▼ The identification of autoantibodies is a primary diagnostic marker for the diagnosis of some autoimmune diseases. The sera of patients with connective tissue diseases commonly contain autoantibodies that target nuclear antigens. As such these antibodies are called antinuclear antibodies (ANAs). Furthermore, the clinical identification of ANAs in patient sera to specific nuclear antigens is a primary tool for the diagnosis of connective tissue diseases. For this purpose specific extractable nuclear antigens (ENAs) are used. The most commonly employed laboratory technique for the detection of ENA antibodies is the enzyme linked immunoabsorbant assay (ELISA). ELISA is a simple technique that can be automated and provides high diagnostic sensitivity for the detection of ENA antibodies comparatively to a counter immunoelectrophoresis (CIE) assay. However, compared to CIE ELISA lacks diagnostic specificity. Therefore there is a demand for an assay with high diagnostic specificity as a secondary diagnostic test for the detection of ENA antibodies. The central aim of this project was to develop and validate a novel CIE assay that used fluorescently labelled ENAs to detect ENA antibodies in patient serum. Development of the assay began with comparative testing of fluorescently labelled and unlabelled antigens to confirm that the immunochemical properties of the antigen remained intact. The CIE assay conditions were optimised for the detection of four ANAs SSA, SSB, RNP/Sm, and Sm using their corresponding ENAs. Assay conditions optimised were flurophore type, gel composition, buffer composition, antigen concentration, the running time, and analytical specificity. Evaluation of 281 clinical sera samples known to have previously tested positive test by ANA indirect immunofluorescence were performed using the optimised CIE assay and ELISA. The inter-rater agreement between the CIE assay and ELISA results was determined. Clinical details were used to retrospectively classify the clinical sera samples into clinical categories and case groups. This data was used for the diagnostic comparison of the CIE assay and ELISA results. There was strong inter-rater agreement between the SSA CIE assay and ELISA results. The diagnostic specificity and positive likelihood ratio values of the SSA CIE assay were superior to those of the ELISA, while diagnostic sensitivity and the negative likelihood ratio values were approximately the same. There was strong inter-rater agreement between the RNP/Sm CIE assay and ELISA. However, it was observed that the statistical measures of assay accuracy (diagnostic specificity and sensitivity, positive likelihood ratio, and negative likelihood ratio) for the RNP/Sm ELISA were superior to those of CIE assay. The diagnostic specificity of the SSB and Sm CIE assays were higher than that of their respective ELISAs. This was also true for the positive likelihood ratio values of the SSB and Sm CIE assays when compared to their respect ELISAs. ▼ In 2003 the National Endowment for the Arts (NEA) created the Shakespeare in American Communities initiative. The purpose of this initiative is to introduce participants to the plays of Shakespeare through live productions and educational workshops. Each year, selected theatre companies receive a matching grant through this program that enables them to create educational activities and performances that are then provided free or at low cost for communities that have traditionally been considered under-served by arts programs. In an introduction to the Shakespeare in American Communities initiative published on the NEA's website in 2007, former NEA Chairman Dana Gioia wrote, "In order to understand American culture or American theater, one must first understand Shakespeare." It is this provocative statement that provides the impetus for the primary research questions of this dissertation. What did the NEA under the chairmanship of Dana Gioia believe that Shakespeare could and should teach citizens about American culture? How did the NEA's marketing of the Shakespeare in American Communities initiative serve to establish a branded product that could improve the reputation of the NEA following the battering it had taken during the culture wars of the 1980s and 1990s? Through interviews with key participants in the initiative, including Chairman Gioia, analysis of the NEA's promotional literature for the program, data compiled from the self-reports of participating theatre companies, and analysis of the media's response to this and other NEA projects, this study provides both a detailed history of the Shakespeare in American Communities initiative as well as a consideration of its position and importance within the overall narrative of the history of the Endowment. This study also includes an analysis of Gioia's understanding and use of the term "culture" and the manner in which that understanding influenced the goals of the NEA under his leadership. This detailed analysis of the Shakespeare in American Communities initiative provides not only an overview of a major Shakespeare performance-based educational program, but also an explanation of how the program was strategically branded and marketed to improve the reputation and status of the NEA at the turn of the twenty-first century. ▼ In this study, we used multi-phosphor-doped sodium glass (MPDG) as color conversion layer, and successfully developed glass phosphor with high color rendering index (CRI) and high reliability. We study the optical properties and microstructure of MPDG in this experiment. We used yellow phosphor of YAG based (Y3AlO12 : Ce3+), green phosphor of LuAG based (Lu3Al5O12 : Ce3+), and red phosphor of nitride based (CaAlClSiN3 : Eu2+) sintering to be glass phosphor at 680â, 700â and 750â, respectively. The study indicates that the yellow and green glass phosphor can keep the same quantum yield (QY) at different sintering temperature. However, the QY of red glass phosphor is significantly reduced with rising temperature. The reason of the lowering QY of red glass phosphor can be explained by microstructure analysis when sintered at high temperature, the low temperature glass will diffuse into the phosphor crystal. Field-Emission Transmission Electron Microscope (FE-TEM) clearly shows that the higher sintering temperature induces thicker diffusion layer and more diffusion into phosphor crystal. Element analysis by Energy Dispersive System (EDS) reveals that, for red glass phosphor, higher percentage of carbon atoms in the diffusion layer under high sintering temperature, indicating more carbide was produced at higher temperature in the mixture of nitride phosphor and glass. The carbide severely lowered the quantum yield of the red glass phosphors. Employing multi-phosphor (yellow, green, red) with doping concentration 0:4:1, and sintered at 680â. Using a blue LED 445nm of wavelength as pumping source. The QY reaches 51%. The Color Coordinator and Correlated Color Temperature (CCT) of white light emitting diode (WLED) with them as color conversion layer is (0.317, 0.315ï¼6336K). The CRI is up to 90, which meets the regulations of U.S. Energy Star that the CRI for interior lighting must exceed 80. The luminescence loss of glass phosphor is 5 to 10 space times lower than the traditional silicone phosphor under the 1008 hours reliability test under 150â and 250â, respectively, which has great potential in lighting system. Este trabalho apresenta os resultados da síntese e caracterização de nanoestruturas de óxido de tungstênio (WO3) obtidos por via ultrassônica em meio não aquoso. Esta técnica tem como vantagens o uso de pouco material e o tempo empregado. A caracterização das nanoestruturas foi realizada por Microscopia Eletrônica de Transmissão (TEM), Microscopia Eletrônica de varredura (MEV), espectroscopia Raman e no infravermelho, termogravimetria e difração de raios-X. A imobilização das nanoestruturas foi realizada em substratos condutores transparentes (ITO) por automontagem camadas por camadas (LbL) e deposição eletroforética (EPD). Foram efetuadas caracterizações eletroquímicas e espectroeletroquímicas com medidas in situ usando UV-Vis. Os resultados eletrocrômicos demostraram a estrutura nanométrica dos filmes obtidos, com parâmetros eletrocrômicos como contraste óptico, tempo de resposta e eficiência eletrocrômica típicos de sistemas nanoestruturados. Alguns aspectos relacionados com o uso de um líquido iônico prótico (PIL) foram mostrados, conferindo melhora na durabilidade dos filmes em relação ao eletrólito H2SO4 1,0 mol L-1. Com o objetivo de modular e incrementar a coloração dos filmes de WO3, foi preparado um viológeno para imobilização superficial através de um grupo ancorador. Os filmes modificados foram investigados por espectroeletroquímica e coordenadas de cor. As coordenadas de cromaticidade foram obtidas por um colorímetro virtual baseado na convenção CIE 1931, dados de cromaticidade para as reações eletrocrômicas dos filmes de WO3 modificado são mostrados em espaços de cor.

The present work describes the synthesis and characterization of WO3 nanoplates in a non-aqueous solvent using ultrasonic irradiation process. This method presents some advantages such as the small amount of reactants and time consumed. WO3 nanoplates have been characterized by TEM and SEM microscopies, Raman and Infrared spectroscopies, thermogravimetric analysis and X-ray diffraction. The nanostructured nanoplates were immobilized by electrostatic layer by layer and Electrophoretic Deposition (EPD) processes onto ITO substrates. Spectroelectrochemical experiments were carried out showing electrochromic performance of the WO3 films such as optical density, response time and chromatic efficiency in a typical way for nanostructured films of WO3. The results using a protic ionic liquid (PIL) as electrolyte, instead of H2SO4, showed improvement in cyclic durability. Cyclic voltammograms were combined with transmittance measurements for all films that undergo typical reversible electrochromic reaction. In order to modulate and improve the colorimetric changes, viologen molecules were prepared with an anchoring group for modifying the surface by the chemisorption of a monolayer. ▼ Teachers during the Baby Boom era (1946-1964) faced mounting pressures to be Cold War role models to the rapidly expanding population of students. Politicians, education scholars, school administrators, and community leaders considered teachers a central component to Cold War success. K-12 teachers were charged with producing new generations of cold warriors ready to defeat communism abroad and overcome social problems in America. Throughout the Baby Boom era, teachers were represented by others and presented themselves as indispensable agents for maintaining and furthering the United States’ Cold War goals and American ideals. As the rhetoric about the centrality of educators to winning the Cold War escalated, teachers gained access to powerful political and cultural discourses to redefine teaching as a serious professional career. American Federation of Teachers (AFT) and National Education Association (NEA) used the discursive space around Americanism, consumerism, democracy, and world leadership to represent teachers as worthy professionals. In an effort to gain public recognition for teachers as professionals, the AFT and NEA attached each of these discourses to specific elements of professionalism in American culture, including occupational autonomy, middle class compensation, public service, and technical expertise. The NEA from its origins in 1857 until the late 1950s adopted a top-down approach to professionalism. NEA leaders argued that the best way to elevate teaching was to advance public education. From its inception in 1916, the AFT adopted a bottom-up approach to teacher professionalism. AFT leaders argued that by advancing teachers’ employment status and developing their political voice then the union would increase the teaching profession’s status in the eyes of administrators, politicians, the media, and the American public. The AFT’s and NEA’s discursive efforts during the Baby Boom era were important factors in the claim and confirmation of teacher professionalism. Together, these organizations utilized prominent Cold War discourses to construct a recognized professional identity for teachers. By 1965, teaching attained specific professional traits including the ability to privilege teaching as a vital public service, to achieve a middle class standard of living for teachers, to regulate the conduct of its members, and to refine the technical expertise required of its practitioners. ▼ The C57BL/6 (B6) mouse is the most commonly used inbred strain in biomedical research, and has served as the basis for various large-scale genetic and genomic projects. Although the parental substrain, C57BL/6J (B6J), originated at The Jackson Laboratory, isolated breeding colonies are now maintained at numerous sites. This separation has resulted in genetic drift that has led to the emergence of phenotypic differences among these colonies. For instance, B6J mice display higher levels of voluntary ethanol consumption and increased locomotor responses to psychostimulants, relative to C57BL/6N mice (B6N). Initial progress has been made in elucidating the genetic bases of these phenotypic differences, as Kumar et al. (2013) identified a single nucleotide polymorphism (SNP) in the Cytoplasmic FMR1-interacting protein 2 (Cyfip2) gene that underlies the differential locomotor response to cocaine exhibited by B6J and B6N mice. The present study compared voluntary ethanol consumption, binge-like drinking, and affective behavior during forced abstinence in B6J and C57BL/6NJ (B6NJ) substrains, and when substrain differences were seen, in CRISPR-Cas9 engineered lines in which the previously identified substrain-specific Cyfip2 SNP was exchanged within the same genetic backgrounds. Results showed that B6J consumed significantly greater quantities of ethanol than B6NJ mice and allelic variation in Cyfip2 greatly contributed to observed substrain differences in two-bottle free-choice ethanol drinking; however, since these differences were completely reversed in males, but not females, it’s likely that sex-specific contributions from other polymorphisms play a role in moderating these effects. Interestingly, there were no significant substrain differences in binge-like drinking or depressive- or anxiety-like behaviors during abstinence. Overall, while B6J and B6NJ mice displayed dramatic differences in voluntary ethanol consumption, which were dependent in part on an identified cocaine-relevant SNP in Cyfip2, no substrain differences were seen in binge- like drinking or affective behavior during abstinence. Die Betriebszeit solcher Quellen wird durch die Abnahme der Quantenausbeute der Photokathode mit der Zeit begrenzt. Die Problematik der Abnahme der Quantenausbeute konnte durch die Reaktion der Kathodenoberfläche mit sauerstoffhaltigen Molekülen sowie durch Ionenbombardement geklärt werden. Im Laufe dieser Arbeit wurden, teilweise zum ersten Mal, Mechanismen untersucht, die zur Entstehung der chemisch aktiven Moleküle und der Ionen beitragen und weitere Effekte, die die Betriebszeit der polarisierten Quellen reduzieren. Die Experimente wurden an einer genauen Kopie der an MAMI vorhandenen polarisierten Quelle durchgeführt. Es wurde demonstriert, dass Erwärmung der Photokathode, Ioneneinfang und Strahlverlust aufgrund der Raumladungskräfte die Kathodenlebensdauer begrenzen können. Der erste Effekt ist Erwärmung der Photokathode. Die Laserleistung wird fast vollständig in Wärmeleistung umgesetzt, was zur Absenkung der Verfügbarkeit der polarisierten Quellen führen kann, und zwar unabhängig davon, ob der Photostrom produziert wird oder nicht. Der zweite Effekt ist Ionenbombardement mit den sowohl in der Beschleunigungsstrecke als auch in der Strahlführung entstehenden Ionen. Es wurde demonstriert, dass der in der Strahlführung entstehende Ionenstrom sogar größer ist als der in der Kanone. Unter bestimmten Bedingungen können die gebildeten Ionen durch das Potenzial des Elektronenstrahls eingefangen werden und die Kanone erreichen und damit zusätzlich zur Zerstörung der negativen Elektronenaffinität beitragen. Der dritte Effekt ist Strahlverlust. Es wurde demonstriert, dass die relativen Strahlverluste kleiner als 1*10-6 sein sollten, um eine Lebensdauer von mehr als 1000 Stunden beim Strom von 100 A zu erreichen, was für die vorhandene Apparatur möglich ist. Zur Erzeugung extrem hoher Ströme wurde zum ersten Mal im Bereich der spinpolarisierten Quellen das Prinzip der „Energierückgewinnung“ eingesetzt. Experimente bei einer mittleren Stromstärke von 11.4 mA und einer Spitzenstromstärke von 57 mA bei 1% Tastverhältnis wurden bereits durchgeführt.

The production of a polarized electron beam with high average current is important for operation of existing polarized sources as well as for future projects. The operation lifetime of the polarized electron sources is limited through decrease of the quantum efficiency. It was found that two mechanisms limit the lifetime of the negative electron affinity photocathode, which are ion backbombardment (created by the ionisation of the residual gas) and chemical poisoning of the photocathode surface by H2O and CO2 gases. O objetivo deste estudo foi avaliar a efic?cia e efeitos toxicol?gicos da associa??o entre a Crotamina (CTA) de Crotalus durissus terrificus e f?rmacos cl?ssicos empregados na terapia da leishmaniose. Estudo preliminar realizado identificou a melhor combina??o entre a CTA e Glucantime? (GLU) ou Pentamidina (PEN) contra as formas promastigotas de L. amazonensis. Doravante, as formula??es com GLU avaliadas foram denominadas de G1 (GLU 300 ?g ?g/mL + CTA 100 ?g/mL) e G2 (GLU 300 ?g ?g/mL + CTA 3,125 ?g ?g/mL) e com PEN denominadas de P1 (PEN 0,05 ?g ?g/mL + CTA 100 ?g ?g/mL) e P2 (PEN 0,05 ?g ?g/mL + CTA 3,125 ?g ?g/mL). In vitro, os tratamentos foram avaliados sobre a inibi??o do crescimento de promastigotas, amastigotas, produ??o de ?xido N?trico, citocinas e citotoxicidade dos tratamentos sobre c?lulas de linhagem. Foram analisadas tamb?m a afinidade molecular e a caracteriza??o molecular da CTA e GLU ou PEN. In vivo, os grupos experimentais foram denominados G5 (GLU 300 mg.kg- 1 + CTA 5 mg.kg-1) e G6 (PEN 4 mg.kg-1 + CTA 5 mg.kg-1) e os tratamentos foram realizados por oito dias alternados. Ap?s 48h do fim do tratamento os animais foram mortos e as patas foram retiradas na primeira articula??o acima da les?o para pesagem. Em seguida, foi realizada a retirada de sangue, tecido da pata, linfonodo, rins, f?gado e ba?o para posteriores experimentos. O sangue de cada animal foi centrifugado e o soro separado para an?lise bioqu?mica e imunol?gica. ▼ The black pericarp trait in grain sorghum [Sorghum bicolor (L.) Moench] is a novel trait with complex inheritance. In addition to its uniform, dark appearance, black sorghum grain contains high levels of favorable phenolic compounds such as 3-deoxyanthocyanidins (3-DOA) and condensed tannins which have applications in the specialty food industry as high-antioxidant food additives, natural food colorants, or natural food preservatives. Previous studies have indicated the trait is not fully penetrant in all environmental conditions. Additionally, black sorghum has acceptable agronomic performance, but is significantly lower yielding than other elite grain sorghum hybrids. Further improvement of black sorghum is dependent on understanding the factors—both genetic and environmental—influencing the expression of this trait. The first of two studies investigated the effect of light shading on grain color and grain composition in black Tx3362. Increased light shading reduced, and in some cases, eliminated the black color resulting in red grain production. In addition, increased shading reduced the concentration of 3-deoxyanthocyanidins, total phenols, tannins, and fiber while increasing fat concentrations. Thus the black pericarp trait and associated high phenolic concentrations are strongly influenced by both intensity and duration of sunlight exposure. In the second study, a generation means analysis was performed to determine the genetic factors affecting the trait. This study concluded grain color and associated grain composition traits were influence by additive, dominance, and epistatic effects. The generation means analysis also determined the black pericarp trait is recessive, controlled by multiple genes, and is moderate to highly heritable. Despite these challenges, there is enough variation in breeding populations between red and black parents for further improvement of the trait. Creation of high yielding hybrids with uniformly dark grain and high levels of phenolic compounds will be possible through standard plant breeding practices. ▼ The different stages of technological evolution are characterized in terms of the dominant type of tools used in each of them. We passed through the `stone age', the `copper age' and the `iron age' and now we are probably in the `electron age'. After having done a wonderful job, today it looks as if electronic technologies have started experiencing their limits. For example, today's `information explosion' and communication problems demand very large bandwidths without cross talk, larger than that electronics can provide. What could be the technology that can tackle the growing needs of tomorrow? Photonics is being studied today as a possible alternate technology for the future. Investigations are on to harness the capability of the photon to carry information and energy. Fortunately, success has already been achieved in the area of communications. Optical fibers, not copper cables, are already being used to carry huge amounts of information across the great oceans. However, among the three “C”s (communication, computing and control), communication is the only one that is currently implying photonic devices. Photonic switches and optical computers are still in the laboratories and not yet in the marketplace. The major bottleneck in this area is not in solving technological problems or in perfecting theoretical understanding, but in developing suitable materials. In this field, materials with non linear optical (NLO) properties seem to be extremely appealing in fact NLO effects allow the manipulation of laser light beams, extending the frequency range provided by normal laser sources into the ultraviolet and infrared regions, and may thus form the basis of all-optical computing devices. In order to be implied in a device, the NLO materials must satisfy some requirement such as good NLO properties, high laser damage threshold, fast optical response time, wide phase matchable angle, architectural flexibility for molecular design and morphology, ability to process into crystals and thin films, optical transparency (no absorption at fundamental and SH wavelengths), synthetically facile, environmental friendly and high mechanical strength and thermal stability. During this three-year research the attention was to the synthesis and the investigation of the second harmonic NLO properties of new organic and inorganic-organic hybrid material. NLO characterization has been performed in solution using the EFISH technique, the Kurtz-Perry method has been implied for the measurements in the solid state while the Maker’s fringe technique has been use for measurements on film. ▼ Increased alcohol consumption over time is one of the characteristic symptoms of Alcohol Use Disorder (AUD). The molecular mechanisms underlying this escalation in intake is still the subject of study. However, the mesocortical and mesolimbic dopamine pathways, and the extended amygdala, because of their involvement in reward and reinforcement are believed to play key roles in these behavioral changes. Multiple gene expression studies have shown that alcohol affects the expression of thousands of genes in the brain. The studies discussed in this document use the systems biology technique of co-expression network analysis to attempt to find patterns within genome-wide expression data from two animal models of chronic, high-dose ethanol exposure. These analyses have identified time-dependent and brain-regions specific patterns of expression in C57Bl/6J mice after multiple exposures to intoxicating doses of ethanol and withdrawal. Specifically, they have identified the PFC and HPC as showing long-term ethanol regulation, and identified Let-7 family miRNAs as potential gene expression regulators of chronic ethanol response. Network analysis also indicates neurotransmitter release and neuroimmune response are very correlated to ethanol intake in chronically exposed mice. Examining gene expression response to chronic ethanol exposure across a variable genetic background revealed that, although gene expression response may show conserved patterns, underlying differences in gene expression influence by genetic background may be what truly underlies voluntary ethanol consumption. Finally, combined network analysis of gene expression in the prefrontal cortex (PFC) of mice and macaques following prolonged ethanol exposure demonstrated that neurotransmission, myelination, transcription, cellular respiration, and, possibly, neurovasculature are affected by chronic ethanol across species. Taken together, these studies generate several new hypothesis and areas of future research into the continued study of druggable targets for AUD. A presente dissertação se dedica a refletir sobre o corpo e o grotesco na arte e na comunicação contemporâneas. O objetivo do estudo foi investigar o grotesco na cena da dança contemporânea com o intuito de compreender como se dá a relação entre o modo de apreensão do grotesco na atualidade e o modo como ele se apresenta cenicamente. Assim, procurou-se investigar que representações da cultura contemporânea podem estar associadas às manifestações do fenômeno grotesco; como a mídia e o consumo de massa participam na disseminação desta estética; e como o corpo cênico na dança se aproxima/reflete estas representações presentes no cotidiano. O fenômeno grotesco, objeto de estudo desta pesquisa, foi investigado em duas companhias de dança: a Lia Rodrigues Companhia de Danças (RJ) e o Grupo Cena 11 de Dança (SC), onde foram observados o tratamento artístico e os dispositivos utilizados para construção do efeito corpo-grotesco nos espetáculos e o posicionamento dos artistas com o uso da estética grotesca nas obras. O embasamento teórico do estudo investiu na compreensão do fenômeno grotesco nos âmbitos histórico, estético/artístico e social e sua contextualização na contemporaneidade. Partiu-se do entendimento de que o corpo é mídia primária, "espaço" de comunicação e de linguagem e a dança cênica é um fenômeno urbano, que espelha as transformações representacionais do início deste século XXI. As abordagens teóricas de Mikhail Bakhtin sobre o realismo grotesco; de Wolfgang Kayser, sobre o grotesco romântico e os estudos de Muniz Sodré e Raquel Paiva, sobre o grotesco na comunicação contemporânea fundamentaram as discussões. Foi relevante refletir sobre as mudanças de paradigmas sociais que vêm se desenhando na atualidade, sobretudo com a influência do desenvolvimento tecnológico e midiático sobre as corporeidades e os processos de subjetivação. As qualidades fluidas e polifônicas associadas à contemporaneidade nortearam o entendimento desses novos paradigmas estéticos do corpo - atribuídos como corporeidades móveis contemporâneas - e, serviram para denominar/compreender como "grotesco líquido" a presença múltipla e volátil dos fenômenos grotescos na atualidade.

▼ This concept of calculating, optimizing, and utilizing a trajectory known as a ``Free Return Trajectory" to facilitate spacecraft rendezvous with Near-Earth Asteroids is presented in this dissertation. A Free Return Trajectory may be defined as a trajectory that begins and ends near the same point, relative to some central body, without performing any deterministic velocity maneuvers (i.e., no maneuvers are planned in a theoretical sense for the nominal mission to proceed). Free Return Trajectories have been utilized previously for other purposes in astrodynamics, but they have not been previously applied to the problem of Near-Earth Asteroid rendezvous. Presented here is a series of descriptions, algorithms, and results related to trajectory initial guess calculation and optimal trajectory convergence. First, Earth-centered Free Return Trajectories are described in a general manner, and these trajectories are classified into several families based on common characteristics. Next, these trajectories are used to automatically generate initial conditions in the three-body problem for the purpose of Near-Earth Asteroid rendezvous. For several bodies of interest, example initial conditions are automatically generated, and are subsequently converged, resulting in feasible, locally-optimal, round-trip trajectories to Near-Earth Asteroids utilizing Free Return Trajectories. Subsequently, a study is performed on using an unpowered flyby of the Moon to lower the overall DV cost for a nominal round-trip voyage to a Near-Earth Asteroid. Using the Moon is shown to appreciably decrease the overall mission cost. In creating the formulation and algorithms for the Lunar flyby problem, an initial guess routine for generic planetary and lunar flyby tours was developed. This continuation algorithm is presented next, and details a novel process by which ballistic trajectories in a simplistic two-body force model may be iteratively converged in progressively more realistic dynamical models until a final converged ballistic trajectory is found in a full-ephemeris, full-dynamics model. This procedure is useful for constructing interplanetary transfers and moon tours in a realistic dynamical framework; an interplanetary and an inter-moon example are both shown. To summarize, the material in this dissertation consists of: novel algorithms to compute Free Return Trajectories, and application of the concept to Near-Earth Asteroid rendezvous; demonstration of cost-savings by using a Lunar flyby; and a novel routine to transfer trajectories from a simplistic model to a more realistic dynamical representation.
