▼ Introduction The combination of metformin and Dipeptidyl Peptidase 4 inhibitors has superior health outcomes as compared to metformin alone. However, the cost may be prohibitive and the combination is being considered as a replacement of metformin alone as first line therapy for management of Type 2 diabetes. The comparative cost effectiveness of either treatment is unknown. Objectives To compare the cost effectiveness of changing the first line therapy in the treatment of Type 2 diabetes mellitus from metformin monotherapy to dual therapy of metformin and Dipeptidyl Peptidase 4 inhibitors in drug naïve patients. Methodology The study was divided into three parts. The first part was a local and international price survey on the prices of metformin and Dipeptidyl Peptidase 4 inhibitors. The second part, a key informant interview with staff at Kenyatta National Hospital to identify the key resource input required in the management of diabetic nephropathy and lastly a Markov chain model is developed to obtain the long term cost and effectiveness of treating type 2 diabetes patients on either metformin monotherapy or metformin/Dipeptidyl Peptidase 4 inhibitor dual therapies. The design was a predictive model based cost effectiveness study. The comparator interventions were metformin monotherapy and metformin/Dipeptidyl Peptidase 4 inhibitor dual therapy. Costing was done from the perspective of the provider and only health care costs were considered. The time horizon was 25 years. A macro-ingredient approach was considered for costing. Effectiveness data was obtained from literature and the measures of effectiveness was the life expectancy and time to development of diabetic nephropathy. A sensitivity analysis was used to determine how variation in the costs of the different therapies affected the overall cost effectiveness ratio. 10 Results The factors affecting the price of metformin andDipeptidyl Peptidase inhibitors were found to be the pack size of the drug (P<0.05) and the country of importation of the drug (P<0.05). The median local price of the daily defined dose of metformin was KShKSh22.8 (International United States Dollar (IUD) 0.48). The median price ratio of the international median price and the local median price of metformin 500mg tablets and 850mg was 1.3 and 8.4 respectively. The median local price of a daily defined dose of Dipeptidyl Peptidase 4 inhibitors was KShKSh 58 (IUD1.22) while that of Fixed Dose Combination of metformin and Dipeptidyl Peptidase 4 inhibitors was KShKSh122 (IUD 2.58). The annual cost of dialysis treatment, with the perspective of the health provider, at Kenyatta National Hospital was found to be KShKSh 1,871,640 (IUD 39,678). The annual cost of treating microalbuminuria and macroalbuminuria was KShKSh 174,360 (IUD 3696) and KShKSh 251,160 (IUD 5324) respectively. O objetivo deste trabalho foi estudar a atividade antioxidante de um extrato padronizado da raiz de Pothomorphe umbellata L. Miq, incorporado em diadermina (creme óleo em água - O/A), sobre a pele de camundongos Hairless e avaliar esta atividade antioxidante em comparação à atividade antioxidante do α-tocoferol, incorporado na mesma base. A peroxidação lipídica foi avaliada como medida de TBARS e de quimiluminescência. Animais tratados com o extrato de Pothomorphe umbellata demonstraram um decréscimo significativo nos parâmetros de peroxidação lipídica, em concentrações de até 0,1% p/p de 4-nerolidilcatecol (o que equivale a uma concentração de 4% p/p de extrato) incorporado na diadermina. Em nosso modelo o decréscimo significativo destes indicadores, nos animais tratados com α-tocoferol, só foi observado em concentrações superiores a 5%. Os resultados indicam uma potente atividade antioxidante do extrato da raiz de Pothomorphe umbellata, sugerindo sua aplicação em formulações cosméticas.

Antioxidants may play a significant role in ameliorating or preventing oxidative damage to skin. In this work antioxidant activity of P. umbellata root extract, with known concentration of 4-nerolidylcathecol, incorporated to a hydrophilic ointment in different concentrations, was evaluated on skin of Hairless mice. This antioxidant activity was compared with that of different concentrations of α-tocopherol (Sigma) incorporated in the same vehicle. Lipid peroxidation of skin homogenates was evaluated as thiobarbituric acid reactants (TBARS) and chemiluminescense (CL). Animals treated with P. umbellata root extract showed a decrease up to 90% in the lipid peroxidation parameters, for concentrations higher than 0.1%. Our model demonstrated a significant decrease in TBARS an CL only for higher concentrations of α-tocopherol (5 and 10% w/w). The results indicate a potent antioxidant activity of the Pothomorphe umbellata root extract, suggesting its application in cosmetic formulations.

▼ Red maple (Acer rubrum), also known as swamp, water or soft maple, is endemic to eastern and central North America and was widely used as traditional medicine by the first peoples. Commercially, its well-known products include maple syrup and high-quality lumber. The potential medicinal benefits of phenolic compounds extracted from the red maple plant, such as glucitol-core containing gallotannins, include antioxidant, and antiglycation effects as well as their importance in cosmetic applications. Plant-derived proteins and peptides are important biomolecules; however to date, there is no published data on the identification of proteins/peptides from red maple leaves. Therefore, the present study focuses on the activity guided purification of proteins from red maple leaves collected in spring and fall seasons. In addition, the focus of this project was in the evaluation of maple leaves employing bottom-up proteomics and De Novo protein profiling by PEAKS Studio-X and Gene Ontology Bioinformatics. The red maple leaves were grounded, defatted in hexane and proteins extracted in 25 mM sodium phosphate buffer pH 6.5. The extracted crude proteins were recovered by precipitation in 80% ammonium sulfate. The first-dimensional chromatography of crude extracted proteins was performed on a gel filtration column (HiLoad 16/600 Superdex200). The separation of crude extract and the partially purified gel filtration fraction was conducted by reversed-phase HPLC. The crude and eluted fractions were analyzed by SDS-PAGE gel electrophoresis. The extract was screened for cytotoxicity activity on Michigan Cancer Foundation-7 breast cancer (MCF-7), M.D. Anderson Metastasis Breast (MDA-MB-231) cancer cell lines and anti-inflammatory activity on murine macrophage (RAW 264.7) cell line from American Type Culture Collection (ATCC). The drug Doxorubicin was used as a positive control whereas untreated cells as a negative control in these experiments Preliminary data revealed that active protein fractions were eluted at two different regions of gel filtration chromatography both in spring and fall leaves. Bottom-up proteomics of crude and active fractions by PEAKS Studio-X and MASCOT bioinformatics database identified over 54 proteins. The Gene Ontology Annotation classified these proteins involved in the biological processing, cellular compartment, and molecular functions. ▼ In the UK, complementary medicines (CMs), including herbal and homoeopathic products, are used extensively and increasingly by patients and the public. This pervasive use of CMs raises concerns related to patient safety which suggest that there is a public need for a health-care provider who can advise on and monitor their safe, effective and appropriate use. The aim of this thesis is to investigate, from both the users' and the pharmacists' perspectives, the potential of the pharmacist to take on such a role. The present work confirms the need for an expert adviser-monitor on CMs, and shows that pharmacists have the potential to undertake this function. Data resulting from the work show that most pharmacies sell CMs, and that users of CMs and pharmacists interact with respect to the use of these types of products. Therefore, there are opportunities for pharmacists to advise patients and the public on the safe, effective and appropriate use of CMs. Furthermore, there are ways in which pharmacists can contribute to increasing the evidence base for CMs, not only by collating current knowledge, but also by conducting community-pharmacy-based clinical research projects. However, the data raise several concerns, such as the extent of pharmacists' knowledge and training, and their professional practice regarding CMs. Pharmacists and the profession need to address these issues. Consequently, it is recommended that pharmacognosy (the study of natural products used in medicine) and areas of complementary/alternative medicine (CAM), particularly CMs, be included on the indicative syllabus for Master of Pharmacy (MPharm) degree programmes. Also, the training needs of practising pharmacists in these areas should be reviewed. Pharmacists should be encouraged to improve their professional practice pertaining to CMs, including reporting adverse drug reactions for herbal products. There is a corresponding need to raise public awareness of the pharmacist as a source of objective, reliable advice and information on CMs. In addition, an ongoing programme of research is suggested to clarify the area of CAM within pharmacy, and to determine the implications of CAM use for pharmaceutical care.

▼ This dissertation was oriented towards the isolation and structure elucidation of novel biologically active metabolites from marine organisms collected in the South Pacific. In this investigation I focused on the characterization and biological evaluation of vitilevuamide, a novel bicyclic depsipeptide isolated from two Fijian ascidians, Polysyncraton lithostrotum and Didemnun cuculliferum. The first chapter reviews cyclic peptides of marine origin along with their biological activity. This provides an overview of the types of peptides isolated from marine sources along with their structural uniqueness and similarities. The subsequent chapters deal with different aspects of the chemical and biological characterization of vitilevuamide. The alcohol extract of the ascidians was fractionated to yield vitilevuamide which displayed potent cytotoxic activity toward the human colon tumor cell line HCT 116. The structure of this peptide was elucidated primarily using nuclear magnetic resonance (NMR) spectroscopic techniques and tandem mass spectrometry (MS/MS). The peptide was partially linearized using alcoholic ammonia in order to facilitate analysis by tandem mass spectrometry. Chapter 3 deals with the synthesis of two not commercially available amino acids. The first is homoisoleucine (Hil) and the second N-methyl methoxinine (Nmm), a novel amino acid. The synthesis of Hil was successfully completed according to reported procedure. In the case of Nmm, several synthetic routes were attempted prior to successful synthesis of the molecule. Chapter 4 deals with stereospecific assignments of the amino acids. Their absolute configurations were determined by HPLC analysis of derivatized constituent amino acids obtained from acid hydrolysis of the peptide. New conditions were determined for the unambiguous assignment of isoleucine (He), Nmm and Hil. Liquid chromatography mass spectrometry (LCMS) was utilized in order to verify the stability of lanthionine (Lan) prior to assignment of stereochemistry. The biological profile of vitilevuamide is discussed in Chapter 5. The cytotoxicity of the peptide was evaluated in a panel of six human tumor cell lines. Vitilevuamide also scored positive in an in-house mechanism based antitumor screen geared towards the detection of tubulin interactive compounds.

Phytoecdysteroids, polyhydroxylated ketosteroids, are the plant analogues of insect growth hormones. Although their role in insect molting is well characterized, their function in plants is less clear. Lacking the properties of classic plant hormones, phytoecdysteroids may be involved in plant growth and defense. One of the main benefits of phytoecdysteroids may be their therapeutic effects on mammals, including humans. Their claimed medicinal properties include anabolic, adaptogenic, hepatoprotective, and hypoglycemic activity. Although ethnobotanical use has been supported by some evidence, the research is quite limited, lacking the scientific rigor necessary to be convincing. Two ecdysteroid containing plants, Ajuga turkestanica, and Spinacia olearaceae (Spinach), were selected as beneficial sources of phytoecdysteroids. Cultivation, analysis of ecdysteroid content, and characterization of anabolic activity were performed to support future medicinal use. Phytoecdysteroids' anabolic activity, one of their most interesting properties due to the claimed lack of androgenic effect, was studied. Anabolic activity was confirmed in animal studies and a cellular model of skeletal muscle. The cellular model was used to characterize ecdysteroids' effect on protein incorporation and to elucidate the signal transduction pathway involved. Ecdysteroid's lack of androgenic activity was confirmed in vivo and in vitro, with ecdysteroids showing no specific binding to the androgen receptor. Identification of mammalian nuclear receptors homologous with the insect nuclear ecdysone receptor led to binding and activation assays of potential receptors using ecdysteroids. The discovery of a lesser known membrane bound G Protein Coupled Receptor (GPCR) insect ecdysone receptor, DoEcR, suggested the existence of a hypothetical mammalian membrane bound GPCR ecdysone receptor. Use of specific inhibitors supported the involvement of G protein signaling, Phospholipase C (PLC), Inositol Phosphate 3 Receptor (IP3R), and Akt. Ecdysteroid stimulated activation of Akt confirmed its role in the anabolic effect. Ecdysteroid generated increases in intracellular calcium were also characterized, with the rapid flux in Ca2+ linked with Akt activation and anabolic activity. The evidence produced suggests the involvement of a putative mammalian GPCR ecdysteroid receptor mediating the anabolic effect through the rapid activation of the PLC/IP3R pathway, generating Ca2+ flux which leads to activation of the Phosphoinositide 3 Kinase/Akt pathway, eventually causing increases in protein incorporation.

La pharmacognosie, science multidisciplinaire étudiant les produits naturels, n'a eu de cesse de s'adapter et d'évoluer parallèlement aux autres disciplines, en particulier les techniques d'analyse chimique et les technologies de l'information. De nouvelles approches complémentaires, comme la pharmacognosie inverse allant des molécules vers les organismes sources, un cheminement en sens inverse de la pharmacognosie, ont été développées. Les données nécessaires à ces études n'ont jamais été aussi nombreuses et hétérogènes. Molécules, activités, cibles, organismes sources et leurs usages traditionnels, sont autant d'informations essentielles à la pharmacognosie. Le but de cette thèse a été de développer une plateforme informatique, regroupant toutes ces informations diverses au sein d'une vaste base de données, accompagnée d'une interface web dédiée et d'outils de chémoinformatique et de fouille de données. Parallèlement, un enrichissement constant de cette base a été réalisé. Cette plateforme permet de rechercher et de consulter les informations, mais également de les croiser afin d'en faire émerger de nouvelles connaissances. Réellement optimisée pour la pharmacognosie, elle simplifie et améliore l'étude des produits naturels en permettant une approche sur de multiples niveaux (ethnopharmacologie, botanique, chimie), offrant de nouvelles possibilités d'interaction entre les données de ces différents domaines comme l'organism hopping (sauts entre organismes), une approche originale de comparaison des organismes selon leurs profils chimiques.

Pharmacognosy, a multidisciplinary science which studies natural products, has greatly evolved in parallel with other disciplines particularly analytical chemistry and information technologies. New complementary approaches like reverse pharmacognosy, from molecules to source organisms (in contrary of pharmacognosy), have been developed. Data necessary for these studies are overwhelming and heterogeneous. Molecules, activities, targets, organisms, traditional uses, are some of essential information for pharmacognosy. The main goal of this thesis was to develop an information platform, including a large database, to collect the diverse data, along with a specific web interface and also mining and cheminformatics tools. A constant enrichment of this database has also been realized. This platform allows to query and browse data, but also to cross them in order to discover new knowledge. Truely optimised for pharmacognosy, it makes easier and enhances the study of natural products, due to a multi-level approach (ethnopharmacology, botany, chemistry). It offers new possibilities of interaction between data from different domains, such as organism hopping, a new concept of organism comparisons according to their chemical profiles.

▼ The genus Illicium L. (Schisandraceae) consists of ca. 40 species that form one of the earliest evolutionary branches of angiosperms. It is represented by evergreen trees and shrubs disjunctively distributed in North America, Mexico, Peru, the West Indies and eastern Asia - with the highest concentration of species in Northern Myanmar and Southern China. While Illicium is commonly used as a source for shikimic acid in the production of oseltamivir (Tamiflu), there is a long history of traditional use of this genus as analgesic, antiemetic, antiseptic, antispasmodic, antirheumatic, anxiolytic, carminative, digestive, and sedative agents. Early phytochemical studies of the Illicium genus focused on the extraction of essential oils but recently a variety of secondary metabolites were reported, including the structurally unique and rarely occurring in nature seco-prezizaane sesquiterpenes, diterpenes, triterpenes, prenylated phenylpropanes, lignans, and neolignans. Illicium angustisepalum is a sparingly studied representative of Illicium. Its classification and identification as a member of this taxon has been based on the morphological characteristics of its flowers, fruits, leaves, pollen and seeds. Until now, only two peer-reviewed articles regarding its chemistry have been published, and no biological activities of this plant were reported or recorded so far. In order to substantiate the chemical characteristics of this plant species, the metabolite fingerprint of I. angustisepalum was established by GC-MS and compared with those of better-known species, I. verum and I. lanceolatum. The existing variations of the volatiles within this taxon were identified. The phytochemical investigations led to isolation and identification of a total of sixteen secondary metabolites, including two novel compounds. The biological potentials of the isolates were evaluated in a battery of in vitro bioassay systems for cytotoxic, antimicrobial, neuroprotective, acetylcholinesterase inhibiting and neurotrophic activities, and led to determination of an extensive in vitro biological profile of I. angustisepalum. ▼ Natural products have served as therapeutic agents in the treatment of many human health ailments for thousands of years. In order to find new therapeutic agents for future human health problems, those in the field of natural product drug discovery will need to take advantage of technological advances in screening capabilities such as high-content imaging-based assays for the identification of promising lead compounds, as well as computational calculations to aid in natural product structure determination and chemical reactivity studies. The research reported in this dissertation demonstrates the applications of these methodological approaches in three applications. Trichomonas vaginalis is a sexually transmitted parasite that infects at least 170 million people worldwide and current treatments are limited to a single therapeutic agent, metronidazole. As a result, there is a need for new drug leads to treat this as well as other anaerobic parasites. To address this need, a high-throughput imaging-based assay was developed to screen a fungal crude extract library for inhibition of T. vaginalis and Ect1/E6E7 human cervical cells. This information was used to determine selectivity indices (SIs), and bioassay-guided fractionation led to the purification of promising leads that inhibit T. vaginalis while remaining safe to human cells. We found that the quinone-containing compounds fusarubin (SI=30) and xanthoquinodin A1 (SI=20) were promising leads, but as quinones are known to have indiscriminate activities, further protocols were developed to probe the potential liabilities of these compounds. Manipulation of oxygen levels led to a loss of potency in fusarubin, while xanthoquinodin A1 inhibited Lactobacillus acidophilus, a beneficial bacterium found in the vaginal tract. As such, we determined that while quinones show antitrichomonal activities, these compounds should be approached with caution when considering them as leads. Aflatoxin B1 (AfB1) is a mycotoxin with tremendous health impacts as it is widely considered one of the most potent liver carcinogens known to humankind, and billions of people worldwide are exposed to it through their diets. While efforts have been made to find small molecules capable of protecting the liver from AfB1, there has been little success. In order to address this problem, we developed a high-throughput and high-content screening assay to identify and test natural products that are capable of protecting HepG2 liver cells, grown as monolayers and spheroids, against AfB1 toxicity. The use of spheroids proved to be excellent for the production of in vivo sensitivity of HepG2 cells under in vitro conditions, and led to the identification of two promising compounds, alternariol-9-methyl ether (purified from an Alternaria alternata isolate) and 1-hydroxy-3-methoxyxanthone, as potential future starting points for the development of new AfB1-protective agents. Deste modo, os objetivos do presente trabalho visaram avaliar os extratos brutos liofilizados de folha (EBLF) e caule (EBLC) quanto a caracterização botânica, o estudo químico e farmacológico, direcionando principalmente, às ações antiúlcera, antioxidante e cicatrizante, e toxicidade destes órgãos vegetais de C. ferrea. A triagem fitoquímica foi realizada com a droga vegetal constituída de folha (DF) e de caule (DC), bem como com os seus EBLF e EBLC. Os métodos empregados foram preconizados por Farnsworth (1966) e Matos (1988) onde foram pesquisados os seguintes compostos: flavonóides, glicósidos cardiotônicos, saponinas, antraderivados, alcalóides, cumarinas, taninos e óleo essencial. Além disso, foi realizada a quantificação de taninos e flavonóides segundo a metodologia proposta na Farmacopéia Européia (2001) e na Farmacopéia Brasileira (2003), respectivamente. Para a avaliação da Toxicidade de C. ferrea foram realizadas a Toxicidade Aguda de ambos os órgãos vegetais, a DL50 do EBLF e a Toxicidade subcrônica do EBLF e EBLC, todos os modelos seguiram a metodologia de Brito (1994). Para análise da atividade antiulcerogênica da espécie em estudo foi realizado o teste da indução de lesão gástrica aguda por etanol/HCI e lesão subcrônica por ácido acético. Grupos Tratados receberam EBLF ou EBLC ou frações ou extratos enriquecidos em flavonóides, Grupo Controle água ou tween 80 e Grupo de Referência Misoprostol ou Cimetidina. Três parâmetros foram avaliados neste modelo: Área Total de Lesão (ATL) , Área Relativa de Lesão (ARL) e índice de Lesão Ulcerativa (ILU). A atividade antioxidante ïn vitrof̈oi medida através da inibição da autoxidação de homogenato de cérebro de Ratos (Stocks et aI., 1974). Os extratos foram solubilizados em etanol 70% e as diluições (0.05-0.003mg/mL) foram efetuadas em etanol 35%. O etanol 35% foi utilizado como controle. E na avaliação da Atividade Cicatrizante, os animais (ratos) sofreram uma incisão na região dorsal com auxílio de punch. Os Grupos Tratados receberam diariamente 1 mL de EBLF ou EBLC, solubilizados a 15% em água, e o Grupo Controle água destilada na mesma proporção por um período de 14 dias. Na triagem fitoquímica foram detectados para ambos os extratos, flavonóides, taninos, além de antraderivados e cumarinas nas Folhas. A porcentagem encontrada de Taninos na DF foi de 7.13% e no EBLF de 23.95% e na DC foi de 2.26% e no EBLC de 11.77%. Já a quantificação de flavonóides foi de 0.0095% na DF, 0.026% no EBLF, 0.00014% na DC e de 0.0017% no EBLC. No teste de toxicidade aguda, somente os animais que receberam EBLF apresentaram alterações comportamentais a partir dos primeiros tempos de observação e morte de 3 animais machos e 2 fêmeas (n=5/sexo). ▼ Prevention of the accumulation of undesirable biological material i.e. biofouling upon a solid surface requires the use of antifouling systems. The solid surface may be a contact lens, an off shore oil rig or a living organism. When chemicals are employed as a mechanism of defense against biofouling, the agents involved are known as antifouling agents. Marine algae must protect themselves from fouling organisms and it is thought that one of the mechanisms used by these organisms is the production of secondary metabolites with an array of biological activities. In vitro studies have shown numerous compounds isolated from marine algae to possess antibacterial, antifungal and antimacrofouling activity. The aim of this study was to evaluate the secondary metabolite extracts of selected Southern African marine macro-algae as a potential source of compounds that inhibit biofilm formation and that could be used as antifouling agents. In this project, marine macro-algae were collected from various sites along the South African coastline. Their extracts were screened for antimicrobial activity against four ubiquitous microorganisms, Staphylococcus aureus, Klebsiella pneumoniae, Mycobacterium aurm and Candida albicans. Results of screening assays guided the fractionation of two Rhodophyta, Plocamium corallorhiza and Laurencia flexuosa. The algae were fractionated using silica gel column chromatography and compounds were isolated by semi-preparative normal phase HPLC. Compound characterization was performed using UV, IR and advanced one- and two-dimensional NMR (¹H, ¹³C NMR, COSY, HSQC, HMBC and NOESY) spectroscopy and mass spectrometry. Ten halogenated monoterpenes including four members of the small class of halogenated monoterpene aldehydes were isolated from extracts of P. corallorhiza. The compounds isolated included the known compounds 3,4,6,7-tetrachloro-3,7-dimethyl-1-octene; 4,6-dibromo-1, 1-dichloro-3,7 -dimethyl-2E,7 octadiene; 4,8-d ibromo-1,1,7 -trichloro-3, 7-dimethyl-2,5Eoctadiene;1 ,4,8-tribromo-3, 7 -dichloro-3,7-dimethyl-1 E,5E-octadiene; 8-bremo-6, 7-dichloro-3,7-dimethyl-octa-2E,4E-dienal; 4-Bromo-8-chloro-3,7-dimethyl-octa-2E,6E-dienal; 4,6- Dibromo-3,7-dimethyl-octa-2E,7-dienal; 2,4-dichloro-1-(2-chlorovinyl)-1-methyl-5-methylidene-cyclohexane and two new metabolites 4,8-chloro-3,7-dimethyl-2Z,4,6Z-octatrien-1-al and Compound 3.47. Alfaviruksia levittävät Aedes -suvun hyttyset ja niitä esiintyy Etelämanteretta lukuunottamatta kaikilla mantereilla. Alfaviruksia on tähän mennessä löydetty 29 lajia ja ne voidaan jakaa uuden ja vanhan maailman viruksiin niiden maantieteellisen esiintyvyyden ja taudinaiheuttamiskyvyn mukaan. Chikunkunyavirus (CHIKV) on yksi vanhan maailman alfaviruksista, jota esiintyy muun muassa Afrikassa ja Aasiassa. Ilmaston lämmettyä se on leviämässä myös eteläiseen Eurooppaan. Ihmisessä se aiheuttaa muun muassa kuumetta, päänsärkyä, ihottumaa ja niveltulehdusta, joka voi kestää useita vuosia ja ne voivat olla hyvinkin kivuliaita. Pienillä lapsilla chikungunya on todettu aiheuttavan myös neurologisia oireita kuten aivotulehdusta. Alfaviruksen genomi koodaa neljää rakenneproteiinia ja neljää replikaatioproteiinia. Replikaatioproteiineista nsP3 sisältää makrodomeeniosan. Makrodomeeniproteiinit ovat eliökunnassa konservoituneita, mutta makrodomeeniproteiinien tarkkaa merkitystä ei vielä tunneta. Makrodomeenien on osoitettu sitovan ADP-riboosia ja sen johdannaisia ja alfaviruksen nsP3-proteiinin on osoitettu olevan tärkeä osa viruksen replikaatiossa. Tutkimuksen tavoitteena oli tutkia makrodomeeniproteiiniin sitoutuvien yhdisteiden käyttöä antiviraalisena yhdisteinä. Tietokonemallinnuksella valittiin antiviraalitutkimuksiin 45 yhdistettä, joiden oletettiin sitoutuvan makrodomeeniproteiiniin. Kilpailevassa sitoutumiskokeessa viisi yhdistettä esti yli 50 % poly-ADP-riboosia (PAR) sitoutumasta MDO1-makrodomeeniproteiiniin, jolla tietokonemallinnus oli tehty. SFV-makrodomeeniproteiinilla tehdyssä kokeessa vain yksi yhdiste esti yli 50 % poly-ADP-riboosin sitoutumisen. SFV-antiviraalikokeessa seitsemällä yhdisteellä inhibitioprosentti oli yli 50 %. Näillä yhdisteillä ei kuitenkaan ollut merkittävää vaikutusta poly-ADP-riboosin sitoutumisen estossa. CHIKV-replikonikokeessa yli 50 % inhibitioprosentti oli viidellä yhdisteellä. Muiden mahdollisia vaikutusmekanismeja tutkittiin selvittämällä estävätkö yhdisteet virusta pääsemästä solun sisään. Tässä kokeessa tutkituista yhdisteistä lähes kaikilla oli vaikutusta viruksen soluun pääsyn estossa. Yleisesti ottaen kyky estää PAR:n sitoutuminen makrodomeeniproteiineihin ja antiviraaliset vaikutukset eivät korreloineet keskenään tutkittavilla yhdisteillä. Vaikka antiviraalista vaikutusta omaavat yhdisteet eivät osoittaneetkaan makrodomeeni-inhibiitiota, työssä löydettiin potentiaalisia antiviraalisia yhdisteitä joiden käyttö viruksen soluun pääsyn estäjinä antaa aihetta jatkotutkimuksille.

Undersökningen genomfördes med hjälp av en genetiskt modifierad däggdjurscellinje som producerar virala replikationsproteiner samt fluorescerande och luminiscerande markörproteiner. I sekundärstudier undersöktes toxisk effekt och förhållandet mellan dos och respons för prov som varit aktiva i den primära undersökningen. På basen av primärundersökningen identifierades 32 prov (8,6 % av den totala sållade mängden) med aktivitet mot Chikungunyavirusets replikation. Alla aktiva prov var ekstrakt eller isolerade föreningar, de syntetiska föreningarna visade sig vara inaktiva. Falska positiva resultat exkluderades på basen av sekundärstudiernas resultat och slutligen kunde nio ej-toxiska prov med doskorrelerad aktivitet identifieras. Metoden som användes i denna undersökning lämpar sig väl för preliminär sållning för att identifiera prov med effekt mot Chikungunyavirusets replikation. Fortsatta studier med bland annat infektiösa virus samt andra virustyper behövs trots detta för att få mer detaljerad information om provens och arternas antivirala inverkan. Proven bör dessutom undersökas med tanke på isolation av aktiva komponenter, hållbar samling eller odling av marina arter samt möjlig syntes och kemisk optimering av komponenterna.

▼ Rhazya stricta Decne. är en liten vintergrön buske som hör till familjen Apocynaceae. Den växer i Sydasien och Mellanöstern och används där i traditionell läkekonst. Växtens alla delar används i olika typer av preparat och bland de varierande användningsändamålen finns vård av infektioner, tarmsjukdomar, klåda och diabetes. R. stricta syntetiserar ett hundratal alkaloider varav endast en bråkdel har studerats närmare. Några av de analyserade alkaloiderna har visat sig ha intressanta farmakologiska egenskaper, så som antimikrobiella och cytotoxiska egenskaper. Då det ofta är både ekonomiskt och ekologiskt ohållbart att odla eller samla stora mängder medicinalväxter från naturen, har man utvecklat cellodlingar från växterna. Cellodlingarnas egenskaper och syntetiserade ämnen kan analyseras och modifieras i laboratorier. I experimentella delen av detta arbete utvecklades ett system där man extraherade, fraktionerade och isolerade alkaloider från torkat cellmaterial av odlade R. stricta hairy root-celler. Målet var att utveckla ett fungerade system för att slutligen kunna identifiera vilka alkaloider de odlade cellerna syntetiserar under givna förhållanden. Alkaloider extraherades ur 26 g torkad och malen cellmassa. Fraktioneringen gjordes med hjälp av mediumtryck vätskekromatografi (eng. medium pressure liquid chromatography, MPLC) och fraktionerna analyserades med hjälp av tunnskiktskromatografi (eng. thin-layer chromatography, TLC) och högtrycksvätskekromatografi (eng. high-performance liquid chromatography, HPLC). Vid rengöringen av alkaloider användes horisontell TLC och preparativ HPLC. Jonparskromatografi användes vid analys av extraktet, fraktionerna och de rengjorda alkaloiderna. Slutligen isolerades sammanlagt fem komponenter ur två fraktioner. En av komponenterna, vincanin, identifierades preliminärt, men vidare analyser måste göras, för att med säkerhet identifiera alla komponenter. Allt som allt verkar hairy root-cellerna syntetisera några tiotal alkaloider med varierande polaritet.
