ARN y compris l’ARN viral prégénomique. Nous avons étudié différentes symétries (T=3 et T=4), ainsi que les états d’oxydation de la capside, et comparé les différences de conformation grâce aux perturbations de déplacements chimiques observées dans les spectres RMN. Nous avons pu identifier les acides aminés impliqués dans les changements conformationnels majeurs entre les différentes préparations. La RMN du solide en détection proton à 100 kHz a récemment émergé comme un outils important pour l’analyse de protéines produites en quantités moindres. Prevodi morajo biti prilagojeni ravni razumevanja otroških bralcev, ki vseh konotacij še ne razumejo. V empiričnem delu smo kulturnospecifične elemente razdelili v različne kategorije. Pri analizi smo se osredotočili na to, katere prevajalske strategije podomačitve je uporabil prevajalec, in ali je pri posodobitvi prišlo do kakšne spremembe v prevodu ali zamenjave prevajalske strategije. Zanimalo nas je tudi, ali so prevodi ustrezni in njihova raba dosledna. V teoretičnem delu smo predstavili otroško literaturo, njen razvoj in prevajanje. Opisali smo kulturnospecifične elemente, njihovo prevajanje ter podomačitve in potujitve. Teoretični del zajema prevajalske strategije po Moni Baker, Jeanu Paulu Vinayu in Jeanu Darbelnetu, ki smo jih uporabili v primerjalni analizi. Slednja je pokazala, da prevajalec kulturnospecifične elemente najpogosteje podomači z uporabo strategije nadomestitve kulturnospecifične prvine oz. adaptacije. Pri posodabljanju prevoda naredi največ sprememb pri prevodih idiomov. Posodobitve prevodov so v večini primerov ustrezne. V obeh prevodih pa pri prevajanju kulturnospecifičnih elementov pride do nedoslednosti.

In the master’s thesis we made a comparative analysis of translations of culturally specific elements in Slovene translations of The Complete Tales by Beatrix Potter. Both translations were made by Branko Gradišnik. The first was released in 1993, and the second in 2002. As we expected both translations wereadapted to children as young readers cannot comprehend all connotations easily. In empirical part we divided culturally specific elements into categories. The analysis then focused on translating strategies. We also analysed changes that occurred in retranslation. We were also interested in adequacy and consistency of the use of culturally specific elements. In theoretical part of the study we presented the main characteristics of children’s literature, its development and specifics of translating. Furthermore, we also presented categories of culturally specific elements, as well as strategies of foreignization and domestication. Theoretical part also includes translation strategies introduced by Mona Baker, and Jean Paul Vinay and Jean Darbelnet which we applied in the comparative analysis. The results of this study showed that in most cases the translator domesticated culturallyspecific elements by using translation by cultural substitution or adaptation, and that in retranslations most changes occurred when translating idioms. Analysis also showed inconsistencies in translating some culturally specific elements.

▼ Objective.Disruptive behaviors are bidirectionally associated with low-quality parent-school interactions (Hoover-Dempsey & Sandler, 1995). However, Non-White children are more likely to be identified by teachers as behaviorally disruptive (Epstein et al., 2005), more likely to have low-quality parent-school interactions (Serpell & Mashburn, 2012), and more likely to be engaged in the justice system (Hockenberry & Puzzanchera, 2013).Considering that non-White parents may interact with schools and teachers differently than White parents (Mariñez-Lona & Quintana, 2009), it is unclear to what extent race might influence the predictive relationship between aspects of parent-school interactions and disruptive behavior problems. Thus, this study will examine how race moderates the reciprocal effects of parent-school interactions on Oppositional Defiant Disorder (ODD) over time. Method. As part of a study on parent help-seeking behaviors, data was collected annually for three years from 75 parents recruited at their child’s first contact with the juvenile court. Regression models assessed the reciprocal relationship between four aspects of parent-school interactions (Engagement, Relationship, Correspondence, and Endorsement) on ODD symptoms over time. Tests of moderation assessed differences in the strength of these relationships between non-White and White children. Results. After controlling for demographic variables and autoregressive effects, both Engagement and Correspondence predicted increasing ODD symptoms. Race significantly moderated the predictive relationship for Engagement and Relationship on ODD symptoms, as well as ODD symptoms on Relationship. Conclusions: This study provides some evidence that parent-school interactions are associated with future behavioral symptoms, and that these relationships differ by race. These results call for greater attention to how schools are engaging with non-White families, particularly those that have come into contact with the justice system. As esp?cies Theobroma grandiflorum, T. cacao e T. subincanum pertencem ao g?nero Theobroma da fam?lia Sterculiaceae. Quimicamente, as esp?cies do g?nero s?o caracterizadas por apresentar triterpenos, esteroides, flavonoides e alcaloides pur?nicos. Estudos realizados no Grupo de Pesquisa de Qu?mica de Biomol?culas da Amaz?nia (Q-BiomA), relataram perfil qu?mico rico em compostos fen?licos nas cascas das sementes de T. grandiflorum. Estes resultados motivaram a execu??o do presente trabalho de compara??o de esp?cies de Theobroma com caracter?sticas f?sicas e qu?micas semelhantes e avalia??o da atividade antioxidante das esp?cies. Os extratos hidroalcoolicos das cascas das sementes das tr?s esp?cies, foram obtidos por macera??o exaustiva, a esp?cie T. grandiflorum apresentou maior rendimento. O perfil cromatogr?fico dos extratos e fra??es foram avaliado por CCDAE, onde foi poss?vel sugerir a presen?a de diferentes flavonoides. A an?lise por EM no modo negativo, evidenciou a semelhan?a das esp?cies T. grandiflorum e T. subincanum. As fa??es foram analisadas por CLUAE-DAD em uma corrida em modo gradiente de 20min, foi poss?vel observar a presen?a da teacrina, ao se comparar os dados obtidos com o padr?o de teacrina, nas esp?cies T. grandiflorum e T. subincanum. Os resultados obtidos das an?lises realizadas com todas as fra??es das esp?cies em estudo, por CLUAE-EMAR/MS2, permitiram identificar diferentes subst?ncias fen?licas: em T. subincanum foram detectados como principais ?ons, m/z 179 (?cido cafeico), m/z 191 (?cido isoc?trico), m/z 329 (?cido vanilico glucos?deo) m/z 431 (vitexina), m/z 477 (hipoleatina-8-O- O-?-D-glucoron?deo), m/z 557 (hipoleatina-8- O-?-D- glucoron?deo 3??-O-sulfato), m/z 461 (isoscutelare?na-8-O-?-D-glucoron?deo), m/z 541 (teograndina I), estas subst?ncias foram relatadas pela primeira vez na esp?cie. A metabol?mica global foi aplicada com m?todos de an?lises de PCA e HCA, onde foi poss?vel confirmar a similaridade de todas as fra??es analisadas, entre as esp?cies T.grandiflorum e T. subincanum e a diferencia??o da esp?cie T. cacao. O extrato hidroalcoolico de T. grandiflorum foi o mais ativo quantitativamente frente aos dois radicais livres, DPPH (44,50 ? 3,35 ?g/mL ? DP) e ABTS (72,17 ? 0,76 ?g/mL ? DP). A utiliza??o de diferentes t?cnicas cromatogr?ficas e diferentes detectores, confirmou a semelhan?a do perfil qu?mico das cascas das sementes das esp?cies T. grandiflorum e T. subincanum, sendo este o ?nico trabalho comparativo das cascas das sementes, um res?duo de ind?strias de alimentos, envolvendo diferentes t?cnicas cromatogr?ficas.

As copaibeiras como s?o conhecidas ?s esp?cies do g?nero Copaifera (Fabaceae) s?o muito apreciadas na medicina popular e na fitoterapia, principalmente, pela utiliza??o do ?leo-resina de copa?ba que possui propriedades terap?uticas, bem como os ch?s das folhas e casas de copa?ba s?o apreciados pelas popula??es tradicionais da Amaz?nia e do cerrado brasileiro. Embora o g?nero Copaifera seja bastante estudado fitoqu?micamente e farmacologicamente, principalmente pelo ?leo-resina, a pesquisa de novos recursos farmacol?gicos ainda se limitam a poucas esp?cies, como a esp?cie end?mica do cerrado Copaifera langsdorffii, seus estudos se direcionam, principalmente nas folhas, assim outras partes vegetais (cascas, galhos, sementes e flores) s?o objetos de pouca ou nenhuma pesquisa. Logo, a metabol?mica n?o direcionada buscou, pela t?cnica de UHPLC-HRMS, analisar qualitativamente o maior quantidade de metab?litos detect?veis no material de estudo e estabelecer similaridades e diferen?as por An?lise de Componentes Principais (PCA). O objetivo do trabalho foi explorar e descrever as caracter?sticas do metaboloma das principais esp?cies (C. langsdorffii Desf., C. multijuga Hayne, C. trapezifolia Hayne, C. lucens Dwyer e C. venezuelana Harms & Pittier) do g?nero Copaifera. Por meio de padr?es de refer?ncia, dados espectrom?tricos de MS2, dados da literatura cientifica e bancos de dados virtuais HMDB e MoNA, os perfis metab?licos das esp?cies foram investigados. Foram detectados 47 metab?litos, entre diversas classes metab?licas, como derivados de ?cido benzoico e ?cido cin?mico, ?cidos graxos, ?cidos diterpenos, flavonoides agliconas, flavonoides glicosilados, um glicos?deo fen?lico, ?cidos galoilqu?nicos e taninos condensados. A PCA observou-se uma forte distin??o no perfil metab?lico das folhas e cascas das esp?cies. As folhas distinguiram-se, principalmente, pela presen?a dos ?cidos galoilqu?nicos di- e tri- substitu?dos e flavonoides glicosilados (derivados de quercetina e canferol) e nas cascas foram detectados principalmente, taninos condensados (derivados de catequina) nas formas de d?mero e tr?mero. Os galhos apresentaram composi??o qu?mica intermedi?ria entre as folhas e as cascas, principalmente devido ? proximidade e extens?o entre os ?rg?os vegetais. As sementes e flores de C. langsdorffii apresentaram composi??es qu?micas divergentes em rela??o aos demais ?rg?os vegetais, trata-se de composi??es com caracter?sticas ?nicas dos ?rg?os e necessitam de maior aten??o fitoqu?mica. Concluiu-se que a abordagem metabol?mica n?o direcionada por UHPLC-HRMS ? uma excelente ferramenta de explora??o metab?lica que possibilitou o perfilamento metab?lico e a detec??o de 47 metab?litos nos ?rg?os vegetais (folhas, cascas, galhos, sementes e flores) das esp?cies e por meio da PCA os metab?litos foram mapeados e os ?cidos galoilqu?nicos (di-, tri-substitu?dos) foram indicadas como biomarcadoras das folhas e os taninos condensados (d?mero e tr?mero) das cascas.

En 2015, l’Organisation Mondiale de la Santé a mis en oeuvre des nouvelles lignes directrices sur la prévention de la transmission de la mère à l'enfant (PTME) du VIH recommandant à toutes les femmes enceintes et allaitantes séropositives, d'initier la trithérapie antirétrovirale à titre de traitement à vie (option B+). Cependant, il existe de plus en plus d'inquiétudes quant à l'efficacité de cette stratégie pour réduire réellement la transmission verticale du VIH à la lumière des défis programmatiques de sa mise en oeuvre ; on observe en particulier des niveaux sous-optimaux de suivi dans les soins chez le couple mère/enfant en Afrique sub-Saharienne. Notre thèse vise à évaluer, d’un point de vue de santé publique, l’engagement dans les soins du couple mère/enfant en prenant l’exemple du Mozambique, pays connu pour avoir l'une des prévalences du VIH les plus élevées au monde. Nous analysons, plus particulièrement trois aspects importants de l’engagement dans les soins : la cascade de soins prénatale, la rétention des femmes enceintes séropositives initiant la trithérapie antirétrovirale et la couverture dudépistage du VIH chez l’enfant exposé.Nous observons dans ce contexte mozambicain une couverture élevée du dépistage du VIH, une amélioration significative de la couverture du traitement antirétroviral au cours du temps et une meilleure acceptation du traitement chez les femmes enceintes séropositives. Cependant,la rétention dans les soins à la fin de la première année du traitement antirétroviral chez les femmes enceintes VIH+ et la couverture du dépistage du VIH chez les nourrissons exposés restent sous-optimales. Nous concluons que les interventions visant à améliorer l’engagementdans les soins du couple mère-enfant doivent être systématiquement encouragées. Enfin, nous soulignons la grande hétérogénéité des définitions des indicateurs utilisées dans les différents programmes de PTME.Ce travail met en évidence les lacunes dans l'engagement du couple mère-enfant dans les soins pour la PTME et ses conséquences sur l'élimination virtuelle de la transmission vertical du VIH. Nous soulignons également l’importance d’une approche consensuelle pour mesurer les indicateurs clés afin de permettre des comparaisons à l’échelle nationale et internationale. Les efforts visant à suivre les femmes sous traitement tout au long de leur vie, en veillant à ce que leurs nourrissons exposés au VIH reçoivent les soins attendus jusqu'à ce que leur statut VIH définitif soit déterminé, constituent la pierre angulaire de l’obtention d'une génération sans Sida pédiatrique d'ici 2030.
