▼ Psychopathy is a personality disorder characterized by antisocial deviance in the context of interpersonal and emotional detachment. The study of psychopathy in non-forensic samples is an area of growing interest, but one that is limited by the fact that most large-scale epidemiological studies, which collect a wealth of data that could further elucidate the phenotypic correlates, constructs, assessments, and etiologic mechanisms in psychopathy, typically do not include direct assessment of psychopathy construct or measurements. However, if facets of psychopathy could be predicted from other measures, such as broadband inventories of normal personality that are often administered in large-scale investigations, data from college epidemiological studies could be brought to bear light on the study of psychopathy. This study is two-fold in the investigation of psychopathy. First, the present study replicated the work of Sellbom and colleagues (2012) three-psychopathy scales derived from the Minnesota Multiphasic Personality Inventory-2-Restructured Form (MMPI-2-RF). These scales were developed to assess psychopathy as conceptualized in the PPI-R and include Global Psychopathy (
Py-T), Impulsive-Antisociality (
Py-IA) and Fearless-Dominance (
Py-FD). Second, the present study built upon the three-psychopathy scales by investigating psychopathy’s construct in relation to Cooke and Michie’s (2001) three-facto model. A sample of 151 participants from a Midwestern university were administrated the PPI-R and MMPI-2-RF. The MMPI-2-RF three scales and construct of psychopathy were evaluated using bivariate correlations. Results support previous studies, regarding the
Py-T,
Py-IA, and
Py-FD scales and the three-factor model of psychopathy.

▼ Many Low Carbon Fuels (LCFs) present unknown spontaneous combustion risks, which must be quantified before their use as fossil fuel replacements. Wood and coal spontaneous combustion is well understood; however, LCFs weather, and subsequent chemical changes could affect their spontaneous combustion properties. LCF spontaneous combustion could lead to accidental fires with possible loss of life, limb andproperty. The spontaneous combustion risks of two LCFs, discarded creosote-treated wooden railway ties and roofing asphalt shingles, were investigated with calorimetry and heat transfer experiments. Chemical changes due to weathering were studied with pyrolysis-Gas Chromatography/Mass Spectrometry (py-GC/MS).
Creosote-treated wooden railway tie dust, roofing asphalt shingle particles, poplar wood pellets, and petroleum coke self-heating were studied with isothermal calorimetry. Railway tie dust and asphalt shingle heat transfer were characterized with a guarded hot plate. Petroleum coke self-heating was consistent with coal, while both poplar pellets and railway tie dust were found to be more reactive compared to oven test results of similar materials. The observed increase in reactivity was probably a result of significant moisture contenint in the pellet and railway tie dust. Critical conditions for spontaneous combustion were evaluated with the Frank-Kamenetskii parameter, assuming an ambient temperature of 40°C and constant moisture content. Kamenetskii calculations indicate that a 1.6 m cube of railway tie dust, or a 58 m cube of asphalt particles, would be unstable and combust.
LCF chemistry may have been affected by weathering, which would cause chemical changes that affect their spontaneous combustion properties. Therefore, railway tie wood and roofing asphalt shingle chemistry were investigated by identifying products of 250° and 550°C pyrolysis with py-GC/MS. Railway tie wood pyrolyzates did not show signs of weathering; in contrast, asphalt pyrolysis produced sulfur dioxide and many oxygenated species, which indicated that the asphalt was extensively oxidized.
Calorimetry showed that self-heating was primarily oxidative — the oxygen content of asphalt probably reduced its self-heating capacity. The railway tie wood was not extensively oxidized and it self-heated in a similar manner to “fresh” wood.

This PhD thesis focus on the study of spin currents and of the spin Hall effect in lateralnano-structures. Lateral spin-valves based on Py/Al, Py/Cu and Py/Au, fabricated by meansof electron-beam lithography, have been optimized and characterized using magneto-resistancemeasurements. Non-local, GMR and Hanle effect measurements have been recorded at 300K and77K. The optimization of these lateral spin-valves allowed the observation of high spin signalamplitudes. Lateral and vertical confinement effects on the spin accumulation, by using confinednon-magnetic channel and AlOx tunnel barriers, were evidenced experimentally and describedtheoretically. Finite Elements Method simulations and analyses based on a 1D diffusion modelhave been developed, allowing the extraction from our experimental data of the effective spinpolarization Peff and of the spin diffusion length lNsf .Finally, the spin Hall effect of materials with high spin Hall angles (Pt, Au alloys) has beenstudied using both hybrid lateral nano-structures and spin pumping ferro-magnetic resonance.

The current work presents the chemical characterization of Brazilian artistic paints. Differently from other matrices, art materials produced in Brazil lack for information in literature that help in their conservation and restoration. Acrylic and oil paint samples, Brazilian and imported, were analyzed by Raman and Fourier transform infrared (FTIR) spectroscopies, pyrolysis coupled with gas chromatography and mass spectrometry (Py-GC/MS) and
scanning electron microscopy coupled with x-ray fluorescence spectrometry (SEM-EDS). Results indicate that Brazilian acrylic paints are composed of polystyrene (PS), differently from imported ones, composed of polymethyl methacrylate (PMMA). Whereas, oil paints present similar composition (linseed oil), Brazilian and imported. Two paintings from the collection of Pinacoteca do Estado de São Paulo were selected for a case study. Both works of art present formation of exudates on their surface, which demands chemical characterization as a previous step to the development of a restoration methodology. The painting \"Mandala da saudade\", by Sara Goldman-Belz, is composed of spray paint (alkyd) and the painting \"Três pessoas\", by Marina Saleme, is composed of oil paint. Results presented in this thesis are the first step towards the establishment of a database for artistic materials produced in Brazil for purposes of art restoration and forensic science.

Biomass is a renewable energy source, which collect and stock CO2. The objectives of this work were to study the thermal behavior of natural biomass and its components (cellulose, hemicellulose and lignin). We were particularly interested in the thermal degradation mechanisms of natural polymers arising, first, during heat treatments (primary mechanisms). This work was carried out with in the framework of the ACI "Biomasters" with the support of Credit Mutuel. The thermal behaviors of biomass and its components were studied by using thermogravimetry (TGA), pyrolysis coupled with mass spectrometry(Py-MS) and pyrolysis coupled with gas chromatography with detection by mass spectrometry (Py-GC/MS). The thermal behaviors of biomass components were shown, by TGA, to be appreciably different and their roles were specified in thermal degradation. We also showed the impossibility of proposing estimated laws of biomass thermalbehavior on the basis of the main component TGA results. With Py-MS, we developed a method allowing to highlight the most representative molecular fragments evolved during biomass thermal degradation. This enabled us to specify the most sensitive components towards metallic catalystaction. The Py-CG/SM analysis of the mixtures showed that vapor compositions in the mixtures are close to simple laws of addition. We complete our work by an…

▼ Further advancements in the offshore wind industry, which come with the use of larger wind turbines at deeper water, require larger support structures. For shallow and intermediate water depths, monopile foundations still remain the most widely-used and cost-effective support structure due to the relative simplicity of fabrication and installation. The aim of this study is to investigate the challenges and feasibility of extending monopile technology for larger wind turbines and deeper water. Preliminary monopile designs to support the DTU 10 MW reference wind turbine were established for water depths 20 m, 30 m, 40 m and 50 m. For large diameter piles, applying lateral soil stiffness derived from API method underpredicts lateral deflections at both seabed and pile toe. To properly account for the pile-structure interaction and the rigid pile behaviour, lateral soil stiffness was derived using finite element software Plaxis 3D. To verify the preliminary design, ULS and FLS analyses were done using 1st order and 2nd order wave models applied with Morison’s equation. For the ULS analysis, the 2nd order wave model predicts a response which is 1.9 times higher than the 1st order prediction for a water depth of 20 m. The sum-frequency effects and higher wave particle acceleration for the 2nd order wave model explain this result. For the FLS analysis, it was found that designing monopiles for higher water depths increases contribution of hydrodynamic loads to fatigue damage. A more sophisticated wave model is desired to increase accuracy in fatigue damage prediction at deeper water. Lastly, a method for predicting total fatigue damage using a fewer number of representative sea states is introduced. A Fatigue Damage Parameter (FDP) is established to correlate fatigue damage to environmental condition by means of thrust, Hs and Tp. Using at least 30% of the total number of conditions results in at least 90% accuracy in damage prediction. The applicability of the method to other structures and environments is left for future work. ____________________________________________________________________________________

▼ Glutathione (GSH) plays essential roles in many
cellular processes, and disturbances
in its homeostasis are
associated with a number of human diseases. GSH export from
cells
is a key component in regulating GSH levels; however the proteins
that mediate
GSH transport and underlying mechanisms are not well
defined. In an attempt to
identify novel GSH transporters, a human
mammary gland cDNA library was
screened for clones capable of
complementing GSH uptake by yeast cells that lack
Hgt1p, the
primary GSH uptake transporter in yeast, although these cells do
retain an
uncharacterized low affinity GSH uptake mechanism. This
screening strategy initially
identified 11 cDNAs that complement
the growth deficiency of the HGT1 deletion
cells in
sulfur-deficient medium supplemented with GSH. From this pool, five
human
genes were identified that were able to increase
intracellular glutathione levels,
namely: PMEPA1, prostate
transmembrane protein, androgen induced 1; LAPTM4α,
lysosomal-associated protein transmembrane 4 alpha; SLC25A1, solute
carrier family
25, member 1; LITAF, lipopolysaccharide-induced TNF
factor; and CYYR1,
cysteine/tyrosine-rich-1.
All of these genes
encode for small integral membrane proteins of unknown function,
although none appear to encode for prototypical GSH transporters.
Nevertheless, they
all increased [3H]GSH uptake by the yeast
cells. The enhanced [3H]GSH uptake rate
was uniformly inhibited by
high concentrations of unlabeled GSH, GSSG, and
ophthalmic acid,
suggesting that it is due to increased activity of an endogenous
yeast
low affinity GSH uptake mechanism. Interestingly, each
protein is predicted to
contain Pro-Pro-x-Tyr (PY) motifs, which
are thought to be important for regulating
intracellular protein
sorting, plasma membrane protein expression, and multiple steps
of
ubiquitin-dependent endocytosis. Indeed, uptake of the endocytotic
markers lucifer
yellow and FM4-64 was enhanced by each of the five
genes. Mutations of the PY
motifs in LITAF largely abolished all
of its effects. In summary, although the results
do not reveal
novel GSH transporters, they identify five PY-containing gene
products
that enhance GSH and endocytotic marker uptake in yeast,
suggesting that these
proteins are involved in cell surface
remodeling.
In addition, a HGT1 overexpressing yeast strain was
characterized. The constitutive
expression of Hgt1p led to
increased GSH uptake and excessive GSH accumulation in
the yeast.
As a result of GSH toxicity, these cells exhibited growth
inhibition in GSH
containing medium, and thus could potentially be
utilized as a model to search for
unidirectional GSH efflux
transporters.

▼ Highly oxygenated compounds in bio-oil produce negative properties that have hampered fuel development. Copyrolysis with plastics has increased hydrogen content in past research.
Py-GC/MS analyses for two wood types (pine and oak) and three plastic types (polystyrene, polypropylene and high density polyethylene) established temperature, heating rate and residence time to produce a typical bio-oil. Analysis of various plastics to wood ratios by
Py-GC/MS showed that a 50:50 wt/wt ratio produced the highest level of low molecular weight compounds best for fuel viscosity. Copyrolysis was performed on a laboratory-scale reactor at these temperature and wood-to-plastic ratios. Copyrolysis lowered bio-oil oxygen content and increased carbon content. Lower water content, acid value and viscosity also resulted, improving bio-oil suitability for fuels. Cross reactions between wood and plastics formed no new chemical species during copyrolysis. These results indicate that copyrolysis of waste plastics with woody biomass has potential for improving bio-oil properties for fuels production.

▼ Despite the importance of soil organic matter (SOM) in C storage and provision of ecosystem services, the magnitude and direction of the response of SOM to climate change remains debated. Particularly contested is the role of biochemical recalcitrance in determining the biological stability of SOM, which in turn, may also vary with climate. Employing a climosequence study design controlling for confounding pedogenic factors, the research described in this thesis aimed to uncover the response of both SOM chemistry and SOM biological stability to changes in climate and associated land use shifts at the grassland-forest ecotone in west-central Saskatchewan. Characterization of SOM chemistry was achieved using two advanced analytical techniques: X-ray absorption near edge structure (XANES) spectroscopy and pyrolysis-field ionization mass spectrometry (
Py-FIMS). Agreements between XANES and
Py-FIMS revealed only minor differences in SOM chemistry resulting from a 0.7 °C mean annual temperature (MAT) gradient and associated broad differences in land use, but revealed a clear influence of depth within soil profiles. In contrast, long-term aerobic incubations revealed that biological stability of SOM varied with both climate and climate-induced differences in land use, but was not largely influenced by depth. Together, these findings suggest a decoupling of SOM chemistry and its biological stability, indicating that factors other than biochemical recalcitrance are the primary drivers of SOM persistence in these soils.

The main goal of this study was to characterize the binding medium used in easel paintings, by high performance liquid chromatography (HPLC) and gas chromatography coupled to mass spectrometry, with pyrolysis (Py-GC/MS). The results were complemented with infrared microspectroscopy (µ-FTIR). Another purpose of this work was the study of aging process, occurred on paintings, by artificial aging of the ink layers. In this work, were studied three types of binding medium: egg yolk, animal glue and linseed oil. The ink layers were prepared with pure binding medium and/or mixtures with different pigments, like, azurite, red lead and lead white. The µ-FTIR had an important role in the identification of binding medium and pigments used. The results obtained by HPLC, using Pico-Tag method, proved that this technique is useful in proteinaceous material identification. From the analysis…

▼ Many of the current analytical techniques originally developed to characterize conventional reservoir rocks and fluid cannot adequately measure shale and source rocks. An example of this is Retort, where it is not feasible to get sufficient fluid from source rock to make useful measurements. The primary interest of this thesis is the exploration of other analytical techniques, two of which are previously unused in the oil and gas industry. These are Thermal Gravimetric Analysis (TGA), Pyrolysis-Gas Chromatography-Mass Spectrometry (
Py-GC-MS), and Nuclear Magnetic Resonance (NMR). The techniques proposed offer valuable insight into the properties of the rock. TGA gives accurate weight of a sample as temperature is increased,
Py-GC-MS is useful for identifying exact molecules in vaporized fluid, and NMR can be used to characterize viscosity and hydrocarbon chain length. The methods using these techniques can be utilized to further confirm mineralogy of a sample, identify the fluid constituents and quantify their weight, analyze changes in a sample between two different states, and calculate the free fluid saturations of oil and gas in shales. Procedures and results for each of these are presented in this thesis to show methodology and give the reader an idea of its useful applications.

Essential oils extract from aromatics, medicinal and perfume plants, are used since ancient times for their medicinal or olfactory properties. They are quite expensive, and owing to the actual growing demand for getting high quality materials at lowest prices, fraudulent essential oils are frequently found on the market. In order to fight this problem, norms exist (AFNOR, pharmacopeas) which have to be respected. However those techniques are often limited. This work allowed great improves in the quality control of citrus and lavender essential oils. The knowledge of the chemical composition of volatiles and polymethoxy-flavones (lemon), of the chirale ratios of alpha-pinene, le sabinene (citrus), linalool et linalyl acetate (bergamot et lavender), isotope ratio analyses of carbon 13, deuterium, of linalool, linalyl acetate (bergamot et lavender), citral, geranyl acetate (lemon) as wall as isotope ratio analyses of deuterium of nitrogen 15 du Nmethyl anthranilate de methyl (mandarin) permitted the detection of various type of frauds. In order to have an efficient methodology, principal component and discriminant analyses have been proved indispensables. It allowed the identification of fraudulent samples via volatiles molecules study. Chirale analysis and isotope ratio highlight frauds on more “discrete” adulterated samples or a better understanding of adulteration’s process

This essay uses the term derealization to investigate Py Sörman’s novel Aloë. Derealization, the author of this essay claims, consists of an awareness of problems regarding the representation of reality that are central to postmodernism. This awareness gives rise to questions regarding the relationship between narrativity and reality, questions that are present in the modern and postmodern novel. Aloë’s subject and narrativity, the essay argues, highlights these questions. The essay aims to prove this by analysing the novel using concepts from narrative theory. The concluding evaluation of this investigation finds that derealization is not Aloë’s most central subject or theme, but finds that questions regarding derealization are present, that they affect the novel’s representation, and that they form an integral part of the novel’s structure.

▼ In healthy cells, huntingtin is primarily found in the cytoplasm; however, upon cellular stress, huntingtin is phosphorylated (phospho-huntingtin) at serines 13 and 16 of the amino-terminal N17 domain and shuttled into the nucleus. Such dynamism in nucleocytoplasmic translocation and post-translational modification suggests an important role for huntingtin in Huntington’s disease (HD) pathogenesis as these phenotypes propose possible mechanisms for disease progression. Huntingtin nuclear import is also facilitated by its proline-tyrosine nuclear localization signal (PY-NLS), which harbours a highly conserved intervening sequence specific to the huntingtin gene. This encouraged a proteome investigation to identify potential protein partners of the PY- NLS. Results of this study revealed that high mobility group box 1 (HMGB1), a cofactor of base excision repair, uniquely bound to the wild-type PY-NLS, but not the PY-NLS KK177/178AA mutant. Immunofluorescence microscopy in human telomerase reverse transcriptase (hTERT) immortalized fibroblast cells using HMGB1- and phospho- huntingtin-specific antibodies revealed a promising association between the two, as changes in nuclear levels of HMGB1 positively correlated with nuclear levels of phospho- huntingtin. This relationship was further confirmed by co-immunoprecipitation of HMGB1 by the PY-NLS and N17 domain. Also, when exogenous oxidative stress was introduced, increased interaction between HMGB1 and huntingtin was observed. This suggests that HMGB1 facilitates the nuclear import of huntingtin in a ROS-dependent manner, and thus, presents a novel avenue to a potential therapeutic target in HD pathogenesis.

▼ The large pool of carbon (C) stored as soil organic matter (SOM) in soils of high-latitude ecosystems contains more organic C than all global vegetation and the atmosphere combined. Global climate change is expected to have especially pronounced effects in these ecosystems, and even small changes in the accumulation and decomposition of their soil C pool driven by heterotrophic microbial activity could profoundly affect atmospheric CO₂ levels and thus the global climate. Because such changes could trigger drastic shifts in the delicate balance of CO₂ between the biosphere and atmosphere, a better understanding of the key regulators of C cycling is urgently needed.
Using advanced molecular and biochemical techniques, I investigated the availability of C substrates and their utilization by microorganisms under controlled but ecologically relevant conditions in soils representative of the boreal landscape. This molecular characterization of SOM revealed that tree species significantly influence SOM genesis by changing its rate of accumulation and organo-chemical composition. More importantly, a strong connection between SOM decomposition and microbial decomposers was observed and shown to be governed by the organo-chemical composition of the SOM. The structural arrangement of cellulose, and particularly its degree of crystallinity, emerged as a key factor determining rates of cellulose hydrolysis and subsequent C decomposition in boreal forest soils.
This work provides some of the first empirical evidence that soil microbial communities in frozen boreal forest soils can hydrolyze cellulose and use the released substrate for both catabolic and anabolic metabolism. These findings, together with results from peat soil experiments, show that both persistent microbial degradation of C (biopolymers and monomers) and the synthesis of new microbial biomass during winter are widespread features in soils of the boreal landscape. More importantly, the results indicated that small differences in winter soil temperatures can have very large implications for the winter C fluxes of boreal soils.
Over the studied temperature range, C substrates were readily utilized and microbial activity was never totally impeded. However, thermodynamic constraints caused strong reductions in metabolic rates at sub-zero temperatures. The rates of these processes at low temperatures are low but their importance should not be neglected given the spatial scale over which they can occur and the prolonged winters these ecosystems experience. Taken as a whole, this thesis provides a valuable contribution to our understanding of microbial C cycling in one of the world's major soil C pools.

▼ The key interactions between elastomer,
filler, and coupler in uncured and cured compounds were studied by
two complementary analytical techniques, low field 1H solid state
nuclear magnetic resonance (LF NMR) and pyrolysis gas
chromatography / mass spectrometry (
PY GC/MS). LF NMR was utilized
to characterize the macrostructure, and more specifically, the
mobility of the elastomer chains in the polymeric matrix, while
PY
GC/MS provided information on the microstructure of the compounds
analyzed. The networks of the compounds studied
were found to have three components: a tightly bound region (Tr), a
loosely bound region (Tm1), and an extractable unbound rubber
region (Tm2). Based upon the mobility of these three regions and
the peak intensity ratios determined from
PY GC/MS, conclusions
were drawn about the nature of the filler-elastomer interaction in
samples with different polymers (PI and SBR), fillers (CB and
silica), couplers (NXT and Si-75) and cure states (uncured and
cured). It was found that the network of the SBR
compounds was more tightly bound and significantly different from
that of PI compounds regards of cure state, filler type, or coupler
addition. This was attributed to the SBR network having restricted
motion due to the phenyl ring on the backbone of the elastomer
chain causing steric effects. In regards to the
PI compounds, the nature of the interaction between elastomer
chains and the different fillers was determined. Strong physical
adsorption interactions between elastomer and filler dominated when
CB was used as the filler. The addition of coupler to this system
resulted in increased mobility of elastomer chains and higher
concentrations of dimer species. This suggested that the coupler
served as a molecular lubricant between the CB and PI chains and
thus reduced physical adsorption effects. A
strong physical adsorption interaction was proposed between silica
and PI in the uncured state. The addition of coupler in the uncured
state resulted in an increased dispersion of the silica aggregates
and less polarization effects of the silica on the PI chains. Upon
curing, the nature of the interaction between coupler, filler, and
elastomer was confirmed to be chemisorption. Furthermore, a more
developed silica-coupler network occurred with coupler loading
based on the increasing mobility of the elastomer chains in the
loosely bound region and higher dimer concentrations. The
silica-coupler network was found to be the most developed when NXT
was used as the coupler based on the
PY
GC/MS.

▼ The most frequent soils in the São Paulo State Coastal Plain are Podzols, characterized by strong to moderate hydromorphic to well-drained podzolization with very well developed podzol-B horizons (Bh or Bhm). Podzolization processes include the effects of hydrology and rooting on profile morphology and the subsequent effects of improved drainage. A Podzol chrono-hydrosequence was described in detail on a cliff at the south coast of Ilha Comprida, a Holocene barrier island, and allowed a subdivision into four distinct groups: poorly-drained profiles, profiles with well-drained B horizons, strongly rooted profiles and superposed profiles. The morphological description and some observations about the exposure cliff were essential for grouping and differentiating the podzol profiles. Some well-drained podzols have OM-depleted mottles that are related to selective decomposition of organic matter (OM) by microorganisms. Such mottles are frequently
associated to root channels. Seventeen profiles were studied, thirteen had depletion mottles scattered along the profile. Most of these mottles are whitish and are located preferentially in the horizons of transition between the E and B horizons, particularly in conditions of good drainage. Such mottles have certain morphological differences and may be grouped according to similarities in their morphology and their position in the profile. Distinct groups are: (a) concentric OM-depleted mottles; (b) circular/tubular OM-depleted mottles (burrows); (c) dotted OM-depleted mottles; (d) ghost OM-depleted mottles; (e) irregular OM-depleted mottles and (f) Fe-depleted mottles. The chemical composition of soil organic matter was studied in detail using pyrolysis in combination with gas chromatography/mass spectrometry (Py-GC/MS). Samples of all horizons of the distinct profiles studied were taken, as well as from the center of the mottle (M) and from the direct surroundings (S). The
processes involved in the genesis of Podzols in the sandy coastal plain are directly related to drainage, the contribution of dissolved organic matter (DOM), the contribution of organic matter derived from roots, the chemical composition of organic matter and its decomposition by microorganisms, causing a large variation in adjacent Podzols. The well-drained Podzols differ in characteristics from the poorly drained ones in composition and deposition of OM, as well as its decomposition, which is directly related to the activity of groups of microorganisms. They also differ in the relative contribution of OM-derived from roots and DOM. There is a wide variation in the characteristics of decomposition by microorganisms between the profiles of Podzols permanently exposed to air and marine spray (the cliffs) on Ilha Comprida and those inland (pits). There are therefore two main processes that change the morphology of Podzols (OM and composition): (a) change in drainage and rooting, and
(b) exposure to air.
