▼ Clostridium perfringens causes necrotic enteritis (NE), resulting in decreased feed efficiency and increased mortality, costing the poultry industry USD 2 billion a year worldwide. The objective of the first trial was to validate the Fung Double Tube (FDT) to detect and enumerate C. perfringens in chicken intestines. Two methods (FDT and petri plates) and three media (Shahidi Ferguson Perfringens [SFP] with egg supplement, polymyxin B [p], and kanamycin [k; E]; SFP with p and k [P]; and SFP with cycloserine [C]) were arranged in a 2 x 3 factorial, resulting in six treatments. The FDT with medium C (5.35 log CFU/g) had significantly (P<0.05) higher C. perfringens counts than any other media/method combination. The objective of the second and third trials was to determine the effect of diet type on the population of C. perfringens in broiler intestines using the FDT. Trial 2 tested: corn-soybean meal (SBM), low-crude protein (19.8%)/high synthetic amino acids (SAA), and barley (56%)-fishmeal (4%; BF). Diets in Trial 3 included: corn-SBM, barley (7.46%), fishmeal (4%), and BF. Diets in Trial 2 contained an antibiotic and a coccidiostat; diets in Trial 3 did not. After 21 days, birds in Trial 2 fed BF had significantly higher (P<0.05) counts (5.96 log CFU/g) of C. perfringens, as compared to all other diets. Both, corn-SBM and SAA diets resulted in 3.89 log CFU/g. In Trial 3, birds fed the corn-SBM diet (2.7 log CFU/g) had significantly lower (P<0.05) counts than broilers fed BF (4.15 log CFU/g). When broilers were fed fishmeal (3.583 log CFU/g) and barley (3.577 log CFU/g) separately, C. perfringens counts were numerically higher compared to the corn-SBM diet, but numerically lower than birds fed BF. Barley and fishmeal inclusion increased the incidence of C. perfringens, and their combination resulted in a cumulative effect. The FDT method is able to detect C. perfringens at higher levels than the conventional petri plate method (P<0.001) and it also proved to be an effective method to detect differences in C. perfringens counts from the intestines of chickens fed different diet. Alle drei Filme behandeln politische Themen, woraus in dieser Arbeit eine engagierte Haltung der beiden abgeleitet wird, die sich fiktionalisierender Methoden und dem Genre des Political Fiction Films bedienen, um mit den präsentierten Inhalten ein großes Publikum zu erreichen. Die Analyse des Films Z zeigt auf, dass die historische Lambrakis-Affäre als Grundlage für einen Film diente, der darauf abzielt, die Mechanismen einer politischen Verschwörung aufzudecken, ohne sich dabei auf eine konkrete historische Situation festlegen zu lassen. Durch diese Herangehensweise wurden zahlreiche Interpretations- und Identifikationsmöglichkeiten eröffnet. L’Aveu setzte sich mit den stalinistischen Schauprozessen auseinander und war von allen an seiner Produktion Beteiligten als Film mit kathartischer Wirkung für die kommunistische Partei intendiert, wenn er auch von vielen nicht in dieser Art rezipiert wurde. Die Abfassung des Drehbuchs auf der Basis der Memoiren des ehemaligen stellvertretenden Außenministers der Tschechoslowakei, Artur London, ermöglichte Jorge Semprún die Auseinandersetzung mit seiner eigenen kommunistischen Vergangenheit. Der Film Section Spéciale, der sich mit der Kollaborationsregierung General Pétains mit einem der dunkelsten Kapitel der französischen Geschichte beschäftigt, hatte geringeren Erfolg als seine beiden Vorgänger. Die Ursachen dafür werden in der geringeren filmischen Attraktivität als filminterne Komponente sowie in der ablehnenden Haltung der Franzosen gegenüber der im Film spürbaren Kritik als filmexterne Komponente erkannt. Der Vergleich der Filme mit Semprúns Prosa erschließt den a-chronologischen, assoziativen Handlungsverlauf als verbindendes formales Element sowie den Primat der Wahrscheinlichkeit vor der Wahrheit als durchgängiges gestalterisches Prinzip. Im letzten Kapitel werden thematische Parallelen zwischen den drei Filmen und Semprúns literarischem Werk untersucht. Dabei kristallisieren sich Antitotalitarismus und Kommunismus, Antisemitismus, die stalinistischen Schauprozesse, das revolutionäre Leben sowie die tendenzielle Nähe zu Film und filmischen Techniken als Schwerpunkte heraus.

Manipulacija v medijih postaja aktualna tema različnih sodobnih raziskav. V magistrski nalogi so predstavljeni različni načini uporabe in učinkov manipulacije v javnih občilih in množičnih medijih. Del njenega delovanja sta tudi senzacionalizem in spektakel, ki sta nadomestila novinarski vrlini objektivnost in resnicoljubnost. Naloga pa med drugim obravnava (manipulativno) vlogo oglaševanja in odnosov z javnostmi v sodobnem novinarstvu in situacije, ko medije obvladuje povečanje moči kapitala nad javnim dobrim. Trdimo, da so v nekaterih primerih mediji spremenili svojo vlogo iz obveščevalca v prodajalca. Temu v veliki meri pripomorejo tudi novinarji, ki pri svojem delu radi opuščajo profesionalne standarde in etične vrednote. Pomemben del naloge se ukvarja z možnimi načini ubranitve pred manipulacijo, kar je običajno spregledana tema. Omejevanje manipulacije je možno na sistemski ravni, pomembno pa je že njeno prepoznavanje. Tega se lahko lotimo s filozofskimi orodji in s tem, da vzpodbujamo bralce, gledalce in poslušalce novic in prispevkov h kritičnemu mišljenju in pretresu, hkrati pa vztrajamo pri tem, da ima javnost pravico biti obveščena o temah, ki jo zadevajo, in to na pravilen in ustrezen način.

Media manipulation has become a current theme of contemporary research. Various media manipulation strategies that influence public and mass media are presented in the thesis. Sensationalism and spectacle are a part of manipulation and they have replaced the virtues of objectivity and veracity in journalism. The thesis also examines (manipulative) influence of advertising and PR in contemporary journalism. Furthermore, the situations in which the power of capital influences public goods are also introduced. It is presumed that some media have changed their role from an informant to the seller of information. Journalists who do not do their job professionally and disregard ethical standards and norms could also be blamed for this shift in media role. Different ways of protection against media manipulation are also presented in the thesis. Namely, this is usually a neglected topic. Control over the manipulation by the media and its restriction is possible on the level of the system, however its identification is of the vital importance. The process of the identifying media manipulation is possible with philosophical instruments and by encouraging readers, viewers and listeners of news, and articles to process information with a critical approach and analysis. We insist that the audience has the right to be well-informed about current events.

En terme de performance, le biocapteur idéal doit avoir très grande sensibilité, basse limite de détection et temps d’analyse qui est extrêmement court. Les biocapteurs sans marquage à base de résonance de plasmons de surface (biocapteurs SPR) possèdent naturellement le temps d’analyse le plus court parmi différent types de biocapteurs. Leur limite de détection n’est cependant pas la plus impressionnante. Il y a donc un besoin pour augmenter considérablement la sensibilité intrinsèque des biocapteurs SPR afin de permettre de plus basses limites de détection. Quelques approches pour exalter la sensibilité optique des biocapteurs SPR dans la configuration « traditionnel » de Krestchmann telles que film SPR bimétallique, plasmons à longues portées et détection dans l’infrarouge proche sont examinées dans ce travail. Des configurations « non traditionnelles » comme guides optiques planaires avec couplage par réseau et structures sub-longueur d’ondes ont été aussi théoriquement étudiées. Nouvelle stratégie de fonctionnalisation de surface à base de graphène qui augmente la sensibilité de reconnaissance biomoléculaire et peut être appliquée à quasiment toute structure SPR a été également démontrée.

In terms of performance, the ideal biosensor should have high sensitivity, low limits of detection, and extremely short analysis time. Label-free surface plasmon resonance (SPR) biosensors naturally offer the shortest analysis time compared to other types of biosensors. On the other hand, the limits of detection of SPR biosensors are not the most impressive. The inherent sensitivity of SPR biosensors thus needs to be significantly improved to allow lower limits of detection. Several approaches for the enhancement of optical sensitivity of SPR biosensors in the “traditional” attenuated total reflection (ATR) Kretschmann configuration such as the use of bimetallic SPR film, long-range surface plasmons, and near-infrared operating wavelength have been investigated in this work. In addition, some “non traditional” configurations for SPR biosensors including grating-coupled planar optical waveguides and arrays of sub-wavelength structures have been theoretically studied. Novel graphene-based surface functionalization strategy with enhanced biorecognition sensitivity that can be applied to virtually any SPR structure has also been demonstrated.

▼ This work deals with the qualitative and quantitative investigation of the interactions of proteins and their specific ligands and other nonspecific molecules. In the first part, the interactions between fluorescent biotin and galactose derivatives and different fluorescent proteins in solution and on glass surfaces were investigated. For this purpose, fluorescent biotin and galactose derivatives were synthesized and characterized. Furthermore, the immobilization of these derivatives was optimized and the interactions of specific ligands with different proteins were examined via fluorescence measurements. We were able to show that the labeled and covalently bound ligands still exhibit binding activity towards their specific proteins. This was proven by a successful energy transfer in solution between fluorescently labelled biotin and streptavidin coated QDot 605. At the glass surfaces we could clearly show the binding of streptavidin coated QDot 605 to a biotin presenting surface by using fluorescence measurements. In the second part of this thesis, the same ligand-protein systems were used. Here we wanted to introduce hyperbranched polyglycerol (hPG) as a scaffold for ligand presentation to improve the protein resistance of surfaces. Biotin- and galactose-hPG derivatives with different degrees of functionalization were synthesized and characterized. The hPG-derivatives were immobilized under optimized conditions onto glass and gold surfaces. Their behavior towards different proteins was investigated with fluorescence measurements and surface plasmon resonance (SPR) spectroscopy. Herein we were also able to prove that covalently bound ligand-hPG derivatives still exhibit binding activity to their specific protein. At the gold surfaces we could verify ligand density dependent protein binding for galactose and biotin presenting surfaces with the help of surface plasmon resonance (SPR). It is known that lectines and sugars interact only weakly and therefore we were not able to prove protein binding to the lactose-functionalized glass surfaces. In case of biotin- functionalized glass surfaces, we were able to detect a strong binding of FITC-streptavidin by fluorescence microscopy. Furthermore, we were able to demonstrate a significant reduction of the interactions of unspecific proteins with hPG-ligand-functiona¬lized compared to ligand-functionalized surfaces. Immer mehr Kinder fangen schon früh in ihrer Kindheit oder spätestens am Anfang der Grundschule an, Fremdsprachen zu lernen, denn die Studien zeigen, je früher man mit dem Fremdsprachenlernen beginnt, desto besser. Damit das Lernen der Sprachen den Kindern interessant ist, ihnen Spaß macht und möglichst effektiv ist, verwenden Lehrer bei der Arbeit verschiedene Medien. Eine der vielen Arten von Medien sind auch Handpuppen, die allen gut bekannt sind. Da das Thema sehr aktuell ist, war das Ziel dieser Magisterarbeit zu erforschen, wie es mit der Verwendung von Handpuppen in der Praxis der Fremdsprachenlehrer des frühen Fremdsprachenlernens ist. Im theoretischen Teil der Arbeit wurde der Begriff ‚Puppe‘ anhand der Fachliteratur definiert und die Bedeutung von Handpuppen für das Kind veranschaulicht. Ebenso wurden verschiedene Arten von Puppen aufgezählt und beschrieben sowie verschiedene Funktionen von Handpuppen beim Unterricht und auch beim frühen Fremdsprachenlernen genauer dargestellt. Zum Schluss wurden die Handpuppen in zwei Gruppen geteilt: diejenigen, die für die Arbeit beim Unterricht geeignet sind, und diejenigen, die es nicht sind. Im empirischen Teil der Arbeit wurde die Untersuchung dargestellt. Diese wurde mithilfe einer Umfrage durchgeführt, in der Fremdsprachenlehrer mitgemacht haben. Die gesammelten Daten, die analysiert und interpretiert wurden, sind dargestellt. Die Umfrage hat gezeigt, dass Fremdsprachenlehrer des frühen Fremdsprachenlernens mit Handpuppen arbeiten. Diese verwenden sie am meisten für die Motivation am Anfang der Unterrichtsstunden und für Dramatisierungen von Geschichten und Märchen. Es wurde auch festgestellt, dass sie von den Puppen am meisten die Handpuppe verwenden und dass sie das Schattenspiel, die Stockhandpuppe und die Marionette nur selten benutzen. Aufgrund der Analyse der Ergebnisse wurden verschiedene Empfehlungen und Hinweise für die zukünftige Arbeit mit Handpuppen formuliert.

Vedno več otrok se tuji jezik začne učiti že zelo zgodaj v otroštvu ali najkasneje na začetku osnovne šole, saj študije kažejo, da prej kot začneš z učenjem tujih jezikov, toliko bolje. Da je učenje le-tega otrokom čimbolj zanimivo, zabavno ter učinkovito, uporabljajo učitelji pri delu različne medije. Ena izmed mnogih vrst medijev so ravno lutke, ki so vsem še kako dobro poznane. Ker je tema o lutkah zelo aktualna, je bil namen magistrskega dela ugotoviti, kako je z uporabo lutk v praksi učiteljev zgodnjega poučevanja. V teoretičnem delu naloge je na podlagi različne strokovne literature opredeljen pojem „lutka“ in ponazorjen pomen lutk za otroka. Naštete in opisane so tudi različne vrste lutk ter podrobneje predstavljene različne vloge lutk pri pouku kot tudi pri samih urah zgodnjega učenja tujih jezikov. Na koncu teoretičnega dela so vrste lutk, razdeljene v dve skupini: tiste, ki so priporočene za delo v razredu, in tiste, ki niso. V empiričnem delu naloge je predstavljena raziskava, ki je bila izvedena s pomočjo anketnega vprašalnika, v katerem so sodelovali učitelji tujega jezika.…

Opinnäytetyö toteutettiin yhteistyössä Suomen Punaisen Ristin kanssa. Opinnäytetyön tarkoituksena oli kuvata sitä, millaisen koulutuksen Suomen Punaisen Ristin palveluksessa olevat sairaanhoitajat saavat ennen katastrofialueelle lähtöä. Lisäksi opinnäytetyössä selvitettiin sairaanhoitajien kokemuksia heidän saamastaan koulutuksesta ja sen merkityksestä katastrofityöskentelyssä. Tavoitteena oli kerätä tietoa siitä, millaisena sairaanhoitajat kokivat Suomen Punaisen Ristin koulutustilaisuudet ja koulutukset sekä järjestön toiminnan. Opinnäytetyö toteutettiin kvalitatiivisella tutkimusmenetelmällä. Aineistonkeruumenetelmänä käytettiin teemahaastattelua. Haastatteluun osallistui neljä Suomen Punaisen Ristin delegaattina työskentelevää sairaanhoitajaa. Opinnäytetyössä käytettiin aineistolähtöistä sisällönanalyysiä. Opinnäytetyön tulokset osoittivat, että sairaanhoitajat suorittivat Punaisen Ristin peruskoulutukset ja jatkokoulutuksilla erikoistumisen delegaatiksi työskentelyä varten. Pohjakoulutusten jälkeen delegaatit lisäsivät osaamistaan kouluttautumalla erilaisin kurssein. Tuloksista kävi ilmi, että sairaanhoitajat kokivat koulutukset laadukkaina, monipuolisina sekä tarpeisiin vastaavina. Tulosten mukaan delegaattien suorittamilla koulutuksilla oli keskeinen merkitys katastrofityöskentelyssä. Lisäksi tuloksista nousi esiin delegaatin sairaanhoitajan työssä hankkiman työkokemuksen merkitys katastrofityöskentelyssä. Tulosten mukaan koulutukset tarjosivat delegaateille konkreettista tietoa sekä toimintatapoja katastrofityön tekemiseen. Suomen Punainen Risti tarjoaa laadukkaita ja delegaattien tarpeita monipuolisesti huomioon ottavia koulutuksia. Opinnäytetyön tuloksia Suomen Punainen Risti voi hyödyntää toimintansa kehittämisessä. Kehittämisehdotuksia Suomen Punaiselle Ristille ovat yksinhuoltajavanhemman tukeminen delegaatin työssä, delegaatin urasuunnittelun parantaminen humanitaarisen työn puolella, koulutusten aikataulutuksen kehittäminen ja paljon tietoa sisältävän koulutuksen jakaminen useampaan koulutukseen.

This study was carried out in cooperation with the Finnish Red Cross. The purpose of this study was to describe what kind of education nurses working for the organisation are given before going to work in a disaster area. This study also charted out the experiences of nurses about the education they were given and its significance in disaster work. The objective of this study was to gather information about how the nurses experienced the received education, courses and their contents and how they saw the actions of the Finnish Red Cross as an organisation. This study was carried out using a qualitative method. The data were collected by thematic interviews of four nurses working as Red Cross delegates. The data were analysed using inductive content analysis. The results suggest that the nurses completed the basic training organised by the Red Cross and specialised by special educations for working as a delegate. ▼ Meine Arbeit beschäftigt sich mit dem deutschsprachigen Dramatiker Ödön von Horváth (* 09.12.1901 Fiume, heutiges Rijeka; † 01.06.1938 Paris). Dabei lege ich ein besonderes Augenmerk auf die Darstellung des Kleinbürgertums zwischen den beiden großen Weltkriegen, welches Horváth in seinen Dramen und Prosatexten anhand seiner Protagonisten beschreibt. Das eigentliche Ziel meiner Arbeit ist es jedoch, ausgesuchte Themen, die Horváth in seinem Werk vor dem Hintergrund der Zwischenkriegsgesellschaft aufzeigt und dramaturgisch, sprachlich, allegorisch und rein inhaltlich verarbeitet, zu analysieren und sie in den Kontext der heutigen Zeit zu stellen, d.h. mit aktuellen Problemen von heute in Beziehung zu setzen, um somit gleichzeitig Bezüge herzustellen, die die zeitliche Unabhängigkeit der Dramen Horváths unterstreicht. Unter zeitlicher Unabhängigkeit verstehe ich den Sinn und Zweck, welche die Existenz der Dramen Horváths auf den aktuellen Spielplänen der Theaterhäuser rechtfertigen und dadurch die Bedeutung dieses Autors untermauern, der nach seinem Tode lange in Vergessenheit geraten war. Meine Arbeit soll folglich keine Milieu-Studie über das Kleinbürgertum der 30er Jahre sein, wobei Kenntnis über diese gesellschaftliche Formation unabdingbar ist, um meine These in die historischen und gesellschaftlichen Umstände der damaligen Zeit einzuordnen und um gleichzeitig Parallelen, bezogen auf die heutige Kultur und Zivilisation zu finden. In meiner Arbeit soll jedoch auf dieses Basis-Wissen aufgebaut werden, um somit einen wesentlichen neuen Beitrag zur Horváth-Forschung zu leisten, der die Aktualität und Wichtigkeit der Werke dieses früh verstorbenen Autors deutlich rechtfertigt. Als Grundlage der Betrachtung dienen die drei Dramen: „Geschichten aus dem Wiener Wald“, „Glaube Liebe Hoffnung“ und „Kasimir und Karoline“, die gleichzeitig zum Verständnis des künstlerischen Schaffens Horváths herangezogen werden. Diese Dramen werden oft auch als Horváths Hauptdramen bzw. große Dramen bezeichnet, da sie heute aufgrund ihrer ständigen Präsenz auf deutschsprachigen Bühnen zur Bedeutung und Namhaftigkeit Horváths als wichtigen Dramatiker nachhaltig beitragen und ihn als „modernen Klassiker“ des deutschsprachigen Theaters auszeichnen. Diese Dramen dienen mir und dem Leser als strukturelle Hilfe und sind quasi der rote Faden, der durch die ersten beiden Teile meiner Arbeit führt. Im ersten Teil meiner Arbeit gehe ich anhand der Darstellung des Menschenbilds Horváths auf den aktuellen Stand der Horváth-Forschung ein. Ein Schwerpunkt liegt dabei in der Darstellung des damaligen Kleinbürgertums, welches die Nachwehen des Krieges existenziell und seelisch zu verkraften hat, sich für den kommenden Krieg rüstet und das damit einhergehende Leid und soziale Elend durch die Verwendung des „Bildungsjargons“ sprachlich rechtfertigt. ▼ Wie bisherige Forschungsergebnisse zeigen, scheint ein unregelmäßiger Frühstücksverzehr bzw. das vollkommene Auslassen des Frühstücks bei Kindern und Jugendlichen möglicher-weise mit einem erhöhten Risiko für Übergewicht und Adipositas einherzugehen und sich negativ auf bestimmte lebensstilassoziierten Faktoren auszuwirken. Die vorliegende Studie setzte hier an, indem sie den Frühstücksverzehr von Schülerinnen und Schülern der 5. bis 8. Schulstufe untersucht hat, sowie auch den Zusammenhang mit einem täglichen Obst- bzw. Süßigkeitenkonsum sowie der täglichen Schlafdauer und der körperlichen Aktivität. Insgesamt haben 299 Mädchen und 353 Burschen im Alter zwischen 10 und 16 Jahren an der vorliegenden Studie im Zeitraum zwischen April bis Mai 2017 teilgenommen. Knapp zwei Drittel (65,8 %) der Teilnehmenden wiesen einen BMI im Normalbereich auf, während bei jeweils 28,8 % Übergewicht bzw. Adipositas und extreme Adipositas beobachtet werden konnte. 29,7% der Schüler gaben an täglich Obst und Gemüse zu sich zu nehmen, bei den Mädchen lag der Anteil bei 42,1 %. 66 Die Ergebnisse zeigten zudem, dass 19,4 % der Teilnehmerinnen und 10,5 % der Teil-nehmer täglich Süßigkeiten und Softdrinks konsumieren. Durch Angabe der zum Frühstück verzehrten Lebensmittel konnten 4 Frühstückstypen unterschieden werden, wobei ein energiereiches Frühstück die höchste Rate an körperlichen Aktivität aufwies (5,2 Tage pro Woche). Ein hingegen aus vorwiegend pflanzlichen Lebensmitteln bestehendes Frühstück war überraschenderweise zu 17,9 % mit dem höchsten Adipositas-Anteil unter allen Teilnehmenden assoziiert. Schülerinnen und Schüler, die ihre erste Mahlzeit meistens zur Gänze ausfallen lassen, wiesen in der vorliegenden Stichprobe den höchsten Anteil an Übergewicht und die zweit-höchste Rate an Adipositas auf. Zudem konnte gezeigt werden, dass die Teilnehmerinnen und Teilnehmer dieser Gruppe den höchsten täglichen Süßigkeiten- und Softdrinkkonsum, sowie die geringste durchschnittliche Schlafdauer angaben.

▼ This research is motivated by the need to understand and parameterize mixing processes that occur during degassing and refilling of oil in large underground strategic petroleum reserve SPR caverns. The oil is pumped from cavern bottom to a degassing station and then returned to the cavern by using a vertical turbulent jet located near the cavern top. This dissertation concerns a laboratory experimental and theoretical modeling program conducted to investigate: (a) mixing mechanisms and precession of turbulent jets in homogeneous fluids (b) mixing of turbulent jet in stratified fluids and © wall attachment of offset jet in a homogeneous fluid. In the first part, a round turbulent axisymmetric jet is discharged in a long cylinder. It is found that the flow does not reach a true steady state, but vacillates periodically. Digital video recordings and particle image velocimetry are used to map the flow structures and velocity/vorticity fields, from which the frequency of jet switching, jet stopping distance, mean flow, turbulence characteristics and the influence of end-wall boundary conditions are inferred. The results are parameterized using the characteristic length D and velocity J^ (½)/D scales based on the jet kinematic momentum flux J and cylinder width D. The scaling laws so developed could be used to extrapolate laboratory observations to SPR flows. In the second part, a turbulent (positively or negatively) buoyant jet is injected vertically into a slender cylinder containing a stratified fluid. The interest is the vertical density distribution in the container and its dependence on time and other parameters. For each case (lighter or heavier jet) the experimental data, when properly non-dimensionalized, could be collapsed into a ‘universal’ time dependent behavior. A theoretical model is advanced to explain the results. Finally, a round turbulent offset jet in a low-aspect ratio cylinder is investigated. Particle Image Velocimetry and flow visualization are used for flow diagnostics. The measurements include the jet penetration (mixing) depth l, jet spreading rate and the mean velocity/vorticity fields for different offset positions ‘Delta’. With the introduction of offset, the flow patterns change drastically. For 0 < Delta/D < 0.2, the jet deflects toward the wall while precessing (as in the axisymmetric case), for 0.2 < Delta/D < 0.4 the jet hugs the wall but with an oscillating tail, and for 0.45 < Delta/D the jet appears as a wall jet. In all cases, the jet is destroyed at a certain distance (mixing or penetration depth) from the origin. This mixing depth takes its lowest value for 0 < Delta/D < 0.2, with l ≈ (3.2-3.6)D, becomes maximum at Delta/D equal to 0.4 with l ≈ 5.2D, and drops to l ≈ 4.5D when the jet is close to the wall. ▼ El TNFα es una citoquina proinflamatoria con diversas funciones en el organismo. Está implicada en múltiples acciones fisiológicas como la hematopoyesis, el sistema inmune y la regresión tumoral. Debido a su importancia, cuando algo falla en su regulación o funcionamiento, tiene graves y diversas consecuencias patológicas. Por ello, es la molécula diana de muchos tratamientos efectivos para enfermedades inflamatorias como el síndrome de Crohn, la aterosclerosis la artritis reumatoide. Sin embargo, a día de hoy no se realiza de manera rutinaria su determinación para el diagnóstico en análisis clínico. Ni siquiera se lleva a cabo el seguimiento de los niveles de esta molécula en pacientes tratados con fármacos terapéuticos anti TNFα. Las únicas técnicas actuales para medir su concentración son la ELISA y las técnicas basadas en ella sólo se utilizan en estudios de investigación debido a su elevado coste. Por un lado, este trabajo ha pretendido impulsar el conocimiento sobre el TNFα mediante una revisión exhaustiva de la bibliografía recogida hasta ahora en artículos y libros científicos. Por otro lado, se ha caracterizado su detección diseñando un protocolo para la formación de una estructura de reconocimiento biológico de la citoquina con el uso de las SAMs mediante técnicas de resonancia plasmónica superficial (SPR). Para ello, se han optimizado todos los parámetros del procedimiento paso por paso. Se ha comenzado con el tipo de SAM, concentraciones y técnicas de formación estudiadas. Después, se ha analizado la adición de distintos bloqueantes de oro y desactivadores de SAM al proceso con el fin de minimizar las uniones inespecíficas. Se ha continuado con el estudio de las condiciones óptimas para la activación de la SAM y la inmovilización del ligando para poder detectar el máximo de analito posible, probando distintas concentraciones de TNFα con las condiciones óptimas obtenidas. Por último, utilizando la estructura de biorreconocimiento de TNFα óptima, se han analizado diferentes muestras de plasma de ratas sanas y con esteatosis inducida proporcionadas por el Hospital Donostia, pudiendo distinguir entre las que tenían concentraciones basales y elevadas de TNFα en plasma con el sistema diseñado. Además, se ha caracterizado cada paso optimizado con técnicas de diversa índole. Este proyecto ha permitido profundizar en el conocimiento del TNFα, los biosensores, las técnicas libres de marcaje y las monocapas autoensambladas, además de la optimización del protocolo de obtención de una estructura de biorreconocimiento para la detección de TNFα mediante técnicas de resonancia plasmónica superficial, obteniendo el diseño y producción de una SAM efectiva para este caso.
