▼ Despite its negative connotation as a toxic gas, carbon monoxide (CO) is involved in anti-inflammatory, cytoprotective and vasodilatory processes, suggesting a possible therapeutic role in many disease states. Endogenous CO production results in a resting percent carboxyhemoglobin (%COHb) level of 0.5 – 1.5% in humans. In smokers, this can rise to 17%. It has been shown that women who smoke have a lower risk of developing pre-eclampsia (PE), which is thought to be due to an increase in CO. PE is a pregnancy-related disease that is characterized by new-onset hypertension and proteinuria. PE is multifactorial, but a common belief is that poor placentation and systemic maternal endothelial dysfunction are involved in its pathogenesis. Menadione (MD), synthetic vitamin K3, increases CO production in rat brain microsomes. This research tested the hypothesis that MD could also raise CO in the blood and tissue of pregnant mice. A dose-response study was first conducted in non-pregnant mice to determine an efficacious, non-lethal dose. After determining their baseline water consumption rate, non-pregnant CD-1 mice were given one of three doses of MD in drinking water for seven days. No significant differences in %COHb between groups were observed. Splenic CO production and mass doubled in the 4 g/L and 6.5 g/L MD groups, compared to the 1.5 g/L and control cohorts. In a separate experiment, the effect of MD on gestational day (GD) 15 placental tissue CO production was tested using two different tissue preparation methods. Non-linear regression showed that while absolute levels of CO were higher in sonicated placentas than whole placentas, the relative increase in CO between concentrations was the same using either method. Lastly, oral administration of 6.5 g/L MD from GD10.5 – 17.5 significantly decreased maternal weight gain and fetal survival. Significant increases in %COHb and splenic CO were seen in the treatment group on GD17.5, with a positive trend for CO production in the liver, placenta and kidney. Overall, the findings demonstrate that MD can induce CO production in vivo. However, further studies in a mouse model of PE at a safer dose must be done to establish the drug’s therapeutic potential.

The well-established vitamin A supplementation program in children virtually reduces xerophthalmia and mortality in children but offers only a transient impact on raising serum retinol. Regular consumption of vitamin A-fortified food is considered a sustainable approach to maintain a healthy serum response of retinol of vulnerable populations. Bananas dominate the diet of many East-African countries where vitamin A deficiency is prevalent, thus making it a great vehicle for biofortification, a breeding technique to improve vitamin A value in the crop and increase dietary intake of vitamin A. Currently, provitamin A carotenoid (α- and β-carotene) biofortified bananas are being developed through genetic modification but its bioefficacy in humans has not been studied. The dominant provitamin A found in the biofortified bananas is α-carotene, followed by β-carotene. The nonsymmetrical α-carotene provides a molecule of active vitamin A (retinol) and its inactive analog (α-retinol). Exclusion of the inactive α-retinol with limited vitamin A activity is important for accurate determination of vitamin A activity of α-carotene in α-carotene containing foods such as provitamin A biofortified bananas. The overall objective of this study was to accurately determine the bioconversion factor of the α-carotene-containing provitamin A biofortified bananas in humans after the consumption of provitamin A biofortified bananas. An ultra-high selective and sensitive high-performance liquid chromatography–quadrupole-time-of-flight–high-resolution mass spectrometric with electrospray ionization in positive mode (HPLC-ESI (+)-QTOF-HRMS) quantitative method was developed and applied to the measurement of the postprandial appearance of provitamin A carotenoids and their retinyl ester bioconversion products as well as α-retinyl palmitate product of α-carotene after the consumption of provitamin A biofortified bananas. A 3µm C30 carotenoid column was used to separate α- and β-carotene, and their respective retinyl ester bioconversion derivative of α-retinol and retinol (i.e., α-retinyl ester and retinyl ester) in postprandial plasma triacylglycerol-rich lipoproteins. Labeled internal standards (d8-α-retinyl palmitate, 13C10-β-carotene, 13C10-retinyl palmitate) were used to account for analysis variability. Twelve healthy women each consumed three 200-g cooked banana fruit as follows: 1) biofortified banana fruit containing 1415.3 µg (2.64 µmol) of total β-carotene equivalents, 2) wild-type banana fruit with a β-carotene reference dose containing 536.8 μg (1.00 μmol) added β-carotene, and 3) wild-type banana fruit with a vitamin A reference dose containing 262.5 μg retinol activity equivalents (0.92 μmol) added retinyl palmitate. ▼ The studies presented in this dissertation explore Cytochrome P450 family 4 (CYP4) enzyme inhibition and metabolism. The structure-activity relationships between CYP4 enzymes and the potent formamidoxime inhibitor, HET0016, was defined, which led to the development of competitive inhibitors that are selective for CYP4F2 and CYP4F3B and do not inhibit CYP4A11. These CYP4 enzymes are principally responsible for biosynthesis of the signaling eicosanoid, 20-HETE. Additionally, HET0016 was established as a mechanism-based inhibitor of CYP4A11, but not of other CYP4 enzymes. Studies to characterize the kinetic properties of this mechanism based inhibitor and identify a reactive species have shown the inactivation is potent and efficient, does not result in metabolite intermediate complex formation or nitric oxide release, and does not destroy the heme cofactor. A carbodiimide or isocyanate reactive intermediate has been proposed that could result in apo-protein adduction. In the second portion of this thesis, it was established that CYP4F2 and CYP4F11 metabolize vitamin K1 and K2 to their respective ω-hydroxy metabolites, and that CYP4F2 sequentially metabolizes vitamin K to the ω-carboxy metabolites. Microsomal alcohol dehydrogenase/ fatty-aldehyde dehydrogenase (ADH/fALDH) or other P450 enzymes may also play a role in formation of ω-carboxy vitamin K after initial ω-hydroxylation by these CYP4 enzymes. For the first time, the amount of CYP4F11 protein in human liver was quantitated, using a new LC-MS/MS method and it was confirmed that the amount of CYP4F2 protein in human liver correlates with the presence of the common CYP4F2*3 (V433M) allelic variant. Menaquinone-4 (MK4) is one form of Vitamin K2, and this work has shown that metabolism of MK4 is distinctly different from that of Vitamin K1 (phylloquinone). P450 enzymes other than those in the CYP4 family were shown to ω-hydroxylate MK4 in vitro and two new oxidized microsomal metabolites of MK4 were characterized. This work has laid groundwork for future studies that should establish the differences in the metabolic fate of the various forms of vitamin K in vivo and help clarify a genetic basis for the inter-individual variability in response to these supplements when used as pharmacotherapeutic agents. ▼ Provitamin A-biofortified maize has been developed to contribute to the alleviation of vitamin A deficiency (VAD), which is prevalent in the sub-Saharan African region where vitamin A- deficient white maize is a leading staple. The biofortified maize is new compared to white maize and as such its grain properties, including milling, nutritional composition (save provitamin A composition) and sensory characteristics, are barely known. There is a challenge in the adoption of the biofortified maize as a food crop due to its low consumer acceptance, which necessitates more consumer studies. Furthermore, there is a need to develop high value commercial food products using provitamin A-biofortified maize to promote its wide spread utilisation and thereby enhance the vitamin A status of the population. The milling and nutritional properties of grains of 34 varieties of provitamin A-biofortified maize were assessed relative to a white variety (control/reference). The milling properties of the biofortified varieties as indicated by the milling index (69.9-112.1) and hectolitre mass (65.8-82.9 kg/hl) were better compared to 93.5 and 78.5 kg/hl of the white variety, respectively. The ash content of one biofortified maize variety PVAH 48 was comparable to that of the white variety (1.02 g/100 g), whilst five biofortified varieties showed significantly high Fe content (25.67-70.33 mg/kg) compared to the white variety (20.67 mg/kg). The protein (9.8-12.8 g/100 g) and lysine (0.16-0.37 g/100 g) content of the biofortified varieties were significantly high compared to 10.5 g/100 g and 0.21 g/100 g of the white variety, respectively. The sensory quality of stiff porridges made with provitamin A maize varieties were evaluated using descriptive analysis and the 5-point facial hedonic test by a trained panel and an untrained consumer panel, respectively. The provitamin A maize porridges were described as having a cooked maize flavour and aroma, sticky, fine with low intensity of residual grain and slight bitter aftertaste. Provitamin A carotenoid retention in the porridges was determined. Provitamin A carotenoid retention in the porridges was considerably high (91-123%). Relative to white maize porridge, the biofortified porridges were fairly acceptable, although their acceptability seemed to be reduced by their stickiness and bitter aftertaste. Leaf powder of Amaranth (Amaranthus cruentus), a vegetable widely consumed by rural communities in Southern Africa and reported to have good nutritional and health-promoting properties, was used to partly replace flours of four biofortified maize varieties at 0%, 1% and 3% (w/w) and extruded into snacks. The effects of Amaranth addition on the quality and health-promoting potential of the snacks was assessed, as well as the physical and sensory quality. The physical and sensory qualities of the extruded snacks, in terms of texture and expansion, tended to decrease with increasing Amaranth concentration. ▼ min A homeostasis control on different levels is essential to maintain retinoid function in peripheral tissues and to ensure optimal vision and requires a well-coordinated team of metabolic enzymes and transport proteins. The production of vitamin A from a chemically diverse set of over 60 putative provitamin A precursors poses a metabolic challenge. We scrutinized enzymatic properties of the two carotenoid cleavage dioxygenases (CCDs), BCO1 and BCO2, which catalyze the metabolism of carotenoids. Recombinant BCO1 split substrates at the C15,C15’ position adjacent to a canonical but not substituted ß-ionone ring site to vitamin A aldehyde, and was not impeded by removal of methyl groups from the substrate backbone. BCO2 split substrates at the C9,C10 position adjacent to assorted ionone ring sites. Kinetic analysis revealed a higher catalytic efficiency of BCO2 with substrates bearing non-canonical ionone rings. Accordingly b- cryptoxanthin, with one canonical and one non-canonical ionone ring site, wasxixexclusively converted by both CCDs to retinoids in mice. Homology modeling and site directed mutagenesis identified amino acid residues at the entrance of the BCO1 substrate tunnel which determined its substrate specificity. Thus, selectivity of CCDs for distinct ionone ring sites guarantees production of vitamin A.To exert its effects, vitamin A must be distributed and transported across cell membranes, a process mediated by the STRA6 protein. Impaired vitamin A transport and homeostasis are associated with type 2 diabetes and Matthew Wood Syndrome. STRA6’s role in vitamin A homeostasis is defined. Stra6-/- mice were raised on diets sufficient or insufficient for vitamin A, with heterozygous sibling controls. Vitamin A levels of eyes, brain and testes, which highly express Stra6, as well as lung and fat, were measured, and consequences of STRA6 deficiency on retinoid-dependent processes were determined. Furthermore, effects of STRA6 deficiency on retinoid homeostasis of the aging mouse were determined. Thus STRA6 is critical for the transport of vitamin A over blood-tissue barriers. Fat and lung rely on dietary vitamin A. In testis and brain, Stra6 expression was regulated by vitamin A, reducing vitamin A consumption in controls when the dietary supply was limited, sequestering it for the eye. ▼ Improving the nutritional content of staple crops through breeding (biofortification) is promoted as cheap and sustainable approach for alleviating vitamin A and other micronutrient deficiencies across the world. Maize is one of the target crops for provitamin A biofortification, since it is a major staple for millions of people in developing countries, and has a great genetic potential for accumulating provitamin A in its grain. This PhD study addressed three research objectives aimed at accelerating the biofortification of maize with provitamin A. To achieve the objectives, a diverse set of inbred lines developed from different crosses having mixed genetic background of temperate and tropical germplasm were employed. In the first research objective, the efficiency of PCR based functional markers proposed for high provitamin A maize breeding was investigated. Out of eight polymorphisms for three key carotenoid biosynthesis genes tested, the 3'TE and 5'TE polymorphisms of the gene crtRB1 showed the strongest association. About 18 percent of the inbred lines studied had the favourable alleles for these two polymorphisms. The second research objective assessed the association of genome-wide SNP markers with provitamin A and non-provitamin A carotenoids using GWAS and pathway level association analysis. This study detected known major and minor effect carotenoid genes (lcyE, crtRB1, ZEP1 and others), plus novel loci, such as ARF20, that may be involved in regulation of carotenoid biosynthesis in maize endsosperm. In the last research objective 24 yellow maize inbred lines were inter-crossed using factorial mating scheme for studying their combining abilities and heterotic effects for carotenoids and key agronomic traits. The result demonstrated the predominance of additive genetic effects in the inheritance of endosperm carotenoid content. Small percentage of non-additive genetic effect was also detected for provitamin A which is important for expression of heterosis in the trait. Hybrids combining good agronomic performance and high provitamin A content were identified. This study will serve as a basis for establishing heterotic pattern in IITA's yellow maize germplasm which is crucial for development of hybrid and synthetic varieties that can meet existing and emerging demands, while incorporating high level of provitamin A in the grain. ▼ Biofortification is a food-based intervention to combat nutrient deficiencies, including vitamin A deficiency (VAD), by increasing the levels of target nutrients in crops through traditional (conventional breeding) and modern genetic manipulation methods. Maize has been selected for biofortification with provitamin A to alleviate the prevalence of VAD in sub-Saharan Africa where white maize, which is devoid of vitamin A, is a leading staple. However, when compared to white maize, provitamin A biofortified (yellow) maize (PABM), consumers in several countries in sub-Saharan Africa found it less acceptable, largely due to their negative perceptions of yellow maize and its unfamiliar sensory properties. A combination of strategies such as food product development, consumer awareness campaigns, and nutrition education could improve consumer acceptance of yellow maize. Two provitamin A biofortified (yellow) maize food products, phuthu (crumbled porridge) and jeqe (steamed bread), and their corresponding white maize products (controls), were evaluated for their acceptability. Consumer acceptability tests were conducted with a consumer sample of 68 untrained panellists of the age range 18-85 years. A 5-point smiley pictorial Hedonic scale was used to evaluate the sensory acceptability of samples of PABM phuthu and Jeqe. The two food products were selected mainly because of their popularity amongst the KZN community. The results showed low acceptability of yellow phuthu compared to white phuthu, whilst the acceptability of yellow jeqe was similar to that of white jeqe. It was not clear why the acceptability of yellow phuthu was lower than that of white phuthu. Therefore, a descriptive sensory analysis was performed to characterise the sensory attributes of yellow phuthu and thereby reduce the influence of its sensory attributes on its acceptability. Eleven trained panellists analysed the sensory properties (attributes) of phuthu made from three varieties of provitamin A biofortified maize hybrids. Descriptive sensory analysis data were subjected to ANOVA, Fisher’s Least Significant Difference (LSD) tests, and Principle Component Analysis (PCA). The results showed that the yellow phuthu samples were characterised by lower intensity of chewiness, crumbliness, roughness, white specks, and had less malleability. The control phuthu had a lower intensity of stickiness and yellow colour compared to the yellow phuthu. The carotenoid pigments in the yellow phuthu were probably responsible for the yellow colour of the biofortified maize phuthu and its stickiness. It is necessary to reduce the intensity of the stickiness of yellow maize phuthu to enhance its acceptability. To change the negative perceptions and lower acceptability of provitamin A biofortified maize compared to white maize, a provitamin A awareness campaign was conducted. Bakgrund: Efter utvecklingen av Bokföringsnämndens K-regelverk, K2 och K3, diskuterades olika för- och nackdelar avseende regelverken bland experter inom redovisningsprofessionen. För mindre företag med möjligheten att välja mellan K2- och K3-regelverket blev valet således inte självklart. En påverkande faktor som redovisningsprofessionen benämner som central är tillväxt, men trots detta är variabeln relativt outforskad. Fortsättningsvis är fastighetsbranschen en av de verksamhetsgrenar som riktade stark kritik mot införandet på grund av regleringen avseende de tvingade komponentavskrivningarna och branschen karaktäriseras även av hög tillväxt. Majoriteten av tidigare forskning studerar större företag och få undersöker tillväxt i relation till redovisningsval, vilket föranleder att denna studie har mindre företag som utgångspunkt för att utforska området ytterligare.Syfte: Syftet med denna uppsats är att bidra med vidare förståelse för företags redovisningsval avseende K2 och K3, och vad som påverkar valet. I denna kontext undersöker studien huruvida det föreligger något samband mellan ett företags tillväxt och dess redovisningsval avseende regelverk. Metod: Undersökningen utgår från en deduktiv ansats och tillämpar en kvantitativ forskningsmetod. Utifrån den teoretiska referensramen formuleras hypoteser som sedan testas av ett Chi-två testet.Slutsats: Studien visar att en antydan till samband mellan mindre företags val av regelverk och tillväxt som påverkande faktor. Däremot kan det konstateras enligt undersökningens statistiska test att det inte föreligger något statistiskt signifikant samband.

Background: After Bokförningsnämndens development of the K-regulations, K2 and K3, different pros and cons were discussed concerning the regulations amongst experts within the accounting profession. For the smaller businesses with the opportunity to choose between K2 and K3, the decision was therefore not obvious. An affecting factor that the accounting profession claims to be important is growth, however the variable has remained relatively unexplored. Huvudsyftet med denna studie är att ta reda på om mindre onoterade aktiebolag som arbetar med forskning och utveckling valt att tillämpa K2- eller K3-regelverket. Delsyftet är att undersöka vilka faktorer som kan inverka på valet. En förhoppning med studien är att denna kan vara till hjälp för de företag som står inför valet mellan K2 och K3. I studien har en kvantitativ metod tillämpats. Empiriskt material har samlats in genom årsredovisningar, via databasen Retriever samt via kontakt med ansvarig person på företagen i urvalet. Insamlad data har sedan analyserats med hjälp statistiska analyser. Variabler som kan tänkas påverka valet av regelverk har tagits fram utifrån redan erkända teorier samt ur tidigare forskning. Variablerna presenterats i en a priori-modell. En undersökningsmodell har sedan utformats då alla tänkbara variabler ur den teoretiska referensramen inte är möjliga att undersöka på grund av metodval. Resultatet i denna studie visar på att övervägande del av företagen valt att tillämpa K3-regelverket trots att det är K2-regelverket som är det regelverk som rekommenderas för denna kategori av företag. Dessutom gjorde studiens tidsram det omöjligt att samla in fler årsredovisningar än de som inkom, detta gav ett stort bortfall som troligtvis påverkat studiens resultat. En liknande studie i höst skulle ge ett bättre empiriskt material och därmed förhoppningsvis ett generaliserbart resultat.

The main objective of this study is to find out the reasons why small and limited companies working with R&D would decide to apply K2 or K3 regulations and to investigate what factors may influence that choice. One hope of the study, if completed successfully and thoroughly, is that it may serve as a guide in order to help companies faced with this choice. The study is based on a quantitative method. Empirical data was collected through annual reports, through the database, Retriever, and by having contact with the CFO. Further, the empiric data was analyzed using statistical analysis and the variables that could affect the choice of framework were found in acknowledged theories as well as previous research regarding accounting choices. The variables are presented in a “A priori-model”. A research model was formed since all possible variables from the theories could not be investigated due to methodological choices. The empirical data has been presented in the form of tables, charts and then discussed regarding previous research. The results of this study show that the majority of the companies decided to apply the K3 regulations, even though the K2 regulation were recommended for that category of company. In addition, the study's timeframe made it impossible to collect any more annual reports than what was received, this may affect the results of this study. A similar study in the fall would give better empirical data.

▼ Zaradi vse večje težnje po razvoju zelenih tehnologij, ki ne obremenjujejo okolja, in postopkov procesiranja materiala, ki ne vsebujejo sledi toksičnih organskih topil, kot je bilo to običajno pri konvencionalnih postopkih, smo se osredotočili na superkritične fluide, ki se v industriji vedno bolj uveljavljajo. Doktorska disertacija je namenjena pridobivanju podatkov o faznih ravnotežjih in topnostih za načrtovanje visokotlačnih procesov, kjer se uporabljajo superkritični fluidi. Proučili smo sisteme polietilen glikola (PEG) z molskimi masami 1 500 g/mol, 4 000 g/mol, 10 000 g/mol, 35 000 g/mol s fluidi argon, propan in žveplovheksafluorid (SF6), vitamin K3/ogljikov dioksid in naftol/ogljikov dioksid. Na podlagi eksperimentov za sisteme PEG/fluid smo ugotovili, da imajo različni fluidi (CO2, propan, SF6, argon) različne termodinamske in transportne lastnosti v binarnem sistemu. Naredili smo obsežno študijo proučevanja topnosti, difuzijskih koeficientov, temperatur tališč, časov desorpcij itd. Vsak fluid ima zaradi svojih edinstvenih lastnosti različen vpliv na binarni sistem PEG/fluid in na lastnosti kot so temperatura tališča, topnost in difuzijski koeficient, zaradi česar lahko npr. pridobivamo tudi produkte z različnimi karakteristikami. Te podatke smo proučili za načrtovanja procesa mikronizacije PEG s procesom PGSS (Particles from Gas Saturated Solutions oz. delci iz raztopin nasičenih s plinom). Ugotovili smo, da je PEG možno mikronizirati s fluidi propan, SF6 in argon pri nižjih tlakih kot s CO2. Mikronizacijo smo izvedli pri pogojih minimalnega tališča PEGa pod tlakom fluida in pri pogojih 328 K in 15 MPa. Poleg tega smo pridobili delce, ki so bolj enakomernih velikosti in pravilnejših krogličastih oblik kot pri mikronizaciji s CO2. Delci, ki so enakomernih oblik in velikosti, so atraktivni za proizvajalce trdnih, praškastih produktov, saj so višje kvalitete. Sistem vitamin K3/CO2 smo proučili pri temperaturah 313 K, 333 K in 353 K in tlakih do 40 MPa ter primerjali z literaturo. Rezultati kažejo, da se topnosti vitamina K3 razlikujejo od podatkov v literaturi, kar pripisujemo uporabi različnih metod vzpostavljanja ravnotežja, vzorčenja in analize. Določili smo tudi parcialne molske volumne, ki so izrazito veliki in negativni pred in v bližini kritične točke, kar kaže na močne interakcije med molekulami topila in topljenca. Z določevanjem parcialnih molskih volumnov lahko sklepamo tudi o pojavu maksimuma topnosti, ki se v primeru topnosti vitamina K3 v CO2 pojavi pri temperaturi 313 K in 333 K. Pri določevanju gostot raztopine vitamina K3 v CO2, smo ugotovili, da so razlike med gostoto binarnega sistema vitamin K3/CO2 in čistega fluida CO2 lahko tudi 100 % ali več. Tališče naftola pod tlakom CO2 ima temperaturni minimum pri tlaku 20 MPa. Topnosti 1-naftola v CO2 pri temperaturah 313 K, 333 K in 353 K so v večini višje od topnosti 2-naftola v CO2. På redovisningsområdet pågår det ständigt förändringar och under den senaste tiden har stort fokus legat på BFN. Nämnden har sedan 2004 arbetat med det så kallade K-projektet där målet är att ta fram samlade regelverk för företag i olika form och storlek. I denna uppsats ligger fokus på K3-regelverket som huvudsakligen är anpassat för större onoterade aktiebolag samt ekonomiska föreningar. Tanken är att regelverket ska baseras på IFRS for SMEs som är IASBs regelverk för små och medelstora företag. I dagsläget finns inget samlat regelverk för dessa bolag i Sverige för upprättande av årsredovisning. Syftet med K3 är att det ska förenkla redovisningsarbetet och på det sättet minska de administrativa kostnaderna. I slutet av år 2010 skickade BFN ut K3 på remiss till 52 instanser. Av dessa valde 28 att skicka in remissvar av olika omfattning. Vi har behandlat 13 utav dessa eftersom resterande inte varit tillräckligt innehållsrika för att bidra med relevant information till uppsatsen. Syftet med uppsatsen har varit att undersöka och redogöra för vilka huvudargument som de olika instanserna har yttrat i sina remissvar gällande K3-regelverket. Studien är av kvalitativ karaktär med en induktiv ansats där empirin har bestått av de utvalda remissvaren. Instanserna har framfört konstruktiv kritik av olika karaktär. Huvudargumenten har vi delat in i fyra olika kategorier. Dessa har varit användarvänlighet, förhållandet till svenska lagar, förhållandet till IFRS for SMEs samt förhållandet till god redovisningssed. Resultatet av studien visar att en stor majoritet anser att användarvänligheten i K3 är undermålig och att strukturen måste förbättras för att kunna tillämpas i praktiken. Flera instanser menar även att BFN i sitt arbete att utforma K3 tagit sig stora friheter att inskränka på regler som anges i ÅRL. I vissa fall har instanser yttrat att normerna i K3 strider mot regeringsformen och ifrågasatt BFNs skrivelser. Positivt ser instanserna dock på att nämnden valt att basera K3 på IFRS for SMEs men att det i flera fall sker avvikelser från det internationella regelverket vilket inte välkomnas av ett antal instanser. Vår slutsats är att K3 i dagsläget inte är tillräckligt välutvecklat för att kunna tillämpas i praktiken eftersom majoriteten av instanserna har sammantaget fler negativa än positiva synpunkter kring regelverket.

År 2004 beslutade BFN att inleda en ny inriktning i sitt normgivningsarbete som kom att benämnas K-projektet. Detta projekt innefattar fyra särskilda ramverk som inkluderar kategorierna K1-K4. K-regelverken omfattar icke noterade företag som är bokföringsskyldiga och nämnda kategorier skall tillämpas beroende på företagsform och företagsstorlek. Regelverket K3 skickades ut på remiss i juni år 2010 och skall vara huvudregelverk för icke noterade företag som avslutar räkenskapsåret med en årsredovisning. IASB publicerade år 2009 ett internationellt redovisningsregelverk som erhållit namnet IFRS for SMEs. I likhet med K3 är detta regelverk även avsett för icke noterade små och medelstora företag som upprättar årsredovisning. Studien har sin utgångspunkt i BFNs påstående om att reglerna i K3, med anpassning till ÅRL och IL, är utformade efter det internationella regelverket IFRS for SMEs. Syftet med studien är att undersöka om det svenska regelverket K3 bygger på det internationella regelverket IFRS for SMEs, genom att utföra en komparativ studie. Studien är utformad enligt en induktiv ansats och är av kvalitativ karaktär. Den teoretiska referensramen innehåller en beskrivning av K3 och IFRS for SMEs och dess normgivande organ. Undersökningen omfattar en jämförelse av sex utvalda kapitel i K3 och motsvarande kapitel som i IFRS for SMEs behandlar likartade områden. Likheter och olikheter regelverken emellan lyfts fram och analyseras med syfte att undersöka om K3 bygger på regler och principer i IFRS for SMEs. Studien visar att K3 i mångt och mycket är utformat i enlighet med IFRS for SMEs och därmed verifierar vi BFNs påstående. Likheter regelverken emellan överväger olikheter och innehållet i K3 skiljer sig från IFRS for SMEs endast i de avseenden K3 anpassats till svensk lag. Vi anser att K3 är ett bevis på att Sverige strävar efter ökad jämförbarhet. Inflytandet av ÅRL och IL i K3 förhindrar dock en fullständig tillämpning av internationella regler och principer. Vi förordar ökad internationell jämförbarhet på redovisningsområdet och förespråkar en lagförändring som tillåter Sverige att i större utsträckning vara en del av ett gemensamt redovisningsspråk.

Historically, vitamin K is known for its role in blood clotting; however in recent years additional functions have been identified including a role in sphingolipid metabolism and as an anti-inflammatory agent. Studies have shown correlations between low vitamin K status and poor cognitive function while several biomarkers of inflammation are inversely correlated with cognition. The ELDERMET cohort, a group of well characterised subjects aged ≥ 64 years, were used in this study. The aim of this study was to identify potential associations between vitamin K status and cognition as well as any correlations between biomarkers of inflammation and cognition using a subgroup of the ELDERMET cohort. In addition the study aimed to ascertain significant differences in variables across living stratification i.e. the community dwelling subjects versus those in long term care. Subjects were divided into 4 groups based on their Mini Mental State Exam (MMSE) score. Vitamin K status was determined by measuring dietary and serum phylloquinone (vitamin K) status. Percent undercarboxylated osteocalcin (% ucOC) was also measured in serum as a long-term indicator of functional vitamin K status. Biomarkers of inflammation including high sensitivity C-reactive protein (hsCRP), Interleukin 6 (IL-6), Interleukin 8 (IL-8) and Tumour Necrosis Factor alpha (TNFα) were measured. Analysis of variance (ANOVA) revealed significant differences between markers of vitamin K status and inflammation and MMSE categories. Positive correlations were found between MMSE and serum and dietary phylloquinone. Inverse correlations were found between MMSE and other variables including %ucOC, hsCRP, IL-6, IL-8 and TNFα. Similarly, significant differences were found across living stratification for biomarkers of inflammation. Analysis of covariance (ANCOVA) showed a significant difference in dietary phylloquinone intake between those with severe cognitive impairment and those with normal cognitive function, and between those with moderate cognitive impairment and normal functionality. In addition, a significant difference between IL-6, TNFα and hsCRP was found especially between those with poor cognitive function and those with normal cognitive ability. This study adds to the growing body of evidence which suggests a role for vitamin K in cognitive function. Findings from this study have the potential to help inform public health policy and dietary recommendations aimed at promoting healthy aging.

▼ Diet and physical activity are two modifiable factors that can curtail the development of osteoporosis in the aging population. One purpose of this study was to assess the differences in dietary intake and bone mineral density (BMD) in a Masters athlete population (n=87, n=49 female; 41.06 ± 5.00 years of age) and examine sex- and sport-related differences in dietary and total calcium and vitamin K intake and BMD of the total body, lumbar spine, and dual femoral neck (TBBMD, LSBMD and DFBMD, respectively). Total calcium is defined as calcium intake from diet and supplements. Athletes were categorized as participating in an endurance or interval sport. BMD was measured using dual-energy X-ray absorptiometry (DXA). Data on dietary intake was collected from Block 2005 Food Frequency Questionnaires (FFQs). Dietary calcium, total calcium, or vitamin K intake did not differ between the female endurance and interval athletes. All three BMD sites were significantly different among the female endurance and interval athletes, with female interval athletes having higher BMD at each site (TBBMD: 1.26 ± 0.10 g/cm2, p<0.05; LSBMD: 1.37 ± 0.14 g/cm2, p<0.01; DFBMD: 1.11 ± 0.12 g/cm2, p<0.05, for female interval athletes; TBBMD: 1.19 ± 0.09 g/cm2; LSBMD: 1.23 ± 0.16 g/cm2; DFBMD: 1.04 ± 0.10 g/cm2, for female endurance athletes). Male interval athletes had higher BMD at all three sites (TBBMD 1.44 ± 0.11 g/cm2, p<0.05; LSBMD 1.42 ± 0.15 g/cm2, p=0.179; DFBMD 1.26 ± 0.14 g/cm2, p<0.01, for male interval athletes; TBBMD 1.33 ± 0.11 g/cm2; LSBMD 1.33 ± 0.17 g/cm2; DFBMD 1.10 ± 0.12 g/cm2 for male endurance athletes). Dietary calcium, total daily calcium and vitamin K intake did not differ between the male endurance and interval athletes. This study evaluated the relationship between calcium intake and BMD. No relationship between dietary or total calcium intake and BMD was evident in all female athletes, female endurance athletes or female interval athletes. In all male athletes, there was no significant correlation between dietary or total calcium intake and BMD at any of the measured sites. However, the male interval athlete group had a negative relationship between dietary calcium intake and TBBMD (r=-0.738, p<0.05) and LSBMD (r=-0.738, p<0.05). The negative relationship persisted between total calcium intake and LSBMD (r=-0.714, p<0.05), but not TBBMD, when calcium from supplements was included. The third purpose of this study was to evaluate the relationship between vitamin K intake (as phylloquinone) and BMD. In all female athletes, there was no significant correlation between vitamin K intake and BMD at any of the measured sites. No relationship between vitamin K and BMD was evident in female interval or female endurance athletes. Similarly, there was no relationship between vitamin K intake and BMD in the male endurance and interval groups. Nosso laboratório, nos últimos anos, tem investido na caracterização de intermediários e produtos finais da via de isoprenóides. Baseando-se na presença das vias 2-C-metil-D-eritritol 4-fosfato e do chiquimato, decidimos verificar se ocorriam as biossínteses de retinóides e vitamina K no ciclo intraeritrocitário do P. falciparum, através da análise de produtos metabolicamente marcados com [1(n)-3H]-pirofosfato de geranilgranila por análises cromatográficas (HPLC e TLC), além da espectrometria de massas. Não identificamos a presença de retinal, retinol e ácido retinóico no ciclo intraeritrocitário do P. falciparum. Já a biossíntese de vitamina K precisa ser estudada com mais profundidade, pois há indícios de biossíntese, em especial da menaquinona-4.

Malaria is one of the most important parasitic diseases in the world. The spread of resistance to the antimalarials impairs the parasite\'s control. Therefore, is necessary the discovery of new metabolic routes to allow new antimalarials development. Our group has been studying the isoprenoid pathway, characterizing the intermediate and secondary products of this pathway. Based on the presence of 2-C-methyl-D-erythritol 4-phosphate and shikimate pathways, we decided to investigate the occurrence of retinoids and vitamin K biosynthesis in P. falciparum, through the chromatography of [1(n)-3H]Geranylgeranylpyrophosphate labeled products of intraerythrocytic stages of P. falciparum, and mass espectometry analysis. The retinal, retinol and retinoic acid were not identified in P. falciparum. The results indicate the menaquinone-4 biosynthesis, although deeper investigation is necessary.

▼ The studies that are presented in this dissertation have; (i) established a mechanism by which warfarin dose response is modulated by VKORC1 genotype under optimized conditions for kinetic analysis of human vitamin K epoxide reductase (VKOR) catalytic activity, (ii) evaluated previously suggested irreversible and reversible mechanisms of VKOR inhibition by vitamin K antagonists (VKAs), and (iii) elucidated the role of tyrosine 139 in the catalytic center and inhibitor binding site of VKOR. Firstly, observation of VKOR activity in human liver microsomes (HLMs) under varied experimental conditions demonstrated that human VKOR is a highly labile enzyme. Under optimal conditions, evaluation of genotyped HLMs for downstream effects of altered VKORC1 mRNA expression revealed a relationship between the VKORC1 B haplotype and increased the catalytic activity and protein expression of VKOR. This demonstrates that warfarin dose is regulated through a transcriptional mechanism wherein higher levels of VKORC1 mRNA translate to increased protein expression and associated enzymatic activity of VKOR, thus requiring a higher dose of warfarin to maintain the same target level of anticoagulation. Secondly, the co-existence of irreversible and reversible modes of VKOR inhibition was re-investigated by the dilution method with the addition of two treatments, 20 mM DTT and 4% BSA, aimed specifically at stringent removal of inhibitor from microsomes. A post-dilution evaluation of VKOR activity in bovine liver microsomes that had been treated with 4-hydroxycoumarin and 1,3-indanedione VKAs demonstrated that they are not irreversible inhibitors, but appear to be tight-binding compounds that exhibit non-competitive type reversible kinetics with respect to vitamin K epoxide (KO). Finally, a recombinantly expressed VKOR carrying a tyrosine→phenylalanine mutation, which was made to test the importance of Y139 as a base or proton donor in the mechanism of KO reduction, had no effect on steady-state kinetic parameters for the formation of vitamin K1 or 3-hydroxyvitamin K (a putative side-product). Any mutation of Y139, however, generated a warfarin-resistant enzyme. Therefore, although this residue is located proximally to the catalytic site of VKOR, we conclude that Y139 is not directly involved in the mechanism of substrate turnover, but has a pivotal role in warfarin binding and inhibition. V doktorski dizertaciji smo raziskovali uporabo ogljikovega dioksida pri visokih tlakih za impregnacijo maščobo-topnih vitaminov. Za impregnacijo smo izbrali vitamina K3 in D3. Predvsem vitamin D3 je izjemno občutljiva substanca, zato zahteva izjemno pozornost. Za impregnacijo obeh vitaminov smo uporabili polisaharidne arogele. Aerogeli so porozni materiali z izjemnimi lastnostmi. Zaradi visokih poroznosti imajo velike specifične površine in nizke gostote. Če te lastnosti združimo še z biokomatibilnostjo ter biorazgradljivostjo polisaharidov, dobimo idealne materiale za farmacevtske aplikacije. Cilj doktorske dizertacije je bil impregnacija vitaminov v polisaharidne aerogele, da bi jih s tem dodatno zaščitili ter tako povišali njihovo stabilnost. Delo je razdeljeno v dva večja dela. V prvem delu smo uporabili superkritični ogljikov dioksid za impregnacijo vitamina K3 in vitamina D3. Vitamin K3 smo uporabili kot modelno substanco, naš glavni cilj pa je bil impregnacija vitamina D3. Raziskali smo termodinamske in kinetične lastnosti obeh vitaminov. Dobljene vrednosti smo razložili z uporabo že razvitih kinetičnih modelov. V prvem delu dizertacije smo določili tudi strukturne lastnosti vzorcev ter preverili sproščanje vitamina in vitro. V drugem delu doktorske dizertacije smo preiskovali impregnacijo vitamina D3 z ogljikovim dioksidom. Tokrat smo uporabili ogljikov dioksid pri različnih pogojih, od superkritičnega do tekočega ogljikovega dioksida. Preiskovali smo kinetično obnašanje pri različnih pogojih ter določili stopnjo degradacije vitamina glede na izbrane pogoje. Z različnimi karakterizacijskimi metodami smo določil vpliv tako superkritičnega kot tekočega ogljikovega dioksida na končne strukturne lastnosti aerogelov. V tem delu dizertacije smo prav tako izvedli sproščanje vitamina D3 in vitro, izvedli pa smo tudi stabilnostno študijo za potrditev povišane stabilnosti vitamina D3 ob impregnaciji v aerogele.

This doctoral dissertation examines utilization of carbon dioxide as solvent for high pressure impregnation of fat-soluble vitamins, in particular vitamin K3 and vitamin D3. Chosen vitamins, especially vitamin D3, are handling challenging since they are highly sensitive substances. Polysaccharide alginate aerogels were used as vitamins’ carriers. Aerogels are airy materials with outstanding properties such as high porosity, high specific surface area and low density. Combined with the biocompatible and biodegradable polysaccharides, they fulfil pharmaceutical demands for ideal candidates. General idea of this dissertation was to achieve protection and prolonged stability of sensitive vitamins against environments by impregnating them into polysaccharide aerogels. The work is divided in main two parts. In the first part, carbon dioxide was used in its supercritical state. The impregnation experiments were conducted with both vitamin K3 and vitamin D3.
