▼ Transdermal drug delivery is an effective alternative to conventional oral and injectable drug delivery routes. It offers painless and convenient once daily or even once weekly dosing for a variety of clinical indications. The major limitation to successful transdermal drug delivery is the efficient barrier properties of the skin. Significant research efforts have been focused on developing strategies to overcome these barrier properties. These strategies include the use of physical and chemical penetration enhancers. Physical skin penetration enhancers use an external energy source to alter the barrier properties of the skin. The current research focuses on some of these physical skin penetration enhancers on a range of drug molecules and peptides.The first technology investigated was Dermaportation that utilised pulsed electromagnetic energy. This technology enhanced the epidermal permeation of naltrexone in vitro as compared to passive diffusion. A 5-fold increase in naltrexone permeation was observed during Dermaportation application when compared to passive administration. Multiphoton tomography-fluorescence life-time imaging microscopy (MPT-FLIM) analysis of the permeation of gold nanoparticles in the presence of Dermaportation revealed increased penetration across ex vivo human skin. These results demonstrated that the channels created by dermaportation must be larger than the 10 nm diameter of the applied nanoparticles.The second technology investigated was an unpowered magnetic film array technology (ETP), which utilised unpowered magnetic energy. Chapter 3 presents enhanced epidermal permeation of urea with ETP. A 4-fold increase in urea penetration was observed across human epidermis in the in vitro permeation study. Optical resonance tomography was used to visualise the changes in epidermal thickness due to urea permeation as an indication of increased hydration. The results revealed an increase in epidermal thickness at 30 min, to 16% for ETP induced urea permeation as compared to 3% with urea from occlusion. These results further substantiated our previous findings that magnetic energy creates hydrophilic diffusion channels or pores in the skin.The third technology investigated was low-frequency sonophoresis that utilises cavitation bubbles as a force to create channels for drug delivery in the skin. Chapter 4 presents enhanced human skin permeation of 5-aminolevuleninic acid in vitro and curcumin dye in vivo with low-frequency sonophoresis. Two different sources of ultrasound devices that generated low-frequency sonophoresis were investigated. MPT-FLIM analysis was utilised to investigate the effects of sonophoresis on human skin in vivo. This revealed that there was substantial disturbance in the epidermal cells due to cavitation by sonophoresis. Permeation of curcumin was found in the deeper layers of the epidermal membrane with 55 kHz sonophoresis and was confined to the more superficial layers of skin with 21 kHz sonophoresis. ▼ The resistance of concrete targets to penetration of high speed projectiles is a topic of high value in the national security and catastrophic design fields. Many methods have been developed to effectively analyze these types of problems. Currently there are a number of numerical codes and constitutive models used to analyze concrete impact and penetration with new methods developed continuously. This research evaluates the accuracy of four analysis codes and five concrete constitutive models. Two Lagrangian analysis programs, EPIC and LS-DYNA, as well as an Eulerian code, CTH, are compared in this work. A developmental version of the Material Point Method is also evaluated in order to study the effectiveness of Arbitrary Lagrangian Eulerian (ALE) modeling methods for concrete impact and penetration. The concrete models evaluated in this research include Holmquist Johnson Cook, Brittle Failure Kinetics, Osborn, Karagozian and Case, and Drucker-Prager. The modeling programs and constitutive models are evaluated by comparing simulation results to a series of concrete impact and penetration experiments. The experimental test data, provided by Sandia National Laboratories, comprises concrete targets of two compressive strengths (3.3 and 5.7 ksi) and two thicknesses and projectiles of two nose shapes. Extensive material testing of the experimental concrete is used to calibrate the constitutive models in each analysis package. An additional parametric study investigates the influence of experimental variables on the most promising analytical configuration. Observations from this research show that the EPIC and LS-DYNA analysis codes are currently best suited for concrete impact and penetration problems. Both codes contain features which allow for realistic modeling and produce accurate results for the experimental impact tests. Recommendations for improving analysis methods specific to concrete impact and penetration are presented. ▼ The research detailed in this dissertation provides a comprehensive analysis of market penetration of privately owned autonomous vehicles (AVs) and elaborates on several dimensions of people’s adoption behavior. Research in the area of autonomous mobility is advancing at a fast pace, heavily centered around estimating the potential impacts of the technology on travel behavior, network operation, and the environment. While highly informative, most of these studies inevitably suffer from the absence of a sound estimation of AVs’ market penetration and establish their assessments based on the speculations regarding people’s adoption behavior. Current research is conducted to address such a gap by focusing on multiple dimensions of people’s AV adoption behavior such as sensitivity of their adoption decision to the vehicle attributes, their willingness to pay, their preferred AV adoption timing, and the potential variance in adoption behavior of people from different generations. The research is designed to thoroughly investigate how individual- and household-level demographics, travel preferences, social factors, and built-environment characteristics affect each dimension of people’s adoption behavior. To collect the required data for the study, a comprehensive web-based stated preference (SP) survey has been designed and conducted in the Chicago metropolitan area. All results presented in this dissertation are based on the data derived from the implemented survey. Various behavioral theories from the well-known utility maximization theory to the innovation diffusion theory have been adopted to improve the behavioral realism of the models. Further, several advanced methodological approaches have been employed to address the limitations of the traditional models. Estimating a heterogeneity-in-means random parameters best-worst model as an advanced alternative of the select-one-choice models, developing a random parameters random thresholds hierarchical ordered probit model for estimating people’s willingness-to-pay for AVs, and developing a joint model of vehicle fuel choice and automation choice to capture the potential correlation between the two decisions and account for the shared unobserved factors that might affect them are amongst the methodological contributions of the current research. ▼ Polymeric nanoparticles (NPs) have demonstrated potential for the delivery of bioactive agents to solid tumors, however, despite decades of research, few polymeric NPs have been able to reach the clinic. Conventional designs have taken a 'one-size fits all' approach, yet several factors play critical roles in defining their nano-bio interactions, including size, shape and surface functionality. Whereas larger NPs between 50-200 nm tend to exhibit enhanced circulation times and tumor accumulation, they are limited in their ability to efficiently permeate tissues. In contrast, ultrasmall NPs (<10 nm) demonstrate markedly improved tissue penetrating capabilities. In order to investigate the roles that ultrasmall NP precise size and surface functionality play in their ability to penetrate a solid tumor we prepared poly(amidoamine) (PAMAM) dendrimers, small (<10 nm) highly branched polymers with tightly controlled size distributions. G2 (2.9 nm), G4 (4.5 nm), and G7 (8.1 nm) dendrimers were prepared containing positive, neutral, and negative surface charge and their penetration evaluated in multicellular tumor spheroids (MCTS) and an in vivo tumor xenograft model. Whereas larger G4 and G7 dendrimers were restricted to the MCTS and tumor periphery even after prolonged treatment times, smaller G2 dendrimers displayed more rapid penetration characteristics. Among charged groups, positive dendrimers exhibited significantly improved accumulation compared to their neutral or negative counterparts. Our findings suggest that dendrimer size is the greatest determinant in its tumor permeation, with NP-cell interactions, endocytosis, and particle flexibility not significantly contributing to penetration depth. Moreover, when the anticancer drug doxorubicin (DOX) was conjugated to dendrimers, DOX penetration demonstrated a similar trend dependent on size, resulting in notable differences in the distribution of dead cells within the MCTS. These findings reveal that dendrimer size and surface functionality determine their tumor penetration behaviors, and provide useful insights in the further development of ultrasmall NPs with precisely controlled tumor distributions. ▼ Seismic piezocone testing (SCPTu) offers an economic and expedient means for the modern collection of geotechnical data during site investigation of soils. It is actually a hybrid test that has all the advantages and the merits of classic cone penetration testing (CPT) where it obtains three continuous readings with the depth: cone tip resistance (qt), sleeve friction (fs), and porewater pressure (u2); plus geophysical component involving downhole testing, where shear wave velocity (Vs) measurements are recorded at 1-m depth intervals. The conducted study aims at making full usage of the four main readings obtained from the seismic piezocone test: cone tip resistance, cone sleeve friction, porewater pressure and downhole shear wave velocity. New links between undrained shear strength and shear wave velocity are investigated and previous relationships between stress history and shear wave velocity are revisited and improved. A unified approach for estimating stress history of wide variety of geomaterials using net cone tip resistance is presented. Predictive equations for detecting sensitive clays from seismic shear wave velocity are introduced with a focus on sensitive and structured clays within North America. Modification of a hybrid spherical cavity expansion theory- critical state soil mechanics solution is introduced addressing the stress history of sensitive and structured clays. A new analytical approach allows the direct assessment of undrained rigidity index from CPTu data which finds value in assessing undrained shear strength, yield stress, and coefficient of consolidation, the latter from piezodissipation tests. The special nature and difficulties of organic clays are discussed and an indirect means of detecting their existence using piezocone testing is presented. Finally, an effort is undertaken to calibrate an analytical model for evaluating the undrained shear strength and stress history of clays using two large databases. ▼ This thesis provides details of the hardware architecture and the software scripting, which are employed to demonstrate penetration testing in a laboratory setup. The architecture depicts an organizational computing asset or an environment.¬¬¬ With the increasing number of cyber-attacks throughout the world, the network security is becoming an important issue. This has motivated a large number of “ethical hackers” to indulge and develop methodologies and scripts to defend against the security attacks. As it is too onerous to maintain and monitor attacks on individual hardware and software in an organization, the demand for the new ways to manage security systems invoked the idea of penetration testing. Many research groups have designed algorithms depending on the size, type and purpose of application to secure networks [55]. In this thesis, we create a laboratory setup replicating an organizational infrastructure to study penetration testing on real time server-client atmosphere. To make this possible, we have used Border Gateway Protocol (BGP) as routing protocol as it is widely used in current networks. Moreover, BGP exhibits few vulnerabilities of its own and makes the security assessment more promising. Here, we propose (a) computer based attacks and (b) actual network based attacks including defense mechanisms. The thesis, thus, describes the way penetration testing is accomplished over a desired BGP network. The procedural generation of the packets, exploit, and payloads involve internal and external network attacks. In this thesis, we start with the details of all sub-fields in the stream of penetration testing, including their requirements and outcomes. As an informative and learning research, this thesis discusses the types of attacks over the routers, switches and physical client machines. Our work also deals with the limitations of the implementation of the penetration testing, discussing over the vulnerabilities of the current standards in the technology. Furthermore, we consider the possible methodologies that require attention in order to accomplish most efficient outcomes with the penetration testing. Overall, this work has provided a great learning opportunity in the area of ethical hacking using penetration testing.

▼ Historically, cognitive scientists and philosophers have accepted a theory of the mind known as modularity, whereby individual thought processes are completely separate and insulated from one another—meaning that cognitions have no influence on perceptions. However, the recent literature has seen a resurgence in support of a thesis of cognitive penetration, which suggests that cognitions can and do influence perceptions in a way that would be impossible if the mind were modular in the traditional sense. In addition to calling the idea of modularity into question, cognitive penetrability raises some passing concerns for the objectivity of scientific observation, and certain philosophical distinctions such as that between cognition and perception. Along similar lines, the literature has also seen an increase in the exploration of cognitive phenomenology, which similarly calls into question the distinction between cognition and perception and requires a model of the mind which is less clear-cut than the modular view. As such, it seems that given the evidence, one cannot accept either penetrability or cognitive phenomenology without accepting the other, given that they both rest on a similar view of the mind. In addition to calling into question the literal distinction between cognition and perception (though it may remain intact on a conceptual level), a subsection of cognitive phenomenology, known as evaluative phenomenology (the unique phenomenal character of emotions) similarly makes ambiguous the philosophical distinction between reason and emotion. Breaking this dichotomy, as well, makes the possible epistemic consequences of penetrability pale in comparison to those implied by cognitive phenomenology. While this is not an answer to the issues raised by penetrability, it does contextualize the difficulties in a way which opens the system up to a deeper understanding. This study examines set-off in newspaper printing and its relation to newsprint properties and other printing parameters, consisting of a review of the literature related to the subject, a theoretical discussion and a set-off process analysis. Laboratory printing tests with laboratory-made paper and ink samples and commercial newsprint and newsink samples were carried out. In addition, the surface of newsprint, the structure of the offset blanket, the ink penetration and its location on the paper surface, and set-off prints were studied with microscopic methods. Ink transfer and ink setting and the situations where set-off is created were also examined. Main targets for this work were: to define the optimum surface characteristics for uncoated newsprint in newspaper printing with the coldset web offset (CSWO) method, and to gain a better insight into the set-off phenomenon. In CSWO printing, the compressibility and conformability of the blanket and the compressibility of the newsprint surface improve the ink coverage. After the printing nip the ink film splits only in the area where it has been in contact with the newsprint surface. In single-colour printing, the penetration of ink that is caused by the pressure in the printing nip is insignificant. In contrast, in multi-colour printing, the ink layer printed later pushes the previously printed ink deeper into the voids of the paper surface. Ink setting decreases set-off. It is the result of solvent separation from the ink layer. Ink setting is defined by the change of the set-off during the delay time. Filler loading improves ink setting. Set-off can be reduced by using less ink, higher printing nip pressure, lower pressure in the folder and in other set-off situations, and chemical pulp containing less compressible DIP-based newsprint. The most important properties of uncoated newsprint for set-off are: optimised roughness in relation to print quality and set-off, low surface compressibility, high specific surface area, good absorption ability of the fines area and uniform formation (no high-density calendering spots).

▼ In addition to its various military applications, shaped charges have been used in oil industry as an oil well perforator (OWP) to connect oil and gas to their reservoirs. The collapse of the liner material under the explosive load produces a hypervelocity jet capable of achieving a deep penetration tunnel into the rock formation. The achieved penetration depends on the OWP design, which includes the geometry and the material of the explosive and the liner as well as the initiation mode and the casing of the shaped charge. The main purpose of this research is to assess the performance of OWP with different design aspects in terms of its penetration depth into concrete material.This research employed the Autodyn finite difference code to model the behaviour of OWPs in the stages of liner collapse, jet formation and jet penetration. The design parameters of OWPs were studied quantitatively to identify the effect of each individual parameter on the jet characteristics and the jet penetration depth into concrete material according to the API-RP43 standard test configuration. In order to validate the Autodyn jetting analysis, this research compared the jetting simulation results of copper OWP liners with those obtained from flash x-ray measurements while the numerical jet penetration into the laminated concrete target was validated experimentally by the static firing of OWPs. Above-mentioned experiments were designed and performed in this project.The validated hydrocode was implemented in this research to study the effects of the concrete target strength, the liner material and the liner shape on the jet penetration depth into concrete targets.For the target strength, the traditional virtual origin (VO) penetration model was modified to include a strength reduction term based on Johnson’s damage number and the effect of the underground confinement pressure using Drucker-Prager model. The VO analytical model is also implemented in the liner material study to account for the jet density reduction phenomena and its induced reduction of jet penetration capability. The jets obtained from machined copper and zirconium liners and from copper-tungsten powder liner all exhibited the density reduction phenomena. The modified VO model considers the non-uniform distribution of jet density based on the jet profile analysis using Autodyn and the experimental soft recovery for some tested liners. The results lead to a modified VO penetration model including the non-uniform jet density effect.For zirconium liner material, numerical and analytical studies were conducted for different flow velocities and different collapse angles in order to determine the boundaries between the jetting and non-jetting phases and whether a coherent or a non-coherent jet will form. ▼ The water-material interface is of fundamental importance in nature and in wide engineering applications. Behaviors such as wettability, adhesion and penetration are influenced by the surface morphology. There is growing need in textured surface leading to increased interests in understanding the surface properties of textured materials. To date, the existing wetting models are limited to well-defined surfaces. The adhesion mechanism between water and textured surfaces is not well-understood. Moreover, stability and kinetic behavior of a water drop on a porous surface has yet to be understood. The objectives of this research are: 1) Revealing the principles behind a water drop and a porous surface; 2) Obtaining understanding about wetting, adhesion and penetration of a water drop; and 3) Identifying key parameters related to wetting for materials design. In this research, the investigation about water-solid interface of textured materials is divided to three parts: wettability and adhesion; stability of penetration; and kinetic behavior of penetration. To examine wettability and adhesion on textured surface, super-hydrophobic surfaces of micro-beads/nano-pores were studied. It is for the first time to consider the synergetic effect of wetting and adhesion in textured surfaces. Next, the stability of a drop was studied to understand the penetration mechanism on hydrophobic porous surfaces. An experimental method combined theoretical approach was used through force balance principles. Results show that a critical condition for spontaneous penetration on a porous surface, which is related to pore size, drop volume and surface wettability. During penetration, the kinetic behavior of a water drop has been investigated to reveal new principles such as the maximum penetrating rate, receding contact angle, slip-stick motion of contact circle. Results have revealed fundamental mechanisms of wettability, adhesion and penetration in water-textured material interfaces. Specific geometric design factors of textured materials, which are related to behaviors in water-solid interface, have been identified. The findings in this research lead to new approaches to optimize various applications of textured surfaces. The semi interlocking masonry (SIM) was originally designed and developed at the University of Newcastle for use in seismically active zones as an energy dissipation device in buildings. Interlocking SIM bricks are the primary units used in mortar-less walls or panel construction and are available in two types: mechanical (which use dowels) and topological (which use particular shapes). The interlocking prevents the relative out-of-plane movement, but allows for longitudinal relative movement along bed joints. As a mortar-less masonry system, SIM has unavoidable gaps in both its bedding and perpend joints. This reduces the overall performance of SIM walls in terms of thermal efficiency and water penetration. The main objective of this study was to evaluate water penetration and the thermal performance properties of dry-stacked SIM walls. Additionally, this study aimed to determine whether an alternative construction technique (i.e., the use gap-fillers) led to any improvement in the properties. Two standard test methodologies (i.e., ASTM E514 and C1363) were used to evaluate water and heat transfer through walls. Non-adhesive putty and foam tape were used as gap-fillers to examine the potential improvement in restricting water and heat transfers and their suitability for intended application. In the water penetration experiment, the performance results of a traditional masonry wall were used as the benchmark against which the results of the SIM wall were compared. The results of the water penetration tests showed a high volume of leaked water through the dry-stacked SIM wall compared to the gap-filled SIM wall. The traditional masonry wall had the least amount of water leakage followed by the topological SIM wall that used putty. However, the area of dampness in the traditional masonry wall was higher than the area of dampness in the gap-filled SIM walls. In the second part of this study, a hot box apparatus was used to evaluate the thermal properties of the gap-filled SIM walls. The results of thermal tests showed that the thermal resistance of the SIM wall was equal to the traditional concrete masonry wall. It was also found that the type of gap-filler used had a slight effect on the thermal performance of the SIM walls. However, the SIM walls with the putty was more thermally efficient.

▼ An understanding and ability to predict permeation behavior is important when applying polyelectrolyte treatments in enhanced oil recovery, papermaking, biomass hydrolysis, catalytic process, chromatographic separation, water treatment etc. In this research, polydiallyldimethylammonium chloride (poly-DADMAC) was used as a probe molecule to investigate major factors affecting penetration of polyelectrolyte into small pores, including time, polyelectrolyte concentration, substrate consistency, molecular mass, pH, conductivity, etc. Streaming potential tests were used innovatively to understand polyelectrolyte diffusion and adsorption on external surface and in nanopore networks. The magnitude change in streaming potential, which was taken as evidence of permeation, increased increasing polyelectrolyte dosage, with decreasing molecular mass. Increasing ionic strength tended to enhance adsorption of high-mass cationic polymers on the outer surfaces, but only a relatively small effect on permeation of high-mass cationic polymer into silica gel (nominal pore sizes of 6 nm or 30 nm) was observed. Adsorption of very-low-mass cationic polymer onto the outer surfaces and inside the 6 nm pore size silica gel was maximized at an intermediate salt level (1000 ÃŽÂ¼S/cm conductivity). Electrokinetic tests also were used to provide evidence of polyelectrolyte desorption from mesoporous material. Another technique, a streaming current titration with a particle charge detector (PCD) was used to quantitatively determine polyelectrolyte adsorption on the external and internal surfaces of silica gel particles at levels of the above factors. The diffusion slowed down as the extension of the time for adsorption experiment. The adsorbed amount of poly-DADMAC was controlled by both the pore size and the surface area. The highest adsorption amount, based on mass of the substrate, was achieved when using silica gel having an intermediate pore size (15 nm) at a relatively high solution concentration of very-low-mass poly-DADMAC. The results were fit well to an assumed Langmuir model of the adsorption process. Streaming potential tests and streaming current tests may produce different result due to the difference in measurement principles. Further investigation may be needed for confirmation. ▼ The pharmaceutical industry is one of the most stable and fastest growing industries in the world. In the last few years, this industry’s revenue has surpassed the one trillion U.S. dollars mark. Despite this massive growth, researchers and scholars across the globe are yet to dedicate their studies to understand how this industry can take advantage of multichannel marketing, which is perceived to be the future of marketing. This study therefore aimed at contributing to the knowledge of this subject matter specifically in regard to multinational pharmaceutical companies in Kenya. The study objective was to analyze the influence of multichannel marketing on market penetration by pharmaceutical multinational companies in Kenya. The researcher sought to find out the different marketing channels that multinational pharmaceutical companies are using to penetrate the Kenyan market and also remain relevant and competitive in the long run. To achieve this objective, the researcher utilized cross-sectional study and descriptive research design to collect, organize and analyze data, and present the research findings. Study population of ten multinational pharmaceutical companies which had functional sales and marketing department locally was used. Questionnaires were used to collect primary data and were sent to at least six people in each of the ten companies including: commercial directors, product managers, sales managers and sales executives, making the total target population to be sixty. 37 questionnaires were filled and returned, representing a response rate of 62%. Secondary data used was from company’s publications and websites in addition to Pharmacy and poisons board website. Data collected was analyzed and findings presented in form of graphs, pie charts and tables. It was found out that multichannel marketing in pharmaceutical industries is not as effective as expected in all other industries. This is because pharmaceutical industry, have a number of factors that influence their consumers’ responsiveness to different marketing channels. In addition, the research findings reveal that despite contemporary marketing channels gaining traction in different industries, the pharmaceutical industry perceive the traditional marketing channels as the most effective channels to penetrate the Kenyan market. The study concluded with recommendations on the need to conduct further study on whether multinational pharmaceutical corporations in Kenya rely on the marketing strategies in their mother countries when developing marketing strategy for a country such as Kenya. In addition, the study recommends that there is a need for multinational pharmaceutical companies to study on ways of increasing their use of contemporary multichannel marketing to reach out to their target consumers.

▼ The purpose of the thesis was to undertake a preliminary product design to assess the feasibility of using drone technology to aid reforestation for automated seed planting. This product would be able to increase productivity and reduce labour costs, especially when applied in difficult to reach terrains around New Zealand. Ultimately, the final product would be a weaponised drone or a swarm of drones that can fire plastic seed capsules into soil to a specific depth to prevent predation and ensure optimal germination conditions. The proof of concept hinges on the success of three objectives: Assessing and controlling the capsule’s biodegradability in soil. A method of mass manufacturing projectile seed capsules that can be delivered at high velocity and the ability of it to be fired into soil to a specific depth to ensure safe germination. Novatein was blended with poly(butylene adipate terephthalate) (PBAT) to aid sheet extrusion and improve water resistance. Biodegradability was tested by measuring the ability of bacteria and fungi to metabolise the polymer blends into CO2 by enclosing samples in soil at 25 °C. PBAT showed very little biodegradation under these conditions, where approximately 3 % of the theoretical carbon content of the sample was converted to CO2. Novatein reached 20 %, and the blends were intermediate, based on the amount of PBAT present. It was concluded that PBAT can effectively control the rate and extent of biodegradation in blends with Novatein. A method for mass manufacturing seed capsules was evaluated by compression moulding spheres to test mouldability and manufacturing lap welded samples to quantify weld strength. Blends of Novatein and PBAT were successfully moulded and welded above the glass transition temperature of the material; optimal parameters were 105 °C with a three minute hold time. Lap welds were tested by varying the compression force and sheet thickness and thereby reducing the weld times to approximately eight seconds. Welded samples were tested for tensile and peel strength, where 0.68 mm thick sheets, welded at 750 N compression force, yielded the highest tensile strength (88 % that of an un-welded samples). Thicker sheets (1.52 mm) welded at 900 N compression force, resulted in the highest peel force. Producing capsules capable of penetrating soil would benefit from thicker sheets using sufficient pressure to prevent peeling at the seam. Finally, seed capsule delivery was assessed using empirical modelling in combination with controlled soil conditions. Models created by Young and Lorenz used a dynamic cone penetrometer (DCP) to measure the soil’s penetration resistance to predict the penetration depth of a known projectile. These models were developed for considerably heavier projectiles (> 2.2 kg) and penetration depths much greater than considered here. Considering the DCP measures the soils resistance to penetration at a depth of one meter, a comparison to a 10th scale model was investigated.
