▼ Les cellules dendritiques (DCs) sont les plus efficaces cellules présentatrices d'antigène. La détection de motifs pathogènes, tel que lipopolysaccharides bactériens et ARNs double-brins (ARNdb) viraux, par les DCs provoque leur maturation et induit de nombreux changements morphologiques et biochimiques permettant aux DCs d'acquérir leurs puissants fonctions activatrices des cellules T. Dans ce travail, les réponses des DCs à l'ARNdb ont été analysées. Nous avons montré que, en réponse à au poly I:C, un analogue synthétique des ARNdb, les DCs montent une réponse de stress intégré spécifique au cours de laquelle le facteur de transcription ATF4 et le cofacteur de la phosphatase 1, GADD34, sont exprimés. Les DCs activées par le poly I:C présentent un profil de transcrits similaire à ce qui est produit au cours d'une ‘unfolded protein response'. GADD34 est important pour contrebalancer la phosphorylation du facteur d'initiation de la synthèse protéique eIF2α par la kinase PKR au cours de l'activation des DCs. Contrairement aux fibroblastes embryonnaires murins, les DCs résistent à l'inhibition de la synthèse des protéines induite en réponse à la stimulation avec poly I:C. Néanmoins, l'expression de GADD34 n'a pas un impact majeur sur la synthèse protéique globale. Par contre, GADD34 a été démontré être absolument nécessaire à la production d'interféron du type I et d'IL-6 par les fibroblastes et les DCs en réponse à l'ARNdb. Cette observation a des implications importantes en liant la détection des pathogènes avec les voies intégrés de réponse au stress.
Dendritic cells (DCs) are the most important antigen presenting cells. In response to inflammatory stimulation, DCs display a distinct pattern of differentiation that exhibits specific mechanisms to control the immune response. In this work the responses to dsRNA were analyzed. We have shown that in response to a mimic of dsRNA, polyriboinosinic:polyribocytidylic acid (poly I:C), DCs mount a specific integrated stress response during which the transcription factor ATF4 and the growth arrest and DNA damage-inducible protein 34 (GADD34), a phosphatase 1 (PP1) cofactor, are expressed. GADD34 is important to counteract phosphorylation of eIF2α by PKR. In contrast to murine embryonic fibroblasts (MEFs), DCs resist to protein synthesis inhibition induced in response to cytosolic dsRNA. Nevertheless, GADD34 expression does not have a major impact on global protein synthesis. Importantly, GADD34 was shown to be absolutely required for type I-IFN and IL-6 production by MEFs and DCs in response to dsRNA. This observation has important implications in linking pathogen detection with the integrated stress response pathways. The importance of this link is further underlined by the extreme susceptibility of GADD34-deficient fibroblasts and neonate mice to Chikungunya virus infection.

▼ We induced in rabbits acute nephrotoxicity by injecting high doses of CsA, aiming to clarify whether CsA has a nephrotoxic effect by pre- and post-glomerular vasoconstriction together with direct toxic action on the epithelial cells of the proximal convoluted tubules and the renal parenchyma. We also investigated whether endogenous interferon induced or released by Poly I:C administration in rabbits, has a protective effect on the renal epithelial cells from CsA toxic action. A total of 38 rabbits were allocated in 5 Groups:1. Group of 8 rabbits, controls.2. Group of 14 rabbits, which received high doses of CsA (25 mg/kg B.W.) in 5 consecutive days and then were investigated at the end of the 5th day. 3. Group of 6 rabbits from the previous Group, which were investigated at the end of the 5th day after the last dose of CsA and then were maintained and investigated again on the 30th day from the 1st dose of CsA. 4. Group of 8 rabbits, which received Poly I:C (5 mg/kg B.W.) on days 1st and 3rd. Poly I:C induces interferon production or releases preformed interferon in the animals’ organism. The rabbits were investigated at the end of the 5th day. 5. Group of 8 rabbits, which received CsA, as in Group of CsA and Poly I:C as in Group of Poly I:C. The rabbits were investigated on the 5th day. The amount of food and water for 24 hour-periods for each rabbit of all Groups was measured. The body weight and the 24 hours urine volume before the study and at the end of it were checked. In the rabbits, which were followed and studied again on the 30th day, the same investigations were performed. Blood and 24 hours urine and a mean urine specimen were collected for investigations. From the blood: Levels of CsA were estimated, complete blood count, serum sodium, blood urea and serum creatinine, uric acid, SGOT, SGPT and γ-GT. From the urine: In the 24 hours urine collections, the urinary activities of AP, LDH and NAG were assayed. An urinalysis was examined and urine sodium and creatinine were estimated. FENa was calculated from sodium and creatinine in serum and urine. The animals were killed at the end of the experiment and the kidneys removed for histological examination with light microscopy.The CsA levels were highly increased after the 5th day of CsA administration. The rabbits which received CsA in the Group of CsA and CsA + Poly I:C lost significant body weight, most probably due to the reduced amount of food taken. The amount of urine was reduced due to the reduced volume of water received and also due to the reduction of GFR from the CsA induced vasoconstriction. The body weight of the rabbits returned to previous levels in the 6 rabbits studied again on the 30th day.Blood urea and serum creatinine were increased in the rabbits received CsA and CsA + Poly I:C, while FENa was reduced compared to controls, as well as the total protein levels. ▼ In the experimental autoimmune encephalomyelitis (EAE) murine model of multiple sclerosis, leukocytes transmigrate through the blood brain barrier into the perivascular space and enter the parenchyma of the central nervous system (CNS). This entry is facilitated by the interaction of very late activation antigen-4 (α4β1-integrin) on activated T cells with vascular cell adhesion molecule-1 that is expressed by inflamed endothelium. It has been demonstrated that α4-integrin antagonism in multiple sclerosis patients can be significantly beneficial, but it can also lead to detrimental CNS infections due to impairing of immune surveillance. In this work, we seek to understand cell type specificity of α4-integrin antagonism in order to improve immune competence in the CNS. We generated the conditional knock out mouse strain Mx1.Cre+ α4-integrinfl/fl on the C57BL/6 background that enables α4-integrin gene ablation by the Cre-lox recombination system upon
poly I:
C treatment. We hypothesize that antagonism of α4-integrin diminishes immunocompetence within the CNS by differentially affecting leukocyte subsets. In order to test this hypothesis, we actively and passively induced EAE on Mx1.Cre+ α4-integrinfl/fl. We observed that disease susceptibility and severity in
poly I:
C treated Mx1.Cre+ α4-integrinfl/fl mice were similar to that of wild type (WT), but the disease onset was significantly delayed. Additionally, while there was decreased migratory capabilities of CD45+ splenocytes from Mx1.Cre+α4-integrinfl/fl mice, the absolute number and composition of leukocytes in the CNS of EAE mice was similar in WT and
poly I:
C treated Mx1.Cre+ α4-integrinfl/fl mice. The presence of Evans Blue dye in the parenchyma of
poly I:
C treated mice suggest a role of
poly I:
C in this migratory effect. On the other hand, adoptively transferred cells from
poly I:
C treated Mx1.Cre+ α4-integrinfl/fl mice did not transfer EAE disease into WT. Based on these data, we concluded that Mx1.Cre+ α4-integrinfl/fl strain is limited to study the role of α4-integrin ablation on immunocompetence, however it is successful for the generation of α4-integrin deficient leukocytes. Finally, we determined optimization techniques for CNS leukocyte isolation that can be of benefit for future analyses.

▼ Control of nuclear RNA stability is an important determinant of gene expression, but the factors involved in nuclear RNA decay remain largely unknown in higher eukaryotes. Here,
I describe our work showing that polyadenosine (
poly(A)) tails can stimulate transcript decay in the nucleus of human cells, a function mediated by the ubiquitous nuclear
poly(A) binding protein PABPN1. We show that PABPN1 is required for the degradation of a viral nuclear noncoding RNA as well as an inefficiently exported human mRNA. Importantly, the targeting of RNAs to this decay pathway requires the PABPN1 and
poly(A) polymerase (PAP)-dependent extension of the
poly(A) tail. Nuclear transcripts with longer
poly(A) tails are then selectively degraded by components of the nuclear exosome.
I also describe our work showing that PABPN1 and PAP are required for the degradation of a variety of nuclear-retained long noncoding RNAs. Taken together, this work uncovers an important pathway in the turnover of RNAs in the nucleus.

▼ Poly(c)-binding proteins (PCBPs) are RNA and
single-stranded DNA
binding proteins known to be involved in a
variety of post-transcriptional control
pathways. Among the
diverse functions of these proteins is the interaction with a
27
nucleotide pyrimidine-rich domain within the 3‘UTR of tyrosine
hydroxylase
(TH) mRNA. Mutations to this domain result in
decreased stability of TH mRNA,
and it has been postulated that
PCBPs play key roles in maintaining TH mRNA
stability and in
regulating TH mRNA translation. In order to further characterize
the role of PCBPs in the post-transcriptional regulation of TH, we
examined the
tissue distribution as well as the subcellular
localization of these proteins.
Fluorescent immunohistochemistry
and confocal microscopy was used to
analyze the distribution of
PCBPs in TH-positive cells of the rat midbrain, locus
coeruleus,
and adrenal gland. PCBP 1 was not found in TH-positive cell bodies
of
the substantia nigra pars compacta (SNc), however this protein
was present in
TH-positive cell processes extending into the
substantia niga pars reticulata
(SNr). In contrast, PCBPs 2, 3,
and 4 were found in select cell bodies of the SNc
as well as in
the TH-positive fibers extending into the SNr. PCBP 3 was found in
TH-positive cell bodies of the locus coeruleus, however PCBPs 1, 2,
and 4 were
not. Finally, all four isoforms were found in the
adrenal with PCBP1 exhibiting a
stronger staining pattern in the
medulla relative to the surrounding cortex. PCBPs
2, 3, and 4
appear to be distributed equally in both the cortex. PCBP3
immunoreactivity was observed in the adrenal medulla, however this
isoform
appeared to be present in a lower concentration relative
to PCBP1.
A number of previous studies have shown that PCBP
isoforms 1, 2, and 4
are widely distributed throughout the cell
and that they shuttle between the
nucleus and cytoplasm. However,
PCBPs have not yet been shown to associate
with specific
cytoplasmic organelles. Given that PCBPs appear to modulate the
stability and translation of TH mRNA, we have speculated that PCBPs
are
localized to cellular compartments that are involved in
translational silencing and
mRNA degradation. Recently, Fujimura
et. al. (2009) have demonstrated through
immunofluorescence
studies in HeLa and Neuro2a cell lines that PCBP2 colocalizes
with
a subset of processing bodies (p-bodies). These cytoplasmic foci
contain mRNA decay factors and translational repressors and are
known to be
involved in mRNA degradation and translation
silencing. Based on these findings,
we tested whether PCBPs
co-localize with p-bodies markers in
catecholaminergic cell lines.
Mouse mesencephalon/neuroblastoma hybridoma
MN9D cells were
immunofluorescently stained with antibodies directed towards
PCBPs
1, 2, and 3, as well as known p-body markers. Confocal microscopy
revealed that PCBPs apparently co-localize with a subset of
p-bodies in MN9D
cells. These findings may provide insight into
the role of these RNA binding
proteins in catecholaminergic cells

▼ Carrier-selective passivating contacts (CSPC) are very promising contact structures for highly efficient silicon solar cell. They provide passivation of silicon surface and high carrier selectivity. So far, superior results have been achieved through the use of a stack of
poly-Si with SiO2. The objective of this project is the optimization of an alternative passivation stack based on carbon alloyed
poly-Si (
poly-SiCx). The alloy is selected since carbon provides improved material resilience against blistering and wet-chemical stability in commonly used chemicals in the silicon industry. The optimized
poly-SiCx contacts are implemented in fornt back contacted (FBC) solar cell. We improve the passivation quality of SiOx/
poly-SiCx contacts optimizing several parameters. Our main focus is on the buffer layer optimization and its deposition methods comparing LPCVD and PECVD technique. We also turn our attention to identify the optimum annealing temperature and thickness of doped layers. Finally, we focus on the influence of the different capping layer composition on hydrogeneation process. With the optimization of the annealing temperature, and thickness of (
i)a-Si layer deposited by LPCVD, we obtain high passivation quality on cell precursor structure:
i-Voc of 713 mV, τeff of 2.14 ms and Jo of 9.5 fA/cm2. On the contrary, during the optimization of buffer layer deposited by PECVD, we firstly focus on the material properties and the bonds present in the deposited layers to minimize the hydrogen contact. After the optimization of the (
i)a-Si:H layer thickness, deposition parameters and annealing temperature on (p)
poly-SiCx symmetrical sample, we obtain the remarking results:
i-Voc of 681 mV, τeff of 1.15 ms and Jo of 31.3 fA/cm2. The potential of SiOx/
poly-SiCx passivation contacts is checked on the device level of FBC solar cells. On FBC solar cells with buffer layer deposited by the PECVD, after an effective post annealing of the cell performed at a temperature of 350°
C, we achieve the remarking parameters; Voc of 659 mV, Jsc of 34.36 mA/cm2, FF of 77.58 % and ηact of 17.56 %. We check also the the influence of different SiNx capping layers on the hydrogenation process. We obtain the best results on the FBC solar cell on which the capping layer is stoichiometric. These results, after the high-temperature port annealing, are Voc of 690 mV, Jsc of 36.18 mA/cm2, FF of 80.38 %, and ηact of 20.06 %. These are also the best FBC
poly-SiCx results which we have obtained in this project. The application of SiOx/
poly-SiCx passivation contacts is investigated also in terms of the IBC solar cell concept. We proposed two alternative fabrication methods based on photolithography, which is crucial for contact formation of IBC solar cells. One of the fabrication methods focuses on a buffer layer deposited by LPCVD as this layer is of higher quality and homogeneity than the interlayer deposited by PECVD. ▼ A hipótese neurodesenvolvimental da esquizofrenia (SZ) tem origem numa perturbação do desenvolvimento ou maturação encefálica no período perinatal. Nesse contexto, objetivo deste trabalho foi avaliar o efeito da suplementação com ômega-3 (ω-3) em dois diferentes períodos de vida (período de crescimento fetal e durante a adolescência) em parâmetros comportamentais e bioquímicos em um modelo de desafio imune neonatal induzido por ácido poliinosínico:policitidílico (
Poly(
I:
C)). Para isso, fêmeas wistar grávidas foram divididas em dois grupos para o tratamento de prevenção a partir do 16° ao 21º dia de gestação: um grupo recebeu ω-3 na dose de 0,8g/Kg via gavagem (grupo O), e o outro recebeu Tween (grupo T) na mesma dose e via. Após o nascimento, do 5º ao 7º dia pós-natal (PN) cada grupo de recém nascidos foi subdivido em dois grupos: um recebeu
poly(
I:
C) na dose de 2mg/kg (grupo OP ou TP) e outro recebeu salina na dose 1 ml/kg (grupo OS ou TS), ambos via intraperitoneal. Na adolescência, do 30º ao 44º dia PN, os animais receberam o segundo tratamento de prevenção com ω-3, seguindo a mesma suplementação da gestação: com ω-3 (grupo OPO, TPO e TSO) ou Tween (grupo OPT, OST,TPT e TST)). No total, foram 7 grupos de tratamento (n~15 animais/grupo): TST, TPT, TSO, TPO, OST, OPT e OPO. No 45º dia PN foram realizados os testes comportamentais de Y-maze, campo aberto e interação social (three-chamber sociability and social novelty). Ao final foi realizada a eutanásia dos animais seguida da a coleta do sangue troncular e dissecação das estruturas cerebrais - córtex pré-frontal (CPF) e hipocampo (HC). O fator neurotrófico derivado do encéfalo (BDNF) e o estresse oxidativo, através das substâncias reativas ao ácido tiobarbitúrico (TBARS) e glutationa peroxidase (GPx), foram avaliadas no soro e nas estruturas cerebrais Os resultados foram expressos em mediana e intervalo interquartil (IQR). A análise foi realizada por Mann-Whitney U para comparações entre dois grupos (TST x TPT) e, para comparações múltiplas, empregou-se Kruskal-Wallis. Os resultados mostram que a administração neonatal de
poly(
I:
C) foi capaz de induzir um comprometimento significativo na interação social e no interesse por novos conspecificos quando comparado ao controle (TST), sendo estes comportamentos considerados como um sintoma negativos da SZ. O desafio imune também aumentou significativamente a peroxidação lipídica no HC. A associação de suplementação ω-3 durante o crescimento fetal e durante a adolescência (OPO) foi capaz de proteger os animais de déficits de interesse social quando comparados ao grupo TPT. Além disso, apresentou níveis séricos significativamente elevados de BDNF e níveis mais baixos de peroxidação lipídica no HC do que o grupo TPT. ▼ The use of pharmaceutically active peptides and proteins has increased significantly over the last 30 years. Topical delivery of growth factors to chronic wounds to enhance wound management and repair have been of particular interest. The aim of the work in this thesis was to evaluate the ability of the PVA-borate hydrogel formulation to deliver biologically active protein payloads. The unique viscoelastic properties of PVA-borate hydrogels are advantageous for application to wounds. The hydrogel can flow into the wound crevasses providing maximal surface area for drug delivery. Yet, they maintain enough cohesive integrity of its cross-linked network to allow for complete and intact removal from the wound, without causing any trauma to newly formed tissue. The ability of the PVA-borate hydrogel to maintain a moist environment and also absorb small volumes of exudate from low exuding wounds also aids wound healing. Several model payloads (BSA, haemoglobin, trypsin and pepsin) were incorporated into the hydrogel. These proteins where found to change the viscoelastic properties of the PVA-borate hydrogel, but, were released in a sustained manner from the hydrogel without undergoing any degradation. This investigation indicated that the PVA-borate hydrogel is capable of providing a sustained release of protein payloads with certain physical and chemical characteristics. Insulin-like growth factor-1 (IGF-1) was incorporated into the hydrogel and found not to effect the viscoelastic properties of the hydrogel. In vitro release studies confirmed that the hydrogel provided a sustained release of IGF-1, which had maintained its biological activity and not experienced any degradation. Cell culture studies confirmed that IGF-1-loaded PVA-borate hydrogels can stimulate fibroblast and keratinocyte migration and proliferation. Therefore, the work in this thesis demonstrates that IGF-1-loaded hydrogels may have a beneficial use in the management and repair of chronic and recalcitrant wounds.

The infection resulted in very high mortality, which causes massive economic loss. Therefore, early detection of pathogen is critical to prevent epidemic outbreak. Retinoic acid inducible gene-
I (RIG-
I) is an intracellular receptor recognizing dsRNA virus. Upon dsRNA virus infection, RIG-
I is induced and over-expressed in protein level that mediating type
I Interferon (IFNs) antiviral signaling. In this study, total RNA extracted from fertilized eggs of Epinephelus coioides was extracted for the reverse transcription-polymerase chain reaction (RT-PCR) amplification of 432 bp cDNA encoding regulatory domain (RD) of RIG-
I protein. The amplified cDNA fragment was then cloned into pQE32 for the prokaryotic expression of 17 kDa His-tagged RIG-
I (His-RIG-
I-RD) fusion protein. His-RIG-
I-RD fusion protein was purified for immunization of New Zealand white rabbit to produce antiserum against RIG-
I protein. Our Western blot result confirmed the specificity of antiserum for recognizing 0.01 μg His-RIG-
I-RD fusion protein at 1:5000 dilution. In addition, 0.02 μg of immunogen His-RIG-
I-RD fusion protein can be readily detected at 1:25000 dilution. Furthermore, this antiserum could detect the increase of a 72 kDa protein in lysate prepared from head kidney of dsRNA viral mimic
poly(
I:
C) injected Epinephelus coioides, but not in uninjected control fish. The immunostaining results of frozen head kidney sections also revealed the RIG-
I immunoreactivity using this antiserum. RIG-
I protein immunoreactivity was prominently increased in melanomacrophage center (MMC) of head kidney from
poly(
I:
C) injected fish compared to that of uninjected control fish. In summary, we have generated a specific antiserum against RIG-
I protein of Epinephelus coioides, which could be a useful reagent for further study of Grouper immune response and signal transduction upon virus infection.

▼ The formation of isotactic
poly(propylene) (iPP) and polycarbonate (PC) nanowires of tunable diameter was demonstrated by melt-wetting the polymer into nanoporous anodic alumina. The crystallization process was analyzed using differential scanning calorimetry. A transition from hetero- to homogeneous crystallization was observed for iPP as the pore diameter decreased. Also, the Avrami exponent decreased as the pore diameter decreased, indicating preferential 1-D crystallization for diameters of 40 nm or less. X-ray diffraction studies showed that the polymer chains crystallize into the ??-phase and that they preferentially orient along the a- and b-axis, perpendicular to the pore wall. Directed crystallization of iPP may enhance the materials electrical and mechanical properties.

▼ Alcohol and cigarettes are commonly used together and
>80% of individuals with an alcohol use disorder (AUD) are also
smokers. Attention bias is the tendency of our perception to be
affected by our recurring thoughts and represents a key area of
research in addiction psychiatry. However, current paradigms
developed to assess attentional biases are limited to individual
cues investigating the general process rather than investigating
how this process might be manifested in terms of selective
attention. It has been previously shown that in smokers with AUD,
there exists a potentiation of attentional processing towards
specific related cues, which in turn, escalates craving of the
preferred substance. The primary aim of this study was to develop a
novel paradigm that integrated a cue-reactivity (CR) session (bar
laboratory) with an eye-tracking (ET) session (virtual laboratory)
to directly measure selective behavioral response and selective
visual attention. In each session, participants’ interaction with
alcohol and cigarette was determined in order to investigate which
substance was associated with an attentional bias. To provide an
additional tool to measure and capture craving from psychological
and visual perspectives, we also explored possible correlations
between time spent with alcohol or cigarette with selective
substance craving. This was a pilot human laboratory study (n = 32)
with moderate to heavy drinking smokers who completed both a CR
session and an ET session using their preferred alcoholic drink and
cigarette brand in order to link behavior, attention and cravings
in dual users. In the CR, when we compare the time spent
interacting with either alcohol or cigarette, there was a
difference between alcohol (p<0.001) and cigarette (p<0.01)
when compared to water, but there was no difference between the two
substances (p’s>0.05). In the ET session, when we compare the
time spent interacting with either alcohol or cigarette, there was
a difference between cigarette and alcohol (p’s<0.001) compared
to water, but there was no difference between the two substances
(p’s>0.05). In the integration of the CR session with the ET
session, time spent interacting with alcohol in the CR session is
correlated to total time fixating on alcohol during the ET session,
p<0.05). However, the time spent interacting with the cigarette
in the CR session trends towards correlation with time fixating on
the cigarette during the ET session (p=0.06). In the analysis of
craving, the time spent with alcohol during CR is correlated both
with cravings for alcohol and cigarettes (p’s<0.05). Finally,
when grouping participants by self-reported preferred substance,
alcohol cravings were higher both for individuals that preferred
alcohol (p <0.05) and for individuals that preferred cigarettes
(p<0.05). With results indicating that the integrated paradigm
is a novel and useful measure to relate behavior and attention in
dual users, a larger study can be developed to further elucidate
the drivers of behavior and attention as well as cravings…

▼ Communicable diseases, including infectious disease, remained the second highest contributor to the total number of disability adjusted life-years (DALYs) lost in 2017 (more than 17% of the 2.5 billion DALYs) (1). The pathogens responsible for these diseases are a diverse array of viruses, bacteria and parasites, each with varying tropism,
i.e. the propensity of a given pathogen to infect a particular cell type, tissue and/or species. At the level of species tropism, some pathogens are more promiscuous than others, infecting a broad range of vertebrates and invertebrates (e.g. the bacterium Salmonella enterica) while on the opposite end of the spectrum are pathogens with highly restricted tropism limited to only humans or humans and select non-human primate (NHP) species (e.g. hepatitis
C virus, HCV). It is the latter category of pathogens that are most problematic for researchers as their very nature hinders study in traditional small animal models, such as mice. Thus, to facilitate such work, it is crucial to identify the determinants of tropism, which at the molecular level can be due to: (
i) necessary factors missing or the ortholog of such a factor being incompatible in a given species, such as receptors for a specific virus; (ii) the presence of dominant restriction factors that block steps of the pathogen’s life cycle, such as its replication within host cells; (iii) the varying abilities of a pathogen to evade and/or disrupt the immune response of a given host. The following dissertation describes telling examples of these determinants, specifically for RNA viruses infecting humans, and how knowledge of such factors is important in generating improved research models as well as in understanding emergent and re-emergent pathogens. This work provides novel insights concerning the impact of cyclophilin A on HCV host tropism and the conservation of innate immune responses observed across diverse NHP species, with implications for the development of a small, immunocompetent animal model for HCV and related pathogens.

▼ Iron is a crucial element that is associated with many metabolic pathways important for life sustaining processes. Polyamines are small positively charged polycations involved in various physiological functions. Both iron and polyamines levels are known to be high in cancer cells which suggests a possible unexplored link between the two metabolic pathways. For the first time, we demonstrate that iron-depletion robustly regulates the expression of 13 polyamine pathway proteins. Iron-depletion also decreased polyamine and S-adenosylmethionine levels (required for spermidine/spermine biosynthesis) and decreased 3H-spermidine uptake in accordance with expression of the polyamine importer, SLC22A16. The “reprogramming” of polyamine metabolism by iron-depletion showed dependence on the proto-oncogene, c-Myc, and tumour suppressor, p53 expression. Furthermore, the ability of iron chelators to inhibit proliferation can be rescued by polyamine supplementation. Collectively, these data demonstrate that iron and polyamine metabolism are closely linked at multiple levels. Moreover, we have identified that the mRNA and protein expression of the iron-containing enzyme, aci-reductone dioxygenase 1 (ADI1), was regulated by iron levels. Cellular iron depletion or deficient ADI1 metalation by the iron chaperone, PCBP1, promotes the proteasomal degradation of ADI1. Collectively, this demonstrates that cellular iron regulates ADI1 stability, a key enzyme involved in methionine salvage, polyamine biosynthesis and proliferation. In addition to regulating ADI1, poly(rC)-binding proteins (PCBPs) have been reported to function as iron-binding chaperones that deliver iron to ferritin. We observed that PCBP2 enhances, while PCBP1 inhibits ferritin 59Fe-loading and ferritin protein expression. Our results suggest that the regulation of ferritin iron-loading by PCBPs may involve a combination of translational regulation and/or iron chaperone activity. Overall this study has demonstrated for the first time the direct interaction between iron metabolism and polyamine metabolism, which is important in understanding how cancer cells can survive adverse environments.

▼ Post-transcriptional processing of all tRNA primary transcripts constitutes an important cellular activity, thought to play a role in regulation of protein synthesis and overall cell fitness. Ribonucleases are the predominant effectors of
these processing events. This dissertation research aimed to achieve greater understanding of the mechanisms involved in tRNA processing, with a focus on the biological role of RNase P through microbiological, genetic and biochemical analysis. We also
attempted to discern the role of polyadenylation in tRNA maturation and identify physiological interactions between RNase P and poly(A) polymerase I, the primary polyadenylation enzyme in E. coli. Based on our analysis of the valU and lysT polycistronic
operons, we have proposed a novel pathway for RNase P-mediated processing of tRNA primary transcripts. The enzyme works in an overall 3’ → 5’ direction, proficiently removing Rho-independent transcription terminators. Other endoribonucleases such as
RNase E play only a minor role in the processing of the large polycistrons. Dramatic reductions in the amount of mature valine tRNA do not result in a similar reduction in the level of aminoacylated valine tRNA. Furthermore, deficient processing at the
5’-ends of tRNAs does not lead to reduced levels of aminoacylated tRNAs, suggesting that a few extra nucleotides at the 5’-termini of tRNAs might not completely inhibit aminoacylation, as previously believed. The work presented also discovered that
inactivation of poly(A) polymerase I suppresses the temperature sensitivity of an RNase P mutant. There is significant polyadenylation of tRNA precursors in an RNase P mutant and removal of the polyadenylation enzyme, PAP I, results in improved
processing of the tRNA precursor 3’-termini. Lack of PAP I leads to increased cellular levels of RNase T, the primary exoribonuclease involved in 3’-end processing of tRNA precursors.

▼ After RNA virus infection, the innate immunity utilizes RIG-
I family of receptors in the cytoplasm to initiate the production of type-
I IFNs and proinflammatory cytokines by mitochondrial protein MAVS, which forms a prion-like structure to recruit TBK1and IKK complex to activate IRF3 and NF-κB, respectively. Herein, we revealed the important roles of CARMA3 and BCL10 in RIG-
I/MAVS signaling pathway for the first time. CARMA3 or BCL10 deficient cells exhibited partially defective NF-κB activation but hyperactivation of TBK1-IRF3 signaling pathway upon ssRNA virus infection. It led to less production of IL6 but more production of type
I IFNs and resulted in CARMA3-deficient mice more resistant to virus infection, including VSV and H1N1 influenza virus. Mechanistic studies showed that CARMA3 and BCL10 physically interacted with MAVS and directly regulated MAVS aggregation. Overexpression of CARMA3 and BCL10 inhibited MAVS aggregation. Consistently, MAVS aggregation more easily occurred in CARMA3 deficient cells after VSV infection. VSV induced the cleavage of CARMA3 and led to more MAVS aggregation. These novel findings not only identified the important role of CARMA3 and BCL10 during influenza virus infection, which may provide the new target for future therapeutic interference but also revealed molecular mechanism by which CARMA3 and BCL10 orchestrated the NF-κB and IRF3 activation to balance the production of type
I IFNs and proinflammatory cytokines

▼ Ce travail de thèse décrit la synthèse de copolymères diblocs amphiphiles par polymérisation radicalaire contrôlée par transfert d'iode en mode inverse (RITP). Dans un premier temps, des homopolymères à base de monomères activés (styrène, acrylate de méthyle, méthacrylate de méthyle), non activés (acétate de vinyle) et fonctionnels (chlorométhylstyrène, acrylate de tert-butyle), ont été synthétisés par RITP. Des aspects tels que le contrôle des masses molaires ainsi que la fonctionnalité en iode du bout de chaîne ont été examinés. Afin de démontrer la compatibilité de la RITP avec d'autres techniques de polymérisation contrôlée/vivante (non radicalaires), le copolymère amphiphile non-ionique poly(styrène)-b-poly(2-méthyl-2-oxazoline) (PS-b-P(MOx)) a été élaboré par la voie toute iode en procédé « one-pot » en combinant de manière consécutive la RITP du styrène avec la polymérisation cationique par ouverture de cycle (CROP) de la 2-méthyl-2-oxazoline. Par ailleurs, des copolymères cationiques poly(styrène)-b-poly(chlorométhylstyrène) quaternisé avec la triéthylamine (PS-b-PCMS+) et anioniques poly(styrène)-b-poly(acide acrylique) (PS-b-PAA-) ont été synthétisés par RITP du styrène suivie de l'ITP du bloc hydrophile. Tous ces copolymères amphiphiles ont été obtenus à partir de polystyrène iodé (PS-I) de faibles masses molaires, allant de 1000 à 3000 g.mol-1, jouant le rôle de macro-amorceurs (CROP) ou de macro-agents de transfert (ITP). L'étude de l'auto-organisation en phase aqueuse de ces copolymères amphiphiles a révélé la formation de micelles pour des concentrations supérieures à la concentration d'agrégation critique (CAC) dont la valeur a été déterminée par diffusion dynamique de la lumière (DDL) et spectroscopie de fluorescence. Enfin, des résultats très prometteurs ont été obtenus dans l'utilisation d'un copolymère cationique amphiphile PS-b-PCMS+ comme agents structurants de matériaux siliciques mésoporeux nanostructurés élaborés par procédé sol-gel en milieu basique. La synthèse de ces copolymères amphiphiles ainsi que leur caractérisation physico-chimique ont mis en exergue que la RITP permet de couvrir la synthèse de différentes catégories de copolymères amphiphiles, tout en étant une technique simple à mettre en œuvre, peu couteuse et robuste.
This manuscript describes the synthesis of amphiphilic diblock copolymers by reverse iodine transfer polymerization (RITP). Firstly, homopolymers derived from activated (styrene, methyl acrylate, methyl methacrylate), non-activated (vinyl acetate) and functional monomers (chloromethylstyrene, tert-butyl acrylate) synthesized by RITP, were studied. ▼ The prevention and treatment of cancer and infectious diseases requires vaccines that can mediate cytotoxic T lymphocyte-based immunity. A promising strategy is protein subunit vaccines composed of purified protein antigens and immunostimulatory adjuvants, such as Toll-like receptor (TLR) agonists. In this research, we developed two new biodegradable polymeric delivery vehicles for protein antigens and TLR agonists, as model vaccine delivery systems. This work was guided by the central hypothesis that an effective vaccine delivery system would have stimulus-responsive degradation and release, biodegradability into excretable non-acidic degradation products, and the ability to incorporate various TLR-inducing adjuvants. The first vaccine delivery system is a cross-linked polyion complex micelle which efficiently encapsulates proteins, DNA, and RNA. The micelle-based delivery system consists of a block copolymer of
poly(ethylene glycol) (PEG) and
poly(L-lysine), cross-linked by dithiopyridyl side groups to provide transport stability and intracellular release. The second delivery system consists of solid biodegradable microparticles encapsulating proteins, nucleic acids, and hydrophobic compounds. The microparticles are composed of pH-sensitive polyketals, which are a new family of hydrophobic, linear polymers containing backbone ketal linkages. Polyketals are synthesized via a new polymerization method based on the acetal exchange reaction and degrade into non-acidic, excretable degradation products. In addition, the technique of hydrophobic ion pairing was utilized to enhance the encapsulation of ovalbumin, DNA, and RNA in polyketal microparticles via a single emulsion method. Using in vitro and in vivo immunological models, we demonstrated that the micelle- and polyketal-based vaccine delivery systems enhanced the cross-priming of cytotoxic T lymphocytes. The model vaccines were composed of ovalbumin antigen and various TLR-inducing adjuvants including CpG-DNA, monophosphoryl lipid A, and dsRNA. The results demonstrate that the cross-linked micelles and polyketal microparticles have considerable potential as delivery systems for protein-based vaccines.

▼ Many fabrication methods for scaffolds made with biodegradable polymers and hydrogels have shown promise for tissue engineering applications. Both possess their own benefits and disadvantages. However, by utilizing both, the advantages of each can be combined into a single hybrid scaffold. By increasing the reaction concentration of a modified synthesis protocol previously published, the lower critical solution temperature (LCST) of a biodegradable, thermos-sensitive hydrogel, poly (ethylene glycol)-poly (N-isopropylacrylamide) (PEG-PNIPAAm), was optimized to its lowest possible concentration for gelation (5 wt. %). The resulting PEG-PNIPAAm was blended with a hydrophobic, biodegradable polymer, poly (ε-caprolactone) (PCL), and electrospun into a 3-dimensional (3D), electrospun hybrid scaffold. The blending ratio of PEG-PNIPAAm:PCL (13:9), scaffold porosity (90%) and fiber diameter (11 μm) where optimized for uniform cell seeding and cell encapsulation within the dissolved hydrogel. During cell seeding, PEG-PNIPAAm dissolved into the media solution, surrounding seeded cells and filling the scaffolds pore volume without the need of a mold. When incubated at 37 °C, the seeded cells are encapsulated within the PEG-PNIPAAm hydrogel. The hydrophobic PCL network remains as a structural support, increasing the compressive, elastic and viscoelastic properties of PEG-PNIPAAm. Including PEG-PNIPAAm within the PCL scaffold, also resulted in the improvement of mechanical characteristics over PCL control scaffolds. Culturing mesenchymal stem cells (MSCs) within hybrid scaffolds for two weeks under chondrogenic conditions increased chondrogenic gene expression and proteoglycan deposition compared to PCL controls with a similar scaffold porosity and fiber diameters. The addition of PEG-PNIPAAm within a 3D structured, PCL network produces a hybrid scaffold with the potential for a variety of tissue engineering applications.

▼ Emulsion polymerization technique is one of the most common techniques employed in industry to synthesize a wide range of polymers due to its versatility, the possibility to control the properties of the final product and because it is an environmentally friendly technique. Nevertheless, large amounts of surfactant are usually required to stabilize the system which can have deleterious effects on water resistance and on the mechanical properties of the polymer. Among the different techniques employed to circumvent this problem, recently, graft copolymerization of a monomer from water-soluble polymers containing amino groups has been employed to synthesize polymer particles with amphiphilic core-shell nanostructures. Applications of these particles include controlled release drugs, water-borne coatings and diagnostic testing.
In this project, water-soluble
poly (N-vinylformamide) (PNVF) of different molecular weights was synthesized and characterized. Subsequently, the basic hydrolysis of these polymers was accomplished to obtain
poly (vinyl amine) (PVAm) in an easy and controlled way. The synthesized
poly (vinyl amines) were then employed as stabilizers in the surfactant-free emulsion polymerization of methyl methacrylate (MMA). The MMA monomer polymerizes via two different ways, grafting from polyvinylamine and homopolymerization, leading to a core-shell type particles with polyvinylamine-based shells. The effect of different variables such as initiator type and concentration, the pH of the reaction and the molecular weight of PVAm’s on the conversion, grafting efficiency and morphology of the polymer particles were investigated. Throughout all the work, an extensive characterization of the polymers obtained in each step was performed in order to clarify possible mechanisms involved in this polymerization process. For the characterization of the polymeric particles obtained during this work were used some characterization techniques such as AF4, NMR, DLS, TEM, SEC/GPC and FT-IR.

▼ Deep-eutectic solvents (DES) are considered novel renewable and biodegradable solvents, with a cheap and easy synthesis, without waste production. Later it was discovered a new subclass of DES that even can be biocompatible, since their synthesis uses primary metabolites such as amino acids, organic acids and sugars, from organisms. This subclass was named natural deep-eutectic solvents (NADES).
Due to their properties it was tried to study the interaction between these solvents and biopolymers, in order to produce functionalized fibers for biomedical applications. In this way, fibers were produced by using the electrospinning technique. However, it was first necessary to study some physical properties of NADES, as well as the influence of water in their properties.
It has been concluded that the water has a high influence on NADES properties, which can be seen on the results obtained from the rheology and viscosity studies. The fluid dynamics had changed, as well as the viscosity.
Afterwards, it was tested the viability of using a starch blend. First it was tested the dissolution of these biopolymers into NADES, in order to study the viability of their application in electrospinning. However the results obtained were not satisfactory, since the starch polymers studied did not presented any dissolution in any NADES, or even in organic solvents. In this way it was changed the approach, and it was used other biocompatible polymers.
Poly(ethylene oxide),
poly(vinyl alcohol) and gelatin were the others biopolymers tested for the electrospinning, with NADES. All polymers show good results, since it was possible to obtain fibers. However for gelatin it was used only eutectic mixtures, containing active pharmaceutical ingredients (API’s), instead of NADES. For this case it was used mandelic acid (antimicrobial properties), choline chloride, ibuprofen (anti-inflammatory properties) and menthol (analgesic properties).
The polymers and the produced fibers were characterized by scanning electron microscope (SEM), Transmission electron microscopy (TEM) and Fourier transform infrared spectroscopy (FTIR). With the help of these techniques it was possible to conclude that it was possible to encapsulate NADES within the fibers.
Rheology it was also study for
poly(ethylene oxide) and
poly(vinyl alcohol), in a way to understand the influence of polymer concentration, on the electrospinning technique.
For the gelatin, among the characterization techniques, it was also performed cytotoxicity and drug release studies. The gelatin membranes did not show any toxicity for the cells, since their viability was maintained. Regarding the controlled release profile experiment no conclusion could be drawn from the experiments, due to the rapid and complete dissolution of the gelatin in the buffer solution. However it was possible to quantify the mixture of choline chloride with mandelic acid, allowing thus to complete, and confirm, the information already obtained for the others characterization technique.

In this thesis, we investigate the electrical properties and reliabilities of
poly-Si TFTs for system on panel application. Roughly, we divide the thesis into two parts, n-type and p-type TFTs respectively. In n-type TFT, we mainly study degradation characteristics of TFTs under dynamic stress. On the other hand, we focus on special negative bias temperature instability (NBTI) degradation for p-type
poly-Si TFTs. Because grain boundary in
poly-Si film and serious self-heating effect due to glass substrate, which has a poor thermal conductivity, the electrical properties and reliabilities of
poly-Si TFTs become more complicated, compare with metal-oxide-semiconductor field effect transistor (MOSFET). Therefore, in the thesis, we found some strange phenomena never observed in a-Si TFT and MOSFET.
In chapter 3, the degradation mechanism of n-channel
poly-silicon thin film transistor (
poly-Si TFT) has been investigated at room temperature under dynamic voltage stress, which simulate under high frequency operation as driving devices. The ON-current of TFT is degraded to as low as 0.3 times of the initial value after 1000 second stress. On the other hand, both the sub-threshold swing and threshold voltage kept well during the AC stress. The current crowding effect was rapidly increased with increasing of stress duration. However, comparing the initial and degraded characteristics at rising temperature, namely, 150◦
C, the ON-current of TFT only decrease to 75 percent of the initial value after 1000 second AC stress. It depicts that creation of effective trap density in tail-states of
poly-Si film is responsible for the electrical degradation of
poly-Si TFT. At high temperature, electron has enough energy to pass the energy barrier created by ac stress and the degradation is less obvious.
In chapter 4, the degradation mechanism of n-channel
poly-silicon thin film transistor (
poly-Si TFT) has been investigated under dynamic voltage stress, which simulate under low frequency operation as pixel switches. Surprisingly, two totally different degradations of TFTs were observed after dynamic stress. Firstly field-effect mobility and driving current increased during early stress. However, a clear and rapid degradation of field-effect mobility occurred instead during later stress. Additionally, the threshold voltage of stressed TFTs strangely shifted to negative direction in later stress, which was never observed in early stress. Finally, we clarify the degradation mechanisms for early and later stress respectively by varied temperature experiments.
In chapter 5, the characteristics of p-type
poly-silicon thin film transistor (
poly-Si TFT) with dynamic bias stress were investigated. The AC stress is operated with the constant drain voltage (15V) and the varying gate voltage (0V~-15V) to degrade the devices. There are some phenomena which cannot be completely explained by typical NBTI mechanism in the experiment. La polycaprolactone est l'un des polyesters aliphatiques la plus couramment utilisée pour des applications médicales grâce à sa perméabilité, sa biocompatibilité et sa non-toxicité. Le but de ce travail est de synthétiser et caractériser la polycaprolactone, ce polymère sera ensuite testé pour l'encapsulation de l'acide ascorbique. Nous avons synthétisé ce polymères par polymérisation en solution et en masse .Les deux échantillons obtenus ont été caractérisés par différentes méthodes d'analyses telles que : la résonance magnétique nucléaire RMN 1H, la spectroscopie infrarouge IR, l'analyse calorimétrique différentielle DSC, Le microscope à balayage électronique MEB, et la rhéologie. L'encapsulation de l'acide ascorbique par les deux échantillons de la polycaprolactone a été effectuée par la méthode d'encapsulation par évaporation du solvant. L'ensemble des microcapsules obtenues ont été caractérisées par la spectroscopie IR, le MEB, et la taille des microcapsules a été déterminée par l'essai de granulométrie

▼ The liquid argon target of the DEAP-3600 dark matter detector is contained by an extremely radiopure acrylic vessel. Alpha decays from the inner surface of the acrylic vessel are a source of background. If a fraction of the alpha energy is observed, or if the recoiling nucleus from the alpha decay is observed, the event will not be separated from a dark matter candidate event. In addition to the low level of inherent contamination from uranium and thorium, the Pb-210 from Rn-222 diffusion during manufacturing must be measured. The limit for the DEAP-3600 acrylic vessel is 1.1 × 10^−20 g/g Pb-210. By vaporizing a large quantity of acrylic and counting the concentrated residue with an ultralow background HPGe well detector and a low background alpha spectrometer, the bulk acrylic was found to have an upper limit of 10^−19 g/g Pb-210. The design, installation, commissioning, operation, and analysis for various aspects of the acrylic assay are described.

poly(ADP-ribose) polymerases consistant en une
population hétérogène de polymères formés à partir du NAD. La
poly(ADP-ribose) glycohydrolase est responsable de la dégradation
du
poly(ADP-ribose). Les activités enzymatiques de ces enzymes
constituent un système de régulation pour différents sentiers
métaboliques. En effet, l'interaction démontrée entre le pADPr et
de multiples protéines a permis de confirmer un rôle de modulateur
de nombreuses voies de signalisation tel que la réparation de
l'ADN, apoptose, cycle cellulaire, surveillance de l'intégrité du
génome, transcription et modulation de la chromatine. Ainsi, nous
avons formulé l'hypothèse que le pADPr pourrait coordonner la
réparation des lésions à l'ADN et la progression du cycle
cellulaire avec la signalisation d'événement apoptotiques. Une
approche efficace constituerait à identifier et caractériser les
protéines (intermédiaires) associées au pADPr selon une logique
temporelle. Les diverses actions du pADPr sur les processus
biologiques dépendent (dans le cas de la majorité du pADPr, soit
celui métabolisé par PARP-1) de la gravité des dommages induits à
l'ADN. Par conséquent, il existe probablement des points de seuil
faisant basculer les voies de signalisation de la réparation vers
la mort cellulaire. Une approche réductionniste, dans ce type de
problème, ne peut apporter des réponses satisfaisantes.
L'utilisation de la protéomique quantitative semble être une
approche plus appropriée. Un volet du travail présenté dans cette
thèse visait à identifier des partenaires des PARP-1, PARP-2 et
PARG dans le but de reconnaître les sentiers biochimiques qui
pourraient inclure une composante de
poly(ADP-ribosylation) dans
leur régulation et définir des interactions fonctionnellement
pertinentes. Par la suite, nous avons établi un réseau dynamique
des complexes associés au pADPr en fonction du temps suivant un
dommage alkylant induit par un stress génotoxique. Ainsi, certains
événements modulés par le pADPr ont été analysés et cartographies.
De plus, nous avons caractérisé un rôle novateur du pADPr dans la
formation des granules de stress suite à un stress génotoxique. En
conséquence, nos résultats ont permis d'édifier les premières bases
pour la biologie des systèmes de la
poly(ADP-ribosyl)ation en
fournissant un répertoire d'interactions protéique
exhaustif.

▼ La compréhension du comportement des actinides dans l’environnement est un enjeu important et très complexe en raison des nombreux phénomènes intervenant dans les processus chimiques. Parmi eux, nous pouvons citer la complexation, notamment via les fonctions carboxylates issues des substances humiques présentes dans les sols. En milieu aqueux, les actinides au degré d’oxydation IV sont très sensibles à l’hydrolyse impactant alors leur mobilité. Au cours de ce travail de thèse, nous avons étudié la réactivité de ces actinides(IV) (Th, U et Np) en présence de ligands organiques de type carboxylate aromatique, afin d’enrichir nos connaissances sur les entités formées. Plus de quarante nouveaux composés à base d’actinides(IV) et d’acides polycarboxyliques ont été synthétisés. Leurs structures cristallographiques ont été déterminées par DRX sur monocristal et leurs descriptions sont rapportées. Le contrôle de l’hydrolyse de ces métaux au sein de nos composés a été effectué par une variation de l’acidité en milieu aqueux ou par ajout contrôlé de quantités d’eau dans des solvants organiques. Certains des composés présentés sont construits autour d’assemblages polynucléaires possédant entre 2 et 38 centres métalliques. Pour ces derniers ({U38} et {Np38}), une nouvelle voie de synthèse a été investiguée. Au cours de l’étude de ces différents composés, nous avons mis en évidence les analogies ou les différences existant entre ces actinides, et avons enrichis notamment nos connaissances sur la cristallochimie du neptunium(IV). Enfin, nous avons montré la capacité de ligands N- et O- donneurs à complexer simultanément l’uranium(IV) avec d’autres métaux di- (Mn) ou trivalent (Ln).
Understanding the behavior of actinides in the environment is an important and very complex issue due to phenomena involved in chemical processes. Among them, one can mention the complexation, via carboxylate functions from the humic substances present in the soils. In an aqueous medium, actinides at the oxidation state IV are very sensitive to the hydrolysis process impacting their mobility. This thesis focuses the reactivity of these actinides (IV) (Th, U and Np) in the presence of organic ligands with aromatic carboxylate functions, in order to improve our knowledge about entities formed and linked to such organic linkers.More than forty new compounds based on actinides (IV) and polycarboxylic acids have been synthesized. Their crystallographic structures were determined by single crystal XRD and their descriptions are reported. The control of the hydrolysis of these metals within our compounds has been managed by varying the acidity in aqueous medium or by addition of controlled amount of water in organic solvents. ▼ Building upon existing synthetic methods, we have optimized the synthesis of Thiele’s methyl ester to an efficient and scalable methodology. As part of a study of chemo- and regioselective transformations within the Thiele’s ester scaffold, we designed and synthesized a new suite of molecular scaffolds incorporating a broad range (from 123° to 176°) of cleft angles. In addition to this, we compared two competing conceptual models for their ability to rationalize the selective formation of Thiele’s ester and two minor regioisomers which arise during the formation of the target product. We found that radical stabilization arguments (based on Deslongchamps’ seminal work) outperformed the classic frontier molecular orbital theory model in predicting the regioselectivity of Thiele’s ester dimerization. When this method was combined with simple steric arguments, we arrived at a general algorithm to rationalize Thiele type dimerization, including all the known homo- and heterodimerizations in the literature as well as a novel phosphine oxide-containing Thiele acid analogue discovered as part of this thesis work. In order to stimulate the use of Thiele’s ester chemistry in a diverse range of applications, we took advantage of our Thiele’s ester methodology to achieve a mono ester-substituted dicyclopentadiene (colloquially referred to as a “half” Thiele’s ester), and used this as the precursor of a novel functionalized polydicyclopentadiene (fPDCPD) ROMP polymer. The resulting fPDCPD has the highest glass-transition temperature reported for any polydicyclopentadiene material and allows for the facile manipulation of the surface chemistry through alteration of the embedded functional group. A long-term goal in the Wulff lab is to use Thiele’s ester as a scaffold for the generation of conformationally restricted (“peramivir-like”) neuraminidase inhibitors. Setting the groundwork for this, we explored the selectivity of various peramivir derivatives toward group-1 vs. group-2 neuraminidase enzymes. To this end, we coupled a wide range of alkyl chains and aromatic rings with different length and size parameters onto the primary amine of peramivir. We found that our de-guanidinylated peramivir analogues showed a rare target selectivity against group-2 neuraminidases instead of group-1 neuraminidases, which might due to the ring geometry of peramivir as well as the reduced electrostatic interaction between the amino group from our analogues and the Asp147-His150 residues from the enzyme. This suggested that it is possible for group-2 neuraminidases to have a more open 150-cavity state than group-1 neuraminidases. Additionally, the respectable IC50 values for these compounds, together with their significantly reduced polarity (relative to peramivir itself) may prove advantageous from a bioavailability standpoint.
